Enterohemorrhagic Escherichia coli O157:H7 from healthy dairy cattle in Mid-West Brazil: occurrence and molecular characterization. Pesquisa Veterinária Brasileira 34(1):24-28. Departamento de Medicina Veterinária, Centro de Ciências Agrárias e Biológicas, Universidade Federal de Goiás, Rodovia BR-364 Km 192 n° 3.800, Pq. Industrial, Jataí, GO 75801-615, Brazil. E-mail: cissanm@yahoo.com.br Enterohemorrhagic Escherichia coli (EHEC) serotype O157:H7 represents the major Shiga toxin-producing E. coli (STEC) strain related to large outbreaks and severe diseases such as hemorrhagic colitis (HC) and the potentially lethal hemolytic uremic syndrome (HUS). The aim of this study was to report the occurrence and molecular characterization of O157:H7 isolates obtained by rectal swab from 52 healthy dairy cattle belonging to 21 farms in Mid-West of Brazil. Detection of 16SrRNA, stx1, stx2, rfbO157, fliCh7, eae, ehxA, saa, cnf1, chuA, yjaA and TSPE4.C2 genes was performed by PCR. The isolates were further characterized by serotyping. Two hundred and sixty E. coli isolates were obtained, of which 126 were characterized as STEC. Two isolates from the same cow were identified as serotype O157:H7. Both isolates presented the stx2, eae, ehxA, saa and cnf1 virulence factor genes and the chuA gene in the phylogenetic classification (virulent group D), suggesting that they were clones. The prevalence of O157:H7 was found to be 1.92% (1/52 animals), demonstrating that healthy dairy cattle from farms in the Mid-West of Brazil are an important reservoir for highly pathogenic E. coli O157:H7.

#22 - Different pathways in the larval development of the crab Ucides cordatus (Decapoda, Ocypodidae) and their relation with high mortality rates by the end of massive larvicultures, 32(4):284-288

Abstract in English:

ABSTRACT.- Silva U.A.T., Cottens K., Ventura R., Boeger W.A. & Ostrensky A. 2012. Different pathways in the larval development of the crab Ucides cordatus (Decapoda, Ocypodidae) and their relation with high mortality rates by the end of massive larvicultures. Pesquisa Veterinária Brasileira 32(4):284-288. Grupo Integrado de Aquicultura e Estudos Ambientais, Universidade Federal do Paraná, Rua dos Funcionários 1540, Curitiba, PR 80035-050, Brazil. E-mail: ubiratansilva@gmail.com One of the most limiting factors affecting the larval rearing of Ucides cordatus in the laboratory is a period of high mortality, which usually occurs late in the course of the larviculture during the metamorphosis from the zoeal to the megalopal phase. The objective of the present research was to analyze the post-embryonic development of U. cordatus on an individual basis and, in particular, to search for patterns linking disturbances in the molting process to the high larval death rates observed in massive larvicultures. A total of 50 larvae were individually reared from hatching to metamorphosis into the megalopal phase under controlled conditions, fed a combination of microalgae and rotifers. The survivorship rate was 70% until zoea V. The 35 surviving zoea V larvae followed two different pathways. Eleven underwent metamorphosis directly to megalopa, eighteen molted to zoea VI and six died as zoea V. In the last molting event, only two zoea VI larvae reached the megalopal stage, while the remaining sixteen died. In further observation under microscope, 13 of the dead zoea VI showed characteristics of the pre-molt stage and pereiopods disproportionably large in relation to the carapace. The observed pattern resembles the Molt Death Syndrome (MDS) described for other decapod species, in which larvae die in the late pre-molt phase of the molting cycle. We suggest that U. cordatus larvae develop disturbances in the molting process similar to the MDS described for other species and that these disturbances are related to a more complex pathway involving the emergence of larval stage zoea VI.

RESUMO.- Silva U.A.T., Cottens K., Ventura R., Boeger W.A. & Ostrensky A. 2012. Different pathways in the larval development of the crab Ucides cordatus (Decapoda, Ocypodidae) and their relation with high mortality rates by the end of massive larvicultures.

#23 - Profile of central nervous system samples from cattle with neurological syndrome and diagnosis of bovine rabies by the service of sanitary defense of Minas Gerais, 2003-2010, 32(4):333-339

Abstract in English:

ABSTRACT.- Oliveira T.S., Bull V., Rezende C.A., Furtini R., Costa E.A., Paixão T.A. & Santos R.L. 2012. [Profile of central nervous system samples from cattle with neurological syndrome and diagnosis of bovine rabies by the service of sanitary defense of Minas Gerais, 2003-2010.] Postal 567, Pampulha, Belo Horizonte, MG 30161-970, Brazil. E-mail: rsantos@vet.ufmg.br Diseases of the central nervous system (CNS) are often reported in cattle in Brazil. Although the State of Minas Gerais has the second largest cattle herd in the country, there is little information from this state concerning neurological diseases that affect cattle. The Laboratório de Saúde Animal of the Instituto Mineiro de Agropecuária (LSA/IMA) is in charge of the diagnosis of neurological diseases of livestock in the State, with emphasis on rabies and transmissible spongiform encephalopathies. A retrospective study was conducted on data from cattle with neurologic diseases evaluated by the LSA/IMA from January/2003 to June/2010, aiming to determine the profile of CNS samples sent for analysis, with emphasis on the diagnosis of bovine rabies. Issues related to the animal (sex, age, breed and type of death) as well as to the sample (method of conservation, person in charge the collection, and region of the CNS sampled) were evaluated. Data on frequency of rabies positive samples were analyzed by Fisher’s exact test. During the period studied, 3,731 samples from cattle with neurological signs were analyzed, with a predominance of females and crossbred cattle. The method of preservation was the main problem encountered with only 25.89% of samples sent both refrigerated and fixed in 10% formaldehyde. There was a gradual decrease in submission of samples during the course of this study. All 3,703 samples were evaluated by direct fluorescent antibody test (DFA) and biological test (BT) for rabies, 41.58% being positivity for rabies, and 282 of those samples being subjected to histopathology examination. The frequency of positivity was influenced by breed, age, and type of death. Composition of the sample significantly influenced the results, with higher frequency of positivity in samples containing three or more CNS fragments by DFA, BT, or histopathology. The medulla, which is the fragment of choice for diagnosis of BSE, has often been mistakenly submitted under refrigeration, but not in 10% formalin. Cerebellum, thalamus, brain stem, and spinal cord had higher frequency of Negri bodies than the cerebral cortex and trigeminal ganglia. The nonsuppurative inflammatory infiltrate was less frequent in the cerebral cortex than in other CNS fragments. In conclusion, CNS samples from cattle with neurological syndrome sent to the animal health protection service of Minas Gerais are heterogeneous, and the preservation method was the major problem hindering and adequate diagnosis. In addition, diagnosis of rabies was influenced by parameters of the animal as well as the CNS sample. Submission of properly preserved fragments from various segments of the CNS contributes to a more accurate diagnosis of rabies in cattle.

Postal 567, Pampulha, Belo Horizonte, MG 30161-970, Brazil.

#24 - Intra-abdominal pressure in dogs, 31(9):812-816

Abstract in English:

ABSTRACT.- Gonçalves R.P.M., Wouk A.F.P.F. & Mira A. 2011. [Intra-abdominal pressure in dogs.] Pressão intra-abdominal em cães. Pesquisa Veterinária Brasileira 31(9):812-816. Programa de Pós-Graduação em Farmacologia, Laboratório de Disfunções Cardiovasculares, Universidade Federal de Santa Catarina, Campus Universitário, Trindade, Bloco D/CCB, Florianópolis, SC 88049-900, Brazil. E-mail: ronaldmoreno.paiva@gmail.com In order to favor the homogeneity of scientific studies and to provide guidelines on the diagnosis and therapy, human basal values of the intra-abdominal pressure (IAP) and their probable thresholds in morbid cases were recently established. However, basal values and changes in the intra-abdominal pressure in animals are still partly unknown and not scientifically confirmed. Veterinarians have the need to expand their knowledge on morbid conditions that develop with Intra-Abdominal Hypertension (IAH) or Abdominal Compartment Syndrome (ACS). The present study aims to test in dogs the technique adopted in human medicine as a model for measurement of intra-abdominal pressure (1) and determine normal values for the species (2). Fifteen known mongrel healthy dogs, males and females and homogenous were tested. The measurement of IAP was done through an indirect technique which involves catheterizing the bladder and using a water column with a graduate ruler in cm of H20 - its final value converted to mm of Hg. The values observed ranged from a minimum sub atmospheric value (below of zero mm of Hg) to 3,75 mmHg. There was a considerable variation within the individual values observed in some animals, bringing up considerations related to body mass, frequency/total volume of the physiologic solution to be administered and inquietude of these dogs during the treatment. The thresholds values found are considered physiologic, indicating reliability of the technique and possibility of its clinical use. The absence of sedation and the use of urethral catheter did not compromised the measurement, however could have resulted in under- or overestimation of the values found.

RESUMO.- Gonçalves R.P.M., Wouk A.F.P.F. & Mira A. 2011. [Intra-abdominal pressure in dogs.]

#25 - Serological profile, viremia and PCV2 shedding in naturally infected pigs of herds with or without postweaning multisystemic wasting syndrome, 31(1):17-22

Abstract in English:

ABSTRACT.- Pinto F.F., Gerber P.F., Oliveira T.M.L., Borges I.A. & Lobato Z.I.P. 2011. [Serological profile, viremia and PCV2 shedding in naturally infected pigs of herds with or without postweaning multisystemic wasting syndrome.] E-mail: ziplobato@vet.ufmg.br A virological and serological cross-sectional study (E1) and a longitudinal study (E2) were performed on herds with (G2 and G3) and without (G1) post weaning multisystemic syndrome (PMWS) in Brazil. Blood, serum, nasal and rectal swabs samples were collected of sows, farrowing piglets, nursery, growing and finishing pigs. In E1, were sampled 40 animals in each category (G1a and G2). In E2, (G1b and G3), 35 farrowing piglets were identified and sampled along the production cycle. Porcine circovirus type 2 (PCV2) antibodies were assayed. A PCR was used to detected PCV2 genome in blood and swabs. In E1, sows had high rates of viremic and seropositives animals, with percentage of sows with high antibodies titers greater than G2. Passive antibodies decline occurred between nursery and growing area with increased viral shedding in swabs and subsequent seroconversion in G1. In G2, the passive antibodies decay occurred in nursery, with a reduction in viral shedding. In E2, the decline of maternal immunity occurred between the 1st and 2nd collection in G1b, and between 2nd and 3rd collections in G3. In both herds, the decay of passive immunity coincided with increased viremia and viral shedding; and seroconversion occurred between the 3rd and 4th collection in both herds with decline of viremia. Viremia and viral shedding was detected in all samples days, 42% of animals sampled in E2 were viremic and all tissue samples collected at slaughter were positive for PCV2. This study confirms the persistence of viremia even in the presence of high titers of antibodies and the serological profile in a herd with or without PMWS may be different, especially with regard to the passive immunity duration.

E-mail: cintiamev@hotmail.com The objective of this study was to evaluate radiographically and ultrasonographically the podotrochlear apparatus using a transcuneal approach in 22 American Quarter horses suffering from navicular syndrome. On the radiographs, different forms of synovial invaginations, calcification of the podotrochlear bursa and deep digital flexor tendon, enthesophyte and osteophyte formation, and changes in the cortical-medullary margin were observed. On the ultrasound scans, changes of the podotrochlear bursa, adhesions and irregularities at the surface of the deep digital flexor tendon, the digital cushion area reduction, calcification of the impar distal sesamoideal ligament, and irregularities of the flexor surface of the distal sesamoid bone were observed. Radiographic and ultrasonographic abnormalities compatible with navicular syndrome were present in all patients. Ultrasonography using transcuneal approach was a practical and efficient method to evaluate podotrochlear apparatus injuries in equine, thus being a complementary method to radiography.

#27 - Facial paralysis and vestibular syndrome in feedlot cattle in Argentina, 29(11):894-898

Abstract in English:

ABSTRACT.- Odriozola E., Diab S., Khalloub P., Bengolea A., Lázaro L., Caffarena D., Pérez L., Cantón G. & Campero C. 2009. Facial paralysis and vestibular syndrome in feedlot cattle in Argentina. Pesquisa Veterinária Brasileira 29(11):894-898. INTA EEA Balcarce, CC 276, (7620) Balcarce, Argentina. E-mail: eodriozola@balcarce.inta.gov.ar This paper reports 6 outbreaks of neurological disease associated with paralysis of the facial and vestibulocochlear nerves caused by intracranial space occupying lesions in feedlot cattle. The clinical signs observed were characterized by head tilt, uni or bilateral drooping and paralysis of the ears, eyelid ptosis, keratoconjunctivitis, and different degrees of ataxia. Morbidity and mortality rates ranged from 1.1 to 50% and 0 to 1%, respectively. Gross lesions observed included yellow, thickened leptomeninges, and marked enlargement of the roots of cranial nerves VII (facial) and VIII (vestibulocochlear). Histopathologically, there was severe, chronic, granulomatous meningitis and, in one case, chronic, granulomatous neuritis of the VII and VIII cranial nerves. Attempts to identify bacterial, viral, or parasitic agents were unsuccessful. Based on the morphologic lesions, the clinical condition was diagnosed as facial paralysis and vestibular syndrome associated with space occupying lesions in the meninges and the cranial nerves VII and VIII. Feedlot is a practice of growing diffusion in our country and this is a first report of outbreaks of facial paralysis and vestibular disease associated with space occupying lesions in Argentina.

E-mail: aclcamara@yahoo.com.br Pituitary abscess syndrome is a neurologic disease responsible for sporadic cases and outbreaks especially in calves leading to high mortality rates. This paper aimed to report the occurrence and the clinical, laboratorial and pathologic findings in three 8 to 11-month-old calves with pituitary abscess syndrome from Mid-Western Brazil. The most important clinical findings were nervous signs of cerebral and brainstem origin with clinical evolution of 7-20 days. Hematology revealed leucocytosis by neutrophilia and hyperfibrinogenemia. Cerebrospinal fluid analysis showed neutrophilic pleocytosis. Arcanobacterium pyogenes was isolated from cerebrospinal fluid. One calf recovered after antibiotic treatment. Mortality rate was 66.6% (2/3). Necropsy findings included single para-hypophyseal abscesses or located in the glandular parenchyma; one calf showed necrotizing rhinitis and another abscedative pneumonia. Histological exams of the central nervous system reveal complete absence of normal pituitary tissue due to the wide necrosis and neutrophilic inflammatory infiltrate. The authors reiterate the importance of adequate management practices to reduce incidence of several diseases especially in calves, including the pituitary abscess syndrome.

A análise do líquido céfalo-raquidiano apresentou pleocitose neutrofílica. O exame histológico do sistema nervoso central revelou ausência quase completa do tecido hipofisário normal, devido à necrose extensa e infiltrado inflamatório neutrofílico difuso concomitante.

E-mail: lucianelaskoski@hotmail.com The gastrointestinal diseases of horses are aggravated by complications such as laminitis. The laminitis etiopathogeny are connected with lamellar basement membrane degradation by matrix metalloproteinases (MMPs). Inflammatory cytokines and leukocytes enzymes can active MMPs. The object of this study was to evaluate morphological changes on lamellar tissue of horses with colic syndrome and its association with clinical and laboratorial parameters. It was observed intensive destruction of lamellar architecture, mainly on animals with severe physical and laboratorial alterations, such as delayed capillary refill time, congested mucous membrane, tachycardia, hemoconcentration and low count of platelet and leukocytes. The results sign to the most likely moment of development of lamellar tissue injuries in horses with colic syndrome, which can be adopted preventive measures against laminitis.

E-mail: davetpat@ufrgs.br Swine producers have historically resected needle teeth of newborn pigs to prevent potential injuries to the sow mammary gland and faces of littermates. However, the possible impact of this practice on pig performance has been questioned. As part of a study, 280 PMWS affected piglets, with 40-70 days of age, were examined for the presence of dental lesions immediately after euthanasia. Most pigs were confirmed as PMWS by the detection of typical microscopic lesions and immunohistochemical pattern. At least one periapical abscess was observed in 58 piglets (20.7%), of which 12 had multiple abscesses. There were 22 (31.4%), 16 (22.9%), 4 (5.7%), 23 (32.9%), and 5 (7.1%) abscesses in 3rd upper incisives, 3rd lower incisives, upper canines, lower canines, and other teeth, respectively. The higher prevalence of dental abscesses observed in 3rd incisives and lower canines is probably associated with the wider area of grinding or clipping to which these teeth were subjected. Among bacteria isolated from samples collected from 65 abscesses, Streptococcus sp. was the most prevalent bacteria and was present in 21.48% and 27.7% of the aerobic and anaerobic isolates, respectively. In the second most frequent group were coryneform microorganisms, which showed higher rate of isolation under anaerobic atmosphere than in aerobic culture. There was preponderance of Gram-positive isolates. It could not be determined whether these bacterial co-infections predisposed pigs to development of PMWS-type lesions or if were subsequent to the PCV2 infection.
