The disease, which affects mainly weaned about 4 month-old pigs, caused the death of at least 14 animals. The property, where the outbreak occurred, had inadequate sanitary and management conditions. Clinically the disease was characterized by wasting, poor development, cough, tachypnoea, dispnoea, diarrhoea, ataxia, tremors after stimulation, decubitus and convulsions. The course of the disease was acute or subacute. The most important post-mortem findings were enlarged lymphnodes, non-collapsed lungs, with consolidated areas mainly in the cranial lobes. Histological lesions consisted mainly of lymphohistiocytic infiltration with multinucleate giant cells in lymph nodes, spleen, Peyer’s patches, kidney, lung and liver, depletion or lymphoid hyperplasia, as well as lymphohistiocytic interstitial pneumonia and areas of secondary bronchopneumonia. The diagnosis was established through observations of the symptoms and typical lesions, and was confirmed by immunohistochemical examination and PCR. The objective of this study was to characterize the epidemiological, clinical and pathological aspects of the outbreak of PMWS, because of the severe direct or indirect economical losses caused by the disease to the world pig industry.

E-mail: fabiolanc@ig.com.br Lyme disease is a worldwide zoonosis caused by Borrelia burgdorferi (Spirochaetae) and transmitted through ticks (Ixodidae). Many species of domestic and wild animals are affected. The dog is an important reservoir in the domiciliary environment. The disease affects the joints and causes alterations in other organs. Blood serum samples from 143 dogs, living in the counties of Seropédica, Itaguaí and Rio de Janeiro, were analyzed with the indirect ELISA test, to search for antibodies against Borrelia burgdorferi latu sensu. Average positive results were 48.25%, 41.9% in Seropédica, 4.9% in Itaguaí and 1.4% in Rio de Janeiro. The titres varied from 1:400 to 1:6400. The prevalence was lower (p< 0.05) in dogs up to one year of age than in older dogs, with significant difference. Between males and females was no statistical difference with positives titres. The result of this survey should draw attention to the occurrence of Borrelia sp in the area studied, considering the importance of Lyme disease as an emerging zoonosis.

#33 - O surto de peste suína africana ocorrido em 1978 no município de Paracambi, Rio de Janeiro

E-mail: tokarnia@ufrrj.br Due to doubts which still persist 25 years after the outbreak of African swine fever (ASF) which ocurred in the county of Paracambi, Rio de Janeiro, Brazil, in 1978, the results obtained through the studies to establish and confirm the diagnosis are presented. These include data on the epidemiology, clinic-pathological aspects, bacteriological, virological and ultramicroscopic examinations, the experimental reproduction of the disease and cross immunity tests with classical swine fever virus performed in Brazil, and on the confirmation with isolation of the virus and determination of its identity at the Plum Island Animal Disease Center, New York, USA. The pigs of the affected herd had been fed untreated remains of meals from airplanes of international lines flying to Brazil from Portugal and Spain where ASF was occurring at the time. According to publication by the Ministry of Agriculture, after the diagnosis of the outbreak of ASF described in this paper, 223 additional outbreaks were reported in Brazil between 1978 and 1979, in all the Brazilian regions (North, Northeast, Central-West, Southeast and South). Further outbreaks were reported in 1981, but their number is not known. The last case was reported to have occurred on November 15, 1981, and on December 5, 1984, Brazil was declared free of ASF. For the diagnosis of ASF 54,002 samples were examined by the Department of Virology of the Institute of Microbiology, Federal University of Rio de Janeiro, from 1978 to 1981, by the techniques of haemadsorption in leucocyte cultures (HAd), direct immunoflorescence in tissue sections (FATS), direct immunoflorescence in cell cultures (FATCC), immuno-electro-osmophoresis (IEOP) and indirect immunoflorescence assay (IIF). Only 4 samples were positive with the FATCC procedure. This is the only technique which includes virus isolation; the origin of these positive samples was not reported, but probably they were from the Paracambi outbreak. From other suspected outbreaks of ASF in Brazil there is no information on the isolation and characterization of the virus isolates. Likewise there is no information available about the epidemiology, clinical signs, and pathology of suspected ASF in other outbreaks. The analysis of all published data on this matter in Brazil, the possibility of false-positive results, the lack of information about isolation and characterization of the virus, as well as the lack of epidemiological, clinical and pathological data of these other supposed outbreaks of ASF strongly suggest that the outbreak of Paracambi was the only occurrence of ASF in Brazil, confirmed by the isolation, identification of the virus, and the determination of its pathogenicity, and that ASF occurred and maintained itself confined to this area probably due to the early diagnosis and the rapid application of efficient control measures by the Brazilian authorities; the slaughter of the animals in the outbreak of Paracambi started as soon as 10 days after the first death, 3 days after the presumptive diagnosis.

E-mail: adivaldo@ufrrj.br The epidemiology of pulmonary and gastrintestinal helmintoses of calves has been studied in cross-bred Zebu-Friesian calves, 6 to 9 month old, with natural infections. The calves were maintained in permanent pasturing in the lowland, with climate Aw, in the state of Rio de Janeiro. The experiments had the duration of 24 months. At the end the animais were bearer of natural infection of several helminth species. The research was based on fecal analysis and necropsies of at least four calves every 28 days. The animais maintained in permanent pasturing showed tendency to have larger helminth populations in autumn and spring. The graphical representation of an ellipse, obtained through the relationship of total rainfall and average minimum temperature per month, was effective in demonstrating the potential for clinical parasitism in autumn and spring.

E-mail: adivaldo@ufrrj.br Foi estudada a epidemiologia das helmintoses pulmonares e gastrintestinais em bezerros mestiços (Zebu x Holandês) mantidos em regime de pastoreio permanente em região de baixada, correspondente ao clima Aw, no Estado do Rio de Janeiro. Os animais tinham entre 6 e 9 meses de idade, e o experimento teve duração de 24 meses. Os animais eram portadores de infecção natural por diversas espécies de helmintos e o trabalho baseou-se na contagem de ovos por grama de fezes e necropsias de pelo menos quatro animais a cada 28 dias. Observou-se a tendência dos animais abrigarem maiores populações de helmintos nas estações de outono e primavera. Os parâmetros bioclimatográficos representados por elipse, obtida por meio da relação precipitação pluviométrica e temperatura média das mínimas, foram eficientes para demonstrar o potencial de parasitose clínica, a qual correspondeu às estações de outono e primavera.

Pesquisa Veterinária Brasileira 21(2):77-80. Depto Parasitologia Animal, Univ. Fed. Rural do Rio de janeiro, Seropédica, RJ 23851-970, Brazil. This studywas carried out in order to investigate the emergence of adult flies of Haematobia irritans in cattle dung maintained in the field and in the laboratory, as well as other flies associated with dung pats. Two groups of cattle dung were used, one in the field and another in the laboratory, each group consisting of five fresh dung pats ofmedium size (about 30 cm in diameter), opaque appearance, presence of a thin externai crust· and firm consistency. The dung pats were covered with emergence cages in pyramidal format, presenting a hole in the superior part where a removable flask was coupled, and the substitution of the dung was done every 2 weeks. A total of 355 specimens of H. irritans was collected, 151 of which were obtained in the field and 204 specimens in the laboratory. These differences probably were dueto smaller temperature variations in the laboratory than in the field, besides the action of arthropodal predators, competitors and parasites of horn fly pupae that probably interfered in H. irritans emergence in cattle dung under field conditions. A higher number of females were observed in both conditions. Besides H. irritans, there were collected other flies associated with bovine manure belonging to the following families: Aulacigastridae, Muscidae, Psychodidae, Sarcophagidae, Sepsidae, Tachinidae and Ulidiidae. Sepsidae was the most abundant, with 5,224 specimens of the total of 8,928 flies collected, followed by Sarcophagidae with 2,235, Muscidae with 1,357, Aulacigastridae with 54, Psychodidae with 46, Ulidiidae with 6 and Tachinidae with 5 specimens.

Foram coletados um total de 355 espécimens de H. irritans, sendo 151 provenientes das gaiolas mantidas no campo e 204 em laboratório. Estas diferenças deveram-se provavelmente à menores oscilações de temperatura verificadas no laboratório do que àquelas ocorridas no campo, além da ação de artrópodes predadores, competidores e parasitóides que, provavelmente, interferiram na emergência de H. irritans nas fezes bovinas mantidas no campo. Foi observado maior número de fêmeas em comparação com machos em ambas as condições investigadas. Além de H. irritans, obteve-se outros dípteros associados às fezes bovinas, pertencentes as seguintes famílias: Aulacigastridae, Muscidae, Psychodidae, Sarcophagidae, Sepsidae, Tachinidae e Ulidiidae. The slow serum agglutination test was applied to 119 healthy pigs for the determination of the possible presence of anti-Yersinia enterocolitica 0:3 agglutinins. Of the 63.9% reactive animals (&ge; 1 :20), 8.4% presented positive titers (&ge;1 :80), suggesting the presence of this pathogen among swine and consequently an additional public health problem.

Sixteen Yersinia enterocolitica serotype 0:3 strains, isolated from pigs from Rio de Janeiro, have been analyzed for genetic and phenotypic markers of pathogenicity. lt was observed that only 6 strains harbored the p YV ( + 70 kb) plasmid and one strain harbored a small cryptic plasmid of about 8.6 kb. Accordingly only strains harboring pYV were calcium dependent in the MOX medium at 37°C. Twelve strains showed pesticin sensitivity and the esculin reaction was negative in 13 strains. PCR analysis of pathogenicity genes using specific primers showed the presence of the ail gene in 14 strains, the irp2 gene in one and the psaA in none. Most of the strains were resistant to ampicillin and carbenicillin, although they were susceptible to sulfazotrin and cefoxitin. For chloramphenicol, tetracycline, kanamycin, gentamicin and nalidixic acid the results varied among the strains.

Foi observado que apenas 6 cepas possuíam o plasmídio de virulência, pYV ( + 70 kb) e apresentavam dependência ao cálcio no meio MOX a 37°C. Um plasmídio críptico de cerca de 8,6 kb foi encontrado em uma cepa. Doze cepas revelaram sensibilidade à pesticina enquanto que apenas três se revelaram capazes de hidrolisar a esculina. Através de PCR com &quot;primers&quot; específicos, foi constatada a presença dos genes ail em 14 cepas, irp2, em 1 cepa e a ausência de psaA em todas as cepas analisadas. Quanto aos quimioterápicos, a quase totalidade das cepas mostrou-se ao mesmo tempo resistente à ampicilina e carbenicilina e sensível ao sulfazotrin e à cefoxitina. As respostas foram variadas frente ao cloranfenicol, tetraciclina, kanamicina, gentamicina e ácido nalidíxo.


#38 - Cross-reactions between Yersinia enterocolitica serotype 9 and Brucella spp in bovine and swine sera, in the area of Rio de Janeiro

Abstract in English:

The extent of antigen interference between Yersinia enterocolitica 0:9 and Bmcella spp was evaluated in 245 bovine serum samples divided into groups according to status of immunization against brucellosis, and in 119 swine serum samples. The specimens were submitted to the plate serum agglutination test, tube serum agglutination test and to the Rose Bengal test for Brucella spp, and to tube sérum agglutination for Y. enterocolitica 0:9. The immune response to Y. enterocolitica was demonstrated and considered significant in terms of its possible effects on the interpretation of serological tests for brucellosis. Agglutinating titers were dose or even similar from a quantitative viewpoint. Since in most cases it was not possible to determine precisely the etiologic agent, it is clearly necessary to set up a laboratory method for the diagnosis of brucellosis that will permit the distinction of specific infections.

O grau de interferência antigênica entre Yersinia enterocolitica 0:9 e Bmcella spp foi analisado em 245 soros de bovinos discriminados quanto ao estado de imunização contra brucelpse, e 119 soros de suínos. Os espécimens foram submetidos às provas de soroaglutinação rápida, soro-aglutinação lenta e antígeno acidificado para Brucella spp e soro-aglutinação lenta para Y. enterocolitica 0:9. A resposta imune a Y. enterocolitica foi evidenciada e considerada significativa em relação aos seus possíveis efeitos na interpretação dos testes sorológicos para brucelose. Os títulos aglutinantes foram aproximados ou mesmo similares do ponto de vista quantitativo. Como não foi possível determinar com precisão, na maioria dos casos, o agente etiológico, é necessário instituir um método laboratorial de diagnóstico de brucelose que permita distinguir as infecções específicas.


#39 - Virulence factors of Yersinia enterocolitica 0:3 isolated from healthy pigs, Rio de Janeiro

Abstract in English:

Virulence factors were determined in 43 cultures of Yersinia enterocolitica serotype 0:3, biotype 4, phagetype VIII, isolated from apparently healthy pigs. Twenty one isolates were from tonsils, twelve from feces, six from colon contents and four from tongues. The in vitro tests consisted of examining autoagglutination at 36ºC, and absorption of Congo Red. The virulence assays of Y. enterocolitica were conducted by in vivo tests, testing the invasiveness into guinea-pig eyes and detection of heat stable enterotoxin in suckling mice. The results revealed a higher positivity percentage of the autoagglutination test (76.7%), followed by absorption of Congo Red (53.5%), invasiveness into guinea-pig eyes (20.9%) and detection of heat-stable enterotoxin (7.0%). A higher correlation was found in vitro test as compareci to in vivo tests, which characterized 53.5% Y. enterocolitica isolates tested as virulent and which allowed the conclusion that the simultaneous utilization of autoagglutination and absorption of Congo Red tests may be relevant for the identification of pathogenic strains.

Foram pesquisados os fatores de virulência em 43 estirpes de Yersinia enterocolitica sorotipo 0:3, biotipo 4, lisotipo VIII isoladas de suínos sadios, sendo 21 amostras provenientes de amigdala, 12 de fezes, seis de conteúdo de cólon e quatro de língua. A avaliação da virulência foi verificada pela invasibilidade em olho de cobaia, detecção de enterotoxina termoestável em camundongo lactente, auto aglutinação a 36 ºC e absorção do vermelho Congo. Os resultados revelaram um maior percentual de positividade no teste de auto-aglutinação (76.7%), seguido da absorção do vermelho Congo (53,9%), invasibilidade em olho de cobaia (20,9%) e detecção de enterotoxina termoestável (7,0%). Evidenciou-se maior correlação entre os testes realizados "in vitro" quando comparados às provas "in vivo", caracterizando 53,5% das amostra testadas, como virulentas. A utilização simultânea dos testes de auto-aglutinação a 36°C e absorção do vermelho Congo na identificação de cepas patogênicas parecem ser importantes.


#40 - Rumen function tests in goats under pastore conditions in the Baixada Fluminense

Abstract in English:

Ihe research was carried out to determine values for rumen functions in goats under pasture conditions in the Baixada Fluminense, Rio de Janeiro. Ruminal samples were colleted using an esophageal tube, from 16 goats during the month of November (spring). The macroscopic characteristics showed that the variation in color was from green to mit-brown, the smell was aromatic, the consistency was slightly viscous, and there was no sedimetation or floating. In the physicochemical evaluations, the average values were: pH measured with a pH meter 7.13 ± 0.06, and with a special indicator paper 7.04 ± 0.07; methylene-blue reduction, 6.21 ± 0.65 minutes; nitrite reduction, 7.55 ± 0.98, 19.68 ± 2.74, and 31.38 ± 4.38 minutes in the tubes with 0.2, 0.5 and 0.7 ml of KN02, respectively; acidity 18.04 ± 1.63 units; glucose fermentation, 0.21 ± 0.06 and 0.47 ± 0.08 ml gas measured at 30 and 60 minutes; cellulose digestion, 50.84 ± 2.58 hours; and chloride 39.26 ± 2.41 mEq/1. Ruminal rcicrobiotic analysis revealed variations in size, density, and active motility of the protozoa, and a mixed population of bacteria with a predominance of the Gram negative forrns.

Esta pesquisa teve o objetivo de determinar valores para as funções ruminais em caprinos sob condições de pastagem na Baixada Fluminense, Rio de Janeiro. Amostras de fluido ruminal foram coletadas através de sonda esofágica, de 16 caprinos, durante o mês de novembro (primavera). As características macroscópicas mostraram que a variação na coloração foi entre o verde e castanho, o odor foi aromático, a consistência levemente viscosa e não houve sedimentação ou flotação. Nos exames físico-químicos, os valores médios foram: pH medido com pH metro 7,13 ± 0.06 e com papel indicador especial 7,04 ± 0,07; redução do azul de metileno 6,21 ± 0,65 minutos; redução de nitritos 7,55 ± 0,98, 19,68 ± 2,74 e 31,38 ± 4,38 minutos em tubos com 0,2, 0,5 e 0,7 ml de KN02, respectivamente; acidez total titulável 18,84 ± 1,63 unidades; fermentação da glicose 0,21 ± 0,06 e 0,47 ± 0,08 ml de gás medidos em 30 e 60 minutos; digestão da celulose 50,84 ± 2,58 horas e cloretos 39,29 ± 2,41 mEq/1. A análise microbiológica ruminal revelou variações no tamanho, densidade e motilidade ativa de protozoários e uma população bacteriana mista com o predonúnio de formas Gram negativas.
