E-mail: rmbvalenca@ig.com.br This study was aimed to determine the prevalence and to identify risk factors associated with infection by Toxoplasma gondii in pigs from commercial swine farms in the state of Alagoas, Brazil. To compose the sample size of prevalence 342 pigs were used, with 312 sows and 30 boars, from of seven swine farms and distributed in five districts of the state of Alagoas. The serological examination for the detection of anti-Toxoplasma gondii antibodies was achieved by Indirect Immunofluorescence (IFAT), using anti-pig IgG-conjugated to fluorescein isothiocyanate. The analysis of risk factors were performed by the application of questionnaires consisting of objective questions relating to the designer, the general characteristics of the property, the production, reproductive and health management. The prevalence was of 26,9% (92/342) of seropositive pigs. The associated factor was the boars introduction on farms in the last five years (p=0,014; OR=1,83; CI=1,13-2,96). It is concluded that infection with Toxoplasma gondii is widespread in commercial pig farms in the state of Alagoas, Brazil. The results suggest the control of cat population, the performance of cleaning and sanitation practices and laboratory testing to detect of infection by T. gondii in animals to be placed on the farms as measures to reduce the infection rates on region studied.

Para compor a amostra do estudo de prevalência foram utilizados 342 suínos, sendo 312 matrizes e 30 varrões, oriundos de sete granjas de ciclo completo e distribuídas em cinco municípios do Estado de Alagoas. O exame sorológico para a pesquisa de anticorpos anti-Toxoplasma gondii foi realizado através da técnica de Imunofluorescência Indireta (RIFI), utilizando-se anticorpos anti-IgG-suíno conjugado ao isotiocianato de fluoresceína. A análise dos fatores de risco foi realizada por meio da aplicação de questionários constituídos por perguntas objetivas referentes ao criador, às características gerais da propriedade, ao manejo produtivo, reprodutivo e sanitário. Determinou-se uma prevalência de 26,9% (92/342) de suínos soropositivos. O fator associado à infecção foi a introdução de reprodutores nas granjas nos últimos cinco anos (p=0,014; OR=1,83; IC=1,13-2,96). Concluiu-se que a infecção por Toxoplasma gondii encontra-se disseminada em suínos criados em granjas tecnificadas no Estado de Alagoas, Brasil. Recomenda-se o controle da população de gatos, a realização de práticas de limpeza e higienização das instalações e realização de testes laboratoriais para diagnóstico da infecção por T. gondii nos animais a serem introduzidos no plantel como medidas de redução dos índices de infecção na região estudada.

E-mail: rinaldo.mota@hotmail.com This paper aimed to identify Toxoplasma gondii infection in house sparrows (Passer domesticus, Linneaus 1758) coming from poultry farms in the “agreste” region of the Brazilian state of Pernambuco. 151 sparrows (Passer domesticus) captured in eight broiler, egg layer and commercial laying poultry farms, were used. Indirect hemagglutination test was used to research anti-T. gondii antibodies. Animals that presented titration of 1:16 were destined to DNA research through Polymerase Chain Reaction (PCR) technique, followed by Nested-PCR. It was observed that, from 151 analyzed samples. 91 (60.3%) were reagents and 60 (39.7%) were not reagents. It was verified, through analysis of the distribution of infected animals frequency per farm, that in only one farm (12.5%) no animal reagent to T. gondii was captured. It was also observed that three (30.00%) of the ten samples destined to DNA research for T. gondii were positive to PCR and four (40.00%) were positive to Nested-PCR. Anti-T gondii antibodies occurrence and the molecular identification of the agent confirmed natural T. gondii infection in sparrows from poultry farms in Brazil. Other studies must be carried out to highlight the real importance of these animals in the epidemiological chain and their efficiency in the transmission of the parasite to felines. Therefore, researches that use parasite isolation and molecular techniques to determine genomic profile of the agent present in these poultry farms are needed.

Para a pesquisa de anticorpos anti-T. gondi utilizou-se o teste de hemaglutinação indireta, aqueles animais que apresentaram titulação 1:16 foram encaminhados para pesquisa do DNA por meio da técnica de Reação em Cadeia da Polimerase (PCR) seguida do Nested-PCR. Das 151 amostras analisadas observou-se que 91 (60,3%) foram reagentes e 60 (39,7%) não reagentes. Na análise da distribuição de freqüência dos animais infectados por granja constatou-se que em apenas uma (12,5%) não foi capturado animal reagente para T. gondii. Das dez amostras que foram encaminhadas para pesquisa do DNA do T. gondii, observou-se que três (30,00%) foram positivas ao PCR e quatro (40,00%) ao Nested-PCR. A ocorrência de anticorpos anti-T. gondii e a identificação molecular do agente confirmam a infecção natural por T. gondii em pardais em granjas avícolas no Brasil. Outros estudos devem ser conduzidos para elucidar a real importância destes animais na cadeia epidemiológica e sua eficiência da transmissão do parasito para felinos. Para tal serão necessárias pesquisas que utilizem técnicas de isolamento do parasito e molecular para determinar o perfil genômico do agente presente nestas granjas.

E-mail: msvaraschin@ufla.br This study determines the seroprevalences of Neospora caninum and Toxoplasma gondii in caprine herds in southern Minas Gerais state and investigates their association with the reproductive performance of the herds analyzed since no data concerning neosporosis in goats in this state are available. Serum samples from 401 goats from 11 farms were subjected to indirect immunofluorescence reaction for the detection of N. caninum and T. gondii. The mean prevalence rates of animals that tested positive were 10.7% and 21.4%, respectively. Significant differences between different age groups were observed. The highest frequencies of positive cases for both parasites were observed in goats over three years of age; in this group, the probability of seropositivity for N. caninum and T. gondii was 2.6 and 4.8 times higher, respectively, when compared to animals up to one year of age. No positive correlation was found between the mean prevalence rate of N. caninum (p-0,553) and T. gondii (p=0.098) and the occurrence of reproductive problems in the herds using T-test. However, farms with a history of reproductive problems exhibited an average of 18.64% animals reactive to N. caninum, compared to 13.97% from farms where those problems were not reported. When reproductive problems were separated in category, the relationship between occurrence of abortion and seroprevalence of N. caninum and T. gondii was significant (p<0.05) using Fisher’s exact test and chi-square test. No difference was seen (p>0.05) between the mean prevalence rate of T. gondii of herds to which cats had access as compared to herds that had no contact with cats using a T-test. However, the mean prevalence rate in farms where cats had access to the herd was 31.01% as compared to 17.34% in farms where there were no cats. Access of dogs was observed in 100% of the farms. The results obtained in this study demonstrate that N. caninum and T. gondii infect goats in southern Minas Gerais and that these parasites can be associated with the occurrence of abortion in this animal species.

Amostras de soro de 401 caprinos provenientes de 11 propriedades foram submetidas à reação de imunofluorescência indireta anti-N. caninum e T. gondii. A prevalência média de animais positivos foi de 10,7% e 21,4%, respectivamente. Houve diferença significativa entre as faixas etárias. Neste quesito os maiores índices de positividade, para ambos os parasitas, foram observados nos caprinos acima de três anos de idade. Estes apresentaram uma probabilidade 2,6 e 4,8 maior, em relação aos animais de até um ano de idade, para N. caninum e T. gondii, respectivamente. No teste T não houve associação positiva entre as médias de prevalência de N. caninum (p=0,553) e T. gondii (p=0,098) com a ocorrência de problema reprodutivo nos rebanhos. Porém, propriedades com histórico de problemas reprodutivos apresentaram uma média de reagentes para N. caninum de 18,64%, se comparadas a 13,97% de criatórios que não relataram essas alterações. Quando os problemas reprodutivos foram separados em categorias a relação entre a ocorrência de aborto e a soroprevalência de N. caninum e de T. gondii foi significativa (p<0,05) por meio dos testes Exato de Fisher e Qui-quadrado. Não houve diferença (p>0,05) entre as médias de prevalência de T. gondii e o acesso de gatos ao rebanho no teste T. Entretanto, a média de prevalência nas propriedades em que os gatos tinham acesso foi de 31,01%, se comparada a 17,34% em criatórios onde não existiam gatos. Foi constatado o acesso de cães em 100% das propriedades. Os resultados obtidos neste estudo demonstram que N. caninum e T. gondii infectam caprinos no Sul de Minas Gerais e que estes parasitas podem estar associados à ocorrência de abortos nessa espécie animal.

E-mail: rinaldo.mota@hotmail.com The aim of this paper was to study the Toxoplasma gondii shedding in the semen of naturally infected rams. Sixty-five rams were initially submitted to anti-T. gondii antibody detection by indirect immunofluorescence (IIF). Serologically positive rams were then submitted to semen collection for T. gondii DNA detection. In the serology, 6/65 (9.2%) rams were positive, while in the nested PCR of semen there were 4/6 (66.6%) positive rams. It can be concluded that detection of the proliferative form of T. gondii in semen of naturally infected rams by the nested PCR technique reinforces the need to investigate possible horizontal transmission of this parasite via semen in sheep.

Foram utilizados 65 reprodutores submetidos inicialmente à pesquisa de anticorpos anti-T. gondii por meio da técnica de imunofluorescência indireta (IFI). Os carneiros sorologicamente positivos foram submetidos à colheita de sêmen para detecção do DNA de T. gondii. Na sorologia observaram-se 6/65 (9,2%) carneiros positivos, enquanto no PCR nested de sêmen 4/6 (66,6%) carneiros foram positivos. Conclui-se que a detecção, por meio da técnica da PCR nested, da forma proliferativa de T. gondii no sêmen de carneiros naturalmente infectados, reforça a necessidade de se pesquisar sobre a possibilidade da transmissão horizontal do parasito via sêmen na espécie ovina.

E-mail: debora@unipar.br This paper aims to analyze the effects of the Toxoplasma gondii infection in the intestinal wall and myenteric plexus of chicken (Gallus gallus). Ten 36-day-old chickens were separated into two groups: control and experimental, orally inoculated with oocysts of the T. gondii strain M7741 genotype III. After 60 days the birds were submitted to euthanasia and had their duodenum removed. Part of the intestinal segments was submitted to histological routine, HE staining, PAS histochemical technique, and Alcian Blue. Qualitative analysis of the intestinal wall and comparative measurements among the groups with respect to total wall thickness, muscle tunic, mucosa, and tunica mucosa were carried out. Caliciform cells were quantified. The other part of the intestinal segments was fixed in formol acetic acid and dissected having the tunica mucosa and the tela submucosa removed. Neurons were stained with Giemsa, counted, and measured. Chickens from the experimental group presented diarrhea and inflammatory infiltrates in the tunica mucosa, thickness reduction of all the parameters assessed in the intestinal wall, and an increase of the number of caliciform cells. There was a ~70% reduction regarding the intensity of myenteric neurons; and the remaining cells presented a reduction of ~2.4% of the perikarion and ~40.5% of the nucleus (p<0.05). Chronic infection induced by T. gondii oocysts resulted in intestinal wall atrophy, mucin secretion increase, death and atrophy of chicken myenteric plexus neurons. Death and atrophy of myenteric plexus neurons may be related with the causes of diarrhea observed in chickens with toxoplasmosis.

Dez galinhas de 36 dias de idade separadas em dois grupos: controle e experimental inoculado com oocistos da cepa M7741 de T. gondii (genótipo III) pela via oral. Após 60 dias os animais foram submetidos à eutanásia e o duodeno coletado. Parte dos segmentos intestinais foi submetida à rotina histológica, coloração por HE e técnica histoquímica de PAS e Alcian Blue. Realizou-se uma avaliação qualitativa da parede intestinal e medidas comparativas entre os grupos da espessura da parede total, túnica muscular, muscular da mucosa e túnica mucosa. As células caliciformes foram quantificadas. Outra parte dos segmentos intestinais foi fixada em formol acético e dissecada retirando-se a túnica mucosa e a tela submucosa. Os neurônios foram corados pela técnica de Giemsa, contados e mensurados. Os animais do grupo experimental apresentaram diarréia e infiltrados inflamatórios na túnica mucosa, redução da espessura de todos os parâmetros avaliados da parede intestinal e aumento do número das células caliciformes. Houve uma redução de ~70% da densidade dos neurônios mientéricos e as células remanescentes sofreram redução de ~2,4% do pericário e ~40,5% do núcleo (p<0,05). A infecção crônica induzida por oocistos de T. gondii levou a atrofia da parede intestinal, aumento da secreção de mucinas, morte e atrofia dos neurônios do plexo mientérico de galinhas. A morte e atrofia dos neurônios do plexo mientérico podem estar envolvidas na causa da diarréia observada em galinhas com toxoplasmose.


#36 - Occurrence of IgG antibodies anti-Neospora caninum and Toxoplasma gondii in female water buffaloes (Bubalus bubalis) raised in the Brazilian state of Pará, 30(5):443-446

E-mail: spatroca@yahoo.com.br To investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.

Obteve-se 153 (40,9%) de animais soropositivos para N. caninum com 100% das propriedades com focos da infecção, enquanto que quatro búfalas (1,1%) foram soropositivas em quatro fazendas para T. gondii e uma búfala (0,27%) foi soropositiva para ambos parasitos. A presença de anticorpos é um indicativo da circulação desses protozoários em búfalos das propriedades estudadas, representando uma fonte de infecção para outros animais, assim como o possível envolvimento em distúrbios reprodutivos nessa espécie.

E-mail: silva_rcd@yahoo.com.br Toxoplasmosis is caused by the obligate intracellular protozoan parasite Toxoplasma gondii and affects warm-blooded vertebrates, including pets and man. Dogs are epidemio-logically important since they act as sentinels for the infection in humans. The present study aimed to determine the presence of antibodies to T. gondii in 205 serum samples from dogs in Ubatuba, Sao Paulo state, Brazil, through indirect immunofluorescence reaction (IFAT), as well as the risk factors related to toxoplasmosis in the animals such as breed, age, sex, access to outdoors, homemade food ingestion, access to untreated water, and contact with rodents. Toxoplasmosis-positive samples accounted for 52/205 (25.4%), with titers ranging from 16 to 256. The serological results presented significant association (P<0.05) with homemade food ingestion (45/118; 38.1%; CI95% 29.9%-47.2%) (OR=7.0; CI95% 3.0-16.6), and with access to outdoors where those that do not have access to the street were prevalent (37/121; 30.6%; CI95% 23.1%-39.3%) (OR=0.5; CI95% 0.2-1.0). These results show that toxoplasmosis in this region is related to problems of sanitary education, mainly concerning the appropriate cooking of foods, since most positive animals did not show significant association with the presence of rodents or untreated water consumption but showed, instead association with ingestion of homemade food. Thus, toxoplasmosis is a public health problem in the studied region, and sanitary measures are needed to control the infection due to the strict relationship between man and dog and the presented risk factors.

Os resultados sorológicos apresentaram associação significativa (P<0,05) com consumo de comida caseira (45/118; 38,1%; IC95% 29,9%-47,2%) (OR=7,0; CI95% 3,0-16,6), e acesso a rua, em que aqueles que não tinham acesso a rua foram prevalentes (37/121; 30,6%; IC95% 23,1%-39,3%) (OR=0,5; CI95% 0,2-1,0). Estes resultados demonstram que a toxoplasmose na região está relacionada com problema de educação sanitária, principalmente quanto ao adequado cozimento dos alimentos, visto que a maioria dos animais positivos não apresentou associação significativa com presença de roedores ou consumo de água não tratada, porém os mesmos permaneciam em casa aos quais fora oferecida comida caseira. Portanto, a toxoplasmose consiste em um problema de saúde pública na região estudada, sendo necessárias medidas sanitárias para o controle da infecção, visto a estreita relação homem-cão e os fatores de risco presentes.

There was positive influence of anti-T. gondii antibody rate due to the older age of the animals and the presence of cats in pens. The risk of transmitting T. gondii to the public health must be considered, once in some properties where the positive serology was verified natural, products such as milk and/or meat are commercialized. Results were discussed at 5% level of significance.

A frequency of 25.5% of serum samples tested positive for dilutions equal to or greater than 1:64. Statistically significant were not observed in relation to sex and age of the animals. The highest antibody level observed was 4,096 (3.0%) and the most frequent was 64 (74%), suggesting that a chronic infection occurred in the population. The high prevalence of the infection found in the swine herds of this region suggests that these herds can be a source of infection for individuals who typically consume this type of raw or badly cooked meat.
