Programa de Pós-Graduação em Ciência Veterinária, Universidade Federal Rural de Pernambuco, Rua Dom Manoel de Medeiros s/n, Dois Irmãos, Recife, PE 52171-900, Brasil. E-mail: whaubtyfranct@yahoo.com.br The objective in the present study was to research the prevalence of anti-Neospora caninum in 812 samples of blood serum of dairy cattle from farms of seven municipalities of microrregions of Itapecuru-Mirim, Middle Mearim and President Dutra, state of Maranhão, Brazil. For the calculation of sample size, it was considered a seroprevalence of 34.7% for N. caninum, with a maximum error of 9.5% and a confidence interval of 95%. To detect antibodies, it was used the technique of Indirect Immunofluorescence (IFI), with the cut-off of 1:200, using as antigen, tachyzoites strain NC-1, maintained in cell culture in the Laboratory of Diagnosis of Parasitism of the Animals, School of Veterinary Medicine of the Federal University of Bahia, Brazil. Of the total samples, it was obtained a prevalence of 50.74%. The titles ranged from 1:200 to 1:6400, distributed as follows: 108 (26.21%) serum samples showed title of 1:200; 132 (32.04%) 1:400; 94 (22.81%) 1:800; 46 (11.16%) of 1:1600; 23 (5.58%) of 1:3200 and nine (2.18%) with titers of 1:6400. Among the microrregiões the Itapecuru-Mirim showed the lowest percentage of animals seropositive (20.69%) and President Dutra the largest (47.66%). It was observed higher prevalence of seropositives in females (46.80%) than in males (52.46%). There was no significant difference (P> 0.05) for the microrregions variables, sex and age. Concluded that the dairy cattle of the regions studied are exposed to infection by N. caninum.

[Equine neosporosis: occurrence of antibodies against Neospora spp. and association between the serological status of the mares and of their offspring.] Roraima 1000, Prédio 44, Camobi, Santa Maria, RS 97105-900, Brazil. E-mail: gugatoscan@hotmail.com Neospora caninum and N. hughesi are protozoa which can infect horses and can cause reproductive and neurological diseases, respectively. The pathogenesis of neosporosis in horses is poorly understood, as well as the sources of horizontal infection of N. hughesi. Furthermore, there are doubts about the role of the vertical transmission of Neospora spp. in maintenance of these parasites in equine populations. In this study, we evaluated: (1) the occurrence of infections by Neospora spp. in a population of mares (in reproductive age) on a farm of Crioula breed horses; and (2) the possible association between the serological status of mares and of their offspring, aiming to investigate, indirectly, the relevance of transplacental transmission for the occurrence of Neospora spp. in these horses. We found a highly significant association between the serological status of mares and their offspring. Although had been exposed to the same environmental risk factors, the descendants of seropositive mares had a higher percentage of seropositivity against Neospora spp. compared to the descendants of seronegative mares. The association between kinship and serological status indicates an influence of vertical (transplacental) infection raising the occurrence of Neospora spp. in the studied equine population.

Roraima 1000, Prédio 44, Camobi, Santa Maria, RS 97105-900, Brazil.

Abstract in English:

ABSTRACT.- Bauermann F.V., Brum M.C.S., Weiblen R. & Flores E.F. 2010. [A monoclonal antibody-based enzyme-linked immunosorbent assay for detection of antibodies to bovine herpesvirus types 1 and 5.] E-mail: eduardofurtadoflores@gmail.com Bovine herpesviruses 1 (BoHV-1) and 5 (BoHV-5) are antigenic and genetically related viruses associated with different clinical syndromes in cattle, including respiratory, reproductive, neurological disease and abortion. Epidemiological studies indicate the widespread distribution of both viruses among Brazilian cattle. Serological diagnosis, that allows the identification of latently infected animals, represents an important tool for individual and herd monitoring. The present article describes the standardization of a monoclonal antibody (MAb)-based immunoenzymatic test (ELISA) for detection of antibodies to BoHV-1 and/or BoHV-5. The initial steps involved the determination of the most suitable MAb, the appropriate dilutions of viral antigen and serum samples, and the cut-off value of the assay. After standardization, the ELISA was validated by testing 506 cattle serum samples previously tested for neutralizing antibodies to BoHV-1 and BoHV-5 by virus neutralizing assay (VN). Comparing to the VN for BoHV-1 antibodies, the ELISA presented sensitivity and specificity of 96.6% and 98.3%, respectively. Positive and negative predictive values were 97.6%, the concordance between the tests was 97.6% and the coefficient of correlation k (kappa) was 0.95, demonstrating an excellent correlation. Comparing to the VN for BoHV-5 antibodies, the ELISA presented 94.3% of sensitivity, 97.9% of specificity, 97.1% of positive predictive value, 95.9% negative predictive value, concordance of 96.4% and kappa coefficient of 0.92. These results demonstrate that the ELISA presents suitable specificity and sensitivity to be used for individual and herd serological diagnosis of BoHV-1 and BoHV-5, thus, representing an alternative for VN assays and imported ELISA kits.

RESUMO.- Bauermann F.V., Brum M.C.S., Weiblen R. & Flores E.F. 2010. [A monoclonal antibody-based enzyme-linked immunosorbent assay for detection of antibodies to bovine herpesvirus types 1 and 5.]

#44 - Occurrence of IgG antibodies anti-Neospora caninum and Toxoplasma gondii in female water buffaloes (Bubalus bubalis) raised in the Brazilian state of Pará, 30(5):443-446

Abstract in English:

ABSTRACT.- Silva S.P., Mota R.A., Faria E.B., Fernandes E.F.T.S., Neto O.L.S., Albuquerque P.P.F. & Dias H.L.T. 2010 [Occurrence of IgG antibodies anti-Neospora caninum and Toxoplasma gondii in female water buffaloes (Bubalus bubalis) raised in the Brazilian state of Pará.] Anticorpos IgG anti-Neospora caninum e Toxoplasma gondii em búfalas (Bubalus bubalis) criadas no estado do Pará. Pesquisa Veterinária Brasileira 30(5):443-446. Laboratório de Investigação e Diagnóstico de Enfermidades Animais, Universidade Federal do Pará, Belém, PA, Brazil. E-mail: spatroca@yahoo.com.br To investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.

#45 - Risk factors and presence of antibodies to Toxoplasma gondii in dogs from the coast of São Paulo State, Brazil, 30(2):161-166

Abstract in English:

ABSTRACT.- Da Silva R.C., Lima V.Y., Tanaka E.M., Da Silva A.V., Souza L.C. & Langoni H. 2010. Risk factors and presence of antibodies to Toxoplasma gondii in dogs from the coast of São Paulo State, Brazil. Pesquisa Veterinária Brasileira 30(2):161-166. Departamento de Higiene Veterinária e Saúde Pública, Faculdade de Medicina Veterinária e Zootecnia, Universidade Estadual Paulista, Distrito de Rubião Júnior s/n, Botucatu, SP 18618-000, Brazil. E-mail: silva_rcd@yahoo.com.br Toxoplasmosis is caused by the obligate intracellular protozoan parasite Toxoplasma gondii and affects warm-blooded vertebrates, including pets and man. Dogs are epidemio-logically important since they act as sentinels for the infection in humans. The present study aimed to determine the presence of antibodies to T. gondii in 205 serum samples from dogs in Ubatuba, Sao Paulo state, Brazil, through indirect immunofluorescence reaction (IFAT), as well as the risk factors related to toxoplasmosis in the animals such as breed, age, sex, access to outdoors, homemade food ingestion, access to untreated water, and contact with rodents. Toxoplasmosis-positive samples accounted for 52/205 (25.4%), with titers ranging from 16 to 256. The serological results presented significant association (P<0.05) with homemade food ingestion (45/118; 38.1%; CI95% 29.9%-47.2%) (OR=7.0; CI95% 3.0-16.6), and with access to outdoors where those that do not have access to the street were prevalent (37/121; 30.6%; CI95% 23.1%-39.3%) (OR=0.5; CI95% 0.2-1.0). These results show that toxoplasmosis in this region is related to problems of sanitary education, mainly concerning the appropriate cooking of foods, since most positive animals did not show significant association with the presence of rodents or untreated water consumption but showed, instead association with ingestion of homemade food. Thus, toxoplasmosis is a public health problem in the studied region, and sanitary measures are needed to control the infection due to the strict relationship between man and dog and the presented risk factors.

E-mail: zacarias@fcav.unesp.br Visceral leishmaniasis is an emergent zoonosis with an increasing number of new cases in Brazil where the domestic dog is an important parasite reservoir in the infectious cycle of Leishmania chagasi. An enzyme-linked immunosorbent assay (ELISA), based upon the use of a total soluble antigenic preparation of L. chagasi, was adapted for the detection of IgM antibodies in the serum of infected dogs. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkboard titrations. The specificity and sensitivity of the ELISA were 100 %. A total of 110 serum samples were taken from dogs in Belo Horizonte, Minas Gerais, Brazil, and examined for anti-L. chagasi IgM antibody by ELISA and indirect fluorescent antibody test (IFAT). About 25% (n=27) of all the dogs tested were found serologically positive for L. chagasi by IFAT, while 89.09% (n=98) were seropositive by ELISA. The results obtained by ELISA and IFAT were significantly different (P<0.01). The combined use of ELISA and IFAT is recommended in order to enable veterinary services to more efficiently detect canine visceral leishmaniasis.

E-mail: franciscojavier.dieguez@usc.es This study was designed to assess the relationship between antibodies against bovine viral diarrhea virus (BVDV) determined in the bulk tank milk (BTM) and the within-herd seroprevalence. We also assessed the efficiency of measuring antibody levels in BTM samples to monitor BVDV infection status in a herd. In the 81 farms included in the study, BTM samples were obtained and blood samples withdrawn from all cattle older than one year. The infection status was then determined in serum and milk using a commercial blocking ELISA based on the detection of anti-p80 antibodies. Apart from these baseline serum and milk samples, another BTM sample was collected from each herd 9 months later, and a third BTM sample obtained 9 months after this. In these second and third milk samples, anti-BVDV antibodies were determined using the same ELISA kit. Statistical tests revealed good agreement between herd seroprevalences (% seropositive animals in the herd) and the antibody levels detected in the BTM samples. During the 18 months of follow-up, the farms with persistently infected cattle at the study outset (14.8% of the herds) showed a significant decrease in BTM antibody titers after virus clearance. Conversely, a significant increase in BTM antibody levels was observed in the herds infected with BVDV during the follow-up period. Our findings indicate that monitoring antibody levels in the BTM is a useful method of identifying changes in the BVDV infection status of a herd.

Evaluation of the indirect fluorescent antibody test and modified agglutination test for detection of antibodies against Toxoplasma gondii in experimentally infected pigs. Pesquisa Veterinária Brasileira 24(4):199-202. Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86050-970, Brazil. E-mail: italmar@uel.br The study determined the sensitivity and specificity of the indirect fluorescent antibody test (IFAT) and modified agglutination test (MAT) for anti-Toxoplasma gondii antibody detection by analyzing sera from 46 experimentally infected pigs. Values for sensitivity were 95.7% (confidence interval 95%: 84.0-99.2%) and for specificity 97.8% (confidence interval 95%: 87.0-99.9%) in both tests. There was an optimum agreement of results between IFAT and MAT evidenced by a Kappa test of 0.86. These results validate these tests for the detection of T. gondii infection in pigs. IFAT and MAT despite methodologies with different characteristics and readings have similar accuracy in pig serum samples.

#50 - Prevalence of antibodies against chicken anaemia virus (CAV) in broiler breeders in Southern Brazil

Abstract in English:

Canal C.W., Ferreira D.J., Macagnan M., Fallavena L.C.B., Moraes H.L.S. & Wald V.B. 2004. Prevalence of antibodies against chicken anaemia virus (CAV) in broiler breeders in Southern Brazil. Pesquisa Veterinária Brasileira 24(2):89-92. Centro de Diagnóstico e Pesquisa em Patologia Aviária (CDPA), Faculdade de Veterinária da Universidade Federal do Rio Grande do Sul, Porto Alegre, RS 91540-000, Brazil. E-mail: claudio.canal@ufrgs.br Chicks infected during the first two weeks of life with chicken anaemia virus (CAV) manifest clinical disease that can be avoided if the breeder hens transfer enough antibodies to their progeny. The objective of the present work was to establish the prevalence and titer of anti-CAV antibodies in some Brazilian broiler hen breeder flocks and verify in which phase of life the birds were infected. A total of 1,709 serum samples from 12 broiler hen flocks vaccinated against CAV and 64 unvaccinated flocks were analyzed for CAV antibodies with an enzyme-linked immunosorbent assay (ELISA). All non-vaccinated breeder flocks were found to be infected with CAV, with 89% of the hens tested presenting antibodies, 52% of these with titers considered high enough to protect their progeny against CAV infection. Likewise, all vaccinated hens had antibody titer to CAV capable of conferring protection to their progeny. Thus, vaccination of hens seems capable of conferring protection to chicks against clinically apparent CAV-associated disease.
