E-mail: rinaldo.mota@hotmail.com This paper aimed to identify Toxoplasma gondii infection in house sparrows (Passer domesticus, Linneaus 1758) coming from poultry farms in the “agreste” region of the Brazilian state of Pernambuco. 151 sparrows (Passer domesticus) captured in eight broiler, egg layer and commercial laying poultry farms, were used. Indirect hemagglutination test was used to research anti-T. gondii antibodies. Animals that presented titration of 1:16 were destined to DNA research through Polymerase Chain Reaction (PCR) technique, followed by Nested-PCR. It was observed that, from 151 analyzed samples. 91 (60.3%) were reagents and 60 (39.7%) were not reagents. It was verified, through analysis of the distribution of infected animals frequency per farm, that in only one farm (12.5%) no animal reagent to T. gondii was captured. It was also observed that three (30.00%) of the ten samples destined to DNA research for T. gondii were positive to PCR and four (40.00%) were positive to Nested-PCR. Anti-T gondii antibodies occurrence and the molecular identification of the agent confirmed natural T. gondii infection in sparrows from poultry farms in Brazil. Other studies must be carried out to highlight the real importance of these animals in the epidemiological chain and their efficiency in the transmission of the parasite to felines. Therefore, researches that use parasite isolation and molecular techniques to determine genomic profile of the agent present in these poultry farms are needed.

Foram utilizados 151 pardais (Passer domesticus) capturados em oito granjas de frango de corte, matrizes e poedeiras comerciais. Para a pesquisa de anticorpos anti-T. gondi utilizou-se o teste de hemaglutinação indireta, aqueles animais que apresentaram titulação 1:16 foram encaminhados para pesquisa do DNA por meio da técnica de Reação em Cadeia da Polimerase (PCR) seguida do Nested-PCR. Das 151 amostras analisadas observou-se que 91 (60,3%) foram reagentes e 60 (39,7%) não reagentes. Na análise da distribuição de freqüência dos animais infectados por granja constatou-se que em apenas uma (12,5%) não foi capturado animal reagente para T. gondii. Das dez amostras que foram encaminhadas para pesquisa do DNA do T. gondii, observou-se que três (30,00%) foram positivas ao PCR e quatro (40,00%) ao Nested-PCR. A ocorrência de anticorpos anti-T. gondii e a identificação molecular do agente confirmam a infecção natural por T. gondii em pardais em granjas avícolas no Brasil. Outros estudos devem ser conduzidos para elucidar a real importância destes animais na cadeia epidemiológica e sua eficiência da transmissão do parasito para felinos. Para tal serão necessárias pesquisas que utilizem técnicas de isolamento do parasito e molecular para determinar o perfil genômico do agente presente nestas granjas.

E-mail: rinaldo.mota@hotmail.com RESUMO.- A paratuberculose (doença de Johne) é uma das doenças de maior importância econômica para ruminantes em vários países e pode representar uma ameaça ao desenvolvimento da pecuária brasileira. É uma doença infecto-contagiosa que provoca enterocolite granulomatosa crônica, incurável e de difícil controle, cujo agente é o Mycobacterium avium subsp. paratuberculosis (MAP). Descreve-se a ocorrência de paratuberculose em um rebanho de búfalos no Estado de Pernambuco, Brasil. Não foi encontrado registro, na literatura, da ocorrência de paratuberculose em búfalos no país. De 100 búfalos, cinco mostravam sinais clínicos característicos da doença. À necropsia de dois animais as lesões estavam restritas ao intestino delgado com evidente espessamento da mucosa, aumento de linfonodos mesentéricos e vasos linfáticos proeminentes e dilatados. À microscopia, observaram-se na mucosa do intestino, infiltrado inflamatório granulomatoso com numerosos macrófagos epitelióides e células gigantes de Langhans, além de bacilos álcool-ácido resistentes (BAAR) visualizados através da coloração de Ziehl-Neelsen (ZN). Nos linfonodos mesentéricos, havia espessamento da cápsula e marcada inflamação granulomatosa. O exame direto pela técnica de ZN para pesquisa do bacilo em esfregaços de fezes, raspado de mucosa intestinal e imprint de linfonodos mesentéricos resultou positivo. A PCR IS900 específico de linfonodo mesentérico e mucosa intestinal revelou amplificação de um fragmento de aproximadamente 110pb, confirmada pela comparação com outras sequências de M. avium subsp. paratuberculosis disponíveis no GenBank.

E-mail: rinaldo.mota@hotmail.com Paratuberculosis (PTB) is a disease of great economical importance for ruminant in several countries and represents a threat to the development of Brazilian livestock. The contagious disease caused by chronic PTB leads to incurable granulomatous enterocolitis of difficult control. PTB is caused by the Mycobacterium avium subsp. paratuberculosis (MAP). No record on the occurrence of paratuberculosis in buffaloes in Brazil could be found. Five of 100 buffaloes in a herd in Pernambuco-Brazil showed clinical signs characteristic of PTB. At necropsy, of two animals the lesions were restricted to the small intestine with thickening and corrugation of the mucosa, increase of mesenteric lymph nodes and prominent lymph vessels. Histopathology revealed granulomatous inflammation infiltrated with numerous epithelioid macrophages, Langhans type giant cells, and clusters of Ziehl-Neelsen (ZN) positive organisms within the intestinal mucosa. In the mesenteric lymph nodes there was thickening of the capsule and marked granulomatous inflammation. Smears of feces and scrapping smears were prepared from intestinal mucosa and cut surface of mesenteric lymph nodes and, stained by the Ziehl-Neelsen method for research of acid fast bacilli, with positive results. Lymph nodes and intestinal mucosa revealed at IS900 specific polymerase chain reaction amplification of a fragment of about 110pb, confirmed by the comparison with other sequences of M. avium subsp. paratuberculosis available in GenBank.

The present paper aimed to accomplish a retrospective study on primary abomasal impaction in 14 cattle from Pernambuco State, northeastern Brazil. Eight moderate cases, without severe abdominal distention and with no rumen compaction, were treated conservatively, and four severe cases, with severe abdominal distention and rumen compaction, were treated surgically. One bull was slaughtered and one cow died without treatment. The greater number of abomasal impaction cases was in Holstein cows with six cases (42.9%), followed by crossbred cattle with five cases (35.8%), and the breeds Brown-Swiss, Nelore and Marchigiana, each with one case (21.3%). The food composition was characterized by low quality fibers and varied greatly among cases. Most frequent clinical signs were apathy, dehydration, hypomotility and ruminal bloat, intestinal hypomotility and scanty or absent feces with mucus. The hematological findings revealed leukocytosis with neutrophilia and hyperfibrinogenemia in most cases. Ruminal fluid analysis showed compromised flora and fauna dynamics and increased chlorine ion concentration. Clinical (4/8) and chirurgical (2/4) recovery rate achieved 50%. The clinical and chirurgical methods remain as viable options for the treatment of light and severe impaction, but the prognosis is always reserved especially when associated to late pregnancy.

In another, 112 (9.28%) cattle out of 1206 were affected, 84 (6.96%) died due to emaciation, and 28 (2.32%) gained weight after the pods had been withdrawn from the feed. Main clinical signs were progressive weight loss, atrophy of the masseter muscles, dropped jaw, tongue protrusion, difficulties in prehending food, tilting the head during mastigation or rumination, salivation, impaired swallowing, and decreased tone of the tongue. The hematology reveals hypoproteinemia and anemia. Gross lesions were emaciation and reduction in size of the masseter muscles, which appear thinner than normal and grayish due muscular atrophy. Degeneration of neurons of the trigeminal motor nuclei, Wallerian degeneration of the trigeminal nerve roots, and muscular atrophy of the masseter muscles with substitution by fibrous tissue were observed on histologic examination. For the prevention of the poisoning is necessary to limit the amount of mesquite beans in animal nutrition. It is also necessary to develop research to determine the economic and sustainability of the use of Prosopis juliflora for animal food, human food or other uses such as charcoal, wood and fuel wood.

Serum samples (n=290) were analyzed to detect Chlamydophila spp. antibodies in 12 farms. Questionnaires were applied to identify risk factors. Frequency of serum-reactive animals were 10.3% (12.0% in ewes and 8.1% in goats) and 1/12 (91.6%) infection focuses were identified. This is the first report on anti-Chlamydophhila abortus antibodies in goats and sheep in Pernambuco and Brazil, respectively. Risk factors associated with goat infection were breed (OR=9.10) and management (OR=6.41). No significant associations in any of the analyzed factors were found for sheep. In summary, Chlamydophila sp. infection is disseminated in sheep and goat herds in the region. Control measures should be established, focusing primarily risk factor identified in this study, to reduce the possibility of infection by the agent.

E-mail: acynnara@gmail.com The objective of this study was to determine normal standards to ruminal fluid characteristics of Santa Inês sheep under pasture conditions in the State of Pernambuco. Fifty samples were collected, using an esophageal tube, during winter (rainy season) and summer (dry season). In the biochemical tests, the average values found in winter and summer were, respectively: pH 6.76±0.21 and 6,59±0.14; methylene blue reduction, 3.20min (±0.76) and 7.76min (±3.00); chloride, 28.14±4,16mEq/L and 24.97±5.65mEq/L; acidity, 21.90±4.38UC and 13.68±2.97UC. Ruminal microbiotic analyses revealed abundant and moderate density of protozoa in winter and summer, respectively. The motility was very active and there were almost 90% of live protozoa in both seasons. Protozoa numbers were 425.373±217.258/mL in winter and 155.375±83.113/mL in summer. There was a mixed population of bacteria with prevalence of Gram-negative forms in both seasons.

ABSTRACT.- Mota R.A., Brito M.F., Castro F.J.C. & Massa M. 2000. [Glanders in horses and mules of the states of Pernambuco and Alagoas, Brazil.] Mormo em eqüídeos nos Estados de Pernambuco e Alagoas_ Pesquisa Veterinária Brasileira 20(4):155-159. Depto Med. Vet., Univ. Fed. Rural de Pernambuco, Rua Dom Manoel de Medeiros s/nº, Dois Irmãos, Recife, PE 52171- 900, Brazil. Based on clinical-pathological, epidemiological and serological aspects, as well as by the isolation of Burkholderia mallei, foci of glanders were diagnosed in equids of the "Zona da Mata" in the states of Pernambuco and Alagoas. Clinically there was hyperthermia, noisy respiration, loss of appetite, progressive loss of weight leading to emaciation, nasal congestion and muco-purulent discharge, erosions, ulcers and scars of the nasal mucosa. The size of submaxillary and cervical lymphonodes was increased, which contained abscesses and fistules exudating purulent material. In the skin, accompanying the lymph vessels, there were firm nodules and abscesses which in chronic cases fistulated and ulcerated Ieaving stellar scars. The histopathology of the nodules at various sites, from the horses as well as from the guineapigs used for the Strauss test, showed granulomatous lesions and pyogranulomas. The results of this study showed that glanders in the region was emerging and further sero-epidemiological studies are urgent for the eradication of the disease.

Com base em aspectos clínico-patológicos, epidemiológicos e sorológicos, bem como pelo isolamento de Burkholderia mallei, diagnosticaram-se focos de mormo em eqüídeos na Zona da Mata dos Estados de Pernambuco e Alagoas. Clinicamente observaram-se hipertermia, respiração ruidosa, inapetência, emagrecimento progressivo até caquexia, congestão nasal e descarga mlicopurulenta, erosões, úlceras e cicatrizes na mucosa nasal. Os linfonodos mandibulares e cervicais superficiais apresentavam-se aumentados de volume, com abscessos e fistulas exsudando material purulento. Na pele, ao longo dos vasos linfáticos, havia formação de nódulos firmes e abscessos que, nos casos crônicos, fistulavam e ulceravam deixando cicatrizes estrelares. A histopatologia dos nódulos de diversos sítios, tanto dos eqüídeos como dos cobaios utilizados na prova de Straus, revelou lesões granulomatosas e piogranulomas. Os dados deste estudo evidenciam a emergência da doença nesta área e permitem recomendar inquéritos soro-epidemiológicos nos Estados de ocorrência dos surtos e nos Estados limítrofes.


#49 - Occurrence of enterotoxigenic Escherichia coli and Salmonella sp in calves in the southern agreste region of the State of Pernambuco, Brazil

Abstract in English:

The diarrheic processes caused by pathogenic enterobacteria are of great economicsanitary importance, with limiting effects on cattle raising. In the present study 106 fecal samples from 52 diarrheic and 54 non-diarrheic calves aged 1 to 90 days were analyzed, in arder to determine the occurrence of enterotoxigenic Escherichia coli (ETEC) and Salmonella sp among calves reared in the agreste region of the state of Pernambuco and to determine the behavior of the isolates in response to 16 antimicrobial agents. The strains were cultured in selective media and submitted to biochemical characterization, serologic identification and detection of heat-stable enterotoxin. E. coli was isolated from all 106 calves studied and five of these strains (4,7%) were proclucers of heat-stable enterotoxin. Salmonellasp was isolated from six (5,7%) samples belonging to serovars Dublin, Muenchen and Infantis. The Salmonella strains were highly sensitive to the drngs tested, whereas the E. co/istrains were more resistent to tetracycline (50,9%), sulfonamide (40,6%), streptomycin (35,9%) and ampicillin (22,6%). Particularly outstancling in the general analysis was the multiresistance of E. coli (52,0%), with a predominance of the tetracycline-sulfonamide association. The present results serve as a warning regarding the incliscriminated use of antimicrobial agents. Low percentages of ETEC and Salmonella in calves show the necessity of other diagnostic procedures to clarify the etiology.

Os processos diarréicos acarretados por enterobactérias patogênicas têm sido incriminados como de grande importância econômica-sanitária, desempenhando um papel limitante na criação de bezerros. Com o objetivo de estudar a ocorrência de Escherichia coli enterotoxigênica e Salmonella sp em bezerros no agreste meridional de Pernambuco e o compottamento dos isolados frente aos antimicrobianos, foram analisadas 106 amostras fecais provenientes de 52 bezerros diarréicos e 54 não diarréicos de 1 a 90 dias de idade. O cultivo das amostras foi realizado em meios seletivos com posterior caracterização bioquímica, identificação sorológica e detecção de enterotoxina termoestável. Para verificação do comportamento dos isolados, foram selecionados 16 antimicrobianos. E. coli foi isolada dos 106 bezerros estudados, sendo cinco (4,7%) produtoras de enterotoxina termoestável e, Salmonella isolada de seis (5,7%) amostras pertencentes aos sorovares Dublin, Muenchen e Infantis. As amostras de Salmonella demonstraram uma elevada sensibilidade frente as drogas testadas, enquanto que as de E. coli revelaram maior resistência à tetraciclina (50,9%); sulfonamida (40,6%); estreptomicina (35,9%) e ampicilina (22,6%). No cômputo geral destaca-se a multirresistência de E. coli (52%), predominando a associação tetraciclina-sulfonamida. Face aos resultados auferidos adverte-se quanto ao uso indiscriminado de antimicrobianos. Os baixos percentuais de ETEC e Salinonella salientam a necessidade de outros procedimentos de diagnóstico visando esclarecer a etiologia das diarréias.


#50 - Sero-epidemiological survey on caprine arthritis-encephalitis in Pernambuco, Brazil

Abstract in English:

An epidemiological survey on dairy goats in the state of Pernambuco, Brazil, was carried out to determine the prevalence of caprine arthritis-encephalitis virus (CAEV) infection. A total of 397 serum samples was examined by the agar gel immunodiffusion test using Maedi/Visna virus antigen. The prevalence was 17.6% (70/397), with at least one positive sample in 19 of 40 studied flocks. Significant differences (X2; p<0.05) were found between the rates of positivity recorded in pure bred (21.1%) (Saanen, Toggemburg and Alpine) and crossbred (9.8%) goats; also between animals older (40.0%) or younger (15.1-16.1%) than 4 and a half years. No differences were observed when the sampled population was grouped according to sex or flock composition (bucks, dams and kids).

Com o objetivo de determinar a prevalência da artrite-encefalite caprina em criações de caprinos leiteiros no Estado de Pernambuco, amostras de soro foram testadas pela imunodifusão em ágar gel, utilizando-se antígeno do vírus Maedi/Visna. A prevalência observada foi de 17,6% (70/397), ocorrendo pelo menos um animal positivo em 19 das 40 criações estudadas. Foi observada maior frequência (X2; p<0,05%) de soropositivos nos animais puros (21,1 %) (Saanen, Toggemburg e Parda Alpina) que nos mestiços (9,8%), bem como o estrato de idade superior a 4 anos e meio (40%) que nos mais jovens (15,1-16,1%). Não houve diferença significativa entre as frequências observadas nos estratos formados segundo os critérios sexo e composição do rebanho (reprodutor, matriz e cabrito(a)).
