#61 - Experimental acute poisoning by bracken fern (Pteridium aquilinum) in cattle, 29(9):753-766

E-mail: claudioslbarros@uol.com.br This experiment was design to address the following points in relation to the acute poisoning by bracken fern (Pteridium aquilinum) in cattle: 1) the severity of the thrombocytopenia in different stages of the intoxication and its relationship to possible deficits in the secondary hemostasis, 2) the relationship between neutropenia and the morphological signs of septicemia occasionally found at necropsy, and 3) the mechanism of anemia and its relationship with blood loss, medium life of erythrocytes and the progress of the disease. The fresh green upper parts of P. aquilinum were fed to four mixed breed calves with average age of 18 months and weights ranging from 190 to 215 kg. A calf of similar age and weight was kept together with the other four under the same conditions, except for the ingestion of P. aquilinum. The four fern-fed calves died with typical features of acute bracken fern poisoning after being fed with the plant for 53-58 days daily doses of 8.0, 8.6, 10.2, and 10.6g/kg body weight totaling at the end of the experiment, respectively, 59.3%, 63.3%, 47.4%, and 47.5% of bracken fern in relation to their body weight. The disease was characterized by fever up to 42.5°C and varying degrees of hemorrhages observed clinically, at necropsy and on histological examination. Death occurred 6-7 days after the onset of fever. The hematological changes consisted mainly of marked thrombocytopenia and neutropenia. Two of the four affected calves had mild anemia. The deficits in secondary hemostasis were mild in each case. There were no significant changes thus permitting to rule out the role of secondary hemostasis in the pathogenesis of the hemorrhages in the bracken fern poisoning. The measurement of fibrin degradation products in the serum showed conflicting results and did no allow for a solid conclusion regarding the role of disseminated intravascular coagulation in the pathogenesis of the hemorrhages in fern poisoning. The cytology and the histopathology of bone marrow from the four poisoned calves revealed marked decrease in the numbers of hematopoietic cells from the three marrow lineages, which characterizes marrow failure due to aplasia; it was concluded that the events of the primary hemostasis due to thrombocytopenia are responsible for the hemorrhages. In blood culture from three affected calves there was growth of Klebsiella oxytoca, Staphylococcus hyicus and Staphylococcus aureus, indicating that septicemia, facilitated by neutropenia could have a role in the death of cattle acutely poisoned due to the ingestion of P. aquilinum. Additional points of the interest in the acute experimental poisoning by bracken fern in cattle reported here were the development of hematuria and the so called laryngeal form.

E-mail: claudioslbarros@uol.com.br Este experimento foi delineado para investigar os seguintes pontos em relação à intoxicação aguda por samambaia (Pteridium aquilinum) em bovinos: 1) a intensidade da trombocitopenia em diferentes momentos da intoxicação e sua relação com possíveis déficits na hemostasia secundária, 2) a relação da neutropenia com as manifestações morfológicas de septicemia ocasionalmente observadas na necropsia, e 3) o mecanismo da anemia e sua relação com a perda de sangue, a vida média eritróide e a evolução da doença. As hastes superiores mais verdes de P. aquilinum foram administradas a quatro bovinos sem raça definida, com idade média de 1,5 ano e pesos entre 190-215 kg. Um bovino de idade e peso semelhantes foi usado como controle e, exceto por não ter recebido P. aquilinum, foi mantido nas mesmas condições que os outros quatro. Os quatro bovinos que receberam a planta morreram com quadro característico da intoxicação aguda por samambaia após receberem durante 53-58 dias, doses diárias de 8,0, 8,6, 10,2 e 10,6g/kg de peso corporal, que totalizaram, ao final do experimento, respectivamente, 112,7, 107,6, 85,7, 90,15 kg da planta, o que corresponde, respectivamente, a 59,3%, 63,3%, 47,4%, 47,5% da planta em relação ao peso dos bovinos. A doença caracterizou-se por febre de até 42,5°C e diversos graus de hemorragias observadas clinicamente, na necropsia e na histopatologia. A morte ocorria 6-7 dias após o início do quadro febril. As alterações hematológicas revelaram trombocitopenia e neutropenia acentuadas. Em dois dos quatro bovinos havia anemia leve. Não houve variações significativas nos tempos de coagulação dos bovinos intoxicados, quando avaliados os fatores de coagulação (secundária), excluindo-se assim a possibilidade da participação de distúrbios da hemostasia secundária na patogênese das hemorragias nessa intoxicação. A determinação dos produtos da degradação da fibrina no soro revelou dados conflitantes, não permitindo concluir se a coagulação intravascular disseminada tem participação na patogênese das hemorragias nessa intoxicação. A citopatologia e histopatologia da medula óssea dos quatro bovinos intoxicados revelaram acentuada diminuição no número de células hematopoéticas das três linhagens medulares, caracterizando insuficiência medular por aplasia; conclui-se que apenas eventos da hemostasia primária devidos a trombocitopenia são responsáveis pelas hemorragias. Na hemocultura de três dos bovinos intoxicados houve crescimento de Klebsiella oxytoca, Staphylococcus hyicus e Staphylococcus aureus, indicando que a septicemia, facilitada pela neutropenia, pode ter participação na causa da morte de bovinos na intoxicação aguda pela ingestão de P. aquilinum. Aspectos adicionais de interesse na reprodução da intoxicação aguda por samambaia em bovinos deste relato incluem o desenvolvimento de hematúria na doença aguda e a apresentação da chamada forma laríngea da doença.

Hepatic fibrosis is characterized by a disruption of normal tissue architeture by cellular lesions, and may alter the GJIC. This work aimed to study the expression and distribution of Cx32 in liver fibrosis induced by the oral administration of dimethylnitrosamine in female Wistar rats. The necropsy of the rats was carried out after five weeks of drug administration. They presented a hepatic fibrosis state. Sections from livers with fibrosis and from control livers were submitted to immunohistochemical, Real Time-PCR and Western-Blot analysis to Cx32. In fibrotic livers the Cxs were diffusely scattered in the cytoplasm, contrasting with the control livers, where the Cx32 formed junction plaques at the cell membrane. Also it was found a decrease in the gene expression of Cx32 without reduction in the protein quantity when compared with controls. These results suggest that there the mechanism of intercellular communication between hepatocytes was reduced by the fibrotic process, which may predispose to the occurrence of a neoplastic process, taken in account that connexins are considered tumor suppressing genes.

The present study consisted in the analyses of the resistance power to the micro-traction of samples of rabbit cortical bones preserved in several means for up to 180 days and fresh. The results showed that the resistance and the preservation time presented an inversely proportional relation, meaning that, the longer the time of preservation, the shorter the physical resistance evaluated in the biomechanical rehearsal of resistance to the micro-traction. The glycerin has presented lower values in relation to the resistance test, showing, after 30 days, only 24.58% of the present power in the fresh bone, and by 180 days, 1.76%. The samples submitted to autoclavation also showed low values by the end of the experiment, while they remained with just 12.31% of the power present in the fresh bone. The bones preserved in homologous plasma, Dakin liquid and the ones cryopreserved showed the best levels of resistance at the end of the experiment, remaining, respectively with 82.47, 70.34 and 66.72% of the maximum power, while compared with the resistance of the fresh bones. The conclusion is that the choice of methodology and time of preservation interfered directly in the biomechanics of the cortical bones, promoting decrease of the resistance capacity to the traction along the period of preservation.

Out of 984 quarter milk (246 cows), 10 (1.0%) were positive for clinical mastitis, 562 (57.1%) for subclinical mastitis and 412 (41.9%) were negative. A total of 81 Staphylococcus spp. isolates were obtained from milk samples from the cows diagnosed with subclinical mastitis. From these, 53 (65.0%) were S. aureus, 16 (20.0%) coagulase-positive staphylococci (CPS) and 12 (15.0%) coagulase-negative staphylococci (CNS). The isolates were further investigated for the presence of toxin genes by multiplex and uniplex PCR. The main gene observed was seg followed by seh, sei and sej. The distribution of these observed genes among the isolates obtained from different areas showed a regional pattern for the SEs. The presence of toxin genes in the strains isolated from bovine milk demonstrates a potential problem for public health.

Morbidity and mortality were 17.9% and lethality was virtually 100%. In 40% of the farms the disease occurred in small outbreaks affecting several cattle per farm and in 60% only one bovine was affected in each farm. Main clinical signs include fever (40-42°C), apathy, drooling, and hemorrhages; the latter were mainly from the gums, nostrils, and gastrointestinal tract. Multiple petechiae were observed in several mucosae and in the skin. Occasionally hematuria and blood in the milk were reported. The disease was invariably fatal after a course of approximately two days. Hematological changes included severe neutropenia, non-regenerative normocytic normochromic anemia and thrombocytopenia. Necropsy findings include hemorrhages o varying degrees in several organs and infarcts in the liver. Both hemorrhages and hepatic infarcts were confirmed histologically; clusters of bacterial rods and thrombosed vessels were associated with the infarcts. Marked bone marrow aplasia was a consistent finding in the four cases in which the marrow was evaluated.

Although R. equi is likely to be found in every horse-breeding farm, the clinical disease is unrecognized in most of them. Capsule components, equi factor, micolic acid and some products encoded by the large 85-90Kb plasmid were described as virulence factors. However, the pathogenesis of R. equi infections and the sensibility of foals are not completely understood. The aim of this study was evaluate the virulence of R. equi isolated from human, horses and environment for mices. Nine strains carrying the 85-90Kb plasmid isolated from foal clinical specimens, one from immunodeficient human patient and six plasmidless strains (four isolated from feces, one from pasture and one from immunodeficient human patient) were inoculated in cyclophosphamide immunossuppressed mice. The pathological changes and viability of R. equi cells in the liver of mice was verified after the 3rd, 6th an 10th day after inoculation for horse and environmental isolates and for R. equi isolates from human patients on the 1st, 3rd and 6th day. During the necropsy procedures, infiltrate of macrophages and pyogranulomatous lesions were detected after the sixth pos-inoculation day in the liver and spleen. In horse isolates, only plasmid positive strains were virulent, but in human isolates both strains (plasmid positive e plasmid negative) were virulent. Both groups of the immunossupressed mice inoculated with R. equi isolated from environment showed pathological changes. All R. equi strains were unable to kill non imunossuppressed mice.

In horse isolates, only plasmid positive strains were virulent, but in human isolates both strains (plasmid positive e plasmid negative) were virulent. Both groups of the immunossupressed mice inoculated with R. equi isolated from environment showed pathological changes. All R. equi strains were unable to kill non imunossuppressed mice.

E-mail: glaukommers@yahoo.com Thirty bovine with neoplasms of the upper digestive tract (UDT) associated with spontaneous ingestion of bracken fern (Pteridium aquilinum) were studied. They were from 27 farms, located in the municipalities of Jaguari (23) and Nova Esperança do Sul (4), Rio Grande do Sul, Brazil. The total cattle population in those farms was 1,090 and large amounts of bracken fern were found in the pastures. Twenty-six of the affected cattle were cows and four were castrated males, 3-13 years of age; most of them were 7-8 years old (46,6%). Clinical signs observed in the affected animals were progressive weight loss, absence of ruminal movements, cough, dysphagia, regurgitation, halitosis, diarrhea, and bloat. Less frequent signs were selective appetite, dyspnea, and salivation. Two bovine died and 28 were submitted to euthanasia in advanced stage of disease and necropsied. The main gross and microscopic alterations were found in identical areas of the UDT. They consisted of papillomas, transforming papillomas, and squamous cell carcinomas (SCCs). Metastases of SCCs to regional lymph nodes and other organs, such as liver and lungs, were also observed (18/30). Twenty-nine bovine had papillomas of various sizes in several areas of the UDT. The digestive papillomatosis ranged from mild (45%), to moderate (38%), to severe (17%). Three developing phases were observed microscopically in the examined papillomas: an early growing phase, a developing phase, and a regressing phase. In 16 cases, there was malignant transformation of papillomas into SCCs. The SCCs were solitary (12/30) or multiple (18/30) and were histologically well, moderately, or poorly differentiated. Grouping the distribution of SCCs of larger extension in the UDT into cranial region (base of the tongue, pharynx/oropharynx, and epiglottis), medial region (esophagus), and caudal region (cardia and rumen), the distribution was cranial in 39%, middle in 16%, and caudal in 45% of the cases. By the same grouping criteria, but considering the total number of times SCCs of varied extensions were diagnosed in the cranial, middle, and caudal regions, the percentages changed to 34%, 26%, and 40%, respectively. The epidemiological and histomorphological evidences found in this study are in agreement with the observations that point out the co-carcinogenesis between bovine papillomavirus type 4 infection and chemicals of bracken fern in the pathogenesis of the SCCs in the UDT of cattle. However, the presence of pre-neoplastic changes and SCCs in situ or in early stages of development, independently of the presence of papillomas, clearly indicates the possibility of development of SCCs from normal epithelium, probably due to the direct action of the chemical carcinogens contained in bracken fern.

[Evaluation of zinc, manganese and iron levels in liver samples of cattle and sheep from various regions of Brazil.] Avaliação das concentrações de zinco, manganês e ferro no fígado de bovinos e ovinos de várias regiões do Brasil. Pesquisa Veterinária Brasileira 18(3/4):107-110. Centro Nacional de Pesquisa de Gado de Corte, Embrapa, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. To complement studies on diseases of cattle and sheep possibly caused by mineral deficiencies, liver samples collected· in several regions of Brazil and which had been analysed for copper and cobalt were now analysed for zinc, manganese and iron. In most regions the levels of zinc were only sporadically low. Only in the States of Ceará and Piauí a greater number of samples had low levels of this mineral, suggesting the occurrence of zinc deficiency, but subclinically, as case histories and clinical pathological studies did not indicate the occurrence of this deficiency. Also the values for manganese were sporadically Iower than those considered normal. Only in the States of Rio de Janeiro (southern part), Espírito Santo (northern part), Ceará (Serra da Ibiapaba) and Amapá, there was a greater incidence of low values of manganese; the sarne reasons mentioned for zinc indicate that deficiency of manganese in these regions occurs subclinically.

Pesquisa Veterinária Brasileira 18(3/4):107-110. Centro Nacional de Pesquisa de Gado de Corte, Embrapa, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. Complementando estudos realizados sobre doenças de bovinos e ovinos possivelmente causadas por deficiências minerais, as amostras de fígado coletadas em diversas regiões do Brasil, que tinham sido analisadas somente para cobre e cobalto, foram agora analisadas para zinco, manganês e ferro. Na maioria das regiões, os valores de zinco só esporadicamente estiveram abaixo do nível considerado normal. Somente dos Estados do Ceará e do Piauí, número maior de amostras apresentou teores baixos para este mineral, sugerindo a ocorrência de deficiência de zinco, mas sob a forma sub-clínica, pois os históricos e estudos clínico-patológicos não indicavam a ocorrência desta deficiência. Os valores de manganês também só esporadicamente estiveram abaixo do nível considerado normal. Somente nas amostras dos Estados do Rio de Janeiro (Sul), do Espírito Santo (Norte), do Ceará (Serra da lbiapaba) e do Amapá, havia uma incidência maior de valores baixos de manganês; os mesmos motivos apontados acima para o zinco, indicam que a deficiência de manganês nestas regiões deve ocorrer sob forma sub-clínica.


#69 - Arterial diffuse intimal thickening associated with enzootic calcinosis of sheep, 18(1):9-15

ABSTRACT.- Vasconcelos R.O., Barros S.S., Russowski D., Grando S.M. & Irigoyen L.F. 1998. Arterial diffuse intimal thickening associated with enzootic calcinosis of sheep. [Espessamento intimal difuso das artérias na calcinose enzoótica dos ovinos.] Pesquisa Veterinária Brasileira 18(1):9-15. Oepto Patologia, Universidade Federal de Santa Maria, 97119-900 Santa Maria, RS, Brazil. Morphometric, immunohistochemical and ultrastructural studies were carried out on the diffuse intimal thickening (DIT) in arteries of 7 sheep with clinical signs of naturally occurring enzootic calcinosis dueto ingestion of the plant Nierembergia veitchii. Arterial lesions consisted of medial deposition of calcium salts and DIT. Calcification of the intima was rare, mild and located near the elastic lamina. By immunohistochemistry a-actin was detected in cells of the media and in cells forming the intimal thickening. Receptors for 1,25(OH)2 vitamin O3 were detected in nuclei of intimal, medial and endothelial cells. DIT was irregularly distributed and was neither proportionally related to the intensity of the underlying mineralization area nor to the thickening of the remaining media. Ultrastructural morphometry in smooth muscle cells (SMCs) of the media and thickened intima revealed, in the latter, an increase of 318% in the volumetric fraction of those organelles involved in synthesis and a proportional decrease in contractile elements when compared to normal values of media cells. There were histological and ultrastructural evidences of modification of SMCs and their migration to the intima, where they proliferated causing DIT. It was concluded that DIT is a consistent component of arteriosclerotic lesions in N. veitchii induced calcinosis of sheep and that the predominant cell in this process is the SMCs originated from its predecessor of the media. It is suggested that the inducing factor for the arterial changes is 1,25(0H)2 03 present in N. veitchii.

Foram feitos estudos morfométrico, imunoistoquímico e ultra-estrutural do espessamento intimal difuso (DIT) das artérias de 7 ovinos com sinais clínicos de calcinose enzoótica espontânea causada pela ingestão da planta Nierembergia veitchii. As lesões caracterizavam-se por deposição de sais de cálcio na média como placas e estrias que, com frequência faziam saliência para a luz, criando rugosidades e irregularidades da íntima. Não foram observadas calcificações na artéria pulmonar e no sistema venoso. Microscopicamente, as calcificações das artérias restringiam-se quase que exclusivamente à média. Na imunoistoquimica, foi verificada a-actina nas células da média e nas do espessamento intimal. Receptores para 1,25(OH)2D3 foram evidenciados nos núcleos das células musculares da média, da íntima e das células endoteliais. As análises morfométricas em microscopia ótica revelaram, nas artérias, DIT irregularmente distribuído sem relação com a intensidade dos processos de calcificação subjacente nem com a espessura da média remanescente. A morfometria das alterações ultra-estruturais das células musculares lisas da média e da íntima espessada, m intimal ostrou que nessas últimas foi verificado aumento de até 318% na fração volumétrica das organelas de síntese em detrimento dos elementos contráteis, quando comparados com os valores das células da média. Essas modificações indicam a transformação das células musculares lisas de um tipo contrátil para células de tipo sintético. Entre essses dois extremos foram evidenciadas diversas formas intermediárias. Foram observadas evidências histológicas e ultra-estruturais da transformação e migração de células da média através das fenestras da lâmina elástica interna, para a íntima, onde proliferam e formam o DIT. Foi concluído que o DIT é um componente constante nas lesões arterioscleróticas nas calcinoses dos ovinos induzidas pela planta N. veitchii e que as células predominantes são células musculares lisas provenientes de predecessoras da média. Foi constatado que o DIT só excepcionalmente apresenta calcificação, vascularização, presença de células espumosas e fendas de colesterol. Não foram vistas áreas de necrose. Sugere-se que o fator indutor das alterações arteriais é o 1,25(OH)zD3 contido em N. veitchii.
