E-mail: felipp.sf@gmail.com Cardiac biomarkers for clinical and experimental heart diseases have previously been evaluated in rabbits. However, several laboratory assays performed and reported with inconsistent results. This study aimed to assess the effects of breed on serum ANP, CRP, and ACE and establish reference interval (RI) for these biomarkers in a large population of healthy rabbits. Ninety-seven adult rabbits from five breeds were included in this study. Assays were performed using specific ELISA commercial kits. The results were statistically analyzed using ANOVA, Tukey test (p<0.05), arithmetic mean, RI of mean, and standard deviation. A significant effect of breed was shown, indicating different RI between breeds for each biomarker. In conclusion, this study demonstrated that breed is an important physiological variable influencing the normal values of cardiac markers in healthy rabbits.

E-mail: felipp.sf@gmail.com Biomarcadores cardíacos têm sido avaliados em coelhos para avaliação clínica e experimental das doenças cardíacas. Entretanto diferentes testes laboratoriais têm sido utilizados e relatados, sem uma confluência de resultados. Os objetivos deste estudo foram verificar os efeitos de diferentes raças de coelhos sobre as concentrações séricas de ANP, CRP e ACE, além de estabelecer intervalor de referência para estes biomarcadores em uma população de coelhos saudáveis. Foram utilizados noventa e sete coelhos de cinco diferentes raças. Os exames foram realizados pela metodologia de ELISA, por meio de kits comerciais específicos. Os resultados foram analisados estatisticamente os testes de ANOVA e Tukey (p<0,05), média aritmética, intervalo de referência da média e desvio padrão. Um efeito significativo da raça foi observado sobre as variáveis estudadas, indicando diferentes intervalos de referência entre as raças para cada biomarcador. Em conclusão, este estudo demonstrou que a raça é uma variável fisiológica importante que influencia os valores normais destes biomarcadores em coelhos saudáveis.


#2 - Lesão vascular mediada pelo receptor AT1 em miocárdio, rins e fígado de ratos, 30(7):605-611

E-mail: mfvailati@yahoo.com.br The systemic aspect of vascular damage induced by angiotensin II (ANG II) has been poorly explored in the literature. Considering the presence of ANG II and its specific receptor AT1, in several organs, all tissues might be potentially affected by its effects. The aims of this study were: To evaluate the early histological changes in the heart, liver and kidneys, produced by ANG II infusion, to evaluate the protective effect of losartan. Wistar rats were distributed into three groups: control (no treatment), treated with ANG II, and treated with ANG II + losartan. ANG II was continuously infused over 72 hours by subcutaneous osmotic pumps. Histological sections of the myocardium, kidneys and liver were stained and observed for the presence of necrosis. There were ANG II-induced perivascular inflammation and necrosis of the arteriolar wall in the myocardium, kidney, and liver by, which were partially prevented by losartan. There was no significant correlation between heart and kidney damage. Tissue lesion severity was lower than that of vascular lesions, without statistical difference between groups. ANG II causes vascular injury in the heart, kidneys and liver, indicating a systemic vasculotoxic effect; the mechanisms of damage/protection vary depending on the target organ; perivascular lesions may occur even when anti-hypertensive doses of losartan are used.

E-mail: mfvailati@yahoo.com.br O aspecto sistêmico da lesão vascular induzida pela angiotensina II (ANG II) tem sido pouco explorada na literatura. Considerando a presença de ANG II e de seu receptor AT1 em diversos órgãos, todos os tecidos poderiam ser potencialmente afetados por esses efeitos. Os objetivos deste estudo foram: avaliar as alterações histológicas iniciais no coração, fígado e rins, produzidas pela infusão de ANG II, e avaliar o efeito protetor do losartan. Ratos Wistar foram divididos em três grupos: controle (sem tratamento), tratados com ANG II, e tratados com ANG II + losartan. A ANG II foi infundida continuamente por 72 horas por meio de mini-bombas osmóticas. Foram realizados cortes histológicos de miocárdio, rim e fígado para coloração e observação para a presença de necrose. Observou-se a presença de inflamação perivascular e necrose de parede arteriolar em miocárdio, rins e fígado, que foram parcialmente prevenidas pelo losartan. Não houve correlação significante entre as lesões observadas no coração e nos rins. A severidade da lesão tissular foi menor quando comparada às lesões vasculares, sem diferença estatística entre os grupos. A ANG II causa injúria vascular no coração, rins e fígado, sugerindo um efeito vasculotóxico sistêmico; os mecanismos de lesão/proteção variam dependendo do órgão afetado; as lesões perivasculares podem ocorrer mesmo quando doses anti-hipertensivas de losartan forem utilizadas.

E-mail: marinabrito@usp.br Duchenne muscular dystrophy (DMD) is a neuromuscular disorder characterized by a continuous muscle necrosis and degeneration with eventual fibrosis and fatty tissue infiltration. Progressive increase in muscle interstitial fibrosis prevents the movement of myogenic cells, which is necessary for myotube formation. Canine model is the best phenocopies of the disease in humans when comparing with others animal models with dystrophy. Anti-fibrotic treatment of DMD patients, targeting the cytokine mediators, TGF-beta, and the treatment with antiinflammatories, may limit muscle degeneration and contribute for the improvement of the course of the illness. This work aimed to verify the possible adverse effects in renal physiology by means of evaluation sanguineous biochemist and arterial pressure, in order to verifying the viability of Losartan (a TGF-beta inhibiter) in affected dogs by muscle dystrophy. It was used four adults dogs, two of each gender. A dose of 50mg of Losartan was orally given once a day. The clinical exams, the kidney function, arterial blood pressure and potassium level did not show any adverse effect through the experimental period. Losartan utilization showed to be a safe therapy for the antifibrotic treatment in adults dogs, not affecting neither the kidney function nor the arterial blood pressure.
