#1 - Reproductive biology, morphology, and morphometry of ovaries and oviducts of Trachemys scripta elegans in Brazilian Cerrado

Abstract in English:

Although Trachemys scripta elegans is an exotic species popular as a pet in Brazil, studies on reproductive biology and capacity are non-existent in the Brazilian Cerrado. This study analyzed ovarian and oviduct characteristics and the egg production capacity of T. scripta elegans grown in this biome. The findings will associate with the size of the specimens and the sexual maturity, aiming at comparisons with native and exotic populations, as well as interspecific and contributing to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. Thus, 39 females had evaluated the body biometry and the morphology and morphometry of the ovaries and oviducts. G2 (N=20): with Class I (>5-10mm) follicles, with Class I and Class II (>10-fold) follicles, 25mm) and G3 (N=9) with Class I, Class II and Class III (>25mm) follicles. Analysis of variance, Scott-Knott’s test, and Pearson’s correlation analysis showed that there was no significant difference between the groups in body biometry; in the mean gonadosomatic index and gonadal morphometry, only the width of the oviducts in the right antimer and the mass and width in the left antimer were higher in G3, the only one that presented eggs. There was positive and harmonic development between body mass, carapace, and plastron, and gonadal growth occurred concomitantly with body growth, indicating a higher reproductive potential and a positive relationship between the size of the litter and the female litter. The gonadosomatic index proved to be an excellent reproductive indicator, and the ovarian evaluation was a better indicator of sexual maturity than the maximum carapace length. Ovaries were irregular structures, without delimitation between the cortical and medullary regions and filled with vitelogenic follicles of different diameters, atresic follicles, and corpora lutea, which reflected the ovarian complexity of the species and the presence of follicular hierarchy. In the scarce stroma, two germinative beds were observed per ovary and the presence of gaps very close to the follicles and associated with the blood vessels. Analysis of gonadal tissue revealed three types of oocytes according to cytoplasmic characteristics: homogeneous, vesicular or vesicular in the cortex with apparent granules. Oviducts were functional and separated, joining only in the final portion to form the cloaca and subdivided into infundibulum, tuba, isthmus, uterus, and vagina. The structure of the uterine tube was composed of serosa, muscular and mucous, which was full of glands. The presence of eggs in the oviducts indicated that the specimens can reproduce in the Brazilian Cerrado. This study provides necessary and relevant information on the reproductive biology and capacity of T. scripta elegans in the Brazilian Cerrado and can contribute to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. The extraction of females with capacity can reduce the annual reproductive yield of the species and decrease its effect on local biodiversity.

Embora Trachemys scripta elegans seja uma espécie exótica popular como animal de estimação no Brasil, estudos sobre biologia e capacidade reprodutivas são inexistentes no Cerrado brasileiro. Este estudo analisou características ovarianas e do oviduto e a capacidade de produção de ovos em T. scripta elegans criadas neste bioma, correlacionando estes achados ao tamanho dos espécimes e a maturidade sexual, visando comparações com populações nativas e exóticas, bem como interespecíficas e contribuir para a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação. Assim, 39 fêmeas tiveram avaliadas a biometria corporal e a morfologia e morfometria dos ovários e ovidutos. De acordo com o tamanho dos folículos ovarianos as fêmeas foram separadas em G1 (N= 10): com folículos Classe I (>5-10 mm), G2 (N= 20): com folículos Classe I e Classe II (>10-25 mm) e G3 (N= 9) com folículos Classe I, Classe II e Classe III (>25 mm). À análise de variância, teste de Scott-Knott e à análise de correlação de Pearson verificou-se que não houve diferença significativa entre os grupos na biometria corporal; no índice gonadossomático médio e na morfometria gonadal, apenas a largura dos ovidutos no antímero direito e a massa e a largura no antímero esquerdo foram maiores no G3, o único que apresentou ovos. Houve desenvolvimento positivo e harmônico entre massa corporal, carapaça e plastrão e o crescimento gonadal ocorreu concomitante ao crescimento corporal, indicando maior potencial reprodutivo e relação positiva entre o tamanho da ninhada de ovos e o da fêmea. O índice gonadossomático mostrou-se um bom indicador reprodutivo e a avaliação ovariana um melhor indicador da maturidade sexual que o comprimento máximo da carapaça. Ovários foram estruturas irregulares, sem delimitação entre a região cortical e medular e repletos de folículos vitelogênicos de diferentes diâmetros, folículos atrésicos e corpos lúteos, que refletiram a complexidade ovariana da espécie e a presença de hierarquia folicular. No estroma escasso foram observados dois leitos germinativos por ovário e a presença de lacunas muito próximas aos folículos e associadas aos vasos sanguíneos. A análise do tecido gonadal revelou três tipos de oócitos de acordo com as características do citoplasma: homogêneo, vesicular ou vesicular no córtex com grânulos aparentes. Ovidutos eram funcionais e separados, unindo-se apenas na porção final para formar a cloaca e subdividiam-se em infundíbulo, tuba uterina, istmo, útero e vagina. A estrutura da tuba uterina era constituída de serosa, muscular e mucosa, a qual era repleta de glândulas. A presença de ovos nos ovidutos indicou que os espécimes podem se reproduzir no cerrado brasileiro. Este estudo fornece informações básicas e relevantes da biologia e capacidade reprodutivas de T. scripta elegans no Cerrado brasileiro e pode contribuir com a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação, uma vez que a extração de fêmeas com capacidade reprodutiva pode contribuir com a diminuição do rendimento reprodutivo anual da espécie e diminuir seu efeito sobre a biodiversidade local.


#2 - Biochemical parameters of the giant anteater (Myrmecophaga tridactyla Linnaeus, 1758) of the Brazilian Cerrado

Abstract in English:

The giant anteater (Myrmecophaga tridactyla) is classified as a vulnerable species on Brazil’s list of species at risk of extinction mainly due to deforestation and forest fires. This has contributed to a considerable increase in detailed clinical case records of the treatment of wild species at veterinary institutions. However, the paucity of serum biochemical profiles of healthy giant anteaters has made it difficult to evaluate these animals, preventing diagnosis, treatment and prognosis. The objective of this work was to collect data about the biochemical profile of healthy giant anteaters from the Brazilian Cerrado raised in captivity, in order to better understand the physiological characteristics inherent to this species. Eighteen analytes from 12 healthy giant anteaters were measured. The following means and standard deviations were found in the biochemical analyses: albumin 3.29±0.33g/dL, ALT 15.49±7.98 IU/L, amylase 1037.92±149.04 IU/L, AST 21, 12±7.50 IU/L, total cholesterol 62.79±20.08mg/dL, HDL cholesterol 14.73±4.98mg/dL, LDL cholesterol 26.60±11.05mg/dL, VLDL cholesterol 2.14±1.06mg/dL, CK 111.61±70.16 IU/L, creatinine 1.05±0.37mg/dL, iron 194.64±81.17µg/dL, GGT 65.18±54.57 IU/L, glucose 103.71±29.63mg/dL, globulins 2.76±0.36g/dL, lipase 28.80±5.11 IU/L,TSP 6.05±0.56g/dL, triglycerides 10.71±5.29mg/dL, and urea 53.46±18.28mg/dL. The values found in this study can be used as references for the laboratory evaluation of giant anteaters living in conditions similar to those of this study. This is one of the first reports of biochemical examinations on giant anteaters of the Cerrado biome.

O tamanduá-bandeira (Myrmecophaga tridactyla) está classificado como espécie vulnerável na lista brasileira de espécies ameaçadas de extinção devido principalmente ao desmatamento e aos incêndios florestais. Tal fato contribuiu com o aumento da casuística de atendimento de espécies silvestres em instituições veterinárias. Porém, a escassez de valores bioquímicos séricos em tamanduás-bandeiras hígidos tem dificultado a avaliação destes animais, impedindo o diagnóstico, tratamento e prognóstico. O objetivo deste trabalho foi fornecer dados sobre o perfil bioquímico de tamanduás-bandeiras saudáveis do cerrado brasileiro, criados em cativeiro, a fim de compreender melhor as características fisiológicas inerentes a esta espécie. Foram mensurados 18 analitos de 12 tamanduás-bandeiras hígidos. As médias e o desvio padrão correspondentes às análises bioquímicas foram: albumina 3,29±0,33g/dL; ALT 15,49±7,98 UI/L; amilase 1037,92±149,04 UI/L; AST 21, 12±7,50 UI/L; colesterol total 62,79±20,08mg/dL; colesterol HDL 14,73±4,98mg/dL; colesterol LDL 26,60±11,05mg/dL; colesterol VLDL 2,14±1,06mg/dL; CK 111,61±70,16 UI/L; creatinina 1,05±0,37mg/dL; ferro 194,64±81,17µg/dL; GGT 65,18±54,57 UI/L; glicose 103,71±29,63mg/dL; globulinas 2,76±0,36g/dL; lipase 28,80±5,11 UI/L; PST 6,05±0,56g/dL; triglicerídeos 10,71±5,29mg/dL; ureia 53,46±18,28mg/dL. Os valores encontrados neste estudo podem ser utilizados como referência para a avaliação laboratorial de tamanduás-bandeiras que vivam em condições similares ao do presente estudo. Este é um dos primeiros estudos a relatar exames bioquímicos em tamanduás-bandeiras do bioma cerrado.

The increasing number of animals that are attended by wildlife rescue centres reinforces the need of knowledge about haematological parameters, especially for those that inhabit Brazilian cerrado biome. For this purpose and in order to establish reference values for this species and also to compare them with previous studies, haematological analysis of captive giant anteaters from Brazilian cerrado were performed. Moreover, the alterations of blood samples after 24 and 48 hours of storage at refrigeration temperatures (4oC) and preserved with two different EDTA concentrations (5% and 10%) were studied. Means and standard deviations of haematological parameters analysed immediately after collection were: RBC: 2,07 x106/µL ± 0,40; hematocrit: 38,08%± 5,93; haemoglobin: 11.33g/dL±2.15; MCV:186.52 fL±21.72; MCHC: 29.68g/dL±2.56; MCH: 55.08pcg±5,94; total leucocytes: 8.142/µL±2.441; neutrophils: 5.913/µL±2.168; lymphocytes: 1.460/µL±740; eosinophil: 522/µL±385; monocytes: 247/µL±176; thrombocytes: 123.458/µL±31.362 and total plasma protein: 6.23g/dL±0.49. This data shows evidence of the existence of important differences between these values and others from other areas, either from Brazil or from other South American countries. Those variations might be connected to environment, genetic, nutritional and/or management factors. Regarding the storage effect analysis, it can be concluded that in giant anteaters, haematological analysis can be performed until 24h after collection without any significant alterations on the haematological parameters, except for thrombocytes. Concerning the different EDTA concentrations, it can be concluded that there are no quantitative differences in haematological variables. Nevertheless, relevant morphologic alterations in blood cells can be observed after a 24h storage period, being most noticeable in the leucocytes. Those alterations can lead to misinterpretation of the results, interfering diagnosis, prognosis and treatment.

Por isso, este trabalho teve por objetivos realizar o hemograma de tamanduás-bandeiras de cativeiro localizados no cerrado brasileiro, a fim de estabelecer valores de referência para essa espécie e compará-los a estudos prévios. Além disso, verificar quais alterações podem ser encontradas em amostras armazenadas por 24 e 48 horas após a colheita, em temperatura de refrigeração (4oC) e tratadas com duas concentrações distintas de EDTA (5% e 10%). A média e o desvio padrão das variáveis hematológicas encontradas nas amostras processadas logo após a colheita foram: hemácias (2,07x106/µL±0,40); volume globular (38,08%± 5,93); hemoglobina (11,33g/dL±2,15); VCM (186,52 fL±21,72); CHCM (29,68g/dL±2,56); HCM (55,08pcg±5,94); leucócitos totais (8.142/µL±2.441); neutrófilos (5.913/µL±2.168); linfócitos (1.460/µL±740); eosinófilos (522/µL±385); monócitos (247/µL±176); plaquetas (123.458/µL±31.362) e proteínas plasmáticas totais (6,23g/dL±0,49). Tais dados permitem afirmar que existem importantes diferenças entre os valores hematológicos destes em relação aos animais provenientes de outras regiões, tanto do Brasil quanto de outros países da América do Sul. Provavelmente, tais divergências estão associadas a fatores ambientais, genéticos, nutricionais e/ou de manejo. Quanto à análise das amostras estocadas, conclui-se que, em tamanduás-bandeiras, as amostras para a realização de hemograma podem ser processadas até 24 horas após a colheita, sem alteração significativa das variáveis hematológicas, com exceção das plaquetas. Com relação às duas concentrações de EDTA, pode-se inferir que não há diferença quantitativa entre ambas para as variáveis hematológicas. Contudo, é possível constatar que o EDTA promove alterações morfológicas relevantes nas células sanguíneas após 24 horas de armazenamento, sendo os leucócitos os mais afetados. Tais alterações, quando relatadas, podem induzir interpretações equivocadas, interferindo no diagnóstico, prognóstico e tratamento.


#4 - Occurrence of antibodies against Leptospira spp. in free-ranging wild canids from the Brazilian savanna, 35(8):734-740

E-mail: thaiscarneiro_25@hotmail.com The Brazilian savanna, also known as Cerrado, is one of the world’s richest and most ecologically invaluable tropical savanna regions. There are few studies in Brazil about the diseases that affect the wild canids of this biome, which may be harmful to wildlife populations and public health. The aim of this study was to evaluate the occurrence of antibodies against Leptospira spp. in three Cerrado wild canids species using the microscopic agglutination test (MAT). Serum samples were tested from 19 crab-eating foxes (Cerdocyon thous), 14 maned wolves (Chrysocyon brachyurus), and seven hoary foxes (Lycalopex vetulus), all free-ranging animals found in the municipalities of Araguari and Uberlândia, Minas Gerais State, and Cumari, Goiás State, Brazil. Fourteen (35%) of these samples were seropositive. The most frequent serovars detected in the samples were Copenhageni and Hardjo, but reactions to the serovars Autumnalis, Grippotyphosa, Hebdomadis, Wolffi, and Icterohaemorrhagiae also occurred. Notwithstanding other reported results, this study is the first to report the presence of antibodies against Leptospira spp. in L. vetulus. The three species of wild canids examined may act as potential hosts for several serovars of leptospira in Brazil’s savanna environment.

Foram testadas amostras de soro sanguíneo de 19 cachorros-do-mato (Cerdocyon thous), 14 lobos guarás (Chrysocyon brachyurus) e sete raposas-do-campo (Lycalopex vetulus) de vida livre das regiões de cerrado de Araguari e Uberlândia, MG, e Cumari, GO. Dessas 40 amostras, 14 (35%) foram soropositivas. Os sorovares detectados com maior frequência foram Copenhageni e Hardjo, porém evidenciou-se também reação aos sorovares Autumnalis, Grippotyphosa, Hbedomadis, Wolffi e Icterohaemorrhagiae. Esse trabalho foi o primeiro a relatar a presença de anticorpos anti-Leptospira spp. em L. vetulus e verificou-se que as três espécies de canídeos utilizadas nessa pesquisa podem atuar como possíveis hospedeiros de diversos sorovares de leptospiras no ambiente de cerrado.

Four groups of pregnant cows received the following treatments: T1- cows and calves not treated, T2- only calves treated, T3- only cows treated, and T4- cows and calves treated. The calves of T2 and T4 were distributed in the following treatments: A- calves treated at 3 to 5 months of age with long action ivermectin, B- treated with ivermectin, and C- control. For the cows, the deworming did not diminish the EPG during lactation and also did not have significant effect on the conception rate, live weight gain and the body weight of their calves at 3 to 5 months of age. The calves of treatment A gained, from the time of treatment to weaning (84 to 108 days), an average of 4.2kg (P=0.0003) and 7.1kg (P<0.0001) more than those of treatment B and C, respectively. The average difference in live weight gain of 2.9kg between the animals of treatment B and C was not significant. The EPG before treatment was not significantly different from the treatments (P=0.8665); but at weaning, the average EPG of the calves from treatment A was lower than for treatment B (P=0.0004) and C (P<0.0001). There was no significant difference in the mean EPG for the calves from treatment B and C.

#6 - Fanniid diptera vectors of Dermatobia hominis eggs in Campo Grande, MS, Brazil, 22(3):114-118

[Fanniid diptera vectors of Dermatobia hominis eggs in Campo Grande, MS, Brazil.] Dípteros fanídeos de ovos de Dermatobia hominis em Campo Grande, Mato Grosso do Sul. Pesquisa Veterinária Brasileira 22 (3):114-118. Laboratório de Ectoparasitologia, Embrapa Gado de Corte, BR 262 Km 4, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. This study was undertaken to determine the epidemiological importance of fanniid flies on warble fly infestations. The local species of Fanniidae as well as those utilized by Dermatobia hominis to carry its eggs were identitied, and the population dynamics of the most abundant species were studied. In order to accomplish this task, five traps with rotten bovine liver as bait were used. These traps were settled into a ciliate forest bordered by pastures and grazed continuously by cattle. The experiment was carried out at Embrapa Gado de Corte, in Campo Grande, Mato Grosso do Sul, Brazil, situated at 20º27‘S and 54°37’W. The capture of the insects was done weekly, from August 9, 1999 to August 3, 2000. A total of 40,629 fanniid flies which belonged to five different species of the genera Fannia were captured: E pusio, F. heydenii, E bahiensis and E longipila, and one species which needs further identification. The most frequent species was E pusio, with 63.20% of the total captured flies, followed by E heydenii with 28.82%. Eggs of D. hominis were observed only on 0.44% (45 specimens) of the E heydenii females captured, mainly on the ones captured in August and September. The average number for D. hominis eggs, per specimen, was 15.98±7.13. The only vector body region, where the eggs of D. hominis were found, was the abdomen. E heydenii predominated during the dry season (May to September) and at the beginning of the rainy season (October and November). The number of specimens bearing eggs of D. hominis was greater at the end of the annual dry season, what explains the high incidence of this parasite in cattle during September and October.

[Fanniid diptera vectors of Dermatobia hominis eggs in Campo Grande, MS, Brazil.] Dípteros fanídeos de ovos de Dermatobia hominis em Campo Grande, Mato Grosso do Sul. Pesquisa Veterinária Brasileira 22 (3):114-118. Laboratório de Ectoparasitologia, Embrapa Gado de Corte, BR 262 Km 4, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. Determinou-se a importância epidemiológica de dípteros Fanniidae na infestação de mosca-do-berne, por meio da identificação das espécies presentes, da determinação daquelas utilizadas por Dermatobia hominis na veiculação de seus ovos, bem como, pelo conhecimento da dinâmica populacional das espécies mais abundantes. Foram utilizadas cinco armadilhas iscadas com fígado bovino cru deteriorado e colocadas em uma mata ciliar margeada por uma área de pastagem com presença constante de bovinos. O estudo foi desenvolvido em uma área da Embrapa Gado de Corte, em Campo Grande, Estado de Mato Grosso do Sul, Brasil, localizada a 20°27‘S e 54°37’W. A captura dos insetos foi realizada semanalmente durante o período de 09/08/1999 a 03/08/2000. Foi capturado um total de 40.629 moscas da família Fanniidae, pertencendo a cinco espécies do gênero Fannia: E pusio, E heydenii, E bahiensis e F. longipila, e uma a ser identificada. A espécie mais freqüente foi E pusio, com 63,20% do total capturado, seguida de E heydenii, com 28,82%. Somente 0,44% do total de femeas de E heydenii (45 exemplares) capturadas, principalmente nos meses de agosto e setembro, portavam ovos de D. hominis e o número médio, por indivíduo, foi de 15,98±7,13. Observaram-se ovos de D. hominis apenas na região abdominal dos vetores. F. heydenii predominou no período seco (maio a setembro) e início do período chuvoso do ano (outubro e novembro). O número de exemplares portando ovos de D. hominis foi maior no final do período seco do ano, o que explica a alta incidência deste parasito em bovinos nos meses de setembro e outubro.
