#1 - Anatomopathological pneumonic aspects associated with highly pathogenic Pasteurella multocida in finishing pigs, 37(10):1091-1100

E-mail: guedesufmg@gmail.com The bacterium Pasteurella multocida is a frequent cause of porcine respiratory disease complex in finishing pigs. Historically, the bacterium is recognized as an opportunistic agent, causing secondary bacterial pneumonia in pigs. Several Brazilian reports have suggested the ability of P. multocida to cause primary pulmonary infection that leads to the death of finishing pigs prior to slaughter. The aim of this study was to evaluate anatomopathological pulmonary findings associated with P. multocida infection that were obtained from animals with clinical respiratory disease and from animals at slaughter. Twenty-five lung samples from 14 herds of finishing pigs with acute clinical respiratory disease and 19 lungs collected at slaughter from a different set of 14 herds were studied. In all lung samples, bacterial isolation was performed, and only samples with pure P. multocida growth were included in the study. Gross and histopathological lesions were evaluated, as well as Influenza A, porcine circovirus type 2 (PCV2) and Mycoplasma hyopneumoniae co-infections. Pleuritis and pericarditis were more often observed in clinical samples (P<0.05). Moreover, there was a numerical trend indicating that pericarditis, lymphadenomegaly and cavity exudates were more often present in clinical samples. Thirteen lung samples were negative to M. hyopneumoniae, Influenza A and PCV2 by immunohistochemistry (IHC), with only P. multocida identified. In these cases, gross lesions such as pleuritis, pericarditis and lymphadenomegaly were always present, and no histologic lesions indicative of other agents such as Actinobacillus pleuropneumoniae, Actinobacillus suis or Haemophilus parasuis were observed. These findings suggest the ability of some P. multocida isolates to cause primary respiratory and systemic infection. However, in this study, it was not possible to determine specific virulence markers to explain these findings.

Historicamente, a bactéria é reconhecida como agente oportunista, causando pneumonia bacteriana secundária. Diversos relatos brasileiros sugerem a habilidade da P. multocida de causar infecção pulmonar primária que leva a mortalidade de animais de terminação antes do abate. O objetivo deste estudo foi avaliar achados anatomopatológicos pulmonares associados com infecção por P. multocida, obtidas de animais acometidos clinicamente por doença respiratória e de animais ao abate. Avaliou-se 25 amostras de pulmão de 14 rebanhos obtidas de animais de terminação com sinais clínicos de doença respiratória aguda, e 19 pulmões coletados ao abate de 14 rebanhos diferentes. Em todos os pulmões, realizou-se isolamento bacteriano, e apenas amostras com crescimento puro de P. multocida foram incluídas no trabalho. Avaliou-se as lesões macro e microscopicamente, assim como co-infecções por Influenza A, Circovirus suíno tipo 2 (PCV2) e Mycoplasma hyopneumoniae. Pleurite e pericardite foram mais frequentemente observadas em amostras clinicas (P<0,05). Ainda, houve tendência numérica indicando a ocorrência de linfadenomegalia e exsudação cavitária, mais presentes em amostras clínicas. Treze amostras de pulmão foram negativas para M. hyopneumoniae, Influenza A e PCV2 por imunoistoquímica (IHQ), com identificação de apenas P. multocida. Nestes casos, lesões macroscópicas como pleurite, pericardite e linfadenomegalia foram sempre presentes, sem lesões histológicas indicativas de outros agentes como Actinobacillus pleuropneumoniae, Actinobacillus suis ou Haemophilus parasuis. Estes achados sugerem a habilidade de alguns isolados de P. multocida de causarem quadro respiratório primário e infecção sistêmica. No entanto, neste estudo, não foi possível determinar marcadores de virulência específicos para justificar tais achados.

E-mail: nelson.mores@embrapa.br In order to understand better the pathological aspects and spread of Pasteurella multocida type A as the primary cause of pneumonia in pigs, was made an experiment with intranasal inoculation of different concentrations of inocula [Group (G1): 108 Colony Forming Units (CFU)/ml; G2: 107 CFU/ml; G3: 106 CFU/ml and G4: 105 CFU/ml], using two pigs per group. The pigs were obtained from a high health status herd. Pigs were monitored clinically for 4 days and subsequently necropsied. All pigs had clinical signs and lesions associated with respiratory disease. Dyspnoea and hyperthermia were the main clinical signs observed. Suppurative cranioventral bronchopneumonia, in some cases associated with necrosuppurative pleuropneumonia, fibrinous pericarditis and pleuritic, were the most frequent types of lesion found. The disease evolved with septicaemia, characterized by septic infarctions in the liver and spleen, with the detection of P. multocida type A. In this study, P. multocida type A strain #11246 was the primary agent of fibrinous pleuritis and suppurative cranioventral bronchopneumonia, pericarditis and septicaemia in the pigs. E-mail: nelson.mores@embrapa.br Para entender melhor os aspectos patológicos e disseminação de Pasteurella multocida tipo A como causa primária de pneumonia em suínos foi realizado um experimento com inoculação intranasal de diferentes concentrações de inóculos [Grupo (G1): 108 Unidades Formadoras de Colônias (UFC)/ml; G2: 107 UFC/ml; G3: 106 UFC/ml e G4: 105 UFC/ml], usando dois suínos por grupo. Esses suínos foram obtidos de um rebanho de alto status sanitário. Os animais foram monitorados clinicamente por quarto dias e subsequentemente necropsiados. Todos os suínos apresentaram sinais clínicos e lesões associadas com doença respiratória. Dispneia e hipertermia foram os principais sinais clínicos observados. Broncopneumonia cranioventral supurativa, em alguns casos associados com pleuropneumonia necrossupurativa, pleurites e pericardite fibrinosa foram mais frequentes. A doença evoluiu com septicemia, caracterizada por infartos sépticos no fígado e baço, com detecção de P. multocida. Neste estudo, P. multocida tipo A isolado 11246 foi agente primário de pleurite fibrinosa e broncopneumonia cranioventral supurativa, pericardite fibrinosa e septicemia em suínos. Todas as concentrações de inóculo utilizado (105-108 UFC/ml) foram capazes de produzir sinais clínicos e patológicos de alterações de pneumonia, pleurites, pericardites e septicemia nos animais.

E-mail: valdutra@ufmt.br Based on diagnostic tests for the detection of nucleic acids without amplification through the use of gold nanoparticles (AuNPs) have been described for various diseases. This study aimed to develop a technique of unmodified AuNPs to detect Actinobacillus pleuropneumoniae (App). We used 70 lung samples from pigs, 17 with and 53 without characteristic lesions of pneumonia, to detect App. The primer used was based on ApxIV gene. The AuNPs test had a sensitivity of 93.8% and specificity of 84.6% when compared with PCR detection. The results showed good agreement between AuNPs and PCR testing, and the technique can be used as an alternative to conventional tests, since it is quick and easy, and does not require implementation infrastructure and skilled labor.

Este trabalho teve como objetivo desenvolver uma técnica de AuNPs não modificada para detecção de Actinobacillus pleuropneumoniae (App). Utilizaram-se 70 amostras de pulmão de suínos, 17 sem lesão e 53 com lesões características de pneumonia, objetivando a detecção de App. O oligonucleotídeo utilizado foi baseado no gene ApxIV. O teste de AuNPs apresentou sensibilidade de 93,8% e especificidade de 84,6% quando comparado com a detecção pela PCR. Os resultados mostraram boa concordância entre os testes de AuNPs e a PCR, sendo que a técnica pode ser utilizada como alternativa aos testes convencionais, já que é de fácil e rápida execução e não exige infraestrutura e mão de obra especializada.

A set of six primers were designed derived from the dsbE-like gene of A.pleuropneumoniae and validate the assay using 9 A. pleuropneumoniae reference/field strains, 132 clinical isolates and 9 other pathogens. The results indicated that positive reactions were confirmed for all A. pleuropneumoniae strains and specimens by LAMP at 63°C for 60 min and no cross-reactivity were observed from other non-A.pleuropneumoniae including Haemophilus parasuis, Escherichia coli, Pasteurella multocida, Bordetella bronchiseptica, Streptococcus suis, Salmonella enterica, Staphylococcus, porcine reproductive and respiratory syndrome virus (PRRSV), and Pseudorabies virus. The detection limit of the conventional PCR was 102 CFU per PCR test tube, while that of the LAMP was 5 copies per tube. Therefore, the sensitivity of LAMP was higher than that of PCR. Moreover, the LAMP assay provided a rapid yet simple test of A. pleuropneumoniae suitable for laboratory diagnosis and pen-side detection due to ease of operation and the requirement of only a regular water bath or heat block for the reaction.

#5 - Serotypes of Haemophilus (Actinobacillus) pleuropneumoniae isolated from swine in Brazil

Thirty-three strains of Haemophilus (Actinobacillus) pleuropneumoniae biovar 1 were isolated from swine lungs with acute or chronic lesions of swine pleuropneumonia, with one exception, which was isolated from the nasal cavity of a pig from a problem herd. These strains originated from 14 herds, ten located in the State of Santa Catarina, three in Rio Grande do Sul and one in São Paulo. Serotyping was done by rapid slide agglutination (RSA), immunofluorescence (IF), counterimmunoelectrophoresis (CIE) and immunodiffusion (ID). Twenty-two (66%) of the 33 strains could be serotyped, 19 belonged to serotype 5 and three to serotype 3. All serotype 3 strains together with those not possible to be serotyped originated from swine of the sarne herd, Santa Catarina, one in which serotype 5 was also isolated. The IF and CIE were the most sensitive tests, while the ID was the least sensitive. Three strains could not be serotyped by the RSA because they auto-agglutinated.

Isolaram-se 33 amostras de Haemophilus (Actinobacillus) pleuropneumoniae, biovar 1, sendo que 32 originaram-se de pulmões de suínos com lesões macroscópicas agudas ou crônicas de Pleuropneumonia Suína (PPS) e uma amostra da cavidade nasal de um suíno de granja problema. Estas amostras foram coletadas em 14 propriedades, sendo dez localizadas no Estado de Santa Catarina, três no Rio Grande do Sul e uma em São Paulo. Para a sorotipagem utilizaram-se as técnicas de soroaglutinação rápida (SAR), imunofluorescência (IF), imunoeletroosmoforese (IEOF) e imunodifusão (ID). Das 33 amostras, 22 (66%) foram sorotipadas e, entre estas, 19 pertenciam ao sorotipo 5 e três ao sorotipo 3. As amostras do sorotipo 3 e aquelas não sorotipadas originaram-se de suínos de uma mesma propriedade do Estado de Santa Catarina, onde também isolou-se o sorotipo 5. Os testes de IF e IEOF foram os mais sensíveis e o de ID apresentou a menor sensibilidade. Três amostras não puderam ser sorotipadas pelo teste de SAR porque auto-aglutinavam.


#6 - Immunoprophylaxis of swine pleuropneumonia with an inactivated vaccine against Haemo-philus pleuropneumoniae

Abstract in English:

The effectiveness of a bacterin in the prevention of pleuropneumonia of swine was tested. The bacterin was prepared from a strain of H. pleuropneumoniae, serotype 5, and adsorved to aluminum hydroxide. Six sows were vaccinated at 60 and 100 days of gestation (VS) and seven were not vaccinated (NVS). Piglets born to VS were divided into three treatment groups: unvaccinated; vaccinated at 40 days of age; and vaccinated at 25 and 40 days of age. Those born to the NVS were divided into two groups: unvaccinated and vaccinated at 25 and 40 days of age. On third of the piglets from each treatment groups was challenged with a strain of H. pleuropneumoniae, serotype 5, at 70 days of age. In piglets born to VS, respiratory symptoms were observed less frequently, H. pleuropneumoniae was found less often in the nasal cavity after challenge, a lower mortality rate was noted, and less severe lesions characteristic of pleuropneumonia were seen than in piglets born to NVS. Piglet vaccination schedules did not interfere with the above mentioned parameters.

Testou-se a eficiência de uma bacterina preparada com H. pleuropneumoniae, sorotipo 5, e adsorvida em hidróxido de alumínio, na prevenção de pleuropneumonia suína. Vacinou-se seis matrizes (MV) aos 60 e 100 dias de gestação, e sete não foram vacinadas (MNV). Os leitões originados das MV foram divididos em três tratamentos: não vacinados, vacinados aos 40 dias e aos 25 e 40 dias de idade. Os originados das MNV foram divididos em dois grupos: vacinados aos 25 e 40 dias e não vacinados. Um terço dos leitões de cada tratamento foi inoculado com H. pleuropneumoniae, sorotipo 5, aos 70 dias de idade. Leitões originados de MV apresentaram menor ocorrência de sintomas respiratórios, menor presença de H. pleuropneumoniae na cavidade nasal apos a agressão, menor índice de mortalidade e menor ocorrência de severidade de lesões compatíveis com a pleuropneumonia suína do que aqueles originados de MNV. Os esquemas de vacinação aplicados aos leitões no interferiram nos parâmetros acima mencionados.
