#11 - A thymidine kinase-deleted bovine herpesvirus 5 establishes latent infection but reactivates poorly in a sheep model, 33(3):331-338

E-mail: eduardofurtadoflores@gmail.com The ability of thymidine kinase (tk)-deleted recombinant bovine herpesvirus 5 (BoHV-5tkD) to establish and reactivate latent infection was investigated in lambs. During acute infection, the recombinant virus replicated moderately in the nasal mucosa, yet to lower titers than the parental strain. At day 40 post-infection (pi), latent viral DNA was detected in trigeminal ganglia (TG) of all lambs in both groups. However, the amount of recombinant viral DNA in TGs was lower (9.7-fold less) than that of the parental virus as determined by quantitative real time PCR. Thus, tk deletion had no apparent effect on the frequency of latent infection but reduced colonization of TG. Upon dexamethasone (Dx) administration at day 40 pi, lambs inoculated with parental virus shed infectious virus in nasal secretions, contrasting with lack of infectivity in secretions of lambs inoculated with the recombinant virus. Nevertheless, some nasal swabs from the recombinant virus group were positive for viral DNA by PCR, indicating low levels of reactivation. Thus, BoHV-5 TK activity is not required for establishment of latency, but seems critical for efficient virus reactivation upon Dx treatment.

Durante a infecção aguda, o vírus recombinante replicou na mucosa nasal em títulos moderados, porém menores do que os da cepa parental. Aos 40 dias pós-infecção (pi) DNA viral latente foi detectado no gânglio trigêmeo (TG) de todos os cordeiros em ambos os grupos. No entanto, a quantidade de DNA do vírus recombinante nos TGs foi 9,7 vezes menor do que do vírus parental, segundo determinação por PCR em tempo real. Assim, a deleção do gene tk (timidina quinase) não produziu efeito aparente sobre a frequência da infecção latente, porém reduziu a colonização do TG. Após a administração de dexametasona (Dx) no dia 40pi, os cordeiros inoculados com o vírus parental excretaram partículas virais infecciosas, contrastando com a falta de infectividade nas secreções nasais dos animais inoculados com o vírus recombinante. Entretanto, alguns suabes nasais dos cordeiros do grupo do vírus recombinante foram positivos para o DNA viral por PCR, indicando baixos níveis de reativação. Assim, a atividade da enzima timidina quinase não é requerida para o estabelecimento de latência pelo BoHV-5, mas parece fundamental para reativação eficiente da infecção latente após tratamento com Dx.


#12 - Prokaryotic expression of a truncated form of bovine herpesvirus 1 glycoprotein E (gE) and use in an ELISA for gE antibodies, 33(1):41-46

E-mail: eduardofurtadoflores@gmail.com This article describes the expression of a truncated form of bovine herpesvirus 1 (BoHV-1) glycoprotein E (gE) for use as immunodiagnostic reagent. A 651 nucleotide fragment corresponding to the amino-terminal third (217 amino acids) of BoHV-1 gE - that shares a high identity with the homologous BoHV-5 counterpart - was cloned as a 6×His-tag fusion protein in an Escherichia coli expression vector. A soluble protein of approximately 25 kDa purified from lysates of transformed E. coli was recognized in Western blot (WB) by anti-6xHis-tag and anti-BoHV-1 gE monoclonal antibodies. In addition, the recombinant protein was specifically recognized in WB by antibodies present in the sera of cattle seropositive to BoHV-1 and BoHV-5. An indirect ELISA using the expressed protein as coating antigen performed comparably to a commercial anti-gE ELISA and was able to differentiate serologically calves vaccinated with a gE-deleted BoHV-5 strain from calves infected with BoHV-1. Thus, the truncated gE may be useful for serological tests designed to differentiate BoHV-1/BoHV-5 infected animals from those vaccinated with gE-negative marker vaccines.

Um fragmento de 651 pares de bases (pb) correspondente ao terço amino-terminal (217 aminoácidos) da gE do BoHV-1 – que compartilha uma alta identidade com a gE do BoHV-5 – foi clonada como proteína de fusão com cauda 6x de histidina em um vetor de expressão em Escherichia coli. Uma proteína solúvel de aproximadamente 25 kDa purificada de lisados de E.coli foi reconhecida em Western blot (WB) por anticorpos monoclonais anti-6xHis-tag e anti-gE. Além disso, a proteína recombinante purificada foi reconhecida em WB por anticorpos presentes no soro de animais soropositivos ao BoHV-1 e BoHV-5. Um ELISA indireto utilizando a proteína recombinante como antígeno apresentou performance comparável a um ELISA gE comercial e foi capaz e diferenciar sorologicamente animais vacinados com uma cepa gE-negativa de BoHV-5 de animais infectados com o BoHV-1. #13 - Reactivation and distribution of bovine herpesvirus 5 DNA in the brain of latently infected sheep, 31(12):1090-1096

E-mail: eduardofurtadoflores@gmail.com The biology of latent infection by bovine herpesvirus type 5 (BoHV-5) has been studied in cattle and rabbits, yet many aspects remain poorly understood. We herein investigated the suitability of lambs to investigate aspects of BoHV-5 latency. Thirteen six-month-old lambs inoculated intranasally (IN) with BoHV-5 strain SV-507/99 (titer of 106.8 TCID50/mL) shed the virus in nasal secretions in titers up 105.5 TCID50/mL, during up to 11 days, developing virus neutralizing (VN) titers of 16 to 128 at day 30 post-inoculation (pi). The inoculated animals developed only a mild serous nasal secretion and transient hyperthermia. Examination of brain sections of five lambs euthanized at day 30 pi by PCR revealed the presence of latent DNA in the trigeminal ganglia (TG, 5 out of five), olfactory bulbs (OB, 5/5), pons (2/5), cerebellum (2/5) and cerebral cortex (1/5). Administration of dexamethasone (Dx, n=4) or flumethasone (FluM, n=4) to eight latently infected lambs at day 65 pi resulted in virus reactivation and shedding by 3 out of 4 individuals in each group. Virus shedding in nasal secretions started at day 3 post-treatment and lasted up to five days (1-5) in Dx treated lambs (titers up to 102.8TCID50/mL), was delayed and lasted up to three days (1-3) in FluM-treated lambs (titers up to 102.1 TCID50/mL). PCR examination of the brains of animals submitted to reactivation, at day 30 post-treatment, showed a pattern of distribution of latent viral DNA fairly similar to that found in those not submitted to reactivation. In summary, the ability of BoHV-5 to establish latent infection, the consistent colonization of TGs and OBs by latent viral DNA and virus reactivation induced by corticosteroid treatment are promising findings towards the use of lambs to study selected aspects of BoHV-5 latency.

Este artigo relata uma avaliação de ovinos jovens como modelo para o estudo da infecção latente pelo BoHV-5. Treze cordeiros com idade entre seis e sete meses, inoculados pela via intranasal (IN) com a cepa SV-507/99 do BoHV-5 (título de 106,8 DICC50/mL) excretaram o vírus em secreções nasais em títulos de até 105,5 DICC50/mL, com duração de até 11 dias, desenvolvendo anticorpos neutralizantes em títulos de 16 a 128 no dia 30 pós-inoculação (pi). Os ovinos inoculados apresentaram apenas secreção nasal serosa leve e hipertermia transitória. O PCR de secções do encéfalo de cinco animais inoculados no dia 30 pi revelou a presença de DNA viral latente nos gânglios trigêmeos (TG, 5 de 5 animais), bulbo olfatório (BO, 5/5), ponte (2/5), cerebelo (2/5), córtex cerebral (1/5). Administração de dexametasona (Dx, n=4) ou flumetasona (FluM, n=4) a oito ovinos no dia 65 pi resultou em reativação e excreção viral por 3 de 4 animais de cada grupo. A excreção viral nas secreções nasais iniciou no dia 3 pós-tratamento e durou entre 1 e 5 dias nos ovinos tratados com Dx (títulos até 102,8TCID50/mL) e foi mais tardia, durando entre 1 e 3 dias nos animais tratados com FluM (títulos de 102,1 TCID50/mL). Uma análise por PCR do encéfalo dos animais submetidos à reativação, no dia 65 pós-infecção, revelou uma distribuição do DNA latente semelhante àquela observada nos animais não submetidos à reativação. Em resumo, a capacidade do BoHV-5 estabelecer infecção latente, a colonização dos TGs a BOs com DNA viral latente e a reativação induzida por corticoides são achados promissores para o uso de cordeiros como modelo para a infecção latente pelo BoHV-5.


#14 - A thymidine kinase-negative bovine herpesvirus 5 is highly attenuated for rabbits, but is neuroinvasive and establishes latent infection, 31(5):389-397

E-mail: eduardofurtadoflores@gmail.com Mutant viral strains deleted in non-essential genes represent useful tools to study the function of specific gene products in the biology of the virus. We herein describe an investigation on the phenotype of a bovine herpesvirus 5 (BoHV-5) recombinant deleted in the gene encoding the enzyme thymidine kinase (TK) in rabbits, with special emphasis to neuroinvasiveness and the ability to establish and reactivate latent infection. Rabbits inoculated with the parental virus (SV-507/99) (n=18) at a low titer (105.5TCID50) shed virus in nasal secretions in titers up to 104.5TCID50 for up to 12 days (average: 9.8 days [5-12]) and 5/16 developed neurological disease and were euthanized in extremis. Rabbits inoculated with the recombinant BoHV-5TKD at a high dose (107.1TCID50) also shed virus in nasal secretions, yet to lower titers (maximum: 102.3TCID50) and for a shorter period (average: 6.6 days [2-11]) and remained healthy. PCR examination of brain sections of inoculated rabbits at day 6 post-infection (pi) revealed a widespread distribution of the parental virus, whereas DNA of the recombinant BoHV-5TKD-was detected only in the trigeminal ganglia [TG] and olfactory bulbs [OB]. Nevertheless, during latent infection (52pi), DNA of the recombinant virus was detected in the TGs, OBs and also in other areas of the brain, demonstrating the ability of the virus to invade the brain. Dexamethasone (Dx) administration at day 65 pi was followed by virus reactivation and shedding by 5/8 rabbits inoculated with the parental strain (mean duration of 4.2 days [1 – 9]) and by none of seven rabbits inoculated with the recombinant virus. Again, PCR examination at day 30 post-Dx treatment revealed the presence of latent DNA in the TGs, OBs and in other areas of the brain of both groups. Taken together, these results confirm that the recombinant BoHV-5TKD is highly attenuated for rabbits. It shows a reduced ability to replicate in the nose but retains the ability to invade the brain and to establish latent infection. Additional studies are underway to determine the biological and molecular mechanisms underlying the inability of BoHV-5TKD to reactivate from latency.

Este estudo relata uma investigação do fenótipo, em coelhos, de uma cepa recombinante do herpesvírus bovino tipo 5 (BoHV-5) defectiva na enzima timidina quinase (TK), com ênfase para a neuroinvasividade e capacidade de estabelecer e reativar a infecção latente. Coelhos inoculados com o vírus parental (SV-507/99, n=18) em baixo título (105,5TCID50) excretaram o vírus nas secreções nasais em títulos de até 104,5TCID50/mL por até 12 dias (média: 9,8 dias [5-12]) e 5/16 desenvolveram doença neurológica e morreram ou foram eutanasiados in extremis. Em contraste, coelhos inoculados com o recombinante BoHV-5TKD em alto título (107,1TCID50) excretaram o vírus em títulos inferiores (máximo 102,3TCID50/mL), por um período menor (média: 6,6 days [2-11]) e permaneceram saudáveis. #15 - Experimental infection of calves with recombinants of bovine herpesvirus 5 defective in glycoprotein E (gE), thymidine kinase (TK) and both, gE/TK, 31(4):319-325

E-mail: eduardofurtadoflores@gmail.com This article describes an investigation on the virulence/attenuation of bovine herpesvirus type 5 (BoHV-5) recombinants deleted in the genes encoding glycoprotein E (BoHV-5gED), thymidine kinase (BoHV-5TKD), and both gE and TK (BoHV-5gEDTKD). Seronegative calves (80 to 90 days-old) inoculated with the parental strain (SV-507/99, n=5) shed virus in nasal secretions for up to 15 days (average 10.8 days). Duration of virus shedding was 11 days for BoHV-5gD, 9.6 days for BoHV-5TKD and 6.2 days for BoHV-5gEDTKD groups. The highest titers were observed between days 1 and 6 post-infection (pi) for SV-507/99 (106.8TCID50/mL), 105.1TCID50/mL (BoHV-5gED), 105.9TCID50/mL (BoHV-5TKD) and 104.7TCID50/mL (BoHV-5gEDTKD). Calves inoculated with the parental virus presented anorexia, profound apathy and loss of body condition. Two calves were euthanized in extremis on days 10 and 11 pi; infectious virus was recovered from several areas of the brain. In contrast, calves inoculated with the recombinants remained healthy and a few presented a mild and transient nasal secretion. Dexamethasone (Dx) administration at day 42 pi resulted in virus shedding by all controls calves (mean duration 3.7 days), by 2/5 of BoHV-5TKD calves (two days) and 2/5 of BoHV-5gED (one day). No virus shedding was detected in BoHV-5gEDTKD calves upon Dx treatment. PCR examination of brain sections of calves euthanized at day 30 post Dx treatment revealed the presence of latent viral DNA widely distributed in the brain of SV-507/99 calves. Latent viral DNA was detected in a few sections (3/30) of the brains of BoHV-5gED calves and was not detected in the brains of calves inoculated with BoHV-5TKD and BoHV-5gEDTKD. These results show that the single BoHV-5 mutants (gE and tk-deleted) are attenuated for calves and establish and/or reactivate latent infection inefficiently. The double mutant BoHV-5gEDTKD is fully attenuated and appears not to establish or not reactivate efficiently from latent infection. Thus, these recombinants, especially the double mutant BoHV-5gEDTKD, display an adequate phenotype for use in modified-live vaccine formulations.

Bezerros soronegativos (80-90 dias de idade) inoculados com o vírus parental SV-507/99 (n=5) excretaram o vírus em secreções nasais por até 15 dias (média de 10,8 dias). Nos animais inoculados com os recombinantes, a duração da excreção viral foi de 11 dias (BoHV-5gED), 9,6 dias (BoHV-5TKD) e 6,2 dias (BoHV-5gEDTKD). Os maiores títulos foram observados entre os dias 1 e 6 pós-inoculação (pi), sendo de 106,8TCID50/mL para o SV-507/99, 105,1TCID50/mL (BoHV-5gED), 105,9TCID50/mL (BoHV-5TKD) e 104,7TCID50/mL (BoHV-5gEDTKD). Os bezerros inoculados com o vírus parental apresentaram anorexia e apatia; três deles mostraram apatia profunda e perda da condição corporal. Dois bezerros foram eutanasiados in extremis nos dias 10 e 11 pi, respectivamente e o vírus foi isolado de várias regiões do encéfalo. Já os bezerros inoculados com os recombinantes permaneceram saudáveis; alguns apresentaram uma secreção nasal serosa transitória. Administração de dexametasona (Dx) no dia 42 pi resultou em excreção viral por todos os bezerros inoculados com o vírus parental (duração média de 3,7 dias), por 2 de 5 bezerros dos grupos BoHV-5TKD (dois dias) e BoHV-5gED (um dia). Os bezerros inoculados com o duplo mutante BoHV-5gEDTKD não excretaram o vírus após o tratamento com Dx. Pesquisa de DNA viral por PCR no dia 30 pós-Dx revelou uma ampla distribuição do DNA do vírus parental no encéfalo; poucas seções (3/30) foram positivas no encéfalo dos animais do grupo BoHV-5gED, e não detectou-se DNA latente no encéfalo dos animais dos grupos BoHV-5TKD e BoHV-5gEDTKD. Esses resultados demonstram que os mutantes simples (gE and tk-deletados) são atenuados para bezerros e estabelecem e/ou reativam infecção latente ineficientemente. Já o duplo mutante BoHV-5gEDTKD é atenuado e parece não estabelecer e/ou não reativar eficientemente a infecção latente. Portanto, os vírus recombinantes, e em especial o duplo mutante BoHV-5gEDTKD apresentam um fenótipo compatível com a sua inclusão em vacinas vivas modificadas.

E-mail: eduardofurtadoflores@gmail.com Bovine herpesvirus 5 (BoHV-5) is an important pathogen of cattle in South America and efforts have been made to produce safer and more effective vaccines. In addition to afford protection, herpesvirus vaccines should allow serological differentiation of vaccinated from naturally, latently infected animals. We previously reported the construction and characterization in vitro of a double mutant BoHV-5 (BoHV-5gE/TK”D) lacking the genes encoding thymidine kinase (tk) for attenuation, and glycoprotein E (gE) as the antigenic marker, as a vaccine candidate strain (Brum et al. 2010a). The present article reports an investigation on the attenuation and immunogenicity of this recombinant in calves. In a first experiment, 80 to 90-day-old seronegative calves (n=6) inoculated intranasally with the recombinant (titer of 107.5TCID50) shed virus in low to moderate titers in nasal secretions for up to 6 days, yet did not develop any respiratory, systemic or neurological signs of infection. At day 30 post-infection (pi) all calves had BoHV-5 specific neutralizing (VN) antibodies in titers of 4 to 8 and were negative for anti-gE antibodies in a commercial ELISA test. Administration of dexamethasone (0.1mg/kg/day during 5 days) to four of these calves at day 42 pi did not result in virus shedding or increase in VN titers, indicating lack of viral reactivation. Secondly, a group of 8-month-old calves (n=9) vaccinated intramuscularly (IM) with the recombinant virus (107.5TCID50/animal) did not shed virus in nasal secretions, remained healthy and developed VN titers from 2 to 8 at day 42 post-vaccination (pv), remaining negative for gE antibodies. Lastly, 21 calves (around 10 months old) maintained under field conditions were vaccinated IM with the recombinant virus (titer of 107.3TCID50). All vaccinated animals developed VN titers from 2 to 16 at day 30 pv. A boost vaccination performed at day 240 pv resulted in a rapid and strong anamnestic antibody response, with VN titers reaching from 16 to 256 at day 14 post-booster. Again, serum samples remained negative for gE antibodies. Selected serum samples from vaccinated animals showed a broad VN activity against nine BoHV-5 and eight BoHV-1 field isolates. These results show that the recombinant virus is attenuated, immunogenic for calves and induces an antibody response differentiable from that induced by natural infection. Thus, the recombinant BoHV-5gE/TKD is an adequate candidate strain for a modified live vaccine.

Além de conferirem proteção, vacinas contra os herpesvírus animais devem permitir a diferenciação sorológica entre animais vacinados e infectados naturalmente. Foi recentemente relatada a construção e caracterização in vitro de um recombinante do BoHV-5 defectivo na enzima timidina quinase (tk) para atenuação e, na glicoproteína E (gE) como marcador antigênico, como potencial cepa vacinal (Brum et al. 2010a). O presente artigo relata uma investigação sobre da atenuação e imunogenicidade do recombinante BoHV-5gE/TKD em bezerros. Em um primeiro experimento, bezerros soronegativos, com 80 a 90 dias de idade (n=6), inoculados pela via intranasal (IN) com o vírus recombinante (título de 107,5TCID50) excretaram baixos títulos de vírus nas secreções nasais por até seis dias, mas não desenvolveram sinais sistêmicos, respiratórios ou neurológicos. No dia 30 pós-infecção (pi), todos os animais possuíam anticorpos neutralizantes contra o BoHV-5, em títulos entre 4 e 8, mas permaneceram negativos para anticorpos contra a gE. Administração de dexametasona (Dx) a quatro desses bezerros no dia 42 pi (0.1mg/kg/dia durante 5 dias) não resultou em excreção viral ou em aumento dos títulos de anticorpos, indicando ausência de reativação viral. Em um segundo experimento, vacinação intramuscular (IM) de bezerros com 8 meses de idade (n=9) com o recombinante (107,5TCID50/animal) não resultou em excreção viral ou em manifestações clínicas. Os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 8 no dia 42 pós-vacinação (PV) e permaneceram negativos para anticorpos anti-gE. Finalmente, 21 bezerros (aproximadamente 10 meses de idade) mantidos a campo foram vacinados com o recombinante (107,3TCID50) pela via IM. Todos os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 16 no dia 30pv. Revacinação desses animais no dia 240 pv provocou uma resposta anamnéstica rápida e intensa, resultando em títulos neutralizantes entre 16 e 256 no dia 14 pós-revacinação. O soro de todos os animais permaneceu negativos para anticorpos contra a gE. Amostras de soro dos animais vacinados apresentaram atividade neutralizante cruzada frente a nove isolados de BoHV-5 e oito de BoHV-1. Esses resultados demonstram que o recombinante é atenuado e imunogênico para bezerros e induz uma resposta sorológica diferenciável da resposta à infecção natural. Portanto, o recombinante se constitui em um potencial candidato a cepa vacinal.

E-mail: eduardofurtadoflores@gmail.com The activity of three anti-herpetic drugs (Acyclovir [ACV], Gancyclovir [GCV] and Foscarnet [PFA]) was tested against bovine herpesvirus 1 (BoHV-1), 2 (BoHV-2) and 5 (BoHV-5) in vitro using the plaque reduction assay. Different drug concentrations were tested against one hundred 50% tissue culture infectious dose (TCID50) of the respective viruses. Drug concentrations lower than 200µg/mL resulted in viability rates of more than 80% for MDBK and Hep2 cells in the MTT test (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). The selectivity index (IS) of the drugs was calculated dividing the concentration of the drug that is cytotoxic for 50% of the cells (CC50) by the concentration of the drug that was effective in reducing by 50% the number of viral plaques (EC50) for the three herpesviruses. Thus, ACV was shown to be moderately active against BoHV-1 (EC50: 112.9mg/mL; IS: 4.5), BoHV-2 (EC50: 114.2mg/mL; IS: 4.5) and BoHV-5 (EC50: 96.9mg/mL; IS: 5.3). GCV was effective against BoHV-2 (EC50: 33.5mg/mL; IS: 16.6), moderately effective against BoHV-5 (EC50: 123.2mg/mL; IS: 4.5) and poorly active against BoHV-1 (EC50: 335.8mg/mL; IS: 1.7). PFA exhibited the highest antiviral activity, being the only drug that, at concentration of 100mg/mL, completely inhibited plaque formation by all three viruses. PFA was the most effective in vitro against BoHV-1 (EC50: 29.5mg/mL; IS: 42.2), BoHV-2 (EC50: 45.2mg/mL; IS: 27.6) and BoHV-5 (EC50: 7.8mg/mL; IS: 160.6). Thus, the results indicate that PFA is a promising candidate for experimental therapeutic testing in vivo against bovine herpesviruses.

Para isso, utilizou-se o teste de redução de placas virais em cultivo celular, testando-se diferentes concentrações dos fármacos frente a 100 doses infectantes para 50% dos cultivos celulares (DICC50) dos respectivos vírus. Pelo teste de MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide), verificou-se que concentrações inferiores a 200µg/mL dos três antivirais resultaram em índices de viabilidade de células MDBK e Hep2 superiores a 80%. Com base na concentração citotóxica para 50% das células (CC50) e na concentração dos fármacos efetiva para inibir em 50% o número de placas virais (EC50), calculou-se o índice de seletividade (IS) dos antivirais para os três herpesvírus. Assim, o ACV demonstrou ser moderadamente ativo frente ao BoHV-1 (EC50: 112,9mg/mL e IS: 4,5), ao BoHV-2 (EC50: 114,2 mg/mL e IS: 4,5) e BoHV-5 (EC50: 96,9mg/mL e IS: 5,3). O GCV apresentou atividade moderada frente ao BoHV-2 (EC50: 33,5mg/mL e IS: 16,6) e, em menor grau, contra o BoHV-5 (EC50: 123,2mg/mL e IS: 4,5), sendo ineficaz frente ao BoHV-1 (EC50: 335,8mg/mL e IS: 1,7). O PFA apresentou atividade antiviral mais pronunciada, sendo o único fármaco que, na concentração de 100µg/mL, inibiu completamente a produção de placas pelos três vírus testados. O PFA foi o mais efetivo in vitro frente ao BoHV-1 (EC50: 29,5mg/mL e IS: 42,2), ao BoHV-2 (EC50: 45,2mg/mL e IS: 27,6) e ao BoHV-5 (EC50: 7,8mg/mL e IS: 160,6). E-mail: eduardofurtadoflores@gmail.com The activity of Foscarnet (PFA) against three bovine herpesviruses (BoHVs) was previously demonstrated in cell culture. Herein we evaluated the effects of PFA on the infection and disease by BoHV-1 and BoHV-5 in a rabbit model. Rabbits inoculated with BoHV-5 in the conjunctival sac (IC) and treated with PFA (100 mg/kg/day) from day 1 to 17 post-inoculation (pi) shed less virus between days 2 and 6 pi comparing to untreated controls; this difference was significant at day 3 pi [F(9,108) = 2,23; P<0.03]. The morbidity and mortality rates of rabbits inoculated with BoHV-5 IC or intranasally (IN) were also significantly reduced in PFA-treated rabbits (50%; 11/22) comparing to untreated controls (95.4%; 21/22) (P<0.0008). In rabbits inoculated IC with BoHV-1, a reduction in virus shedding was observed in PFA-treated animals between days 1 and 4 pi; 6 and 7 pi. In addition, PFA-treated rabbits presented a longer incubation period and a shorter clinical course comparing to untreated controls (P<0.005 and P<0.04, respectively). The frequency and severity of ocular signs were also reduced in the PFA-treated group. These results demonstrate that PFA is effective against BoHV-1 and BoHV-5 in vivo and open the way towards its use in experimental therapy of herpetic infections in domestic animals.

The application of molecular diagnostic techniques represents an important contribution for the study of BoHV. This paper describes the detection of BoHV-5 and BoHV-1 by a specific multiplex PCR assay in 76 paraffin-embedded samples from central nervous system (CNS) of cattle with neurological disorders. The samples were divided into 2 groups according to the histological features: Group 1 was composed of 40 cases of necrotizing meningoencephalitis (characteristic of BoHV infection), and Group 2 was composed of 36 cases of nonspecific nonsuppurative meningoencephalitis. Positive results for BoHV-5 accounted for 40% of the samples in the group 1 and 33% in the group 2. No detection of BoHV-1 was recorded.

Com base nas alterações histológicas, as amostras foram separadas em 2 grupos: o Grupo 1 era composto de 40 amostras de bovinos com meningoencefalite necrosante característica da infecção por BoHV; no Grupo 2 estavam 36 amostras de casos com encefalite não-supurativa inespecífica. Identificação de BoHV-5 foi constatada em 40% das amostras do grupo 1 e em 33% das amostras do grupo 2. Não houve amplificação de DNA de BoHV-1 em nenhuma amostra.

E-mail: proehe@gmail.com The serum neutralization (SN) test is the gold standard method to measure neutralizing antibodies to bovine herpesviruses. However, in view of the further subdivisions of bovine herpesviruses in types/subtypes, defining which virus to use at challenge in SN tests may be difficult. In view of that, this study was carried out to re-evaluate (SN) sensitivity with different types/subtypes of bovine herpesviruses types 1 (BoHV-1) and 5 (BoHV-5) as challenge viruses. Bovine sera (n=810) were collected from two distinct geographic regions and tested by SN with three type 1 viruses (BoHV-1.1 strains “Los Angeles” and “EVI123/98”; BoHV-1.2a strain “SV265/96”) and three type 5 viruses (BoHV-5a strain “EVI88/95”; BoHV-5b strain “A663” and BoHV-5c “ISO97/95”). SN tests were performed with a 1 hour incubation of the serum-virus mixtures at 37ºC against 100 TCID50 of each of the viruses. SN sensitivity varied greatly depending on the challenge virus used in the test. The highest sensitivity (327 positive/810 total sera tested; 40.37%) was attained when the positive results to the six viruses were added together. No association could be found between any particular type or subtype of virus and the sensitivity of the test. When positive results to each single strain were considered, SN sensitivity varied from 41.7% to 81.7%, depending on the virus and the geographic region of origin of the sera. Variation was detected even when challenge viruses belonged to the same subtype, where disagreement between positive results reached 41%. These results indicate that one hour incubation SN tests against single viruses, as performed here, may display a significantly low sensitivity (p=0.05); performing SN tests against a number of different viruses may increase considerably SN sensitivity. Furthermore, the choice of virus used for challenge is critical in SN tests. In addition, sera from different geographic regions may give rise to disagreeing results with different strains of BoHV-1 and BoHV-5. This might be particularly relevant for control programs and in international trade, were maximum sensitivity should be targeted.

Soros bovinos (n=810) foram coletados de duas regiões geográficas distintas e testados frente a amostras do tipo 1 (BoHV-1.1: amostras “Los Angeles” e “EVI123/98”, BoHV-1.2a: amostra “SV265/96”) e três amostras do tipo 5 (BoHV-5a: “EVI88/95”; BoHV-5b: “A663” e BoHV-5c “ISO97/95”). Os testes de SN foram realizados com incubação de 1 hora a 37ºC da mistura soro-vírus, frente a 100 doses infectantes para 50% dos cultivos celulares (DICC50) de cada um dos vírus. A sensibilidade da SN variou grandemente em função do vírus utilizado no teste. A maior sensibilidade (327 soros positivos/810 soros testados; 40.37%) foi alcançada quando os resultados positivos frente aos seis diferentes vírus foram somados. Nenhuma associação foi detectada entre determinado tipo/subtipo de vírus e a sensibilidade do teste. Quando resultados positivos frente a cada vírus foram considerados isoladamente, a sensibilidade da SN variou entre 41,7% a 81,7%, dependendo do vírus de desafio e da região geográfica de origem das amostras de soro. Variação foi detectada mesmo quando as amostras de desafio pertenciam a um mesmo subtipo; a discrepância entre os resultados positivos atingiu até 41%. Estes resultados indicam que testes de SN contra amostras isoladas de vírus podem apresentar uma sensibilidade notadamente baixa; o emprego de diferentes amostras de vírus de desafio pode aumentar consideravelmente a sensibilidade da prova. Além disso, a escolha da amostra de vírus para a realização do teste é crítica. Outro achado importante é que sorors de diferentes regiões geográficas podem dar resultados discordantes frente a diferentes amostras de BoHV-1 e BoHV-5. Estes achados são particularmente relevantes para programas de controle destas infecções e para o comércio internacional, onde a sensibilidade deve ser maximizada.
