E-mail: alage@vet.ufmg.br The present update deals with two of the most important sexually transmitted diseases of cattle: bovine genital campylobacteriosis and bovine genital trichomonosis. Epidemiological aspects, mainly their distribution in Brazil, alongside with their diagnosis in cattle are presented and commented. The main points in their diagnoses, including the description of the techniques and the interpretation of the results are also reviewed. Finally the control and prevention of both diseases are discussed.

Também são revisados aspectos importantes de diagnóstico, incluindo as técnicas e interpretação dos resultados, além de medidas de controle para ambas as doenças.

E-mail: aiesca@cpap.embrapa.br An indirect enzyme-linked immunosorbent assay was developed to detect antigen-specific secretory IgA antibodies to Campylobacter fetus subsp. venerealis in bovine vaginal mucus with a protein extract of the Campylobacter fetus subsp. venerealis by the acid glycine extraction method. Mean optical density measurement (l=450 nm) was 0.143±0.9. The most immunoreactive protein bands of the Campylobacter fetus subsp. venerealis or Campylobacter fetus subsp. fetus recognized by IgA in immunoblotting, using bovine vaginal mucus samples, migrate at 42.6 kDa. The protein that migrates at 93 kDa was recognized exclusively for C. fetus subsp. venerealis. A positive vaginal mucus sample of a cow from negative herd recognized antigens of C. jejuni subsp. jejuni e C. fetus subsp. fetus.

E-mail: aiesca@cpap.embrapa.br Foi padronizado um ensaio imunoenzimático do tipo indireto para detecção de imunoglobulina A (ELISA IgA) anti- Campylobacter fetus subp. venerealis em muco cérvico- vaginal bovino utilizando um extrato protéico de Campylobacter fetus subsp. venerealis produzido pelo método de extração ácida pelo tampão de glicina (0,2M; pH2,2). A média dos valores de densidade ótica (DO450) foi de 0,143±0,09. As bandas protéicas dos antígenos de Campylobacter fetus subsp. venerealis e de Campylobacter fetus subsp. fetus melhor reconhecidas pela IgA do muco cérvico- vaginal migraram em 42,6 kDa mas a proteina evidenciada em 93 kDa foi reconhecida exclusivamente pelo Campylobacter fetus subsp. venerealis. Os anticorpos presentes na amostra de muco vaginal testada no “immunoblotting” que apresentou resultado positivo no ELISA IgA, reconheceu antígenos de C. jejuni subsp. jejuni e C. fetus subsp. fetus.

E-mail: agueda.vargas@gmail.com Bovine genital campylobacteriosis is a common venereal disease of cattle; the prevalence of this disease can be underestimated mostly because of the nature of the etiological agent, the microaerobic Campylobacter fetus subspecies venerealis. The purpose of the current study was to evaluate the utilization of polymerase chain reaction (PCR) in the diagnosis of genital campylobacteriosis in samples obtained from bull prepuce aspirate, cow cervical mucus, and abomasum contents of aborted fetuses, collected into enrichment medium. Five different DNA extraction protocols were tested: thermal extraction, lysis with proteinase K, lysis with guanidine isothiocyanate, lysis with DNAzol, and lysis with hexadecyltrimethylammonium bromide (CTAB). The specificity, sensitivity, and technical application of the PCR assay were also evaluated with clinical samples and compared to bacterial isolation by standard culture. DNA extraction by the CTAB protocol provided better results in PCR, and it was able to detect 63 colony-forming units per ml of C. fetus. Out of 277 clinical samples tested, 68 (24%) were positive for Campylobacter fetus using PCR, while only 8 (2.8%) of the samples were positive by bacterial isolation in solid medium, proving the superiority of the PCR technique when compared to the standard isolation method, and providing evidence for its usefulness as a better screening test in cattle for the diagnosis of bovine genital campylobacteriosis.

A prevalência desta doença pode ser subestimada na maioria das vezes pela natureza microaeróbica do agente etiológico, Campylobacter fetus subspecies venerealis. O propósito do presente estudo foi avaliar a utilização da reação de polimerase em cadeia (PCR) no diagnóstico de campilobacteriose genital em amostras obtidas de aspirado prepucial de touros, muco cervical de vacas e conteúdo abomasal de fetos abortados, coletados em meio enriquecido. Cinco protocolos diferentes de extração de DNA foram testados: termo extração, lise com proteinase K, lise com guanidine isothiocyanate, lise com DNAzol e lise com hexadeciltrimetilamônio brometo (CTAB). A especificidade, sensibilidade e a aplicação da técnica da PCR foram também avaliadas com amostras clínicas e comparadas com bactérias isoladas por cultura padrão. DNA extraído pelo protocolo de CTAB demonstrou os melhores resultados na PCR, e foi capaz de detectar 63 unidades formadoras de colônias de C. fetus por ml de meio. Das 277 amostras clínicas testadas, 68 (24%) foram positivas para Campylobacter fetus pela PCR, enquanto 8 (2,8%) das amostras foram positivas por isolamento bacteriológico, provando a superioridade da técnica de PCR quando comparada com métodos padrão de isolamento, e fornecendo evidências de sua utilização como um teste de melhor projeção para diagnóstico em campilobacteriose genital bovina.

E-mail: fervogel@smail.com The aim of this study was to determine the occurrence of anti-Neospora caninum antibodies in serum samples collected from 260 bovine fetuses, between July 2007 and March 2008, in an abattoir in the municipality of Santa Maria, Rio Grande do Sul, Brazil. An indirect fluorescent antibody test was used to detect anti-N. caninum immunoglobulins G (IgG) and M (IgM), using a cut-off of 1:25. Considering IgG or IgM detection, 15% (39/260) of the samples tested were found positive. Among the positive samples, 38 (97.4%) were positive for IgG and 6 (15.4%) for IgM. However 5 (12.8%) of it were positive for both IgG and IgM. These results are in agreement with the proven ability of N. caninum to fetal infection. IgM testing was few relevant to detect N. caninum transplacental infection through fetal bovine serum analisys.

E-mail: fervogel@smail.com O objetivo deste estudo foi determinar a ocorrência de anticorpos anti-Neospora caninum em 260 amostras de soro coletadas de fetos bovinos de julho de 2007 a março de 2008, em abatedouro do município de Santa Maria, Rio Grande do Sul, Brasil. Para detecção de anticorpos anti-N. caninum, a técnica de imunofluorescência indireta foi utilizada tanto para a detecção de imunoglobulinas G e M. Amostras com títulos e” 25 foram consideradas positivas. Das 260 amostras testadas, 15% (39/260) foram positivas para anticorpos anti-N. caninum. Destas, em 38 (97,4%) foi detectada a presença de IgG anti-N. caninum e em seis (15,4%) de IgM. Em cinco amostras (12,8%) detectaram-se ambos, IgG e IgM. Os resultados reafirmam a habilidade do N. caninum em determinar infecção fetal. A pesquisa de IgM foi de limitada importância na detecção da infecção via transplacentária em soro fetal bovino.

Os exemplares tiveram o sistema arterial preenchido com solução aquosa de, a 50% de Neoprene Látex, corado e, em seguida foram submetidos à fixação em solução aquosa, a 10%, de formaldeído. Os lobos torácicos do timo foram vascularizados por ramos diretos das artérias torácica interna direita (63,33%) e esquerda (53,33%), subclávia esquerda (3,33%), vertebral esquerda (3,33%), cervical superficial direita (3,33%) e esquerda (3,33%), carótida comum esquerda (3,33%), coronária direita (3,33%) e pelos troncos braquiocefálico (33,33%) e costocervical (3,33%). Observou ainda os ramos indiretos das artérias torácica interna direita (70%) e esquerda (76,67%), subclávia esquerda (23,33%), cervical superficial esquerda (3,33%) e do arco aórtico (6,67%).

E-mail: limaemm@unb.br The origins and distributions of the arteries that vascularization the thoracic lobes of the thymus, right and left, were studied in 30 swine of the lineage C40, fetus, being 12 males and 18 females. The animals had the arterial system filled with aqueous colored solution in a 50% of Neoprene Latex, and than were fixed in 10% aqueous solution formaldehyde. The thoracic lobes of thymus were vascularization by direct branches of the right (53.33%) and left (63.33%) internal thoracic arteries, left subclavian (3.33%), left vertebral (3.33%), right (3.33%) and left (3.33%) superficial cervical, left common carotid (3.33%), right coronary (3.33%) and brachiocephalic trunk (33.33%) and costocervical trunk (3.33%). The right internal thoracic arteries (70%) and left internal thoracic (76.67%), left subclavian (23.33%), left superficial cervical (3.33%) of the aortic arch (6.67%) provided indirect branches for the correspondent thoracic lobes of the thymus in each side.

Nevertheless, only PCV2 has been associated with different clinical syndromes, thus representing a major problem to the pig industry. The association of cases of swine abortions and stillborns with PCV1 and PCV2 and PPV was studied retrospectively (2005-2007). Additional pathogens were also investigated in lesioned fetuses. The studied litters included stillborn piglets and several mummified fetuses of varied sizes. Ventricular dilatation, myocardial pale areas, and mesocolic edema were the gross lesions. Escherichia coli was detected as co-infecting with PCV2 the cases in which mesocolic edema was seen. Microscopic lesions included non-suppurative myocarditis, myocardial necrosis and fibrosis, mineralization foci and intranuclear inclusion bodies in cardiomyocytes, and interstitial mononuclear pneumonia. Samples from 7 (5.78 per cent) of 121 aborted fetuses and stillborn piglets had lesions consistent with a viral cause and showed both positive anti-PCV2 immunostaining as well as PCV2-PCR. In samples from 3 (2.47 per cent) of these 7 fetuses, co-infection with PPV was confirmed by Nested-PCR. Both viruses were detected in fetuses at different stages of gestation. Viral antigens of PCV2 were detected by immunohistochemistry mainly in macrophages and myocytes. PCV1 individually was not detected in any of these affected fetuses, but it was associated with PCV2 and/or PPV in some of them. These findings indicate that PCV2 alone or in association with PPV should be kept in mind when investigating causes of infectious abortion in pigs in Brazil.

#18 - Susceptibilidade antimicrobiana de Campylobacter fetus subsp. venerealis isolado de bovinos

[Antimicrobial susceptibility of Campylobacter fetus subsp. venerealis isolated from cattle.] Susceptibilidade antimicrobiana de Campylobacter fetus subsp. venerealis isolado de bovinos. E-mail: agueda@ccr.ufsm.br Venereal campylobacteriosis is associated with infection of Campylobacter fetus subsp. fetus and Campylobacter fetus subsp. venerealis. The etiological agent is transmitted by natural bull breeding or artificial insemination using contaminated semen. The present study aimed to determine the in vitro susceptibility of C. fetus subsp. venerealis isolates to antimicrobial drugs generally used in clinical and semen treatment. Reference strains of C. fetus subsp. fetus and C. fetus subsp. venerealis and 21 C. fetus isolates were tested. The susceptibility test was performed by using the modified Kirby-Bauer diffusion disc method. C. fetus subsp. fetus reference strain was resistant to lincomycin and penicillin. C. fetus subsp. venerealis was susceptible to all antimicrobial tested, with exception to C.C. KrewerDnalidixic acid. C. fetus subsp. venerealis samples were sensible to amikacin, ampicillin, cefalotin, streptomycin, gentamycin, penicillin and tetracycline. Drug resistance was observed on 42.86% of lincomycin, 4,76 % of enrofloxacin, and 100% to nalidixic acid. In addition 4.76% of the isolates showed intermediate susceptibility to enrofloxacin, neomycin, polimixin B and 9.52% to lincomycin. E-mail: agueda@ccr.ufsm.br Venereal campylobacteriosis is associated with infection of Campylobacter fetus subsp. fetus and Campylobacter fetus subsp. venerealis. The etiological agent is transmitted by natural bull breeding or artificial insemination using contaminated semen. The present study aimed to determine the in vitro susceptibility of C. fetus subsp. venerealis isolates to antimicrobial drugs generally used in clinical and semen treatment. Reference strains of C. fetus subsp. fetus and C. fetus subsp. venerealis and 21 C. fetus isolates were tested. The susceptibility test was performed by using the modified Kirby-Bauer diffusion disc method. C. fetus subsp. fetus reference strain was resistant to lincomycin and penicillin. C. fetus subsp. venerealis was susceptible to all antimicrobial tested, with exception to C.C. KrewerDnalidixic acid. C. fetus subsp. venerealis samples were sensible to amikacin, ampicillin, cefalotin, streptomycin, gentamycin, penicillin and tetracycline. Drug resistance was observed on 42.86% of lincomycin, 4,76 % of enrofloxacin, and 100% to nalidixic acid. In addition 4.76% of the isolates showed intermediate susceptibility to enrofloxacin, neomycin, polimixin B and 9.52% to lincomycin. #19 - Efficiency of a vaccine against Bovine Campylobacteriosis with autochdtonous cultores in oily adjuvant

Abstract in English:

The study of a vaccine containing 13 cultures of Campylobacter fetus subsp. venerealis and one intestina/is was carried out. These cultures were isolated in Rio de Janeiro State from animals suffering of reproductive diseases, from an aborted fetus and from the foreskin cavity of infected bulis. Those cultures were inactivated by a 0,5% formalin solution and treated by heating at 37ºC for 24h in oily adjuvant. The immunogenicity of the vaccine was assayed on two groups of eight half-breed virgin Holstein aging over two year old weighing over 300 kg. This was performed by infecting subcutanously 5.0 m1 of the vaccine in both groups. The second group animals received a 5.0 m1 booster 14 days later. lndirect immunofluorescence and serum-agglutination test showed an optimal antibody production at 30th and 36th days, respectively for animals vaccinated with one and two doses. Sixty days after the first vaccination the heifers were infected with 4 x 106 cells (culture RJ 14, Campylobacter fetus subsp. venerealis) at the moment of artificial insemination at the cervicovaginal region. The reproductive efficacy of 100 and 75%, for one and two doses respectively, with three inserninations, showed to be the best way to assay immunity confered by the vaccine. On the other hand, for the eight heifers composing group II, 15 inseminations were necessary for na efficiency of 100%, or 1.8 semen doses, for fertilization. The result was superior to that of group III, in which 16 inseminations were necessary for pregnancy in 7 out of 8 heifers; na efficiency of 7 5%, or 2.2 semen doses, for fertilization. The control group of 8 heifers presented only 4 pregnancies from 19 inserninations, or 4. 7 semen doses, for fertilization. The opsonizating effect of IgG was the responsible by the absence of microorganisms in the cervico mucous of the vaccinated heifers. However the presence of rnicroorganisms in some heifers did not affect the reproductive efficacy in these animals.

O presente estudo trata de uma vacina contendo 13 culturas de Campylobacter fetus subsp. venerealis e uma cultura do Campylobacter fetus subps. intestinalis, isoladas no Estado do Rio de Janeiro, provenientes de animais com problemas de reprodução, de um feto abortado e da cavidade prepucial de touros infectados, inativada pelo formal a 0,5% e normalizada pelo calor à 37°C por 24h em adjuvante oleoso. Para a pesquisa da ação imunogênica da vacina foram empregados dois grupos compostos por oito novilhas virgens mestiças de holandês, com idades acima de dois anos e pesos acima de 300 kg. A vacinação realizou-se com uma aplicação de 5,0 m1 da vacina na região cervical via subcutânea, sendo que no segundo grupo, houve uma vacinação com mais 5,0 m1 com intervalo de 14 dias entre as doses. Os testes de imunofluorescência indireta e de soroaglutinação, indicavam haver um ótimo pique na média da produção de anticorpos para o grupo vacinado com uma dose ao 309 dia e no 369 dia para o grupo vacinado com duas doses. Após 60 dias da primeira vacinação, os animais foram contaminados com 4 x .106 células da amostra RJ 14 do Campylobacter fetus subsp. venerealis no momento da primeira inseminação artificial na região anterior da vagina. A eficâcia de 100 e 75% para uma dose e duas doses da vacina respectivamente com base no desempenho reprodutivo obtido com no máximo três inseminações, demonstrou ser o melhor método de avaliação da imunidade conferida pela vacina. Por outro lado, para as oito novilhas que compunham o grupo II foram necessárias 15 inseminações para se obter a eficiência de 100%, ou seja, 1,8 dose de sêmen por fecundação. Tal resultado foi superior ao do grupo III, no qual, dentre oito novilhas, sete ficaram gestantes, sendo necessárias 16 inseminações para se obter uma eficiência de 75%, ou seja, 2,2 doses de sêmen por fecundação. O grupo controle apresentou apenas quatro novilhas gestantes e requereu 19 inseminações, ou seja, 4, 7 doses de sêmen por fecundação. A opsonização pelo IgG foi a responsável pela ausência do microrganismo no muco cérvico-vaginal de novilhas vacinadas, porém, a presença de microrganismo no muco cérvico-vaginal de algumas novilhas não afetou a eficiência r*eprodutiva desses animais.
