#11 - Influence of prolonged flaxseed (Linum usitatissimum) consumption over epididymis and testicle histoarchitecture of Wistar rats, 37(6):650-656

E-mail: chagas.m@gmail.com Flaxseed is considered a functional food with several health benefits. However, because of its high phytoestrogen content, flaxseed influences hormone metabolism and affects the gonadal biomorphology. In this study, computerized histomorphometry was used to evaluate seminiferous and epididymal tubules, considering the different regions of the epididymis (head, body and tail) of rats subjected to a prolonged diet of flaxseed. Young adult male Wistar rats (n=20) were divided into 2 groups during their lactation period: Control Group (CG), fed casein-based meals and Flaxseed Group (FG), fed a 25% flaxseed meal. After 250 days of continuous ingestion, the animals were euthanized and a blood sample was collected. The testicles and epididymis were removed and fixed in buffered formalin solution. The samples were subjected to routine histological paraffin techniques and stained with hematoxilin and eosin. Immunostaining was performed using an antivimentin antibody for Sertoli cell identification. For morphometry, images of the slides were scanned and analyzed using Image J to determine the epithelial height, tubular and luminal diameter and tubular and luminal area. In the hormonal evaluation, FG had a higher serum concentration of estrogen (P=0.001), but no change was observed in the concentration of testosterone. The morphometric assay of seminiferous tubules and epididymal regions revealed no significant differences between the analyzed groups. Similarly, Sertoli cell quantification showed no significant differences in the FG (P=0.98). These results revealed that the continuous and prolonged intake of 25% flaxseed meals from gestation to 250 days of age, even with a significant increase in serum levels of estradiol, does not exert adverse effects on the testicular and epididymal structure or on the cells participating in the spermatogenesis of rats.

Entretanto, devido ao seu elevado teor de fitoestrógenos, esta semente pode influenciar no metabolismo hormonal e interferir na biomorfologia gonadal. Neste estudo, utilizamos a histomorfometria computadorizada para avaliar os túbulos seminíferos e epididimários, considerando as diferentes regiões do epidídimo (cabeça, corpo e cauda) de ratos submetidos a uma dieta prolongada de semente de linhaça. Foram utilizados ratos Wistar machos adultos jovens (n=20) divididos em 2 grupos, durante o período de lactação: Grupo Controle (GC) a base de caseína e Grupo Linhaça (GL) alimentados com 25% de semente de linhaça. Ao final de 250 dias de ingestão contínua, os animais foram sacrificados e amostra de sangue foi coletada. Os testículos e epidídimos foram retirados e fixados em formol tamponado. As amostras foram submetidas ao processamento histológico de rotina para parafina e coradas em hematoxilina e eosina. Foi feita a imunomarcação com anticorpo antivimentina para identificação das células de Sertoli. Para morfometria, as imagens das lâminas foram digitalizadas e analisadas pelo software ImageJ para obtenção dos dados de altura epitelial, diâmetro e área tubular e luminal. Na avaliação hormonal o GL teve maior concentração de estrógeno sérico (p=0,001), mas nenhuma mudança na concentração de testosterona foi observada. Nos parâmetros morfométricos dos túbulos seminíferos e das regiões epididimárias, não houve diferenças significativas entre os grupos analisados. Da mesma forma, a quantificação das células de Sertoli não apresentaram diferenças significativas no GL (p=0,98). Estes resultados mostraram que o consumo contínuo e prolongado de 25% de semente de linhaça desde período gestacional até 250 dias de idade, mesmo com o aumento significativo nos níveis séricos de estradiol, não exerceram efeitos adversos sobre a estrutura testicular e epididimária, assim como nas células participantes da espermatogênese em ratos.

Other rats were fasted for two weeks. Group 2 was applied to a group immediately after starvation (day 14). Groups 3 to 6 were refed in days 16 till 22, respectively. At the end of each period, blood and tissue samples were taken and histopathological and serum analysis, including serum electrolytes (calcium, phosphorus, sodium, potassium), the energy parameters (glucose, insulin, cortisol) and the liver enzymes (ALT, AST, ALP) were determined. Insulin decreased by starvation and then showed an increasing trend compared to starvation period, which the highest amount of this parameter was observed eight days post-refeeding. Serum glucose level showed the opposite pattern of insulin. Histopathological examination of the tissue sections revealed clear vacuoles after starvation and refeeding, in which the severity of lesions gradually decreased during refeeding. The cortisol level decreased by starvation and then increased during refeeding. Also, potassium and phosphorus concentrations declined by refeeding and the serum sodium and potassium levels were changed in the relatively opposite manner. The calcium level decreased by starvation and then increased during refeeding. These results could help recognize and remedy the refeeding syndrome.

E-mail: bebel@ufv.br Osteoporosis is a multifactorial disease of high prevalence and has great impact on quality of life, because the effects on bone structure increase the risk of fractures, what may be very debilitating. Based on the observation that patients with depression have lower bone mineral density than healthy individuals, many studies have indicated that stress could be an aggravating factor for bone loss. This study evaluates the effect of a protocol of chronic mild stress (CMS) on parameters of bone assessment in male and female rats. Five 5-monh-old rats of each sex underwent a schedule of stressor application for 28 days. Stressors included cold, heat, restraint, cage tilt, isolation, overnight illumination, and water and food deprivation. Five rats of each sex were kept under minimum intervention as control group. The animals were weighed at beginning and end of the period, and after euthanasia had their bones harvested. Femur, tibia and lumbar vertebrae were analyzed by bone densitometry. Biomechanical tests were performed in femoral head and diaphysis. Trabecular bone volume was obtained from histomorphometric analysis of femoral head and vertebral body, as well as of femoral midshaft cross-sectional measures. Not all parameters analyzed showed effect of CMS. However, tibial and L4 vertebral bone mineral density and cross-sectional cortical/medullar ratio of femoral shaft were lower in female rats submitted to the CMS protocol. Among male rats, the differences were significant for femoral trabecular bone volume and maximum load obtained by biomechanical test. Thus, it could be confirmed that CMS can affect the balance of bone homeostasis in rats, what may contribute to the establishment of osteopenia or osteoporosis.

Este estudo avalia o efeito de um protocolo de estresse moderado crônico (EMC) em parâmetros de avaliação óssea em ratos machos e fêmeas. Cinco animais de cada sexo, com cinco meses de idade, foram submetidos a um cronograma de aplicação de estressores durante 28 dias. Os estressores incluídos foram: frio, calor, contenção, inclinação da gaiola, isolamento, iluminação durante a noite e privação de água e ração. Cinco animais de cada sexo foram mantidos com um mínimo de intervenção como grupo controle. Os animais foram pesador no início e no final do período, e após eutanásia tiveram seus ossos coletados. Fêmur, tíbia e vértebra lombar foram analisados por densitometria óssea. Testes biomecânicos foram realizados na cabeça e na diáfise do fêmur. Volume trabecular ósseo foi obtido a partir de análise histomorfométricas da cabeça do fêmur e do corpo vertebral, bem como medidas da seção transversal diáfise femoral. Nem todos os parâmetros avaliados sofreram efeito do protocolo de EMC. No entanto, a densidade mineral óssea da tíbia e da vértebra L4 e a razão osso cortical/medula da seção transversal da diáfise femoral foram menores nas fêmeas submetidas ao protocolo. Entre os ratos machos, as diferenças foram significativas no volume trabecular ósseo da cabeça femoral e na carga máxima obtida no teste biomecânico. Assim, confirma-se que o protocolo de EMC pode afetar o equilíbrio da homeostase óssea em ratos, o que pode contribuir para o estabelecimento de osteopenia ou osteoporose.

E-mail: tharcia_kiara@hotmail.com The aim of this study was to evaluate the effect of Aqueous Extract from Peanut (EAA) in weight, serum biochemistry and liver histology of Wistar rats with normal hearing and a high fat diet. The survey was conducted using 40 male Wistar rats divided into four groups (n=0): GA (high fat diet), GB (fat diet+EAA), GC (normolipídica diet), and GD (normolipídica diet+EAA). After 8 weeks, the rats were euthanized and blood samples were collected to evaluate biochemical data (total cholesterol and its fractions, triglycerides, urea, creatinine, AST, ALT and glucose) and liver fragments for histological analysis. The animals of the GB group had a lower weight gain when compared with GA (XGB versus XGA= p<0.05), but CG and GD did not obtain statistical differences. The rats that received EAA had a reduction in cholesterol levels (XGB= versus XGA= p<0.05 vs. XGA and XGD= p<0.01), triglycerides (=XGB versus XGA and =XGD versus XGA= p<0,001) and more discreetly ALT levels. Blood glucose, urea and creatine remained within the reference values. Liver samples analyzed, the rats of different groups showed no histopathological changes. In conclusion, the EAA had preventive effects on weight gain and dyslipidemia.

Após 8 semanas, os animais foram eutanasiados e foram coletadas amostras sanguíneas para a avaliação de dados bioquímicos (Colesterol total e suas frações, triglicerídeos, uréia, creatinina, AST, ALT e glicemia) e fragmentos do fígado para análise histológica. Os animais do grupo GB tiveram um ganho de peso inferior quando comparados ao GA (XGB= versus XGA= p<0,05), já os grupos GC e GD não obtiveram diferenças estatísticas. Os animais que receberam o EAA tiveram uma redução nos níveis de colesterol (XGB= versus XGA= p<0,05 e XGD= versus XGA= p<0,01), dos triglicerídeos (XGB= versus XGA e XGD= versus XGA= p<0,001) e mais discretamente dos níveis de ALT. A glicemia, uréia e creatina permaneceram dentro dos valores de referência. As amostras hepáticas analisadas, dos ratos dos diferentes grupos, não apresentaram alterações histopatológicas. Conclui-se que O EAA apresentou efeitos preventivos sobre o ganho ponderal e dislipidemia.

E-mail: julianaarandas@hotmail.com This study aimed to evaluate the effect of oophorectomy on the morphometric variables of cardiomyocytes and quantification collagen in Wistar rats. Twenty rats were used and divided into two groups: GI - ovariectomized and GII - sham-ovariectomized. After recovery from anesthesia the animals were housed in separate cages and under the conditions of light / dark cycle 12/12h for a period of six months. After the experimental period, the animals were anesthetized, the heart removed and immersed in 10% formaldehyde. The fragments of the ventricles were subjected to histological analysis and stained with hematoxylin and eosin. Histomorphometric analysis (cell area and nuclear area and volume) were performed by light microscopy and ImageJ software version 1.44. The data were submitted to ANOVA and when significant, complemented by Student’s t test (p<0.001). There was a significant decrease in cellular and nuclear areas of the GI cardiomyocytes compared to GII and the area occupied by collagen was greater in cardiac muscle GI, when compared to GII. Therefore, it is concluded that the changes observed in the GI group cardiomyocytes, suggest a higher apoptotic activity in this group, due to decreased serum estrogen levels caused by ovariectomy and increased area occupied by collagen in oophorectomized group is associated the cardioprotective effect of estrogen.

Vinte ratas foram utilizadas e separadas em dois grupos: GI – ooforectomizada e GII – sham-ooforectomizada. Após a recuperação anestésica os animais foram acomodados em gaiolas e separados nas condições de ciclo claro/escuro 12/12h por um período de 6 meses. Passado o período experimental, os animais foram anestesiados, o coração retirado e mergulhado em formaldeído a 10%. Os fragmentos dos ventrículos foram submetidos ao processamento histológico e corados com hematoxilina e eosina. As análises histomorfométricas (área celular e área e volume nuclear) foram realizadas pelo microscópio de luz e software ImageJ versão 1.44. Os dados foram submetidos à análise ANOVA e quando significantes, complementados pelo teste t de student (p<0,001). Observou-se uma diminuição significativa das áreas celular e nuclear dos cardiomiócitos do grupo GI quando comparado ao grupo GII, bem como a área ocupada, pelo colágeno, no músculo cardíaco foi maior em GI, quando comparado a GII. Diante disso, conclui-se que as alterações observadas nos cardiomiócitos do grupo GI, sugerem uma maior atividade apoptótica nesse grupo, devido à diminuição dos níveis séricos do estrógeno provocado pela ooforectomia e o aumento da área ocupada pelo colágeno, no grupo ooforectomizado, está associada ao efeito cardioprotetor do estrógeno.


#16 - Histomorphometric analysis of the Achilles tendon of Wistar rats treated with laser therapy and eccentric exercise, 35(Supl.1):39-50

E-mail: msouza@ufv.br Low-level laser therapy is recommended for the treatment of tendinopathies despite the contradictory results related to the ideal dose of energy, wavelength and time of application. This study aimed to assess the effects of laser therapy and eccentric exercise on tendinopathy of the Achilles tendon of Wistar rats. Forty-eight adult male rats were randomly distributed into four groups (L= laser; E= eccentric exercise; LE = laser and eccentric exercise; and R= rest). Laser therapy (904nm/3J/cm²) and/or eccentric exercise (downhill walking; 15o incline treadmill; 12m/min; 50min/day) was started 24h after induction of unilateral tendinopathy and remained for 20 days. At 3, 7, 14 and 21 days after lesion induction, three rats from each group were euthanized and the tendons were collected for histological and morphometric analyses. There was no difference among groups or among times for the characteristics hemorrhage (p=0.4154), fibrinous adhesion formation (p=0.0712), and organization of collagen fibers (p=0.2583) and of the connective tissue (p=0.1046). For these groups, regardless of the time, eccentric exercise led to epitenon thickening (p=0.0204), which was lower in the group treated with laser therapy. Histological analysis revealed differences (p=0.0032) in the number of inflammatory cells over time. They were more numerous in the group that only exercised. This result was confirmed by morphometric analysis, which showed a significant interaction (groups x time) for this characteristic. Eccentric exercise increased (p=0.0014) the inflammatory infiltrate over time (3 and 21 days). However, association with laser therapy reduced inflammatory reaction. On the other hand, the combination of the treatments increased angiogenesis in morphometric (p=0.0000) and histological (p=0.0006) analyses compared with the other groups, while the isolated application of low-level laser reduced this characteristic over time. Animals maintained at rest presented the lowest amount (p=0.0000) of fibroblasts, according to the morphometric analysis. However, histological evaluation showed a significant group x time interaction (p=0.0024). Greater amounts of fibroblasts were observed in groups E, L and LE on the 7th, 14th and 21st days, respectively. The animals that received laser therapy and were exercised showed a greater (p=0.0000) amount of collagen fibers over time. Laser therapy at a dose of 3J and at a wavelength of 904nm, starting 24h after surgical induction of tendinopathy in Wistar rats, is suitable for angiogenesis and prevention of tendon thickening, which can be associated with the intensity of inflammatory process. When associated with eccentric exercise, the therapy has the advantage of increasing the amount of collagen fibers, reducing fibrinous adhesions and inflammatory infiltrate, despite prolonging angiogenesis. Therefore, eccentric exercise performed concomitantly with laser therapy improves the histological properties of the injured tendon.

E-mail: msouza@ufv.br O laser de baixa potência é recomendado para o tratamento das tendinopatias apesar dos resultados contraditórios no que se refere a dose de energia ideal, comprimento de onda e tempo de aplicação. O objetivo desse estudo foi avaliar os efeitos da laserterapia e do exercício excêntrico na tendinopatia do tendão calcanear comum de ratos Wistar. Foram utilizados 48 ratos machos adultos. Os animais foram distribuídos aleatoriamente em quatro grupos (L=laser; E=exercício excêntrico; LE=laser e exercício excêntrico, e R=repouso). Após 24h da indução de tendinopatia unilateral, foi iniciada laserterapia (904nm/3J/cm²) e/ou exercício excêntrico (caminhada em declive; esteira com 15o de inclinação; 12m/min; 50min/dia) que permaneceu por 20 dias. Aos 3, 7, 14 e 21 dias após indução da lesão, três ratos de cada grupo foram submetidos à eutanásia, e os tendões foram obtidos para análises histológica e morfométrica. Não houve diferença entre grupos nem entre tempos nas características hemorragia (p=0,4154), formação de aderência fibrinosa (p=0,0712) e organização das fibras colágenas (p=0,2583) e do tecido conjuntivo (p=0,1046). Considerando os grupos, independentemente do tempo, foi observado que o exercício excêntrico ocasionou espessamento do epitendão (p=0,0204), que foi menor no grupo submetido à laserterapia. A análise histológica revelou diferença (p=0,0032) na quantidade de células inflamatórias ao longo do tempo, sendo observado em maior quantidade no grupo apenas exercitado. Esse resultado foi confirmado pela análise morfométrica, que demonstrou haver interação significativa (grupos x tempo) nessa característica, na qual o exercício excêntrico aumentou (p=0,0014) o infiltrado inflamatório ao longo do tempo (3 e 21 dias), porém, quando associado à laserterapia, ocorreu redução da reação inflamatória. Por outro lado, a associação dos tratamentos ocasionou maior angiogênese observada nas análises morfométrica (p=0,0000) e histológica (p=0,0006), quando comparada com os demais grupos, enquanto a aplicação isolada do laser de baixa potência reduziu essa característica ao longo do tempo. Os animais mantidos em repouso foram os que apresentaram menor (p=0,0000) quantidade de fibroblastos, na análise morfométrica, porém na avaliação histológica foi observada interação significativa entre grupo e tempo (p=0,0024). Maior quantidade de fibroblastos foi observada nos grupos E, L e LE, no 7o, 14o e 21o dias, respectivamente. Os animais que receberam laserterapia e foram exercitados apresentaram maior (p=0,0000) quantidade de fibras colágenas ao longo do tempo. A laserterapia na dose de 3J e comprimento de onda de 904nm, iniciada 24h após indução cirúrgica de tendinopatia em ratos Wistar é adequada para angiogênese e para evitar o espessamento do tendão, que pode estar relacionado com a intensidade do processo inflamatório. Quando associada ao exercício excêntrico, a terapia apresenta a vantagem de aumentar a quantidade de fibras colágenas, reduzir a aderência fibrinosa e infiltrado inflamatório, apesar de prolongar a angiogênese. Portanto, o exercício excêntrico realizado concomitantemente com a laserterapia melhora as propriedades histológicas do tendão lesionado.

Its presence in the placental uterine environment is related to immunological tolerance to the semi-allograft because it prevents proliferation of maternal immune cells, either by the lack of this amino acid or by the action of its catabolites, such as the quinolinic acid, which is particularly toxic for T lymphocytes. Little is known regarding the influence of hormones and cytokines on the expression of IDO in the maternal fetal interface. Therefore, the hypothesis that some hormones and interleukins present in uteroplacental region could have an effect on the expression of IDO on cultured cells was tested. Cells derived from the fetal maternal interface from Wistar rats were kept in culture and supplemented with estradiol and interferon-&#947;. Expression of the enzyme was assessed by flow cytometry at periods of 4, 24 and 48 hours and confirmation of the presence of protein by immunohistochemistry. The results showed an increasing of IDO expression after the addition of estrogen (9.03±0.81 to 11.25±0.25) and interferon-&#947; (9.03±0.81 to 20.43±0.60). The effect of interferon-&#947; was expected as reported in the literature, however, elevated IDO expression by estrogen represents new information on possible mechanisms involved in the enzyme activation. These findings could provide a better understanding of IDO contribution on maternal-fetal tolerance and may collaborate to future therapeutic modulation of this enzyme

Sua presença no ambiente uterino placentário está relacionada à tolerância imunológica ao semi-aloenxerto, pois impede a proliferação de células imunológicas maternas, seja pela falta do aminoácido, ou pela ação de alguns catabólitos oriundos da quebra do triptofano, como o ácido quinolínico, que é tóxico principalmente para os linfócitos T. Pouco se conhece sob a influência de substâncias (hormônios e citocinas) presentes na interface materno fetal e a expressão dessa enzima. Por esta razão, formulou-se a hipótese de que hormônios e interleucinas presentes na região uteroplacentária poderiam exercer algum efeito na expressão da IDO. Células oriundas da interface materno fetal de ratas Wistar foram mantidas em cultivo, onde receberam suplementação com estradiol e interferon-&#947;. A expressão da enzima foi avaliada pela técnica de citometria de fluxo nos períodos de 4, 24 e 48 horas e confirmação da presença proteica por imuno-histoquímica. Os resultados mostraram um aumento na expressão de IDO após a adição de estrógeno (9,03±0,81/11,25±0,25) e interferon-&#947; (9,03±0,81/20,43±0,60). O efeito do interferon-&#947; já era esperado como relatado na literatura, contudo, a elevação da expressão da IDO pela adição do estrógeno constitui nova informação sobre possíveis mecanismos envolvidos na ativação da enzima. O melhor esclarecimento desses achados poderia contribuir para uma melhor compreensão da participação dessa enzima na tolerância materno-fetal e para uma futura modulação terapêutica da mesma.


#18 - Teratogenic effects of Prosopis juliflora in rats and analysis of the toxicity of the pods, 34(11):1089-1093

E-mail: rmtmed@uol.com.br The objective of this study was to determine the possible teratogenic effects of Prosopis juliflora, check if there is a loss in toxicity between pods stored and newly collected and determine whether there are differences in toxicity between the pods collected in different localities. Thirty pregnant female Wistar rats were randomly separated into five groups: a control (G1) and four experimental (G2, G3, G4 and G5), each with six animals. The animals in groups G2 and G3 were fed diets containing 70% of pods of P. juliflora newly collected in two different municipalities. The groups G4 and G5 were fed beans prepared with the same origins, but stored for a period of 6 months. The control group received the same diet without pods of P. juliflora. In the control group the number of defects per litter (1.16 ± 0.98) was significantly lower than the experimental groups (14.00 ± 2.96, 6.16 ± 2.22, 7.66 ± 2.94 and 4.66 ± 1.63 for G2, G3, G4 and G5, respectively) indicating that the plant is teratogenic. No significant differences were observed in the frequency of malformations and number of fetuses born between groups receiving pods from different locations. However, the number of defects in the groups who received the freshly harvested pods was significantly different from the number observed in rats that received the beans after storage, suggesting that the teratogenic effect of the plant decreases during storage. We conclude that the pods of P. juliflora are teratogenic for Wistar rats and that the teratogenicity decreases with storage.

Trinta ratas prenhes da linhagem Wistar foram separadas, aleatoriamente, em cinco grupos: um controle (G1) e quatro experimentais (G2, G3, G4 e G5). Os animais dos grupos G2 e G3 foram alimentados com ração contendo 70% de vagens de P. juliflora recém-coletadas em dois municípios diferentes. Os grupos G4 e G5 foram alimentados com ração preparada com vagens das mesmas procedências, porém armazenadas por um período de 6 meses. O grupo controle recebeu ração sem vagens de P. juliflora. No grupo controle o número de malformações por ninhada (1,16 ± 0,98) foi significativamente menor do que os dos grupos experimentais (14,00 ± 2,96, 6,16 ± 2,22, 7,66 ± 2,94 e 4,66 ± 1,63 para os grupos G2, G3, G4 e G5, respectivamente) indicando que a planta é teratogênica. Não foram observadas diferenças significativas na frequência de malformações e no número de fetos nascidos entre os grupos que receberam vagens de diferentes localidades. No entanto, o número de malformações nos grupos que receberam as vagens recém-colhidas (65,80 ± 21,20 a, 67,59 ± 18,10 a), foi significativamente maior que o observado nas ratas que receberam as vagens após o armazenamento (35,74±10,10b, 21,85±5,13c) sugerindo que o efeito teratogênico da planta diminui durante o armazenamento. Conclui-se que as vagens de P. juliflora são teratogênicas para ratas Wistar e que a fetoxicidade das mesmas diminui com o armazenamento.

E-mail: valeria@dmfa.ufrpe.br The dexamethasone, a synthetic glucocorticoid, has the ability to cross the placenta by increasing the level of movement of corticosteroids from mother to fetus during pregnancy. When administered in the late stages of pregnancy can produce effects undesirable on placental formation. The present study aimed to investigate the effect of administration of dexamethasone (0.8mg/day/animal) in the first five days of pregnancy, on placental development in rats. We used 30 albino rats, divided into two groups: Group I - pregnant rats without the application of dexamethasone, sacrificed to the 7th and 14th day. Group II - rats subjected to the application of dexamethasone in the first five days of pregnancy, sacrificed to the 7th and 14th day. The results showed that dexamethasone did not affect the number and histology of the implantation sites, but promoted changes in the disk placental causing hypertrophy in trophoblastic giant cell layer. No changes were found in the content of collagen, but there was interference with the metabolism of glycogen in spongiotrophoblast. The morphometry of lines showed, a difference between groups in the region of labyrinth and trophoblast giant cell. However, in morphometry of points there was a difference between groups in the region of labyrinth.

Os resultados mostraram que a dexametasona não afetou o número e a histologia dos sítios de implantação, porém, promoveu alteração no disco placentário ocasionando hipertrofia na camada de células trofoblásticas gigantes. Não foram evidenciadas alterações no teor de colágeno, porém houve interferência no metabolismo do glicogênio no espongiotrofoblasto trofospongio. Na morfometria de linhas houve diferença entre os grupos na região de labirinto e células trofoblásticas gigantes, porém a morfometria de pontos só ratificou as alterações percebidas na região do labirinto.

E-mail: hill@vm.uff.br The increase of heart collagen fibers in diabetics is a well known fact, but the consequences are not defined. The aim was to quantify the cardiac collagen fibers in normal and diabetic rats treated with vitamin C. We selected 32 Wistar rats, 16 diabetic animals induced endovenously with streptozootocin, and 16 healthy animals, half of them, diabetics and normals, were treated with vitamin C for 90 days. After the experimental proceeding, the hearts were removed and processed accordingly to conventional protocol for optical microscopy and specific staining for collagen. The results showed that the diabetic rats presented increase in the number of cardiac collagen fibers, but the ones treated with vitamin C showed little accumulation of fibers. It could be concluded that treatment with vitamin C is important for the prevention of heart failure in diabetic animals.

E-mail: hill@vm.uff.br O aumento do conteúdo de fibras colágenas no coração de diabéticos é um fato bastante conhecido, suas conseqüências ainda são objeto de estudo e causam certa controvérsia, portanto este trabalho objetivou estudar a variação na quantidade das fibras de colágeno cardíacas em animais normais e diabéticos tratados pela vitamina C. Para isso foram selecionados 32 ratos Wistar, 16 diabéticos induzidos pela injeção endovenosa de estreptozootocina e 16 normais, sendo metade deles tratados com Vitamina C (diabéticos e normais) por um período de 90 dias. Após período experimental, os corações foram retirados e processados segundo protocolo convencional para microscopia óptica e coloração específica para colágeno. Os resultados mostram que animais diabéticos apresentam maior quantidade de fibras de colágeno cardíacas e que o tratamento com a vitamina C determinou um menor acúmulo na quantidade dessas fibras.
