E-mail: lirigatto@yahoo.com.br From the Tropic of Capricorn to Equator, the seasonality of domestic cat is known to be absent, i.e., these animals are considered non-seasonal breeders at these regions. We hypothesized that this particularity might have some influence on in vitro embryo production. The aim of this experiment was to determine the percentage of cleavage and morulae and blastocyst formation produced from oocytes recovered from queen ovaries of three distinct status - follicular, luteal or inactive - during two different reproductive seasons experienced by cats in southeast of Brazil (22°53’09” S and 48°26’42” W) – non breeding season (NBS), comprehending January to March; and breeding season (BS), August to October. Thirty queens were neutered. Ovaries were classified according to their status and were sliced in PBS for cumulus oocyte complex (COC) releasing. Grade I COC were washed three times in H-MEM supplemented with BSA, glutamine, sodium pyruvate, cysteine, streptomycin and penicillin. Oocytes were incubated in groups of 20-30 in 400µL of DMEM supplemented with FSH, LH, estradiol, IGF-I and basic fibroblast growth factor under mineral oil for 30 or 36 hours at 38°C in humidified environment of 5% de O2, 5% CO2 and 90% N2. COC were fertilized in Ham’s F-10 medium supplemented with BSA, cysteine, pyruvate and streptomycin/penicillin (culture medium) with fresh semen selected through swim up technique. Eighteen hours later, the presumptive zygotes were denuded, the percentage of cleavage was determined and every 10 zygotes were transferred to 100mL drops of culture medium for culture during three days. After 72 hours of culture the percentage of morulae formation was evaluated and these structures were transferred to drops of the same culture medium. At the eighth day of culture blastocyst formation was analyzed. During NBS, from a total of 272 (inactive), 162 (luteal) and 134 (follicular) fertilized oocytes, the percentage of cleaved zygotes, morulae and blastocysts derived from inactive ovaries were 24.63, 16.54 and 8.09 respectively; for those derived from luteal ovaries, the percentage was 21.6, 12.96 and 8.64, and for those from follicular ovaries, they were 24.62, 16.41 and 8.21. Considering BS, from a total of 102 (inactive), 198 (luteal) and 86 (follicular) fertilized oocytes, the relative frequency (%) of cleaved zygotes, morulae and blastocysts derived from inactive ovaries were 64.7, 41.17 and 23.53 respectively; for those derived from luteal ovaries, the percentage was 64.14, 40.41 and 23.73, and for those from follicular ovaries, they were 63.95, 39.54 and 24.41. The results of this experiment demonstrate that no statistically significant difference (P<0.05) was verified in the frequency of cleaved embryos and morulae and blastocyst formation when comparing the three ovarian conditions in the same season. However the breeding season presented better results considering cleavage and morulae and blastocyst formation.

Nós hipotetizamos que esta particularidade possa ter alguma influência sobre a produção embrionária in vitro. O objetivo deste experimento foi determinar a porcentagem de clivagem e formação de mórulas e blastocistos produzidos a partir de oócitos recuperados de ovários de gatas em três condições - folicular, lútea ou inativa – durante duas estações reprodutivas pelas quais gatas passam na região sudeste do Brasil (22°53’09” S e 48°26’42” O) - estação não reprodutiva (ENR), que compreende os meses de janeiro a março; e estação reprodutiva (ER), agosto à outubro. Trinta gatas foram castradas. Os ovários foram classificados de acordo com sua condição e foram fatiados em PBS para liberação dos complexos cumulus oophorus (COC). COC grau I foram lavados três vezes em H-MEM suplementado com BSA, glutamina, piruvato sódico, cisteína, estreptomicina e penicilina. Os oócitos foram incubados em grupos de 20-30 em 400 µL de DMEM suplementado com FSH, LH, estradiol, IGF-I e fator de crescimento fibroblástico básico sob óleo mineral por 30 ou 36 horas em atmosfera úmida de 5% de O2, 5% CO2 e 90% N2 a 38°C. Os COC foram fertilizados em meio Ham’s F-10 suplementado com BSA, cisteína, piruvato e estreptomicina/penicilina (meio de cultura) com sêmen fresco selecionado através da técnica de swim up. Dezoito horas depois, os presumíveis zigotos foram denudados, a porcentagem de clivagem foi determinada e cada 10 zigotos foram transferidos para gotas de 100 mL de meio de cultura para cultivo durante 3 dias. Após 72 horas de cultivo, a porcentagem de formação de mórulas foi avaliada e estas estruturas foram transferidas para gotas do mesmo meio de cultivo. No oitavo dia de cultivo a formação de blastocisto foi avaliada. Durante a ENR, de um total de 272 (inativo), 162 (lútea) e 134 (folicular) oócitos fertilizados, a porcentagem de clivagem de zigotos, formação de mórulas e de blastocistos derivados de ovários inativos foi 24,63, 16,54 e 8,09 respectivamente; para aqueles oriundos de ovários na condição lútea, a porcentagem foi de 21,6, 12,96 e 8,64, e para aqueles provenientes de ovários na fase folicular, foi de 24,62, 16,41 e 8,21. Considerando a ER, de um total de 102 (inativo), 198 (lútea) e 86 (folicular) oócitos fertilizados, a frequência relativa (%) de zigotos clivados, mórulas e blastocistos derivados de ovários na condição inativa foi de 64,7, 41,17 e 23,53 respectivamente; para aqueles oriundos de ovários na condição lútea, a porcentagem foi de 64,14, 40,41 e 23,73, e para aqueles provenientes de ovários na fase folicular, foi de 63,95, 39,54 e 24,41. Os resultados deste experimento demonstraram que não houve diferença estatística significante (P < 0.05) na frequência de embriões clivados e na formação de mórulas e blastocistos quando comparadas as três condições ovarianas dentro da mesma estação. Entretanto, a ER apresentou resultados melhores considerando as taxas de clivagem e formação de mórula e de blastocisto se comparada à ENR.

E-mail: angelicawouters@yahoo.com.br Senecio spp. poisoning is an important cause of illness and death of cattle in Rio Grande do Sul, Brazil, and is often manifested by neurologic clinical signs and histological brain changes. Histological evaluation was performed on liver and brain samples of ten cattle naturally poisoned by Senecio sp. Samples of cerebrum, brainstem and cerebellum were stained with periodic acid-Schiff (PAS), and immunohistochemistry was carried out, employing anti-fibrillary acidic protein (GFAP), anti-S100 protein and anti-vimentin antibodies. The histological finding in the brain included mild to severe vacuolation in the white matter and the junction of gray and white matter, characterized as spongy degeneration. Histochemical and immunohistochemical staining revealed no significant findings compared with the brains of eleven adult cattle without liver and/or brain changes used as controls.

Foi efetuada avaliação histológica em fígado e encéfalo de dez bovinos naturalmente intoxicados por Senecio sp. e foram realizadas as técnicas de histoquímica de ácido periódico de Schiff e de imuno-histoquímica, com emprego dos anticorpos anti-proteína fibrilar ácida (GFAP), anti-proteína S100 e anti-vimentina em amostras de telencéfalo, tronco encefálico e cerebelo. Na histologia do SNC observou-se vacuolização discreta a acentuada em substância branca e/ou junção entre substância branca e cinzenta, caracterizada como degeneração esponjosa. Na avaliação histoquímica e imuno-histoquímica não houve diferenças significativas em relação aos achados em encéfalo de onze bovinos controle, adultos, sem alterações hepáticas e/ou encefálicas, avaliados pelas mesmas técnicas.


#13 - Records of performance and sanitary status from a dairy cattle herd in southern Brazil, 31(1):1-9

[Records of performance and sanitary status from a dairy cattle herd in southern Brazil.] Registros de desempenho e de status sanitário de um rebanho bovino leiteiro no Sul do Brasil. Pesquisa Veterinária Brasileira 31(1):1-9. Setor de Patologia Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Av. Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@ufrgs.br Over the last decades, the emphasis on the health of dairy cows has changed from an individual to a herd level. In this scenario, the role played by the recording system and its interpretation by veterinarians has gained primordial importance. The records of productive and reproductive performance and of sanitary status from a southern Brazilian dairy cattle herd have been presented and discussed. The period of study was 2000-2009. Mean values per lactation period were 349D 8436M 290F 275P 201SCS (D: days in lactation, M: kg of milk yield, F: kg of fat, P: kg of protein and SCS: somatic cell score in 1000 cells/ml of milk). Major indexes of reproductive efficiency included age at first calving (31 months), services per conception (2.1), intercalving interval (428 days), calving to conception interval (146 days), mean annual rates of parturitions (76.2%), fetal losses (9.8-19.0%), and stillbirths (3.6%), apart of voluntary waiting period (94 days). Main information on sanitary status of the herd was associated with the mean prevalence of common disorders of dairy cattle such as anaplasmosis (29.8%), mastitis (27.8%), digital diseases (26.3%), ovarian cysts (21.3%), placental retention (19.7%), postpartum uterine infections (10.6%), and calf diarrhea (23.7%) and pneumonia (16.8%), among others. In addition, culling reasons (low reproductive performance [56.3%] and udder/mastitis problems [33.6%]), causes of cattle deaths (anaplasmosis [16.4%] and leukosis [11.4]), and the impact of cattle diseases such as tuberculosis, leukosis, and neosporosis on the herd have also been presented and succinctly discussed. Os registros de eficiência produtiva e reprodutiva, bem como os de ordem sanitária observados em um rebanho bovino leiteiro no Sul do Brasil foram apresentados e discutidos. O período estudado foi 2000-2009. Os dados de produção leiteira média incluíram, por período de lactação de 349 dias, 8436 kg de leite, 290 kg de gordura e 275 kg de proteína, com média de contagem de células somáticas de 201000 células/ml de leite. Idade ao primeiro parto (31 meses), serviços por concepção (2,1), intervalo entre partos (428 dias), intervalo parto-concepção (146 dias), taxas médias anuais de parição (76,2%), perdas fetais (9,8-19%) e natimortos (3,6%), além de período de espera voluntária (94 dias) foram alguns dos índices de desempenho reprodutivo registrados. Informações sanitárias do rebanho incluíram prevalência média de doenças comuns em bovinos leiteiros tais como anaplasmose (29,8%), mastites (27,8%), doenças digitais (26,3%), cistos ovarianos (21,3%), retenção de placenta (19,7%), infecções uterinas (10,6%), diarreia (23,7%) e pneumonia (16,8%) em terneiras, entre outras. Além disso, razões de descartes (baixo desempenho reprodutivo [56,3%], alterações de úbere [33,6%], etc.), causas de morte (anaplasmose [16,4%], leucose [11.4], etc.) e o impacto de doenças de bovinos como tuberculose, leucose e neosporose sobre o rebanho foram também apresentados e sucintamente discutidos. E-mail: gugatoscan@hotmail.com Neospora caninum and N. hughesi are protozoa which can infect horses and can cause reproductive and neurological diseases, respectively. The pathogenesis of neosporosis in horses is poorly understood, as well as the sources of horizontal infection of N. hughesi. Furthermore, there are doubts about the role of the vertical transmission of Neospora spp. in maintenance of these parasites in equine populations. In this study, we evaluated: (1) the occurrence of infections by Neospora spp. in a population of mares (in reproductive age) on a farm of Crioula breed horses; and (2) the possible association between the serological status of mares and of their offspring, aiming to investigate, indirectly, the relevance of transplacental transmission for the occurrence of Neospora spp. in these horses. We found a highly significant association between the serological status of mares and their offspring. Although had been exposed to the same environmental risk factors, the descendants of seropositive mares had a higher percentage of seropositivity against Neospora spp. compared to the descendants of seronegative mares. The association between kinship and serological status indicates an influence of vertical (transplacental) infection raising the occurrence of Neospora spp. in the studied equine population.

A patogenia da neosporose é pouco conhecida nos equinos, bem como as fontes de infecção horizontal de N. hughesi. Além disso, há dúvidas quanto ao papel da transmissão vertical de Neospora spp. na sua manutenção em populações equinas. Neste estudo avaliaram-se: (1) a ocorrência da infecção por Neospora spp. na população de éguas em idade reprodutiva em um haras de cavalos da raça Crioula; e (2) a possível associação entre o status sorológico destas éguas com o de suas crias, como meio de investigar, indiretamente, a relevância da transmissão transplacentária na ocorrência da infecção por Neospora spp. nestes animais. A associação entre o status sorológico das éguas e o de suas crias foi altamente significativa. Os animais descendentes de éguas soropositivas tiveram maior ocorrência de anticorpos anti-Neospora spp. do que os descendentes de éguas soronegativas, embora expostos aos mesmos fatores de risco ambientais. A associação entre parentesco em primeiro grau e status sorológico indica a influência da infecção vertical (transplacentária) na ocorrência de Neospora spp. na população equina estudada.

In the first outbreak 12 sheep were affected, 5 of them showed blindness, three sheep died (Sheep 1-3) and two were euthanized 6 months after the onset of clinical manifestation (Sheep 4 and 5). In the second outbreak 26 goats were affected, from which six survived despite blindness and one was euthanized. Clinically the animals showed depression, ataxia, motor incoordination, decreased or absent pupil reflexes. In some animals this clinical picture developed to bilateral blindness, with no reaction to threat and bilateral irresponsive midriasis. In the ophthalmic examination retinal vessel atrophy and hyperreflexia were observed. The histological examination showed myelin edema leading to status spongiosus in the central nervous system and compressive neuropathy of the optic nerve, associated with retinal degeneration and/or atrophy. This report aims to describe the epidemiologic, clinic and pathologic aspects of closantel overdosage in sheep and goats.

E-mail: franciscojavier.dieguez@usc.es This study was designed to assess the relationship between antibodies against bovine viral diarrhea virus (BVDV) determined in the bulk tank milk (BTM) and the within-herd seroprevalence. We also assessed the efficiency of measuring antibody levels in BTM samples to monitor BVDV infection status in a herd. In the 81 farms included in the study, BTM samples were obtained and blood samples withdrawn from all cattle older than one year. The infection status was then determined in serum and milk using a commercial blocking ELISA based on the detection of anti-p80 antibodies. Apart from these baseline serum and milk samples, another BTM sample was collected from each herd 9 months later, and a third BTM sample obtained 9 months after this. In these second and third milk samples, anti-BVDV antibodies were determined using the same ELISA kit. Statistical tests revealed good agreement between herd seroprevalences (% seropositive animals in the herd) and the antibody levels detected in the BTM samples. During the 18 months of follow-up, the farms with persistently infected cattle at the study outset (14.8% of the herds) showed a significant decrease in BTM antibody titers after virus clearance. Conversely, a significant increase in BTM antibody levels was observed in the herds infected with BVDV during the follow-up period. Our findings indicate that monitoring antibody levels in the BTM is a useful method of identifying changes in the BVDV infection status of a herd.

We also assessed the efficiency of measuring antibody levels in BTM samples to monitor BVDV infection status in a herd. In the 81 farms included in the study, BTM samples were obtained and blood samples withdrawn from all cattle older than one year. The infection status was then determined in serum and milk using a commercial blocking ELISA based on the detection of anti-p80 antibodies. Apart from these baseline serum and milk samples, another BTM sample was collected from each herd 9 months later, and a third BTM sample obtained 9 months after this. In these second and third milk samples, anti-BVDV antibodies were determined using the same ELISA kit. Statistical tests revealed good agreement between herd seroprevalences (% seropositive animals in the herd) and the antibody levels detected in the BTM samples. During the 18 months of follow-up, the farms with persistently infected cattle at the study outset (14.8% of the herds) showed a significant decrease in BTM antibody titers after virus clearance. Conversely, a significant increase in BTM antibody levels was observed in the herds infected with BVDV during the follow-up period. Our findings indicate that monitoring antibody levels in the BTM is a useful method of identifying changes in the BVDV infection status of a herd.

This paper reports a spongy degeneration in two Sindhi calves, a female and a male. Both calves were offspring from the same bull, and the herd had high degree of inbreeding. The female calf was born normal, but progressive nervous signs were observed since 2 months old. The male calf had progressive nervous signs since it was born. Both calves were euthanized with severe nervous signs when 4 months old. At necropsy the liver was pale in the male calf. Histological alterations were mild to accentuated vacuolization, mainly in the deep cerebral cortex, cerebellar white matter, grey matter of the spinal cord, internal capsule, and brain stem. Fatty degeneration was observed in the liver of both animals. On electron microscopy it was found that the status spongiosus was caused by astrocytic edema. It is suggested that the disease is caused by a metabolic hereditary error.

Status spongiosus was observed on the deeper layers of the cerebral cortex and subcortical white matter. Six male sheep were divided in three groups of two sheep each. Group 1 (Sheep 1 and 2) received daily doses of 6 g/kg/bw of the dry plant, during 30 days; Group 2 (Sheep 3 and 4) received daily doses of 3 g/kg/bw, during 60 days; and Group 3 was the control group. Sheep 1 was euthanatized 30 days after the start of the ingestion. Only cardiac arrhythmia was observed clinically, and no lesions were observed at necropsy. Total amounts (g/kg/body weight) o(;the plant consumed by each animal were 75 during 60 days (Sheep 6), 125 during-25 days (Sheep 4), 13,0 during 13 days (Sheep 7), 150 during 60 days (Sheep 5), 180 during 18 days (Sheep 3), 330 during 33 days (Sheep 2), and 375 during 25 days (Sheep 1 ). Six sheep were fed the green leaves while Sheep 7 was fed dried A. glazioviana leaves. Five sheep (Sheep 1-4 and 7) developed clinical signs 8 to 16, days after the first administration of the plant. The clinical course varied from 6 to 50 days and clinical signs included apathy, loss of appetite, and increased heart and respiratory rates; affected sheep were ·reluctant to move and showed a low carriage of the head, instability of the hind limbs, and staggering gait. Some sheep supported their weight against the wall of the stable. Sheep 1, 3, and 4 died spontaneously; paddling movements preceded death. Sheep 2 and 7 were euthanatized when presenting marked clinical signs of the toxicosis. Sheep 5 and 6 did not get clinically ill and were euthanatized along with the control sheep 103 days after the first administration of the plant. All sheep were necropsied and several of their organs, including heart and brain, were evaluated histologically. Additionally, fragments of the myocardium of Sheep 3 and 4 were evaluated by transmission electron microscopy. To provide more controls for the myocardial changes, the hearts were collected from six animals at an abattoir known to slaughter sheep from regions free of A. glazioviana. These hearts were processed for histology and examined in the same way as the other eight. All seven treated (plant fed) sheep had gross changes at necropsy. There were firm, irregular white or yellow areas in the myocardium of all treated sheep. These pale areas were more evident on the cut surface of the heart. Hydrothorax and/or hydropericardium were observed in five animals (Sheep 1-4 and 7). Nutmeg livers were seen in Sheep 1 and 7. This latter sheep also had ascites. The main histopathological findings in treated sheep were degenerative/necrotic changes in the myocardium. The early changes consisted of acute swollen myocardial fibers associated with bizarre nuclei. Apparently this change progressed until the sarcoplasm disappeared and an empty collapsed sarcolemmal tube remained surrounded by interstitial connective tissue and fibrosis. Overt necrosis (hyaline and floccular) associated with mono nuclear infiltration and phagocytosis of fiber fragments were also observed. Varying degrees of spongy degeneration were seen in the white matter of the brain of Sheep 1, 3, 4 and 7. Mitochondrial swelling and loss of density of the mitochondrial matrix were the earliest ultrastructural changes observed. In advanced lesions cardiac cells had vacuoles in the sarcoplasm, rupture of the sarcolemma and necrosis with intervening macrophages and fibroblasts associated with increased collagen.

Seis ovinos recéberam folhas verdes e o Ovino 7 recebeu folhas dessecadas. Cinco ovinos (Ovinos 1-4 e 7) desenvolveram sinais clínicos. 8 a 16 dias após a primeira administração da planta. A evolução do quadro clínico foi de 6 a 50 dias e os sinais clínicos. incluíram apatia, perda do apetite, taquicardia e taquipnéia. Os ovinos afetados apresentavam relutância em mover-se, cabeça baixa, instabilidade dos membros pélvicos e andar cambaleante. Alguns dos ovinos afetados apoiavam o corpo contra a parede da baia. Os Ovinos 1, 3 e 4 morreram espontaneamente; movimentos de pedalagem precederam a morte. Os Ovinos 2 e 7 foram submetidos à eutanásia com sinais clínicos avançados da toxicose. Os Ovinos 5 e 6 não adoeceram e foram sacrificados junto com o controle 103 dias após a primeira administração da planta. Todos os ovinos foram necropsiados e vários órgãos, incluindo coração e encéfalo, foram avaliados histologicamente. Além disso, fragmentos de miocárdio dos Ovinos 3 e 4 foram avaliados por microscopia eletrônica de transmissão. A fim de se obter mais controles para a avaliação das lesões do miocárdio, coletaram-se os corações de seis ovinos abatidos para consumo em um abatedouro que sabidamente recebe ovinos originários de uma região livre de A. glazioviana. Esses corações foram processados para histologia e examinados da mesma maneira que os outros oito. Todos os ovinos tratados (que receberam a planta) apresentaram lesões macroscópicas. Havia áreas firmes, irregulares, branco-amareladas no miocárdio de todos os ovinos tratados. Essas áreas pálidas eram mais bem vistas na superfície de corte do coração. Hidrotórax e/ou hidropericárdio foram observados em cinco animais (Ovinos 1-4 e 7). O fígado dos Ovinos 1 e 7 tinha aspecto de nozmoscada. O Ovino 7 apresentava ascite. Os principais achados histopatológicos nos ovinos tratados foram alterações degenerativas/necróticas no miocárdio. As alterações iniciais consistiam de tumefação aguda das fibras cardíacas, associada a núcleos com formas bizarras. Essa alteração aparentemente progredia até o desaparecimento do sarcoplasma, resultando num tubo sarcolemal vazio e colapsado envolto por tecido conjuntivo intersticial e fibrose. Necrose franca (hialina e flocular) associada à infiltração mononuclear e fagocitose de fragmentos de fibras foi também observada. Graus variáveis de degeneração esponjosa foram observados na substância branca do encéfalo dos Ovinos 1, 3, 4 e 7. Tumefação de mitocôndrias e perda da densidade da matriz das mitocôndrias foram as alterações ultra-estruturais mais precoces. Em lesões avançadas os cardiomiócitos apresentavam vacúolos no sarcoplasma, ruptura do sarcolema e necrose com aparecimento de macrófagos e fibroblastos associados a aumento de colágeno.
