E-mail: jpatarro@ufv.br Exclusive chemicals dependence for the control of Rhipicephalus (Boophilus) microplus has become one of the largest scientific and economical concerns in recent years, and as a result, research to vaccine development are being undertaken. The objective of this study was evaluating the lymph nodes response of cattle immunized at field with the rSBm7462 anti-R. (B.) microplus peptide. Fourteen crossbred cattle (Bos taurus x Bos indicus), aged 4-10 months, were used. The animals were maintained on two farms in the north of Minas Gerais state and received three immunizations with the peptide rSBm7462 applied subcutaneously at 30-day intervals. Pre-scapular lymph nodes were collected surgically 15 days after each immunization and fixed in formalin for 18 hours, then, they were embedded in Paraplast subsequently and the samples were stained with Hematoxylin-Eosin (HE) technique for cellular events observation. On the other hand, in order to antigens identifying in immunized animals lymph nodes, the immunohistochemistry (IHC) with peroxidase-anti peroxidase (PAP) method was performed. Lymph node response of cattle inoculated was evaluated by analysis of germinal centers (GC) formation, medullary cords hyperplasia (MC) and antigen rSBm7462 presence in PAP+ cells. This study shows that the recombinant peptide rSBm7462 induces a T-dependent adaptive immune response characterized on secondary lymphoid tissues by structure formation for affinity and immunological memory.

E-mail: jpatarro@ufv.br A dependência exclusiva de compostos químicos para o controle de Rhipicephalus (Boophilus) microplus tornou-se uma das maiores preocupações científicas e econômicas dos últimos anos, e como consequência, estão sendo realizadas pesquisas para o desenvolvimento de vacinas. O objetivo deste trabalho foi avaliar a resposta de linfonodos de bovinos imunizados a campo com o peptídeo rSBm7462 anti R. (B.) microplus. Foram utilizados 14 bovinos mestiços (Bos taurus x Bos indicus), com idades entre 4-10 meses, mantidos em duas propriedades rurais do norte do estado de Minas Gerais. Os animais receberam três imunizações do peptídeo rSBm7462, aplicados por via subcutânea, com intervalo de 30 dias. Após 15 dias de cada imunização, os linfonodos pré-escapulares foram coletados e fixados por 18 horas em formol. Posteriormente, foram incluídos em Paraplast e as amostras foram coradas pela técnica hematoxilina-eosina (HE) para a observação de eventos celulares. Para a identificação do antígeno nos linfonodos dos animais imunizados, foi realizada a técnica de imuno-histoquímica (IHQ) com o método peroxidase-anti-peroxidase (PAP). A resposta de linfonodos dos bovinos inoculados foi avaliada pelas análises de formação de centros germinais (CG), hiperplasia de cordões medulares (CM) e a presença do antígeno rSBm7462 em células PAP+, demonstrando que o peptídeo recombinante rSBm7462 induz uma resposta imune adaptativa T-dependente, caracterizada nos tecidos linfóides secundários pela formação de estruturas que conferem afinidade e memória imunológica.

The Colombian Middle Magdalena is an enzootic region for babesiosis, but there is no previous research providing detail on its transmission cycle. This study aims to assess some Babesia sp. infection indicators in cattle and ticks from the area, by using direct microscopic and molecular techniques to detect the infection. In the cattle, 59.9% and 3.4 % positivity values for B. bigemina and mixed infection (B. bovis + B. bigemina) were found respectively. In ticks, the positivity of B. bigemina reached 79.2% and 9.4% for the mixed infection. The degree of infestation in the region was 3.2 ticks per bovine. There was positive correlation between tick control acaricide frequencies and infestation in bovines. This leads us to infer that control periodicity greater than 90 days, in stable zones, is an abiotic factor that benefits the acquisition of protective immunity in calves, the natural control of the infection and eventual disease absence. It is necessary to monitor the disease by applying new entomological and parasitological indicators showing the complexity of this phenomenon.

E-mail: raquel.rech@embrapa.br The interpretation of the changes observed at necropsy is an important step for the success of the final diagnosis. This article aims to describe and illustrate the anatomical structures, non-lesions, artifacts, lesions without clinical significance and postmortem changes observed in domestic and wild pigs. Moreover, the article also recommends collection techniques of organs or structures for the diagnosis of diseases affecting this species. The main anatomical structures and non-lesions described are marginal lingual papillae, pars oesophagea of the stomach, torus pyloricus, and well demarcated lobular pattern of the liver (gastrointestinal system); tonsils of the soft palate, gastric lymphoid tissue, Peyer’s patches of the small intestine, marginal folds of the spleen (hematopoietic system); prominent mediastinum testis and placental areolae (reproductive system); pulmonary atelectasis and periople (fetus); and carpal glands (integumentary system). The discussed artifacts related to euthanasia are petechiae on the surface of the lung and kidney, false anemia due to exsanguination, subdural hemorrhage due to cerebral concussion, pseudoinfarcts of the spleen, and cerebriform appearance of the small intestine. Lesions without clinical significance described are renal cysts, lymph nodes with iron pigment, papillomas and hemangiomas on the scrotum, osseous metaplasia in the mesentery, and hyperemia of the gastric mucosa. Commonly found postmortem changes are livor mortis, pale muscles, pseudomelanosis, and serosanguinous fluid in the thoracic and abdominal cavities of fetuses.

E-mail: camilafb@usp.br Caprine arthritis encephalitis (CAE) and caseous lymphadenitis (CL) have high incidence and transmissibility in small ruminants. Since both virus have tropism for macrophages and monocytes and affect the innate immune response, it is believed that CAE can predispose the animal to infection by Corynebacteruim pseudotuberculosis, the etiological agent of CL. To confirm this hypothesis, we evaluated phagocytosis from the monocyte-macrophage cells from 30 Saanen goats. Goats were uniformly divided in two groups according to results of agar gel immunodiffusion test for CAE virus (CAEV). Peripheral blood mononuclear cells were isolated by density gradient centrifugation and the monocyte-macrophage cells were isolated from the mononuclear cells by their adhesion properties in plaques. Afterwards, phagocytosis of C. psudotuberculosis was performed for two hours at 37°C, 5% of CO2, and assessed by microscopic visualization. There was no difference in the percentage of monocyte-macrophage cells that phagocytozed C. bovis between groups (P=0.41). However, when phagocytosis rates were classified according to the number of C. pseudotuberculosis phagocyted, the percentage of monocyte-macrophage cells that internalized more than 12 bacteria were higher in serologically CAEV positive animals compared to the serologically negative ones (P<0.001). Furthermore, a positive and significant correlation (r = 0.488; P = 0.006) between the percentage of monocyte-macrophage cells that internalized more than 12 bacteria and the percentage of monocyte that were carrying out phagocytosis was also encountered in serologically CAEV positive goats, however the same were not observed in serologically negative ones. These results demonstrated an alteration in the intensity of C. pseudotuberculosis phagocytosis by monocytes-macrophages from goats infected by CAEV. Thus, these results indicated that goats infected with CAEV may be more susceptible to CL.

E-mail: camilafb@usp.br A Artrite Encefalite Caprina (AEC) e a Linfadenite Caseosa (LC) possuem alta incidência e transmissibilidade em pequenos ruminantes. Como ambas possuem tropismo por monócitos-macrófagos e afetam mecanismos da resposta inata do hospedeiro, acredita-se que a AEC predispõe o animal a infecções por Corynebacteruim pseudotuberculosis, agente etiológico da LC. Para confirmar esta hipótese, avaliou-se a fagocitose de células da série monócito-macrófago de cabras naturalmente infectadas pelo vírus da AEC (VAEC). Para tanto, foram utilizadas 30 cabras da raça Saanen, alocadas em dois grupos distintos, com 15 animais cada, conforme a sororreatividade de anticorpos séricos antivírus da AEC. Células mononucleares de sangue periférico foram isoladas por gradiente de densidade e plaqueadas para isolamento de células da série monócito-macrófago. Posteriormente, o ensaio de fagocitose de C. pseudotuberculosis foi realizado, após incubação por duas horas a 37°C a 5% de CO2, e a visualização da fagocitose foi identificada por microscopia óptica. O presente estudo não encontrou diferença na porcentagem de monócito-macrófagos que realizaram fagocitose entre os diferentes grupos (P = 0,41). Todavia, a análise quantitativa de bactérias fagocitadas, demonstrou maior capacidade fagocítica pelos macrófagos-monócitos do grupo sororreagente ao vírus da AEC. Correlação entre monócitos fagocitando e macrófagos que fagocitaram mais de 12 bactérias foi observado neste grupo (r = 0,488; P = 0,006), não sendo o mesmo encontrado no grupo de animais sorroreagentes negativos. Os dados demonstram aumento na intensidade da fagocitose de macrófagos de animais infectados com o vírus da AEC.

E-mail: dominguesln@gmail.com The effective control of Haematobia irritans (horn fly) and Rhipicephalus (Boophilus) microplus (cattle tick) is crucial for increasing the productivity of livestock industry in Brazil, but still remains a challenge. A greater knowledge about parasite control in the field is necessary to establish control strategies closer to farmer’s reality and easier to be performed. This study characterized the practices adopted for controlling such ectoparasites on 23 cattle ranches from the Triângulo Mineiro and Alto Paranaíba region in the state of Minas Gerais, through interviews. Most interviewees recognized the importance of these parasites to livestock, however, many problems related to parasite control were observed. Control of horn flies and cattle tick was a routine in 17.4% and 95.7% of the ranches, respectively, triggered by high infestations on animals. More than six insecticide/acaricide treatments were applied yearly in all cattle ranches controlling horn flies and in 76.5% of the ranches controlling the cattle tick. Pesticide products were applied mostly by manual backpack sprayers (63.6%) without restraining the animals in 45.5% of ranches. Product dilution following manufacturer’s recommendations was performed by 45.5% of interviewees, but 63.6% of them used lower doses per animal than technically recommended. Associations of organophosphates and pyrethroids were the main products used to control both parasites. Although pesticide use was a routine practice, the use of personal protective equipment (PPE) was not common among surveyed people. Most interviewees were able to recognize epidemiological characteristics of the parasites; however, parasite control measures adopted at most cattle ranches tend to jeopardize the efficacy of products, susceptibility of parasites and sustainability of control programs.

Um maior conhecimento das práticas adotadas no combate destes parasitos faz-se necessário para que se possam estruturar estratégias de controle mais próximas da realidade do produtor rural e mais fáceis de serem executadas. Este estudo caracterizou, através de entrevistas, as práticas adotadas no controle desses ectoparasitos em 23 propriedades da mesorregião do Triângulo Mineiro e Alto Paranaíba, Minas Gerais. A maioria dos entrevistados reconheceu a importância desses parasitos para a atividade pecuária, entretanto, diversos problemas relativos ao controle parasitário foram observados. O controle da mosca-dos-chifres e do carrapato-do-boi era uma prática rotineira em, respectivamente, 17,4% e 95,7% das propriedades, geralmente realizado em função de elevado grau de infestação dos animais. Mais de seis aplicações de ectoparasiticidas eram realizadas por ano em todas as propriedades que empregavam tratamentos para o controle da mosca e em 76,5% daquelas que combatiam o carrapato. Os produtos eram aplicados principalmente com bombas costais manuais (63,6%) e sem a contenção dos animais em 45,5% das propriedades. A diluição dos produtos segundo recomendações dos fabricantes era realizada por 45,4% dos entrevistados, porém, 63,6% aplicavam um volume de solução por animal menor do que o tecnicamente recomendado. Utilizava-se principalmente a associação de piretróides e organofosforados para o controle de ambos os parasitos. Apesar de utilizar rotineiramente ectoparasiticidas, o uso de equipamento de proteção individual (EPI) não era comum entre os entrevistados. A maioria dos entrevistados conhecia algumas características epidemiológicas dos parasitos, entretanto, o controle parasitário adotado na maior parte das propriedades tende a comprometer não apenas a eficácia dos tratamentos, mas a suscetibilidade dos parasitos e a sustentabilidade do controle.

No significant difference was found in cell viability, the phagocytosis and spreading rates by mononuclear phagocytes between the distinct groups. Therefore, the H2O2 release by leukocytes was higher in the milk samples from healthy mammary glands compared to those infected with Streptococcus spp. or Corynebacterium spp. However, when the H2O2 release by phagocytes in 1mL of milk according to SCC mL-1 of each sample was estimated, it was found that milk samples from infected, infected with Staphylococcus spp. and bacteriological negative quarters with high SCC were higher than the healthy ones. It was also observed a positive correlation among SCC and cell viability or phagocytosis and spreading rates, and a negative correlation between H2O2 release and cell viability or SCC. In face of, it can be concluded that the SCC, as well as their function and the cell viability, are related to mammary gland health.

Ao estimar a concentração de H2O2 mL-1 leite observou-se que as amostras de quartos mamários positivos no exame bacteriológico, infectados por Staphylococcus spp. e negativos no exame bacteriológico com alta celularidade foram maiores que aquelas provenientes de quartos mamários sadios. Observou-se também correlação positiva entre a CCS e a viabilidade celular e os índices de fagocitose e espraiamento; e correlação negativa entre a liberação de H2O2 e a CCS e a viabilidade celular. Conclui-se que a CCS, assim como a sua viabilidade e função, são conceitos intimamente relacionados com a saúde da glândula mamária.

E-mail: gerlanemcosta@yahoo.com.br This research aimed to describe the macroscopic and microscopic liver of tambaqui, Colossoma macropomum, Teleost freshwater Family Characidae, of great economic interest for the Amazon basin. We used six juveniles aged between six month and one year, from the small holding Esteio, Alta Floresta/MT, that develops mainly fish farming. The body was photographed in situ, described macroscopically, and fragments were removed and processed by routine histological techniques through paraffin embedding and HE staining. The liver, located ventrally to the swim bladder and craniodorsally to the stomach, is brownish red and consisted of three lobes, the right lateral, the left lateral and the ventral lobe. Microscopically, the parenchyma consists of hepatocytes varying from irregular rounded hexagonal to round forms with a large and central nucleus, and arranged in linear strings limited by sinusoids and radiating to central veins, but with absence of liver lobules. The central veins are distributed throughout the parenchyma, while the portal space consists in most cases only of a hepatic vein and bile duct; elsewhere exist artery and duct. Formation of portal triads was not founde. Melano macrophages were frequently seen dispersed throughout the central parenchyma. The morphofunctional study of the digestive system of fishes of the Amazon basin is important to obtain knowledge about their weight gain, large scale production for human consumption and preservation of the species, and has also its importance for being used as bioindicators today.

Foram utilizados seis (6) exemplares jovens com idade entre seis meses e um ano, oriundos da Chácara Esteio, Alta Floresta, MT, que desenvolve principalmente a prática da piscicultura. O órgão foi fotodocumentado in situ e descrito macroscopicamente, em seguida procedeu-se a retirada de fragmentos deste, que foram processados pelas técnicas histológicas rotineiras para inclusão em parafina e coloração de HE. O fígado localizou-se ventral à bexiga natatória e craniodorsalmente ao estômago, apresentou coloração amarronzada a vermelho, constituído por três lobos hepáticos, o lobo lateral direito, o lobo lateral esquerdo e o lobo ventral. Microscopicamente, o parênquima era constituído por hepatócitos com formato que variou do arredondado irregular hexagonal ao redondo com núcleo grande e central, arranjados em cordões lineares limitados por sinusóides que irradiam para veias centrais, e com ausência de lóbulos hepáticos. As veias centrais estavam distribuídas pelo parênquima, enquanto que o espaço porta, na maioria das vezes, era constituído apenas por uma veia hepática e o ducto biliar, em outros locais foi observado, uma artéria e um ducto. Não foi observada a formação de tríades portais. Foram frequentemente observados melano macrófagos centrais dispersos pelo parênquima. O estudo morfofuncional do Aparelho Digestório de peixes da bacia Amazônica, se faz pertinente com vistas ao conhecimento do aproveitamento de ganho de peso e produção em alta escala para consumo humano e preservação da espécie, além da importância de estarem sendo utilizados como bioindicadores atualmente.

E-mail: julemelo@hotmail.com The effects of endotoxemia on the antioxidant activity in alveolar macrophages of Wistar rats were evaluated. Twenty-four male rats, 90-120 days of age, were separated into 2 groups: control and endotoxemic. To the endotoxemic animals was administered, intraperitoneally, a lipopolyssaccaride at dosage of 1mg/kg body weight. Twenty-four hours after this procedure, blood was collected for total and differential leukocytes counts. In addition, bronchoalveolar lavage was collected for total and differential leukocyte counting. From this lavage macrophages were isolated for the dosage of superoxide and superoxide dismutase. The endotoxemia increased the total leukocyte counts and the number of neutrophils in the peripheral blood and bronchoalveolar lavage of the rats. There was an increased superoxide production without changing the superoxide dismutase. Our findings indicate that endotoxemia induces lung inflammatory response. However, it does not alter the antioxidant activity in adult rats. This fact not only enhances host response against infectious agents, but might also contribute to the pathogenesis of pulmonary injury.

Foram utilizados 24 ratos machos, com idade entre 90 e 120 dias, os quais foram divididos em dois grupos: controle e endotoxêmico. O grupo endotoxêmico foi submetido à injeção intraperitonial de lipopolissacarídio na dose de 1mg/kg de peso corporal. Após 24 h, coletou-se sangue para contagem total e diferencial de leucócitos; lavado broncoalveolar para contagem total e diferencial dos leucócitos e, a partir de macrófagos isolados deste lavado, foram realizadas as dosagens de superóxido e superóxido dismutase. A endotoxemia aumentou a contagem total de leucócitos e o número de neutrófilos no sangue periférico, no lavado broncoalveolar, e aumentou a produção de superóxido sem modificar a produção da superóxido dismutase. Esses resultados sugerem que a endotoxemia induz a uma resposta inflamatória no pulmão. Contudo, não altera a atividade antioxidante em ratos adultos. Tal fato potencializa a resposta contra agentes infecciosos pelo hospedeiro, mas também pode contribuir na patogênese de injúria pulmonar.

Samples were seeded on Sabouraud dextrose agar with chloramphenicol after be obtained by the introduction of urethral probe through the esophagus. Based on morphology and biochemical reactions (API 20C) Candida was confirmed; it was still searched the production of proteinase and phospholipase, virulence factors for Candida species. Candida spp. were isolated from 57.5% parrots, being 72.2% from birds with ingluvitis and 45.5% from without ones. Twenty-five strains of Candida were isolated, 60% and 40%, respectively from parrots with and without ingluvitis, and were speciated: 28% C. humicola, 24% C. parapsilosis, 20% C. guilliermondii, 20% C. famata, and 8% C. albicans. These results demonstrate that C. albicans is not the most frequent species isolated, and it is the first report that shows C. guilliermondii, C. famata, and C. humicola causing infection in parrots. Many isolates presented filamentation (76%), 100% produced proteinase and 68% phospholipase. The observation of Candida spp. producing virulence factors reinforce the pathogenic role of these yeasts in the cases studied.

The distribution of these findings by organ system were as follows: digestive (79/335 [23.6%]), striated muscle and skeleton (47/335 [14.0%]), nervous (37/335 [11.0%]), respiratory (35/335 [10.4%]), integument (31/335 [9.3%]), hematopoietic (24/335 [7.2%]), cardiovascular (13/335 [3.9%]), reproductive (12/335 [3.5%]), urinary (7/335 [2.1%]), and endocrine (3/335 [0.9%]). The cause of death was not possible to be determined in 47 (14.0%) necropsied horses. Displacements of the intestines (17/79 [21.5%]) were the main findings in digestive system, followed by obstruction and impactation (14/79 [17.7%]). Torsion were the type of displacement more frequently observed in the intestines (14/17 [82.4%]). Among those the more prevalent affected the small intestine (7/14 [50%]). Most horses dying from fractured bones were 1-5-year-old. The most prevalent diseases in the nervous system were leukoencephalo-malacia and trypanosomiasis, whereas respiratory depression due to anesthesia was the leading cause of death related to the respiratory system. Equine infectious anemia was the most diagnosed infectious disease and the main reason leading to euthanasia.
