In addition to knowing its life cycle, knowledge of parasite’s epidemiology is essential to establish management strategies. Epidemiological studies on resistance to acaricides in Rio Grande do Sul as well as in the rest of Brazil are scarce. Moreover, the large geographical area and the structural deficiency with respect to the use and access to databases make reliable data difficult to obtain. The present study surveyed the perception by cattle breeders in the southern region of Rio Grande do Sul in regards of identification of R. (B.) microplus populations that are difficult to manage using acaricides, as well as the risk factors for the selection of resistant tick populations. Tick management data on beef cattle in 85 properties of seven municipalities were collected. The results revealed that the difficulty in tick management correlated positively with levels of education of the farm owners (up to elementary school, OR = 3.67 and p = 0.01) and with the yearly number of acaricide applications (over four, with OR = 4.05 and p= 0.006). These results also suggest that properties with more than 100 beef cattle under extensive farming conditions in the southern region of Rio Grande do Sul show characteristics that may contribute to longer acaricide lifetimes as compared to other regions in the Country.

The implantation of xenologous amniotic membrane was performed bilaterally in 90 rats. In the same day of the surgery each right eye started receiving topical CsA twice a day. The left eye received no medication and served as a control. The evaluation of corneal neovascularization was performed by computerized image analysis and histopathological evaluation at 1, 3, 7, 15, 30 and 60 days postoperatively. For the image analysis 10 animals were used per time period, and for the histopathological examination, five animals were used per time period. Image analysis found that corneal neovascularization began on the 3rd postoperative day, reached its peak on the 7th day, and then progressively and rapidly decreased. Statistic analysis indicated that neovascularization of the CsA treated eye on the 7th day was significantly higher than that observed in untreated eyes. On the 30th day, however, this pattern was reversed with the neovascularization observed in the CsA treated eyes declining to the low levels observed on the 3rd day. The degree of neovascularization in the untreated eyes on the 30th day declined to the baseline levels found on day 3 at the 60th day. Histopathological analysis indicated that deposition of collagen in the implanted tissue was completed by the 15th day. Therefore, we concluded that (1) equine amniotic membrane in rat corneal stroma produced an intense neovascularization until the 15th day postoperatively and then regressed, (2) deposition of collagen of the implanted tissue was completed on the 15th day postoperatively, and (3) use of CsA was associated with increase in the corneal neovascularization initially, followed by a quick and intense regression.

E-mail: camilelug@gmail.com Cryptococcus neoformans and C. gattii are associated with dry bird excreta but rarely recovered from birds’ digestive tract. The objective of the present study was (1) to verify the existence of C. neoformans and C. gattii in crop and cloaca of wildlife and captivity birds hypothesizing about a possible primary source of this yeast in the excreta, and (2) to determine the fungi’s invasive capability in avian species through latex agglutination. For that purpose, 172 cloacal and 77 crop samples of domestic pigeon, Passerine, and Psittacine birds were collected. None of these samples was positive, suggesting that the yeast is not saprobiotic in the digestive tract of these birds. Only one out of 82 serum samples collected from pigeons and Psittacine birds was positive (title 1:2) showing that Cryptococcus sp. probably has a low invasive capability in birds, and is thus considered only a dry excreta colonizer.

A number of FMD vaccine candidates have been tested and a replication-defective human adenovirus type 5 (Ad5) vector containing the FMDV capsid (P1-2A) and 3C protease coding regions has been shown to completely protect pigs against challenge with the homologous virus (FMDV A12 and A24). An Ad5-P1-2A+3C vaccine for FMDV O1 Campos (Ad5-O1C), however, only induced a low FMDV-specific neutralizing antibody response in swine potency tests. Granulocyte-macrophage colony-stimulating factor (GM-CSF) has been successfully used to stimulate the immune response in vaccine formulations against a number of diseases, including HIV, hepatitis C and B. To attempt to improve the FMDV-specific immune response induced by Ad5-O1C, we inoculated swine with Ad5-O1C and an Ad5 vector containing the gene for porcine GM-CSF (pGM-CSF). However, in the conditions used in this trial, pGM-CSF did not improve the immune response to Ad5-O1C and adversely affected the level of protection of swine challenged with homologous FMDV.

[Relationship between foamy macrophages in the liver of cattle and the ingestion of Brachiaria spp.] Relação entre macrófagos espumosos ("foam cells") no fígado de bovinos e ingestão de Brachiaria spp no Brasil. Pesquisa Veterinária Brasileira 19(2):79-83. Depto Patologia Clínica Veterinária, Faculdade de Veterinária, UFRGS, Cx. Postal 15094, Porto Alegre, RS 91540-000, Brazil. To establish the etiological relationship and the appearance of foamy macrophages in the liver of cattle from tropical regions of Brazil, Iiver samples from the files of the Pathology Section of Embrapa-Projeto Sanidade Animal, Rio de Janeiro, were reviewed. A total of 55 liver samples of cattle which died from different causes between 1970 and 1991 were reexamined. Only samples of animals which grazed known pastures were reviewed. Foamy macrophages were not seen in the samples from 1970 to 1975, although 40 samples (72%) were from this period. Foamy macrophages were observed from 1976 on, coinciding with the introduction of Brachiaria decumbens from Australian seeds into Brazil. Some samples were from cattle with histories of photosensitization, which were at that time attributed to Pithomyces chartarum. The results of this study indicate that the liver changes are related to prolonged ingestion of Brachiaria spp.

O material utilizado provinha de investigações sobre causas de mortandades em bovinos nas regiões Norte, Centro-Oeste e Sudeste do Brasil, realizadas de 1970 a 1991. Foram estudados 55 fígados de bovinos afetados por enfermidades variadas. Somente foram usados casos em que o tipo de pastagem era conhecido. Essa alteração não foi encontrada de 1970 até o final de 1975, embora 40 amostras (72, 7%) tenham sido coletadas nesse período. A presença de macrófagos espumosos, observada a partir de 1976, coincidiu com a introdução da gramínea Brachiaria decumbens var. australiana no Brasil. Algumas amostras eram provenientes de bovinos que apresentaram histórico de fotossensibilização, na época atribuída ao fungo Pithomyces chartarum. Os achados indicam que essas alterações hepáticas são relacionadas com a ingestão de Brachiaria spp.

Samples of liver, kidney, spleen, and hepatic, mesenteric, retromandibular, prescapular and mediastinic lymph nades, collected from 12 groups of cattle of different ages from the State of Mato Grosso, were studied. A total of 120 bovines was examined. According to their history, the animals were kept in pastures where Brachiaria decumbens and Brachiaria brizantha were the predominant plants. Grossly there was a yellowish liver which was more evidente after 24 hr. fixation with formalin. Hepatic and mesenteric lymph nades had normal size, howeve1; on the cut surface there were whitish grooves, extending from the cortex to the medulal; and multiple small white nodular areas in the medulal: Beside these areas there were red, sometimes hemorrhagic, foci. Microscopically all animals had cells with foamy citoplasm in the liver and in the hepatic and mesenteric lymph nades, of ten forming multinucleated cells. In the lymph nodes these infiltrates were adjacent to necrotic, hemorrhagic areas. In the liver the foamy cells were irregularly disseminated throughout the parenchyma or formed nodules around the central vein. These cells did not stain with PAS but weakly with oil red. Ultrastructurally foamy cells and hepatocytes showed negative images of crystals involved partially or totally by membranes. Mediastinic, retromandibular and prescapular lymph nades of the sarne animals showed no gross and microscopic alterations.

Macroscopicamente o fígado desses animais mostrava coloração amarelada, mais evidente após 24 horas de fixação em formol a 10%. Nos linfonodos hepáticos e mesentéricos foram evidenciadas, na superfície de corte, estriações esbranquiçadas de forma radiada na cortical e medular com pequenas áreas brancas nodulares multifocais principalmente na medular. Em muitos casos, associado com essas áreas foram vistos focos de aspecto hemorrágico. Microscopicamente foram encontradas, no fígado, linfonodos hepáticos e mesentéricos, células com citoplasma espumoso, muitas das quais multinucleadas. Nos linfonodos hepáticos e mesentéricos estes infiltrados estavam associados a áreas de necrose e hemorragia. No fígado, as células de citoplasma espumoso estavam presentes em todo parênquima, de forma irregular, geralmente formando nódulos ao redor da veia centrolobula1: Estas células não se coravam pelo ácido periódico de Schiff (PAS) e apenas fracamente na coloração de gordura pelo Oil Red O. Na ultraestrutura as células de citoplasma espumoso apresentavam fendas, parcial ou totalmente delimitadas por membrana, que representam a imagem negativa de cristais, presentes também no citoplasma dos hepatócitos.


#38 - Analysis of the antigenicity of constituents of Boophilus microplus' saliva

Abstract in English:

The macromolecules present in the Boophilus microplus saliva were separated by Electrophoresis on Polyacrylamide Gels and transferred to nitrocellulose paper. Some aspects were studied in both situations. Using Coomassie Brilliant Blue 250R to stain the proteins in the gel, 6 and 11 bands were detected on nondenaturing and denaturing systems respectively. Three glicoprotein and one lipoprotein bands were detected when Beta-mercaptoethanol was used (denaturing system). In this situation, three antigenic bands were observed in nitrocellulose paper after immunoenzymatic procedure when infested bovine or hyperimmune rabbit or mouse sera were used. All of these were glicoproteins.

A saliva (pool) de Boophilus microplus foi submetida a eletroforese em gel de poliacrilamida e logo a seguir procedida a transferência para papel de nitrocelulose, a fim de que a antigenicidade pudesse ser observada. A eletroforese revelou a existência de seis bandas quando em condições de não desnaturação e 11 bandas em condições desnaturantes. Neste último caso, três bandas eram glicoprotefuas e uma lipoprotefua. A revelação imunoenzimática no papel de nitrocelulose evidenciou três bandas quando se usaram soros de bovinos infestados por esse ixodídeo ou soro de coelhos e camundongos imunizados com a citada saliva.


#39 - Effects of some pastore grasses on infestive larvae of the cattle tick Boophilus microplus

Abstract in English:

The pasture species Andropogon gayanus, Melinis minutiflora and Brachiaria brizantha were studied with respect to their anti-tick properties through repellent and/or lethal mechanisms affecting infestive larvae of Boophilus microplus (Can., 1887). After two months cultivation in pots, the grasses were infested with 10-day old larvae under laboratory conditions. Observations of larvae were made at 2, 7 and 15 days after infestation and collections made at 5, 10 and 20 days after infestation. A. gayanus did not show any prejudicial affects against the larvae, under these conditions. M. minutiflora and B. brizantha showed themselves to be potentially highly lethal to larvae. M. minutiflora showed itself to be highly repellent to larvae of B. microplus. This was considered to reduce its potential as a possible anti-tick pasture as many larvae were caused to avoid its lethal effect. B. brizantha did not show this repellency and consequent disadvantage. It is thus considered to warrant further study under experimental and natural conditions to assess its value as a biological tick control agent for possible inclusion in integrated pest management (IPM) schemes for B. microplus in the tropics.

Abstract in Portuguese:

As espécies forrageiras Andropogon gayanus, Melinis minutiflora e Brachiaria brizantha foram estudadas em relação a suas propriedades "anti-carrapato" através de mecanismos de repelência e/ou letalidade afetando larvas infestantes de Boophilus microplus (Can., 1887). Após dois meses do cultivo em vasos, as gramíneas foram infestadas com larvas de 10 dias de idade, em condições de laboratório. Foram realizadas observações das larvas aos 2, 7 e 15 dias pós-infestação (dpi) e coletas aos 5, 10 e 20 dpi. A. gayanus não apresentou qualquer efeito prejudicial sobre as larvas, ao contrário, M. minutiflora e B. brizantha demonstraram ser potencialmente letais às larvas. M. minutiflora apresentou também um elevado poder de repelência às larvas de B. microplus, sendo esta propriedade considerada prejudicial ao seu potencial "anti-carrapato" em função de muitas larvas evitarem sua ação letal; B. brizantha não mostrou ser repelente e conseqüentemente não apresentou esta desvantagem. Desta forma, justificam ser realizados em B. brizantha estudos adicionais sob condições naturais e experimentais, a fim de avaliar seu real valor no controle biológico, assim como sua inclusão no manejo integrado de pragas (1PM) nas regiões tropicais.
