E-mail: adiaz@mdp.edu.ar The composition and distribution of the glycoconjugates (GCs) secreted by the epithelium of ovarian lamellae with reference to the reproductive biology of Genypterus blacodes (Schneider, 1801) through lectin histochemistry is here discussed. In this species, the epithelial cells that line the ovarian cavity presented sharp morphological variations along the reproductive cycle related to the mucus secretion that accompanies oocyte maturation. During spawning season, residues of mannose and N-acetylglucosamine were detected in the glycocalyx of those cells using lectinhistochemistry. N-acetylgalactosamine and fucose were also observed in the same zone. The greatest variations in the lectinhistochemical pattern were found in the apical cytoplasm composition in comparison to the basal zone of the cells. The results of the present study were discussed by comparing their possible functional implications.

Nesta espécie, as células epiteliais que revestem a cavidade do ovário apresentou acentuada variação morfológica ao longo do ciclo reprodutivo relacionados com a secreção de muco que acompanha a maturação do oócito. Durante a época de desova, de resíduos de manose e N-acetilglicosamina foram detectados no glicocálix dessas células usando histoquímica de lectinas. N-acetilgalactosamina e fucose também foram observados na mesma zona. As maiores variações no padrão de lectinas foram encontradas na composição do citoplasma apical, em comparação com a zona basal das células. E-mail: eldadias@uft.edu.br This study aimed to evaluate the threshold of detection of fluorescent multiplex PCR coupled with capillary electrophoresis for detection of infectious agents in semen samples from experimentally infected with decreasing concentrations of the bacteria Brucella abortus, Leptospira interrogans serovar pomona, Campylobacter fetus and Haemophilus somnus. Samples of bovine semen were experimentally infected with decreasing concentrations of bacteria obtained from serial dilutions in the base 10 so as to obtain samples containing a long time until 10-7 bacteria/mL from the initial concentration of Lepstospira pomona, Brucella abortus, Haemophilus somnus and Campylobacter fetus. The dilutions were made individually for each bacterium, as well as in different concentrations needed to standardize the multiplex PCR test. DNA extractions of all solutions containing sperm and bacteria analyzed in this study were performed according to protocol described by Heinemann et al. (2000). The multiplex PCR products were analyzed by electrophoresis on 8% polyacrylamide gel and capillary electrophoretic separation system for automated equipment in the analysis of DNA fragments MegaBACE. We observed amplification of fragments of 193pb, 330pb, 415pb and 400bp from the DNA of B. abortus, L. pomona, H. somnus, C. fetus respectively. The analysis by capillary electrophoresis of multiplex PCR products of DNA for simultaneous detection of the four pathogens was observed by detecting the dilution of 10-3 bacteria / mL times the initial concentration of the stock solution of each bacterium. The multiplex PCR coupled with capillary electrophoresis was first used for the direct diagnosis of four pathogenic bacteria in semen, proving to be a rapid method to detect bacteria that cause reproductive disorders.

E-mail: eldadias@uft.edu.br Este estudo teve como objetivo avaliar o limiar de detecção da técnica de PCR multiplex fluorescente aliada a eletroforese capilar na detecção de agentes infecciosos em amostras de sêmen experimentalmente contaminadas com concentrações decrescentes das bactérias Brucella abortus, Leptospira interrogans sorovar pomona, Campylobacter fetus e Haemophilus somnus. Amostras de sêmen bovino foram experimentalmente contaminadas com concentrações decrescentes de bactérias obtidas através de diluições seriadas na base 10 de modo a obter-se amostras contendo desde 1 vez até 10-7 bactérias/mL a partir da concentração inicial de Leptospira pomona, Brucella abortus, Campylobacter fetus e Haemophilus somnus. As diluições foram efetuadas individualmente para cada bactéria, bem como nas diferentes concentrações necessárias para a padronização do teste de multiplex PCR. As extrações de DNA de todas as soluções contendo espermatozóides e bactérias analisadas no presente estudo foram realizadas segundo protocolo descrito por Heinemann et al. (2000). Os produtos de PCR multiplex foram avaliados por eletroforese em gel de poliacrilamida 8% e separação eletroforética por sistema capilar em equipamento automático de análise de fragmentos de DNA MegaBace. Observou-se a amplificação de fragmentos de 193pb, 330pb, 400pb e 415pb a partir do DNA de B. abortus, L. pomona, H. somnus, C. fetus, respectivamente. Na análise por eletroforese capilar de produtos da PCR multiplex do DNA para detecção simultânea dos quatro patógenos observou-se a sinal de positividade até a diluição de 10-3 bactérias/mL vezes da concentração inicial da solução estoque de cada bactéria. A técnica de PCR multiplex aliada à eletroforese capilar foi usada pela primeira vez para o diagnóstico direto de quatro bactérias patogênicas no sêmen, demonstrando ser um método rápido na detecção de bactérias causadoras de doenças reprodutivas.

E-mail: juliana_gamba@yahoo.com.br Morphological analysis of the ostrich eggshell is important because several factors can be related to morphological hatchability of eggs of this species in Brazil. Therefore, 60 samples of apparently normal eggs were analyzed (eggs hatched and with embryonic death). The observations were allowed to establish that: the average thickness of the eggshell, the porosity of the shell of the eggs, and correlate these variables with hatchability. The results were statistically analyzed by Duncan’s test, which showed that eggs with less porosity also have a lower hatchability. The average porosity of hatched eggs is 19.87 pores/cm2, and of eggs with embryonic death is pores/cm2 16.78. There was no statistical difference between the thickness of the shell in the regions studied, no interaction between viability and the regions studied.

Para tanto foram analisadas 60 amostras de ovos aparentemente normais (ovos eclodidos e com morte embrionária). Foram efetuadas observações que permitiram estabelecer: a espessura média da casca do ovo, a porosidade da casca destes ovos, e correlacionar estas variáveis com a eclodibilidade. Os resultados foram analisados estatisticamente mediante o teste de Duncan, o que mostrou que ovos com menor porosidade tem também uma menor eclodibilidade. A porosidade média dos ovos eclodidos é de 19,87 poros/cm2, e dos ovos com morte embrionária é de 16,78 poros/cm2. Não houve diferença estatística entre a espessura da casca nas regiões estudada, nem interação entre viabilidade e as regiões.

E-mail: alejandrabarrera.ufrgs@yahoo.com In early lactation dairy cattle suffer metabolic alterations caused by negative energy balance, which predisposes to fatty liver and ketosis. The aim of this study was to evaluate the metabolic condition of high yielding dairy cows subjected to three treatments for preventing severe lipomobilization and ketosis in early lactation. Fifty four multiparous Holstein cows yielding >30 L/day were divided into four groups: control (CN= no treatment), glucose precursor (PG= propylene-glycol), hepatic protector (Mp= Mercepton®), and energy supplement with salts of linolenic and linoleic faty acids (Mg-E= Megalac-E®). Treatments were administrated randomly at moment of calving until 8 weeks postpartum. Blood samples were collected on days 1, 7, 14, 21, 28, 35, 42 and 49 postpartum. Body condition score (BCS) was evaluated at the same periods and milk yield was recorded at 2nd, 4th, 5th, 6th, 7th, and 8th weeks of lactation. Concentrations of non-esterified fatty acids (NEFA), albumin, AST, ß-hydroxybutyrate (BHBA), cholesterol, glucose, total protein, urea and triglycerides were analyzed in blood samples. Cut-off points for subclinical ketosis were defined when BHBA &#8805;1.4 mmol/L and NEFA &#8805;0.7 mmol/L. General occurrence of subclinical ketosis was 24% during the period. An ascendant curve of cholesterol and glucose was observed from the 1st to the 8th week of lactation, while any tendency was observed with BHBA and NEFA, although differences among treatments were detected (p&#8804;0.05). BCS decreased from a mean of 3.85 at 1st week to 2.53 at 8th week of lactation (p=0.001). Milk yield was higher in the Mg-E group compared with the other treatment groups (p&#8804;0.05) Compared with the CN group, the treatments with Mp and PG did not show significant differences in blood biochemistry and milk yield. Cows receiving PG and Mg-E showed higher values of BHBA and NEFA (P<0.05), indicating accentuated lipomobilization. Supplementation with Mg-E also resulted in significant higher concentrations of cholesterol, BHBA, urea, AST and lower values of glycemia. This performance may be explained by the highest milk yield observed with this treatment. Treatments with PG and Mp did not improve milk yield, compared with control cows, but did not show metabolic evidence of ketosis, fat mobilization or fatty liver. These results suggest that treatment with Mg-E improves milk production but induces a higher negative energy balance leading to moderated lipomobilization and ketone bodies production, increasing the risk of fatty liver.

O objetivo deste estudo foi avaliar o status metabólico de vacas leiteiras de alta produção submetidas a três tratamentos para prevenir severa lipomobilização e cetose no início da lactação. Cinquenta e quatro vacas de raça Holandesa multíparas produzindo >30 L/dia foram divididas em quatro grupos: controle (CN= sem tratamento), precursor de glicose (PG= propileno-glicol), protetor hepático (Mp= Mercepton®) e suplementação com sais de ácidos linolênico e linoléico (Mg-E= Megalac-E®). Amostras de sangue foram coletadas nos dias 1, 7, 14, 21, 28, 35, 42 e 49 do pós-parto. A condição corporal foi avaliada nos mesmos períodos e a produção de leite foi registrada nas semanas 2, 4, 5, 6, 7 e 8 de lactação. As concentrações de ácidos graxos não esterificados (AGNE), albumina, AST, ß-hidroxibutirato (BHB), colesterol, glicose, proteína total, uréia e triglicerídeos foram determinadas nas amostras de sangue. Pontos de corte para diagnosticar cetose subclínica foram definidos quando BHB &#8805;1,4mmol/L e AGNE &#8805;0,7mmol/L. A ocorrência geral de cetose subclínica foi de 24% durante o período. Uma curva ascendente de colesterol e de glicose foi observada desde a 1ª até a 8ª semana de lactação, enquanto que nenhuma tendência foi observada com BHB e AGNE, embora diferenças entre os tratamentos foram detectadas (p&#8804;0,05). A condição corporal diminuiu de uma media de 3,85 na 1ª semana até 2,53 na 8ª semana de lactação (p=0,001). A produção de leite foi superior no grupo de Mg-E comparado com os demais tratamentos. Comparado com o grupo CN, os tratamentos de Mp e PG não mostraram diferenças significativas na bioquímica sanguínea nem na produção de leite (p&#8804;0,05) As vacas que receberam PG e Mg-E mostraram maiores valores de AGNE, indicando uma acentuada lipomobilização. A suplementação com Mg-E também resultou em maiores concentrações de colesterol, BHB, uréia, AST e menores valores de glicemia. Este resultado pode ser explicado pela maior produção de leite observada com este tratamento. Os tratamentos com PG e Mp não melhoraram a produção de leite, comparados ao grupo CN, mas também não mostraram evidências metabólicas de cetose, alta lipomobilização nem infiltração gordurosa hepática. Os resultados sugerem que o tratamento com Mg-E melhora a produção de leite, mas induz um balance energético negativo maior levando a moderada lipomobilização e produção de corpos cetônicos, aumentando o risco de fígado gorduroso.

E-mail: daniapag@yahoo.com.br In recent years the ferret (Mustela putoriu s furo) has become popular pet featuring a ever growing population in Brazil, and being increasingly found in veterinary clinics. This study aimed to evaluate the ultrasound anatomy of abdominal lymph nodes of healthy ferrets. We used 20 animals, nine of which males and eleven females, with overall mean age of three years. We located the mesenteric lymph nodes in 100% of the ferrets, the pancreatic-duodenal lymph nodes and the splenic ones in 55%, the gastric lymph node in 20%, and the hepatic lymph node in 5%. We measured their lengths. It is concluded that the location and sonographic characteristics of the abdominal lymph nodes in ferrets are very similar to the abdominal lymph nodes of cats. This is a preliminary orientation for the location of abdominal lymph nodes of healthy ferrets

E-mail: daniapag@yahoo.com.br Nos últimos anos o furão (Mustela putorius furo) tornou-se um conhecido animal de estimação sendo observada uma população em constante crescimento no Brasil, e por conseqüência cada vez mais presente em clínicas veterinárias. O presente trabalho teve como objetivo avaliar a anatomia ultrassonográfica dos linfonodos abdominais de furões-europeus hígidos. Foram utilizados 20 animais, dentre os quais nove eram machos e onze fêmeas, com idade média total de três anos. Localizaram-se em 100% dos furões os linfonodos mesentéricos, em 55% dos animais os linfonodos pancreático-duodenal e esplênico, em 20% o linfonodo gástrico e em 5% o linfonodo hepático. Conclui-se que a localização e características ultrassonográfica dos linfonodos abdominais em furões são muito similares aos linfonodos abdominais de gatos, sendo este estudo uma orientação preliminar para a localização dos linfonodos abdominais de furões hígidos.

Pesquisa Veterinária Brasileira 31(8):702-706. Laboratório de Patologia Veterinária, Campus Jataí, Universidade Federal de Goiás, Jataí, GO 75801-458, Brazil. E-mail: santanafjf@yahoo.com An outbreak of poisoning by Senna occidentalis in wild boars in Goiás, Brazil, is described. Out of 80 wild boars, 15 were affected and one died. Clinical signs included apathy, muscle tremors, incoordination, reluctance to move, sternal or lateral recumbence, and paresis and spastic paralysis, especially in the hind limbs. Gross lesions were characterized by moderate to severe paleness of the skeletal muscles of the hind and thoracic limbs and loin, and were marked in the thigh muscles. Histologically, a monophasic, mild to moderate, multifocal floccular muscle degeneration and necrosis with skeletal fiber sarcoplasmic fragmentation was observed. In transverse sections there were multiple swollen and hypereosinophilic muscle fibers. In addition, mild to moderate, diffuse microvacuolar degeneration was seen in the liver. Creatine phosphokinase serum activity was severely elevated in two evaluated wild boars. In one of them, there was elevation of glutamic oxaloacetic transaminase serum activity.

E-mail: santanafjf@yahoo.com Descreve-se um surto de intoxicação por Senna occidentalis em javalis no Estado de Goiás. De um rebanho de 80 javalis, 15 adoeceram e um morreu. Os sinais clínicos observados foram apatia, prostração, ataxia, tremores musculares, incoordenação, relutância em mover-se, decúbito esternal ou lateral, paresia e paralisia espástica, principalmente dos membros pélvicos. As principais alterações macroscópicas consistiam de palidez moderada a acentuada da musculatura esquelética dos membros pélvicos e torácicos e no lombo, especialmente nos músculos da coxa. Microscopicamente, observou-se degeneração e necrose flocular, multifocal, leve a moderada, monofásica, com fragmentação de fibras na musculatura esquelética. Nos cortes transversais, havia fibras musculares tumefeitas e hipereosinofílicas. Adicionalmente, havia degeneração microvacuolar hepatocelular difusa, leve a moderada. A atividade sérica da CK estava acentuadamente elevada em dois javalis avaliados e da TGO aumentou em um javali afetado.

[Ultrasonographic evaluation of the external ear in dogs.] Avaliação ultrassonográfica do meato acústico em cães. Pesquisa Veterinária Brasileira 30(4):335-339. E-mail: tilde@ufpr.br The purpose of this study was to determine the viability of ultrasonographic evaluation of the external ear in dogs. The thickness and architecture of the ear wall and tympanic bulla of 10 Labrador Retrievers without ear disease was evaluated. The examination was performed with an ultrasound system and a 14 MHz bidimentional linear-array transducer and general anesthesia, before and after the infusion of saline solution into the ear canal. The maxillary artery was identified with color Doppler and helped to establish the location of the tympanic bulla in all dogs. The proximal wall was less well-defined when compared with the distal wall of the ear canal. Dogs with increased ceruminal content within the ear canal showed a hypoechoic epithelium. The tympanic membrane was not identified. It was concluded that ultrasound evaluation is feasible and needs to be explored as clinical diagnostic tool for the diagnosis of ear diseases in dogs.

[Ultrasonographic evaluation of the external ear in dogs.] Avaliação ultrassonográfica do meato acústico em cães. Pesquisa Veterinária Brasileira 30(4):335-339. E-mail: tilde@ufpr.br O propósito deste trabalho foi determinar a viabilidade da ultrassonografia para avaliar o canal auditivo externo de cães, aferindo a espessura e arquitetura da parede do conduto auditivo e bula timpânica. Foram avaliados 10 cães da raça Retriever do Labrador não portadores de otite externa. O exame foi realizado com o aparelho de ultrassonografia bidimensional e transdutor linear de 14 MHz, com todos os animais sob efeito de anestesia geral. O exame foi efetivado antes e depois do conduto ser preenchido com solução salina, promovendo uma janela acústica anecóica. A artéria maxilar foi identificada com o Doppler colorido o que facilitou a localização da bula timpânica em todos os cães. A definição da imagem da parede proximal ao transdutor foi pior quando comparada à parede distal r em todos os animais, isto devido à proximidade da parede com a face de leitura do transdutor. Nos cães que apresentavam uma maior quantidade de cerúmen a superfície do epitélio estava com uma camada hiperecóica mais evidente. A membrana timpânica não foi identificada em nenhum animal. Conclui-se que ultrassonografia é viável e deve ser mais pesquisada na contribuição diagnóstica das doenças óticas em cães.

However, circulatory arrest, event occasioned by this technique, may produce serious metabolic and neurological consequences to the patient. In this study, 12 mongrel dogs were used, divided into two groups, A and B, which were submitted to 7 and 8 minutes of total circulatory arrest, respectively, using “Inflow Occlusion” technique. Normothermia was tried during surgical procedures. Clinical and behavior evaluation were performed after surgery to both groups, and biochemical data were collected to compare pre and post-operatory moments. There were two transoperatory deaths in Group B. Transitory clinical problems were observed in group A until moment M7 (48 hours after surgery), and in Group B these problems were more intense and seen even after M7; and permanent blindness in one animal of Group B was present during all follow up period. Despite all alterations found during the study, it might be safe to use “Inflow Occlusion” technique for periods up to 7 minutes, however, it is contra-indicated for longer periods.

Blood samples were obtained for 5-day intervals to hematocrit analysis. At the end of the experimental period, the sheep were orquiectomized. Testes and epididymides from these animals were studied anatomopathologically. Samples from these tissues of Sheep 1, 4 and 5 were taken to polymerase chain reaction (PCR). Clinical parameters remained for the infected group above the values observed in the control group during the experimental period. Parasitaemia was observed on day 3 post-infection, and the highest values occurred between day 6 and 10, and day 15 and 18 post-infection. Sheep 1 and 4 showed severe anemia on day 25 post-infection. All sheep of the infected group showed flabby and palid testes. Histologically, moderate to severe testicular degeneration, multifocal epididymitis and hyperplasia of epididymal epithelium were observed. The result of T. vivax PCR analysis in the testes and epididymal tissues was positive in 100% of the samples of the experimentally infected sheep. Epididymal and testicular lesions associated with the presence of the parasite in these tissues, shown by PCR, suggest the participation of T. vivax in the pathophysiological mechanism of reproductive damage.

Among diarrheic calves, Escherichia coli (69.0%) was the most common agent found, following by Cryptosporidium spp. (30.0%), coronavirus (16.0%), and rotavirus (11.0%). In the control group, E. coli, Cryptosporidium spp. and coronavirus were detected in 66.7%, 10.0% and 3.3% of the samples, respectively. Salmonella spp. and strongylids were not found in any of the calves from either group. The K99 fimbrial only was detected in E. coli strains from diarrheic calves (5.8%). Enrofloxacin, norfloxacin, and gentamicin were the most effective among the antimicrobials tested. The weight of 210-day-old calves did not show statistic differences between diarrheic and non-diarrheic calves.
