#41 - Perinatal lesions caused in lambs by feeding Tetrapterys multiglandulosa (Malpighiaceae) to sheep at different stages of their pregnancy, 30(1):73-78

E-mail: eqrural@nin.ufms.br In order to verify the effects of non-lethal doses of Tetrapterys multiglandulosa on ovine fetuses, experimental poisoning in sheep at different days of pregnancy was performed. Green leaves of shooting plants were administered to 9 pregnant ewes divided into three experimental groups. Sheep from Group 1 received four doses of 5g/kg of fresh leaves for 4 days; those from Group 2 received 10g/kg for 2 days; Group 3 sheep received a dose of 20g/kg for one day, and sheep from Group 4 did not receive the plant and served as negative controls. To check the plant toxicity, a male sheep (positive control) received 5g/kg until the onset of clinical signs. No signs of poisoning were observed in pregnant ewes; three lambs died 1-5 days after birth, and a fourth lamb died within 3 months after have been born. The positive control died after 38 days of daily consumption of the plant, presenting tachypnea, tachycardia, drowsiness, incoordination, weakness and sudden death. All four dead lambs and the positive control sheep were necropsied. The gross lesions were whitish areas in the myocardium, increased lobular pattern of the liver, dark red lungs, metabolization of pericardial fat, and ruminal distention with free gas. Histological findings were cardiac fibrosis, cardiomyocyte necrosis, pulmonary congestion and edema, and spongy degeneration in subcortical cerebral white matter. Tetrapterys multiglandulosa resulted toxic for ovine fetuses at doses that were not suffient to induce clinical signs in the pregnant ewes, demonstrating that the poisoning may be a cause of death of lambs soon after birth, even several months after the ingestion of the plant has been discontinued.

As ovelhas do Grupo 1 consumiram quatro doses de 5g/kg de folhas verdes durante quatro dias; ovinos do Grupo 2 consumiram duas doses de 10g/kg durante dois dias, o Grupo 3 consumiu uma dose de 20g/kg em um único dia e os ovinos do Grupo 4 não consumiram a planta (controle-negativo). Para testar a toxicidade da planta um ovino macho consumiu 5g/kg até manifestação dos sintomas. Não foram observados sinais clínicos da intoxicação nas ovelhas, mas três cordeiros tiveram morte perinatal, e um cordeiro morreu com três meses de idade; o ovino controle-positivo morreu após 38 dias de consumo diário da planta e os sinais clínicos foram taquipnéia, taquicardia, sonolência, incoordenação e fraqueza. Todos os quatro cordeiros e o ovino controle-positivo foram necropsiados e os achados de necropsia foram áreas esbranquiçadas no miocárdio, evidenciação do padrão lobular do fígado, pulmão vermelho enegrecido e rúmen acentuadamente distendido com presença de gases livres. Os achados histopatológicos foram fibrose cardíaca, necrose dos cardiomiócitos, congestão e edema pulmonar, congestão hepática centrolobular e degeneração esponjosa na região subcortical do encéfalo. Tetrapterys multiglandulosa demonstrou ser tóxica para os fetos ovinos em dosagens que não foram suficientes para induzir sinais clínicos nas ovelhas prenhes, e que a intoxicação pode ocorrer com morte de cordeiros logo após o parto, mesmo após meses de ter cessada a ingestão da planta.

E-mail: claudioslbarros@uol.com.br Polioencephalomalacia (PEM) of ruminants is a complex disease. The term indicates a morphological diagnosis where severe neuronal necrosis results in softening of cerebral grey matter. Initially though as a single disease caused by thiamine deficiency, PEM is currently believe to have several causes and different pathogenic mechanisms or a single pathogenic organism triggered by different agents are responsible for the disease. In this paper the possible causes and pathogenesis of PEM in ruminants are critically reviewed and discussed. Also are reviewed the epidemiology, clinical signs, gross and histological findings, methods of diagnosis, treatment and control.

O termo indica um diagnóstico morfológico em que necrose neuronal grave resulta em amolecimento da substância cinzenta do cérebro. Interpretada no início como uma doença única, causada por deficiência de tiamina, acredita-se hoje que várias causas e diferentes mecanismos patogênicos, ou um único mecanismo patogênico disparado por diferentes agentes, sejam responsáveis pelo aparecimento da doença. Neste artigo, as possíveis causas e a patogênese de PEM em ruminantes são criticamente revisadas e discutidas. Também são revisadas a epidemiologia, os sinais clínicos, os achados macro e microscópicos e os métodos de diagnóstico, tratamento e controle.

[Experimentally amprolium-induced polioencephalomalacia in sheep.] Polioencefalomalacia experimental induzida por amprólio em ovinos. Pesquisa Veterinária Brasileira 29(9):747-752. E-mail: claudioslbarros@uol.com.br In order to establish an experimental model for the study of the etiology, pathology, and pathogenesis of polioencephalomalacia in ruminants, the condition was induced in five sheep by oral administration of amprolium at daily doses of 500 and 1,000mg per kg of body weight respectively for 28-59 days and for 13-39 days. All sheep died or were euthanized in extremis after illness of 3-7 days. Clinical signs included depression, incoordination, midriasis, grinding of the teeth, blindness, and laying down with opisthotonus and paddling movements. Drooling and a sawhorse stance were observed in one sheep and myoclonus in another one. Main gross lesions were restricted to the central nervous system and included swelling of the brain with flattening of telencephalic gyri, and hemorrhages in the parietal and occipital lobes of the telencephalon, in the submeningeal areas of the spinal cord and in the mesencephalon. Histologically, there was segmental laminar neuronal necrosis (red neurons) associated with edema, swelling of endothelial cells, hemorrhages and infiltration by foamy macrophages (gitter cells). These changes were more marked in the frontal, parietal and occipital telecephalic lobes and there was sharp demarcation between the lesions and the adjacent normal neuropile. Additionally, similar, but less marked lesions were seen in the mesencephalon, thalamus and hippocampus. Considering the consistent reproducible aspects of polioencephalomalacia in sheep using amprolium, this may be an useful model for the study of the disease.

Todos os ovinos morreram ou foram eutanasiados in extremis após um curso clínico de 3-7 dias. Os sinais clínicos incluíam depressão, incoordenação, midríase, bruxismo, cegueira e decúbito com opistótono e movimentos de pedalagem. Salivação excessiva e posição de cavalete foi observada em um ovino e mioclonias em um outro. Os principais achados de necropsia restringiam-se ao sistema nervoso central e incluíam tumefação do encéfalo com achatamento dos giros telencefálicos e hemorragias nos lobos parietal e occipital do telencéfalo; as hemorragias ocorriam também nas áreas submeníngeas da medula espinhal e do mesencéfalo. Histologicamente, havia necrose segmentar laminar de neurônios (neurônios vermelhos) associada a edema, tumefação de células endoteliais, hemorragias e infiltração por macrófagos espumosos (células gitter). Essas alterações eram mais marcadas nos lobos frontal, parietal e occipital do telencéfalo e havia uma demarcação abrupta entre as lesões e o neurópilo normal adjacente. Adicionalmente, lesões semelhantes, mas menos acentuadas, eram observadas no mesencéfalo, tálamo e hipocampo. Levando em consideração a reproducibilidade regular dos aspectos da polioencefalomalacia em ovinos pela administração de amprólio, esse modelo pode ser útil para o estudo da doença.

[Bovine polioencephalomalacia: epidemiology, clinical signs and distribution of lesions in the brain.] Polioencefalomalacia em bovinos: epidemiologia, sinais clínicos e distribuição das lesões no encéfalo. Pesquisa Veterinária Brasileira 29(7):487-497. Departamento de Patologia, Universidade Federal de Santa Maria, 97105-900, Santa Maria, RS, Brazil. E-mail: claudioslbarros@uol.com.br Thirty one cases of polioencephalomalacia (PEM) diagnosed from 1999-2008 in cattle from the Southern (13 cases) and Midwestern (18 cases) Brazil were studied. Morbidity (0.04%-6.66 %), mortality (0.04%-6.66 %), and lethality (50%-100%) rates were similar in both regions studied. There was no clear association between PEM cases and age, sex or seasonality. Cases occurred mainly in cattle raised at pasture; in the Southern the disease affected mainly young cattle (one-year old or less) while mainly older cattle (three-year-old or older) were affected in the Midwest. Clinical signs more frequently observed included blindness, incoordination, circling, opisthotonus, recumbence and peddling movements. Clinical course varied from 12 hours to 8 days (average three days and a half). In 11 cases no gross changes were observed in the brain. Main gross findings in the brain of remaining cases included congestion with swelling and flattening of gyri, softening and yellow discoloration of cerebral cortex, hemorrhagic foci in the brain stem, cerebellum and telencephalon, and cerebellar herniation. The main histopathological changes were in the cortex of occipital, parietal and frontal telencephalic lobes; however less prominent and less frequently found lesions occurred in the hippocampus, basal nuclei, thalamus, midbrain, and cerebellum. The type of microscopic cortical lesions was consistent in all cases and included segmentar laminar neuronal necrosis (red neurons), spongiosis, swollen of vascular endothelial nuclei, Alzheimer type II astrocytes and infiltration of gitter cells. In 20% of the cases there was mild lymphohistiocytic cellular infiltrate and in 13% of the cases there was mild infiltrate by neutrophils and eosinophils. Additionally, mild to moderate necro-hemorrhagic lesions were observed in 49% of the cases in the basal nuclei, in 39% of the cases in brain stem and in 26% of the cases in the thalamus. Brain lesions were consistently found in the cortical laminae of the occipital, parietal and frontal telencephalic lobes. In such locations, most frequently affected cortical layers both by neuronal necrosis and edema were external and internal granular layers. Both gyri and sulci were equally affected.

E-mail: claudioslbarros@uol.com.br RESUMO.- Trinta e um casos de polioencefalomalacia (PEM) diagnosticados de 1999-2008 em bovinos do Sul (13 casos) e Centro-Oeste (18 casos) brasileiros foram estudados. As taxas de morbidade (0,04%-6,66 %), mortalidade (0,04%-6,66 %) e letalidade (50%-100%) foram semelhantes em ambas as regiões estudadas. Não houve uma associação clara entre os casos de PEM e a idade, sexo dos bovinos e sazonalidade. Os casos ocorreram principalmente em bovinos criados de forma extensiva em pastagem. Na Região Sul a doença afetou principalmente bovinos jovens (um ano de idade ou menos), enquanto que principalmente bovinos mais velhos (3 anos de idade ou mais) foram afetados no Centro-Oeste. Os sinais clínicos mais frequentemente observados incluíram cegueira, incoordenação, andar em círculos, opistótono, decúbito e movimentos de pedalagem. A evolução do quadro clínico variou de 12 horas a 8 dias (media 3 dias e meio). Em 11 encéfalos não foram observadas alterações macroscópicas; as principais alterações macroscópicas nos outros casos incluíam congestão com tumefação e achatamento das circunvoluções, amolecimento e amarelamento do córtex telencefálico, focos de hemorragia no tronco encefálico, cerebelo e telencéfalo e herniação cerebelar. As principais alterações histológicas ocorreram no córtex dos lobos telencefálicos occipital, parietal e frontal; no entanto, lesões menos acentuadas e menos frequentemente observadas ocorreram no hipocampo, núcleos da base, tálamo, mesencéfalo e cerebelo. O tipo de lesão microscópica cortical era consistente em todos os casos e incluía necrose neuronal (neurônio vermelho) laminar segmentar, espongiose, tumefação do núcleo das células endoteliais, astrócitos Alzheimer tipo II e infiltração por células gitter. [Mucosal disease associated with generalized dermatitis in cattle, Mato Grosso do Sul, Brazil.] Doença das mucosas associada à dermatite generalizada em bovinos, Mato Grosso do Sul. Pesquisa Veterinária Brasileira 28(6):285-292. Departamento de Medicina Veterinária, Faculdade de Medicina Veterinária e Zootecnia, Universidade Federal de Mato Grosso do Sul, Av. Senador Filinto Müller 2443, Campo Grande, MS 79070-900, Brazil. E-mail: eqrural@nin.ufms.br This paper reports epidemiological, clinical, pathological and laboratory diagnostic aspects of a form of dermatitis associated with mucosal disease (MD) in cattle. It also focuses on the methods used for identifying persistently infected (PI) animals and on the impact of the disease on fertility and weaning rates in the affected herd. Cases of dermatitis associated with MD were diagnosed in two 12 and 24-month-old Nelore calves belonging to a beef cattle farm that operates the full cycle of production (calving, rearing, finishing) in Mato Grosso do Sul, Midwest Brazil. The clinical signs exhibited by affected cattle 0included slow, progressive weight loss; formation of diffuse skin crusts in multiple body areas; skin dryness; multiple ulcerations on the gums and dorsal surface of the tongue which evolved to longitudinal fissures; formation of keratinized projections; and detachment of hoof horn. In addition, diarrhea affected one animal in the late stage of the disease. Necropsies also revealed longitudinal erosions in the esophagus. Histological examination showed coagulation necrosis foci in esophageal and lingual mucosae, with neutrophil and lymphocyte infiltration. Skin lesions consisted of epidermal coagulation necrosis associated with neutrophil infiltration and hyperkeratosis. In both cases, clinical diagnosis was confirmed by the isolation and identification of cytopathic and noncytopathic biotypes of the bovine viral diarrhea virus (BVDV) and immunohistochemical detection of viral antigens in formalin fixed tissues. Out of 300 cattle that had contact with the affected animals, 38 were found to be seropositive - in high neutralizing titers - to BVDV. Blood samples from 1,025 young animals and 40 bulls from the farm were examined for the presence of BVDV to identify potential PI animals. The virus was isolated from blood of three calves in the initial test and, 12 months later, from two of them which had remained on the farm. Only one of these calves was found to be BVDV-positive by immuno-histochemical testing performed on ear-tissue samples. In the year following the birth of PI calves, the herd underwent decreases in fertility and weaning rates, which later returned to their previous levels. The resulting data demonstrate the presence of BVDV infection in beef herds in Mato Grosso do Sul and provide evidence as to include the disease in the differential diagnosis of causes of generalized dermatitis in cattle.

#46 - Intoxicação de bovinos por Tetrapterys multiglandulosa (Malpighiaceae) em Mato Grosso do Sul

[Poisoning of cattle by Tetrapterys multiglandulosa in Mato Grosso do Sul, Brazil.] Intoxicação de bovinos por Tetrapterys multiglandulosa (Malpighiaceae) em Mato Grosso do Sul. Pesquisa Veterinária Brasileira 26(3):139-146. E-mail: eqrural@nin.ufms.br Two outbreaks of poisoning by Tetrapterys multiglandulosa in cattle and the experimental reproduction of the toxicosis in sheep are described. Both outbreaks occurred on the same farm in the municipality of Bataiporã, state of Mato Grosso do Sul, Brazil. The first outbreak occurred in July-October 2004 and involved a cattle population at risk of 290 pregnant cows, which were introduced into a 60 hectare pasture with a legal reservation area heavily infested by T. multiglandulosa. Of these, 230 cows (79.3%) aborted, had stillbirths or delivered weak calves that died few days after birth. Seven cows died, and one cow and a 10-day-old calf were necropsied. The second outbreak occurred in September-October 2005, 40 days after 285 2-year-old heifers were introduced into the same pasture infested by T. multiglandulosa and where the first outbreak had occurred in the previous year. Nine heifers got sick and died, and three of then were necropsied. Clinical signs of affected cattle, including a 10-day-old calf, were marked lethargy, loss of weight with distension of the abdomen (ascites), subcutaneous dependant edema, distended and pulsating jugular veins, dyspnea and cardiac arrhythmia. Necropsy findings included a round and dilated heart with whitish and firm areas in the myocardium, and changes related to cardiac failure such as cavitary edema, nutmeg liver, pulmonary edema, a large blood clot in the left ventricle. Histopathological changes included necrosis and fibrosis in the myocardium, chronic passive hepatic centrolobular congestion, pulmonary edema, and spongy degeneration in the white matter of the brain. Experimental sheep died 29 (Sheep 1) and 35 (Sheep 2) days after being fed average daily doses of T. multiglandulosa corresponding to 14g/kg (Sheep 1) and 7,5 g/kg (Sheep 2) per day. Clinical signs were observed from the 7th day (Sheep1) and the 4th day (Sheep 2) of the experiment and included tachycardia, arrhythmia, lethargy and head pressing. Necropsy and histopathologic findings in both experimental sheep were very similar to those observed in affected cattle of the two spontaneous outbreaks.

E-mail: eqrural@nin.ufms.br Two outbreaks of poisoning by Tetrapterys multiglandulosa in cattle and the experimental reproduction of the toxicosis in sheep are described. Both outbreaks occurred on the same farm in the municipality of Bataiporã, state of Mato Grosso do Sul, Brazil. The first outbreak occurred in July-October 2004 and involved a cattle population at risk of 290 pregnant cows, which were introduced into a 60 hectare pasture with a legal reservation area heavily infested by T. multiglandulosa. Of these, 230 cows (79.3%) aborted, had stillbirths or delivered weak calves that died few days after birth. Seven cows died, and one cow and a 10-day-old calf were necropsied. The second outbreak occurred in September-October 2005, 40 days after 285 2-year-old heifers were introduced into the same pasture infested by T. multiglandulosa and where the first outbreak had occurred in the previous year. Nine heifers got sick and died, and three of then were necropsied. Clinical signs of affected cattle, including a 10-day-old calf, were marked lethargy, loss of weight with distension of the abdomen (ascites), subcutaneous dependant edema, distended and pulsating jugular veins, dyspnea and cardiac arrhythmia. From a total population at risk of 1,359 cattle, 54 1-18-month-old calves from both sexes and several breeds were affected and 50 died spontaneously or were euthanatized while moribund. The highest frequency of cases was in recently weaned calves or calves submitted to other stressing factors. General rates of morbidity, mortality and lethality were respectively 3.97, 3.67 and 92.59%. Clinical courses varied from 3-10 days and included depression, nasal and ocular discharge, grinding of teeth, circling, blindness, fever, nistagmus, trembling, anorexia, dysphagia, drooling, incoordination, head pressing, rough hair coat, tachycardia, tachypnea, abdominal pain, melena, falls, recumbency, opisthotonus, convulsions and paddling. Nineteen calves were necropsied. Necropsy findings were characterized by hyperemia of leptomeninges, swollen rostral portions of the telencephalon, and flattening of frontal lobes gyri; frequently in these frontal areas there were segmental brown-yellow discoloration and softening (malacia) of the cortex. In cases with more protracted clinical courses there were extensive swelling, softening and hemorrhaging of the telencephalic frontal lobes. Microscopically, all affected cattle had a necrotizing non-suppurative meningoencephalitis with variable distribution among the 19 cases and among the various telencephalic regions of the same case. The severity of these changes were more marked, in decreasing order of intensity, in the telencephalic frontal cortex, basal ganglia (nuclei), thalamus, brain stem, parietal telencephalic cortex, occipital telencephalic cortex and cerebellum. Perivascular inflammatory infiltrate consisted predominantly of lymphocytes, plasm cells, and less frequently of neutrophils. Additional microscopic findings included variable degrees of gliosis, edema, neuronal necrosis in the telencephalic cortex characterized by shrinking and eosinophilia of perikaria and nuclear picnosis (red neuron); basophilic intranuclear inclusion bodies in astrocytes and neurons (21.05% of the cases); sattelitosis; and neuronophagia. [Polioencephalomalacia in cattle in the states of Mato Grosso do Sul and São Paulo.] Polioencefalomalacia em bovinos nos estados de Mato Grosso do Sul e São Paulo. Pesquisa Veterinária Brasileira 20(3):119-125. Depto Patologia, Fac. Veterinária, Universidade Federal de Pelotas, 96010-900 Pelotas, RS, Brazil. Thirty outbreaks of polioencephalomalacia (PEM) were diagnosed from August 1993 to October 1997. Twenty nine occurred in the state of Mato Grosso do Sul and one in São Paulo. The disease affected cattle from 4 months to 7 years of age. Morbidity rates ranged from 0.02% to 14.28% and case fatalities from 42.5 % to 100%. The disease was not seasonal. All outbreaks occurred in cattle grazing Brachiaria spp, except one affecting feedlot cattle. Only neurologic signs were observed, and the course of the disease varied from 12 hours to 4 days in untreated cattle. Most animals treated with thiamine anel dexamethazone recovered. Moderate cerebral edema was observed at necropsy. Cell changes were characterized by laminar necrosis and gitter cells. In two cases moderate cortical anel submeningeal hemorrhages were observed. Nine cases had perivascular anel submeningeal infiltrations by eosinophils. Sodium ion concentration of the cerebrospinal fluid was elevated in one case. The cause of PEM in the region was not determined, but the results suggest that in some cases sodium chloride intoxication/water deprivation could play a role in the etiology of the disease. PEM represents 4.78% of cattle disease diagnosed at the Pathology Laboratory of the Federal University of Mato Grosso do Sul.

Pesquisa Veterinária Brasileira 20(3):119-125. Depto Patologia, Fac. Veterinária, Universidade Federal de Pelotas, 96010-900 Pelotas, RS, Brazil. Trinta focos de polioencefalomalacia (PEM) foram diagnosticados no período de agosto de 1993 a outubro de 1997. Vinte e nove focos ocorreram no estado de Mato Grosso do Sul (MS) e um no estado de São Paulo (SP). Foram afetados bovinos de 4 a 84 meses de idade. A morbidade dos rebanhos afetados variou ele 0,02% a 14,28% e a letalidade de 42,5% a 100%. A doença não apresentou uma sazonalidade e ocorreu em vários municípios do MS. Em todos os focos estudados os animais afetados eram criados em regimes de criação extensiva, com exceção de um foco. Os sinais clínicos observados foram exclusivamente nervosos e a evolução dos casos variou de 12 horas a 4 dias. A maioria dos animais tratados com tiamina e dexametasona recuperou-se. Histologicamente, as lesões consistiam de necrose laminar do córtex cerebral. Adicionalmente em 2 casos observaram-se hemorragias submeningeanas e corticais, e em 9 casos presença ele infiltrado de eosinófilos. A dosagem de sódio no líquor apresentou-se elevada em um caso. A etiologia da PEM não está esclarecida, porém em alguns casos a intoxicação por cloreto de sódio/privação de água pode estar envolvida na etiologia da enfermidade. A PEM representa 4,78% dos casos ele enfermidades de bovinos diagnosticadas no Laboratório de Anatomia Patológica da UFMS. Com bases nos resultados, verifica-se a importância da enfermidade no diagnóstico diferencial de outras doenças com quadro clínico neurológico no MS, principalmente a raiva, a meningoencefalite por herpesvírus bovino tipo 5 (HVB-5) e o botulismo.
