E-mail: phelipe.favaron@yahoo.com.br Understanding of the reproductive function is essential for both, the establishment of appropriate management systems, and for the use of new species as animal models. In this study, we used light and electron microscopy to characterize the sexual development stages of the guinea pig (Cavia porcellus) in specimens of 30, 45 and 90 days of age. We observed the differentiation of spermatocytes only through transmission electron microscopy in the leptotene, zygotene and pachytene phases of meiosis, in 30-day-old animals. During puberty, there was differentiation of the germinative epithelium and formation of the acrosome. Spermatozoa, however, were not detected. Thus, we could infer that puberty happens after 45 days of age. Sexual maturity was evident in 90-day-old specimens. Our results showed that changes in the testicular germinative epithelium during the postnatal sexual development in guinea pig led to morphological changes, including the ones related to the development of Leydig and Sertoli cells, which are directly related to puberty. In this work, we provide new morphological subsidies for a better understanding of reproductive parameters of this species, enabling its use as an animal model in the field of the reproductive biology.

E-mail: phelipe.favaron@yahoo.com.br A função reprodutiva é um fator de vital compreensão tanto para o estabelecimento de sistemas apropriados de manejo, quanto para o uso de novas espécies como modelos animais. Neste estudo através da microscopia de luz e eletrônica caracterizou-se a fase de desenvolvimento sexual do porquinho-da-Índia (Cavia porcellus) em espécimes de 30, 45 e 90 dias de desenvolvimento. Nos animais de 30 dias, a diferenciação dos espermatócitos foi visualizada somente na microscopia eletrônica de transmissão em leptóteno, zigóteno e paquíteno. Durante a puberdade, houve diferenciação do epitélio germinativo, formação do acrossoma, porém não foram evidenciados espermatozóides, assim, infere-se que a puberdade acontece a partir dos 45 dias de idade. A maturidade sexual foi evidente aos 90 dias de idade. Nossos resultados mostraram que ao longo do desenvolvimento sexual pós-natal do porquinho-da-Índia, mudanças no epitélio germinativo testicular levam há alterações morfológicas, inclusive com relação ao desenvolvimento das células de Sertoli e de Leydig, as quais estão diretamente relacionadas com a puberdade. Assim, novos subsídios morfológicos são fornecidos para um melhor entendimento dos parâmetros reprodutivos desta espécie, a fim de viabilizar sua utilização como modelo animal no campo da biologia reprodutiva.

Fifty-seven rats were divided into six groups, having 6 to 11 rats. Food was provided ad-libitum until three months and then the first group was considered without starvation (day 0). Other rats were fasted for two weeks. Group 2 was applied to a group immediately after starvation (day 14). Groups 3 to 6 were refed in days 16 till 22, respectively. At the end of each period, blood and tissue samples were taken and histopathological and serum analysis, including serum electrolytes (calcium, phosphorus, sodium, potassium), the energy parameters (glucose, insulin, cortisol) and the liver enzymes (ALT, AST, ALP) were determined. Insulin decreased by starvation and then showed an increasing trend compared to starvation period, which the highest amount of this parameter was observed eight days post-refeeding. Serum glucose level showed the opposite pattern of insulin. Histopathological examination of the tissue sections revealed clear vacuoles after starvation and refeeding, in which the severity of lesions gradually decreased during refeeding. The cortisol level decreased by starvation and then increased during refeeding. Also, potassium and phosphorus concentrations declined by refeeding and the serum sodium and potassium levels were changed in the relatively opposite manner. The calcium level decreased by starvation and then increased during refeeding. These results could help recognize and remedy the refeeding syndrome.

E-mail: bebel@ufv.br Osteoporosis is a multifactorial disease of high prevalence and has great impact on quality of life, because the effects on bone structure increase the risk of fractures, what may be very debilitating. Based on the observation that patients with depression have lower bone mineral density than healthy individuals, many studies have indicated that stress could be an aggravating factor for bone loss. This study evaluates the effect of a protocol of chronic mild stress (CMS) on parameters of bone assessment in male and female rats. Five 5-monh-old rats of each sex underwent a schedule of stressor application for 28 days. Stressors included cold, heat, restraint, cage tilt, isolation, overnight illumination, and water and food deprivation. Five rats of each sex were kept under minimum intervention as control group. The animals were weighed at beginning and end of the period, and after euthanasia had their bones harvested. Femur, tibia and lumbar vertebrae were analyzed by bone densitometry. Biomechanical tests were performed in femoral head and diaphysis. Trabecular bone volume was obtained from histomorphometric analysis of femoral head and vertebral body, as well as of femoral midshaft cross-sectional measures. Not all parameters analyzed showed effect of CMS. However, tibial and L4 vertebral bone mineral density and cross-sectional cortical/medullar ratio of femoral shaft were lower in female rats submitted to the CMS protocol. Among male rats, the differences were significant for femoral trabecular bone volume and maximum load obtained by biomechanical test. Thus, it could be confirmed that CMS can affect the balance of bone homeostasis in rats, what may contribute to the establishment of osteopenia or osteoporosis.

Com base na observação de que pacientes com depressão têm menor densidade mineral óssea que indivíduos saudáveis​​, muitos estudos têm indicado que o estresse pode ser um fator agravante para a perda óssea. Este estudo avalia o efeito de um protocolo de estresse moderado crônico (EMC) em parâmetros de avaliação óssea em ratos machos e fêmeas. Cinco animais de cada sexo, com cinco meses de idade, foram submetidos a um cronograma de aplicação de estressores durante 28 dias. Os estressores incluídos foram: frio, calor, contenção, inclinação da gaiola, isolamento, iluminação durante a noite e privação de água e ração. Cinco animais de cada sexo foram mantidos com um mínimo de intervenção como grupo controle. Os animais foram pesador no início e no final do período, e após eutanásia tiveram seus ossos coletados. Fêmur, tíbia e vértebra lombar foram analisados por densitometria óssea. Testes biomecânicos foram realizados na cabeça e na diáfise do fêmur. Volume trabecular ósseo foi obtido a partir de análise histomorfométricas da cabeça do fêmur e do corpo vertebral, bem como medidas da seção transversal diáfise femoral. Nem todos os parâmetros avaliados sofreram efeito do protocolo de EMC. No entanto, a densidade mineral óssea da tíbia e da vértebra L4 e a razão osso cortical/medula da seção transversal da diáfise femoral foram menores nas fêmeas submetidas ao protocolo. Entre os ratos machos, as diferenças foram significativas no volume trabecular ósseo da cabeça femoral e na carga máxima obtida no teste biomecânico. Assim, confirma-se que o protocolo de EMC pode afetar o equilíbrio da homeostase óssea em ratos, o que pode contribuir para o estabelecimento de osteopenia ou osteoporose.

E-mail: anaflaviamabo@gmail.com The purpose of this study is to evaluate healthy conscious guinea pigs as a model for electrophysiology assessment and to describe normal electrocardiographic patterns in controlled laboratory environment, establishing the best QT formula for this method. Electrocardiographic recordings of fifty adult conscious guinea pigs were obtained using a computerized electrocardiography. The electrocardiographic measurements of three different tracings were analyzed. The results obtained established normal mean and range values for the parameters: heart rate, waves and intervals of P-QRS-T deflections, as well as the mean cardiac axis. Groups were separated by body weight: group 1 gathered animals with 500-699g and group 2 with animals 700-900g. No differences were found when measurements were compared between groups, showing no significant difference between weight/body sizes to the electrocardiographic parameters (P<0.05). The mean corrected QT values (QTc) obtained using diverse formulae were significantly different (P<0.05), were the most consistent was Van der Water (QTcV). QTcV values were strongly correlated (r=98) and 95% confidence interval 185.7 to 195.2ms.Considering its simplicity and reliability, the QTcV was deemed the most appropriate to be used for the correction of QT interval in conscious guinea pigs.The results of this study also suggest that the values found can be used as reference for the species.

E-mail: anaflaviamabo@gmail.com O objetivo desse estudo foi avaliar cobaios hígidos e conscientes como modelos para estudos de eletrofisiologia e descrever os padrões eletrocardiográficos normais em ambiente laboratorial controlado, estabelecendo a melhor fórmula QT para esse método. Gravações eletrocardiográficas de cinquenta cobaios adultos conscientes foram obtidas usando eletrocardiografia computadorizada. As medidas eletrocardiográficas de três trechos diferentes foram analisadas. Os resultados estabeleceram média e desvio padrão para os parâmetros: frequência cardíaca, ondas e intervalos P-QRS-T, assim como o eixo cardíaco médio. Grupos foram separados de acordo com o peso: grupo 1 incluiu animais com 500-699g e o grupo 2 animais de 700-900g. Nenhuma diferença foi encontrada quando as medidas foram comparadas entre os grupos, mostrando que não há diferença significativa entre peso/tamanho corporal com os parâmetros eletrocardiográficos (p<0.05). As médias corrigidas dos valores do QT (QTc) obtidas usando diferentes fórmulas foram significativamente diferentes (p<0.05), sendo a mais consistente a de Van der Water (QTcV). Valores de QTcV fortemente correlacionam com o QT (r=98), com intervalo de confiança a 95% de 185.7 a 195.2 ms. Considerando a simplicidade e confiabilidade, o QTcV foi considerado apropriado para correção do intervalo QT em cobaios conscientes. Os resultados do presente estudo também sugerem que os valores encontrados possam ser utilizados como referência para essa espécie.

E-mail: tharcia_kiara@hotmail.com The aim of this study was to evaluate the effect of Aqueous Extract from Peanut (EAA) in weight, serum biochemistry and liver histology of Wistar rats with normal hearing and a high fat diet. The survey was conducted using 40 male Wistar rats divided into four groups (n=0): GA (high fat diet), GB (fat diet+EAA), GC (normolipídica diet), and GD (normolipídica diet+EAA). After 8 weeks, the rats were euthanized and blood samples were collected to evaluate biochemical data (total cholesterol and its fractions, triglycerides, urea, creatinine, AST, ALT and glucose) and liver fragments for histological analysis. The animals of the GB group had a lower weight gain when compared with GA (XGB versus XGA= p<0.05), but CG and GD did not obtain statistical differences. The rats that received EAA had a reduction in cholesterol levels (XGB= versus XGA= p<0.05 vs. XGA and XGD= p<0.01), triglycerides (=XGB versus XGA and =XGD versus XGA= p<0,001) and more discreetly ALT levels. Blood glucose, urea and creatine remained within the reference values. Liver samples analyzed, the rats of different groups showed no histopathological changes. In conclusion, the EAA had preventive effects on weight gain and dyslipidemia.

Após 8 semanas, os animais foram eutanasiados e foram coletadas amostras sanguíneas para a avaliação de dados bioquímicos (Colesterol total e suas frações, triglicerídeos, uréia, creatinina, AST, ALT e glicemia) e fragmentos do fígado para análise histológica. Os animais do grupo GB tiveram um ganho de peso inferior quando comparados ao GA (XGB= versus XGA= p<0,05), já os grupos GC e GD não obtiveram diferenças estatísticas. Os animais que receberam o EAA tiveram uma redução nos níveis de colesterol (XGB= versus XGA= p<0,05 e XGD= versus XGA= p<0,01), dos triglicerídeos (XGB= versus XGA e XGD= versus XGA= p<0,001) e mais discretamente dos níveis de ALT. A glicemia, uréia e creatina permaneceram dentro dos valores de referência. As amostras hepáticas analisadas, dos ratos dos diferentes grupos, não apresentaram alterações histopatológicas. Conclui-se que O EAA apresentou efeitos preventivos sobre o ganho ponderal e dislipidemia.


#46 - Bacterial cellulose and bacterial cellulose/polycaprolactone composite as tissue substitutes in rabbits’ cornea, 36(10):986-992

ABSTRACT.- Sepúlveda R.V., Valente F.L., Reis E.C.C., Araújo F.R., Eleotério R.B., Queiroz P.V.S. & Borges A.P.B. 2016. Bacterial cellulose and bacterial cellulose/polycaprolactone composite as tissue substitutes in rabbits’ cornea. Pesquisa Veterinária Brasileira 36(10):986-992. Programa de Pós-Graduação em Medicina Veterinária, Departamento de Veterinária, Universidade Federal de Viçosa, Avenida Peter Henry Rolfs s/n, Viçosa, MG 36570-900, Brazil. E-mail: andrea@ufv.br In order to test the performance of bacterial cellulose/polycaprolactone composite (BC/PCL) and pure bacterial cellulose (BC) as tissue substitutes in rabbits’ cornea, a superficial ulcer containing 5mm in diameter and 0.2mm deep was made in the right cornea of 36 rabbits, then a interlayer pocket was created from the basis of this ulcer. Twelve rabbits received BC/PCL membrane and 12 were treated with BC membranes, both membranes with 8mm in diameter. The remaining rabbits received no membrane constituting the control group. The animals were clinically followed up for 45 days. Three animals of each group were euthanized at three, seven, 21, and 45 days after implantation for histological examination of the cornea along with the implant. Clinical observation revealed signs of moderate inflammatory process, decreasing from day 20th in the implanted groups. Histology showed absence of epithelium on the membranes, fibroplasia close to the implants, lymph inflammatory infiltrate with giant cells, collagen disorganization, with a predominance of immature collagen fibers in both groups with implants. Although inflammatory response is acceptable, the membranes used does not satisfactorily played the role of tissue substitute for the cornea during the study period.

E-mail: andrea@ufv.br Com objetivo de testar o desempenho do compósito celulose bacteriana/policaprolactona (CB/PCL) e da celulose bacteriana pura (CB) como substitutos teciduais em córnea de coelhos, foi realizada uma úlcera superficial de 5 mm de diâmetro e 0,2 mm de profundidade na córnea direita de 36 coelhos, criando-se um bolso interlamelar a partir da base dessa úlcera. Doze animais receberam a membrana do compósito CB/PCL e 12 foram tratados com membranas de CB, ambas com 8 mm de diâmetro, os coelhos restantes não receberam nenhuma membrana, constituindo o grupo controle. Os animais foram acompanhados clinicamente até 45 dias. Três animais de cada grupo sofreram eutanásia aos três, sete, 21 e 45 dias após o implante das membranas para análise histológica da córnea juntamente com o implante. À observação clínica, houve sinais de processo inflamatório moderado, diminuindo a partir do 20º dia nos grupos implantados. A histologia demonstrou ausência de epitélio sobre as membranas, fibroplasia próxima aos implantes, infiltrado inflamatório linfo-histiocitário com células gigantes, desorganização do colágeno, com predominância de fibras imaturas de colágeno em ambos os grupos com implantes. Embora a resposta inflamatória seja aceitável, as membranas utilizadas não desempenharam satisfatoriamente o papel de substituto tecidual para a córnea, no período estudado.

E-mail: felipp.sf@gmail.com Cardiac biomarkers for clinical and experimental heart diseases have previously been evaluated in rabbits. However, several laboratory assays performed and reported with inconsistent results. This study aimed to assess the effects of breed on serum ANP, CRP, and ACE and establish reference interval (RI) for these biomarkers in a large population of healthy rabbits. Ninety-seven adult rabbits from five breeds were included in this study. Assays were performed using specific ELISA commercial kits. The results were statistically analyzed using ANOVA, Tukey test (p<0.05), arithmetic mean, RI of mean, and standard deviation. A significant effect of breed was shown, indicating different RI between breeds for each biomarker. In conclusion, this study demonstrated that breed is an important physiological variable influencing the normal values of cardiac markers in healthy rabbits.

Entretanto diferentes testes laboratoriais têm sido utilizados e relatados, sem uma confluência de resultados. Os objetivos deste estudo foram verificar os efeitos de diferentes raças de coelhos sobre as concentrações séricas de ANP, CRP e ACE, além de estabelecer intervalor de referência para estes biomarcadores em uma população de coelhos saudáveis. Foram utilizados noventa e sete coelhos de cinco diferentes raças. Os exames foram realizados pela metodologia de ELISA, por meio de kits comerciais específicos. Os resultados foram analisados estatisticamente os testes de ANOVA e Tukey (p<0,05), média aritmética, intervalo de referência da média e desvio padrão. Um efeito significativo da raça foi observado sobre as variáveis estudadas, indicando diferentes intervalos de referência entre as raças para cada biomarcador. Em conclusão, este estudo demonstrou que a raça é uma variável fisiológica importante que influencia os valores normais destes biomarcadores em coelhos saudáveis.

E-mail: fernanda_fmos@hotmail.com This study describes the development of the central nervous system in guinea pigs from 12th day post conception (dpc) until birth. Totally, 41 embryos and fetuses were analyzed macroscopically and by means of light and electron microscopy. The neural tube closure was observed at day 14 and the development of the spinal cord and differentiation of the primitive central nervous system vesicles was on 20th dpc. Histologically, undifferentiated brain tissue was observed as a mass of mesenchymal tissue between 18th and 20th dpc, and at 25th dpc the tissue within the medullary canal had higher density. On day 30 the brain tissue was differentiated on day 30 and the spinal cord filling throughout the spinal canal, period from which it was possible to observe cerebral and cerebellar stratums. At day 45 intumescences were visualized and cerebral hemispheres were divided, with a clear division between white and gray matter in brain and cerebellum. Median sulcus of the dorsal spinal cord and the cauda equina were only evident on day 50. There were no significant structural differences in fetuses of 50 and 60 dpc, and animals at term were all lissencephalic. In conclusion, morphological studies of the nervous system in guinea pig can provide important information for clinical studies in humans, due to its high degree of neurological maturity in relation to its short gestation period, what can provide a good tool for neurological studies.

No total, 41 embriões e fetos foram analisados macroscopicamente e por microscopia de luz e eletrônica. O fechamento do tubo neural foi observado no dia 14 e o desenvolvimento da medula espinhal e diferenciação das vesículas primitivas do sistema nervoso central foram observados no dia 20. Histologicamente, o tecido cerebral indiferenciado foi observado como uma massa de tecido mesenquimal entre os dias 18 e 20 e no 25º dia o tecido no interior do canal medular apresentou maior densidade. No dia 30 o tecido cerebral apresentou-se diferenciado, período no qual a medula espinhal preenchia todo o canal vertebral e foi possível observar os estratos cerebral e cerebelar. No dia 45 as intumescências cervical e lombar foram visualizadas e os hemisférios cerebrais estavam divididos, com uma clara distinção entre substância branca e cinzenta no cérebro e cerebelo. O sulco mediano dorsal da medula espinhal e a cauda equina foram evidentes apenas no dia 50. Não houve diferenças estruturais significativas em fetos de 50 e 60 dpc e animais a termo eram todos lisencefálicos. Estudos morfológicos do sistema nervoso em guinea pig podem fornecer informações importantes para estudos clínicos em seres humanos devido ao alto grau de maturidade neurológica em relação ao seu período de gestação curto, fato que servir como excelente ferramenta em estudos neurológicos.

E-mail: kellyroballo@gmail.com The olfactory epithelium (OE) is a promising source of stem cells (OESC) for therapeutic use in veterinary and human medicine, especially in diseases correlated with the peripheral (spinal cord) and central (brain and brainstem) nervous system (CNS), because of its ability to differentiate into neurons, astrocytes and oligodendrocytes cells. In humans, OESC has been used primarily in therapeutic trials for degenerative diseases such as Alzheimer and Parkinson. In animals, the frequency of corresponding cases of chronic or acute neurodegenerative diseases is very low, because of the difficulty of a definitive diagnosis; thus, the focus of cell therapy research are mostly mechanical spinal cord injuries. Due to the lack of normalization and selection of the best methodologies for comparative studies, this review aims to analyze recent reports on the potential use of stem cells from the olfactory epithelium in cell therapies and to discuss the main challenges and future prospects in veterinary medicine.

E-mail: kellyroballo@gmail.com O epitélio olfatório (EO) é uma fonte promissora de células-tronco (CTEO) para o uso terapêutico na medicina veterinária e humana, especialmente em doenças correlacionadas com o sistema nervoso periférico (medula espinhal) e central (cérebro e tronco encefálico) , pois as CTEO possuem a capacidade de se diferenciar em células do sistema nervoso, tais como: neurônios, oligodendrócitos e astrócitos. Em humanos estas células são utilizadas em ensaios terapêuticos de doenças degenerativas como o Alzheimer e Parkinson. Em animais a casuística relativa das doenças neurodegenerativas crônicas ou agudas é baixa, devido à dificuldade de diagnóstico definitivo, desta forma o enfoque das pesquisas com terapia celular são em sua grande maioria em lesões mecânicas na medula espinhal. Devido à falta de padronização e seleção das melhores metodologias que permitam confrontação de estudos, esta revisão busca reunir as mais recentes publicações, descrevendo o potencial uso das células-tronco do epitélio olfatório em terapias celulares, discutindo os principais desafios e perspectivas futuras com enfoque na medicina veterinária.


#50 - Role of the autologous mesenchymal stem cells compared with platelet rich plasma on cicatrization of cutaneous wounds in diabetic mice, 36(7):617-624

Role of the autologous mesenchymal stem cells compared with platelet rich plasma on cicatrization of cutaneous wounds in diabetic mice. Pesquisa Veterinária Brasileira 36(7):617-624. Departamento de Clínica e Cirurgia Veterinária, Universidade Federal do Piauí, Campus Socopo, Teresina, PI 64049-550, Brazil. E-mail: argolo_napoleao@ufpi.edu.br Chronic cutaneous lesions affect 15% of diabetic human patients and represent a risk 15 to 46 times larger of limb amputations compared to people with normal glycemia. It is assumed that half of these amputations could be prevented by early treatment of wounds, for example, with proper cell therapy. Objectives: In this study, the action of the autologous transplant of mesenchymal stem-cells (MSC) was evaluated compared to the treatment with autologous platelet rich plasma (PRP) in the cicatrization of cutaneous lesions induced in diabetic mice. These animals were previously treated with streptozootocin to induce diabetes mellitus and round wounds of 1.5cm in diameter were created in the posterior region. Diameters of the wounds and healing time were evaluated during 30 days and the results were submitted to variance analysis and Tukey’s test average. It was noticed that the animals treated with MSC presented a more accelerated cicatrization of the cutaneous lesion than the animals treated with PRP. However, the treatment with PRP presented better results than just the daily asepsis of the lesions with saline or covering them with semi-permeable bandage. Besides, the use of semi-permeable bandage kept the cutaneous lesions of diabetic mice did not interfere negatively with cicatrization, proved to be harmless to use, but kept the cutaneous lesions more hydrated than the ones exposed to the environment.

Supõe-se que a metade destas amputações poderia ser evitada por meio do tratamento precoce das feridas cutâneas com, por exemplo, uma adequada terapia celular. Objetivos: Neste estudo, a ação do transplante autólogo de células estaminais mesenquimais (MSC) foi avaliada em comparação com o tratamento com plasma rico em plaquetas autólogo (PRP) na cicatrização de lesões cutâneas induzidas em camundongos diabéticos. Estes animais foram previamente tratados com estreptozotocina para induzir diabetes mellitus e feridas redondas de 1,5 cm de diâmetro foram criadas na região posterior. Os diâmetros dos ferimentos e tempo de cicatrização foram avaliados durante 30 dias e os resultados foram submetidos à análise de variância e média pelo teste de Tukey. Verificou-se que os animais tratados com MSC apresentam uma cicatrização mais acelerada da lesão cutânea que do que os animais tratados com PRP. No entanto, o tratamento com PRP apresentou melhores resultados do que apenas a assepsia das lesões diariamente com solução salina ou cobrindo-os com atadura semi-permeável. Além disso, a utilização de atadura semi-permeável mantidas as lesões cutâneas de camundongos diabéticos não interfere negativamente com a cicatrização, provou ser inofensiva para usar, mas manteve as lesões cutâneas hidratadas mais do que os expostos ao meio ambiente.
