#51 - Expression and distribution of connexin 32 in rat liver with experimentally induced fibrosis, p.353-357

Abstract in English:

ABSTRACT.- Santos-Rodrigues A., Dagli M.L.Z., Avanzo J.L., Moraes H.P., Mackowiak I.I. & Hernandez-Blazquez F.J. 2009. Expression and distribution of connexin 32 in rat liver with experimentally induced fibrosis. Pesquisa Veterinária Brasileira 29(4):353-357. Departamento de Cirurgia, Setor de Anatomia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: alexsantos@usp.br The connexin 32 (Cx32) is a protein that forms the channels that promote the gap junction intercellular communication (GJIC) in the liver, allowing the diffusion of small molecules through cytosol from cell-to-cell. Hepatic fibrosis is characterized by a disruption of normal tissue architeture by cellular lesions, and may alter the GJIC. This work aimed to study the expression and distribution of Cx32 in liver fibrosis induced by the oral administration of dimethylnitrosamine in female Wistar rats. The necropsy of the rats was carried out after five weeks of drug administration. They presented a hepatic fibrosis state. Sections from livers with fibrosis and from control livers were submitted to immunohistochemical, Real Time-PCR and Western-Blot analysis to Cx32. In fibrotic livers the Cxs were diffusely scattered in the cytoplasm, contrasting with the control livers, where the Cx32 formed junction plaques at the cell membrane. Also it was found a decrease in the gene expression of Cx32 without reduction in the protein quantity when compared with controls. These results suggest that there the mechanism of intercellular communication between hepatocytes was reduced by the fibrotic process, which may predispose to the occurrence of a neoplastic process, taken in account that connexins are considered tumor suppressing genes.

#52 - Monitoring bovine viral diarrhea virus (BVDV) infection status in dairy herds, p.588-592

Abstract in English:

ABSTRACT.- Diéguez F.J., Yus E., Sanjuán M.L., Vilar M.J. & Arnaiz I. 2008. Monitoring bovine viral diarrhea virus (BVDV) infection status in dairy herds. Pesquisa Veterinária Brasileira 28(12):588-592. Unidad de Epidemiología y Sanidad Animal, Instituto de Investigación y Análisis Alimentarios, Facultad de Veterinaria s/n, Lugo 27002, Spain. E-mail: franciscojavier.dieguez@usc.es This study was designed to assess the relationship between antibodies against bovine viral diarrhea virus (BVDV) determined in the bulk tank milk (BTM) and the within-herd seroprevalence. We also assessed the efficiency of measuring antibody levels in BTM samples to monitor BVDV infection status in a herd. In the 81 farms included in the study, BTM samples were obtained and blood samples withdrawn from all cattle older than one year. The infection status was then determined in serum and milk using a commercial blocking ELISA based on the detection of anti-p80 antibodies. Apart from these baseline serum and milk samples, another BTM sample was collected from each herd 9 months later, and a third BTM sample obtained 9 months after this. In these second and third milk samples, anti-BVDV antibodies were determined using the same ELISA kit. Statistical tests revealed good agreement between herd seroprevalences (% seropositive animals in the herd) and the antibody levels detected in the BTM samples. During the 18 months of follow-up, the farms with persistently infected cattle at the study outset (14.8% of the herds) showed a significant decrease in BTM antibody titers after virus clearance. Conversely, a significant increase in BTM antibody levels was observed in the herds infected with BVDV during the follow-up period. Our findings indicate that monitoring antibody levels in the BTM is a useful method of identifying changes in the BVDV infection status of a herd.

#53 - Alterações nos parâmetros hematológicos de Gallus gallus domesticus experimentalmente infectados por Borrelia anserina, p.527-532

Abstract in English:

ABSTRACT.- Lisbôa R.S., Guedes Júnior D.S., Silva F.J.M., Cunha N.C., Machado C.H. & Fonseca A.H. 2008. [Alterations in hematological parameters of Gallus gallus domesticus experimentally infected with Borrelia anserina.] E-mail: raquellisboa@ufrrj.br Avian spirochaetosis is a cosmopolite acute septicemic disease of many avian species, caused by Borrelia anserina Sakharoff, 1891. The present study assesses the estimate of the hematological alterations of Gallus gallus domesticus experimentally infected with B. anserina by vector Argas (Persicargas) miniatus. Twenty-seven fowls of the species G. g. domesticus, 67 days old, were randomly allocated into three groups composed by nine animals each. One group was exposed to B. anserina infected ticks (Group 1), other one to ticks free of this agent (Group 2), and another group not exposed to ticks (Group 3). Blood smears of the fowls were taken daily, since the first day the fowls were exposed to the ticks, up to the 25th day after exposure (DAE). Blood samples were collected three days before exposure, and three, eight and 18 DAE, for hematologic tests. The examination of Group 1 smears revealed large number of spirochaetes. Group 2 and 3 blood smears were negative during the whole period under exam. In agreement with the hematological evaluation results, the fowls exposed to infected ticks showed a normocytic normochromic anemia in eight DAE, leucocytosis with heterophilia and monocytosis concomitant with the spirochaetemia. We concluded that B. anserina infection determined on fowls of Group 1 hematological alterations compatible with bacterial infection of moderate gravity, developing to self-cure, in the experimental conditions established in this study.

Prof. Dr. Orlando Marques de Paiva 87, Bloco 12/14, São Paulo, SP 05508-270, Brazil. E-mail: lilianm@usp.br With the purpose of establishing echocardiographic measurements in Holstein calves, 25 calves, 8 to 28 days of age and body weight ranging from 27 to 57 kg, were used. The echocardiographic examination was proceeded in B and M-modes to obtain the following parameters, in diastole and systole: right ventricle (2.05±0.13cm and 1.59±0.13cm) and left ventricle internal diameter (3.91±0.09cm and 2.52±0.13cm), and interventricular septum (1.24±0.04cm and 1.62±0.06cm) and left ventricle free wall thickness (0.92±0.04cm and 1.50±0.05cm). The values for both left and right atria in systole were 2.97±0.12cm and 4.110.21cm, respectively. The left diastolic (67.90±3.65ml), systolic (25.32±3.05ml) and ejection (42.58±2.46ml) volumes, cardiac output (3857±339ml/min), aortic root diameter (2.52±0.05cm), E-point septal separation (0.65±0.08cm), left ventricle ejection time (0.39±0.02s), fractional shortening (36.27±2.40%) and ejection fraction (64.67±3.22%) were also calculated. There was a mean positive linear correlation (66.4%, P<0.01) between the aortic root diameter and the bodyweight, mean negative linear correlation (P<0.01) heart rate (69.1%) and cardiac output (62.4%). There was a tendency of the calves in between the left ventricle ejection time and presenting a smaller left chamber diameter, although maintained the relationship between myocardial wall thickness and functional indexes.

#55 - Imunoglobulinas no trato respiratório de bezerros sadios durante o primeiro mês de vida, p.487-490

Abstract in English:

ABSTRACT.- Bertagnon H.G., Da Silva P.E.G., Wachholz L., Leal M.L.R., Fernandes W.R. & Benesi F.J. 2007.[Immunoglobulin in the respiratory tract of healthy calves during their first month of life.] Imunoglobulinas no trato respiratório de bezerros sadios durante o primeiro mês de vida. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: febencli@usp.br The immunoglobulin variation profile in lavages from the broncoalveolar and tracheo-bronchial regions of 20 healthy newborn Holstein male calves was studied. They were fed with colostrum and distributed into 2 groups, 10 animals each. Group 1 underwent the nasotracheal catheterization technique to get the bronchoalveolar lavage (BAL), and Group 2 underwent the tracheocenthesis to collect the tracheobronchial lavage (TBL), both procedures being carried out at a 7-day-interval, starting on the first days up to about one month of life. Higher IgG contents, as compared to IgA, were noted across the respiratory tract. These immunoglobulins were impacted by the site of the respiratory tract washed, as well as by the calves’ life time in weeks. Higher immunoglobulin contents were detected in TBL, as well as higher IgM and IgA rates, as compared to BAL. The BAL immunoglobulins showed a tendency to be reduced in TBL.

#56 - Estudo comparativo de éguas repetidoras ou não de cio através da avaliação histológica do endométrio e das concentrações plasmáticas de progesterona, p.506-512

Abstract in English:

ABSTRACT.- Eigenheer-Moreira J.F., Fernandes F.T., Queiroz F.J.R, Pinho T.G.& Ferreira A.M.R. 2007. [Comparative study of repeat breeds and healthy mares through endometrial histology and plasmatic progesterone concentrations.] E-mail: joana.vet@gmail.com.br The study aimed to compare endometrial histology and plasmatic progesterone (P4) concentration of repeat breeds and healthy mares. The hypothesis was that there is a correlation between infertility and endometrial histology and P4 concentration in both groups. A total of 36 Campolina and Mangalarga Marchador mares in reproductive age (3-23 years) were used, 11 of them were healthy mares (Control group, 7 embryo recipient and 4 embryo donors), and 25 repeat breeders (10 embryo recipient and 15 embryo donors), classified as based on their reproductive history. Endometrial and blood samples were collected for respectively histological and plasma progesterone concentration evaluation. The endometrial samples obtained after biopsy were fixed in Bouin’s fluid, processed, included in paraffin, and stained with Hematoxylin-Eosin (HE) for histopathological examination. Plasmatic progesterone concentrations were evaluated by enzyme immunoessay (ELISA). There was no correlation between progesterone concentration and fertility. But there was a positive correlation between age and fertility, as older mares had major tendency of subfertility than younger ones. There was also a correlation between biopsy categories and fertility, as more histological alterations were found, higher were the chances for the mares to be subfertile. However not all mares classified as Category I and II maintained pregnancy until parturition. Other factors could influence pregnancy maintenance. In the same way, not all mares in Category III were infertile. The endometrial biopsy was shown to be an easy and cheap diagnostic technique with minimal discomfort to the animals and, together with other data, to be a very important component in the investigation of mare fertility.

Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: juvet@usp.br Herbst corpuscles of the palatine mucosa of ostrich were studied by light microscopy. The corpuscles are composed of an outer core, inner core and central nerve terminal. The outer core presents numerous lamellae, while the inner core shows compact structure of cytoplasm sheets. The corpuscles are elongate or oval in shape and are surrounded by bundles of collagen fibers. Each lamella is composed of a dense network of thick fibrils. The terminal axons are located along the axis and form a bulb terminal. The fibers of external core stained by Picrosirius and examined by polarized light microscopy revealed to be green in color like type I collagen fibers, and at the periphery is a large amount of collagen type III. The corpuscles are surrounded by flat cells and dense collagen fibers at the periphery.

E-mail: santanafjf@yahoo.com Six outbreaks of oropharyngeal lesions caused by drenching guns in sheep are reported. In 2 outbreaks in Rio Grande do Sul the morbidity was 6.25% and 15%, and the lesions observed after antihelmintic treatment in adult sheep were characterized by a fistula extending from mouth to skin of the face. All sheep died or were euthanatized after continuous weight loss for up to 8 months. Another outbreak in the same State affected 26 out of 360 adult sheep after being dosed with molasses, causing lesions in the oral mucosa followed by malignant edema of the oral and subcutaneous tissues. All sheep died within 10 days after treatment. Another case observed in Rio Grande do Sul affected one out of 88 adult ewes. The animal died with hemorrhages from the nose and mouth 8 days after antihelmintic treatment. At necropsy an ulcerated lesion covered by a clot of fibrine was found in the oral mucosa. The lesion had a fistulated tract with purulent exudate extending through the soft tissues in direction to the right eye. Two outbreaks occurred in the State of Goiás 16-20 days after antihelmintic treatment in 5-9 months old lambs. Morbidity was 11.43% and 15.62% and fatality rate was 7.14% and 9.38%. Main clinical signs were nervous signs caused by abscesses with a diameter of 0.5-3cm, containing purulent exudates in the oropharynx, soft palate and adjacent muscles extending to the meninges through the atlantoaxial joint. Not-purulent meningitis was observed histologically. The need to use proper drenching guns and pay attention during dosing to avoid lesions of the oropharynx is pointed out.

E-mail: bombonat@usp.br The thymus is an organ with great immunological relevance during foetus life and neonatal period, It is the predecessor of lymphopoiesis and shows high lymphopoietic activity, being also the largest lymphoid organ during intrauterine life and at birth. In this study, the morphological aspects of the prenatal development of dog thymus were described in foetus of different ages and sexes. Twenty-four foetus of mongrel domestic dogs, males and females, were divided into four age groups. The thymus presented two lobes linked by a rose-colored tissue connection that was localized in the cranial mediastinal space. The cranial portion protrudes slightly beyond the first pair of ribs. The cells that formed the thymus parenchyma were well organized. Concentric aggregates named Hassal corpuscles were observed, involved by a thin capsule of connective tissue thickened by adipose tissue. The vessels size gradually increased with the foetus age, being larger in animals that were closer to birth. The volume (Vref) and size parameters of female thymus were larger than in males.

#60 - IgA production, coliforms analysis and intestinal mucosa morphology of piglets that received probiotics with viable or inactivated cells, p.241-245

Abstract in English:

ABSTRACT.- Rodrigues M.A.M., Oliveira D.A., Taketomi E.A. & Hernandez-Blazquez F.J. 2007. IgA production, coliforms analysis and intestinal mucosa morphology of piglets that received probiotics with viable or inactivated cells. Pesquisa Veterinária Brasileira 27(6):241-245. Departamento de Cirurgia, Setor de Anatomia, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. Email: fjhblazq@usp.br Two types of probiotics were used in piglets. One product is a mixed culture of viable Lactobacillus acidophilus, Enterococcus faecium e Bifidobacterium bifidum. The second product is composed of inactivated Lactobacillus acidophilus cells. The piglets received two weekly oral doses for 30 days while a control group did not receive probiotics. All piglets were euthanized at the 30th day of life and the mesenteric lymph nodes, the small intestine, and blood samples were collected. The tissue samples were studied by light microscopy and the blood serum was analyzed by ELISA method. The treatment with the probiotic with viable cells produced higher serum levels of IgA (P<0.05) and more IgA expressing cells were found in the mesenteric lymph nodes than observed in the inactivated cells treatment or control groups (P<0.05). Also, intestinal villi were longer, crypts were deeper (P<0.05) and fecal coliform count was lower than found in the inactivated product (P<0.05). These results suggest that viable probiotics are more efficient than inactivated probiotics to induce immunostimulation and intestinal modifications in piglets, thus improving their health and development.
