#81 - Virulence factors and antimicrobial resistance in Staphylo-coccus aureus isolated from bovine mastitis in Rio de Janeiro, p.369-374

E-mail: miliane@ufrrj.br The study was conducted to characterize pheno-genotypically the virulence factors and resistance pattern of Staphylococcus aureus isolates from milk samples of cows with subclinical mastitis. All hemolytic isolates presented beta-hemolysin, and 38% of the non-hemolytic isolates were able to express hemolysins in the presence of a beta-hemolytic strain. The amplification of the coa-gene displayed four different size polymorphisms with about 400 bp, 600 bp, 700 bp and 900 bp. The spaA gene that encodes the IgG-binding region of protein A revealed sizes of 700 bp and 900 bp. The amplification of region X from spaA yielded a single amplicon for each isolate with the prevalent amplicon size being of 180 bp. Amplification of sae gene yielded an amplicon size of 920 bp in 71% of the isolates. Antibiotic resistance pattern revealed that 42% S. aureus were susceptible to all antimicrobials tested. Seven different antibiotic patterns were observed. Our results indicated that 47% and 25% of S. aureus strains exhibited resistance to penicillin and oxacillin respectively. All oxacillin-resistant isolates were mecA-positive.

The aim of present study was investigate the milk pathogens, sensitivity and multiple drug resistance of isolates, somatic cell count and residues of drugs in milk, from cattle with and without mastitis, come from four little organic dairy farms in State of São Paulo, Brazil. Were used 148 cattle on the middle period of lactation. From these, two showed clinical mastitis, 72 subclinical mastitis and 74 without signs of mammary inflammation (control). Staphylococcus aureus (25.7%), Streptococcus spp. (21.4%), Corynebacterium bovis (12.9%), Streptococcus agalactiae (4.3%) and Staphylococcus spp (4,3%) were the more-frequent microorganisms isolated from animals with mastitis. Aspergillus spp. was isolated from one animal. Ceftiofur (95.2%), oxacillin (84.2%), gentamicin (76.3%) and cefoperazone (70.3%) were the more effective drugs. High resistance of isolates were found to penicillin (53.5%), ampicillin (41.6%) and neomycin (38.6%). Multiple drug resistance to three or more drug was observed in 40 (39.6%) isolates. Media of somatic cell count encountered in animals with mastitis and controls were 175,742.67cs/mL and 58,227.6 cs/mL, respectively. Antimicrobials residues in milk were detected in four (2.7%) animals. The present findings showed the low somatic cell count of animals, indicative of good quality of milk. However, pointed the need of control measures for contagious pathogens of bovine mastitis and more attention for prohibition of antimicrobial use in organic dairy farms.

E-mail: mateus.costa@univasf.edu.br Aiming the evaluation of sensitivity profiles of pathogen bacteria responsible for fish diseases, 51 bacterial isolates from Jundiá (Rhamdia quelen) belonging to the genus Acinetobacter spp. (8), Aeromonas spp. (15), Edwardsiella spp. (2), Enterobacter spp. (2), Klebsiella spp. (1), Plesiomonas spp. (5), Pseudomonas spp. (1), Staphylococcus spp. (11), and Vibrio spp. (6), were tested against antimicrobial agents used for treatment of bacterial fish diseases. All samples were processed at the Laboratory of Bacteriology, Department of Preventive Veterinary Medicine, UFSM. From 51 bacteria isolated from jundiá fishes (Rhamdia quelen) 51 (100%) were sensitive to gentamycin, 49 (96,08%) to sulphazotrin, 47(92,16%) to chloramphenicol, 43 (84,31%) to tetracylin, 43 (84,31%) to naldixic acid, 31 (60,78%) to nitrofurantoin, 22 (43,14%) to erytromycin, 22 (43,14%) to ampicillin, 15 (29,41%) spiramycin, 13 (25,50%) to cholystin, and 5 (3%) to penicillin G. With exception of an isolate of Staphylococcus spp., the bacteria analyzed in the present study were resistant to one or more antimicrobial agents tested. Knowledge of the sensitivity profile of bacteria involved in infectious processes in fish will allow rational antimicrobial treatment that will contribute to the control of fish diseases in Rhamdia quelen without causing great risks to public health and the environment.

#84 - Antimicrobial resistance and R-plasmid in Salmonella spp from swine and abattoir environments

E-mail: nslazaro@aol.com Salmonella serovars isolated from swine are of particular interest not only because of the pathogenic potential for this animal species, but also due to its relevance with regard to public health. On basis of the profile of resistance to antimicrobials, 13 Salmonella strains were selected which belonged to the serovars Muenster (7), Derby (4), Typhimurium (1), and Braenderup (1). They were isolated from healthy swine as well as from the abattoir environment in the state of Rio de Janeiro. All strains of Salmonella were subjected to bacterial conjugation, and the E. coli K12 Nalr Lac+ F standard strain was used as receptor, with the purpose to verify the ability to transfer the resistance marks. Gene transfer phenomenon was detected in seven strains, and except Salmonella Typhimurium which transconjugated to Sm, Tc and Su, the remaining ones were characterized by transferring mark Su only. By plasmidial analysis of strains used and their respective transconjugants, 63 Kb plasmid was found, which was probably related to S. Typhimurium resistance.

Plasmids varying in size from 1.2 Kb to 42 Kb were detected in 37 (97.36%) of the 38 samples, corresponding to 11 different profiles (P1- P11), varying from 1 to 6 plasmids per model. The number and plasmids diversity was greater than the resistance marks for strains. The plasmid of 2.85 Kb was the most frequent, being present in 83.78% of the 37 strains; only the one of 7.5 Kb was detected at the two slaughterhouses. There was no parallelism between resistance pattern and plasmidial profile, and a same antibiotype was found in several plasmidial profiles. The results of the present investigation, allowed us to conclude that the plasmid characterization is an useful and simple tool for the epidemiological typing of this sorovar.

#86 - Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis, 23(2):47-51

The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

[Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis.] Perfil de susceptibilidade a antimicrobianos em amostras de cocos Gram-positivos, catalase negativos, isoladas de mastite subclínica bubalina. Pesquisa Veterinária Brasileira 23(2):47-51. Depto Epidemiologia e Saúde Pública, Instituto de Veterinária, UFRRJ, Seropédica, RJ 23890-000, Brazil. Estudou-se o perfil de susceptibilidade a antimicrobianos em cocos Gram-positivos catalase negativos (21 amostras de Lactococcus garvieae e 6 de Enterococcus gallinarum), isoladas do leite de fêmeas com mastite subdínica e pertencentes a uma população composta por seis rebanhos bubalinos localizados no Estado do Rio de Janeiro. O teste utilizado foi o da difusão de discos em agar Müller Hinton, segundo recomendações do National Committee for Clinicai Laboratory Standards - NCCLS, tendo sido testados discos com ampicilina (10mg), cefalotina (30mg), cefotaxima (30mg), cefoxitina (30mg), cloranfenicol (30mg), eritromicina (15mg), gentamicina (10mg), nitrofurantoína (300mg), norfloxacina (10mg), penicilina (10 UI), tetracidina (30mg) e vancomicina (30mg). Os resultados evidenciaram que em se tratando de Lactococcus garvieae, o antimicrobiano mais eficiente foi o nitrofurantoína com 85,71% de sensibilidade, seguido da cefotaxima (61,90%), vancomicina (52,38%), norfloxacina (47,62%) e cefalotina (47,62%). A maior resistência foi desenvolvida frente a penicilina e ampicilina, com 95,24% de resistência para os dois antimicrobianos testados. O perfil de susceptibilidade desenvolvido pelas amostras de Enterococcus gallinarum, mostrou baixa sensibilidade frente aos antimicrobianos testados, onde os maiores índices foram observados frente eritromicina e gentamicina, com 33,34% de sensibilidade para ambos; quanto à resistência desenvolvida, foi possível observar 100% de resistência com relação a vancomicina e tetraciclina, seguindo-se cloranfenicol, penicilina, ampicilina, cefoxitina, cefalotina, cefotaxima, norfloxacina e nitrofurantoína, todas evidenciando uma resistência de 83,33% das amostras testadas.


#87 - Bacterial isolations from "Cara inchada"-lesions of cattle

Abstract in English:

Attempts were made to study a possible involvement of bacteria in the periodontal lesions of "Cara inchada" in cattle (CI). Corynebacterium pyogenes could be isolated from all of 23 CI-positive bovines and from the tissue samples of 1 of 22 CI-negative controls. Bacteroides mekzninogenicus occurred together with C. pyogenes in all CI-lesions and only in 1 of the CI-negative tissue samples. Bacteroides bivius was cultivated from 13 and Fusobacterium nucleatum from 9 of the 23 CI-positive, but from none of the CI-negative animals. Actinomyces israelii occurred more frequently in the samples of the CI-negative than those of the CI-positive animals. The isolated bacteria were consistently susceptible in vitro to penicillin G, tetracycline and erythromycin. Repeated intragingival injections of C. pyogenes alone and together with B. melaninogenicus into 5 calves and 2 young sheep did not produce progressive periodontal lesions. However, 2 of the calves developed distinct and 2 slight retractions of the gingiva near the sites of injections. This did not indicate a primary role of the isolated bacteria in "Cara inchada" of cattle, but could suggest some bacterial involvement in the development of the periodontal lesions.

Foi conduzido um estudo para investigar o possível envolvimento de agentes bacterianos em lesões de "cara inchada" em bovinos (CI). Corynebacterium pyogenes foi isolado das lesões peridentárias de todos os 23 bezerros com CI necropsiados para fins de estudo bacteriológico, e somente 1 das 22 biópsias da gengiva feitas em bezerros negativos para a doença. Bacteroides melaninogenicus ocorreu, junto com C. pyogenes, em todas as lesões peridentárias, mas somente em 1 das biópsias dos animais sadios de controle. Bacteroides bivius foi isolado de 13 e Fusobacterium nucleatum de 9 dos 23 bezerros com CI, mas de nenhum dos animais de controle. Actinomyces israelii ocorreu mais freqüentemente nas biópsias da gengiva dos bezerros de controle do que nas lesões peridentárias dos animais com CI. As bactérias isoladas mostraram-se, in vitro, sempre sensíveis à penicilina G, à tetraciclina e à eritromicina. Repetidas inoculações intra-gengivais de C. pyogenes, realizadas em 5 bezerros e 2 cordeiros, e de C. pyogenes junto com B. melaninogenicus, nã'o causaram lesões peridentárias progressivas; mas em 2 bezerros observaram-se, na região das inoculações, retração moderada e, em 2 outros, retração leve da gengiva. Estes resultados não indicam que as bactérias isoladas tenham papel primário na etiologiada "cara inchada" dos bovinos, mas parecem sugerir algum envolvimento destas bactérias na patogênese das lesões peridentárias.
