E-mail: eldinemneto@hotmail.com The objective was to study the effects of ruminal lactic acidosis in goats, induced experimentally, on the clinical manifestations, the physical, chemical and microbiological characteristics of the ruminal fluid. Ten 1 to 2 year old male goats, cross bred (SRD), with 25 kg body weight, in good health condition and housed in a stall, were rumen-fistulated. After establishing normal pattern for the variables studied, the acidosis was induced with the administration of 15g sucrose/kg body weight. Clinical and laboratorial observations were accomplished at intervals of 4, 8, 12, 16, 24, 32, 48, 72, 96, 120 and 144 hours, after the induction (PI). Already 4 hours PI the animals showed clinical signs of apathy, anorexia, increased heart frequency, ruminal atony, abdominal distention and diarrhea. The characteristics of the ruminal fluid suffered alterations, as reduction of the pH (below 5.0), and its color became a milky aspect, acid smell and aqueous consistency. The time of sedimentation activity (TAS) was reduced and the dynamics of the fauna and flora were committed, with the prevalence of Gram-positive bacteria. At the end of the observation period the analyzed variables were not yet fully re-established in some animals.

ABSTRACT.- Mendonça C.L., Vieira D., Kohayagawa A., Schenk M.A.M., Madruga C.R. & Afonso J.A.B. 2003. [Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen, reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

RESUMO.- Mendonça C.L., Vieira D., Kohayagawa A., Schenk M.A.M., Madruga C.R. & Afonso J.A.B. 2003. [Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.]

#83 - Oxidative metabolism of the neutrophils in sheep treated with sodium monensin and experimentally submitted to ruminal acidosis, 22(4):129-134

Abstract in English:

ABSTRACT.- Afonso J.A.B., Ciarlini P.C., Kuchembuck M.R.G., Kohayagawa A., Feltrin L.P.Z., Ciarlini L.D.R.P., Laposy C.B., Mendonça C.L. & Takahira R.K. 2002. [Oxidative metabolism of the neutrophils in sheep treated with sodium monensin and experimentally submitted to ruminal acidosis.] Postal 152, Garanhuns, PE 55292-901, Brazil. Ruminal acidosis is due to excessive ingestion of carbohydrates of rapid fermentation without previous adaptation of the microorganisms, causing severe metabolic disturbances to the animals. The objective of the present study was to assess the neutrophilic oxidative metabolism in sheep treated with sodium monensin in experimentally induced ruminal lactic acidosis. A total of 18 mate sheep, half-bred (Ideal x Merino), fistulated in the rumen, were used; nine of them received 33 mg/kg of the ionophore diet per day, for 30 days; the others were controls. The acidosis was induced by supplying 15g of sucrose/kg of body weight. The clinical evaluation and the rumen and blood samples were obtained before (Oh) and at 6, 12, 24 and 48 hours post-induction. In both groups, all the animals presented clinical manifestations of ruminal lactic acidosis 6 hours after the induction. From this period on, a significant pH decrease (P<0.05) was observed in the ruminal fluid, which reached levels below 5. There were relevant differences (P<0.05) between the groups 12 hours after the induction, when the sheep treated with monensin had higher values than those of the control group. During this period, the oxidative metabolism of the neutrophils remained inhibited, and the reestablishment of this function only occurred in the sheep which received monensin. Blood pH, plasmatic glucose and the ionizable calcium suffered alterations within its levels. The seric cortisol concentration rose significantly (P<0.05) in both groups, although differences (P<0.05) between them were found at the end of the observation period. The treatment with monensin did not influence the oxidative metabolism of the neutrophils inhibited by the lactic acidosis; however, a faster recovery of this metabolism was verified in the animals treated with the ionophore.

Pesquisa Veterinária Brasileira 22(4):129-134. Clinica de Bovinos, UFRPE, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil.

Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil.

#85 - Occurrence of enterotoxigenic Escherichia coli and Salmonella sp in calves in the southern agreste region of the State of Pernambuco, Brazil

Abstract in English:

The diarrheic processes caused by pathogenic enterobacteria are of great economicsanitary importance, with limiting effects on cattle raising. In the present study 106 fecal samples from 52 diarrheic and 54 non-diarrheic calves aged 1 to 90 days were analyzed, in arder to determine the occurrence of enterotoxigenic Escherichia coli (ETEC) and Salmonella sp among calves reared in the agreste region of the state of Pernambuco and to determine the behavior of the isolates in response to 16 antimicrobial agents. The strains were cultured in selective media and submitted to biochemical characterization, serologic identification and detection of heat-stable enterotoxin. E. coli was isolated from all 106 calves studied and five of these strains (4,7%) were proclucers of heat-stable enterotoxin. Salmonellasp was isolated from six (5,7%) samples belonging to serovars Dublin, Muenchen and Infantis. The Salmonella strains were highly sensitive to the drngs tested, whereas the E. co/istrains were more resistent to tetracycline (50,9%), sulfonamide (40,6%), streptomycin (35,9%) and ampicillin (22,6%). Particularly outstancling in the general analysis was the multiresistance of E. coli (52,0%), with a predominance of the tetracycline-sulfonamide association. The present results serve as a warning regarding the incliscriminated use of antimicrobial agents. Low percentages of ETEC and Salmonella in calves show the necessity of other diagnostic procedures to clarify the etiology.

#86 - Virulence factors of Yersinia enterocolitica 0:3 isolated from healthy pigs, Rio de Janeiro

Abstract in English:

Virulence factors were determined in 43 cultures of Yersinia enterocolitica serotype 0:3, biotype 4, phagetype VIII, isolated from apparently healthy pigs. Twenty one isolates were from tonsils, twelve from feces, six from colon contents and four from tongues. The in vitro tests consisted of examining autoagglutination at 36ºC, and absorption of Congo Red. The virulence assays of Y. enterocolitica were conducted by in vivo tests, testing the invasiveness into guinea-pig eyes and detection of heat stable enterotoxin in suckling mice. The results revealed a higher positivity percentage of the autoagglutination test (76.7%), followed by absorption of Congo Red (53.5%), invasiveness into guinea-pig eyes (20.9%) and detection of heat-stable enterotoxin (7.0%). A higher correlation was found in vitro test as compareci to in vivo tests, which characterized 53.5% Y. enterocolitica isolates tested as virulent and which allowed the conclusion that the simultaneous utilization of autoagglutination and absorption of Congo Red tests may be relevant for the identification of pathogenic strains.
