Abstract in English:

ABSTRACT.- Santos R.A., Afonso J.A.B., Mendonça C.L. & Simão, L.C.V. 2007. [Clinical aspects and characteristics of the milk in sheep with mastitis experimentally induced with Staphylococcus aureus.] E-mail: carlaze@uol.com.br The objective was to study the clinical aspects and the physical-chemical characteristics of the milk in sheep with mastitis experimentally induced with Staphylococcus aureus. For such, were used 10 Santa Inês primiparity ewes, weighing 30 kg, clinically healthy and housed in a stall. After establishing the normality patterns for the studied variables, the animals were inoculated into one teat of the udder with an inoculum of 1.0x104ufc/ml of S. aureus, while the other gland served as control. The clinical observations were accomplished in intervals of 12, 24, 36, 48, 60, 72, 84, 96, 108, 120, 132, 168, 180, 288 and 336 hours after the inoculation of the agent (PI). All the animals presented clinical systemic manifestations, and in the inoculated glands with more intensity from 24 hours on after the inoculation. There were significant alterations (P<0.05) in the production and physical-chemical composition of the milk in relation to the controls, with reduction of volume and fat. The pH, the chlorates and CCS reached very high indexes, accompanied by the CMT test reaction. The bacteriological exam revealed the presence of S. aureus during the phase of infection. After the treatment of the sheep 36 hours PI, one animal came to death 48h PI, and the others showed clinical recovery; however there was no physiologic re-establishment of the inoculated udders which lost their function.

E-mail: zafalon@iz.sp.gov.br Electrical conductivity measured by a hand-held meter and chloride concentration of milk were studied as auxiliary methods for diagnosis of bovine subclinical mastitis in the identification of affected mammary quarters where Staphylococcus aureus and Corynebacterium sp were later isolated. Tests were made during 2 years in Holstein cows of a dairy farm producing type C milk, where milking was performed once a day. Sensitivities of electrical conductivity and chloride concentration tests from mammary quarters, where Corynebacterium sp was isolated (65.3% and 78.3%, respectively), were superior to the found in mammary quarters where S. aureus was identified (55.4% and 68.2%, respectively). The efficacies of the two diagnostic tests were similar. Statistical significance was demonstrated with regression analysis of both tests of healthy mammary quarters and subclinical mastitis quarters infected with Staphylococcus aureus.

E-mail: lafranco@vet.ufg.br With the objective to investigate a possible association between foot diseases, clinical mastitis and/or metritis and predisposing factors for foot diseases, 5300 dairy cows from 80 intensive and semi-intensive farms were used. In 325 (6.13%) cows only foot disease was diagnosed, in 35 (0.66%) foot disease and clinical mastitis, in 52 (0.98%) foot disease and metritis, in 28 (0.53%) foot disease, clinical mastitis and metritis, in 128 (2.42%) only metritis, in 165 (3.11%) only clinical mastitis, and in 89 (1.68%) cows metritis and clinical mastitis. Rapid changes in the diet, high exposure time of hoof horn to slurry and wet conditions, concrete floors, use of footbaths, low frequency of claw trimming, irregular quarantine, and no attention to health aspects during acquisition of the animals were considered as major risk factors for the occurrence of those diseases. The McNemar test for dependent samples showed significant statistical differences between the occurrence of foot disease, clinical mastitis and metritis, in spite of the poor association between those diseases. It was concluded that there was no expressive relationship between foot diseases, clinical mastitis and metritis in lactent cows.

#94 - Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis, 23(2):47-51

Abstract in English:

ABSTRACT.- Vianni M.C.E. & Lázaro N.S. 2003. [Profile of antimicrobial susceptibility in strains of Gram positive cocci, negative catalase, isolated from buffalo subclinical mastitis.] The susceptibility of antimicrobials was studied in Gram positive and catalase negative cocci (21 samples of Lactococcus garvieae and 6 Enterococcus gallinarum), isolated from the milk of cows with subclinical mastitis, belonging to six buffalo herds in the State of Rio de Janeiro. The test used was diffusion of disks in agar Müller Hinton, according to recommendations of the National Committee for Clinicai Laboratory Standards - NCCLS. There were tested disks with ampicillin (10mg), cefalotin (30mg), cefotaxime (30mg), cefoxitin (30mg), doranfenicol (30mg), eritromycin (15mg), gentamycin (10mg), nitrofurantoin (300mg), norfloxacin (10mg), penicillin (1 O IU), tetracydin (30mg) and vancomycin (30mg). The results showed that with Lactococcus garvieae, the most efficient antimicrobial was nitrofurantoin, revealing 85.71% sensibility, followed by cefotaxime (61.90%), vancomycin (52.38%), norfloxacin (47.62%) and cefalotin (47.62%). The highest resistance was developed against penicillin and ampicillin, with 95.24% resistance for the two antimicrobials. The susceptibility profile developed by the strains of Enterococcus gallinarum showed low sensibility against the tested antimicrobials; the highest resistance observed was against eritromycin and gentamycin, with 33.34% sensibility for both. The antimicrobial evaluation showed 100% resistance against vancomycin and tetracyclin, followed by cloranfenicol, penicillin, ampicillin, cefoxitin, cefotaxim, norfloxacin and nitrofurantoin; all of them showed a resistance of 83.33% with the samples tested.

#95 - Mastitis caused by Mycoplasma bovis in dairy cattle, 21(4):143-145

Abstract in English:

ABSTRACT.- Pretto L.G., Müller E.E., Freitas J.C., Mettifogo E., Buzinhani M., Yamaguti M. & Salvador R. 2001. [Mastitis caused by Mycoplasma bovis in dairy cattle] Mastite bovina por Mycoplasma bovis em rebanhos leiteiros. Pesquisa Veterinária Brasileira 21(4):143-145. Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86051-970, Brazil. In this study 713 cows were examined. The animals were from three dairy farms in northern Paraná and the southwest of the State of São Paulo. From these cows, 137 had mastitis. On the three farms, 8 cows (1-12%) with Mycoplasma bovis mastitis were detected. Four were treated with tylosin and oxytetracyçlin and three with enrofloxacin There was no response to the treatments, and these animals were culled during the lactation period. One cow treated with enrofloxacin almost totally recovered milk production, but elimination of M. bovis continued during the lactation, and there was no bacteriological cure. This cow had a normal milk production in the next lactation period, without elimination of M. bovis. Culling of positive animals, the bacteriological study anel correct application of preventive practices for contagious mastitis controlled the dissemination of M. bovis to other animals.

Depto Medicina Veterinária Preventiva, Universidade Estadual de Londrina, Cx. Postal 6001, Londrina, PR 86051-970, Brazil.

#97 - Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens, 18(1):39-44

Abstract in English:

ABSTRACT.- Brito M.A.V.P., Brito J.R.F., Souza H.M. & Vargas O.L. 1998. [Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens.] Samples of bulk tank milk from 33 herds were collected at the dairy processing plant and cultured, as a means of detecting specific (contagious) bovine mastitis pathogens. Somatic cell counts (SCC) were made on a Fossomatic 90. Two and three weekly consecutive samples were obtained from 13 and 12 herds, respectively. Only one sample was examined from eight herds. Three daily consecutive samples of bulk milk and individual quarter samples from all lactating cows from four herds (A, B, C and D) were also examined. Milk from individual quarters were cultured on blood agar while tank milk samples were cultured on TKT, Mannitol Salt, MacConkey agars and Sabouraud containing chloramphenicol. Staphylococcus aureus was recovered from 26 of the 33 herds sampled in the dairy processing plant. Nine of these samples also contained Streptococcus agalactiae. Nine herds had SCC above 500,000 ml1 The remaining 23 herds had SCC levels below 400,000 ml1 S. aureus and S. agalactiae were isolated from five of the nine herds with high SCC, S. agalactiae from one and S. aureus from three. Six herds had SSC below 200,000 ml1 S. aureus and S. agalactiae were isolated from one, S. aureus from three, while the other two were negative for both pathogens. The results of herds A, B, C and D sampled at the farms showed that S. aureus was isolated from 1.8%, 19.2%, 17.0% and 8.4% of the animals and 0.9%, 5.9%, 5.4% and 2.2% of the mammary quarters, respectively. S. agalactiae was isolated from herds A, C and D. Within these herds the percentages of isolation were, respectively, 1.8%, 10.6% and 8.4% for the cows and 0.46%, 3.8% and 3.7% for the mammary quarters. S. aureus was recovered from all three bulk tank cultures from herds A, B and D. Only the third sample from herd C was positive for S. aureus. S. agalactiae was recovered from all samples collected from herd D, two samples from herd C and one sample from herd A. Coliforms were isolated from all tank samples from herds A, B, C and D and from all but one sample collected in the processing plant. Yeasts were recovered from 16 herds sampled at the processing plant and from all tank samples from herds A, B, C, and D. Neither coliforms or yeasts were isolated from the individual animals of herds A, B, C and D. These findings indicate that the milk was contaminated during or after milking, probably due to deficient hygiene and cleaning procedures. The analysis of the bulk tank milk cultures showed that the test was sensitive enough to detect contagious mastitis pathogens. The sensitivity of the test increased when more than two consecutive samples were examined.

RESUMO.- Brito M.A.V.P., Brito J.R.F., Souza H.M. & Vargas O.L. 1998. [Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens.]

#98 - Sensitivity and specificity of the Califomia Mastitis Test as a diagnostic tool for subclinical mastitis in quarter somatic cell count estimation

Abstract in English:

The California Mastitis Test (CMT) is a simple and widely used diagnostic tool for subclinical mastitis. lt is used even in areas where laboratory facilities are available for diagnosis and monitoring program purposes. CMT usually score 1-5, where 1 indicates a completely negative reaction and 2-5 increasing degrees of inflammatory reaction of the udder. The reactions 2-5, 3-5 or 4-5 may be considered as indicative of subclinical mastitis, because they are related to an increase of the somatic cell count (SCC) in the milk. This variation, however, may produce either false-positive or false-negative results. The aim of this study was to assess the sensitivity and specificity of the CMT in comparison with SCC. A total of 3,012 quarter milk samples from 760 lactating cows were examined. CMT was evaluated at the moment of sampling at cow side and SCC in the laboratory, with a fluoro-opto-electronic method (Fossomatic 90). The average CCS (x 1,000 cells/ml) for CMT scores were: 1 (79.9), 2 (333.5), 3 (670.3), 4 (1,354.0) and 5 (4,455.6). Three options for CMT interpretation were evaluated in relation to a range of CCS, starting from 100,000 cells/ml: (a) 1 versus 2, 3, 4, and 5; (b) 1 and 2 versus 3, 4 and 5; and (e) 1, 2 and 3 versus 4 and 5. The sensitivities of CMT scoring to detect quarters with SCC >200,000/ml were 79%, 61% e 34%for options a, b ande, respectively. The sensitivities of CMT scores for SCC >500,000/ml for options a, b ande, were, respectively, 93%, 82% e 54%. The sensitivity of CMT scores to identify subclinical mastitis was satisfactory (above 80%) when options b ande were used. The interpretation of CMT score 3 as negative for subclinical mastitis was considered as adequate (sensitivity around 80%) only when CCS ranged between 1,200,000 and 1,400,000 cells/ml. The specificities of CMT scoring for CCS of 200,000 and 500,000 were, respectively, 90% and 80% (option a), 97% and 90% (option b) and 99% and 97% (option e).

#99 - Influence of subclinical staphylococci mastitis on physical, chemical and cellular milk characteristics

Abstract in English:

One hundred twenty-six milk samples from 63 apparently healthy cows, but positive in the California Mastitis Test were submitted to determinations of pH, acidity, density, butter-fat, total solids, non-fat solids, cryoscopic point, caseine level, chloride level And polymorphonuclear leukocytes. Forty-one cows were infected by coagulase-positive Staphylococcus and 22 by coagulase-negative Staphylococcus. The results obtained in the milk sample analysis from the healthy quarters and infected by coagulase-positive Staphylococcus showed variations of all constituents investigated. However only the pH values (F=4.17*) and the polymorphonuclear leukocytes (F=ll.35**) showed significant differences. On the other hand, between the milk samples from healthy quarters and infected quarters by coagulase-negative Staphylococcus, only the polymorphonuclear leukocytes (F=16.29**) showed significant differences.

#100 - Influence of mastites on the physic-chemical features of goats' milk

Abstract in English:

Mille samples from tbree goat breads (Dusk Alpine, Saanem and Toggenburg) with milk production averaging 2.66 kg/day and from crossbred goats were examined during 12 months for physic-chemical features. The results for individual milk and the mixture were respectively: density 1,030.5 and 1,033.5 at 15ºC; acidity 15.67 and 15,84°D; average percentage of fat 4.00 and 4.1 %; total dry extract 12.42 and 12.95%; fatty dry extract 8.83 and 8.73%; cryoscopic indexes -0.569 and -0.585ºC. The average percentage of chlorides was normal (0.21 and 0.19%); lactose 3.27 and 3.42%; caseíne (PTN) 2.68 and 2.52%; total proteine (PTN) 3.59 and 3.43%. The results obtained with milk from goats with mastitis showed the following alterations caused by experimental infection by Staphylococcus aureus: lowered values for density (1,022.5 and 1,023.0 at 15ºC) and acidity (14.50 and 6.1 ºD); increased values for chlorides (0.34 and 0.39%); and reduced values for lactose (1.00 and 1.12%).
