Intoxication by Cassia occidentalis (Leguminoseae) in swine

Abstract in English:

Two outbreaks of intoxication by Cassia occídentalis in pigs occurred on a farm located in the municipality of Águas de Chapecó, Santa Catarina, Brazil. The ingestion occurred after com mixed with C occidentalis seeds, hervested on a plantation with large amounts of the plant in the fruiting stage, was introduced into the diet. In July, 1983, from a total of 1200 pigs, 460 were affected and 420 died as a consequence of the disease. In June, 1984, of 800 animals, 40 were affected and 38 died. Clinical signs were characterized by anorexia, apathy, ataxia, diarrhea, vomiting, dyspnea, and lateral recumbency, with death occurring between eight and twelve days after ingestion was initiated. Postmortem examination of skeletal and cardiac muscles showed areas of paleness together with normal areas; the liver was pale and enlarged. Histological examination showed degeneration of skeletal and cardiac muscles and vacuolization of hepatocytes. The disease was experimentally reproduced in four swine, three ingesting seeds of C occidentalis comprising 20% of the ration, and one that ingested ration containing 10% seeds. The four animmals died seven to eight days after the start of the experiment. Clinical signs and pathological alterations were similar to those observed in field cases.

Descrevem-se 2 surtos de intoxicação por sementes de Cassia occidentalis em suínos, em uma granja localizada no município de Águas de Chapecó, Santa Catarina. A ingest:ro ocorrem após ser introduzido na alimentação das animais, milho misturado com sementes de C. occidentalis, colhido em uma lavoura com grandes quantidades da planta em estágio de frutificação. Em julho de 1983 adoeceram 460 suínos e morreram 420 de um total de 1200 e, em junho de 1984 adoeceram 40 e morreram 38 de um total de 800. Os sinais clínicos, observados 3 dias após o início da ingestão caracterizaram-se por anorexia, apatia, ataxia, diarréia, vômitos, urina amarelo escura, dispneia, decúbito lateral e morte 8 a 12 dias após o início da ingestão. As principais lesões macroscópicas caracterizaram-se por áreas de palidez intercaladas com áreas de coloração normal nos músculos esqueléticos e cardíaco, e fígado aumentado de tamanho e com coloração mais clara que a normal. Histologicamente observou-se degeneração dos músculos esqueléticos e cardíaco e vacuolização dos hepatócitos. A doença foi reproduzida experimentalmente em 4 suínos, 3 que ingeriram sementes de C. occídentalis misturadas a 20% com a ração e 1 que ingeriu as sementes a 10%. Os 4 animais morreram 7 a 8 dias após o início da administração. Os sinais clínicos e patologia observados foram similares à dos casos espontâneos.


Comparison between the serum neutralization and the immunodiffusion tests for the detection of antibodies for Aujeszky's disease virus in swine

Abstract in English:

A comparison was made between the plate immunodiffusion (ID) test and the micro-serum neutralization (SN) test in the detection of antibodies for Aujeszky's disease virus (ADV) in pig sera, using a total of 813 sera derived from five infected herds. The plate ID test was as sensitive and specific as the micro SN test in detecting positive sera that possessed neutralizing activity only when tested undiluted. Sera with this titer generally reacted by bending the precipitation line formed between the antigen and the reference serum. Sera with SN titers equal to or greater than eight, usually gave two lines of precipitation. The plate ID test was equally efficient and specific in detecting antibodies resulting from a natural infection or from vaccination with an inactivated oil-emulsion vaccine, as well as maternally-derived antibodies in the sera of piglets from vaccinated sows. Of 813 sera assayed in the micro SN test, 295 (36.3%) contained ADV antibody, 382 (47%) were negative and 136 (16.7%) were toxic. The sarne sera assayed in the plate ID test showed 347 (42.7%) positive for precipitating antibodies and466 (57.3%) negative. The major limitation of the SN test was the excessive percentage of sera (16.7%) that were toxic for the indicator cells (chicken embryo fibroblasts), due mainly to bacterial contamination and/or hemolysis as a result bad handling and storage of samples before arriving to the laboratory. The disagreement between the number of positive sera detected by the two tests in favor of the plate ID test, was due to the fact that 52 sera that were positive by this test were toxic when assayed by SN. Under the experimental conditions, the plate ID test was both sensitive and specific for the detection of antibodies for ADV, and as well as being economic, simple and rapid to perform, there is the advantage that it can be used to test moderately contaminated and/or hemolysed sera that are toxic for the indicator cells in the SN test.

A comparação do teste de imunodifusão (ID) em placa e o microteste de soroneutralização (SN), na detecção de anticorpos para o vírus da doença de Aujeszky (VDA) em soros suínos, foi realizada em 813 soros oriundos de cinco plantéis infectados com o VDA. O teste de ID em placa foi altamente sensível e especifico, detectando como positivos soros que, no micro-teste de soroneutralização, apenas reagiam quando eram testados sem diluir. Os soros com este título reagiam, geralmente, dobrando a linha de precipitação formada entre o antígeno e o soro de referência. Soros com títulos SN de oito ou superiores apresentavam freqüentemente duas linhas de precipitação. O teste de ID foi igualmente eficiente e específico na detecção de anticorpos da infecção natural, da vacinação com vacina inativada oleosa, bem como de anticorpos transferidos da porca para os leitões via colostro. De 813 soros submetidos ao teste de SN, 295 (36,3%) revelaram anticorpos, 382 (47%) eram negativos e 136 (16,7%) eram tóxicos. Os mesmos soros submetidos ao teste de ID, revelaram 347 (42,7%) positivos para anticorpos precipitantes enquanto que, 466 (57,3%) eram negativos. A maior limitação do teste de SN foi a excessiva percentagem de soros tóxicos (16,7%) para as células indicadoras (fibroblastos de embrião de galinha), principalmente, devido a contaminação bacteriana e/ou hemólise causada por deficiente dessoragem e estocagem antes de serem enviados ao laboratório. A discordância entre o número de soros detectados como positivos para anticorpos em favor do teste de ID, foi devido ao fato de que 52 soros positivos por este teste foram tóxicos no teste de SN. Nas atuais condições, o teste de ID foi sensível e específico na detecção de anticorpos para o VDA e tem a vantagem de ser econômico, simples e rápido de realizar, além de poder testar soros moderadamente contaminados e/ou hemolisados que são tóxicos para as culturas celulares utilizadas no teste de SN.


Equine protozoal myeloencephalitis

Abstract in English:

The occurrence of two cases of equine protozoal myeloencephalitis in a breeding establishment in southem Brazil is described. One of the horses, a 10-year-old thoroughbred mare, presented progressive incoordination of gait in the hind limbs for 15 days. Initially she was lame and dragged her toes while galloping. She was treated with high dosis of dexamethazone which seemed to deteriorate her clinical condition. She was then euthanized and necropsied. Reddened, soft, granular areas appeared at the cut surface of spinal cord segments. Microscopically these areas corresponded to marked inflammatory and degenerative changes among which protozoal microorganisms were detected. Milder inflammatory changel were also found in the brain. The other affected animal was also a 10-year-old throughbred mare which presented similar clinical sigos progressing to death within 60 days. No post-mortem examination was performed on this mare.

É notificada a ocorrência de dois casos de mieloencefalite eqüina por protozoário num Haras do Rio Grande do Sul. Um dos animais, uma égua Puro Sangue de Corrida de 10 anos, apresentou incoordenação progressiva dos membrosposteriores por um período de 15 dias. Os sinais começaram com claudicação. O animal arrastava as pinças dos membros posteriores ao galopar. Os sinais clínicos agravaram-se após terapia com altas doses de dexametasona. O animal foi sacrificado e, na necropsia, áreas avermelhadas, amolecidas e granulares foram detectadas na superfície de corte da medula espinhal. Microscopicamente essas áreas correspondiam a lesões inflamatórias e degenerativas acentuadas em meio as quais percebiam-se microrganismos protozoários. Lesões inflamatórias mais discretas eram observadas também no cérebro. O outro animal, também uma égua Puro Sangue de Corrida de 10 anos, apresentou sinais clínicos semelhantes que progrediram para a morte em 60 dias. Nesse caso, não foi realizada necropsia.


Experimental poisoning by Mascagnia pubiflora (Malpighiaceae) in rabbits

Abstract in English:

The dried and powdered leaves or fruit of Mascagnia pubiflora Griseb., a plant toxic for cattle, were administered by stomach tube to 14 and 9 rabbits, respectively. The plant, stored in the shade at room temperature for two to nine months after collection in the State of Mato Grosso do Sul, was shown to be toxic for this species also. The fruit proved to be approximately six times more toxic than the leaves. Six grams of the dried leaves per kilogram of body weight killed four of five rabbits, 4 g/kg killed three of eight, while 2 g/kg did not kill the one rabbit receiving that dose. Regarding the fruit, doses of 1 g/kg or above killed the five rabbits in each group, but 0.5 g/kg did not kill the four rabbits of that group. The first symptoms of poisoning with leaves were noted between 6h 02min. and 45h 39min., and with fruit between 2h 18min. and 20 hours after administration. The course of the poisoning lasted two to three minutes in the case of the leaves, and one to two minutes in the case of the fruit. Toe main symptoms, identical for rabbits receiving leaves or fruit, were those of "sudden death": the rabbits made sudden violent, uncontrolled movements and fell on their side; respiration became difficult and the animals died. In both groups of rabbits post-mortem examination showed congestion of the liver, and secondly, congestion of the lungs, while histology revealed degenerative and vascular alterations in the liver, kidneys and heart. These experiments show that the rabbit can be used as a small experimental animal in the continuation of the studies on the toxic properties of M pubiflora, and in the identification work of the toxic principles. It is probable that the leaves and fruit contain the sarne toxic elements.

As folhas e os frutos dessecados de Mascagnia pubiflora (Juss.) Griseb., planta tóxica a bovinos, foram administrados por sonda gástrica a 14 e 9 coelhos, respectivamente. A planta,·colhida em setembro de 1984 no Estado de Mato Grosso do Sul e guardada na sombra à temperatura ambiente durante 2 a 9 meses, demonstrou possuir toxidez também para essa espécie animal. Em relação à dose letal das folhas, 6 g/kg mataram 4 de 5 coelhos, 4 g/kg mataram 3 de 8 coelhos, 2 g/kg não mataram o único coelho que recebeu a planta nessa dosagem; em relação aos frutos, doses a partir de 1 g/kg mataram todos os 5 coelhos, enquanto que 0,5 g/kg não matou nenhum dos 4 coelhos. Dessa maneira, os frutos foram cerca de 6 vezes mais tóxicos que as folhas. Os coelhos mostraram os primeiros sintomas de intoxicação, no caso das folhas, entre 6h 02min. e 45h 39min., e no caso dos frutos, entre 2h 18min. e 20 horas, após a sua administração. A evolução do quadro clínico, no caso das folhas, foi de 2 a 3 minutos, e no caso dos frutos, de 1 a 3 minutos. O quadro clínico foi o mesmo, tanto nos coelhos que receberam as folhas, como nos que receberam os frutos. Esse quadro foi o da "morte súbita", isto é, os coelhos, de repente faziam movimentos desordenados violentos, caíam de lado, tinham a respiração difícil e logo morriam. Também os achados de necropsia e as alterações histopatológicas eram os mesmos para os animais intoxicados pelas folhas e pelos frutos. O achado de necropsia mais comum foi congestão hepática, em segundo lugar foi congestão pulmonar. Nos exames histopatológicos os órgãos principalmente afetados foram fígado, rim e coração, sob forma de alterações degenerativas e vasculares. Esses experimentos mostram que o coelho pode ser usado como animal experimental de pequeno porte na continuação dos estudos sobre a ação tóxica da planta, bem como nos trabalhos de isolamento e identificação de seus princípios ativos. É provável que as folhas e os frutos encerrem os mesmos princípios tóxicos.


Enzymatic and endotoxic activities of bacteria isolated from periodontal lesions of calves with "Cara inchada"

Abstract in English:

Bacteria isolated from periodontal lesions of calves with "cara inchada" were examined for enzymatic and endotoxic activities. Black-pigmented cultures of Bacteroides produced deoxiribonuclease, collagenease, chondroitin sulfatase, fibrinolysin, gelatinase, hyaluronidase, lipase and protease. Trypsin-like proteolytic activities were demonstrated in cultures of black-pigmented asaccharolytic Bacteroides that were also high producer of collagenase. Actinomyces israelii, A. pyogenes and Fusobacterium nucleatum showed weak production of hydrolytic enzymes. Biological activities of lipopolysaccharides extracted from Bacteroides melaninogenicus, Bacteroides spp. and F. nucleatum were examined by the ability to produce a Shwarzman-reaction in rabbits. Endotoxic lipopolysaccharides from F. nucleatum had higher biological activities than those from Bacteroides. These findings suggest a possible role of the bacterial enzymes and endotoxins in the development of periodontal lesions in young cattle.

Estudos foram realizados para determinar as atividades enzimáticas e endotóxicas de bactérias isoladas de lesões peridentárias de bezerros com "cara inchada". Amostras "pigmentadas" de Bacteroides hidrolisaram o maior número de substratos, produzindo desoxiribonuclease, colagenase, sulfato de condroitinase, fibrinolisina, gelatinase, hialuronidase, lipase e protease. Atividade proteolítica semelhante à da tripsina foi observada nas amostras assacarolíticas de Bacteroides, que foram também mais ativas na produção de colagenase. Actinomyces israelii, Actinomyces pyogenes e Fusobacterium nucleatum foram pouco ativos na produção de enzimas hidrolíticas. Atividade biológica de lipopolissacarídeos extraídos de Bacteroides spp., B. melaninogenicus e F. nucleatum foi testada pela capacidade de produção da reação dérmica de Shwarzman em coelhos. Endotoxina de F. nucleatum foi mais ativa, quando comparada com a de Bacteroides. Os resultados sugerem uma possível participação de enzimas e endotoxinas bacterianas no desenvolvimento das lesões peridentárias da "cara inchada'' dos bovinos.
