#11 - Quantification of growth factors in horse skin treated with platelet-rich plasma, 34(6):599-612

E-mail: msouza@ufv.br Platelet-rich plasma (PRP) is a product derived from total blood centrifugation, rich in bioactive factors, such as growth factors. Despite largely used in healing processes, there is a controversy whether the therapy is effective in promoting skin healing. The objective of this study was to quantify and compare the concentrations of the factors TGF-&#946;1 and PDGF-BB in PRP, blood plasma and skin, at different phases of the healing process of skin treated or not with PRP. Seven healthy crossbred 16 to 17-year-old geldings (16.14±0.63) were used. Three quadrangular-shaped lesions (6.25cm2) were surgically induced into the right and left gluteal regions of all animals. Twelve hours after induction of the wounds, 0.5mL of the PRP was administered in each of the four edges of the wound in one of the gluteal regions (Treated group = TG) randomly chosen. The contralateral region was used as control (CG). The wounds were submitted to daily cleaning with Milli-Q water, and samples were obtained with a 6mm diameter biopsy Punch. Six skin biopsies were obtained, the first carried out immediately after the production of the wound (T0), and the others 1 (T1), 2 (T2), 7 (T3), and 14 (T4) days after the lesion was induced. The sixth biopsy (T5) was obtained after complete healing of the skin, which occurred at about day 37 (36.85±7.45, CG; 38.85±6.46, TG). EDTA blood samples were also obtained, at all the times mentioned. Quantification of TGF-&#946;1 and PDGF-BB growth factors on the skin, PRP, and blood plasma was carried out by the ELISA technique. Data were statistically analyzed by the t test, Pearson correlation and regression, at a significance level of 5%. No difference was found between the groups in the values of the two growth factors measured on the skin, at the different times. Also, no correlation was found between the amount of growth factors present in the skin and plasma. On the other hand, a positive correlation was observed between PRP and skin in the treated group, for the growth factors TGF-&#946;1 (r=0.31) and PDGF-BB (r=0.38), as well as between both growth factors present in PRP (r=0.81). Considering the growth factor concentrations at T0, the highest skin values (p<0.05) of TGF-&#946;1, in both groups, occurred at T3 and T5. Higher values (p<0.05) of PDGF-BB occurred at T4 (TG) and T5 (CG). No plasma changes occurred at the concentration of these factors in relation to T0, suggesting that PRP does not cause a systemic effect when the procedures adopted in this research are used. Local administration of PRP in the volume studied, 12 h after surgical induction of cutaneous wound gluteal equine does not cause higher concentrations of the growth factors TGF-&#946;1 and PDGF-BB in the plasma and skin during the healing process.

E-mail: msouza@ufv.br O plasma rico em plaquetas (PRP) é um produto derivado da centrifugação do sangue total, sendo rico em fatores bioativos, como os de crescimento. Apesar da ampla utilização em processos cicatriciais, há controvérsia sobre a eficácia da terapia na cicatrização cutânea. O objetivo desse estudo foi quantificar e comparar a concentração dos fatores TGF-&#946;1 e PDGF-BB no PRP, plasma sanguíneo e pele, durante diferentes fases do processo de cicatrização da pele tratada ou não com PRP. Foram utilizados sete equinos machos castrados, mestiços, hígidos, com idade entre 16 e 17 (16,14±0,63) anos. Três lesões em formato quadrangular (6,25cm2) foram produzidas cirurgicamente nas regiões glúteas direita e esquerda de todos os animais. Doze horas após indução das feridas, 0,5mL do PRP foi administrado em cada uma das quatro extremidades das feridas de uma das regiões glúteas (Grupo tratado = GT), escolhida aleatoriamente. A região contralateral foi utilizada como controle (GC). As feridas foram submetidas à limpeza diária com água Milli Q, e amostras foram obtidas mediante biópsias realizadas com Punch de 6mm. Foram obtidas seis biópsias de pele, sendo a primeira realizada logo após a produção da ferida (T0), e as demais com 1 (T1) 2 (T2) 7 (T3) e 14 (T4) dias após a indução da lesão. A sexta biópsia (T5) foi obtida após completo fechamento da pele, que ocorreu aproximadamente aos 37 dias (36,85±7,45, GC; 38,85±6,46, GT). Também foram obtidas amostras de sangue com EDTA em todos os tempos mencionados. A quantificação dos fatores de crescimento TGF-&#946;1 e PDGF-BB na pele, PRP e plasma sanguíneo foi realizada pela técnica ELISA. Os dados foram analisados estatisticamente pelo teste t, correlação de Pearson e regressão, utilizando nível de significância de 5%. Não houve diferença entre os grupos, nos valores dos dois fatores de crescimento mensurados na pele, nos diferentes tempos. Também não houve correlação entre a quantidade dos fatores de crescimento presentes na pele e no plasma. Por outro lado, correlação positiva foi observada entre PRP e pele no grupo tratado, para os fatores de crescimento TGF-&#946;1 (r=0,31) e PDGF-BB (r=0,38), bem como entre ambos os fatores de crescimento presentes no PRP (r=0,81). Considerando as concentrações dos fatores de crescimento no T0, os maiores valores cutâneos (p<0,05) do TGF-&#946;1, em ambos os grupos, ocorreram nos tempos T3 e T5. Valores mais elevados (p<0,05) do PDGF-BB ocorreram no T4 (GT) e T5 (GC). No plasma não houve alteração nas concentrações desses fatores em relação ao T0, o que sugere que o PRP não acarreta efeito sistêmico, quando os procedimentos adotados na presente pesquisa são utilizados. A administração local de PRP no volume estudado, 12 h após indução cirúrgica de ferida cutânea na região glútea de equinos não ocasiona maiores concentrações dos fatores de crescimento TGF-&#946;1 e PDGF-BB no plasma sanguíneo e pele, durante o processo de cicatrização.

E-mail: guilherme.oberlender@uffs.edu.br The study aimed to quantify the concentrations of free IGF-I in serum and fluid of ovarian follicles in pre-pubertal gilts and describe the ovarian morphology by measuring the size of the ovaries and counting the number of surface follicles. Ovaries (n=1,000) from pre-pubertal gilts were obtained immediately after slaughter. A total of 10 samplings were performed, with ovaries obtained from 50 females for each collection. The follicles situated on the surface of each ovary were classified as small (SFs, 2 to 5mm in diameter) or large (LFs 6 to 10mm in diameter) and the follicular fluid was obtained by follicle aspiration. The collection of serum samples was performed after the gilts exsanguination using sterile tubes. From the pool of serum and follicular fluid obtained from 50 females, the concentration of free IGF-I was determined in each sample using an enzyme immunoassay kit (ELISA). The description of ovarian morphometry was performed in 100 ovaries from randomly selected gilts. The larger and smaller lengths of ovaries were measured, and the total number of SFs and LFs present on the surface of each ovary were also counted. The IGF-I concentration was greater (P<0.05) in LFs (170.92±88.29 ng/mL) compared with SFs (67.39±49.90ng/mL) and serum (73.48±34.63ng/mL). The largest and smallest length of the ovaries was 26.0±3.0 and 19.0mm ±2.0mm, respectively. The number of SFs (70.86±25.76) was greater (P<0.01) than LFs (6.54±5.26). The study concluded that LFs present greater levels of IGF-I when compared with SFs and blood, which is related to increased activity of the LFs and its differentiation to ovulation. In addition, ovaries of pre-pubertal gilts have a higher number of SFs compared to LFs. Therefore, our study demonstrated unique data regarding the physiological concentration of free IGF-I in ovarian follicles, that can be used in future research to evaluate the addition of this hormone in the in vitro production media of porcine embryos with the goal to improve the technique efficiency.

Os folículos localizados na superfície de cada ovário foram classificados em pequenos (FPs, 2-5mm de diâmetro) ou grandes (FGs, 6-10mm de diâmetro) e o fluido folicular foi obtido por aspiração dos folículos. A coleta do soro foi realizada após a exsanguinação das fêmeas com o uso de tubos estéreis. A partir do pool de fluido folicular e do soro obtido das 50 fêmeas, determinou-se a concentração de IGF-I livre em cada amostra por meio de kit de ensaio imunoenzimático (ELISA). A descrição da morfometria ovariana foi realizada em 100 ovários provenientes de fêmeas escolhidas aleatoriamente. Foi mensurado o comprimento maior e menor dos ovários e, também, contabilizado o número total de FPs e FGs presentes na superfície de cada ovário. A concentração de IGF-I foi superior (P<0,05) nos FGs (170,92±88,29ng/mL) em comparação com os FPs (67,39±49,90ng/mL) e o sérico (73,48±34,63ng/mL). O comprimento maior e menor dos ovários foi de 26,0±3,0mm e 19,0±2,0 mm, respectivamente. O número de FPs (70,86±25,76) foi maior (P<0,01) em comparação com os FGs (6,54±5,26). Conclui-se que FGs apresentam níveis de IGF-I superiores aos FPs, e ao sangue, sendo isso relacionado a maior atividade dos FGs e à diferenciação que os mesmos sofrem para a ovulação. Além disso, ovários de fêmeas suínas pré-púberes apresentam elevado número de FPs em comparação aos FGs. Portanto, nosso estudo demonstrou dados originais a respeito da concentração fisiológica de IGF-I livre em folículos ovarianos, que podem ser utilizados em futuras pesquisas para avaliar a adição desse hormônio nos meios de produção in vitro de embriões suínos com o objetivo de melhorar a eficiência da técnica.

E-mail: mitika.hagiwara@gmail.com Currently, the serovar Copenhageni is the representative of serogroup Icterohaemorrhagiae maintained in synanthropic rodents found most frequently in dogs and humans in metropolitan areas of Brazil. Despite some authors have suggested the existence of cross-protection between serovars included in the same serogroup, this condition has not yet been sufficiently clarified for serovars Icterohaemorrhagiae and Copenhageni. In the present work, 2 to 6-year-old dogs, vaccinated at 60, 90 and 120 days of age and thereafter, revaccinated annually with commercial vaccine containing Canicola, Icterohaemorrhagiae, Grippotyphosa and Pomona bacterins were evaluated as to the immune status against leptospirosis before and 30 days after revaccination. Mycroscopic agglutination test (MAT) and in vitro growth inhibition test (GIT) were performed to search for agglutinating anti-Leptospira antibodies and neutralizing anti-Leptospira antibodies, respectively for serovars Canicola and Icterohaemorrhagiae, and additionally, for serovar Copenhageni, not included in the vaccine. The results showed that the immunity conferred by the vaccine to serovar Icterohaemorrhagiae is more lasting than that observed for serovar Canicola, since neutralizing antibody titers >1.0 log10 were observed before the booster vaccination with no substantial increase after revaccination. As for the serovar Canicola, revaccination resulted in a considerable increase in neutralizing antibody titer when compared to the one observed previously to the revaccination (p=0.001). The analysis of the data obtained by GIT allowed us to conclude that dogs given vaccine containing Icterohaemorrhagiae bacterin did not produce neutralizing antibodies against serovar Copenhageni enough to inhibit leptopiral growth at the same level as occurred for the homologous serovar. Despite this, the GIT titer found for serovar Copenhageni before and after revaccination showed that at least, some level of protection could be expected for dogs vaccinated with serovar Icterohaemorrhagiae bacterin, not a complete cross protection.

E-mail: mitika.hagiwara@gmail.com Na atualidade, o sorovar Copenhageni é o representante do sorogrupo Icterohaemorrhagiae, mantido por roedores sinantrópicos, que tem prevalecido nos cães e seres humanos das grandes metrópoles brasileiras. A despeito de alguns autores sugerirem a existência de proteção cruzada entre sorovares incluídos em um mesmo sorogrupo esta condição ainda não foi suficientemente esclarecida para os sorovares Icterohaemorrhagiae e Copenhageni. No presente trabalho cães adultos com dois a seis anos de idade primo-vacinados com três doses intervaladas de 30 dias a partir dos 60 dias de idade e revacinados anualmente com vacina anti-leptospirose polivalente contendo os sorovares Canicola, Icterohaemorrhagiae, Grippotyphosa e Pomona foram revacinados com a mesma vacina e aos 30 dias da revacinação foram submetidos aos testes de soroaglutinação microscópica (SAM) e de inibição do crescimento de leptospiras in vitro (TICL), para avaliação comparativa dos níveis de anticorpos produzidos para os sorovares Canicola, Icterohaemorrhagiae e Copenhageni. Os resultados obtidos indicaram que a imunidade conferida pela vacina para o sorovar Icterohaemorrhagiae é mais duradoura que a observada para o sorovar Canicola, já que títulos de anticorpos neutralizantes >1,0 log10 foram observados antes do reforço vacinal não havendo substancial aumento após a revacinação. Quanto ao sorovar Canicola, a revacinação resultou em considerável aumento do título de anticorpos neutralizantes quando comparado ao momento anterior a revacinação (p=0,001). A análise dos valores encontrados após a revacinação demonstrou claramente que cães revacinados com bacterina produzida com o sorovar Icterohaermorrhagiae não apresentam aumento do título de anticorpos inibidores do crescimento contra o sorovar Copenhageni, em nível suficiente para inibir o crescimento de leptospiras. Apesar disso, os títulos de anticorpos inibidores de crescimento anti-Copenhageni encontrados antes e após a revacinação demonstraram que, pelo menos certo grau de proteção contra a infecção por esse sorovar pode ser esperado para os cães vacinados com bacterinas do sorovar Icterohaemorrhagiae, não sendo, no entanto, uma proteção cruzada completa.


#14 - Effect of oral administration of Propionibacterium acnes on growth performance, DTH response and anti-OVA titers in goat kids, 33(1):5-10

These substances can increase resistence to infection by different types of microorganisms, reducing dependence of antibiotics used in livestock animals. Recent reports have demonstrated the positive effect of Propionibacterium acnes (P. acnes) to control animal diseases. In this study, we evaluated the effect of the non-specific immunostimulant P. acnes on immunological functions and growth performance in goat kids. Twenty five goat kids served as control group (A) and another 25 animals received P. acnes being the experimental group (B). Kids were challenged with ovalbumin (OVA) to assess humoral immunity. To assess in vivo cell immunity, delayed type hypersensitivity (DTH) test with phytohemagglutinin (PHA) was used, clinical signs and body weight were recorded each week until 9 weeks of age when the experiment ended. Blood samples were obtained to analyze serum proteins fractions and anti-OVA specific antibodies. No clinical signs of disease and no differences (p>0.05) on body weight between groups were recorded (7.32±0.81 kg in group A, 7.13±0.65 kg in group B). Goat kids from group B had more total protein (59.8±5g/l) and albumin levels (32.8±3.3g/l) than goat kids from group A (56.6±5.7 g/l, 29.6±3.9 g/l respectively) (p<0.05). DTH response in goat kids from group B on day 42 was higher (p<0.05) than group A. At day 63, goat kids from group receiving P. acnes had higher percentage (85.4) of anti-OVA IgM titers (p<0.05) than control group (57.7). In conclusion, the results showed that oral administration of P. acnes to goat kids improved some aspects of the immune system of the animals and it could be used to control goat diseases.

E-mail: baccarin@usp.br Osteochondrosis is the most common orthopaedic developmental disorder in horses and has been traditionally defined as a multifactorial disease. The disease had a very dynamic character in which the repair process starts almost immediately after the lesion has formed. The intensity and efficacy of this repair process is strongly age-related and is determined by the turnover rate of the components of the extracellular matrix of the articular cartilage. Since the insulin-like growth factor-I (IGF-1) acts regulating the growth of articular cartilage, this study aimed to describe the serum levels of IGF-1, insulin and glucose in healthy Lusitano foals and foals with osteochondrosis, from birth to 18 months of age. At one month 76.08% of the foals showed radiographic signs of osteochondrosis, but at age of 18 months the percentage dropped to 16.2%. The IGF-1 concentrations reached peak values at age of 14 to 16 months, equivalent to the onset of puberty. The IGF-1 concentrations in foals with osteochondrosis were lower than in healthy 2 to 5-month-old foals (P<0.05), and the insulin at age of 2 months (P<0.05). On farms with high incidence of osteocondrosis is recommended that measurement of serum IGF-1 and insulin occur during the first 6 months of age, to have enough time to establish preventive and therapeutic measures.

Caracteriza-se por ser uma doença dinâmica, onde o processo de reparo inicia-se quase que imediatamente após a formação da lesão osteocondral. A intensidade e eficácia deste reparo são fortemente relacionados à idade, e estão associados a reposição dos componentes da matriz cartilagínea. Uma vez que o fator de crescimento tipo insulina-1 (IGF-1) age regulando o crescimento da cartilagem articular, este estudo objetivou descrever os níveis séricos de IGF-I, insulina e glicose em potros Puro Sangue Lusitano hígidos ou com osteocondrose, desde o nascimento até 18 meses de idade. Dos potros acompanhados, 76,08% apresentaram sinais radiográficos de osteocondrose com um mês de idade, porém somente 16,2% mantiveram as lesões aos 18 meses. As concentrações de IGF-1 alcançaram picos entre 14 e 16 meses, equivalente ao início da puberdade. As concentrações de IGF-1 nos potros com osteocondrose foram menores que potros sadios, aos 2 e 5 meses (P<0,05), e os de insulina aos 2 meses (P<0,05). Em criatórios com alta ocorrência de osteocondrose recomenda-se que as mensurações dos níveis séricos de IGF-1 e insulina ocorram durante os primeiros 6 meses de idade, para que haja tempo hábil de se estabelecer medidas profiláticas e de tratamento.

The effects in calves of cow Cu supplementation in the last one-third gestation period were also evaluated. Five supplementation trials, with a total of 300 calves, were carried out. Two groups of 30 calves were randomly assigned to each trial, one group was parenterally supplemented (SG) and the other was not supplemented (NSG). Trials began when calves were three-month-old and ended at weaning time. At each sampling calves were weighed and blood was taken to determine Cu concentrations in plasma, Whole Blood (WB), Red Cells (RC) and Packed Cell Volume (PCV). Liver samples from six animals of each group were taken both at the beginning and at the end of the trial. In two trials the mothers of the SG received Cu supplementation at the last one- third gestation period. Four of the five trials exhibited low ADG in the NSGs. In these groups, plasma Cu concentration decreased rapidly before low ADG was detected, which occurred with values remaining below 25µg/dl. The decrease of RC Cu concentration was considerably slow. WB showed an intermediate position. PCV in the SGs was higher than in the NSGs in all trials. Cow supplementation was insufficient to generate a liver storage able to last after calves reached the 3 months of age. These data could be useful to predict the risk of low ADG in grazing calves.

Em cada ensaio os bezerros foram pesados e mostras do sangue foi tomada para determinar as concentrações Cu no plasma, sangue total (WB), eritrócitos (RC) e hematócrito (PCV). Amostras de fígado foram colhidas em seis animais de cada grupo tanto no início quanto no final do ensaio As mães do SG receberam suplementação de Cu no último terço da gestação em dois ensaios. Quatro dos cinco ensaios apresentavam baixa ADG nos NSGs. Nesses grupos, a concentração plasmática de Cu diminuiu rapidamente antes que a baixa do ADG fossei detectada, o que ocorreu com os valores abaixo 25 µg/d. A diminuição da concentração de RC Cu foi bastante lenta. WB mostrou uma posição intermédia. PCV nos SGs foi mais elevado do que nos NSGs em todos os ensaios. A suplementação das vacas foi insuficiente para gerar um armazenamento hepático capaz de durar até os 3 meses de idade dos bezerros. Estes dados poderiam ser úteis para prever o risco de baixa ADG em bezerros em pastagem.


#17 - Effect of medroxy-progesterone acetate on follicular growth and endometrial cycloxygenase-2 (COX-2) expression during the bovine estrous cycle, 30(7):581-585

E-mail: valerio.portela@gmail.com The objective of this study was to evaluate the effect of medroxy-progesterone acetate (MAP) with or without estradiol benzoate (EB) on follicular growth during the estrous cycle in cattle. In the first experiment, Hereford cows were synchronized with a synthetic analogue of PGF2 alpha and were treated with two different doses of MAP (250 or 500 mg) with or without EB for 7 days starting on day 8 of the estrous cycle. Follicular growth was inhibited (P<0.05) in all cows except controls and those receiving 250mg MAP without EB. Seventy-five percent of the animals (15/20) showed estrus on days 21 and 22 of the cycle rather than at MAP withdrawal, demonstrating that these treatments did not induce estrus. To determine whether the EB treatment altered endometrial sensitivity to oxytocin and thus the luteolytic cascade, multiparous pre-synchronized cows received 5 mg of EB followed 6 hours later with 50 IU of oxytocin (OT; n=9). Eight hours after EB injection, endometrial fragments were collected from the cows on days 4, 13 and 17 of the estrous cycle and COX-2 gene expression was measured by PCR. EB increased COX-2 mRNA levels only on day 17 of the estrous cycle (P<0.05). In conclusion, MAP alone or associated with EB is able to suppress bovine follicular growth. However, EB in the presence of MAP is not efficient to induce luteolysis in cows when injected on day 8 of the estrous cycle.

Observou-se uma inibição do crescimento folicular (P<0,05) em todas as vacas, exceto no grupo controle e no grupo que recebeu 250mg de MAP sem BE. Os 75% dos animais não exibiu estro no momento da remoção do MAP, mas sim nos dias 21 e 22 do ciclo, demonstrando que os tratamentos não induziram cio. Para se determinar se o tratamento com BE alterou a sensibilidade endometrial à ocitocina e, assim, a cascata luteolítica, vacas multíparas pré-sincronizadas receberam 5mg de BE, seguidos, após 6 horas, de 50 UI de ocitocina (OT; n=9). Oito horas após a administração de BE, colheram-se fragmentos endometriais das vacas, nos dias 4, 13 e 17 do ciclo estral, mensurando-se a expressão gênica de COX-2 através de PCR. O BE aumentou os níveis de RNAm de COX-2 apenas no dia 17 do ciclo estral (P<0,05). Em conclusão, o MAP isolado ou associado a BE é capaz de suprimir o crescimento folicular bovino. Entretanto, o BE, na presença de MAP é ineficaz na indução da luteólise bovina, quando injetado no oitavo dia do ciclo estral.

Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. From 242 pigs with PMWS, 57 (23.5%) showed purulent lesions in the middle ear. Among 119 pigs with attrition, only 1 (0.7%) presented the lesion. In 24 control pigs, middle ear lesions were not detected. The most frequently isolated agents from the lesions were Arcanobacterium pyogenes (43%), á–hemolytic Streptococci (37.2%) and Pasteurella multocida (27.9 %) out of 86 materials. The frequent occurrence of purulent lesions in the middle ear of PMWS affected pigs suggests that PCV2 infection may increase the susceptibility to bacterial otitis. The low occurrence of this disease in piglets with attrition suggests that otitis does not represent a significant cause for depressed growth in pigs from growing and finishing age. The isolation of A. pyogenes, á-hemolytic Streptococci and P. multocida from most of the lesions emphasizes the importance of these organisms as causal agents of otitis media in swine.

The Sm-poisoned rabbits had an increase of bone resorption in the endosteal surface of the cortical zone and also in the surface covered by osteoblasts of the primary and secondary spongiosa of the trabecular bone compartment. Moreover, the epiphyseal growth plates in long bones appeared narrower than in the control rabbits, with reduction of the proliferative and hyperthrophic chondrocyte zones. The electron microscopic study revealed a significant decrease of proteoglycans in the hyperthrophic chondrocyte zone evidenced by a significant reduction of rutenium red positive granules in the poisoned rabbit. Altogether, these data suggest that cell differentiation may play a pivotal role in the pathogenesis of Sm-induced bone lesions.
