E-mail: savasco@usp.br The diagnostic value of the cutaneous allergic response to tuberculin in piglets experimentally sensitized intramuscularly with the oily suspensions of heat inactivated M. bovis or M. avium was investigated. Ninety-one animals were used and divided into four groups: groups A and B were formed each with 25 individuals, and groups C and D, with 21 and 20 individuals, respectively, balancing the characteristics of race, ancestry, age and sex. At the age of 30 days, all the animals were submitted to the screening test with the use of M. bovis PPD, by the intradermal route at the base of the ear and no reaction was detected. Sixty days after the screening tuberculin test, animals of the group A were injected intramuscularly with 0.5 mL of oily suspension of M. avium D4 strain; animals of the group B received 0.5 mL of an oily suspension of M. bovis, AN5 strain; group C (control I) received 0.5 mL of an oily adjuvant; and the individuals of the group D (control II) received 0.5 mL of saline solution. Following 30 days of sensitization, comparative skin reactions were measured by the variation in skin thickness with a caliper at 0h, 24h, 48h an 72h after applications of tuberculins. In the comparative test measured at 48 or 72h, the reaction was considered negative when the difference of the reactions between bovine PPD and avian PPD was less than 6.7 mm; suspected or inconclusive, when the difference stood in the range of 6.7 to 7.5 mm; and positive according to the type of PPD, considering tuberculosis the M. bovis PPD and mycobacteriosis the M. avium PPD, when the difference of the reaction was greater than 7.5 mm.

E-mail: savasco@usp.br Foi investigado o valor diagnóstico da resposta alérgica cutânea em leitões experimentalmente sensibilizados, pela via intramuscular, com suspensões oleosas de Mycobacterium bovis ou M. avium inativados pelo calor. Foram utilizados 91 animais, divididos em quatro grupos: grupos A e B, cada um com 25 indivíduos, grupos C e D com 21 e 20 indivíduos respectivamente, balanceando-se as características de raça, linhagem, faixa etária e sexo. Aos 30 dias de idade, todos os animais foram submetidos a uma triagem com a aplicação de tuberculina PPD bovina, pela via intradérmica na base da orelha e não houve qualquer tipo de reação. Decorridos 60 dias do teste tuberculínico de triagem, o grupo A recebeu injeção intramuscular de 0,5 mL de uma suspensão oleosa de M. avium estirpe D4; o grupo B recebeu 0,5 mL de uma suspensão oleosa de M. bovis estirpe AN5; o grupo C (controle I), recebeu 0,5 mL do adjuvante oleoso; e o grupo D (controle II), recebeu 0,5 mL de solução fisiológica. Após 30 dias da sensibilização foi realizada a prova de tuberculinização comparativa com reação medida pela variação da espessura da pele com cutímetro de mola às 0h, 24h, 48h e 72h, após a aplicação das tuberculinas. No teste comparativo, lido às 48 ou 72 horas, a reação foi considerada negativa quando a diferença das reações entre o PPD bovino e o PPD aviário foi menor que 6,7 mm; suspeito ou inconclusivo quando a diferença se situou na faixa de 6,7 a 7,5 mm; e positiva de acordo com o tipo de PPD, considerando-se tuberculose para PPD M. bovis e micobacteriose para PPD M. avium, quando a diferença da reação foi superior a 7,5 mm.

E-mail: akaolar@usp.br The Duchenne’s muscular dystrophy (DMD) in humans is a X-linked neuromuscular disease, of recessive character, caused either by the absence or dysfunction of the dystrophin. Clinically, it is characterized by severe alteration in the skeletal musculature, resulting in precocious death. In Golden Retriever dogs, the mutation that takes to the muscular dystrophy happens spontaneously and the extensive homology among the pathogenesis of DMD and of Golden Retriever muscular dystrophy allows to qualify the dog as the main substitute of humans in the clinical tests of new therapies. The deficient myocardium in distrofin is more vulnerable to the pressure overload and the patients with DMD can develop dilated cardiomyopathy, arterial hypertension and the electrocardiogram can come distinctly abnormal. In the present study, 38 healthy Golden Retriever dogs were evaluated by electrocardiographic exam with the purpose to obtain parameters for the standardization of the electrocardiogram in the referred breed, what hereafter can serve as reference in the identification of bearer or affected dogs. Electrocardiographic values obtained were within normal values and reference for the various breeds of dogs, and the variables weight and age significantly altered heart rate and amplitude of the QRS complex.

E-mail: akaolar@usp.br A distrofia muscular de Duchenne (DMD) em humanos é uma alteração neuromuscular hereditária, de caráter recessivo, ligada ao cromossomo X e causada pela ausência ou disfunção da distrofina. Clinicamente, caracteriza-se por grave alteração na musculatura esquelética, resultando em morte precoce do indivíduo acometido. Em cães da raça Golden Retriever, a mutação que leva à distrofia muscular ocorre espontaneamente e a extensa homologia entre a patogênese da DMD e da distrofia muscular do Golden Retriever permite qualificar o cão como o principal substituto de humanos nos testes clínicos de novas terapias. O miocárdio deficiente em distrofina é mais vulnerável à sobrecarga de pressão e os pacientes com DMD podem desenvolver cardiomiopatia dilatada, hipertensão arterial e o eletrocardiograma pode se apresentar distintamente anormal. No presente estudo, foram avaliados exames eletrocardiográficos de 38 cães da raça Golden Retriever clinicamente sadios (20 animais de até 12 meses de idade e 18 animais entre 12 e 36 meses de idade), com a finalidade de se obter parâmetros para a padronização do eletrocardiograma nessa referida raça, o que futuramente poderá servir de referência na identificação de cães portadores ou afetados pela distrofia muscular. Os valores eletrocardiográficos obtidos encontraram-se dentro dos valores de normalidade e referência para as diferentes raças de cães; e as variáveis peso e idade alteraram significativamente a freqüência cardíaca e a amplitude do complexo QRS.

E-mail: angsaf@usp.br Electroretinogram (ERG) is an objective non invasive diagnostic method to evaluate retinal function which permits the early detection of lesions at retinal external layers, even before the appearance of clinical signs. In dogs, ERG is mostly utilized for preoperative evaluation in patients presenting cataracts; characterization of disturbances causing blindness, among the utilization of dogs as animal models in scientific research. Several factors interfere in ERG responses, such as the ERG machine, light stimulation, electrode, time spent on dark adaptation, pupil size, means opacity, sedation or anesthetization, species, breed and age. The purpose of this study was to standardize the full field ERG in sedated dogs, according to the International Society for Clinical Electrophysiology of Vision (ISCEV) protocol, using Ganzfeld and bipolar electrodes. Two hundred thirty threes ERGs were performed in 147 female and 86 male with ages from 1 to 14 years old. Among those 233 dogs, 100 presented cataracts in different stages of maturation, 72 were diabetic and presented mature or hypermature cataracts, 26 presented ERG compatible to progressive retinal degeneration, 3 presented ERG compatible to sudden acquired retinal degeneration syndrome; for 32 dogs no abnormal ERG was registered and they were considered as having normal retinal function. Sedation was capable to induce a good immobilization with no bulb rotation, resulting uniform retinal stimulation, using Ganzfeld. Veris system successfully registered all 5 ISCEV responses, simultaneously from both eyes, at the same time. Full field ERG became a fundamental ophthalmic exam, then, its standardization is mandatory to allow comparison between ERGs from different laboratories. The reliability and reproducibility of this protocol has shown very good responses, using ISCEV protocol, Veris system, Ganzfeld and Burian Allen electrodes in sedated dogs.

E-mail: angsaf@usp.br Eletrorretinograma (ERG) é o meio diagnóstico objetivo e não-invasivo para avaliar a função retiniana e detectar precocemente, em várias espécies, lesões nas suas camadas mais externas. As indicações mais comuns para ERG em cães são: avaliação pré-cirúrgica de pacientes com catarata, caracterização de distúrbios que causam cegueira, além de servir como importante modelo para o estudo da distrofia retiniana que acomete o homem. Vários são os fatores que podem alterar o ERG tais como: eletrorretinógrafo, fonte de estimulação luminosa, tipo do eletrodo, tempo de adaptação ao escuro, tamanho pupilar, opacidade de meios e protocolo de sedação ou anestesia; além da espécie, raça e idade. Objetivou-se com este estudo padronizar o ERG para cães submetidos à sedação, seguindo o protocolo da International Society for Clinical Electrophysiology of Vision (ISCEV), utilizando Ganzfeld e eletrodos Burian Allen. Foram realizados 233 eletrorretinogramas em cães, 147 fêmeas e 86 machos, com idades entre um e 14 anos. Dos 233 cães examinados, 100 apresentavam catarata em diferentes estágios de maturação, 72 eram diabéticos e apresentavam catarata madura ou hipermadura, 26 apresentaram eletrorretinograma compatível com degeneração retiniana progressiva, três apresentaram eletrorretinograma compatível com síndrome da degeneração retiniana adquirida subitamente e 32 não apresentaram lesão retiniana capaz de atenuar as respostas do ERG, sendo considerados normais quanto à função retiniana. A sedação foi capaz de produzir boa imobilização do paciente sem rotacionar o bulbo ocular, permitindo adequada estimulação retiniana bilateralmente, com auxílio do Ganzfeld. O sistema eletrodiagnóstico Veris registrou com sucesso e simultaneamente de ambos os olhos, as cinco respostas preconizadas pela ISCEV. Como o ERG de campo total tornou-se exame fundamental na rotina oftalmológica, sua padronização é indispensável quando se objetiva comparar resultados de laboratórios distintos. A confiabilidade e reprodutibilidade deste protocolo foi demonstrada com a obtenção de registros de ótima qualidade utilizando protocolo padrão da ISCEV, eletrorretinógrafo Veris, Ganzfeld e eletrodos Burian Allen nos cães submetidos à sedação.

E-mail: marcelloroza@gmail.com Eleven dogs and four cats with buccodental alterations, treated in the Centro Veterinário do Gama, in Brasilia, DF, Brazil, were submitted to cone beam computed tomography. The exams were carried out in a i-CAT tomograph, using for image acquisition six centimeters height, 40 seconds time, 0.2 voxel, 120 kilovolts and 46.72 milliamperes per second. The ideal positioning of the animal for the exam was also determined in this study and it proved to be fundamental for successful examination, which required a simple and safe anesthetic protocol due to the relatively short period of time necessary to obtain the images. Several alterations and diseases were identified with accurate imaging, demonstrating that cone beam computed tomography is a safe, accessible and feasible imaging method which could be included in the small animal dentistry routine diagnosis.

E-mail: marcelloroza@gmail.com Onze cães e quatro gatos, portadores de alterações buco-dentárias e atendidos no Centro Veterinário do Gama, em Brasília, DF, foram submetidos à tomografia computadorizada de feixe cônico. Os exames foram realizados em um tomógrafo i-CAT, utilizando para aquisição das imagens, altura de seis centímetros, tempo de 40 segundos, 0,2 voxel, 120 kilovolts e 46,72 miliampéres por segundo. O melhor posicionamento dos animais para realização do exame foi definido neste estudo. Esse é um fator fundamental para a realização do exame, que necessitou um protocolo anestésico simples e seguro, em função do tempo mínimo necessário à obtenção das imagens. Várias alterações e enfermidades foram identificadas, com extrema acurácia, credenciando a tomografia computadorizada de feixe cônico como um exame seguro, acessível e exeqüível e que pode ser incorporado à rotina odontológica das clínicas de pequenos animais.

One polyclonal antibody and two monoclonal antibodies were used. The following reagents were evaluated for antigen retrieval: XIV protease, proteinase K and citrate buffer (pH 6.0) boiling at 100°C during 15 minutes in bain-marie. Detection of rabic antigen was possible with the three antibodies tested. The polyclonal antibody was superior to the monoclonal antibodies, demonstrating good results with the three antigen retrieval protocols. The highest intensity staining was obtained with the citrate buffer and heat. The immunohistochemistry technique demonstrated the presence of viral antigens in the cytoplasm of neurons, in form of aggregates or with round or oval shape. The antigens were found as single or multiples inclusion bodies in the neurons. Immunohistochemistry is a fast method that can be used in routine procedures in cases where rabies is suspected, especially when the brain is submitted to the laboratory as formalin-fixed fragments or when samples could not be immediately shipped. The technique is also useful for retrospective studies.

[Desenvolvimento e padronização de um ELISA indireto para o diagnóstico sorológico de peste suína clássica.] Pesquisa Veterinária Brasileira, 19(3/4):123-127. Equipe de Virologia, FEPAGRO- Centro de Pesquisas Veterinárias Desidério Finamor, Caixa Postal 2076, Porto Alegre, RS 90001-970, Brazil. An indirect enzyme linked immunoassay (ELISA-I) was developed and standardized for the serological diagnosis of classical swine fever (CSF). For the comparison, nine hundred and thirty-seven swine serum samples were tested by serum neutralization followed by immunoperoxidase staining (NPLA), considered ·as the standard. Of these, 223 were positive and 714 negative for neutralizing antibodies to classical swine fever virus (CSFV). In relation to the NPLA, the ELISA-I presented a 98.2% sensitivity; 92.86% specificity, 81.11% positive predictive value, 99.4% negative predictive value and a 94.1% precision. Statistical analysis showed a very strong correlation (r=0,94) between both tests. When compared to a commercially available ELISA kit, the performance of both, in relation to the NPLA, was simila1: It was concluded that the ELISA-I is suitable for large scale screening of antibodies to classical swine fever virus, although it does not distinguish antibodies to classical swine fever vírus from those induced by other pestiviruses.

[Desenvolvimento e padronização de um ELISA indireto para o diagnóstico sorológico de peste suína clássica.] Pesquisa Veterinária Brasileira, 19(3/4):123-127. Equipe de Virologia, FEPAGRO- Centro de Pesquisas Veterinárias Desidério Finamor, Caixa Postal 2076, Porto Alegre, RS 90001-970, Brazil. Um ensaio imunoenzimático do tipo ELISA indireto (ELISA-I) foi desenvolvido e padronizado para o diagnóstico sorológico de peste suína clássica. Na comparação foram utilizadas novecentas e trinta e sete amostras de soros suínos, as quais foram testadas pelo teste de soroneutralização seguido de revelação por imunoperoxidase (NPLA), tomado como padrão, resultando em 223 amostras positivas e 714 negativas. Em relação ao NPLA, o ELISA-I apresentou sensibilidade de 98,21%, especificidade de 92,86%, valor preditivo positivo de 81, 11%, valor preditivo negativo de 99,4% e precisão de 94, 1 %. A análise estatística dos resultados revelou uma correlação muito forte (r=0,94) entre os dois testes. Quando comparado com um "kit" de ELISA disponível comercialmente, a performance de ambos em relação ao NPLA foi similar. Concluiu-se que o ELISA-I é um teste apropriado para triagem em larga escala de soros para a detecção de anticorpos contra o Vírus da Peste Suína Clássica (VPSC), embora não seja capaz de diferenciar entre anticorpos induzidos pelo VPSC ou outros pestivírus.
