E-mail: davetpat@ufrgs.br This paper reports the natural poisoning by Sida carpinifolia (guanxuma, chá-da-índia) in cattle in Rio Grande do Sul, Brazil. Five cattle were affected in the period 2001-2008. Clinical signs included weight loss, incoordination, walking difficulty, generalized tremors, frequent falls, and death. Microscopically, the main changes were vacuolation of Purkinje neurons in the cerebellum, pancreatic acinar cells, and thyroid follicular cells. Transmission electron microscopy revealed vacuoles bordered by membrane containing finely granular material. Lectin histochemistry showed positive staining in neurons with the lectins Concanavalia ensiformis (Con-A), Triticum vulgaris (WGA), and Succinyl Triticum vulgaris (sWGA).

Foram afetados cinco bovinos no período 2001-2008. O quadro clínico foi caracterizado por emagrecimento, incoordenação, dificuldade de locomoção, tremores generalizados, quedas frequentes e morte. Microscopicamente, as principais alterações foram vacuolização dos neurônios de Purkinje do cerebelo, das células acinares do pâncreas e das células foliculares da tireoide. A microscopia eletrônica evidenciou vacúolos com conteúdo finamente granulado e delimitado por membrana. Na lectina-histoquímica, observou-se marcação em neurônios com as lectinas Concanavalia ensiformis (Con-A), Triticum vulgaris (WGA) e Succinyl Triticum vulgaris (sWGA).

E-mail: davetpat@ufrgs.br Spontaneous poisoning of livestock by Baccharis coridifolia affects mostly hungry or curious animals that have not entered in contact with the plant previously. The plant occurs usually in dried hilly soils of Rio Grande do Sul and neighboring states or countries. An indistinguishable and sporadic form of the disease has been associated with the ingestion of Baccharis megapotamica, a species occurring in moist areas. This communication reports the spontaneous poisoning of four lambs after ingestion of B. megapotamica var. weirii. Clinical signs were observed in three lambs, the other was found dead. A rapid clinical disease characterized by anorexia, colic, dehydration, and diarrhea resulted in death after 8-24 hours. Necropsy revealed significant changes in the rumen, in which serosal edema, and submucosal echimotic hemorrhages were observed. Microscopically, the rumen showed submucosal edema and mucosal tumefaction, vacuolization, and necrosis. Diagnosis was based on clinical, pathological and epidemiological findings.

E-mail: davetpat@ufrgs.br As intoxicações por Baccharis coridifolia afetam especialmente animais de fazenda famintos ou curiosos e que não haviam tido contato prévio com a planta. B. coridifolia ocorre usualmente em terrenos secos de coxilhas no Rio Grande do Sul e estados ou países vizinhos. Uma forma indistinguível e esporádica da doença tem sido associada com a ingestão de Baccharis megapotamica que ocorre em áreas úmidas. Relata-se a intoxicação natural de quatro cordeiros após ingestão de Baccharis megapotamica var. weirii. A doença foi observada em uma propriedade localizada em Barra do Ribeiro, Rio Grande do Sul. O rebanho era composto por 220 ovinos, os quais eram mantidos em área de pastagem nativa sem qualquer suplementação. Uma rápida doença clínica caracterizada por anorexia, cólica, diarréia e desidratação causou a morte de três cordeiros em um período de 8 a 24 horas, o outro foi encontrado morto. A necropsia revelou alterações significativas no rúmen, no qual havia edema de serosa e hemorragias equimóticas na submucosa. Microscopicamente, o rúmen apresentou edema de submucosa, além de edema, tumefação, vacuolização e necrose de mucosa. O diagnóstico foi fundamentado nos achados clínicos, patológicos e epidemiológicos.

E-mail: davetpat@ufrgs.br Fulguração é a morte de animais pela ação de correntes elétricas atmosféricas ou raios durante tempestades. Quatro bovinos provenientes de dois rebanhos foram encontrados mortos nas proximidades de árvores de Eucalyptus sp. imediatamente após registros de tempestades no Estado do Rio Grande do Sul. Nos locais haviam árvores com linhas de queimadura recente no tronco, cascas parcialmente soltas, além de restos de galhos e folhas espalhados sob as copas das árvores. Não foram encontradas lesões macroscópicas e microscópicas significativas, nem indícios de enfermidades tóxicas ou infecciosas. Tais achados sugerem morte súbita por fulguração nos bovinos.

E-mail: davetpat@ufrgs.br Lightning strike may cause sporadic deaths of animals that have been exposed to high-voltage electric currents during thunderstorms. Four healthy adult cattle from two herds were found dead next to eucalypt trees immediately after storms in Rio Grande do Sul, Brazil. There were recent burns and loose barks on the trunks of the trees, apart numerous branches and leaves scattered on the ground under the canopy of the trees. No gross or microscopic lesions were observed. In addition, there was no evidence of any toxic or infectious disease. These findings suggest that lightning strike caused the death of those animals.

[Clinical and pathological aspects in cattle affected by rabies with special reference to the rabies antigen mapping by immunohistochemistry.] Aspectos clínicos e patológicos em bovinos afetados por raiva com especial referência ao mapeamento do antígeno rábico por imuno-histoquímica. Pesquisa Veterinária Brasileira 29(11):899-904. Setor de Patologia Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Av. Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@ufrgs.br This retrospective study included clinical and pathological findings from 15 cattle affected by rabies. Thirteen of the 15 cases were confirmed by direct immunofluorescence. Cattle between 4 months and 8 years of age were affected. Clinical course ranged from 3 to 7 days. Paralytical form was the most common clinical picture and included incoordination, paresis, and paralysis of the pelvic members, besides recumbence, paddling, and death. The main histopathological findings were lymphoplasmacytic meningoencephalitis associated with characteristic Negri bodies in 86.6% of the cases. All cases showed anti-rabies immunostaining, which were most prominent in the brainstem including medulla oblongata, pons, and midbrain, besides trigeminal ganglion. Positive labeling was present within axons, dendrites, and perikarya of neurons as aggregates of granules or round formations associated with varying numbers of inclusion bodies. Immunostaining was also observed in the Purkinje neurons and their processes in the molecular layer, in the neurons of of the brainstem, and deep layer of the telencephalic cortex. Immunohistochemistry may be an important auxiliary tool for the diagnosis of rabies, especially in circumstances in which refrigeration cannot be adequately maintained, and in cases characterized by nonsuppurative meningoencephalitis with absence of inclusion bodies.

Todos os casos foram positivos na imuno-histoquímica para raiva, cujas reações foram mais evidentes no tronco encefálico, incluindo bulbo, ponte e mesencéfalo, além de gânglio trigêmeo. A imuno-histoquímica demonstrou o vírus da raiva em axônios, dendritos e pericário de neurônios, como agregados de grânulos ou em formações arredondadas associadas com números variáveis de corpúsculos de inclusão virais nos neurônios. Houve também marcação nos neurônios de Purkinje e de seus processos na camada molecular, nos núcleos do tronco encefálico e camada profunda do córtex telencefálico. A imuno-histoquímica pode ser importante ferramenta diagnóstica no diagnóstico da raiva, especialmente em situações nas quais não é possível manter refrigeração adequada das amostras e em casos com meningoencefalite não-supurativa e ausência de corpúsculos de inclusão.

Samples of this by-product, in which pure cultures of A. clavatus grew, originated from two farms where outbreaks of A. clavatus poisoning in cattle had occurred. The onset of symptomatology was 2 to 6 days after dosage with the contaminated beer by-product or pure A. clavatus culture. The clinical course lasted from one and a half to 12 days. Clinical signs were predominantly of locomotor and respiratory nature and included muscle tremors, hyperesthesia, and progressive tachypnea, rigidity of the pelvic limbs, posterior weakness, and recumbency. One sheep also showed occasional knuckling of fetlocks of the hind limbs. Gait abnormalities and tremors were more pronounced after exercise. In 6 of 7 sheep, appetite and dypsia were maintained until close to death or euthanasia. The main histological findings consisted of chromatolytic neuronal degeneration and necrosis in selected nuclei of the brain stem, the ventral horn of the spinal cord and the spinal, trigeminal, stellate and celiac ganglions. Three sheep also presented slight degenerative and necrotic changes in muscles of the pelvic and thoracic limbs.

The highest intensity staining was obtained with the citrate buffer and heat. The immunohistochemistry technique demonstrated the presence of viral antigens in the cytoplasm of neurons, in form of aggregates or with round or oval shape. The antigens were found as single or multiples inclusion bodies in the neurons. Immunohistochemistry is a fast method that can be used in routine procedures in cases where rabies is suspected, especially when the brain is submitted to the laboratory as formalin-fixed fragments or when samples could not be immediately shipped. The technique is also useful for retrospective studies.

The disease occurred in June-July 2006 on a farm located in the county of Encruzilhada do Sul, Rio Grande do Sul, Brazil. Although there were 11 cattle in the same paddock, none of them was affected. High numbers of compact masses containing up to 150 larvae were scattered in the paddock where the animals were grazing. Most affected sheep showed severe apathy during 24-36 h before death, but weakness, muscular tremors and depression were also observed. Necropsy was performed on six sheep and the main macroscopic lesions were hemor-rhages in the subcutaneous tissues, endocardium, gallbladder wall, and abomasal mucosa. In all animals was found hydrothorax, hydropericardium, ascites, and mild jaundice. Edema in the abomasal folds, mesentery, perirenal tissues, and gallbladder wall were also seen. The livers were yellowish with disseminated pinpoint hemorrhages in the parenchyma and had an enhanced lobular pattern. Perreyia flavipes larval body fragments and heads were found in the forestomach contents of the six sheep. Feces were scant, dry and formed balls coated by mucus and streaks of blood. Similar contents were also present at the end of the cecum. Prominent microscopic lesions included severe and diffuse periacinar or massive necrosis of hepatocytes associated with multifocal random hemorrhages. Diffuse necrosis of lymphoid follicles in lymph nodes and Peyer´s patches, lymphoid depletion and necrosis in germinative centers of the spleen, and diffuse vacuolization in the renal tubular epithelia were also seen.

In July 2004, eight Corriedale sheep showed abortion and stillbirth in the third trimester of gestation. Of these reproductive losses, one stillborn male was submitted to the Laboratory of Veterinary Pathology for necropsy investigation. The direct immunofluorescence test for Leptospira sp. was negative. No significant bacteria was isolated from lung and liver by aerobic and microaerobic cultures. Macroscopic lesions were not found in any fetal tissue. The histological lesions were observed mainly in the brain and heart and consisted primarily of severe multifocal nonsupurative encephalitis and nonsuppurative myocarditis. Schizonts of a protozoan parasite consistent with Sarcocystis sp. were found in the endothelial cells and vascular endothelium in several organs. Many schizonts with merozoites arranged in a rosette-like pattern were observed in brain and kidney tissues. In sections stained with periodic acid-Schiff (PAS), the limiting membrane of some schizonts appeared to be weakly PAS-positive. Merozoites and nuclei were PAS-negative. Protozoa did not react immunohistochemically to the antibody anti-Toxoplasma gondii; however, cross-reactivity was observed with Neospora caninum antibody. These findings were consistent with the diagnosis of Sarcocystis sp.

Other histological changes included lymphoplasmacytic interstitial nephritis, necrotizing lymphadenitis and periportal lympho-plasmacytic cell infiltrates. While bacteriological and direct immunofluorescent antibody tests for Leptospira sp. were negative in samples from all cases, immunohistochemical and PCR procedures for Toxoplasma gondii reacted positively in most of them. Antibody titles against T. gondii varying from 1:512 to 1:2048 were detected in serum samples from the mother goats of these aborted (1), stillborn (3) or dead newborn (2) kids. This paper describes the clinical, pathological, serological, molecular and immunohistochemical findings of a Toxoplasma gondii-infected goat flock.
