#1 - Experimental infection by Trypanosoma vivax in goats in the Brazilian semiarid: detection of T. vivax DNA in colostrum and assessment of lactogenic transmission

Abstract in English:

This study aimed to identify the presence of Trypanosoma vivax DNA in the colostrum of infected goats and to explore the possibility of transmission for neonates fed using colostrum collected from infected goats. We used twelve goats in the final third of gestation with an age of approximately 24 months. Six goats were inoculated intravenously with 0.5mL of blood containing approximately 1.25x105 trypomastigotes of T. vivax, and six remained uninfected. The presence of T. vivax in colostrum was evaluated by Polymerase Chain Reaction (PCR). The possibility of T. vivax transmission by colostrum was assessed by feeding six neonates born of serologically negative goats using colostrum from infected goats. Peripheral blood from neonates was collected daily for thirty days to assess the T. vivax presence through the examination of Giemsa-stained smears of leukocyte layers with the buffy coat technique (BCT) and by PCR. The results of a direct examination of colostrum were negative, but PCR confirmed the presence of T. vivax DNA in all infected goats. Additionally, lactogenic transmission by colostrum was not demonstrated once both BCT and PCR of neonate peripheral blood were negative.

#2 - Listeria innocua and serotypes of Listeria monocytogenes isolated from clinical cases in small ruminants in the northwest of Uruguay

Abstract in English:

Listeriosis is an infectious disease caused by bacteria of the genus Listeria, the neurological form being more common in ruminants. There are many reports of listeriosis in small ruminants in the region that includes Brazil, Argentina and Uruguay. However, these diagnoses were mainly based on histological lesions in the central nervous system (CNS) without the isolation and characterization of the involved Listeria strains. The aim of this study was to report sheep and goats listeriosis cases from 2016 to 2021 in northwestern Uruguay. The diagnosis was made according to lesions observed at histopathology, plus Listeria isolation in CNS, identifying it at specie and serotype level. Nine animals (n=9) of three outbreaks and five sporadic cases of listeriosis were studied. Sheep was the species with more cases in relation to goats, and adults were the category most affected. Cases occurred in spring and less frequently in winter. All presented neurological clinical signs and the lesions in the CNS were consistent with suppurative meningoencephalitis and micro-abscesses in the brainstem. In eight of nine CNS samples, Listeria strains were isolated (seven L. monocytogenes and one L. innocua). All the L. monocytogenes isolates carried the inlA gene; serotyping showed that four strains belonged to serotype 1/2b, two isolates belonged to serotype 4b, and one to serotype 1/2a. Considering that listeriosis is a common disease in this region and the fact that isolates are scarcely recovered from small ruminants, it would be important to emphasize the need for Listeria isolation to better characterize the strains that affect animals. Not only to improve knowledge about the epidemiology of disease but also with the objective of developing serotype specific vaccines for animal use.

#3 - Detection and genotyping of Giardia duodenalis infecting pigs and small ruminants in the state of Piauí, northeastern Brazil

Abstract in English:

This study performed a molecular detection and characterization of Giardia duodenalis infecting pigs, goats and sheep in rural and peri-urban communities in the state of Piauí, northeastern Brazil, and proposed phylogenetic relationships among the characterized parasites. We assessed 52 fecal samples from pigs, 13 from goats, and 10 from sheep. A fragment of the β-giardin locus was PCR-amplified and sequenced. Overall, PCR-based G. duodenalis positivity was 11/52 (21.2%) in pigs, 2/13 (15.4%) in goats, and 2/10 (20%) in sheep. Seven out of 15 successfully amplified samples could be sequenced: three from pigs, two from goats, and two from sheep. Parasites from different hosts were found to belong to sub-assemblage AII. The phylogenetic analyses of the original G. duodenalis AII β-giardin sequences obtained from distinct host species and sequences of G. duodenalis recovered from humans available in GenBank suggest that the parasites are genetically related, supporting a local scenario of cross-host transmission.

#4 - Susceptibility of goats to poisoning by Tephrosia cinerea

Abstract in English:

Spontaneous and experimental poisoning by Tephrosia cinerea in the northeastern semiarid region of Brazil has only been described in sheep. Pathologically, such poisoning leads to ascites and centrilobular liver fibrosis. However, these effects require an experimental study in goats. This study aimed to determine the goats’ susceptibility to the ingestion of T. cinerea and the minimum toxic dose, describing the main clinical and anatomopathological findings. Poisoning was reproduced experimentally in one sheep that received 10g/kg of the ground plant and in two goats, the first receiving a dose of 5g/kg and the second receiving 10g/kg of the ground plant. The sheep presented abdominal distension 34 days after beginning the ingestion of the plant, developing sternal decubitus, breathing difficulty, opisthotonos, mandibular trismus, salivation, dysphagia, vocalization, and pedaling movements on the 50th day of the experiment. Fluid accumulation was observed in the abdominal cavity and liver via necropsy, with an irregular, slightly whitish capsular surface. Histologically, the main lesions observed in the liver were moderate fibrosis, marked sinusoidal distension, accompanied by marked hemorrhage, sometimes forming bridges between the centrilobular regions, associated with a dissociation of hepatocyte cords. There were discrete Alzheimer’s type II astrocytes in the gray matter in the region of the occipital cortex in the nervous system. Goat 2 showed apathy, drowsiness, and weight loss; on the 62th day, lateral decubitus evolved to sternal decubitus, with a rotation of the neck towards the flank. At necropsy, marked edema was observed on the face and dewlap, and a slight accumulation of liquid; slightly yellowish material was observed in the abdominal cavity. There were discrete blackened areas on the capsular surface in the liver. Histologically, the liver showed mild centrilobular fibrosis associated with mild dissociation of hepatocyte cords and mild vacuolar degeneration of the hepatocyte cytoplasm. Goat 1 showed no clinical signs; at necropsy, discrete multifocal areas were observed in the liver on the capsular surface. Histologically, diffuse intracytoplasmic vacuolar degeneration of hepatocytes was detected. The clinical picture and anatomopathological findings differ between the species, proving the lower susceptibility of goats to Tephrosia cinerea ingestion (compared to sheep), with differences in the pathogenesis and epidemiological aspects of poisoning.

#5 - Copper deficiency in dairy goats and kids

Abstract in English:

The clinical, pathological and reproductive aspects of an outbreak of copper deficiency in dairy goats and kids from the semiarid region of Pernambuco, Brazil are described. Ten adult dairy goats with clinical signs of deficiency and four kids presenting enzootic ataxia born from copper deficient does were separated from the herd, and examined. In the dairy goats, the average serum concentration of copper was 6.1±2.8mmol/L and iron was 39.5±8.2mmol/L. In kids, the average serum concentration of copper was 3.8±0.9mmol/L and iron was 38.5±4.1mmol/L. Clinical signs in dairy goats consisted of pale mucous membranes, anemia, emaciation, diarrhea, achromotrichia, brittle hair and alopecia. The main reproductive alterations consisted of prolonged anestrus, embryonic resorption and high indices of retained placenta. The kids born from copper deficient dairy goats were weak, and presented neonatal or late ataxia until 70 days of life. Six dairy goats and four kids were necropsied. Most ovaries examined were small, firm and did not present viable follicles on their surface. Microscopically, there was reduction of viable follicles in addition to disorganization of follicular and stromal structures, with marked follicular atresia. Microscopically, changes in kids with enzootic ataxia consisted of neuronal chromatolysis and axonal degeneration, mainly in neurons of the spinal cord. In this study, the source of high iron was not identified, but it is known that outbreaks of copper deficiency can occur due to excess iron intake, mainly when adequate mineral supplementation is not provided for the goat herds.

#6 - Proteomics characterization of Staphylococcus spp. from goat mastitis and phenogeno-typical assessment of resistance to beta-lactamics

Abstract in English:

Mastitis occupies a prominent place among the diseases that affect dairy herds due to economic problems and public health. Staphylococcus spp. are infectious agents more involved in the etiology of caprine mastites, especially coagulase-negative Staphylococcus. Nineteen isolates of Staphylococcus spp. were obtained from subclinical caprine mastitis. All isolates were characterized by MALDI-TOF MS, being 47.36% (9/19) identified for S. epidermidis, 15.78% (3/19) for S. warneri, 10.52% (2/19) for S. aureus and S. caprae and 5.26% (1/19) for S. lugdunensis, S. simulans, and S. cohnii. All isolates characterized by MALDI-TOF were subjected a to polymerase chain reaction (PCR) for the 16S rRNA gene of Staphylococcus spp. to confirm the gender. After determining the species, tests for phenotypic detection of resistance to beta-lactams were carried out simple disk diffusion oxacillin, cefoxitin, penicillin G and amoxicillin + clavulanic acid, agar “screen” oxacillin and microdilution (MIC) cefoxitin. The disk diffusion test showed a strength of 58% (11/19) for penicillin G, 26.31% (5/19) for cefoxitin and 26.31% (5/19) for oxacillin. All strains were susceptible to amoxicillin + clavulanic acid and agar “screen” oxacillin. In the MIC, 63.15% (12/19) of the samples were cefoxitin resistant (MIC >4.0µg/ml). Then isolates were subjected to detection of the mecA resistance genes and regulators (mecl and mecRI), mecC and blaZ. Two samples of Staphylococcus epidermidis had the mecA gene. All isolates were negative for the mecA gene variant, mecl, mecRI, mecC and blaZ. These findings reinforce the importance of this group of microorganisms in the etiology of subclinical mastitis in goats and open perspectives for future research to investigate the epidemiology of the disease.

#7 - Effect of flunixin or ketoprofen in caudectomy by elastration in lambs: pain and neutrophil function

Abstract in English:

Painful procedures can affect the function of innate immune cells, such as neutrophils and macrophages, increasing the risk of infectious diseases. The present work aimed to verify if the analgesics flunixin meglumine or ketoprofen can attenuate the pain/discomfort of newborn lambs submitted by elastration tail docking and thereby avoid the impairment of blood granulocytes function. Twenty-one neonate lambs were divided into three treatments: the control group (n=7), not subjected to caudectomy; the flunixin group (n=7), subjected to caudectomy under local anesthesia and analgesia with two doses of flunixin meglumine; and the ketoprofen group (n=7), subjected to caudectomy under local anesthesia and two doses of ketoprofen. Pain indicators were observed by pain posture score (PS), the number of vocalizations (V), frequency of the movement of the ears (EF), and respiratory rates (RR), observed by a 10 minutes videos for each time points: -15min, 6h, 48h, and 144h. At the same time points, the reactive oxygen species (ROS) production and phagocytosis of blood granulocytes were measured by flow cytometry. At 6h after caudectomy, there was a pain indicator increase (RR, V, and PS), a blood granulocyte percentage increase, and a granulocytes phagocytosis reduction for both groups. At 48h, the ketoprofen group spend more time in pain posture and, at 144h, they exhibited a ROS production granulocyte reduction without signs of pain. We conclude the flunixin meglumine and ketoprofen did not prevent the acute pain/discomfort caused by caudectomy, because the groups showed a pain behavior and impaired of the innate immune response however, the flunixin meglumine was effective in controlling the chronic pain and their effects on blood granulocytes function in compare ketoprofen.

#8 - Effects of a single trace minerals injection on antioxidant and immune response, resistance to endoparasites, health and growth of newborn Boer kids

Abstract in English:

This study evaluated the effects of injectable trace minerals (ITM) on antioxidant and immune response, resistance to endoparasites, health and growth of newborn Boer kids. Forty-six Boer kids [24 males and 22 females; 3.94±1.03kg of body weight (BW); 6.2±2.4 d of age] were enrolled in the study. Kids were stratified by type of birth (twins or singlet), sex, and BW and assigned to 1 of 2 treatments: one subcutaneous injection (0.1mL/4.5kg of BW) of (1) saline solution or (2) ITM (60, 10, 5, and 15mg/mL of Zn, Mn, Se and Cu, respectively). Blood samples were collected on d 0, 7, 14, 28 and 56. Feces samples were collected on d 56 and BW on d 0, 28 and 56. Kids were checked daily for signs of diarrhea. ITM kids had greater (P<0.01) plasma concentration of superoxide dismutase and tended (P=0.06) to have greater plasma concentration of glutathione peroxidase. ITM kids had greater (P=0.05) concentration of eosinophils, but no differences (P≥0.11) were observed for other hemogram variables. The ITM application did not affect (P≥0.11) the EPG count. However, ITM kids had less (P=0.02) cumulative incidence of diarhea until d 42 (3.85 vs. 25.93±6.8% for ITM vs. Saline kids, respectively) but no differences (P>0.10) were observed after d 42. The ITM application did not affect (P≥0.40) the growth of kids (0.071 vs. 0.065±0.005kg/day for ITM vs. Saline kids, respectively). Thus, the ITM application, increased the plasma concentration of antioxidant enzymes and eosinophils, decreased the incidence of diarrhea only in the middle of the experiment, but did not affected the EPG count and growth of Boer kids.

#9 - Spontaneous poisoning by Ateleia glazioviana (leg. Papilionoideae) in sheep and goats in the West region of Santa Catarina

Abstract in English:

This study reported the epidemiological data and the clinical and pathological conditions of spontaneous poisoning by Ateleia glazioviana in sheep and goats in the West region of Santa Catarina. The small ruminants were located in a place where there was a large amount of young plants of A. glazioviana and showing signs of consumption. The stock of sheep was composed of 250 animals, of which 45 died. In the goat herd, there were 28 animals, and of these, 27 died, and one was sacrificed. The main clinical signs were weight loss, fatigue, slow-walk, and eventually submandibular and facial edema. Some animals were found dead, and others died suddenly after being moved. Abortions and the birth of weak lambs were also observed. In total, five sheep and one goat were necropsied. The main macroscopic lesions were associated to eccentric cardiac hypertrophy and moderate, multifocal white areas in the epicardium and myocardium. Two sheep showed marked edema in the subcutaneous tissue of the head and neck and it could be noted hydrothorax and hydroperitoneum and liver with a nutmeg-like appearance. Histological changes were relevant in the myocardium. They consisted of fibrosis and myofiber necrosis associated with macrophage infiltrate, multifocal, low and marked swelling of cardiomyocytes with loss of fibrillarystriae, and increased nuclear volume. In two sheep, there was still congenital hepatic, as well as marked centrilobular and diffuse areas.

#10 - Ocular and oral lesions caused by Tacinga inamoena in sheep and goats in Northeast Brazil

Abstract in English:

Two outbreaks of traumatic injuries in goats and sheep associated with grazing and ingestion of the cactus Tacinga inamoena have recently been reported in the state of Paraíba, Brazil. In the first of these, which was detected in 2019 during a preliminary inspection of a herd of 70 animals, it was found that 15 showed certain ocular complications, and these animals were subjected to general physical and ophthalmic examinations. The clinical findings included excessive lacrimation, blepharitis, photophobia, corneal opacity, hyphema, corneal neovascularization, corneal ulcers, and adherence of gloquids to the bulbar conjunctiva and cornea. Large amounts of T. inamoena were found to be present in several areas in which these animals had been grazed and had been observed consuming the fruits of this plant. In the second outbreak during the following year, two sheep from a total of 100 animals were identified as having oral lesions associated with grazing in areas within which T. inamoena was growing. One of these animals was referred to a veterinary hospital, and on physical examination was found to show apathy, anorexia, a body score of 1.5, and the presence of gloquids within the fur. With respect to the oral cavity, halitosis, severe sialorrhea, extensive hemorrhagic ulcers, and painful tenderness on palpation were detected. Given the unfavorable prognosis, the sheep was subsequently euthanized. At necropsy, multifocal to coalescent plaques were detected on the upper and lower lips, oral mucosa, tongue, and hard and soft palates. These were generally elevated and firm and characterized by an irregular, ulcerated surface, sometimes crusty, with a yellow to light brown center and erythematous edges. The findings of this study highlight that the consumption of T. inamoena can cause serious oral and ophthalmic lesions in small ruminants in the northeastern semi-arid regions of Brazil, and can potentially contribute to substantial large economic losses. This problem tends to be exacerbated by the invasive behavior of T. Inamoena, particularly in areas in the process of environmental degradation.
