E-mail: fonteque@cav.udesc.br The poisonous snakes of the genera Crotalus and Bothrops have been kept in captivity with the purpose of extracting poison for the production of immunobiological. Knowledge of the physiology of these animals and serum proteins concentration changes are important for early identification of major diseases which lead to states of hypoproteinemia and hyperproteinemia. The objective was to determine the concentration of total protein and serum protein electrophoresis profile of Crotalus durissus terrificus (rattlesnake) in captivity. Blood samples were taken from the ventral coccygeal vein of 21 adult and healthy snakes divided into groups: Group 1 with 12 males, weighing in average 588.89±193.55g, and Group 2 with nine females, weighing in average 708.33±194.04g. The total serum concentration of protein was determined by the method of refractometry and agarose gel electrophoresis. The total protein values in the serum for females was 4.82±0.72, for males 4.51±0.50 and males and females 4.64±0.61, identified by four fractions (g/dL): albumin, a, b and g-globulin. Additionally the albumin/globulin ratio was calculated. The female snakes showed higher values for the variables, albumin and the albumin/globulin (AG) differed significantly (P<0.05) from the group of male snakes, but there was no clinical significance.

The commercial drugs used were moxidectin 0.2%, albendazole, levamisol chlorhydrate, ivermectin and an aqueous extract of the purgative potato (Operculina hamiltonii). The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. In larval culture Haemonchus, followed by Bunostomum, Trichostrongylus, and Oesophagostomum, were identified even after treatments.

[Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Avaliação clínica e hematológica em bezerros Nelore infectados experimentalmente com isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil. Pesquisa Veterinária Brasileira 23(2):52-60. Clínica de Bovinos, Campus Garanhuns, Univ. Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen,reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

O presente trabalho teve por objetivo estudar comparativamente as alterações clínicas e hematológicas desencadeadas por isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil em bezerros Nelore infectados experimentalmente. Foram utilizados 18 bezerros com idade entre sete e nove meses, isentos de anticorpos contra Babesia sp. e criados livres de carrapatos. Os outros 15 bezerros foram subdivididos em três grupos de cinco animais, que foram subinoculados com 1,0x1010 EP dos respectivos isolados. Foram avaliadas as alterações clínicas e hematológica por meio da determinação da parasitemia, do hemograma, do fibrinogênio plasmático, da contagem de reticulócitos, da análise descritiva da medula óssea e da fragilidade osmótica eritrocitária, no decorrer de 30 dias, perfazendo um total de sete momentos de observação. O acompanhamento da resposta imunológica pelo teste de imunofluorescência indireta foi realizado diariamente até o 10º dia pós-inoculação (DPI) e posteriormente no 15º, 20º, 25º e 30º DPI. Clinicamente, observou-se uma manifestação muito branda da doença. Os achados laboratoriais revelaram baixos níveis de parasitemia; decréscimo nos valores do eritrograma; ausência de reticulócitos; diminuição inicial na contagem total dos leucócitos, neutrófilos e linfócitos com posterior elevação do número destas células; hipercelularidade da série eritrocítica e decréscimo da relação mielóide:eritróide mais acentuada entre o 8º e 12º DPI e um aumento da fragilidade osmótica eritrocitária nos grupos inoculados com os isolados sudeste e nordeste. Nenhum dos três isolados de B. bigemina desencadeou a forma clínica característica da enfermidade, apesar de induzirem uma resposta imune humoral.

Ruminal acidosis is due to excessive ingestion of carbohydrates of rapid fermentation without previous adaptation of the microorganisms, causing severe metabolic disturbances to the animals. The objective of the present study was to assess the neutrophilic oxidative metabolism in sheep treated with sodium monensin in experimentally induced ruminal lactic acidosis. A total of 18 mate sheep, half-bred (Ideal x Merino), fistulated in the rumen, were used; nine of them received 33 mg/kg of the ionophore diet per day, for 30 days; the others were controls. The acidosis was induced by supplying 15g of sucrose/kg of body weight. The clinical evaluation and the rumen and blood samples were obtained before (Oh) and at 6, 12, 24 and 48 hours post-induction. In both groups, all the animals presented clinical manifestations of ruminal lactic acidosis 6 hours after the induction. From this period on, a significant pH decrease (P<0.05) was observed in the ruminal fluid, which reached levels below 5. There were relevant differences (P<0.05) between the groups 12 hours after the induction, when the sheep treated with monensin had higher values than those of the control group. During this period, the oxidative metabolism of the neutrophils remained inhibited, and the reestablishment of this function only occurred in the sheep which received monensin. Blood pH, plasmatic glucose and the ionizable calcium suffered alterations within its levels. The seric cortisol concentration rose significantly (P<0.05) in both groups, although differences (P<0.05) between them were found at the end of the observation period. The treatment with monensin did not influence the oxidative metabolism of the neutrophils inhibited by the lactic acidosis; however, a faster recovery of this metabolism was verified in the animals treated with the ionophore.

A acidose láctica ruminal é causada pela ingestão excessiva de carboidratos de fermentação rápida sem uma prévia adaptação dos mocroorganismos, podendo com isso gerar distúrbios metabólicos graves aos ruminantes. Este trabalho tem por objetivo estudar o metabolismo oxidativo dos neutrófilos em ovinos tratados com a monensina sódica na acidose láctica ruminal induzida experimentalmente. Foram empregados 18 ovinos, machos, mestiços (Ideal x Merino), fistulados no rúmen; dos quais nove receberam 33 mg/kg da dieta do ionóforo ao dia, durante 30 dias, os demais ovinos pertenceram ao grupo controle. A acidose foi induzida fornecendo 15g de sacarose/kg de peso corporal. A avaliação clínica e as amostras de rúmen e sangüíneas foram obtidas antes (momento controle) 0h e às 6h, 12h, 24h e 48h pós-indução. Em ambos os grupos os animais apresentaram manifestações clínicas de acidose láctica ruminal 6 horas pós-indução. A partir deste período se observou uma diminuição significativa (p< 0,05) do pH no fluido ruminal, que alcançou valores inferiores a cinco, e diferenças significativas (p< 0,05) entre os grupos a partir das 12h pós-indução foram constatadas, nos quais os ovinos tratados com a monensina apresentaram valores superiores em relação aos do grupo controle. Durante este período ocorreu uma inibição do metabolismo oxidativo dos neutrófilos nos animais estudados, ocorrendo restabelecimento desta função somente nos ovinos que receberam a monensina. O pH sangüíneo, a glicose plasmática e o cálcio ionizável sofreram alterações nos seus índices. A concentração do cortisol sérico elevou-se de forma significativa (p< 0,05) em ambos os grupos, porém diferenças (p< 0,05) entre eles foram constatadas ao final no período de observação. O tratamento com a monensina não influenciou o metabolismo oxidativo dos neutrófilos, inibido pela acidose láctica; porém se verificou uma recuperação mais rápida deste metabolismo nos animais em que receberam o ionóforo.


#25 - Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax, 21(4):157-161

[Clinical and laboratorial evaluation of Nellore cattle experimentally infected with Trypanosoma vivax] Avaliação clínico and laboratorial de bovinos nelore infectados experimentalmente com Trypanosoma vivax. Pesquisa Veterinária Brasileira 21(4):157-161. Embrapa Gado de Corte, Cx. Postal 154, Campo Grande, MS 79002-970, Brazil. In arder to evaluate the clinical-laboratorial alterations, six Neil ore calves were inoculated with 107 Trypanosoma vivax isolated from Poconé region, Mato Grosso, Brazil. The animals were evaluated daily for rectal temperature, packed cell volume (PCV), parasitemia, antibody production, calor of mucous membranes, behavior and appetite. Blood and serum samples for biochemical evaluation for aspartate aminotransferase (AST), alkaline phbsphatase (AF), gamma glutamyltransferase (GGT), cholesterol, urea, creatinine, creatine kinase (CK), calcium, phosphorus and proteinogram were collected on days 4, 8, 12, 16, 23 and 30 post inoculation (DPI). During the following 6 months rectal temperature, PCV and parasitemia were evaluated weekly. T. vivax was evidenced from 1 DPI in all calves and persisted until day 30 in five of six animals. A remarkable decrease (p<0.05) of PCV mean value (25%) was observed on 10 DPI. The animals presented no alterations in their clinical or serum biochemical state during the trial. Seroconversion took place 6 and 8 DPI, and all the animals remained seropositive during the 30 days of experiment. In all the experimental animals the occurrence of T. vivax infection was verified, characterized by the increase of corporal temperature, presente of the blood protozoa and reduction of the globular volume, without altéations in the other variables analyzed. Nellore calves, when experimentally inoculated with T. vivax, are able to establish a balance between host-parasite relationship.

Avaliaram-se as alterações clínico-laboratoriais de seis bezerros Nelore, de ambos os sexos, inoculados experimentalmente com 107 organismos viáveis de Trypanosoma vivax, isolados de bovinos da região de Poconé, Estado de Mato Grosso. Os animais foram observados diariamente, durante 30 dias, quanto aos parâmetros de temperatura retal, volume globular (VG), parasitemia, produção de anticorpos, coloração de mucosas, comportamento e apetite. Deteminaram-se os níveis séricos de aspartato aminotransferase (AST), fosfatase alcalina (FA), gama glutamiltransferase (GGT), creatina kinase (CK), colesterol, uréia, creatinina, cálcio, fósforo e o perfil eletroforético das proteínas séricas aos 4, 8, 12, 16, 23 e 30 dias pós-inoculação (DPI). Durante os 6 meses seguintes, os animais foram observados semanalmente, avaliando-se a temperatura retal, o VG e a parasitemia. T. vivax foi evidenciado a partir do terceiro e quarto DPI em todos os bezerros e persistiu até o 30º DPI em cinco dos seis animais em estudo. Ocorreu um decréscimo significativo (p<0,05) do valor médio do VG (25%) aos dez DPI. Os animais não apresentaram qualquer alteração no quadro clínico, bem como na avaliação da bioquímica sérica durante o período experimental. A soroconversão ocorreu aos 6 e 8 DPI, permanecendo todos os animais soropositivos nos 30 dias experimentais. Bovinos nelores jovens, infectados experimentalmente com T. vivax, foram capazes de estabelecer um equilíbrio na relação hospedeiro-parasita.


#26 - Clinical pathological data and analyses of mineral elements of cattle affected by epizootic botulism in the State of São Paulo

In arder to study laboratorial aspects of beef cow mortality, a syndrome popularly known as "doença da vaca caída", examens were made of blood, cerebrospinal fluid, serum, bone and liver samples from 32 naturally affected 4 to 9 year old cows, 27 belonging to the Nellore breed and5 were crossbred Nellore, all originating from farms located in municipalities near Botucatu, State of São Paulo. Laboratory determinations were analysed by descriptive statistics and included hematological values, total plasma protein, plasma fibrinogen, cerebrospinal fluid analysis, and concentration measurements of serum calcium, phosphorus, magnesium, sodium, potassium, chloride, total protein, albumin, globulin, alkaline phosphatase, aspartate aminotransferase, gama-glutamyltransferase and creatine kinase activities, included bane ash percentage and concentrations of calcium, phosphorus and magnesium, and also hepatic levels of copper, zinc, iron, manganese and cobalt. In addition, mouse bioassays and complement micro-fixation tests were performed to detect botulinum toxins in liver samples. The results indicated leukocytosis (13,3±3,9 x103/mm3) with neutrophilia (8,9±3,2 x103/mm3), hypocalcemia (7,8±1,7mg/dl), hypophosphatemia (3,6±1,6mg/dl), hypoalbuminemia (2,9±0,9g/cll), increased creatine kinase activity (691,0±829,7 UI/1), and reduced ash percentage (60,3±1,9%) and low phosphorus (17,2±0,4%) in bane. The other values were all within normal limits. The diagnosis of botulism, involving type C and D toxins, was confirmed as the cause of the mortality in the region of study, what is strongly consistent with the other laboratorial findings.

Com o objetivo de verificar alguns aspectos laboratoriais da mortalidade de vacas de corte, popularmente conhecida como "doença da vaca caída", estudaram-se amostras de sangue total, líquor, soro sanguíneo, osso e fígado colhidas de 32 fêmeas bovinas (27 da raça Nelore e 5 mestiças), entre 4 e 9 anos de idade, naturalmente acometidas e pertencentes a propriedades rurais localizadas em municípios próximos a Botucatu, Estado de São Paulo. Procederam-se as determinações e o estudo estatístico descritivo das variáveis: 1) hematológicas, de proteínas plasmáticas totais e fibrinogênio; 2) dos aspectos físicos, bioquímicos e celularidade no líquor; 3) de cálcio, fósforo, magnésio, sódio, potássio; cloretos, proteína total, albumina, globulina e atividades da fosfatase alcalina, aspartato aminotransferase, gama-glutamiltransferase e creatina quinase no soro sanguíneo; 4) de cinzas, cálcio, fósforo e magnésio no osso, e, 5) de cobre, zinco, ferro, manganês e cobalto no fígado, empregando-se as provas de inoculação em camundongos e de microfixação de complemento para a detecção da presença de toxinas botulínicas nas amostras de tecido hepático. Os resultados apontaramcomo alterações, leucocitose (13,3±3,9 x103/mm3) com neutrofilia (8,9±3,2 x103/mm3), hipocalcemia (7,8±1,7mg/ dl), hipofosfatemia (3,6±1,6mg/dl), hipoalbuminemia (2,9±0,9g/dl), elevação na atividade da creatina quinase (692,0±829,7 UI/1) e reduções na cinza (60,3±1,9%) e fósforo (17,2±0,4%) ósseos, como indicativos do estado carencial de fósforo, apresentando-se as demais variáveis dentro dos limites de variação considerados normais para a espécie. O diagnóstico de botullsmo, com envolvimento das toxinas C e D, foi confirmado como a causa da mortalidade investigada na área de abrangência do estudo, e encontra coerência com o conjunto de resultados.
