E-mail: ceambrosio@usp.br [Estudo do complexo valvar atrioventricular cardíaco esquerdo em búfalos (Bubalus bubalis) da raça Jafarabadi.] Foram estudados os complexos valvares atrioventricular esquerdo de 30 búfalos da raça Jafarabadi, machos e adultos, sem alterações cardíacas, provenientes de abatedouros do Estado do Paraná. Os corações foram examinados em seu terço médio para acesso ao complexo valvar, que foi submetido a estudos de área, número e tipificação de cordas tendíneas. Os resultados demonstram que este complexo é formado por duas cúspides principais e quatro cúspides acessórias, apresentam em sua formação de 2-3 músculos papilares, nos quais se inserem de 10-25 cordas tendíneas, que se fixam em cúspides voltadas para a parede do ventrículo. A área total deste complexo apresenta uma média de 38,56cm2 com um mínimo de 24,96cm2 e um máximo de 55,54cm2. Estatisticamente não há relação entre número de cordas e a área da cúspide onde estas estão inseridas, nem com o número de músculos papilares dos quais elas provem.

E-mail: ceambrosio@usp.br Atrioventricular valve complex of 30 Jafarabadi water buffaloes, adult males were studied in this research with no heart diseases. The animals were obtained from a slaughterhouse in Brazilian State of Parana. The hearts were opened at the third portion affording access to the valve complex. The complexes had its area, number and type of tendinous cords submitted to analysis. The results showed that the complex is composed by two cusps and four accessory cusps, two or three papillary muscles in which 10-25 tendinous cords fix on the cusps that face the ventricle wall. The total area of the complex was on average 38.56cm2, with a minimum of 24.96cm2 and a maximum of 55.54cm2. Statistically, no relation between the number of cords and the cusps’ area where they are inserted or with the number of papillary muscle where they originated from was observed.

Pesquisa Veterinária Brasileira 29(9):725-730, Universidade Estadual de Maringá, Av. Colombo 5790, Maringá, PR 87020-900, Brazil. E-mail: aemurakami@uem.br An experiment was carried out to evaluate the effect increasing levels of yeast wall and broiler breeders’ age levels on lymphoid organs weight, cellular immune response and hematological parameters in broilers. A total of 3,360 Cobb broilers were allotted, in a completely randomized design and a 2x5 factorial arrangement, and two controls, compound of two broiler breeders age (34 and 57 weeks of age) and five yeast wall levels (zero, one, two, three and four kg of yeast wall/ton of diet). Broiler breeders’ age affected all studied variables. The inclusion of 3kg of yeast wall/ton of meal increased, at 57 weeks age broiler breeders, more intense inflammatory reaction when compared to control diet; however the circulated heterophils and lymphocytes numbers were not increased. In conclusion, the use of yeast wall, associated or not to sorghum on broilers diet still needs complementary studies, for example, purified components of yeast wall (MOS and ß-glucano).

E-mail: aemurakami@uem.br Um experimento foi conduzido para avaliar o efeito de níveis crescentes de parede de levedura e idade das matrizes reprodutoras sobre o peso dos órgãos linfóides, a resposta imune celular e o perfil hematológico de frangos de corte. Foram utilizados 3.360 pintos de corte da linhagem Cobb, distribuídos em delineamento inteiramente casualisado, em esquema fatorial 2x5, mais dois controles, sendo duas idades de matrizes (34 e 57 semanas de idade) e cinco níveis de suplementação de parede de levedura (zero, um, dois, três e quatro kg de parede de levedura/tonelada de ração). A idade das matrizes influenciou a resposta de todas as variáveis. A inclusão de 3kg de parede de levedura/tonelada de ração promoveu, na progênie de reprodutoras de 57 semanas, reação inflamatória mais intensa quando comparada a dieta controle, no entanto não houve aumento significativo no número de heterófilos e linfócitos circulantes. Conclui-se que a utilização da parede de levedura associada ao sorgo ou não em rações de frangos de corte ainda necessita de estudos complementares, que incluam, por exemplo, os componentes purificados da parede de levedura (MOS e ß-glucano).

Considering a sampling universe of 2,491 cows, blood samples were collected from 563 dairy cows in 57 farms, which were randomically selected in proportion to the number of animals, using a random stratified sampling system. For each property the number of selected cows was proportional to the herd size. Abortion or other reproductive disorders were not considered as criteria for selecting the animals, and seropositivity was determined by indirect enzyme-linked immunosorbent assay (ELISA). A high association (p=0.006) between seropositivity and racial patterns was found. The analysis revealed that in comparison of pure black-and-white Holstein cows versus Zebu (p=0.0028), Holstein cows showed 2.65 times greater odds for seropositivity. In the same way, by comparing black-and-white Holstein versus Zebu + crossbreed Zebu/Holstein (p=0.01), it was noted that there is 2.23 times more chance for seropositivity in Holstein cattle. There were no significant differences concerning the comparison of Holstein cattle versus crossbreed Zebu/Holstein (p=0.08) or Zebu versus crossbreed Zebu/Holstein (p=0.11). This study supports the hypothesis that there is a close association between cattle breeds and the frequency of infection by N. caninum.

The three breeds of sheep showed similar cellular response in the small intestine mucosa (P>0.05). There was extensive variation among sheep in the parasitological and inflammatory cell variables, even in lambs of the same breed. In general, animals presenting less inflammatory cells had a larger worm burden, higher fecal egg counts, and larger T. colubriformis worms. The inflammatory cells possibly impaired the parasite’s establishment, development, and survival.

The objective of the present work was to establish the prevalence and titer of anti-CAV antibodies in some Brazilian broiler hen breeder flocks and verify in which phase of life the birds were infected. A total of 1,709 serum samples from 12 broiler hen flocks vaccinated against CAV and 64 unvaccinated flocks were analyzed for CAV antibodies with an enzyme-linked immunosorbent assay (ELISA). All non-vaccinated breeder flocks were found to be infected with CAV, with 89% of the hens tested presenting antibodies, 52% of these with titers considered high enough to protect their progeny against CAV infection. Likewise, all vaccinated hens had antibody titer to CAV capable of conferring protection to their progeny. Thus, vaccination of hens seems capable of conferring protection to chicks against clinically apparent CAV-associated disease.

The aim of the present work was to evaluate the association among the kinetics of humoral immune response, the recovery ofviable leptospiras and the intensity ofthe tissue lesions in mice selected for high (H) and low (L) production of antibodies (selection N-A) and in mice from the outbreed Balb/c Iine, inoculated with pathogenic Leptospira interrogans serovar icterohaemorrhagiae. Toe H and L lines showed modifications in some steps ofthe immune response, mainly in relation to the macrophagic activity, representing extreme phenotypes found in heterogeneous natural populations. Mice were sacrificed in eight moments after the infection. Analysis of the results revealed that from the 7th day after the infection, on Iine H mice presented antibodies titles significantly higher as compared to the L line mice, maintaining the multispecific effect Balb/c tine mice showed intermediàte results between the two Iines. Antibodies production worked as a timiting factor to the infection, becausewhen a greater leptospiras recovery was obtained, at the initial phase of the infection, the titles of antibodies were elevated. Inflammatory and degenerative process led to similar lesions in the organs ofinfected mice. Only a variation in the degree oftissue compromising was observed according to the tine. H tine mice showed more extensive lesions than L and Balb/c lines mice. For the Balb/c tine mice, the lesions were moderate. As a mie, H and Balb/c mice lines showed a Th2 profile of immune response, with higher indexes of antibody production and gravity ofthe lesions, while L line mice presented a Th 1 profile ofimmune response.

O presente trabalho teve por finalidade associar a cinética da resposta humoral à recuperação de leptospiras viáveis e à intensidade das lesões teciduais em camundongos geneticamente selecionados para bons (High) e maus (Low) produtores de anticorpos (seleção IV-A), além de camundongos outbreed, Balb/c, inoculados com amostra patogênica de Leptospira interrogans sorovar icterohaemorrhagiae. As linhagens High e Low (seleção IV-A) apresentam modificações em alguns compartimentos da resposta imune, principalmente em relação à atividade macrofágica, representando fenótipos extre mos encontrados em populações naturais heterogêneas. Os camundongos foram sacrificados em oito momentos após a infecção. A análise dos resultados revelou que a partir do 7º dia após a infecção, os camundongos da linhagem High apresentaram elevação nos títulos de anticorpos estatisticamente significantes quando comparados aos camundongos da linhagem Low, mantendo assim o efeito multiespecífico. Os camundongos Balb/c apresentaram resultados intermediários entre as duas linhagens. A produção de anticorpos colaborou como fator limitante à infecção, pois quando obtevese maior recuperação de leptospiras, na fase inicial da infecção, os títulos de anticorpos encontravam-se em elevação. As lesões observadas nos órgãos de camundongos infectados consistiram basicamente nos mesmos processos inflamatórios e degenerativos, que não se alteraram, variando apenas o grau de comprometimento tecidual, de acordo com a linhagem. A linhagem high apresentou lesões mais extensas que as apresentadas pelas linhagens low e Balb/c, sendo que nesta última as lesões foram moderadas. De forma geral a linhagem High e Balb/c apresentaram um perfil de resposta Th2, com o maior índice de produção de anticorpos e gravidade das lesões, enquanto a linhagem Low apresentou um perfil de resposta Th 1.


#37 - Occurrence and distribution of Clostridium botulinum type C and D spores in buffalo breeding areas of the Baixada Maranhense, Maranhão, Brazil, 18(3/4):127-131

[Occurrence and distribution of Clostridium botulinum type C and D spores in buffalo breeding areas of the Baixada Maranhense, Maranhão, Brazil.] Ocorrência e distribuição de esporos de Clostridium botulinum tipos C e D em áreas de criação de búfalos na Baixada Maranhense. Pesquisa Veterinária Brasileira 18(3/4):127-131. Depto Apoio, Produção e Saúde Animal, Unesp - Campus de Araçatuba, Cx. Postal 533, Araçatuba, SP 16015-050, Brazil. As botulism is a common disease in butfaloes raised in the low lands of the State of Maranhão, Brazil, the occurrence of Clostridium botulinum spores was evaluated in butfalo breeding areas of 4 municipalities in the "Baixada Maranhense". Twenty eight samples of faeces, mud and soil were collected and divided into 140 subsamples, being 40 of faeces, 65 of mud and 35 of soil. Botulinum toxin was detected in the filtrates of 104 cultures (74.28%) from 140 subsantples through the inoculation of mice. Using the microcomplement fixation technique for the identification of C. botulinum toxins, type C (14.29%), D (82.14%) and CD complex (3.57%) were found. No significant ditferences (P>0.05%) between faeces, mud and soil samples were observed. There was a high contamination with C. botulinum spores of the butfalo faeces, mud and soil in the areas studied. Identification of other types and subtypes of C. botulinum was not attempted.

Botulismo é enzoótico na criação de búfalos da Baixada Maranhense, Estado do Maranhão. No presente trabalho foram realizados estudos para verificar a ocorrência e distribuição de esporos de Clostridium botulinum tipos C e D em amostras de solo, limo e fezes de búfalos, colhidas aleatoriamente em áreas inundáveis da criação de búfalos nessa Baixada. A evidenciação de esporos foi realizada em 40 amostras de fezes, 65 de limo e 35 de solo, provenientes de quatro municípios, pelo cultivo em meio de cultura com carne cozida e posterior inoculação do sobrenadante filtrado em camundongo, na tentativa de verificação da presença de toxina botulínica. A tipificação de amostras positivas foi realizada pela microfixação de complemento. Os resultados revelaram que 104 (74,28%) das 140 amostras examinadas foram positivas para a presença de esporos de C. botulinum pelo teste indireto. Não houve diferença significativa (P>0,05) entre os valores obtidos quando das análises das amostras de solo (77,1%), limo (60,0%) e fezes (95,0%). Das 28 amostras de solo, limo é fezes positivas, que foram utilizadas para a tipificação, quatro (14,29%) foram classificadas como tipo C, 23 (82, 14%) como tipo D e uma (3,5%) como pertencente ao complexo CD. Os resultados revelaram uma alta contaminação ambiental por C. botulinum em áreas de criação de búfalos da Baixada Maranhense. A identificação de outros tipos e de subtipos de C. botulinum não foi realizada.


#38 - Drug resistance in Salmonella isolated from chicken and turkey breeder flocks

Abstract in English:

Studies were conducted to determine drug resistance in Salmonella isolated using the plate dilution method. The following drugs were used: ampicillin, sulphodiazine, oxytetracycline, kanamycin, chloramphenicol, gentamicin, polymixin B, nalidixic acid, dehydrostreptomycin and nitrofurazone. Twentyfour Salmonella strains isolated from apparently healthy chicken and turkey breeder flocks were examined. The following strains were tested: Eleven S. typhimurium, two S. saintpaul, two S. eimsbuettel and nine S. arizonae 18:z4,z32:- (Ar. -7:1, 7, 7:-). All these strains presented a resistance level below 2 μg of the base per ml of the culture medium when following drugs were used: gentamicin, ampicillin, kanamycin, polymycin. A resistance level of 2 or 5 μg/ml was found with the following: streptomycin, chloramphenicol, nalidixic acid and oxytetracycline. The strain isolated from turkeys showed a resistance level of 5 μg/ml with nitrofurazone while those isolated from chickens showed a resistance of 10 μg/ml. The only exception was S. typhimurium strain 185 which showed resistance to 20 μg/ml of the same drug. All these strains tested were resistant to sulphadiazine.

Utilizando-se o método de diluição em placa, foram determinados os níveis de resistência de amostras de Salmonella isoladas de galinhas e perus às seguintes drogas: ampicilina, sulfadiazina, oxitetraciclina, sulfato de canamicina, cloranfenicol, sulfato de gentamicina, poliximina B, ácido nalidíxico, sulfato de diidroestreptomicina e nitrofurazona. Foram analisadas 24 amostras de samonelas isoladas de espécimens colhidas de um lote de perus e de outro de galinhas reprodutoras aparentemente normais. Foram usadas as seguintes amostras: onze S. thyphimurium, duas S. saintpaul, duas S. eimsbuettel e nove S. arizonae 18: z4,z32:- (Ar. 7-1,7,8:-). Todas as amostras apresentaram níveis de resistência inferiores a 2 μg/ml frente às seguintes drogas: gentamicina, ampicilina, canamicina, polimixina; e níveis de 2 ou 5 μg/ml, frente às seguintes: estreptomicina, cloranfenicol, ácido nalidíxico, e oxitetraciclina. As amostras isoladas de perus apresentaram níveis de 5 μg/ml à nitrofurazona, enquanto que naquelas oriundas de galinhas, os níveis alcançaram até 10 μg/ml. A única exceção foi apresentada por S. typhimurium 185 que se mostrou resistente a 20 μg/ml da mesma droga. Todas as amostras estudadas foram resistentes à sulfadiazina.


#39 - Analysis of the mean burdens, of adult Dictyocaulus viviparus and Trichuris discolor in male calves, Holstein-Friesian and Holstein-Friesian x Guzera

Abstract in English:

An analysis of the mean burdens of adult Dictyocalus viviparus and Trichuris discolor with relation to genetic breeding types Holstein-Friesian (HVB) x Guzera, usrng six:ty male calves, demonstrated significantly lower mean burdens of D. viviparus in the pure-bred HVB, who also had the highest mean burdens of T. discolor. No significant difference was seen between the types 5/8 and 7/8. The F1 generation (1/2 HVB x 1/2 Guzera) is apparently susceptible to both species of helminths, the B1 (3/4 HVB) less so.

Analisando a carga média de Dictyocaulus viviparus e Trichuris discolor em sessenta bezerros machos traçadores de diferentes graus de sangue Holandês vermelho-branco x Guzerá, verificou-se que HVB apresentou cargas médias baixas para D. viviparus e altas para T. discolor. Não se encontrou diferenças significativas entre os tipos 5/8 e 7 /8. F1 (1/2 HVB x 1/2 Guzerá) e B1 (3/4 HVB) demonstraram maior e menor susceptibilidade às duas espécies de helmintos respectivamente.
