E-mail: luizuenf@yahoo.com.br Rabies is a neurological disease, but the rabies virus spread to several organs outside the central nervous system (CNS). The rabies virus antigen or RNA has been identified from the salivary glands, the lungs, the kidneys, the heart and the liver. This work aimed to identify the presence of the rabies virus in non-neuronal organs from naturally-infected vampire bats and to study the rabies virus in the salivary glands of healthy vampire bats. Out of the five bats that were positive for rabies in the CNS, by fluorescent antibody test (FAT), viral isolation in N2A cells and reverse transcription – polymerase chain reaction (RT-PCR), 100% (5/5) were positive for rabies in samples of the tongue and the heart, 80% (4/5) in the kidneys, 40% (2/5) in samples of the salivary glands and the lungs, and 20% (1/5) in the liver by RT-PCR test. All the nine bats that were negative for rabies in the CNS, by FAT, viral isolation and RT-PCR were negative for rabies in the salivary glands by RT-PCR test. Possible consequences for rabies epidemiology and pathogenesis are discussed in this work.

Antígeno ou RNA do vírus da raiva já foram detectados em vários órgãos, tais como glândula salivar, pulmão, rim, coração e fígado. O presente trabalho teve como objetivo identificar a presença do vírus da raiva em órgãos não neuronais de morcegos hematófagos infectados naturalmente, e pesquisar a presença do vírus na glândula salivar de morcegos hematófagos sadios. Dos cinco morcegos positivos para a raiva no SNC pelas técnicas de imunofluorescência direta e isolamento viral em células N2A, 100% (5/5) foram positivos para a raiva nas amostras de língua e coração, 80% (4/5) no rim, 40% (2/5) nas amostras de glândula salivar e pulmão, e 20% (4/5) no fígado pela técnica de RT-PCR. Todos os nove morcegos negativos no SNC, pela imunofluorescência e isolamento viral, foram negativos na glândula salivar pela RT-PCR. Possíveis consequências para a epidemiologia e patogênese da raiva são discutidas.

E-mail: olney@uenf.br Pythiosis is caused by Pythium insidiosum and the occurrence of disease in horses was described in the North and Northwest State of Rio de Janeiro, Brazil. The disease was described in cattle, sheep, humans, and horses in different states and regions across the country. This paper describes the development of IgY and IgG polyclonal antibodies, in chicken and rabbits, respectively against proteins extracted from kunkers and hyphae of P. insidiosum from affected horses. The proteins were recognized by chicken, rabbit and horse antibodies by immunodiffusion and Western blot against majority bands of 27 and 43 KDa, and titrated by ELISA. The antibodies IgY developed by the first time against Brazilian strains of P. insidiosum may represent a valuable tool in the detection of antigens of the pathogen and contribute to further studies aimed at immunotherapy and knowledge about this disease in endemic areas in Rio de Janeiro and in Brazil.

E-mail: bcs85@hotmail.com The efficacy of a polyvalent bacterin vaccine against Aeromonas hydrophila, Pseudomonas aeroginosa and Enterococcus durans administered by different routes in Nile tilapia was assessed by analyzing hematological and immunological parameters 7 and 21 days after vaccination. Treatments consisted of: non-vaccinated tilapia; tilapia vaccinated by intraperitoneal injection with 2x108 formalin-inactivated bacteria·mL-1; tilapia vaccinated orally with 2x107 formalin-inactivated bacteria·g-1, feed for 5 days; tilapia vaccinated by immersion bath in 2x107 formalin-inactivated bacteria·mL-1, for 20 minutes. Vaccinated fish groups presented higher hematocrit, number of erythrocytes and leukocytes than the non-vaccinated group. Serum agglutination titer of intraperitoneally vaccinated fish was higher on both evaluation periods for the three bacteria strains. Only on day 21 post-vaccination fish from the oral and immersion vaccination groups presented higher serum agglutination titer than the non-vaccinated fish for A. hidrophyla and E. durans. Serum antimicrobial activity in vaccinated fish was higher for P. aeroginosa and E. coli than in non-vaccinated fish on both evaluation periods. The different vaccine administration routes stimulated hematological and immunological responses in Nile tilapia 21 days post-vaccination, but intraperitoneal vaccination presented higher total number of leukocytes, lymphocytes and serum agglutination titer.

Os tratamentos consistiram de: tilápias não vacinadas; tilápias vacinadas via injeção intraperitoneal (i.p.) contendo 2x108 bactérias inativadas·mL-1; tilápias alimentadas com ração contendo vacina na proporção de 2x107 bactérias inativadas·g-1, durante 5 dias; tilápias vacinadas por banho de imersão em 2x107 bactérias inativadas·mL-1, durante 20 minutos. Os peixes vacinados apresentaram maior porcentagem de hematócrito, número de eritrócitos e leucócitos durante o período do experimento, em relação aos não vacinados. O título de aglutinação do soro dos peixes vacinados intraperitonealmente foi superior nos dois períodos de avaliação para todas as bactérias. Apenas no 21º dia os peixes vacinados oralmente e por imersão apresentaram título de aglutinação superior aos peixes não vacinados para A. hidrophyla e E. durans. O soro dos peixes vacinados apresentou maior atividade antimicrobiana para P. aeroginosa e E. coli, do que os peixes não vacinados, nos dois períodos. As diferentes vias de administração da vacina estimularam a resposta hematológica e imunológica da tilápia do Nilo após 21 dias de vacinadas, sendo que o número total de leucócitos, linfócitos, título aglutinante do soro dos vacinados i.p. foi maior do que os demais.

Forty baby parrots of genus Amazona, species aestiva and amazonica that were apprehended from wild animal traffic were used: 18 presented ingluvitis and 22 other alterations, but showing general debilitation. Samples were seeded on Sabouraud dextrose agar with chloramphenicol after be obtained by the introduction of urethral probe through the esophagus. Based on morphology and biochemical reactions (API 20C) Candida was confirmed; it was still searched the production of proteinase and phospholipase, virulence factors for Candida species. Candida spp. were isolated from 57.5% parrots, being 72.2% from birds with ingluvitis and 45.5% from without ones. Twenty-five strains of Candida were isolated, 60% and 40%, respectively from parrots with and without ingluvitis, and were speciated: 28% C. humicola, 24% C. parapsilosis, 20% C. guilliermondii, 20% C. famata, and 8% C. albicans. These results demonstrate that C. albicans is not the most frequent species isolated, and it is the first report that shows C. guilliermondii, C. famata, and C. humicola causing infection in parrots. Many isolates presented filamentation (76%), 100% produced proteinase and 68% phospholipase. The observation of Candida spp. producing virulence factors reinforce the pathogenic role of these yeasts in the cases studied.

E-mail: crisruiz@scamilo.edu.br The coronary arteries of the goats have been used to researches about to pharmacological actions, heart failure after connection of coronary arteries and others. Twenty-seven goat hearts, both sexes, fixed in formalin at 10% has been used in this study that analyzed the coronary arteries pattern in goats and if the same is similar to the human coronary arteries pattern. The left coronary artery is dominant and in relation to the length demonstrated average 1.15cm emitting two branches: interventricular paraconalis and circumflex. The interventricular paraconalis branch with average length 10.5cm issued branches to the both ventricles fairly, ending more frequently in the interventricular subsinuosis sulcus. The circumflex branch with average length 8.5cm issued branches to left ventricle and right atrium fairly. The left marginal branch is not constant. The right coronary artery, with average length 6.1cm issued branches to right ventricle and right atrium fairly, but the interventricular subsinuosis branch with average length 3.1cm is variable and can present one long branch, one short branch or one double branch, with the huge part for the right ventricle.

[Influence of Copaifera langsdorffii oil on the repair of a surgical wound in the presence of foreign body.] Influência do óleo de Copaifera langsdorffii no reparo de ferida cirúrgica em presença de corpo estranho. This oil is used for the treatment of skin wound due to its recognized antiinflammatory and wound healing effects. Despite, its popular use, there are few published data about the therapeutic effect of this medicinal plant. The aim of the study was to evaluate the topic treatment effect of the Copaíba oil on the process of skin repair inflammation induced by a foreign body subcutanously implanted. Sixty BALB/c mice were submitted to a 1cm linear incision and a 12mm circle coverslip was subcutaneously implanted. Four treatments groups were established: control, sterile saline (C); vehicle control, sterile mineral oil, (VC); treatment 1 (T1), mineral oil plus copaiba oil (V/V), and treatment 2 (T2) copaiba oil. The evaluations were performed at pre-determined time points (1, 3, 5, 7 and 14 days). It was possible to find fibroblasts, epithelial cells proliferation, re-epithelization and newly formed blood vessels in all groups, however, all oil treated groups (T1 and T2) did not present re-epithelization at three days post surgical incision. On days 5 and 7, a higher intensity of edema and hyperemia on the groups T1 and T2 was observed, besides that, the T1 and T2 groups presented a serous cellular scab on the wounds that was absent on the C and VC groups. The inflammatory reactions among the groups C and VC showed more mononuclear cells than the T1 and T2 groups that presented a mixed cell patter composed from both mono and polymorphonuclear cells. Although the surgical wounds were re-epithelizaded, in the groups T1 and T2, they were covered by a serous cellular crust and the dermis tissue still presented an intense mononuclear cell inflammatory focus. Fourteen days after of the surgical incision, the gross aspects on groups C and VC were similar and on groups T1 and T2, despite wound to be completely closed and without crusts, the skin those animals was thickened. Furthermore, the dermis on group T2 presented moderate fibrosis, while the other groups presented slightly ones. The results demonstrated that topical treatment with C. langsdorffii oil debilitated the normal process of a wound repair in the presence of a foreign body.

For this anatomic study about coronary arteries, thirty hearts of Landrace pigs, of both sexes, 5 to 6 months old, weighing 80 to 110 kg, were fixed in 10% formalin. The coronary arteries and their branches were dissected until the visible ramifications. There was one left coronary artery, with length of 0.4-1.2cm, giving off 2 (80%) or 3 (20%) branches. The paraconal interventricular branch, with length of 10-16cm, emitted 16-25 branches, 52.3% for the right ventricle and 47.7% for the left ventricle. The circumflex branch, with length of 7-15cm, emitted 4-13 branches, 55.6% for the left ventricle and 44.4% for the left atrium. There was one right coronary artery, with length of 7.5-11.5cm, which emitted 12-21 branches, 57.4% for the right ventricle and 42.6% for the right atrium. The subsinuosus interventricular branch, length of 3-11cm, emitted 9-22 branches, 50.9% for the right ventricle and 49.1% for the left ventricle. The number of branches of the paraconal interventricular branch and the number of branches of the subsinuosus interventricular branch was similar for both ventricles. The results obtained in the present study compared with the literature indicate that the distribution of the coronary branches in swine and in humans is similar.

Fifty samples were collected, using an esophageal tube, during winter (rainy season) and summer (dry season). The predominant colors were an olive green in the rainy season and a nut-brown in the dry season. The smell was aromatic, but was stronger in winter. A slight viscous consistence was found in most of the samples, with a greater proportion in winter. The sedimentation and flotation time was 6.73min (±1.63) in the rainy period and 3.15min (±0.72) in the dry period. In the biochemical tests, the average values found in winter and summer were, respectively: pH 6.76±0.21 and 6,59±0.14; methylene blue reduction, 3.20min (±0.76) and 7.76min (±3.00); chloride, 28.14±4,16mEq/L and 24.97±5.65mEq/L; acidity, 21.90±4.38UC and 13.68±2.97UC. Ruminal microbiotic analyses revealed abundant and moderate density of protozoa in winter and summer, respectively. The motility was very active and there were almost 90% of live protozoa in both seasons. Protozoa numbers were 425.373±217.258/mL in winter and 155.375±83.113/mL in summer. There was a mixed population of bacteria with prevalence of Gram-negative forms in both seasons.

[Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Avaliação clínica e hematológica em bezerros Nelore infectados experimentalmente com isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil. Pesquisa Veterinária Brasileira 23(2):52-60. Clínica de Bovinos, Campus Garanhuns, Univ. Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen,reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

O presente trabalho teve por objetivo estudar comparativamente as alterações clínicas e hematológicas desencadeadas por isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil em bezerros Nelore infectados experimentalmente. Foram utilizados 18 bezerros com idade entre sete e nove meses, isentos de anticorpos contra Babesia sp. e criados livres de carrapatos. Três animais foram previamente inoculados com 2,0x109 eritrócitos parasitados (EP) para cada isolado. Os outros 15 bezerros foram subdivididos em três grupos de cinco animais, que foram subinoculados com 1,0x1010 EP dos respectivos isolados. Foram avaliadas as alterações clínicas e hematológica por meio da determinação da parasitemia, do hemograma, do fibrinogênio plasmático, da contagem de reticulócitos, da análise descritiva da medula óssea e da fragilidade osmótica eritrocitária, no decorrer de 30 dias, perfazendo um total de sete momentos de observação. O acompanhamento da resposta imunológica pelo teste de imunofluorescência indireta foi realizado diariamente até o 10º dia pós-inoculação (DPI) e posteriormente no 15º, 20º, 25º e 30º DPI. Clinicamente, observou-se uma manifestação muito branda da doença. Os achados laboratoriais revelaram baixos níveis de parasitemia; decréscimo nos valores do eritrograma; ausência de reticulócitos; diminuição inicial na contagem total dos leucócitos, neutrófilos e linfócitos com posterior elevação do número destas células; hipercelularidade da série eritrocítica e decréscimo da relação mielóide:eritróide mais acentuada entre o 8º e 12º DPI e um aumento da fragilidade osmótica eritrocitária nos grupos inoculados com os isolados sudeste e nordeste. Nenhum dos três isolados de B. bigemina desencadeou a forma clínica característica da enfermidade, apesar de induzirem uma resposta imune humoral.


#50 - Anthelmintic resistance in goat herds in the State of Ceará, 19(3/4):99-103

ABSTRACT.- Vieira L.S. & Cavalcante A.C.R. 1999. [Anthelmintic resistance in goat herds in the State of Ceará.] Resistência anti-helmíntica em rebanhos caprinos no Estado do Ceará. Pesquisa Veterinária Brasileira 19(3/4):99-103. Embrapa-CNPC, Km 4 da Estrada Sobral-Groaíras, Cx. Postal 0-10, Sobral, CE 62011-970, Brazi. Goats of 45 farms in the State of Ceará, Brazil, were treated with anthelmintics for gastrointestinal nematodes, and their resistance to the anthelmintics was evaluated. On each farm 30 kids were weighed, ear-tagged and divided into three groups of ten. The first group received oxfendazole at 4.75mg/kg, the second levamisole at 7.5mg/kg, and the third group remained untreated as control. All goats were drenched according to their individual body weight. Fecal samples were collected from all animals (treated and control) on the day of treatment and 7 days later, to provide material for egg counts and larval cultures. Among 34 surveyed herds 20.6% showed levamisole resistance, 17.6% showed resistance to benzimidazole, and 35.3% had multiple resistance. At the time of the assessment 52.9% of the farmers were using broad spectrum anthelmintics. Only 26.5% of the surveyed herds had nematode populations susceptible to the anthelmintics assessed. The results of larval cultures showed that larvae surviving the treatment with oxfendazole were mainly Haemonchus sp and, to a lesser extent, Oesophagostomum sp; those surviving levamisole treatment were Haemonchus sp, Oesophagostomum sp and Trichostrongylus sp.

Pesquisa Veterinária Brasileira 19(3/4):99-103. Embrapa-CNPC, Km 4 da Estrada Sobral-Groaíras, Cx. Postal 0-10, Sobral, CE 62011-970, Brazi. Um levantamento em nível de campo sobre resistência anti-helmíntica em nematódeos gastrintestinais de; caprinos foi realizado em 34 rebanhos no Estado do Ceará. Em cada rebanho foram separados 30 cabritos, de ambos os sexos, com idade variando de 1 a 6 meses, os quais foram individualmente pesados, identificados e distribuídos em três tratamentos: 1) Oxfendazole na dose de 4, 75mg/kg; 2) Levamisole na dose de 7,5 mg/kg e 3) Controle (não medicado). Os anti-helmínticos foram administrados de acordo com o peso individual de cada animal e, a dosagem utilizada para cada produto foi a recomendada pelo laboratório fabricante. Foram colhidas fezes dos animais de todos os tratamentos, para OPG e coprocultura, no dia da medicação e 7 dias após. Dos 34 rebanhos avaliados, 7 (20,6%) apresentaram resistência aos imidazóis, 6 (17,6%) aos benzimidazóis e 12 (35,3%) revelaram resistência múltipla. Apenas em 9 rebanhos (26,5%), os nematódeos foram sensíveis aos anti-helmínticos avaliados. Através do questionário aplicado detectou-se que 52,9% dos caprinocultores entrevistados usavam anti-helmínticos de amplo espectro. Os resultados das coproculturas mostraram que os gêneros sobreviventes à medicação com oxfendazole foram principalmente Haemonchus sp, seguido em menor frequência por Oesophagostomum sp, enquanto que ao cloridrato de levamisole sobreviveram Haemonchus sp, Oesophagostomum sp e Trichostrongylus sp.
