The disease is officially considered exotic in Brazil, but recent serological surveys and the isolation of the agent suggest it may occur in our herds. The aim of this study was to evaluate three different formulations of Herrold’s egg yolk agar with mycobactin J (HEYM) and four faecal culture protocols considering their ability for Map growth as well as cost and ease of application. Three formulations of HEYM were inoculated with two suspensions of Map. Spiked faeces and naturally contaminated faecal samples were treated by the four faecal culture protocols. Centrifugation protocol and HEYM recommended by OIE showed the best results on the recovery of Map.

Tests were made during 2 years in Holstein cows of a dairy farm producing type C milk, where milking was performed once a day. Sensitivities of electrical conductivity and chloride concentration tests from mammary quarters, where Corynebacterium sp was isolated (65.3% and 78.3%, respectively), were superior to the found in mammary quarters where S. aureus was identified (55.4% and 68.2%, respectively). The efficacies of the two diagnostic tests were similar. Statistical significance was demonstrated with regression analysis of both tests of healthy mammary quarters and subclinical mastitis quarters infected with Staphylococcus aureus.

[Clinical and pathological aspects of bovine paratuberculosis in Rio Grande do Sul, southern Brazil.] Aspectos clínicos e patológicos da paratuberculose em bovinos no Rio Grande do Sul. Pesquisa Veterinária Brasileira 19(3/4):109-115. Setor de Patologia Veterinária, Depto Patologia Clínica Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Av. The clinical and pathological aspects of bovine paratuberculosis in a dairy herd in the county of Capela de Santana, Rio Grande do Sul, southern Brazil, are described. Clinical signs in eight cows out of 345 cattle included chronic diarrhea refractory to treatment, progressive emaciation and decreased milk production. Necropsy findings included severe and diffuse thickening of the mucosa of the small intestine with exaggerated reticular pattern, reflected in the serosal surface, similar to cerebral gyri. Intestinal loops were filled with whitish watery contents. The lymphatic vessels of the intestinal serosa and of the mesentery were thickened and tortuous. The mesenteric lymph nodes were enlarged and, on cut surface, there were oozing of milky fluid. Plaques of mineralization were evident in the intima of larger arteries, in the endocardium of the heart valves and in the ruminal serosa. There was also edema of the abomasal folds and atrophy of the caudate lobe of the liver. The main histopathological findings were granulomatous enteritis, lymphadenitis and lymphangitis. The inflammatory infiltrate was composed of macrophages, epithelioid cells and Langhans’ giant cells containing large numbers of acid-fast bacilli. Vascular lesions included degeneration, calcification and collagen proliferation of the intima and media of larger arteries. Other microscopk lesions included calcification of the ruminal serosa, diffuse hepatocellular atrophy and multifocal granulomatous hepatitis. Mycobacterium paratuberculosis was cultured in Herrold’s médium enriched with mycobactin from all the eight samples of intestinal scrapings submitted to bacteriological examination.

As principais lesões macroscópicas, observadas nos oito animais eutanasiados e necropsiados, incluíam intestino delgado com acentuado espessamento da parede e superfície mucosa de aspecto reticulado, semelhante às circunvoluções cerebrais, lesão essa perceptível através da serosa. A luz intestinal estava preenchida por conteúdo fluido e de aspecto leitoso. Os vasos linfáticos do mesentério mostravam-se mais evidentes, sendo que alguns tinham aspecto varicoso. Os linfonodos mesentéricos estavam aumentados de volume e, ao corte, fluía grande quantidade de líquido leitoso: Focos de mineralização foram observados na íntima das artérias, nas válvulas cardíacas e na serosa do rúmen. Havia também edema das dobras do abomaso e do mesentério e atrofia do lobo caudado do fígado. As principais lesões microscópicas incluíam enterite, linfadenite e linfangite granulomatosas que se caracterizavam por infiltrado inflamatório composto por macrófagos, células epitelióides e células gigantes de Langhans que continham grande quantidade de bacilos álcool-ácido resistentes. As lesões vasculares consistiam em degeneração e mineralização das túnicas íntima e média das artérias de grande calibre associadas à proliferação de colágeno. Havia calcificação da serosa do rúmen, atrofia hepatocelular difusa e hepatite granulomatosa multifocal. Mycobacterium paratuberculosis foi cultivado em meio de Herrold enriquecido com micobactina a partir de raspados do intestino em todas as oito amostras enviadas para exame bacteriológico.


#64 - Characterization of anaerobic bacteria isolated from periodontal "cara inchada"-Iesions of cattle

Abstract in English:

Anaerobic, non-sporulating, Gram negative bacteria were isolated from periodontal "cara inchada" - lesions and characterized. The capacity of fermentation and hydrolysis of different substrates by 52 isolates was studied by means of the API 20 A - System. Fifteen isolates which formed black pigmented or brown non-fermenting colonies, were classified as Bacteroides asaccharolyticus according to their growth pattem in CDC culture medium and their morphologic staining properties, as well as to their biochemical and physiologic profile. Twenty-three isolates which showed the sarne colonial characteristics, but were non-fermentative, were c1assified as Bacteroides spp. Based on these results, 8 other cultores were indentified as non-pigmented Bacteroides spp. and 6 isolates as Fusobacterium nucleatum. In addition, 68 strains of the 4 groups of bacteria isolated were studied regarding their growth in BGA (Bacteroides gingivalis agar) and the formation of a fluorescent pigment. No correlation between the growth on BGA and the fluorescence of the brown or black-pigmented cultores was found. Furthermore, although the API 20 A - System allowed an extensive characterization of the isolates, it could not be sufficient for definite taxonomic purposes.

Foram caracterizadas amostras de bactérias anaeróbias não esporuladas Gram negativas isoladas de quatro bezerros com peridontite ("cara inchada"). A capacidade de fermentação ou hidrólise de diferentes substatos de 52 amostras foi verificada pelo Sistema API 20 A. De acordo com o crescimento em meio de cultivo CDC, das características morfotintoriais e do perfil bioqufmico e fisiológico, 15 amostras formadoras de colônias pigmentadas de negro ou marrom não fermentativas foram classificadas como Bacteroides asaccharolyticus e 23, com as mesmas características das colônias, no entanto fermentativas, como Bacteroides spp. Com base nos resultados, foram ainda caracterizadas 8 amostras como Bacteroides spp, não pigmentados e 6 como Fusobacterium nucleatum. Adicionalmente, outras . 68 amostras de bactérias pertencentes aos 4 grupos isolados, foram estudadas quanto à capacidade de crescimento em meio de cultura seletivo BGA (Bacteroides gingivalis agar) e de formação de pigmento fluorescente, perfazendo-se assim 120 amostras. Não foram observadas correlações entre o crescimento em BGA e a fluorescência das amostras pigmentadas de marrom ou negro. Além disso, ainda que o Sistema API 20 A tenha permitido uma caracterização extensa das amostras isoladas, este não podia ser eficaz para fins taxonômicos.


#65 - Histologic study and isolation of mycobacteria from tuberculosis-like lesions in cattle in southern Brazil

Abstract in English:

One hundred and fifty-nine lesions macroscopically similar to tuberculosis, observed in cattle slaughtered from 13 counties located in southem Rio Grande do Sul, were studied histologically. Tuberculosis was diagnosed in 147 (92.45%), lesions, club-forming granulomas in 7 (4.4%), eosinophilic abscess in 2 (1.26%), and abscess in 3 (1.89%). Fifty-nine specimens with histological lesions of tuberculosis were cultured for mycobacteria, obtaining 40 isolates, all of them classified as Mycobacterium bovis. Based in these results and the data related with condemnation due to tuberculosis in Rio Grande do Sul, the prevalence of the disease in beef cattle in the State during 1980-1987 was estimated in 0.6%. However, marked differences were observed between different areas. The higher prevalences (2.23% to 3.39%) occurred in the regions focated in the coastland of the "Lagoa dos Patos", middle prevalences (0.97% to 1.95%) in the regions located West and Northwest near Porto Alegre, and the lower prevalences in the other regions of the State (0.12% to 0.71 %). The annual prevalence of the disease has been decreasing sistematically from 1980 (0.95%) to 1987 (0.42).

Cento e cinqüenta e nove lesões observadas em frigoríficos como macroscopicamente similares à tuberculose, de bovinos provenientes de 14 municípios da região sul do Rio Grande do Sul, foram estudadas histologicamente. Tuberculose foi diagnosticada em 147 (92,45%) lesões, actinogranuloma em 7 (4,4%), abscesso eosinofílico em 2 (1,26%) e abscesso em 3 (1,89%). Em 59 desses materiais, com lesões histológicas de tuberculose, foram realizadas culturas para micobactérias, obtendo-se 40 isolamentos, todos eles tipificados como Mycobacterium bovis. Com base nos resultados do estudo histológico e bacteriológico foram analisados os dados de condenação por tuberculose de todos os municípios do Rio Grande do Sul, no período 1980-1987, estimando-se a prevalência da doença, em gado de corte no Estado, em aproximadamente 0,6%. No entanto detectaram-se marcadas variações, entre regiões, na freqüência de lesões macroscopicamente similares à tuberculose, observando-se maiores prevalências (2,23% a 3,39%) nas microrregiões homogêneas localizadas no litoral da Lagoa dos Patos, prevalências médias nas 6 microrregiões próximas a Porto Alegre, no sentido oeste e noroeste (0,97% a 1,95%), e as menores prevalências nas restantes microrregiões do Estado (0,12% a 0,71%).


#66 - Serologic and allergic monitoring of experimental infections in goats by Corynebacterium pseudotuberculosis

Abstract in English:

The six month serologic and allergic monitoring of 26 experimentally infected goats, by intradermic inoculation of 5, 20 and 100 Corynebacterium pseudotuberculosis cells, showed the development of both humoral and cellular imune responses at the sarne time. Only one goat did not develop antibodies, but showed a strong allergic reaction, during 22 weeks post-infection. The antibodies, investigated by the synergistic hemolysis inhibition test (Knight 1978), were first detected at the 3rd week and were most elevated at the 7th week post-infection. In 8 animals the antitoxins were persistent until the 29th week; in 7 animals the antibodies could not be detected between the 12th and the 24th week and in other 9 goats between the 5th and 8th week post-infection. Three animals did not show antibodies in any phase after the infection. The allergic reaction, evaluated by the lymphadenization test (Langenegger et al. 1987) in six-week intervals, showed the highest value between the 4th and the 6th week post-infection and then decreased slowly, following generally the antibody level. One third of the infected goats lost the antitoxin titre and the allergic reaction at the 9th and 10th week, having selfcures. A gradual decrease of the antitoxins titre and the allergic sensitization was observed until the total disappearance in some goats with chronic lymphnode abscesses which did not ulcerate, despite live C. pseudotuberculosis still being in the lesion.

O monitoramento sorológico e alérgico de 26 caprinos infectados experimentalmente por inoculação intradérmica de 5, 20 e 100 bactérias de Corynebacterium pseudotuberculosis, durante 6 meses, revelou que as respostas imunológicas humoral e celular desenvolveram-se paralelamente, exceto em um animal no qual não foram detectadas antitoxinas, porém forte reação alérgica até a 22ª semana pós-infecção. A pesquisa de antitoxinas feita através da técnica da Inibição da Hemálise Sinérgica (Knight 1978), revelou a presença de anticorpos a partir da 3ª e atingiu maior concentração na 7ª semana pós-infecção. Em 8 animais a presença destas persistiu até a 29ª semana; em 7 desapareceu entre a 12ª e 24ª e em 9 entre a 5ª e 8ª semana. Três animais não reagiram com título igual ou superior à diluição 1:20. A linfadenização (Langenegger et al. 1987), feita em intervalos de 6 semanas, revelou reações maiores entre a 4ª e 6ª semana pós infecção. Depois a sensibilidade alérgica decrescia, pouco a pouco, acompanhando grosseiramente o declínio do nível das antitoxinas. Em um terço dos animais houve autocuras a julgar pela regressão total das antitoxinas e perda da sensibilidade alérgica a partir da 9ª e 10ª semana pós-infecção. Em alguns caprinos cujas lesões ganglionares se tornavam crônicas sem ulcerar, observou-se também progressiva regressão, tanto das antitoxinas quanto da reação alérgica, até o seu desaparecimento, apesar da presença de C. pseudotuberculosis na lesão.


#67 - Levels of immunoprotection induced in goats against Corynebacterium pseudotuberculosis using several types of vaccines

Abstract in English:

In order to evaluate the immune response of goats against Corynebacterium pseudotuberculosis, 23 groups, comprising 196 animals, were immunized with several antigenic preparations obtained from the bacteriae, at different time intervals, and using various kinds of adjuvants. The results indicated that the rate of immunoprotection in the immunized animals challenged after 120 days was below 60%. The animals challenged between 8 and 10 months showed protection levels ranging from 44.5 to 22.3%. The lowest rate of incidence of the disease appeared in the anima1s challenged between 30 and 60 days after the last immunization.

Com o objetivo de observar a imunoproteção desenvolvida em caprinos contra Corynebacterium pseudotuberculosis esses animais foram submetidos a diferentes esquemas de imunização, sendo utilizados inóculos de diferentes preparados antigênicos desta bactéria em intervalos de tempo variados e com diversos adjuvantes. Os resultados revelaram que o percentual de imunoproteção nos animais imunizados e desafiados 180 dias após estava abaixo de 60%. Já os animais desafiados dez meses após a primeira inoculação apresentaram níveis de imunoproteção entre 44,5 e 22,3%. O menor índice da doença foi observado nos animais desafiados entre 30 e 60 dias após a última inoculação do preparado antigênico.


#68 - Outbreak of mycobacteriosis in a froggery caused by Mycobacterium marinum

Abstract in English:

An outbreak of mycobacteriosis (tuberculosis) caused by Mycobacterium marinum in a colony of Rana catesbeiana in the State of Rio de Janeiro is described. The disease was introduced in the froggery by infected animals and, in 3 months, about 50% of 160 animals became contaminated and died. The sick frogs had shown clinically progressive anorexia, apathy, adynamia, having the head lowered and lost weight with the evolution of the disease. The necropsy had shown nodular whitish caseous lesions in livers, spleens, kidneys, rarely in lungs, with variable number and size of the nodules. Microscopically, the lesions were characterized as granulomas in different evolutioning stages. These had origin in epithelioid cell agglomerations which became caseous necrosis later. The so initiated nodules were surrounded by some macrophage cells and by concentric connective tissue layers, limiting the nodules themselves and from the normal adjacent organ tissues. A great number of isolated or aggregated acid-fast bacteria was seen scattered in the nodule structure. The identification of M. marinum was based on the cultural and biochemical behaviour, with special regard to the photochromogenicity.

Foi descrito um surto de micobacteriose causado por Mycobacterium marinum numa criação de rãs da espécie Rana catesbeiana no Estado do Rio de Janeiro. A doença foi introduzida no ranário através da compra de rãs infectadas e propagou-se em 3 meses a cerca de 50% das rãs de um lote de 160 animais que adoeceram e morreram. Clinicamente as rãs doentes demonstravam progressiva anorexia, apatia, adinamia e postura cabisbaixa, perdendo peso a medida que a doença evoluia. A necrópsia revelou lesões nodulares caseosas de cor esbranquiçada no fígado, baço e rins, raramente nos pulmões, variando o número e tamanho dos nódulos. Microscopicamente as lesões eram constituídas por granulomas, em vários estágios de evolução. Estes iniciavam-se por aglomerados de células epitelióides que posteriormente eram levados a necrose por caseificação. Os nódulos assim formados eram envoltos por alguns macrófagos e por camadas concêntricas de tecido conjuntivo que delimitavam os nódulos entre si e do tecido normal adjacente do órgão. Grande número de bastonetes isolados ou formando agregados bacterianos, álcool-ácido resistentes, encontravam-se entre as estruturas do nódulo. A identificação de M. marinum baseou-se no comportamento cultural e bioquímico, destacando-se a fotocromogenicidade.


#69 - Reproduction of caseous lymphadenitis in goats by small numbers of Corynebacterium pseudotuberculosis

Abstract in English:

Caseous lymphadenitis was reprodued experimentally by intradennic inoculation of 5, 20 and 100 washed Corynebacterium pseudotuberculosis bacteria in 26 normal goats. Between day 2 and 9 after inoculation, there was local erythema, papula and skin ulceration and, at day 15, a small enlargement of the local lymph node. Between day 45 and 180, independent of the infective dose rate, the infection developed to an abscess in the local lymph node of about one third of the goats. A similar number showed a pronounced enlargement of the local lymph node, but these later became normal again. The remainder of the goats showed an early return to normal after the initial signs of infection. All 26 goats, except 2, presented an antitoxin titre during some phase of the infection. The experimental·infection was shown to develop as the natural. It was concluded that selfcures, demonstrated experimentally, must also occur in the field, making clinical and immunological diagnosis difficult.

A linfadenite caseosa foi reproduzida experimentalmente através da inoculação intradérmica de 5, 20 e 100 bactérias lavadas de C. pseudotuberculosis em 26 caprinos normais. A infecção manifestou-se, inicialmente, por formação de eritema, pápula, pústula e ulceração local da pele entre o 2º e o 9º dia pós-inoculação e no 15º dia por um ligeiro aumento do linfonodo satélite. Posteriormente, entre o 45º e o 180º. dia, independentemente da dose infectante usada, a infecção evoluiu para abscedação do linfonodo local em cerca de 1/3 dos caprinos. Em outro número similar, observou-se acentuado aumento do volume do linfonodo mas que depois regrediu e, finalmente, nos restantes houve rápida normalização após os sinais iniciais da infecção. Dos 26 caprinos apenas 2 não apresentaram antitoxinas no soro durante nenhuma fase da infecção. Foi demonstrado com o presente experimento que a infecção evoluiu simulando a natural e concluiu-se que as curas espontâneas observadas também devem ocorrer nas infecções de campo, dificultando o diagnóstico clínico e imunológico.


#70 - Evaluation of an inactivated vaccine against caseous lymphadenitis of goats kept under extensive management

Abstract in English:

The experiment was carried out during a 2-year period (Oct. 1981 to Oct. 1983), using 163 goats of both sexes with 6 to 12 months of age, The objective was to evaluate the immunogenicity of a vaccine against caseous lymphadenitis, in northeastem Bahia. The vaccine was developed, utilizing the local strain 1002 Curaçá of Corynebacterium pseudotuberculosis isolated from na abscess of a goat. For the preparation, tryptose broth, enriched with yeast extract and lactalbumin, was used. Inactivation was effected with 0.5% formaldehyde, and aluminum gel phosphate was used as an adjuvant. The cell volume varied from 0.65 to 0.7%. The experiment consisted of the following treatments: A1 - animals vaccinated every 6 months; A2-animals vaccinated once a year; and A3 - animals not vaccinated. The goats were vaccinated by subcutaneous inoculation of 3 ml vaccine into each axillar region, and post-mortem examination was made at the end of the experiment. The results showed that 72.1% of the control animals (A3) had moderate to severe lesions in different lymphnodes, and in some cases in the internai organs. Of the vaccinated animals, only 25.9% and 18.9%, at 6 and 12 months interval respectively, showed discrete lesions which were restricted to no more than three lymphnodes per animal. There was a signif cant difference (P<0,05) between the vaccinated and the non-vaccinated animals, but no difference between treatment A1 and A2. It can be estimated that the vaccination gave a protection of 77% against the disease.

Foram utilizados 163 caprinos de ambos os sexos, de 6 a 12 meses de idade, com o objetivo de avaliar a imunogenicidade de uma vacina contra a linfadenite caseosa, no Nordeste do Estado da Bahia. A vacina foi elaborada com a cepa 1002 Curaçá de C. pseudotuberculosis, isolada de abscesso de caprino, cultivada em caldo tryptose, enriquecida com extrato de levedura mais lactalbumina e inativada com formaldeído a 0,5% durante 7 dias, utilizando-se como adjuvante o gel fosfato de alumínio. O volume celular variou de 0,65 a 0,70%. O experimento constituiu-se dos seguintes tratamentos: A1 - animais vacinados semestralmente; A2 - vacinados anualmente; A3 - animais não vacinados. Os animais vacinados foram inoculados via subcutânea com 3 ml da vacina em cada região axilar e, ao final do experimento, foram necropsiados. Os resultados mostraram que 72,1 % dos caprinos do grupo testemunha A3 se infectaram naturalmente e apresentaram lesão moderada a severa em vários linfonodos e, em alguns casos, nos órgãos internos. Dos vaciandos, apenas 25,9 e 18,9%, respectivamente, para os revacinados com 6 e 12 meses de intervalo, apresentaram lesões discretas que se restringiram a não mais de três linfonodos por animal. Houve diferença significativa (P<0,05) entre os animais vacinados e não vacinados mas não houve diferença entre os tratamentos A1 e A2. Pode-se estimar que a vacina proporcionou 77% de proteção contra a doença.
