#1 - Fusobacterium necrophorum predominates in the microbiota of mandibular dental abscess in Blastocerus dichotomus

Abstract in English:

Dental abscess in ruminants is an acute polymicrobial infection, usually resulting from periodontal disease or endodontic infection, with consequences for animal health and welfare. The present study aimed to describe the bacterial microbiota of dental abscesses in Blastocerus dichotomus. Biological material from mandibular dental abscesses, punctured with a sterile syringe and needle during routine veterinary curative procedures or necropsies, was collected from three ex-situ marsh deer. Bacteria were identified using high-throughput sequencing of the 16S ribosomal RNA gene. The three specimens had the presence of facial bulging, and two died because of severe emaciation with a history of progressive weight loss. Bacteroides (38.6%), Fusobacterium (36.65%), and Porphyromonas (7.49%) represented the most abundant genera and Fusobacterium necrophorum (35.69%), Porphyromonas levii (3.12%) and Porphyromonas gulae (1.78%) were among the ten most represented species in the microbiota of mandibular abscess in Blastocerus dichotomus. These molecular findings demonstrate a broader diversity of species in the polymicrobial nature of dental abscesses in B. dichotomus than was previously reported when culture-dependent methods were used in the diagnosis.

#2 - Casuistry of xenarthrans treated at the Veterinary Hospital of the Federal University of Pará, Brazilian Amazon

Abstract in English:

The present study aimed to analyze the casuistry of xenarthrans treated at the veterinary hospital of the Federal University of Pará to quantify the most important cases for clinical care and identify the main species, sex, age group, origin, and destination of the animals admitted. The data were collected from January 2013 to August 2020. The variables analyzed were the number of animals by species, sex, age group, weight (kilograms), form of admission (rescue or guardianship), person responsible for the admission of the animal, origin, circumstances of admission, length of stay (days), and evolution or destination of the animal (release, captivity, death, euthanasia, or escape). Fisher’s exact test was used to determine whether disease incidence was related to sex and whether animal mortality was influenced by sex and disease. The chi-square test was used to determine whether mortality differed by age group. Ninety-two (92) xenarthrans were included: 66 Bradypus variegatus, 10 Choloepus didactylus, 15 Tamandua tetradactyla, and one Dasypus novemcinctus. The animals were mainly from the municipality of Castanhal (70%) and were acquired through rescue (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla), a significant proportion of which was per natural persons (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla). The most important circumstances that led to the admission of the animals were as follows: animals for examination (42%, n=39), young animals at risk (9%, n=8), hit by a vehicle (8%, n=7), respiratory disease (7%, n=6), fall from a tree (5%, n=5), and electrotrauma (5%, n=5). Release was the most common destination of animals after hospitalization (59% B. variegatus, 50% C. didactylus, and 66% T. tetradactyla). The sex of the animals did not affect the incidence of disease or mortality in any species. In contrast, mortality of B. variegatus was significantly higher in animals admitted with disease (p=0.000). Mortality in cubs of this species was higher than that in juveniles and adults (p=0.003).

#3 - Periodontal diseases and tooth wear in sheep flocks in the State of Goiás, Brazil

Abstract in English:

Dental and periodontal affections consist of health concerns of great importance in sheep flocks, as they affect the health and welfare of animals and represent one of the main causes of premature slaughter. Studies on the occurrence of these conditions in sheep are scarce in Brazil. The present study aimed to evaluate the prevalence of periodontal diseases and tooth wear in sheep flocks in the State of Goiás. For this, a clinical oral examination was conducted on 325 sheep from seven farms in six municipalities of the State. Periodontal lesions, characterized by gingival recession in at least one incisor tooth, were observed in 48.3% (157/325) of the sheep, while in cheek teeth it occurred in 13.2% (43/325) of the evaluated sheep. Dental wear on cheek teeth was identified in 69.2% (255/325) of the sheep, and 10.7% (35/325) of the ovines showed dental wear on incisor teeth. Sheep older than 36 months had an increase in the frequency of tooth wear. A total of 171 (52.62%) out of the 325 examined sheep had signs of bleeding on probing, which is compatible with gingivitis, and 69 (40.36%) were older than 36 months. Thirty-eight (11.69%) out of the 325 evaluated ovines had gingivitis in all teeth, among which 21 (55.26%) were older than 36 months. Logistic regression analysis allowed the observation that age was associated with the occurrence of gingivitis. Less frequent changes such as periodontal pockets and loss of at least one tooth were also observed in 4.9% (16/325) and 17.8% (58/325) of the sheep, respectively. It indicates that the presence of periodontal diseases and tooth wear is a common occurrence in the evaluated flocks. Therefore, there is a need to include the oral health of sheep in the routine of the farm as an essential preventive measure to improve the quality of life, animal welfare, and zootechnical indices.

#4 - Type C botulism outbreak in free-ranging waterfowl in Goiás

Abstract in English:

Botulism is generally a fatal disease caused by ingestion of neurotoxins produced by Clostridium botulinum. The present study describes the epidemiological, clinical, and laboratory aspects of a type C botulism outbreak in free-living aquatic birds residing in an urban park in Quirinópolis, Goiás, Brazil. Among a population of approximately 80 waterfowl, a total of 30 birds, including ducks (Cairina moschata), teals (Anas platyrhynchos), and geese (Anser cygnoides), died within 10 days. Of these, six birds showed signs of flaccid paralysis of the pelvic limbs, eyelids, neck, and wings. To confirm the suspicion of botulism, four lake water samples, two samples of the feed consumed by the birds, and samples of serum, intestinal content, stomach content, and liver tissue from two teals that died after presenting clinical signs were analyzed. Using bioassay and neutralization with homologous antitoxin in mice, it was possible to detect the presence of botulinum toxin type C in a water sample and in the intestinal content of one of the necropsied teals. Additionally, the presence of C. botulinum type C was identified in the lake water using polymerase chain reaction. Based on the clinical signs and laboratory results, a diagnosis of botulism caused by botulinum toxin type C was confirmed with probable transmission by lake water.

#5 - Spontaneous poisoning by Dodonaea viscosa (Sapindaceae) in cattle in Southern Brazil

Abstract in English:

In this study, an outbreak of spontaneous poisoning by Dodonaea viscosa (D. viscosa) in a herd of dairy cattle in the municipality of Capão do Leão, Rio Grande do Sul, was investigated. Three deaths occurred in a batch of 16 Jersey cattle, aged between three and four years, kept in a native field. The clinical signs observed were apathy, decreased production, and anorexia, with death occurring within approximately 48 h after the onset of signs. The three cattle were necropsied, and tissue samples were sent for histopathological examination. Necropsy findings included serosanguineous fluid in the abdominal cavity, intestines with congested serosa, and marked mesenteric edema. The mucosa of the abomasum of two of the animals was hemorrhagic with bloody content, and among the ruminal content of a bovine, leaves with morphological characteristics compatible with D. viscosa were observed. The livers of the three animals were enlarged, with accentuation of the lobular pattern. Histologically, centrilobular coagulation necrosis with congestion and hemorrhage was observed in the liver. Vacuolization and degeneration of hepatocytes were observed in the mid-zonal and periportal regions. The diagnosis of poisoning by D. viscosa leaves was based on epidemiological data, necropsy findings, and histopathological alterations. The presence of the plant in the rumen and in the grazing site of the affected cattle was essential for the diagnosis.

#6 - Granulomatous pneumonia due to pythiosis in captive South American coatis (Nasua nasua)

Abstract in English:

Pythiosis is an emerging infectious disease affecting captive and free-ranging wild animals. We report granulomatous pneumonia due to Pythium insidiosum in two South American coatis (Nasua nasua), who were found dead without any clinical records. Severe granulomatous pneumonia associated with pleural effusion was revealed in the necropsy. Microscopically, variably sized granulomas and pyogranulomas presented negative hyphae profiles at the periphery of their necrotic cores. Grocott methenamine silver stain highlighted these structures, and immunostain (anti- P. insidiosum) was strongly positive. Molecular analysis by polymerase chain reaction amplified P. insidiosum specific DNA. These findings characterized P. insidiosum as a cause of granulomatous pneumonia in coatis and proved that pythiosis needs to be considered in the differential diagnosis of respiratory diseases affecting this species in endemic areas.

#7 - Meta-analysis of brucellosis vaccinology in natural hosts

Abstract in English:

Brucellosis is a relevant zoonotic disease for which the most important tool for control is vaccination of susceptible animals. Assessment of vaccine efficacy in natural hosts is based on prevention of abortion and Brucella infection in organs of immunized animals. A meta-analysis of experimental vaccination of Brucella spp. natural hosts was performed, including 45 PubMed and/or Scopus-indexed publications, representing 116 individual experiments. Difference of risk was calculated as an indicator of protection, and a temporal analysis (1980-2016) demonstrated that experimental vaccines tested on natural hosts provided levels of protection that were stable over the past decades. The meta-regression model developed in this study included different vaccine categories (attenuated, inactivated, mutant, subunit, and vectored) considering the difference of risk as the dependent variable. The subcutaneous route of vaccination provided better protection when compared to the intramuscular and oral routes of vaccination. Surprisingly, inactivated vaccines provided better protection than live naturally attenuated vaccine strains (spontaneous mutations) that were considered the reference, whereas subunit vaccines provided lower levels of protection. This is the first meta-analysis of Brucella vaccinology in the natural hosts. These results are useful for the development of new vaccination protocols for controlling animal brucellosis.

#8 - Pathogenic potential of Brucella ovis field isolates with different genotypic profile and protection provided by the vaccine strain B. ovis ΔabcBA against B. ovis field isolates in mice

Abstract in English:

Brucella ovis causes economic and reproductive losses in sheep herds. The goal of this study was to characterize infection with B. ovis field isolates in a murine model, and to evaluate protection induced by the candidate vaccine strain B. ovis ΔabcBA in mice challenged with these field isolates. B. ovis field strains were able to colonize and cause lesions in the liver and spleen of infected mice. After an initial screening, two strains were selected for further characterization (B. ovis 94 AV and B. ovis 266 L). Both strains had in vitro growth kinetics that was similar to that of the reference strain B. ovis ATCC 25840. Vaccination with B. ovis ΔabcBA encapsulated with 1% alginate was protective against the challenge with field strains, with the following protection indexes: 0.751, 1.736, and 2.746, for mice challenged with B. ovis ATCC25840, B. ovis 94 AV, and B. ovis 266 L, respectively. In conclusion, these results demonstrated that B. ovis field strains were capable of infecting and inducing lesions in experimentally infected mice. The attenuated vaccine strain B. ovis ΔabcBA induced protection in mice challenged with different B. ovis field isolates, resulting in higher protection indexes against more pathogenic strains.

#9 - Ultrasound monitoring of the uterus and ovaries of dominant and subordinate females of yellow-breasted capuchin (Sapajus xanthosternos) and robust tufted capuchin (Sapajus robustus) in captive colonies during the ovarian cycle and anestrus periods

Abstract in English:

The yellow-breasted capuchin (Sapajus xanthosternos) and robust tufted capuchin (Sapajus robustus) are endangered species due to destruction of their natural habitat and predatory chase. However, it is still necessary to elucidate some details of their reproductive physiology in order to obtain better indices in the assisted reproduction of these species. This study aimed to evaluate the ovarian cycle of 13 dominant and subordinate females of S. xanthosternos (n=8) and S. robustus (n=5) using sagittal and transversally scanned ultrasound of their uterus and ovaries. Sonograms were performed every seven days for two months. The ovarian cycle phase and anestrous condition were confirmed by colpocytology. Our results showed different uterine parameters (craniocaudal diameter, dorso-ventral diameter, and transverse diameter) (P<0.05) between anestrous subordinate females and other ovarian cycle phases and social classes. The mean of uterine volume was higher in dominant females than subordinate females in all cycle phases (P<0.05), except in follicular phase. During anestrus, endometrial width was smaller in subordinate females than in dominant females (P<0.05). Subordinate females showed differences in endometrial measures (P<0.05) between anestrous period and follicular and luteal periods. Ovarian measures in dominant females were higher than in subordinate females only during anestrus (P<0.05). In the subordinate females, ovarian parameters were different (P<0.05) between anestrus and follicular and luteal phases. Dominant females showed higher volume of right ovary compared to volume of the left ovary during anestrus and follicular phase (P<0.05). Follicles and corpus luteum were distinguished by ultrasonography in most exams (86.11%). During anestrus, measurable ovarian structures were not observed in both ovaries in dominant and subordinate females. In conclusion, the methodology used in this study allowed to evaluate the ovarian cycle in S. xanthosternos e S. robustus females and that cycle phase/anestrus and social class of the female influenced the size of the uterus and ovaries.

#10 - Chemical composition and antimicrobial activity of two extract of propolis against isolates of Staphylococcus spp. and multiresistant bacterials

Abstract in English:

There is a growing need to discover and develop alternative therapies for the treatment of mastitis caused by Staphylococcus spp. and multidrug-resistant bacterial infections. This study examined the chemical composition and antimicrobial potential of two propolis extracts (EPA and EPB) against seventy-seven isolates of Staphylococcus spp. obtained from subclinical bovine mastitis; three clinical strains of MRSA and two from clinical strains of S. aureus ATCC, identified as S. aureus ATCC 6538 and S. aureus ATCC 25923. The total phenolic content was determined by the Folin-Ciocalteau method, the total flavonoid content by the Dowd method and the phenolic profile was quantified by HPLC-DAD. The MBC values of the extracts were evaluated by broth microdilution method. The amount of total phenolic and flavonoid compounds was higher in EPA than EPB. Both extracts revealed the presence of caffeic, coumaric, cinnamic, ferulic and 3,4-dihydroxybenzoic acids, with higher concentrations of coumaric and cinnamic acids. Staphylococcus spp. isolates were susceptible to EPA (90.9%), EPB (83.1%) and oxacillin (80.5%). The oxacillin susceptible isolates were also susceptible to EPA (70.1%) and EPB (80.6%), whereas those oxacillin-resistant strains were also susceptible to EPA (40.0%) and to EPB (26.7%). MBC ranged from 34.3 to 68.7μm/mL for EPA and from 68.7 to 137.5μg/mL for EPB. Both extracts inhibited significantly (100%) the clinical strains of MRSA, S. aureus ATCC 6538 and S. aureus ATCC 25923 at the concentration of 68.7μg/mL. It is concluded that both extracts of propolis, whose main constituents are coumaric and cinnamic acids, have high antimicrobial activity against the microorganisms studied, and EPA also against oxacillin-resistant strains. These findings reinforce its potential use for the treatment of bovine mastitis.
