#1 - Mortality caused by gastrointestinal nematodes in beef cattle submitted to an inadequate sanitary protocol

Abstract in English:

The aim of this study was to describe the epidemiological, clinical, and pathological findings of two outbreaks of gastrointestinal nematode infections in beef cattle kept under an inadequate sanitary protocol. An outbreak of trichostrongylosis occurred in Amambai, Mato Grosso do Sul (MS), from May to July 2016. The herd consisted of 3,000 Nellore cows, and of these, fifteen died after showing emaciation and diarrhea and remaining in recumbency. At necropsy, the abomasum showed edema in the folds, white, raised areas, multifocal to coalescent, and small ulcers. Histopathology showed larvae compatible with trichostrongylid nematodes were present inside the abomasum glands. Trichostrongylus axei was identify in the abomasum. The hemonchosis outbreak began in October 2018, in a breeding and finishing property in Santa Rita do Pardo, MS. Of 4,000 cattle aged 8 to 18 months, 673 became ill and 117 died. Clinical signs were emaciation, weakness, dehydration, submandibular edema, and soft stools. At necropsy, large numbers of nematodes were found in the abomasum; they were morphologically classified as Haemonchus placei. Both outbreaks were caused by failures in the parasite control protocol. However, in Outbreak I, frost and immune stress caused by lack of food may have contributed to the deaths. In Outbreak I, the main failure in the devermination protocol was the use of anthelmintics without carrying out an efficacy test. In Outbreak II, there was no parasite control protocol in the rearing and finishing property, and in the breeding property, there was resistance to the anthelmintic used (ivermectin). The two outbreaks highlight the importance of gastrointestinal nematode control protocols in cattle and demonstrate that infections by T. axei and H. placei can be lethal for this species.

O objetivo deste trabalho foi descrever os achados epidemiológicos, clínicos e patológicos de dois surtos de verminose em bovinos de corte mantidos sob protocolo sanitário inadequado. Um surto de tricostrongilose ocorreu em Amambai, Mato Grosso do Sul (MS), de maio a julho de 2016. O rebanho de cria era composto por 3.000 vacas Nelore, das quais, quinze vacas adultas morreram após apresentarem emagrecimento, diarreia e permanecerem em decúbito. Na necropsia, o abomaso apresentava edema das pregas, áreas brancacentas, elevadas, multifocais a coalescentes e pequenas úlceras. Na histopatologia, larvas compatíveis com nematódeos tricostrongilídeos estavam presentes no interior das glândulas do abomaso. A recuperação de helmintos do abomaso demonstrou que Trichostrongylus axei foi o principal agente etiológico. O surto de hemoncose iniciou-se em outubro de 2018, em uma propriedade de recria e terminação em Santa Rita do Pardo, MS. Do total de 4 mil bovinos com 8 a 18 meses, 673 adoeceram e 117 morreram. Os sinais clínicos foram emagrecimento, fraqueza, desidratação, edema submandibular e fezes pastosas. Na necropsia, havia grande quantidade de nematódeos no abomaso, que foram classificados morfologicamente como Haemonchus placei. Ambos os surtos foram causados por falhas no protocolo de controle parasitário, porém, no Surto I, geadas e estresse imunológico causado pela falta de alimento podem ter contribuído para as mortes. No Surto I, a principal falha no protocolo de desverminação foi o uso de anti-helmínticos sem a realização de teste de eficácia. No Surto II, não havia protocolo de controle parasitário na propriedade de recria e terminação, e na propriedade de cria, havia resistência ao anti-helmíntico utilizado (ivermectina). Os dois surtos ressaltam a importância dos protocolos de controle de nematódeos gastrintestinais em bovinos e demonstram que infecções por T. axei e H. placei podem ser letais para esta espécie.

E-mail: fortesfs@gmail.com The selection and growing spread of resistant nematodes to the most commonly used anthelmintics, benzimidazoles (BZs), imidazoles and macrocyclic lactones (MLs), constitutes a serious obstacle of small ruminants production worldwide. The use of efficient and sensitive methods for detection and monitoring of anthelmintic resistance in the field becomes urgent, especially for the BZs and MLs groups due to its frequent resistant reports. Obtaining an early and accurate diagnosis of resistance is extremely important to aid decision-making regarding parasite control programs, with the objective to preserve the lifespan of existing products, and to limit the development of resistance in nematode populations. The in vivo tests and the more recent in vitro tests have been developed for the detection of nematode resistant to the major anthelmintic groups. However, the availability of validated in vitro tests and its practical use is still very limited. Although the faecal egg-count reduction test (FECRT, in vivo - indirect) is the primary method of choice for the detection of resistance in the field it has being criticized for its results and is receiving significant modifications. Moreover, the development of molecular techniques from genomic changes have generated considerable advances in this research area, with the use of mutations at codons 167, 198 and 200 of β-tubulin gene as the main SNPs (single nucleotide polymorphisms) associated with BZs resistance. This review aims to discuss the available diagnostic methods for the detection of anthelmintic resistance in nematodes of small ruminants, highlighting key developments and obstacles to its use in the laboratory and in the field.

E-mail: fortesfs@gmail.com A seleção e a crescente disseminação de nematoides resistentes aos anti-helmínticos mais comumente utilizados, benzimidazóis (BZs), imidazotiazóis e lactonas macrocíclicas (LMs), constituem um sério entrave na produção de pequenos ruminantes em todo o mundo. O uso de métodos eficientes e sensíveis para a detecção e o monitoramento da resistência anti-helmíntica no campo torna-se urgente, especialmente para os grupos de BZs e LMs, devido aos constantes relatos de resistência. A obtenção de um diagnóstico preciso e precoce da resistência é extremamente importante para auxiliar a tomada de decisão em programas de controle parasitário, com o objetivo de preservar a vida útil dos produtos e limitar o desenvolvimento da resistência nas populações de nematoides. Os testes in vivo e, mais recentemente, os testes in vitro têm sido desenvolvidos para a detecção de nematoides resistentes aos principais grupos de anti-helmínticos. No entanto, a disponibilidade de testes in vitro validados e o seu uso prático ainda são muito limitados. Embora o teste de redução na contagem de ovos nas fezes (TRCOF, in vivo - indireto) seja o principal método de escolha para a detecção de resistência no campo, vem recebendo críticas quanto à validade dos resultados, e passa por significativas modificações. Além disso, o desenvolvimento de técnicas moleculares a partir de alterações genômicas gerou avanços consideráveis nessa área de investigação, com o uso de mutações nos códons 167, 198 e 200 do gene da β-tubulina como principais SNPs (polimorfismos de nucleotídeo único; do inglês Single Nucleotide Polymorphisms) associados à resistência aos BZs. A presente revisão tem o objetivo de discutir os métodos de diagnóstico disponíveis para a detecção de resistência anti-helmíntica em nematoides de pequenos ruminantes, destacando progressos e obstáculos para seu uso na rotina laboratorial e no campo.

Despite the successful development of pesticides against endo and ectoparasites found in domestic ruminants, these parasites are still the major problem of the herbivore production system. The purpose of this study was to know the population of gastrintestinal parasites and their influence on weight gain of calves kept in organic and conventional grazing. Thus, organic and conventional calves were randomly selected in 2008 and 2009. The fecal egg count (FEC) indentified the following genders of helminths: Haemonchus, Trichostrongylus, Oesophagostomum, Cooperia, Strongyloides, Trichuris and oocysts of Eimeria. There was no significant difference (p>0.05) between FEC in organic and conventional animals. Calves younger than 6 months showed significant higher infection (p<0.05) than calves between 7 and 12 months of age. The weight gain observed during the study was of 327g/day and 280g/day for conventional and organic systems animals, respectively. Consequently, the combination of sustainable practices of grazing associated with the selective application of anthelmintics may be a feasible alternative for nematode control in a conventional system and in transition to an organic one.

Não foi observada diferença significativa (p>0,05) entre a FEC nos animais orgânicos e convencionais. Animais com idade inferior a seis meses apresentaram infecção significativamente superior (p<0,05) aos animais com 7-12 meses de idade. O ganho de peso observado durante o estudo foi de 327g/dia e 280g/dia para os animais dos sistemas convencional e orgânico, respectivamente. #4 - Evaluation of resistance in selected field strain of Haemonchus contortus to ivermectin and moxidectin using the Larval Migration on Agar Test, 33(2):183-187

E-mail: fortesfs@gmail.com Haemonchus contortus is one of the most common and economically significant causes of disease in small ruminants worldwide, and the control programs of parasitic nematodes - including H. contortus - rely mostly on the use of anthelmintic drugs. The consequence of the use of this, as the sole sanitary strategy to avoid parasite infections, was the reduction of the efficacy of all chemotherapeutic products with a heavy selection for resistance. The widespread of anthelmintic resistance and the difficulty of its early diagnosis has been a major concern for the sustainable parasite management on farms. The objective of this research was to determine and compare the ivermectin (IVM) and moxidectin (MOX) effect in a selected field strain of H. contortus with a known resistance status, using the in vitro larval migration on agar test (LMAT). Third stage larvae of the selected isolate were obtained from faecal cultures of experimentally infected sheep and incubated in eleven increasing diluted concentrations of IVM and MOX (6, 12, 24, 48, 96, 192, 384, 768, 1536, 3072 and 6144µg/mL). The dose-response sigmoidal curves were obtained using the R2 value of >0.90 and the lethal concentration (LC50) dose for the tested anthelmintic drugs using a four-parameter logistic model. The LC50 value for MOX was significantly lower than IVM (1.253µg/mL and 91.06µg/mL), identifying the H. contortus isolate as considerably less susceptible to IVM compared to MOX. Furthermore, the LMAT showed a high consistency (p<0.0001) and provided to be a useful diagnostic tool for monitoring the resistance status of IVM and MOX in H. contortus field isolate, as well as it may be used for official routine drug monitoring programs under the Ministry of Agriculture (MAPA) guidance.

O objetivo desta pesquisa foi determinar e comparar o efeito da ivermectina (IVM) e da moxidectina (MOX) em um isolado de campo selecionado de H. contortus com um estado de resistência conhecido, utilizando o teste in vitro de migração de larvas em ágar (LMTA). Larvas de terceiro estágio de um isolado de H. contortus selecionado foram obtidas a partir de culturas de fezes de ovinos infectados experimentalmente e incubadas em onze concentrações diluídas crescentes de IVM e MOX (6, 12, 24, 48, 96, 192, 384, 768, 1536, 3072 e 6144µg/mL). As curvas sigmoides de dose-resposta foram obtidas utilizando o valor de R2 >0,90 e a dose de concentração letal (CL50) para as drogas anti-helmínticas testadas, utilizando um modelo logístico de quatro parâmetros. O valor de CL50 para MOX foi significativamente menor do que para IVM (1,253µg/mL e 91,06µg/mL), identificando o isolado de H. contortus como consideravelmente menos suscetível à IVM em comparação à MOX. Além disso, o LMTA mostrou uma alta consistência (p<0,0001) e pode ser uma ferramenta útil de diagnóstico para monitorar o status de resistência de IVM e MOX em isolado de campo de H. contortus, assim como ser utilizado de forma oficial e em rotina para programas de monitoramento das drogas sob a demanda do Ministério da Agricultura (MAPA).

E-mail: alfredosps@hotmail.com This study was conducted to find effective anthelmintic treatments on a multi-resistant population of gastrointestinal nematodes of sheep, which was found resisting to all the classes of nematicidal drugs available in the Brazilian market. We took as a starting point the data obtained in a previous study performed in the targeted flock, in which were tested conventional treatments. Thus, using the calculated efficacy of the drugs (tested previously) for the genera of nematodes found in the flock, we selected drugs which, combined or used in higher doses, could improve the efficacy of the treatments. We evaluated the anthelmintic action of each treatment using the percentage of reduction of eggs in the feces, along with larvae cultures. A combination of moxidectin 1% + disophenol 20% showed 99% of efficacy, and combinations of moxidectin 1% + trichlorphon 10% and moxidectin 1% + levamisole phosphate 22.3% showed efficacy over 90%. Combinations of drugs, with different mechanisms of action, can provide effective anthelmintic treatments on multidrug-resistant populations of gastrointestinal nematodes of sheep.

Valendo-se da eficácia calculada de tratamentos convencionais a partir de um estudo prévio e, assim, conhecendo-se sua ação sobre diferentes gêneros de nematódeos presentes no rebanho, selecionaram-se drogas as quais, utilizadas em combinações ou em altas doses, pudessem aumentar significativamente a eficácia dos tratamentos. Utilizou-se o percentual de redução da contagem de ovos nas fezes e as culturas de larvas para avaliar a ação anti-helmíntica dos tratamentos testados. A combinação de moxidectina 1% + disofenol 20% apresentou 99% de eficácia, e as combinações de moxidectina 1% + triclorfon 10% e de moxidectina 1% + fosfato de levamisol 22,3% superaram os 90% de eficácia. A utilização de combinações de princípios ativos, com diferentes mecanismos de ação, pode promover a recuperação da eficácia dos tratamentos anti-helmínticos sobre populações multirresistentes de nematódeos gastrintestinais de ovinos.

Pesquisa Veterinária Brasileira 31(1):65-71. Hospital Veterinário, Centro de Saúde e Tecnologia Rural, Universidade Federal de Campina Grande, Patos, PB 58700-970, Brazil. E-mail: valery.medeiros@hotmail.com This paper reviews some important concepts to be considered for the control of gastro-intestinal nematode parasites of sheep and goats, including the epidemiology of this parasitic disease in the Brazilian semiarid, and the occurrence of anti-helminthic resistance in the region. Considering the widespread use of preventive treatments during the dry season, new alternatives for the control of gastrointestinal parasites are recommended, taking into account the parasite integrated control system and the use of methods to allow the presence of susceptible parasites in the refugia. Differences between sheep and goats are discussed, taking into account the higher susceptibility of goats and the necessity to use larger doses in goats than in sheep for most anti-helminthics.

Descrevem-se as características epidemiológicas desta parasitose no semiárido e o aparecimento de resistência aos anti-helmínticos na região. São propostas alternativas para o controle da doença levando em consideração, principalmente, a adoção de técnicas de controle integrado de parasitas com uma mudança nos sistemas de tratamento preventivo para sistemas que permitam a sobrevivência de parasitas susceptíveis na refugia. São discutidas as diferenças entre caprinos e ovinos, considerando que os caprinos são mais susceptíveis e que para maioria das drogas devem ser tratados com doses maiores.

E-mail: atsocamil@yahoo.com.br The aim of this paper was to evaluate the resistance of nematodes to some anthelmintics in dairy goats in Cariri Paraibano, Brazil. A total of 144 female goats, over 8 months of age, were used in the dry season and 120 ones during the rainy season, divided into four groups: Group I untreated, Group II treated with albendazole 10%, Group III treated with ivermectin 1%, and Group IV treated with levamisole phosphate 18.8%, at doses recommended by the manufacturer. To evaluate the resistance, the reduction in egg count per gram of feces and the cultivation of larval helminths. The fecal samples were collected on the day of treatment and, after 7, 14 and 21 days of treatment. In the group treated with albendazole, 61%, 11% and 24% of efficiency was observed in the dry period, and 55%, 14% and 12% in the rainy season, at 7, 14, and 21 days respectively. In the group treated with ivermectin, efficacy was 14%, 70% and 66% for the dry period, and 76%, 34% and 71% for the rainy season, at 7, 14 and 21 days respectively. The group treated with phosphate Levamisole showed efficacy rates of 89%, 79% and 73% in the dry period, and 76%, 69% and 67% in the rainy season, at 7, 14 and 21 days. The results indicate that the gastrointestinal nematodes of some dairy goats in Cariri Paraibano are not sensitive to the active Albendazole, Levamisole and Ivermectin. During the study, presence of specimens of the genera Haemonchus, Trichostrongylus, Oesophagostomum and Strongyloides was identified.

E-mail: atsocamil@yahoo.com.br RESUMO.- Objetivou-se avaliar a resistência de nematódeos a alguns anti-helmínticos em rebanhos caprinos no Cariri Paraibano. Foram utilizados 144 animais no período seco e 120 animais no período chuvoso, fêmeas com idade acima de oito meses de vida e lactantes, distribuídos em 4 grupos: Grupo I não tratado, Grupo II tratado com albendazole a 10%, Grupo III tratado com ivermectina a 1% e Grupo IV tratado com fosfato de levamisole a 18,8%, nas doses recomendadas pelos fabricantes. Para avaliar a resistência, aplicou-se o teste de redução na contagem de ovos por grama de fezes (RCOF) e o cultivo de larvas de helmintos. As amostras fecais foram coletadas no dia do tratamento (dia base) e 7, 14 e 21 dias após o tratamento. No grupo tratado com Albendazole, observaram-se eficácias de 61%, 11% e 24% no período seco e de 55%, 14% e 12% no período chuvoso, aos 7, 14, e 21 dias, respectivamente. No grupo tratado com Ivermectina, a eficácia foi de 14%, 70% e 66% para o período seco, e de 76%, 34% e 71% para o período chuvoso, aos 7, 14 e 21 dias, respectivamente. O grupo tratado com fosfato de Levamisole apresentou percentuais de eficácia de 89%, 79% e 73% no período seco e de 76%, 69% e 67% no período chuvoso, aos 7, 14 e 21 dias, respectivamente. Os resultados obtidos indicam que os nematódeos gastrintestinais de alguns rebanhos caprinos no Cariri Paraibano não são sensíveis aos princípios ativos Albendazole, Levamisole e Ivermectina. Durante o período de estudo foi identificada a presença de parasitas dos gêneros Haemonchus, Trichostrongylus, Oesophagostomum e Strongyloides.

The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. In larval culture Haemonchus, followed by Bunostomum, Trichostrongylus, and Oesophagostomum, were identified even after treatments.

ABSTRACT.- Vieira L.S. & Cavalcante A.C.R. 1999. [Anthelmintic resistance in goat herds in the State of Ceará.] Resistência anti-helmíntica em rebanhos caprinos no Estado do Ceará. Pesquisa Veterinária Brasileira 19(3/4):99-103. Embrapa-CNPC, Km 4 da Estrada Sobral-Groaíras, Cx. Postal 0-10, Sobral, CE 62011-970, Brazi. Goats of 45 farms in the State of Ceará, Brazil, were treated with anthelmintics for gastrointestinal nematodes, and their resistance to the anthelmintics was evaluated. On each farm 30 kids were weighed, ear-tagged and divided into three groups of ten. The first group received oxfendazole at 4.75mg/kg, the second levamisole at 7.5mg/kg, and the third group remained untreated as control. All goats were drenched according to their individual body weight. Fecal samples were collected from all animals (treated and control) on the day of treatment and 7 days later, to provide material for egg counts and larval cultures. Among 34 surveyed herds 20.6% showed levamisole resistance, 17.6% showed resistance to benzimidazole, and 35.3% had multiple resistance. At the time of the assessment 52.9% of the farmers were using broad spectrum anthelmintics. Only 26.5% of the surveyed herds had nematode populations susceptible to the anthelmintics assessed. The results of larval cultures showed that larvae surviving the treatment with oxfendazole were mainly Haemonchus sp and, to a lesser extent, Oesophagostomum sp; those surviving levamisole treatment were Haemonchus sp, Oesophagostomum sp and Trichostrongylus sp.

RESUMO.- Vieira L.S. & Cavalcante A.C.R. 1999. [Anthelmintic resistance in goat herds in the State of Ceará.] Resistência anti-helmíntica em rebanhos caprinos no Estado do Ceará. Pesquisa Veterinária Brasileira 19(3/4):99-103. Embrapa-CNPC, Km 4 da Estrada Sobral-Groaíras, Cx. Postal 0-10, Sobral, CE 62011-970, Brazi. Um levantamento em nível de campo sobre resistência anti-helmíntica em nematódeos gastrintestinais de; caprinos foi realizado em 34 rebanhos no Estado do Ceará. Em cada rebanho foram separados 30 cabritos, de ambos os sexos, com idade variando de 1 a 6 meses, os quais foram individualmente pesados, identificados e distribuídos em três tratamentos: 1) Oxfendazole na dose de 4, 75mg/kg; 2) Levamisole na dose de 7,5 mg/kg e 3) Controle (não medicado). Os anti-helmínticos foram administrados de acordo com o peso individual de cada animal e, a dosagem utilizada para cada produto foi a recomendada pelo laboratório fabricante. Foram colhidas fezes dos animais de todos os tratamentos, para OPG e coprocultura, no dia da medicação e 7 dias após. Dos 34 rebanhos avaliados, 7 (20,6%) apresentaram resistência aos imidazóis, 6 (17,6%) aos benzimidazóis e 12 (35,3%) revelaram resistência múltipla. Apenas em 9 rebanhos (26,5%), os nematódeos foram sensíveis aos anti-helmínticos avaliados. Através do questionário aplicado detectou-se que 52,9% dos caprinocultores entrevistados usavam anti-helmínticos de amplo espectro. Os resultados das coproculturas mostraram que os gêneros sobreviventes à medicação com oxfendazole foram principalmente Haemonchus sp, seguido em menor frequência por Oesophagostomum sp, enquanto que ao cloridrato de levamisole sobreviveram Haemonchus sp, Oesophagostomum sp e Trichostrongylus sp.
