#1 - Acute-phase protein concentration in bronchoalveolar lavage fluid from healthy horses

Abstract in English:

Bronchoalveolar lavage fluid (BALF) was analyzed to obtain information on leakage of acute-phase proteins from the blood into the respiratory lumen and about local synthesis. Ceruloplasmin, transferrin, albumin, α1-antitripsin, immunoglobulin G heavy, immunoglobulin G light, immunoglobulin A, haptoglobin, acidic glycoprotein, and P23 were measured in BALF from 30 horses without inflammatory disease by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). In serum, the same proteins were identified except for α1-antitrypsin. In conclusion, this study demonstrated that polyacrylamide gel electrophoresis (SDS-PAGE) can be used for the determination of acute-phase proteins in BALF samples from horses. In healthy horses, the values are very low, but they can be compared with reference values to assist in the diagnosis of animals with respiratory diseases.

O líquido obtido através da lavagem broncoalveolar (LBA) foi analisado para obter informações sobre as proteínas da fase aguda. Ceruloplasmina, transferrina, albumina, α1-antitripsina, imunoglobulina G pesada, imunoglobulina G leve, imunoglobulina A, haptoglobina, glicoproteína ácida e P23 foram medidas nos LBA de 30 cavalos sem doença inflamatória por eletroforese em gel de poliacrilamida com dodecilsulfato de sódio (SDS-PAGE). No soro, as mesmas proteínas foram identificadas, exceto a α1-antitripsina. Em conclusão, este estudo demonstra que a eletroforese em gel de poliacrilamida (SDS-PAGE) pode ser usada para a determinação de proteínas de fase aguda em amostras de LBA em cavalos. Em cavalos saudáveis, os valores são muito baixos, no entanto, podem ser comparados e auxiliar no diagnóstico de animais com doenças respiratórias.


#2 - ELISA of amyloid A in paired bronchoalveolar lavage fluid and serum samples of healthy horses

Abstract in English:

Pulmonary disorders are common in horses, and treatment efficiency depends on an adequate diagnosis. Amyloid A is the most sensitive indicator of pathology in horses. The objective of this study was to establish the concentration of amyloid A of bronchoalveolar lavage fluid (BALF) in healthy horses. Health condition of horses was considered normal based on physical examination, complete blood count, biochemical parameters, and BALF cytology. Blood and BALF were collected from thirty adult female horses. Amyloid A concentrations in serum and BALF were measured using commercial ELISA tests. Amyloid A was detected in serum (mean ± SD = 3.71±2.51) and BALF (mean ± SD = 0.000745±0.000785) of all horses. In conclusion, SAA can also be measured in bronchoalveolar fluid, affording early detection of respiratory infections or inflammatory conditions.

Distúrbios pulmonares são comuns nos cavalos e a eficiência do tratamento depende de um diagnóstico adequado e precoce. A amilóide A é um biomarcador sensível na deteccção de patologias inflamatórias e infecciososa em cavalos. O objetivo deste estudo foi estabelecer a concentração de amilóide A no líquido broncoalveolar (LBA) em cavalos saudáveis. Os cavalos foram considerados saudaveis baseado nos achados de normalidade do exame físico, hemograma, parâmetros bioquímicos e citologia do LBA. Sangue e LBA foram coletados de 30 fêmeas equinas adultas. Os níveis de Amilóide A no soro e no LBA foram mensurados por meio do teste de ELISA. A amilóide A foi detectada no soro (média ± DP = 3,71±2,51) e no LBA (média ± DP = 0,000745±0,000785) de todos os animais. Conclui-se que a amilóide A também pode ser mensurada no LBA, auxiliando no diagnóstico precoce de processos inflamatórios e infecciosos pulmonares.


#3 - Fungal pneumonia in dogs and cats with pulmonary clinical signs in southern Brazil

Abstract in English:

Fungal pneumonia has been a differential diagnosis in dogs and cats with pulmonary clinical signs, posting also a public health risk to respective owners. Since a common-source environmental exposure may result in infection, dogs and cats may also act as sentinels for both animal and human disease. Accordingly, the present study has aimed to evaluate the occurrence of fungal pneumonia in dogs and cats with pulmonary clinical signs from May 2013 to February 2015 in southern Brazil. Thoracic radiographs, blood work and non-bronchoscopic Bronchoalveolar Lavage (BAL), followed by cytology and microbiologic analysis were performed in all animals. One dog was positive in the BAL fluid culture for a pathogenic fungus (Cryptococcus neoformans) and another dog to an opportunistic fungus (Candida parapsilosis). Both dogs have presented cough as the prior clinical sign and showed thoracic radiographic changes. In conclusion, the occurrence of fungal pneumonia in southern Brazil in dogs and cats through of this study was relatively low. The BAL fluid culture may allow diagnosis of fungal pneumonia caused by opportunistic or pathogenic fungi and this diagnosis should be a concern in dogs and cats with pulmonary clinical signs.

Pneumonia fúngica tem sido um diagnóstico diferencial em cães e gatos com sinais clínicos pulmonares, sendo também um risco de saúde pública para seus respectivos proprietários. Uma vez que uma fonte comum de exposição ambiental pode resultar em infecção, cães e gatos podem agir como sentinelas da doença para ambos, pessoas e animais. Assim, o presente estudo teve como objetivo avaliar a ocorrência de pneumonia fúngica em cães e gatos com sinais clínicos pulmonares, de maio de 2013 a fevereiro 2015 no sul do Brasil. Radiografias torácicas, exame de sangue e lavado broncoalveolar (LBA) não broncoscópico seguido de análise citológica e microbiológica foram realizados em todos os animais. Um cão foi positivo na cultura do fluido do LBA para um fungo patogênico (Cryptococcus neoformans) e outro cão para um fungo oportunista (Candida parapsilosis). Ambos os cães apresentaram tosse como sinal clínico principal e alterações radiológicas torácicas. Em conclusão, a ocorrência de pneumonia fúngica no sul do Brasil em cães e gatos por meio deste estudo foi relativamente baixa. A cultura do LBA pode possibilitar o diagnóstico de pneumonia fúngica causada por fungos oportunistas ou patogênicos e este diagnóstico diferencial deve ser uma preocupação em cães e gatos com sinais clínicos pulmonares.

E-mail: 19.schneider@gmail.com Cytology is an important supplementary test used for the detection of changes in the mobile profile of the respiratory tract and aid in the diagnosis of diseases. However, as the first few months of life make up the period of adjustment to extrauterine life resulting in possible changes in cellular populations, it is essential to the standardization of the characteristics common to healthy animals, enabling the identification and analysis of any change in the expected frame. The objective of this research was to characterize the cellular profile by cytology of bronchoalveolar lavage obtained weekly for bronchoscopy of ten calves during the first three months of life. Statistical analysis was performed using the Minitab® 15.0 software, using the paired T test for parametric samples, and Mann-Whitney for nonparametric samples, considering significance level as P≤0,05. The results obtained showed that the profile of bronchoalveolar lavage of healthy calves during the first 90 days of life showed a predominance of macrophages, with medium average of 63.17%, followed by 33.69% neutrophils. However it was observed significant decrease in percentage of macrophages (P=0.04) and increased neutrophils (P=0,05) over the times, proven by the strong negative correlation between percentages of macrophages and neutrophils along the times. There was no difference between the percentages of giant cells, epithelial cells, lymphocytes, eosinophils and basophils during the experiment. Although the results are influenced by environmental factors and management, the results of this research provide subsidies for the identification of critical changes in cell profile of calves affected by respiratory diseases.

No entanto, como os primeiros meses de vida compõem o período de adaptação à vida extrauterina resultando em possíveis alterações das populações celulares, é essencial a padronização das características comuns aos animais sadios, possibilitando a identificação e análise de qualquer mudança no quadro esperado. O objetivo desta pesquisa foi caracterizar o perfil celular por citologia do lavado broncoalveolar obtido semanalmente por broncoscopia de dez bezerros durante os três primeiros meses de vida. A análise estatística foi realizada utilizando-se o software Minitab® 15.0, empregando-se o teste T pareado para as amostras paramétricas e Mann-Whitney para as amostras não paramétricas, considerando nivel de significância P≤0,05. Os resultados obtidos apontaram que o perfil celular do lavado broncoalveolar de bezerros hígidos durante os primeiros 90 dias de vida apresentou predomínio de macrófagos, com média geral de 63,17%, seguido de 33,69% de neutrófilos. Porém observou-se diminuição significativa na porcentagem de macrófagos (P=0,04) e aumento de neutrófilos (P=0,05) ao longo dos momentos, comprovada pela forte correlação negativa entre as porcentagens de macrófagos e neutrófilos ao longo dos momentos. Não houve diferença entre as porcentagens de células gigantes, células epiteliais, linfócitos, eosinófilos e basófilos durante o experimento. Apesar dos resultados serem influenciados por fatores ambientais e de manejo, os resultados dessa pesquisa fornecem subsídios para a identificação de alterações críticas que descaracterizem o perfil celular de bezerros acometidos por doenças respiratórias.

E-mail: joziana@dmv.ufla.br Bronchoalveolar lavage (BAL) is a procedure that retrieves cells and other elements from the lungs for evaluation, which helps in the diagnosis of pulmonary diseases. The aim of this study was to perform this procedure for cellular analysis of BAL fluid alterations during experimental infection with Aelurostrongylus abstrusus in cats. Fourteen cats were individually inoculated with 800 third stage larvae of A. abstrusus and five non-infected cats lined as a control group. The BAL procedure was performed through the use of an endotracheal tube on the nineteen cats with a mean age of 18 months, on 0, 30, 60, 90, 120, 180 and 270 days after infection. Absolute cell counts in the infected cats revealed that alveolar macrophages and eosinophils were the predominant cells following infection. This study shows that the technique allows us to retrieve cells and first stage larvae what provides information about the inflammatory process caused by aelurostrongylosis.

A contagem absoluta de células dos animais infectados revelou que macrófagos alveolares e eosinófilos constituíram os tipos celulares predominantes durante todo o período de infecção. Os resultados deste trabalho demonstram que a técnica permite recuperar células e larvas de primeiro estádio de A. abstrusus, fornecendo informações importantes sobre o processo inflamatório causado na aelurostrongilose.

E-mail: dellalibera@usp.br The recognition of the impact of certains forms of stress in the animal well being and organic equilibrium, represents a challenge to the adoption of good practices. Thus, this research verified the impact of a painful rotine of the cattle husbandry can cause on pulmonary and systemic immunity. Was evaluated hemogram and cortisol, in four moments, being M1, M6, M7 e M8 (respectively seven days before and one, three and eight days after the painful challenge) and bronchoalveolar cytology,obtained by bronchoscopy, in the moments of M1, M6 e M8. There was a reduction of the erythrogram values in the first day after the challenge, compatible with the anaplasmosis and aggravated by the blood loss during the surgery and an influx of leukocytes to the pulmonary region. Eight days after the challenge, became evident the cortisol increase, enerating leukocytosis by neutrophilia and monocytosis in the blood with a probable reduction of chemotaxis to the lungs, turning the respiratory tract potentially more susceptible to infections, suggesting that this practice, even accompanied by analgesic protocol, increases the risk of pneumonia, affecting the animal well-being.

Assim, a presente pesquisa verificou o impacto de um desafio doloroso rotineiro dos bovinos na imunidade pulmonar e sistêmica. Avaliou-se hemograma e cortisol, em quatro momentos, sendo M1, M6, M7 e M8 (respectivamente sete dias antes e um, três e oito dias depois do desafio doloroso) e citologia broncolaveolar, obtida por broncoscopia, nos momentos M1, M6 e M8. Houve uma redução dos valores do eritrograma no primeiro dia após o desafio, compatível com anaplasmose e agravada pela perda de sangue durante a cirurgia e um influxo de leucócitos para a região pulmonar. Oito dias após o desafio, evidenciou-se aumento de cortisol, gerando uma leucocitose por neutrofilia e monocitose no sangue com provável redução de quimiotaxia para o pulmão, tornando o trato respiratório potencialmente mais susceptível a infecções, sugerindo que esta prática de manejo, mesmo acompanhado de protocolo analgésico, pode ser considerada um fator de risco a penumonias afetando o bem estar animal.

Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: febencli@usp.br The neonatal calf is a critical moment for adaptation of the newborn to extra uterine life. The respiratory tract is functionally very demanded and often affected by disease, resulting in direct loss of their function and causing serious economic losses in livestock. The basic point to reduce these losses is appropriate clinical evaluation of neonates; but the diagnosis based solely in physical examination is very difficult to establish. The use of complementary analysis such cytology of the respiratory tract becomes an important diagnostic tool; however their findings must be standardized in the face of different techniques employed. This research studied the dynamics of the cellularity of the bronchoalveolar and tracheobronchial region obtained through lung lavage harvested by nasotracheal catheterization technique and tracheocenthesis respectively, during the first month of life of healthy calves. The tracheobronchial cytology was influenced by the time, showing decreased number of alveolar macrophages and greater number of neutrophils, possibly increased by local irritation caused by the technique, which was repeated sequentially, and/or through greater stimulation of inhaled microorganisms deposited in this region. In the bronchoalveolar region no variation in the cellular constituents in function of time was found. The results allowed the conclusion the cell population of the tracheobronchial region has changed over the week-old calves, possibly due to the technique used and/or to the normal region physiology, represented by higher magnitudes of neutrophils. Otherwise, the cells of the broncholaveolar region showed a stable behavior during the first month of life of newborn calves, presenting numerical predominance of alveolar macrophages.

E-mail: febencli@usp.br O período neonatal dos bezerros é um momento crítico para adaptação do recém-nascido à vida extra uterina e o sistema respiratório, um dos mais exigidos funcionalmente, é frequentemente afetado por enfermidades, redundando no prejuízo direto da sua função e acarretando perdas econômicas importantes na pecuária. O ponto básico para reduzir estas perdas, é representado pela adequada avaliação clínica dos neonatos, todavia o diagnóstico baseado exclusivamente no exame físico é muito difícil de ser estabelecido. O uso de exames complementares como a citologia do trato respiratório torna-se uma ferramenta diagnóstica importante nestes casos, porém faz-se necessário, padronizar seus achados frente às diferentes técnicas empregadas para a sua obtenção. Assim, o presente estudo propôs-se acompanhar as variações dos constituintes celulares da região traqueobrônquica e broncoalveolar obtidos por lavados respiratórios pelos métodos de traqueocentese e por colheita nasotraqueal respectivamente, durante o primeiro mês de vida de bezerros sadios. Observou-se alteração no quadro citológico ao longo do tempo, quando a região traqueobrônquica foi lavada, expresso por diminuição da porcentagem de macrófagos alveolares, com aumento de neutrófilos, possivelmente, por maior irritação local provocada pela técnica, que se repetiu sequencialmente e/ou por maior estimulo de microorganismos inalados depositados nesta região. Na região broncoalveolar, não encontraram-se variações nos constituintes celulares em função do tempo. Os resultados permitiram a conclusão que a população celular da região traqueobrônquica modificou-se ao longo das semanas de vida dos bezerros, possivelmente pela técnica empregada e/ou fisiologia normal da região, sendo representadas por maiores magnitudes de neutrófilos. De modo diverso, na região broncolaveolar, as células evidenciaram um comportamento estável durante o primeiro mês de vida dos bezerros neonatos, apresentando predomínio numérico dos macrófagos alveolares.
