#1 - Epigenetic characterization of the H19/IGF2 locus in calf clones placenta

Abstract in English:

Somatic Cell Nuclear Transfer (SCNT-Cloning) is a promising technique in many areas and is based on genetically identical individuals. However, its efficiency is low. Studies suggest that the leading cause is inadequate epigenetic reprogramming. This study aimed to characterize the methylation pattern of the exon 10 regions of the IGF2 gene and the Imprinting Control Region (ICR) of the H19 gene in the placenta of cloned calves. For this study, female and male cloned calves presenting different phenotypes were used. Genomic DNA from these animals’ placenta was isolated, then treated with sodium bisulfite and amplified to the ICR/H19 and IGF2 loci. PCR products were cloned into competent bacteria and finally sequenced. A significant difference was found between controls and clones with healthy phenotypes for the ICR/H19 region. In this region, controls showed a hemimethylated pattern, as predicted in the literature due to this region has an imprinted control, while clones were showed less methylated. For the IGF2, no significant differences were found between controls and clones. These results suggest that different genomic regions in the genome may be independently reprogrammed and that failures in reprogramming the DNA methylation patterns of imprinted genes may be one of the causes of the low efficiency of SCNT.

A Transferência Nuclear de Células Somáticas (TNCS-Clonagem) é uma técnica promissora em várias áreas, e se baseia na produção de indivíduos geneticamente idênticos. No entanto, sua eficiência é baixa. Estudos sugerem que a principal causa seja uma reprogramação epigenética inadequada. O objetivo desse trabalho é caracterizar o padrão de metilação da região éxon 10 do gene IGF2 e da Região Controladora de Imprinting (ICR) do gene H19 na placenta de bezerros clonados. Para a execução do trabalho foram selecionados clones bovinos fêmeas e machos, apresentando diferentes fenótipos. O DNA da placenta desses animais foi extraído, e em seguida foi tratado com bissulfito de sódio e amplificado para os loci ICR/H19 e IGF2. Os produtos da PCR foram clonados em bactérias competentes e, por fim, sequenciados. Foi encontrada uma diferença significativa entre os controles e os clones com fenótipos saudáveis para a região da ICR/H19. Nesta região, os controles tiveram um padrão hemimetilado, como previsto pela literatura, devido essa região ser imprinted. Enquanto os clones encontravam-se menos metilados. Para a região do éxon 10 do IGF2, não foi encontrada diferença significativa entre controles e clones. Estes resultados sugerem que as diferentes regiões do genoma podem se reprogramar independente umas das outras e que falhas na reprogramação do padrão de metilação do DNA de genes imprinted podem ser uma das causas da baixa eficiência da TNCS.


#2 - Occurrence of virulence-related sequences and phylogenetic analysis of commensal and pathogenic avian Escherichia coli strains (APEC), p.533-540

E-mail: wds@unicamp.br The presence of iron uptake (irp-2, fyuA, sitA, fepC, iucA), adhesion (iha, lpfAO157/O141, lpfAO157/O154, efa, toxB) and invasion (inv, ial-related DNA sequences and assignment to the four main Escherichia coli phylogenetic groups (A, B1, B2 e D) were determined in 30 commensal E. coli strains isolated from healthy chickens and in 49 APEC strains isolated from chickens presenting clinical signs of septicemia (n=24) swollen head syndrome (n=14) and omphalitis (n=11) by PCR. None of the strains presented DNA sequences related to the inv, ial, efa, and toxB genes. DNA sequences related to lpfAO157/O154, iucA, fepC, and irp-2 genes were significantly found among pathogenic strains, where iucA gene was associated with septicemia and swollen head syndrome and fepC and irp-2 genes were associated with swollen head syndrome strains. Phylogenetic typing showed that commensal and omphalitis strains belonged mainly to phylogenetic Group A and swollen head syndrome to phylogenetic Group D. Septicemic strains were assigned in phylogenetic Groups A and D. These data could suggest that clonal lineage of septicemic APEC strains have a multiple ancestor origin; one from a pathogenic bacteria ancestor and other from a non-pathogenic ancestor that evolved by the acquisition of virulence related sequences through horizontal gene transfer. Swollen head syndrome may constitute a pathogenic clonal group. By the other side, omphalitis strains probably constitute a non-pathogenic clonal group, and could cause omphalitis as an opportunistic infection. The sharing of virulence related sequences by human pathogenic E. coli and APEC strains could indicate that APEC strains could be a source of virulence genes to human strains and could represent a zoonotic risk.

#3 - Antigenic differences of clones of Moraxella bovis

Abstract in English:

Some antigenic differences of clones of Moraxella bovis recovered from a 6 months old bullock with infectious keratoconjunctivitis were studied through different techniques. The clones were classified biochemically by standard procedures. Seven polyclonal sera were produced in rabbits and tested against 12 clones by double immunodiffusion. Six monoclonal antibodies produced against M. bovis GF9 were used to quantify titles contained in five clones through the hemagglutination inhibition technique. Six clones were used in an ELISA with the panel of monoclonal antibodies. Immunodiffusion detected that only one clone was different from the others, while hemagglutination showed that the five clones studied were different. ELISA showed no clear cut differences among the clones. It can be concluded that the M. bovis population present in the animal studied is antigenically heterogeneous and that the hemagglutination inhibition technique may detect these differences.

Foram estudadas diferenças antigênicas de clones de Moraxella bovis recuperados de um terneiro com 6 meses de idade, acometido pela primeira vez de ceratoconjuntivite infecciosa bovina. Sete soros policlonais produzidos em coelhos foram testados contra 12 clones por imunodifusão dupla. Seis anticorpos nionoclonais produzidos a partir da amostra GF9 de M. bovis foram usados para verificar títulos de cinco clones através da técnica da inibição da hemoaglutinação. O teste ELISA foi utilizado para detectar reações de seis clones. Através da imunodifusão verificou-se que apenas um clone foi diferente dos outros. Contudo, pela técnica de inibição da hemoaglutinação observou-se que os cinco clones estudados eram diferentes. O teste ELISA não demonstrou diferenças claras entre os clones. Pode-se concluir que a população de M. bovis presente no globo ocular do animal estudado é antigenicamente heterogênea.
