#1 - Expression patterns of mesenchymal stem cell-specific proteins in adipose tissue-derived cells: possible immunosuppressing agent in partial allograft for restoring the urinary bladder in rabbits

Abstract in English:

Adipose tissue-derived stem cells (ADSCs) are an attractive source of mesenchymal stem cells (MSCs) for use in tissue engineering and clinical applications. This paper focuses on the characterization of ADSCs used as immunosuppressive agent in rabbits undergoing partial allograft for urine bladder restorage. For this study highlighted the characterization of the ADSCs used as immunosuppressive agents in rabbits submitted to partial allograft for restoration of the urinary vesicle, using 25 animals, six months old, New Zealand. ADSCs at the third peal were characterized by the MSC-specific CD105, CD73 and CD90 expression and by the absence of the hematopoietic marker CD45, as revealed by flow cytometry analysis. Moreover, ADSCs were efficient in preventing allograft rejection from the urinary bladder, as judged by biochemical, clinical and ultrasonography analysis. Together, these results compose characterization of protein expression profiles and immunosuppressive functionality of ADSCs in rabbits, which had undergone partial allografts of the urinary bladder, foreseeing future applications in clinical practice.

As células mesenquimais derivadas de tecido adiposo (ADSCs) são uma fonte atraente de células-tronco mesenquimais (MSCs) para uso na engenharia de tecidos e suas aplicações clínicas. Este trabalho destacou a caracterização das ADSCs utilizadas como agentes imunossupressores em coelhos submetidos a aloenxerto parcial para restauração da vesícula urinária, sendo utilizados 25 animais, de seis meses de idade, Nova Zelândia. As ADSCs, após o terceiro repique, foram caracterizadas pela expressão específica de MSC CD105, CD73 e CD90 e pela ausência do marcador hematopoiético CD45, tal como revelado por análise de citometria de fluxo. Além disso, os ADSCs foram eficientes na prevenção da rejeição de aloenxertos da vesícula urinária, conforme avaliado por análises clínica, bioquímica e ultrassonográfica. Juntos, esses resultados compõem a caracterização dos perfis de expressão proteica e a funcionalidade imunossupressora de ADSCs em coelhos, que sofreram aloenxertos parciais da bexiga, prevendo futuras aplicações na prática clínica.


#2 - Comparative study of distinct techniques to determine differential leukocyte counts in milk

Abstract in English:

Milk somatic cell count (SCC) is the basis of mastitis and milk quality control programs, however it not differentiate the distinct leukocyte populations which in turn can improve the diagnosis of mastitis. Thus, the present study aimed to evaluate different techniques used to measure the distinct leukocyte populations in milk in attempt to improve the diagnosis of mastitis. Here, milk samples from 31 dairy cows (124 quarter milk samples) were used. The differential leukocytes count was determined by cytocentrifugation, direct microscopy smears, and monoclonal antibodies by flow cytometry. The automatic SCC was also performed. The results showed a positive and significant correlation between the proportion of polymorphonuclear leukocytes determined by all techniques and automatic cell count; although a discrete higher correlation between flow cytometry and automatic SCC was found. Furthermore, the present study reinforces the idea that macrophages were the predominant cell type in mammary gland with low SCC. The proportion of each leukocyte population differ among techniques, probably due to the subjectivity of the examiner in the evaluation of the differential leukocyte counts by cytocentrifugation and direct microscopy smears, which emphasize that flow cytometry can be a useful and feasible tool in the diagnosis and control of mastitis.

A contagem de células somáticas (CCS) é um parâmetro amplamente utilizado para monitorar a saúde do úbere e a qualidade do leite, porém não diferencia as distintas populações leucocitárias. Portanto, a diferenciação das populações celulares no leite pode aprimorar o diagnóstico da mastite bovina. Dessa forma, o objetivo do presente trabalho foi avaliar as diferentes técnicas de contagem diferencial de leucócitos no leite para diagnosticar precisamente a mastite. Para tal, foram utilizadas 31 vacas da raça holandesa preta e branca em lactação (124 quartos mamários). Foram empregadas a contagem automática de células somáticas, e a contagem diferencial de leucócitos pelas técnicas de citocentrifugação, contagem diferencial de leucócitos por esfregaço direto, e citometria de fluxo com a utilização de anticorpos monoclonais específicos para identificação de cada população leucocitária. Os resultados demonstraram correlação positiva e significativa entre a proporção de leucócitos polimorfonucleares pelas diferentes técnicas e a contagem automática de células somáticas, sendo observada uma correlação discretamente mais forte com a citometria de fluxo. Além disso, foi demonstrado que os macrófagos são a população predominante no leite oriundo de glândula mamária com baixa CCS. Observaram-se também diferenças na proporção das distintas populações leucocitárias entre as distintas técnicas, resultado da possível subjetividade do examinador na contagem diferencial de leucócitos pelas técnicas de citocentrifugação e contagem microscópica direta por esfregaços, o que reforça que a citometria de fluxo pode ser uma ferramenta confiável no controle e diagnóstico da mastite.

E-mail: karinamedici@yahoo.com.br The particular apocrine secretion of goat milk different from the merocrine one observed in cows, may lead to errors in interpreting cellularity evaluations in the milk of this species. Thus, the objective of the present trial was to determine Somatic Cell Counts by means of one indirect methods, the California Mastitis Test (CMT), and direct methods, flow cytometry and direct microscopic count using methyl green-pyronine-Y stain, beyond comparing the methods of cellular counting. A total of 102 samples from 51 Saanen, Brown Alpine and Toggenburg female goats, bred in the state of São Paulo, were analyzed. Goats were separated in groups according to the phase of lactation and to physical examination of the mammary gland, and milk examination. Samples were divided into two aliquots, and were collected after California Mastitis Test evaluation. One aliquot was used in automatic cell counts, and the other, in direct microscopic count using methyl green-pyronine-Y stain. CMT results were as follows: 74.5% of the samples were negative, 8.8% yielded traces, 8.8% were weak positive (1), 6.8% were distinct positive (2) and 0.9% were strong positive (3). Medians of somatic cell counts in goat milk as evaluated by means automatic cell counter and direct microscopy, and grouped according to the different CMT scores, were as follows: 181,000, 578,000, 628,000, 1,421,500, and 5,542,000 cells/mL of milk and 74,991, 271,396, 71,420, 640,995, and 5,049,394 cells/mL of milk in scores negative, traces, 1, 2 and 3, respectively. Medians obtained in automatic cell counts and direct microscopic counts, grouped according to the phase of lactation were 159,500; 508,000; and 277,500 cells/mL of milk and 62,493; 89,275; and 146,411 cells/ml of milk, respectively. The correlation between the automatic and microscopic methods for somatic cell counts was 88%. Based on the results obtained, it could be concluded that there were differences between the automatic and microscopic methods for somatic cell counts, being this most adequate for the determination of the celularidade in the cellularity of goat milk.

Portanto, o presente trabalho teve o objetivo de determinar a contagem de células somáticas pelo método indireto California Mastitis Test (CMT), e por métodos diretos, incluindo a contagem por citometria de fluxo e a contagem microscópica direta, através da coloração de verde de metil e pironina-Y, além de comparar os métodos de contagem celular. Foram analisadas 102 amostras de 51 fêmeas caprinas, das raças Saanen, Parda Alpina e Toggenburg, criadas no Estado de São Paulo. Os animais foram categorizados segundo a fase da lactação, exame físico da glândula mamária e exame do leite. As amostras foram colhidas, após a realização do exame Califórnia Mastitis Test, em duas alíquotas, uma destinada à contagem celular automática e a outra, a contagem microscópica direta, utilizando-se o corante verde de metil e pironina-Y. De acordo com os diferentes escores do CMT, observou-se 74,5% de amostras negativas, 8,8% de amostras com escore traços, 8,8% de amostras ligeiramente positivas (+), 6,8% de amostras fracamente positivas (++) e 0,9% de amostras fortemente positivas (+++). Os valores medianos das contagens de células somáticas presentes no leite de cabras, avaliadas através de contador automático e microscopia direta, e analisadas de acordo com os diferentes escores do CMT, foram, respectivamente, 181.000, 578.000, 628.000, 1.421.500 e 5.542.000 células/mL de leite e 74.991, 271.396, 71.420, 640.995 e 5.049.394 células/mL de leite, nos escores negativo, traços, +, ++ e +++. Os valores medianos obtidos através da contagem de células somáticas pelo método automático e microscópico direto, de acordo com as fases de lactação foram de 159.500, 508.000 e 277.500 células/mL de leite, e 62.493, 89.275 e 146.411. A correlação obtida entre a contagem celular automática e microscópica direta foi de 88%. A partir dos resultados observados pode-se concluir que existe diferença na contagem celular determinada através do método automático e microscópico sendo este último o mais adequado para a determinação da celularidade no leite de cabras.

Synovial fluid was collected from the metacarpophalangeal joints of athletic horses before exercise and 3, 6, 24 hours after exercise, and as well as from the control group (without exercise). Apoptosis/necrosis percentage, TNF-a and PGE2 were determined by annexin V/PI assay, bioassay (L929) and ELISA, respectively. The results showed that total leukocyte count was higher in the athletic group when is compared with the control group. Three hours after the exercise was done there were increases of cellular apoptosis (P>0.05) and necrosis (P<0.05) percentage, PGE2 concentration (P<0.05) and protein concentration (P<0.05), and the TNF-a level has dropped. The athletic group showed moderate level of joint inflammation after the strenuous exercise. This articular tissue response to biomechanical insult due to the exercise, with high intensity after 3 hours after training associated with normality after 24 hours, reveals the articular adaptation to physical stress in athletic horses.
