#1 - Safety and efficacy of allogeneic bone marrow mesenchymal stem cells for treatment of canine leukopenia induced by canine parvovirus infection

This study aims to establish a therapy strategy for canine leukopenia induced by canine parvovirus (CPV) infection through intravenous infusion of allogeneic bone marrow mesenchymal stem cells (BMMSCs) and to evaluate the therapeutic effect of BMMSCs on canine parvovirus. Forty healthy 2-month-old dogs were randomly divided into four groups including the BMMSC treatment group (A), conventional treatment group (B), CPV infection group (C), and a normal control group (D). Then the A, B, and C groups were orally infected with CPV (103.25 TCID50/mL) at 1mL/kg, and the D group received the same dose of saline. After the onset of infection, Group A received mesenchymal stem cells (MSCs) and rehydration as the treatment; Group B was treated with anti-inflammatory therapeutics and rehydration; and Group C and D were injected with the same dose of physiological saline. The level of leukocytes rebounded significantly after the treatment with BMMSCs and returned to reference numbers on Day 3 after treatment, which was significantly higher than that in the conventional treatment group. The concentrations of IL-2 and IFN-α were gradually increased during the treatment, and the BMMSC treatment group exhibited significantly higher IL-2 and IFN-α concentrations than the conventional treatment group on Days 3 and 4. The expression of the virus in the blood gradually decreased during the treatment, and the BMMSC treatment group displayed a faster decrease than the conventional treatment group. These results showed the advantages of BMMSC treatment over conventional treatment. This study provides a new BMMSC treatment strategy for canine leukopenia induced by CPV infection and reveals the mechanism by which BMMSC increases leukocytes after CPV infection.

Este estudo tem como objetivo estabelecer uma estratégia terapêutica de leucopenia canina induzida pela infecção por parvovírus canino (CPV) através de infusão intravenosa de células tronco mesenquimais da medula óssea alogênica (BMMSCs) e avaliar o efeito terapêutico de BMMSCs no parvovírus canino. Quarenta cães saudáveis de dois meses de idade foram divididos aleatoriamente em quatro grupos: o grupo de tratamento de BMMSCs (A), o grupo de terapia convencional (B), o grupo de infecção por CPV (C) e um grupo controle (D). Os grupos A, B e C foram infectados oralmente com CPV (103.25 TCID50/mL) a 1mL/kg e o D recebeu a mesma dose de soro fisiológico. Após o início da infecção, o grupo A recebeu uma dose de derivadas da medula óssea (CTMMO) e hidratação como tratamento, o grupo B foi tratado com terapia anti-inflamatória e hidratação, e grupos C e D foram injetados com a mesma dose de soro fisiológico. As concentrações de IL-2 e IFN-α aumentaram gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou concentrações significativamente maiores de IL-2 e IFN-α do que o grupo de tratamento convencional nos Dias 3 e 4. A expressão de vírus no sangue diminuiu gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou uma diminuição mais rápida do que o grupo de tratamento convencional. Esses resultados mostraram as vantagens do tratamento BMMSC em relação ao tratamento convencional. Este estudo fornece uma nova estratégia de tratamento da BMMSC para leucopenia canina induzida pela infecção por VCP e revela o mecanismo pelo qual a BMMSC aumenta os leucócitos após infecção por VCP.


#2 - Experimental infection by Trypanosoma vivax in goats in the Brazilian semiarid: detection of T. vivax DNA in colostrum and assessment of lactogenic transmission

Abstract in English:

This study aimed to identify the presence of Trypanosoma vivax DNA in the colostrum of infected goats and to explore the possibility of transmission for neonates fed using colostrum collected from infected goats. We used twelve goats in the final third of gestation with an age of approximately 24 months. Six goats were inoculated intravenously with 0.5mL of blood containing approximately 1.25x105 trypomastigotes of T. vivax, and six remained uninfected. The presence of T. vivax in colostrum was evaluated by Polymerase Chain Reaction (PCR). The possibility of T. vivax transmission by colostrum was assessed by feeding six neonates born of serologically negative goats using colostrum from infected goats. Peripheral blood from neonates was collected daily for thirty days to assess the T. vivax presence through the examination of Giemsa-stained smears of leukocyte layers with the buffy coat technique (BCT) and by PCR. The results of a direct examination of colostrum were negative, but PCR confirmed the presence of T. vivax DNA in all infected goats. Additionally, lactogenic transmission by colostrum was not demonstrated once both BCT and PCR of neonate peripheral blood were negative.

Este estudo teve como objetivo identificar a presença de DNA de Trypanosoma vivax no colostro de cabras infectadas experimentalmente e verificar a possibilidade de transmissão para neonatos alimentados com colostro coletado de cabras infectadas. Foram utilizadas doze cabras no terço final de gestação com idade aproximada de 24 meses. Seis cabras foram inoculadas intravenosamente com 0,5mL de sangue contendo aproximadamente 1,25x105 tripomastigotas de T. vivax, e seis permaneceram não infectadas. A presença de T. vivax no colostro foi avaliada por Reação em Cadeia da Polimerase (PCR). A possibilidade de transmissão de T. vivax pelo colostro foi avaliada através da alimentação de seis neonatos nascidos de cabras sorologicamente negativas com colostro de cabras infectadas. Foi coletado diariamente o sangue periférico dos neonatos, por trinta dias para avaliar a presença de T. vivax através do exame de esfregaços de camadas leucocitárias coradas por giemsa, pela técnica BCT e por PCR. Os resultados do exame direto do colostro foram negativos, mas a PCR confirmou a presença de DNA de T. vivax no colostro em todas as cabras infectadas. Além disso, a transmissão lactogênica pelo colostro não foi demonstrada, uma vez que tanto a BCT quanto a PCR do sangue periférico do neonato foram negativas


#3 - Evaluation of infection by Borrelia sp. in domestic and wild mammals and ticks from the Catimbau National Park, Pernambuco

Spirochetes in the genus Borrelia (Spirochaetes) are the etiological agents of two zoonotic diseases that have rodents as reservoirs: the Lyme disease, transmitted by ixodid ticks (Ixodidae), and tick-borne relapsing fever, which is transmitted mostly by argasid ticks (Argasidae). The information on the epidemiology of pathogens that affect animals is scarce, especially in places that have been barely studied, such as the Caatinga biome. Thus, in the present study, we aimed to detect DNA of Borrelia in the blood of wild and domestic mammals, as well as in ticks collected in the Catimbau National Park, State of Pernambuco, Northeast Brazil. Blood was collected from 77 captured wild animals belonging to Didelphidae (45), Echimyidae (21), Cricetidae (7), Caviidae (2), and Dasypodidae (2) families. Additionally, blood samples from 120 sheep, 119 goats, 70 dogs, and 47 cattle were obtained. A total of 286 ticks were collected in the environment and identified as Ornithodoros cf. tabajara (188) and Ornithodoros rietcorreai (98); in addition, larvae of Ornithodoros spp. were collected on Thrichomys laurentius (4 specimens) and Monodelphis domestica (1); larvae of O. rietcorreai on Didelphis albiventris (2) and T. laurentius (1); Amblyomma parvum on T. laurentius (4); Rhipicephalus sanguineus (sensu lato) on dogs (121) and Rhipicephalus microplus on cattle (47). All blood samples, 120 free-living ticks, 46 O. rietcorreai and 74 O. cf. tabajara, and 12 R. sanguineus s.l. were submitted to DNA extraction to perform a polymerase chain reaction (nested PCR), aiming at amplifying a fragment of the flaB gene present in all species of Borrelia. Only one free-living specimen of O. cf. tabajara (1.3%) was positive. Negative detections in mammals may be related to the absence of infection or the low concentration of the spirochetes in the hosts. The low prevalence of Borrelia found in ticks can be explained by the low infection rates that these spirochetes depict in their vectors. New studies focusing on the detection of antibodies, as well as on the sequencing and isolating Borrelia spp. should be encouraged for a better understanding of the epidemiology of these bacterial agents in the region.

No gênero Borrelia (Spirochaetes) encontram-se dois grupos de agentes responsáveis por duas doenças zoonóticas, que têm roedores como reservatórios e acometem humanos: o grupo de Lyme, transmitido por carrapatos ixodídeos, e a febre recorrente, que pode ser transmitida por carrapatos argasídeos ou piolhos. Poucas são as informações sobre epidemiologia e agentes patogênicos que acometem animais, podendo representar uma riquíssima fonte de informações, principalmente em locais nunca estudados, pois podem ser bioindicadores de qualidade do ambiente e saúde. Desta forma, no presente estudo foi realizada a pesquisa de Borrelia spp. em amostras de sangue de mamíferos silvestres e domésticos, assim como de carrapatos, do Parque Nacional do Catimbau, Pernambuco. Foi realizada a captura e colheita sanguínea de 77 animais pertencentes às famílias Didelphidae (45), Echimyidae (21), Cricetidae (7), Caviidae (2) e Dasypodidae (2). Adicionalmente, amostras de sangue foram obtidas de 120 ovinos, 119 caprinos, 70 cães e 47 bovinos. Foram coletados 286 carrapatos no ambiente, identificados como Ornithodoros cf. tabajara (188) e Ornithodoros rietcorreai (98); além de carrapatos coletados nos animais: Ornithodoros spp. em Thrichomys laurentius (4 espécimes) e Monodelphis domestica (1); O. rietcorreai em Didelphis albiventris (2) e T. laurentius (1); Amblyomma parvum em T. laurentius (4); Rhipicephalus sanguineus (sensu lato) em cães (121) e R. microplus em bovinos (47). Todas as amostras sanguíneas, 120 carrapatos de vida livre, sendo 46 O. rietcorreai e 74 O. cf. tabajara, e 12 R. sanguineus s. l. foram submetidos à extração de DNA para realização da reação em cadeia da polimerase (nested-PCR), visando amplificação de um fragmento do gene flaB presente em todas as espécies de Borrelia. Não houve amplificação nas amostras sanguíneas dos animais, no entanto em um espécime de O. cf. tabajara de vida livre (1,3%) foi detectado o DNA de Borrelia. A não detecção nos mamíferos pode estar relacionada com a ausência de infecção ou à baixa concentração do agente nos hospedeiros. A baixa prevalência encontrada nos carrapatos pode ser justificada devido à baixa infecção nos vetores. Novos estudos focando na determinação de anticorpos, assim como no sequenciamento e isolamento de Borrelia devem ser estimulados para uma melhor compreensão da epidemiologia do agente na região.


#4 - Pathology of Spirorchiidae (Digenea: Schistosomatoidea) infection in green turtles (Chelonia mydas) on Campos Basin, Rio de Janeiro

Abstract in English:

Spirorchidiasis is one of the most common diseases in green turtles. Eggs and the adult form of trematodes are mainly found in the heart, liver, lung, spleen, and gastrointestinal tract. The present study aimed to describe the pathological aspects of Spirorchiidae trematode infection in green turtles (Chelonia mydas) in the Campos Basin, Rio de Janeiro. The current study was based on the necropsy of 99 green turtles (Chelonia mydas, Chelonidae) from April to December 2021. Epidemiological data were obtained from the Aquatic Biota Monitoring Information System (SIMBA). A histopathological examination was performed at the “Setor de Anatomia Patológica” (SAP) from “Universidade Federal Rural do Rio de Janeiro” (UFRuralRJ). Tissue fragments (buccal papilla, esophagus, trachea, lung, heart, kidney, bladder, stomach, liver, spleen, intestines, gonads, adrenal, thyroid, pancreas, salt glands, muscle, and skin) were sampled for histological examination. The green turtles had lesions compatible with parasitic infection in 89% (88/99) cases. Out of 88 turtles, 87 were juveniles (99%), 75% (66/88) were females, and 25% (22/88) were males. Adult parasites and their eggs were found to cause granulomatous tissue reactions in 14% (12/88) of turtles. Egg-associated granulomatous inflammation was found in 86% (76/88) of the turtles. The organs with the highest number of egg-associated granulomas in order of occurrence were: the spleen, small intestine, adrenal gland, pancreas, and lungs. The adult trematodes were observed in histological lesions in order of occurrence, mainly in the esophagus, stomach, and small intestine. Out of the total number of turtles parasitized, 33% (29/88) of deaths occurred due to multiple causes such as fractures, interaction with fishing, predation, and fibropapillomas. The other 67% (59/88) presented multisystemic granulomatous disease caused by Spirorchiidae infection as a cause of death. Evidence is presented that spirorchidiasis is highly prevalent in young green turtles, is responsible for extensive chronic lesions, and responsible for significant debilitation and mortality.

A espirorquidíase é uma das doenças mais comuns em tartarugas verdes. Os ovos e a forma adulta dos trematódeos são encontrados principalmente no coração, fígado, pulmão, baço e trato gastrointestinal. O objetivo do presente estudo foi descrever os aspectos patológicos da infecção por trematódeos Spirorchiidae em tartarugas verdes (Chelonia mydas) na Bacia de Campos, Rio de Janeiro. O presente trabalho foi baseado na necropsia de 99 tartarugas verdes (Chelonia mydas, Chelonidae) no período de abril a dezembro de 2021. Os dados epidemiológicos foram obtidos do Sistema de Informação de Monitoramento da Biota Aquática (SIMBA). Realizou-se exame histopatológico no Setor de anatomia Patológica (SAP) da Universidade Federal Rural do Rio de Janeiro (UFRuralRJ). Os fragmentos de tecidos (papila bucal, esôfago, traqueia, pulmão, coração, rim, bexiga, estômago, fígado, baço, intestinos, gônadas, adrenal, tireoides, pâncreas, glândulas de sal, músculo e pele) foram coletados e fixados em formalina a 10% tamponada, processados rotineiramente para exame histológico. Em 89% (88/99) dos casos, as tartarugas-verdes apresentaram lesões compatíveis com infecção parasitária. Das 88 tartarugas, 87 eram jovens (99%), 75% (66/88) correspondiam a fêmeas, 25% (22/88) a machos. Em 14% (12/88) das tartarugas foram encontrados parasitas adultos e seus ovos causando inflamação granulomatosa. Em 86,4% (76/88) das tartarugas foram encontradas lesões granulomatosas associadas aos ovos dos trematódeos. Os órgãos com maior quantidade de granulomas associados a ovos foram: baço, intestino delgado, adrenal, pâncreas e pulmões. Trematódeos adultos foram principalmente observados nas lesões histológicas, em ordem de ocorrência, no esôfago, estômago e intestino delgado. Do total de tartarugas parasitadas, 33% (29/88) das mortes ocorreram em consequência de múltiplas causas como fraturas, interação com pesca, predação e fibropapilomas. As outras 67% (59/88) apresentaram como causa de morte a doença granulomatosa multissistêmica causada pela infecção por trematódeo da família Spirorchiidae. São apresentadas evidências de que a espiroquidiase é altamente prevalente em tartarugas verdes jovens no Rio de Janeiro, responsável por extensas lesões crônicas, quadro clínico de debilidade e mortalidade significativas.


#5 - Real-time PCR quantification and histopathological findings of Mycoplasma hyopneumoniae infection in the lungs of pigs slaughtered in São Luís, Maranhão, Brazil

Abstract in English:

Porcine enzootic pneumonia (PES), mainly caused by the bacteria Mycoplasma hyopneumoniae, is the main cause of respiratory problems in pigs. Infection by M. hyopneumoniae leads to production losses and the predisposition of affected animals to secondary infections, which may result in the condemnation of carcasses and organs due to lung lesions at the time of slaughter. The objective of the research was to evaluate the infection by M. hyopneumoniae in pigs submitted to slaughter in São Luís Island/MA, using molecular and histopathological diagnostic methods. One hundred fifty lung samples were collected from inspected (n=65) and non-inspected (n=85) slaughter pigs on São Luís Island, Maranhão, from July 2019 to August 2021. Of the 150 DNA samples collected, 121 showed an amplified product for Cyt B in the PCR assay. Thus, 121 samples were submitted to qPCR of M. hyopneumoniae, of which 44 (36.36%) showed positive results. The mean amount of bacterial load ranged from 1.20 × 101 to 7.20 × 104, with a mean of 1.73 × 104 copies. Of the reagent samples, 81.81% (36 samples) were obtained from non-inspected slaughter, while 18.18% (8 samples) were obtained from slaughterhouses. In the histopathological analysis, 44 positive qPCR samples were evaluated, of which 28 (63.63%) presented results compatible with the main inflammatory process associated with the presence of M. hyopneumoniae, that is, bronchial-associated lymphoid tissue hyperplasia (BALT). Three samples that showed the highest bacterial load (qPCR: 5.63 × 10³, 2.19 × 104 and 7.23 × 104) showed more evident lesions in this study. The microscopic findings associated with the quantifications indicated a relationship between the amount of bacterial load and the presence of microscopic lesions; higher bacterial load in lung tissue is associated with increased histopathologic staining for BALT hyperplasia. In conclusion, the results point to the circulation of the etiological agent in the sampled animals and the need for preventive measures on pig farms in Maranhão with the involvement of producers, sanitary defense and inspection agencies.

A pneumonia enzoótica suína (PES), causada principalmente pela bactéria Mycoplasma hyopneumoniae, é a principal causa de problemas respiratórios em suínos. A infecção por M. hyopneumoniae leva a perdas produtivas e a predisposição dos animais acometidos a infecções secundárias, o que pode resultar em condenação de carcaças e órgãos por lesões pulmonares no momento do abate. O objetivo da pesquisa foi avaliar a infecção por M. hyopneumoniae em suínos submetidos ao abate na Ilha de São Luís, por meio de métodos diagnósticos moleculares e histopatológicos. Para isso, foram coletadas 150 amostras de pulmão de suínos de abate inspecionado (n=65) e não inspecionado (n=85) na Ilha de São Luís/Maranhão, no período de julho de 2019 a agosto de 2021. Das 150 amostras de DNA coletadas, 121 apresentaram produto amplificado para Cyt B no ensaio de PCR. Assim, 121 amostras foram submetidas à qPCR de M. hyopneumoniae, das quais 44 (36,36%) apresentaram resultados positivos. A quantidade média de carga bacteriana variou de 1,20 × 101 a 7,20 × 104, com média de 1,73 × 104 cópias. Das amostras reagentes, 81,81% (36 amostras) foram obtidas de abate não inspecionado, enquanto 18,18% (8 amostras) foram obtidas em abatedouro. Na análise histopatológica, foram avaliadas 44 amostras positivas para qPCR, das quais 28 (63,63%) apresentaram resultados compatíveis com o principal processo inflamatório associado à presença de M. hyopneumoniae, ou seja, hiperplasia do tecido linfóide associado ao brônquio (BALT). Três amostras que apresentaram maior carga bacteriana (qPCR: 5,63 × 10³, 2,19 × 104 e 7,23 × 104) foram mais evidentes neste estudo. Os achados microscópicos associados às quantificações indicaram uma relação entre a quantidade de carga bacteriana e a presença de lesão microscópica; a maior carga bacteriana no tecido pulmonar está associada a maior alteração histopatológica para hiperplasia BALT. Em conclusão, os resultados obtidos sinalizam para a circulação do agente etiológico nos animais amostrados e a necessidade de medidas preventivas nas criações de suínos do estado do Maranhão com envolvimento dos produtores, órgãos de defesa sanitária e inspeção.


#6 - Cutaneous fungal infections secondary to avian pox in Northeast Brazil

Abstract in English:

This study describes the epidemiological and clinical-pathological aspects of outbreaks of avian pox diagnosed in poultry associated with fungal co-infections. A retrospective study was carried out, and cases of avian pox with suspected associated fungal infection were selected. The slides were subjected to routine and special histochemical staining in cases of suspected fungal infection, in addition to performing the immunohistochemical technique. Macroscopically, there were nodular lesions with crust formation, and histologically, the lesions were characterized by hyperplasia of the spinous layer associated with multiple eosinophilic intracytoplasmic inclusions. In seven cases, morphologically compatible structures with fungi were verified through black impregnation with GMS and strongly stained in pink with PAS. The morphotintorial aspects were suggestive of fungi belonging to the genus Aspergillus sp. and Candida sp. The diagnosis of fungal co-infections was confirmed through immunohistochemistry, with positive immunostaining for fungi of the genus Aspergillus sp. (five cases) and Candida sp. (two cases). It is concluded that the occurrence of fungal co-infections secondary to the cutaneous lesions of avian pox represents a complicating factor of the disease, favoring the weakness of these animals and death. Therefore, the investigation of associated secondary agents is necessary.

Este artigo descreve os aspectos epidemiológicos e clínico-patológicos de surtos de varíola aviária diagnosticada em aves domésticas associados a coinfecções fúngicas. Foi realizado um estudo retrospectivo e selecionados os casos de varíola aviária com suspeita de infecção fúngica associada. As lâminas foram submetidas às colorações histoquímicas de rotina e especiais, nos casos de suspeita de infecção fúngica, além de realização da técnica de imuno-histoquímica. Macroscopicamente, haviam lesões nodulares com formação de crostas, e histologicamente, as lesões caracterizavam-se por hiperplasia da camada espinhosa associada a múltiplas inclusões intracitoplasmáticas eosinofílicas. Em sete casos verificou-se a presença de estruturas morfologicamente compatíveis com fungos, através de impregnação em preto por GMS e coradas fortemente em rosa por PAS. Os aspectos morfotintoriais foram sugestivos de fungos pertencentes aos gêneros Aspergillus sp. e Candida sp. O diagnóstico das coinfecções fúngicas foi confirmado através da imuno-histoquímica, com imunomarcação positiva para fungos dos gêneros Aspergillus sp. (cinco casos) e Candida sp. (dois casos). Através desses dados, pode-se concluir que a ocorrência de coinfecções fúngicas secundárias às lesões cutâneas de varíola aviária representam um fator complicador da doença, favorecendo a debilidade desses animais e óbito. Sendo assim, é necessária a investigação de agentes secundários associados.


#7 - Efficacy of disinfectants to inactivate H1N1 influenza A virus isolated from pigs

Abstract in English:

The aim of this study was to access the efficacy of four disinfectants to inactivate influenza A [H1N1] 0 hour and 72 hours after disinfectant dilution. A pandemic H1N1 influenza virus isolated from a pig with respiratory disease was used to obtain inoculums containing 6.4log10 EID50/mL; 5.4log10 EID50/mL; 4.4log10 EID50/mL and 3.4log10 EID50/mL. Suspension test was composed of 400µL of viral inoculum, 100µL of organic load and 500µL of each individually diluted disinfectant and incubated for ten minutes of contact time. After a neutralizing step, each mixture was filtered on a 0.22µm membrane and 0.2mL was inoculated in six 9-day-old embryo chicken egg through allantoic route. The allantoic fluid from eggs was harvest for RT-PCR and hemagglutination test. The experiment was repeated 72 hours after disinfectant dilution. On the first assessment with fresh disinfectant, influenza virus was inactivated by oxidizing compost disinfectant and phenolic disinfectant in all virus concentrations, the quaternary ammonium compound (QAC) and glutaraldehyde association inactivated the virus up to a concentration of 5.4log10 EID50/mL. QAC disinfectant did not eliminate virus viability. Seventy-two hours after disinfectants were diluted, oxidizing compost disinfectant and QAC and glutaraldehyde association disinfectant demonstrated the same result as the evaluation with fresh disinfectant solution. Phenolic disinfectant inactivated viral inoculum up to a concentration of 5.4log10 EID50/mL. QAC had no effect on inactivating 3.4log10 EID50/mL of influenza virus. In conclusion, three of the four disinfectants tested were effective to inactivate pandemic H1N1 influenza virus in the presence of organic load. Test result performed 72hours after disinfectant dilution suggest a decrease in the effectiveness of one disinfectant.

O objetivo deste trabalho foi avaliar a eficácia de quatro desinfetantes em inativar o vírus da influenza A [H1N1] 0-hora e 72-horas após a diluição dos produtos. Um vírus H1N1 pandêmico isolado previamente de um suíno com doença respiratória foi utilizado e foram obtidas quatro concentrações de inóculo contendo 6,4log10 EID50/mL; 5,4log10 EID50/mL; 4,4log10 EID50/mL and 3,4log10 EID50/mL. Para compor o teste em suspensão foram adicionados 400µL de inóculo viral, 100µL de matéria orgânica e 500µL de cada desinfetante diluído individualmente e a mesma foi incubada por 10 minutos. Após a etapa neutralizante, a suspensão foi filtrada em membrana 0,22µm e 0,2mL foi inoculado em seis ovos de galinha embrionados de nove dias de incubação, via rota alantóide. O fluido alantóide foi colhido após 72 horas para testes de hemaglutinação e RT-PCR. O mesmo protocolo experimental foi repetido usando as soluções desinfetantes 72 horas após a diluição. O vírus da influenza foi inativado pelo composto oxidante e também pelo desinfetante fenólico em todas as concentrações virais testadas 0-hora após diluição. O desinfetante com associação de amônia quaternária e glutaraldeído inativou o vírus na concentração de até 5,4log10 EID50/mL. O desinfetante à base de amônia quaternária não inativou o vírus. Os resultados 72-horas após a diluição não diferiram quando comparado com 0-hora, exceto o desinfetante fenólico, o qual inativou o vírus da influenza somente até a concentração 5,4log10 EID50/mL. Concluindo, três dos quatro desinfetantes testados foram efetivos ao inativar o vírus da influenza [H1N1] pandêmico na presença de matéria orgânica. Os resultados do teste com produtos diluídos após 72 horas sugerem redução da efetividade em, pelo menos, um desinfetante.


#8 - Prevalence and factors associated with Dirofilaria immitis infection in dogs in Sertão Paraibano, Northeast Brazil

Dirofilaria immitis is a heart and large vessel parasite that mainly affects domestic dogs and has shown a re-emerging zoonosis in recent years. The objective of this study was to determine the prevalence, associated factors, and geographic areas of D. immitis in dogs in the city of Sousa, Paraíba, Northeast Brazil. A total of 320 dogs were selected and evaluated, 160 domiciled and 160 wandering, from the 32 districts of the city. Clinical examination, blood collection, and epidemiological data retrieval were performed for each animal. The sanitation conditions of the environment were visually observed at the time of evaluation. Three methods were used to investigate the morphometric diagnosis of microfilariae: capillary blood smear (ESC), peripheral blood smear (ESP), and modified Knott test. The data were subjected to univariate and multivariate statistics for the observation of risk factors and qualitative assessment of the examinations. Of the 17.5% (56/320) of animals testing positive for D. immitis, 25% (40/160) were wandering and 10% (16/160) were domiciled dogs. Positive dogs were found in 24 of the 32 neighborhoods evaluated, with Angelim and Doutor Zezé having the highest percentages of 1.56% (5/320) each. Only the categories of cardiac alterations (OR 6.231 [1,539-25,236]) and stray dogs (OR 2.463 [1,281-4,735]) demonstrated potential risk factors for infection. Of the 56 positive animals, 28 were positive in the three tests, and another 28 showed variance between methods and/or between prepared slides. No other filaridae were observed. The city of Sousa is considered to have a significant prevalence of infection by D. immitis, and forms of control and prophylaxis are required to reduce the risks of transmission to animals, as well as to humans.

Dirofilaria immitis é um parasito de coração e de grandes vasos que acomete principalmente cães domésticos, também causador de uma zoonose reemergente nos últimos anos. O objetivo deste trabalho foi determinar a prevalência, fatores associados e áreas geográficas de D. immitis em cães na cidade de Sousa, Paraíba, Nordeste do Brasil. Para isso, foram selecionados e avaliados 320 cães, sendo 160 domiciliados e 160 errantes, provenientes dos 32 bairros da cidade. Foi realizado o exame clínico, coleta de sangue e dados epidemiológicos de cada animal. O ambiente foi observado visualmente quanto as condições de saneamento no momento de avaliação. Para pesquisa de microfilárias, foram realizados três métodos: esfregaço sanguíneo capilar (ESC), periférico (ESP) e teste de Knott-modificado, associado ao diagnóstico morfométrico das microfilárias. Os dados foram submetidos a estatísticas uni e multivariadas para observação de fatores de risco e avaliação qualitativa dos exames. Obteve-se prevalência de 17,5% (56/320) de animais positivos para D. immitis, sendo 25% (40/160) errantes e 10% (16/160) domiciliados. Dos 32 bairros avaliados, em 24 foram encontrados cães positivos, sendo Angelim e Doutor Zezé, os bairros com maior percentual 1,56% (5/320) cada. Somente as categorias de alterações cardíacas (OR 6,231 [1.539-25.236]) e cães errantes (OR 2,463 [1.281-4.735]) demonstraram potencial fator de risco para infecção. Dos 56 animais positivos, 28 apresentaram positividade nos três testes, e outros 28 demonstraram variância entre métodos e/ou entre lâminas confeccionadas. Não foram observados outros filarídeos. Considerou-se que a cidade de Sousa apresenta prevalência significativa de infecção por D. immitis, sendo necessário estabelecer formas de controle e profilaxia, para reduzir os riscos da transmissão para animais, como também para humanos.


#9 - Efficacy of antimicrobial therapy in association with vaccination on the bacteriological cure of subclinical mastitis caused by Staphylococcus aureus in lactating cows

Abstract in English:

The objective of this study was to evaluate the efficacy of the antibiotic therapy associated with vaccination on the microbiological cure rate of subclinical mastitis caused by Staphylococcus aureus in lactating dairy cows. A total of five herds, from which 72 cows (120 mammary quarters - MQ) were diagnosed with S. aureus subclinical mastitis, were included in this study. Cows were randomly allocated to one of three treatment groups: a) Control (no treatment); b) ATB (antibiotic therapy); and c) ATB+VAC (antibiotic therapy plus vaccination against S. aureus). Intramammary treatment consisted of twice-daily infusion of ampicillin 75mg + cloxacillin 200mg, for 5 days. Parenteral treatment was done by injection of a single dose (7.5mg/kg) of enrofloxacin, on the first day of the treatment protocol. Vaccinated cows received three doses of a commercial vaccine 14 days before treatment (d-14), on the first day of treatment protocol (d1), and 14 days after the treatment protocol (d+14). Non-treated cows had a lower cure rate (0.06) than cows treated with ATB (0.84) and ATB+VAC (0.85). No difference in cure rate was observed between cows treated with ATB and ATB+VAC. On the other hand, vaccinated cows had lower somatic cell count (SCC) after 28 days of the treatment protocols (4.76 log10) than non-treated cows (5.37 log10). In conclusion, treatment with intramammary ampicillin and cloxacillin, associated with intramuscular enrofloxacin presented a high cure rate for SCM caused by S. aureus during lactation. The use of vaccination against S. aureus in association with antibiotic therapy did not increase the cure rate of MQ during lactation, but it was effective in reducing the SCC when compared to non-treated MQ. Although to ensure that the decrease of the SCC in ATB+VAC group was associated with the vaccination, the study should have included an additional group of only vaccinated cows, without antimicrobial therapy, with was not done in the present study, and therefore is one of the limitations of the experimental protocol used.

O objetivo deste estudo foi avaliar a eficácia da antibioticoterapia associada à vacinação sobre a taxa de cura microbiológica de mastite subclínica causada por Staphylococcus aureus em vacas leiteiras em lactação. Foram selecionados 5 rebanhos, dos quais 72 vacas (120 quartos mamários, QM) foram diagnosticadas com mastite subclínica por S. aureus e foram alocadas aleatoriamente em um de três grupos de tratamento: a) Controle (sem tratamento); b) ATB (antibioticoterapia); e c) ATB+VAC (antibioticoterapia mais vacinação contra S. aureus). O tratamento intramamário consistiu em infusão de ampicilina 75 mg + cloxacilina 200 mg duas vezes ao dia, durante 5 dias. O tratamento parenteral foi feito por injeção de uma dose única (7,5 mg/kg) de enrofloxacina, no primeiro dia do protocolo de tratamento. As vacas vacinadas receberam três doses de uma vacina comercial, 14 dias antes do tratamento (d-14), no primeiro dia do protocolo de tratamento (d1) e 14 dias após o protocolo de tratamento (d+14). A taxa de cura das vacas não tratadas foi menor (0,06) do que das vacas tratadas com ATB (0,84) e ATB+VAC (0,85). Não foi observada diferença de taxa de cura entre vacas tratadas com ATB e ATB+VAC. Por outro lado, as vacas vacinadas apresentaram menor a contagem de células somáticas (CCS) após 28 dias de tratamento (4,76 log10) do que em vacas não tratadas (5,37 log10). Em conclusão, o tratamento com ampicilina e cloxacilina intramamária, associados à enrofloxacina intramuscular, apresentou alta taxa de cura para MSC causada por S. aureus durante a lactação. A utilização da vacinação contra S. aureus associada à antibioticoterapia não aumentou a taxa de cura dos QM durante a lactação, mas foi eficaz na redução do CCS quando comparada à QM não tratados. Entretanto, para ter certeza que a diminuição da CCS no grupo ATB+VAC estivesse associada à vacinação, o estudo deveria ter incluído um grupo adicional de apenas vacas vacinadas, sem terapia antimicrobiana, o que não foi feito no presente estudo e, portanto, é um das limitações do protocolo experimental utilizado.


#10 - Clinical and pathological aspects of an outbreak of Streptococcus suis serotype 9 infection in pigs

Abstract in English:

can cause severe systemic inflammatory disease in pigs, mainly during the nursery phase. Streptococcus suis is a reemergent pathogen, and outbreaks of its inducing disease represent significant economic losses for the pig industry worldwide. In this study, we described the clinical, pathological, and molecular aspects of an outbreak of S. suis infection with atypically high mortality. The outbreak occurred in nursery farms integrated into a cooperative in the state of Paraná, Brazil. Of the 30 nurseries, 10 were severely affected by the pathogen and had high economic losses. Clinical signs usually started approximately 10 days after weaning and were mainly characterized by acute nervous and locomotor disorders. The mortality of the affected batches usually ranged between 8% and 10%, but in some cases, it reached 18%. Nine piglets were submitted to post mortem examination. Macroscopically, the synovial joints were enlarged and contained fibrinous exudates. In the central nervous system, there was hyperemia of the leptomeningeal vessels associated with deposition of fibrin and purulent exudate in the leptomeninges. In three piglets, there was thickening of the choroid plexus associated with dilation of the lateral ventricles. Microscopic lesions were characterized mainly by fibrinosuppurative inflammation, which involved the synovial membranes, leptomeninges of the brain, and spinal cord. Furthermore, it also affects the choroid plexus, ependyma, nerve roots, and central canal of the spinal cord. S. suis was isolated from the cerebrospinal fluid, meningeal swabs, and/or synovial fluid of 8/9 piglets, and typified as serotype 9 by multiplex PCR.

Streptococcus suis é um patógeno Gram positivo que habita o trato respiratório superior e pode causar doença inflamatória sistêmica grave em suínos, principalmente durante a fase de creche. Streptococcus suis é um patógeno reemergente e surtos representam perdas econômicas significativas a suinocultura mundial. Neste estudo descrevemos os aspectos clínicos, patológicos e moleculares de um surto de infecção por S. suis com mortalidade atipicamente alta. O surto ocorreu em creches integradas a uma cooperativa do estado do Paraná, Brasil. Das 30 creches, 10 foram severamente afetadas pelo patógeno e apresentavam elevadas perdas econômicas. Os sinais clínicos iniciavam em torno de 10 dias após o desmame e eram caracterizados principalmente por sinais clínicos nervosos e locomotores agudos. A mortalidade dos lotes afetados variava entre 8% e 10%, mas em alguns casos ultrapassava 18%. Nove leitões foram submetidos ao exame post mortem. Macroscopicamente, as articulações sinoviais estavam aumentadas e continham exsudato fibrinoso. No sistema nervoso central havia hiperemia dos vasos leptomeníngeos associada a deposição de fibrina e exsudato purulento nas leptomeninges. Em três leitões havia espessamento do plexo coroide associado a dilatação dos ventrículos laterais. As lesões microscópicas eram caracterizadas principalmente por inflamação fibrinossupurativa que envolvia as membranas sinoviais, as leptomeninges do cérebro e medula espinhal. Além disso, também afetava o plexo coroide, epêndima, raízes nervosas e canal central da medula espinhal. S. suis foi isolado do líquido cefalorraquidiano, suabe de meninge e/ou líquido sinovial de 8/9 leitões e tipificado como sorotipo 9 por PCR multiplex.
