E-mail: faleirosufmg@gmail.com Leucocytes recruitment to tissues is an essential part of the innate immune response and an unregulated process can result in tissue damage. Thus, leucocytes infiltration has been implicated in the pathogenesis of acute laminitis. The objectives of this stud were to determine the effect of an antagonist for CXCR1/2, a chemokine receptor for neutrophils attraction on clinical signs and hematological parameters in horses given oligofructose to induce laminitis. Twelve horses were given oligofructose (10g/kg bw PO) in time 0 and divided into two groups: one treated (30mg/kg bw. ICXCR1/2 IV, times 6, 12, 18 e 24 h) and the other not treated. Cardiac and respiratory frequency, rectal temperature, mucous membrane colour, digital pulse, hoof sensitivity and Obel’s grade of lameness were recorded. Values for RBC, WBC and blood glucose, BUN, creatinin, ALT, AST, alkaline phosphatase, GGT, total bilirubin and serum protein concentrations were measured on times 0, 6, 12, 18, 24, 36, 48, 60 e 72 h. All the horses given oligofructose developed signs of endotoxemia like diarrhea, fever and leukocytosis and laminitis. Also, CXCR1/2 antagonist treatment did not cause any adverse effects. However, this substance when injected intravenously (30mg/kg) 6/6 hours in 4 applications, did not ameliorate clinical and hematological signs of endotoxemia.

E-mail: faleirosufmg@gmail.com O recrutamento de leucócitos aos tecidos é uma parte essencial da resposta imune inata e esse processo de forma desregulada pode resultar em lesões aos tecidos. Assim, a infiltração de leucócitos tem sido implicada na patogênese de laminite aguda em equinos. Os objetivos dessa pesquisa foram verificar a ação da ICXCR1/2 sobre os sinais clínicos e parâmetros hematológicos de cavalos com laminite induzida por oligofrutose. Doze equinos receberam oligofrutose (10g/kg de peso vivo PO) no tempo 0 e foram divididos em 2 grupos: tratados (30mg/kg p.v. ICXCR1/2 IV, nos tempos 6, 12, 18 e 24 h) e não tratados. As frequências cardíaca e respiratória, temperatura retal, coloração de membranas mucosas, presença e intensidade de pulso digital, sensibilidade ao exame com pinça de casco e grau de claudicação segundo Obel, bem como parâmetros hematológicos e bioquímicos (hemograma e as concentrações sanguíneas de glicose, uréia, creatinina, ALT, AST, FA, GGT, bilirrubina total e proteína total) foram aferidos nos tempos 0, 6, 12, 18, 24, 36, 48, 60 e 72 horas . O modelo usando oligofructose foi adequado para induzir sinais de laminite e de sinais de endotoxemia, como diarreia, febre e leucocitose em cavalos sem raça definida de origem nacional. Também, não foram observadas quaisquer reações adversas clínicas ou hematológicas relacionadas ao uso intravenoso do antagonista de CXCR1/2, contudo essa substância, quando administrada na dose de 30mg/kg de peso vivo, 4 vezes ao dia, por 4 aplicações, não foi capaz de prevenir os sinais clínicos e as alterações hematológicas causadas pela administração de oligofructose nos equinos deste estudo.

E-mail: aurea@dmv.ufrpe.br This study was undertaken with the aim to evaluate the p53 expression, applying the immunohistochemical technique to malignant primary mammary neoplasms histopathologically diagnosed in female dogs, and to investigate exon 8 of the Tp53 suppressor gene for mutation types by means of PCR-RFLP pattern of bands. Nineteen healthy mammary glands were used as a control group (group 1). Samples from 18 cases diagnosed with malignant mammary tumors (group 2), and the contralateral mammary glands (group 3) were collected during the UFRPE Veterinary Hospital routine. The tumors were diagnosed by histopathology and subdivided into grades of malignity. The streptavidin-biotin peroxidase method was used to analyze the immunohistochemical expression of p53, evaluated according to the location and intensity of stain. Expression of p53 protein was not observed in the samples of group 1. On the contrary, it was observed in all malignant tumors; the protein p53 was localized either only in the nucleus or in the nucleus and in the cytoplasm, in samples of group 2. In group 3, expression of p53 protein was observed in the tumors (2 samples) and in normal mammary tissues (4 samples). For the molecular analyses, genomic DNA was extracted and submitted to PCR-RFLP with the following endonuclease enzymes: AluI, BsoBI, DdeI and SmaI. The band pattern showed polymorphism between groups, but not between histological variants of tumors. This polymorphism detected mutations in the fragment studied - exon 8 of Tp53 - which could account for changes in nucleotides, located in the restriction sites of the endonuclease enzymes. In conclusion, the immunoexpression of p53 had no relationship with histological subtype or malignity grade, but it can be related to the presence of mammary tumors in female dogs. The PCR-RFLP technique can be an important tool for the study of mammary carcinogenesis in bitches because the polymorphism obtained may allow early diagnosis in tissues of mammary glands.

O método da estreptoavidina-biotina peroxidase foi utilizado para a análise da expressão de p53 por imuno-histoquímica, de acordo com a localização e intensidade da coloração. A expressão da proteína p53 não foi observada nas amostras do Grupo 1, mas foi encontrada em todas as amostras de tumores malignos (Grupo 2) seja só no núcleo, ou também no citoplasma. No Grupo 3, a expressão foi observada em quatro amostras normais e em duas que apresentavam tumor. Para a análise molecular, o DNA genômico foi extraído e submetido à PCR-RFLP com as seguintes endonucleases: AluI, BsoBI, DdeI e SmaI. O padrão de bandas foi polimórfico entre os grupos, mas não entre as variantes tumorais. Esse polimorfismo detectou mutações no fragmento estudado - éxon 8 do gene Tp53 - que podem resultar em alterações nos nucleotídeos, localizados nos sítios de restrição das enzimas. Esses achados levam a conclusão de que a imunoexpressão da p53 não tem relação com o subtipo histológico ou grau de malignidade do tumor, mas sim com a presença dos tumores no tecido mamário de cadelas. A PCR-RFLP pode ser usada como importante ferramenta para o estudo da carcinogênese mamária na cadela, possibilitando gerar diagnósticos precoces através do polimorfismo obtido com endonucleases de restrição pré-selecionadas.

E-mail: lopescwg@ufrrj.br Toxoplasmosis is one of the most common parasitic zoonoses throughout the world. Infection in man and animals varies in different geographical areas influenced by many environmental conditions. Seroprevalence of Toxoplasma gondii infection in cattle in Brazil ranges from 1.03 to 71%. A cross-sectional survey was carried out in 58 out of 453 farms in the South Fluminense Paraiba Valley, State of Rio de Janeiro, Brazil. Over 3-year-old cattle (n=589) from dairy herds were selected for blood collection and detection of anti-T. gondii antibodies by indirect immunofluorescence reaction (IFA) with initial titration of 1:16; titers ³ 64 were considered positive. Univariate analysis of risk factors showed that cats in contact with cattle, cats in contact with drinking water, and number of cats were associated with T. gondii seroprevalence. Logistic regression revealed a two-fold increased risk for infection of cattle (p=0.0138) through larger number of cats (>3) compared with low numbers of cats (1-2) on the farm. In contrast, the presence of chickens was considered a protective factor (p=0.025).

Infecções em seres humanos e em animais variam nas diferentes áreas geográficas influenciadas pelas condições ambientais. A soroprevalência da infecção por Toxoplasma gondii em bovinos no Brasil varia de 1,03 a 71,0%. O estudo transversal foi realizado em 58 de um total de 453 propriedades na região Sul Fluminense do estado do Rio de Janeiro. Vacas leiteiras acima de 3 anos de idade (n=589) foram selecionadas para coleta de sangue e a detecção de anticorpos anti-T. gondii foi feita pelo teste de imunofluorescência indireta (IFI) com titulação inicial de 1:16 e títulos ³ 64 foram considerados positivos. Após análise univariada dos fatores de risco, gatos em contato com bovinos, em contato com a água de beber dos animais e o número de gatos foram associados com a soroprevalência de T. gondii. A regressão logística demonstrou que o número maior de gatos (>3) teve um risco duas vezes maior (p=0,0138) que propriedades que tinham um número menor de gatos (1-2). Em contraste, a presença de galinhas foi considerada um fator de proteção (p=0,025).

Testes and epididymides from these animals were studied anatomopathologically. Samples from these tissues of Sheep 1, 4 and 5 were taken to polymerase chain reaction (PCR). Clinical parameters remained for the infected group above the values observed in the control group during the experimental period. Parasitaemia was observed on day 3 post-infection, and the highest values occurred between day 6 and 10, and day 15 and 18 post-infection. Sheep 1 and 4 showed severe anemia on day 25 post-infection. All sheep of the infected group showed flabby and palid testes. Histologically, moderate to severe testicular degeneration, multifocal epididymitis and hyperplasia of epididymal epithelium were observed. The result of T. vivax PCR analysis in the testes and epididymal tissues was positive in 100% of the samples of the experimentally infected sheep. Epididymal and testicular lesions associated with the presence of the parasite in these tissues, shown by PCR, suggest the participation of T. vivax in the pathophysiological mechanism of reproductive damage.

[Anatomical-surgical arterial segments of the kidney in agouti (Dasyprocta prymnolopha).] Segmentos anátomo-cirúrgicos arteriais do rim de cutia (Dasyprocta prymnolopha). Pesquisa Veterinária Brasileira 28(5):249-252. Departamento de Morfofisiologia Veterinária, Centro de Ciências Agrárias, Universidade Federal do Piauí, Teresina, PI 64049-550, Brazil. The renal arteries were injected with stained acetate vinyl, followed by procedures of acid corrosion in order to obtain vascular casts. It was found that the renal artery is always single and bifurcated into ventral and dorsal sectorial arteries. The sectorial arteries reached the kidneys (100% of the cases) through the hilus. These vessels gave origin to segmental branches responsible for kidney irrigation. At the right kidney, the ventral sectorial arteries gave origin to 3 (60% of the cases), 4 (35%) and 5 (5%) segmental branches; the dorsal sectorial arteries gave origin to 3 (30%), 4 (45%), 5 (20%) and 6 (5%) segmental arteries separated by a vascular sector. At the left kidney, the ventral sectorial arteries originated 2 (10%), 3 (55%) or 4 (35%) segmental branches; the dorsal sectorial arteries gave origin to 3 (25%), 4 (50%) and 5 (25%) segmental branches. Based on the arterial distribution of agouti kidneys, independent sections and arterial segments were found, so that it is possible to accomplish partial kidney resection surgery.

[Clinical, epidemiological and pathological signs of natural infection in cattle by Trypanosoma vivax in Paraíba, Brazil.] Aspectos clínicos, epidemiológicos e patológicos da infecção natural em bovinos por Trypanosoma vivax na Paraíba. Pesquisa Veterinária Brasileira 28(1):63-69. Departamento de Ciências Animais, Universidade Federal Rural do Semi-árido, BR 110 Km 47, Caixa Postal 147, Mossoró, RN 59625-900, Brazil. E-mail: jaelsbatista@hotmail.com Two outbreaks of trypanosomiasis by Trypanosoma vivax, reported in cattle, occurred on two farms located in the state of Paraíba, northeastern Brazil. The epidemiology, clinical signs and pathology of the disease were studied from May 2005 to November 2006. T. vivax was identified morphologically and by polymerase chain reaction test (PCR). The affected cattle presented anorexia, depression, fever, anemia, weight loss, reduction in milk production, transitory blindness, abortion and some nervous signs as nystagmus, tetany and bruxism. All cattle that presented nervous signs died. Necropsy findings were enlarged lymph and spleen, serous atrophy of the fat depots, proeminence of the splenic white pulp, hydropericardium and pericardial petechiae and ecchymoses on the epicardium. Histologically there was meningoencephalitis. The treatment of the clinical cases with diminazena aceturate was efficient with clearance of the parasite from the blood or disappearance of clinical signs in up to 2 months after the beginning of the outbreak on the two farms studied. The epidemiologic factors favoring the occurrence of the outbreak were the abundance of mechanical vectors, as tabanids and Stomoxys sp., and the introduction into the herd of cattle from farms where the disease occurred. It is suggested that the semiarid of the Brazilian Northeast is an enzootic instability region for trypanosomiasis due to its prolonged periods of drought and high temperatures, constituting during most part of the year an unfavorable environment for the development of vectors.

All cattle that presented nervous signs died. Necropsy findings were enlarged lymph and spleen, serous atrophy of the fat depots, proeminence of the splenic white pulp, hydropericardium and pericardial petechiae and ecchymoses on the epicardium. Histologically there was meningoencephalitis. The treatment of the clinical cases with diminazena aceturate was efficient with clearance of the parasite from the blood or disappearance of clinical signs in up to 2 months after the beginning of the outbreak on the two farms studied. The epidemiologic factors favoring the occurrence of the outbreak were the abundance of mechanical vectors, as tabanids and Stomoxys sp., and the introduction into the herd of cattle from farms where the disease occurred. It is suggested that the semiarid of the Brazilian Northeast is an enzootic instability region for trypanosomiasis due to its prolonged periods of drought and high temperatures, constituting during most part of the year an unfavorable environment for the development of vectors.

The brain of an one year old mal e calf which died with signs of neurological disease was submitted to the laboratory for rabies diagnosis. Microscopical findings included moderate mielitis, mild meningoencephalitis with perivascular cell cuffing and Negri inclusion bodies in Purkinje cells of the cerebellum. Rabies virus infection was further confirmed by the direct fluorescent antibody test as well as by mouse inoculation. ln addition, a herpesvirus was isolated from brain tissues. The isolate was antigenic and genetically characterized as bovine herpesvirus type 5 (BHV-5). It was not possible to determine whether BHV-5 played an active role in the outcome of the infection, since, the virus might have been present in a latent form in neural tissues. This is the first report of a mixed rabies/ BHV-5 infection in calves.

O encéfalo de um bezerro macho, de um ano de idade, que morreu com sinais de doença neurológica, foi submetido ao laboratório para diagnóstico de raiva. O exame histopatológico revelou mielite moderada, meningoencefalite leve com infiltração perivascular e corpúsculos de Negri nas células de Purkinje do cerebelo. A infecção pelo vírus rábico foi ainda confirmada por imunofluorescência diretá e por inoculação em camundongos. Além disso, um herpesvírus foi isolado dos tecidos do encéfalo. O vírus isolado foi antigênica e geneticamente caracterizado como Herpesvírus bovino tipo 5 (BHV-5). Não foi possível determinar se o BHV-5 teve algum papel ativo no desfecho da enfermidade, uma vez que o vírus poderia estar presente em forma latente nos tecidos neurais. Este é o primeiro relato de uma infecção mista pelo vírus da raiva e BHV-5 em bovinos.

E-mail: callado@altavista.net Small ruminant lentiviruses (SRLV), whose prototypes are Caprine Arthritis-Encephalitis virus (CAEV) and Maedi-Visna virus, are the causative agents of slow progressive degenerative diseases of goats and sheep (infected animals), responsible for significant economic losses. These viruses cause persistent infections with long periods of incubation and induce inflammatory and degenerative lesions. The lesions are induced in target organs of the host such as joints, CNS, lungs and mammary glands dueto viral replication in cells of the monocyte/macrophage lineage which is the main target cell. Infections occur particularly in the young and are acquired through ingestion of virus in milk or colostrum from infected does or ewes. The induction of immune response is variable and does not protect against the infection. Diagnosis is primarily based on the presence of SRLV antibodies usually detected by agar gel immunodiffusion (AGID) or enzyme linked immunosorbent assays (ELISA). As no vaccine is available, most often employed schemes to prevent spread of SRLV are based on segregation or/and culling of positive animals associated with management practices, especially the offspring. The strategies of SRLV for dealing with the immune system make difficult to accomplish diagnosis of infection, control or prevention of the viral spread. This review shows aspects of SRLV based on their phylogenetic studies of fields isolates, clinical, and immunopathological features.

Estes vírus causam infecções persistentes com período de incubação longo e causam inflamatórias e degenerativas. As lesões são induzidas em tecidos específicos do hospedeiro como articulações, pulmões, CNS e glândulas mamárias devido à replicação virai em células da linhagem monocítico-fagocitária que são as principais células-alvo. A infecção ocorre principalmente durante os primeiros meses de vida, através da ingestão de vírus no leite ou colostro de cabras ou ovelhas infectadas. A indução da resposta imunológica é variável e não protege contra a infecção. O diagnóstico é baseado primariamente na detecção de anticorpos para SRLV, geralmente por imunodifusão em gel de agar (AGID) e enzyme linked immunosorbent assay (ELISA). O diagnóstico e separação ou descarte dos animais soropositivos associado ao uso de certas práticas de manejo, especialmente das crias, são os principais meios implementados para prevenir a disseminação de SRLV, uma vez que ainda não existe vacina contra o vírus. As estratégias adotadas pelos SRLV para enfrentar o sistema imune dificultam o diagnóstico da infecção, controle ou prevenção da disseminação de SRLV. Esta revisão apresenta alguns aspectos das lentivíroses de pequenos ruminantes baseadas em estudos filogenéticos de amostras isoladas, aspectos clínicos e imunopatológicos.


#19 - A monoclonal blocking ELISA for the serological diagnosis of bovine herpesvirus type 1 (BHV-1) infections, 21(1):33-37

[A monoclonal blocking ELISA for the serological diagnosis of bovine herpesvirus type 1 (BHV-1) infections] ELISA de bloqueio monoclonal para o diagnóstico sorológico de infecções pelo herpesvírus bovino tipo 1 (BHV-1). Pesquisei Veterinária Brasileira 21(1):33-37. Centro de Pesquisá Veterinária Desidério Finamor (CPVDF), FEPAGRO, Cx. Postal 2076, Porto Alegre, RS 90001-970, Brazil. A monoclonal antibody-based blocking enzyme linked immunosorbent assay (ELISA-M) was developed and standardized for the detection of antibodies to irífectious bovine rhinotracheitis virus (Bovine Herpesvirus type 1; BHV-1). A total of 266 samples of bovine sera (148 negative and 118 positive) were tested and compared with the results of a standard sérum neutralization (SN) test. The ELISA-M was adjusted to 92.37% sensitivity, 92.56% especificity, 93.83% negative predictive value, 90.83% positive predictive value and to na accuracy of 92.48%, with an agreement index (k) equal to 0.85. The main advantages presented by the ELISA-M were its practicality and rapidity in performance. This test was shown to be a suitable alternative to SN tests in the detection of BHV-1 antibodies in cattle. However, the ELISA was unable to discriminate between BHV-1 and bovine herpesvirus type 5 (BHV-5) antibodies.

Foram utilizadas nesta avaliação 266 amostras de soros bovinos, sendo 148 negativos e 118 positivos em testes de soroneutralização (SN). Em comparação com este último, o ELISA-M demonstrou uma sensibilidade de 92,37%, especificidade de 92,56%, valor preditivo positivo de 90,83%, valor preditivo negativo de 93,83% e precisão de 92,48%. O índice de concordância (k) entre os testes foi de 0,85. O ELISA-M apresentou como vantagens a rapidez e a praticidade de execução. Com base nestes resultados, o ELISA-M foi considerado uma alternativa apropriada para o diagnóstico sorológico de infecções pelo BHV-1. Entretanto, o teste não foi capaz de diferenciar anticorpos induzidos por BHV-1 ou BHV-5.
