#21 - Stomoxys calcitrans outbreaks in pastured beef cattle in the state of Mato Grosso do Sul, Brazil, 30(11):945-952

E-mail: thadeu@cpap.embrapa.br The expansion of the alcohol industry has led to the installation of ethanol plants in areas traditionally occupied by beef cattle in the Brazilian Midwest. Stable fly (Stomoxys calcitrans) outbreaks associated with alcohol plants have been reported in Nelore cattle in Mato Grosso do Sul, Brazil, in the last two years. Visits to livestock ranches and alcohol plants were held in mid-November 2009 at the end of S. calcitrans outbreaks in the counties of Angélica and Ponta Porã, MS. Interviews, surveys and collections of immature stages of flies were conducted at the sites and the entomological material was taken to the laboratory for further emergency. High stable fly infestations and cattle bunching behavior were observed during visits. Stable fly breeding sites were found and emergence of adult flies occurred from material collected from both cattle ranches and alcohol plants. The set of information, on-site observation, and sampling results made possible a preliminary epidemiological approach on the dynamics of S. calcitrans outbreaks as well as a discussion of potential risk factors.

E-mail: thadeu@cpap.embrapa.br A expansão da indústria sucroalcooleira tem levado à instalação de usinas de álcool em áreas tradicionalmente ocupadas pela pecuária de corte na região Centro-Oeste do País. Surtos pela mosca-dos-estábulos (Stomoxys calcitrans) em bovinos Nelore têm sido relatados nos últimos dois anos em Mato Grosso do Sul, associados a estas usinas. Visitas em propriedades pecuárias e usinas foram realizadas em meados de novembro 2009, ao final de surtos por S. calcitrans ocorridos nos municípios de Angélica e Ponta Porã, MS. Entrevistas, observações e coletas de imaturos de dípteros foram realizadas nos locais e o material entomológico coletado foi levado ao laboratório para posterior emergência. Elevadas infestações pela mosca-dos-estábulos e comportamento de agrupamento dos bovinos foram observados. Sítios de reprodução foram encontrados nos locais e a emergência de S. calcitrans foi constatada nas amostras coletadas tanto nas fazendas como nas usinas. O conjunto de informações, observações in loco e resultados das amostragens possibilitaram realizar uma abordagem epidemiológica preliminar sobre a dinâmica dos referidos surtos por S. calcitrans e discutir potenciais fatores de risco.

E-mail: alschild@terra.com.br The study aimed to characterize morphological patterns of 59 liver samples of Senecio spp. poisoned cattle from 35 outbreaks, observed in southern Rio Grande do Sul, Brazil, from 2000 to 2009. The lesions were associated with epidemiological changes during these years. The climate changes concerning accumulated rain and mean temperature during the different seasons were analyzed. The macroscopic and histological lesions were classified into 6 different patterns. The macroscopic classification was made according to capsular pattern, hepatic cut surface discoloration, and the presence of nodules. The histological classification was based on the distribution of fibrosis, the amount of megalocytes in 10 high magnification fields, and on bile duct proliferation. Pattern 1 was characterized by a whitish liver, diffuse fibrosis, severe bile duct proliferation, and discrete megalocytosis; pattern 2 was characterized by nodules consisting of groups of hepatocytes surrounded by fibrosis, severe bile duct proliferation, and discrete to mild megalocytosis; pattern 3 was characterized by a macro-nodular aspect to the cut surface with hepatic lobules surrounded by a thin septa of fibrous tissue, severe bile duct proliferation, and mild megalocytosis; pattern 4 was characterized by a non-nodular surface with marble aspect, mild to severe bile duct proliferation, and megalocytosis; pattern 5 was characterized by a non-nodular surface and bridging or diffuse fibrosis, mild megalocytosis, and severe bile duct proliferation; and pattern 6 was characterized by a non-nodular surface, severe megalocytosis, discrete bile duct proliferation, and incipient fibrosis of the portal system, central vein or among hepatocyte cords. The results of macroscopic and histological liver analysis showed that patterns 1, 2 and 4 were the most frequently observed. The results of this study demonstrated that the macroscopic lesion observed in Senecio poisoned cattle is variable. Histologically this variation is related to the amount and distribution of fibrosis, megalocytosis and bile duct proliferation observed in each liver. Age of the cattle, evolution period of poisoning and clinical signs did not interfere on the pattern of lesions observed. On the other hand, climatic conditions probably had influence on increased disease prevalence due to major availability of Senecio spp. plants.

E-mail: alschild@terra.com.br O presente estudo teve como objetivo caracterizar os diferentes padrões morfológicos hepáticos em 59 fígados de bovinos intoxicados por Senecio spp. provenientes de 35 surtos observados na área de influência do Laboratório Regional de Diagnóstico da Faculdade de Veterinária da Universidade Federal de Pelotas (LRD) de 2000-2009, dos quais havia registro fotográfico das lesões macroscópicas, fazendo uma relação com as variações epidemiológicas observadas no período. Foram analisadas as variações climáticas referentes à precipitação pluviométrica e temperatura média nas diferentes estações do ano ocorridas neste intervalo de tempo. As alterações hepáticas macro e microscópicas foram agrupadas em padrões morfológicos semelhantes. Foi considerada como critérios de classificação macroscópica a coloração das superfícies capsular e de corte e a presença ou não de nódulos; e como critérios para a classificação histológica a quantidade e distribuição da fibrose, o número de megalócitos por campo de maior aumento, o grau estimado de proliferação de ductos biliares e a presença de hepatócitos vacuolizados. O Padrão 1 caracterizou-se por fígado esbranquiçado, fibrose difusa, acentuada proliferação de células de ductos biliares e megalocitose discreta; o Padrão 2 por presença de nódulos correspondentes a grupos de hepatócitos ou lóbulos hepáticos circundados por tecido conjuntivo fibroso, acentuada proliferação de células de ductos biliares e megalocitose de discreta a moderada; o Padrão 3 por superfície de corte com aspecto macronodular correspondente a lóbulos hepáticos circundados por delgado cordão de tecido conjuntivo, acentuada proliferação de ductos biliares e megalocitose moderada; o Padrão 4 por superfície sem nodulações e com aspecto marmorizado e histologicamente por fibrose em ponte, megalocitose e proliferação de células de ductos de moderadas a acentuadas; o Padrão 5 por superfície sem nodulações e microscopicamente por fibrose em ponte a difusa, megalocitose moderada e acentuada proliferação de células de ductos; e, o Padrão 6 por superfície sem nodulações e microscopicamente por megalocitose acentuada, proliferação de ductos discreta e fibrose em estágio inicial localizada nos espaços porta e veia centrolobular ou como colágeno imaturo entre os cordões de hepatócitos. Os resultados da análise macroscópica e histológica dos fígados dos bovinos demonstraram que os Padrões 1, 2 e 4 foram os mais frequentemente encontrados. Os resultados deste trabalho demonstram que a lesão hepática macroscópica que ocorre em bovinos intoxicados por Senecio spp. é variável e que histologicamente esta variação está relacionada à distribuição e a quantidade da fibrose, à quantidade de megalócitos e à proliferação biliar observadas em cada fígado. A idade dos animais, o tempo de evolução da intoxicação e os sinais clínicos não interferem nos padrões de lesões hepáticas encontradas. Por outro lado, as condições climáticas provavelmente tiveram influência no aumento da freqüência da doença devido ao aumento da disponibilidade da planta.

The symptomathology was characterized by bloody diarrhea with peracute course. Postmortem findings were pale organs and tissues, reticulate aspect of the ileum with its mucosa moderately thickened nad contained large amounts of partially coagulated blood. Histopathological examination revealed hyperplastic epithelial cell proliferation of the Lieberkühn crypts, sometimes associated with inflammatory and necrotic alterations. Immunohistochemistry demonstrated the presence of large amounts of bacteria in the cytoplasm of the cryptal epithelial cells. L. intracellularis was also seen by ultramicroscopic examination. These outbreaks occurred in 1987 and this is the first report of the disease in the state of Rio de Janeiro.

E-mail: claudioslbarros@uol.com.br Cases of trypanosomiasis by Trypanosoma evansi were diagnosed in horses in the state of Rio Grande do Sul, Brazil, between 2003 and 2004. In one stud farm (Farm A) with 125 horses, 52 died. Additionally, around 80 mares were sent to Farm A to be bred. Of those, 66 became ill and 56 died after being returned to their farms of origin. Twenty one horses clinically affected by the disease were observed. Clinical signs included loss of weight (despite voracious appetite), lethargy, incoordination and instability of hindlimbs, atrophy of the large muscles of the hindlimbs, muscle weakness and paleness of mucosae. Specimens of T. evansi were detected in the blood drawn from four affected horses. Normocytic normochromic anemia with PCVs ranging from 15 to 31%, leucocytosis due to lymphocytosis associated to large atypical lymphocytes was observed in several affected horses. High levels of antibodies against T. evansi were detected in the serum of six horses from Farm A. Eight horses presented encephalic neurological signs such as circling, ataxia, blindness, excitation, falls, listlessness, proprioception deficits and head tilt. One horse assumed a “dog-seating position”. Necropsy findings included muscle atrophy, enlargement and lymphoid hyperplasia of the spleen and lymphnodes, edema and softening of the white and grey matter of the brain. Histologically, an overwhelming necrotizing panencephalitis was observed in the seven horses with encephalic signs. This panencephalitis was characterized by marked edema, demyelination and necrosis and perivascular infiltrates of 6-10 layers of lymphocytes and plasm cells affecting both the white and gray matter. Several plasm cells in the inflammatory infiltrate contained numerous eosinophilic globules in their cytoplasm (Mott cells). Similar histological lesions were observed in the spinal cord of the horse with the “dog-seating position”. The brains of five horses with the encephalic signs were submitted to immunohistochemistry stain by the streptavidin-biotin technique. In all of those five brains moderate to abundant specimens of T. evansi in the perivascular spaces and neuropile were marked by the specific antibody. Epidemiological, clinical, hematological, and pathological aspects of equine trypanosomiasis caused by T. evansi are discussed.

[Botulism outbreaks in cattle in Brazil associated with contaminated water] Surtos de botulismo em bovinos no Brasil associados à ingestão de água contaminada. Pesquisa Veterinária Brasileira 21(2):43-48. Depto Apoio, Produção e Saúde Animal, Unesp-Campus de Araçatuba, Cx. Postai 533, Araçatuba, SP 16050-680, Brazil. Botulism in cattle occurs by ingestion of botulinum toxin C and/or D. Seven outbreaks of the poisoning in the states of Mato Grosso do Sul and São Paulo are reported. The clinical and pathological data, epidemiology and laboratory findings indicate a possible intake of the toxin through contaminated water. The average mortality rate was 20.1%, with 99.2% lethality and 31.62% morbidity. From about 9,000 cattle envolved in the outbreaks, 2,844 animals died, predominantly with a byperacute and acute clinical picture. The high morbidity rates were observed within a short period and affected all categories of cattle, with a clinical and pathological picture characterized by paresis and paralysis of the muscles of locomotion, swallowing and mastigation, with abscence of gross lesions at post-mortem examination. The outbreaks were related to the presence of decomposed animal carcasses or vegetal material in the drinking water. Botulinum toxins C and/or D were detected in water samples, víscera and blood serum of a considerable number of materials examined.

Pesquisa Veterinária Brasileira 21(2):43-48. Depto Apoio, Produção e Saúde Animal, Unesp-Campus de Araçatuba, Cx. Postai 533, Araçatuba, SP 16050-680, Brazil. Botulismo em bovinos resulta da ingestão de toxina previamente formada. No presente trabalho são descritos sete surtos da intoxicação onde os dados clínico-patológicos, epidemiológicos e os achados laboratoriais indicaram a possível ingestão da toxina através da água contaminada. O coeficiente médio de mortalidade foi de 20, 1%, com letalidade de 99,92%, e morbidade de 31,62%. Dos cerca de 9.000 bovinos envolvidos nos surtos, 2.844 morreram com quadro clínico predominantemente superagudo e agudo. A alta morbidade e mortalidade foram registradas num curto período de tempo e envolveram todas as categorias animais, com quadro clínico-patológico caracterizado por paresia e paralisia da musculatura da locomoção, deglutição e mastigação e ausência de lesões macroscópicas. As circunstâncias em que ocorreram os surtos estiveram relacionadas com a existência de carcaças de animais decompostas ou matéria orgânica vegetal na água de dessedentação. Foram detectadas toxinas botulínicas C e/ou D nas coleções de água, nas vísceras e no soro sangüíneo de parte considerável dos materiais examinados.


#26 - Outbreaks of periodontitis in calves ("Cara inchada") related to soil cultivation

Abstract in English:

"Cara inchada" of cattle (CI) is a periodontitis which occurs in young animals during tooth eruption and is related to tree clearance atid sowing pasture grasses in certain areas of Central Brazil once covered by forest or savanna vegetation. The findings of outbreaks of CI on two farms in the State of São Paulo are reported. In these outbreaks the calves showed severe emaciation, diarrhea, various periodontal lesions, halitosis and loss of premolar teeth of the upper jaw. On one farm the disease occurred after feeding sugar cane harvested from a newly cultivated area, in which the prevalence of periodontal lesions was 61.5% and mortality 22.1%. On the other farm, the outbreak occurred after cattle grazed on recently improved pastures. In the dry season the animals were also fed supplements of forage and silage grown on the sarne farm. The prevalence of CI in the latter was 51.9% and mortality 3.6%. It is concluded that there is a relationship between new outbreaks of periodontitis and soil and pasture improvement, i.e. ploughing, harrowing, application of lime and fertilizers.

"Cara inchada" dos bovinos (CI) é uma periodontite que os acomete na fase de dentição e está associada à formação de pastagens em determinadas áreas antes cobertas de mata ou com vegetação de cerrado no Brasil Central. Foram registrados dois surtos da doença em propriedades situadas no Estado de São Paulo, após a reforma de áreas anteriormente positivas para CI, e utilização das forrageiras cultivadas na alimentação animal. Nas duas propriedades surtos da doença haviam sido observados há mais de 10 anos atrás, após a formação inicial das pastagens em área de cerrado; no entanto, nos últimos anos não havia mais registros da ocorrência da periodontite. No ressurgimento da doença os bezerros apresentaram emagrecimento acentuado, diarréia, lesões peridentárias de diferentes intensidades, odor fétido bucal e perda dos dentes premolares decíduos maxilares. Numa das propriedades a doença ocorreu após a utilização de cana-de-açúcar, cultivada no ano anterior, na alimentação dos bezerros. A prevalência de animais com lesões peridentárias foi de 61,5%, com coeficiente de mortalidade de 22,1%. Na outra propriedade o surto ocorreu após a reforma dos pastos e das áreas destinadas à capineira e silagem de milho. A prevalência da doença foi de 51,9%, com mortalidade de 3,6%. Numa análise retrospectiva pode-se associar o ressurgimento da periodontite em bezerros nas duas propriedades com a reforma das áreas através de aragem, gradeação, calagem e adubação do solo.


#27 - Occurrence of outbreaks of Senecio spp. (Compositae) poisoning in cattle in Rio Grande do Sul, Brazil

Abstract in English:

Three outbreaks of Senecio spp. poisoning in cattle in Rio Grande do Sul, Brazil, are described. At least 13 bovines died due to the intoxication in the counties of Santo Augusto, São Sepé and Júlio de Castilhos. Clinical signs included depression, abdominal straining, abdominal pain, loss of appetite and nervous signs such as incoordination of gait, circling and agressivity. At necropsy the most consistent lesions were edema of the body cavities, mesentery, abomasal folds, gall bladder wall and subcutaneous tissue of the ventral parts of the body, fibrous and hard livers and firm and white spotted kidneys. Microscopic lesions included different degrees of fibrous proliferation and megalocytosis in the tiver and kidney with ductal hyperplasia in the former a protein loosing glomerulopathy in the latter. Vacuolar and hyalin degeneration were seen in the myocardial fibers and spongy degeneration in the white matter of the brain. Associated with the pastures where the affected animals had been grazing, abundant quantities of two species of Senecio were found, namely S. brasiliensis and S. selloi. No sheep were grazing at the sarne pastures where cattle tosses occurred.

São descritos três surtos de intoxicação por Senecio spp. em bovinos no Rio Grande do Sul. Os surtos ocorreram nos municípios de Santo Augusto, São Sepé e Júlio de Castilhos, causando pelo menos 13 mortes. Sinais clínicos dos animais afetados incluíam depressão, tenesmo, dor abdominal, anorexia e sinais nervosos como incoordenação motora, andar· em círculos e agressividade. As lesões mais consistentemente vistas na necropsia foram edemas cavitários, do mesentério, das pregas do abomaso, da parede da vesícula biliar, do tecido subcutâneo das partes ventrais do organismo e endurecimento do fígado e rim por proliferação fibrosa. O rim mostrava ainda manchas brancas no córtex. As lesões microscópicas consistiam de diferentes graus de proliferação fibrosa e megalocitose no fígado e rim, com hiperplasia ductal no primeiro e glomerulopatia com perda de proteína neste último. Degeneração vacuolar e hialina de fibras cardíacas e degeneração esponjosa da substância branca do encéfalo foram também observadas. As pastagens, onde os animais afetados tinham estado, encontravam-se intensamente infestadas por duas espécies de Senecio, S. brasiliensis e S. selloi. Nessas pastagens não havia ovinos.


#28 - Isolation and identification of Aujeszky's disease virus from outbreaks in swine in the State of Santa Catarina

Abstract in English:

From April 1983, through December 1985, 18 isolations of Aujeszky's disease virus (ADV) were made-in the State of Santa Catarina, Brazil, from the brains of piglets (17 samples) anda dog, all of which had shown nervous symptoms. All viruses were isolated either in primary cultures of chicken embryo fibroblasts or in a swine kidney cell line. Identification of the. isolated agents was made both by direct immunofluorescence (IF) on coverslips and by virus neutralization (VN) in microplates, using reference serum specific for ADV. All 18 isolates induced a cytopathic effect (CPE) typical of herpesvirus, first noticeable 24 hours after inocula tion and characterized by the rounding up and high refractiveness of affected cells. Direct IF demonstrated intense fluorescence of the entire cell monolayer in cultures inoculated with 10% and 1 % brain suspensions, while 0.1 % suspensions allowed the formation and visualization of bright fluorescent plaques amidst dark areas of uninfected cells. The presence of ADV was also confirmed in the VN test using the third passage of each of the 18 isolates. Characteristic herpesvirus CPE was observed in cultures inoculated with mixtures of the isolates and negative reference serum, while no CPE was seen in cultures inoculated with mixtures of the isolates and positive reference serum. Extracts prepared from the cell fraction and from the fluids of infected cultures contained two antigens· that were demonstrable in the immunodifusion test. Similar extracts prepared from uninfected cultures did not contain these antigens.

De abril de 1983 a dezembro de 1985, 18 amostras do vírus da doença de Aujeszky (VDA) foram isoladas do cérebro de leitões (17 amostras) e de um cão que apresentavam sintomatologia nervosa. Os vírus foram isolados em culturas primárias de fibroblastos de embrião de galinha ou em uma linhagem celular contínua derivada de rim de suíno. A identificação dos agentes isolados foi realizada utilizando-se os testes de imunofluorescência (IF) direta em lamínulas e de vírus neutralização (VN) em microplacas com soros de referência específicos para o VDA. As 18 amostras induziram efeito citopático típico de vírus herpes observado a partir de 24 horas após a inoculação e caracterizado pelo arredondamento e alta refratividade das células afetadas. O teste de IF direta, mostrou fluorescência intensa das monocamadas inoculadas com suspensões cerebrais de 10% e 1 %, enquanto que, as suspensões a 0,1 % permitiram a formação e visualização de placas fluorescentes rodeadas de áreas opacas de células não infectadas. A presença do VDA foi também confirmada no teste VN realizado com a terceira passagem das 18 amostras isoladas. Efeito citopático característico de vírus herpes foi observado nas culturas inoculadas com mistura das amostras virais e soro de referência negativo, enquanto que, as culturas inoculadas com mistura das mesmas amostras virais e soro de referência positivo não desenvolveram efeito citopático. Extratos preparados tanto da fração celular como dos fluídos das culturas infectadas continham dois antígenos, os quais foram evidenciados no teste de imunodifusão. Extratos preparados de culturas não infectadas não continham esses antígenos.
