E-mail: eduardofurtadoflores@gmail.com Bovine herpesvirus 5 (BoHV-5) is an important pathogen of cattle in South America and efforts have been made to produce safer and more effective vaccines. In addition to afford protection, herpesvirus vaccines should allow serological differentiation of vaccinated from naturally, latently infected animals. We previously reported the construction and characterization in vitro of a double mutant BoHV-5 (BoHV-5gE/TK”D) lacking the genes encoding thymidine kinase (tk) for attenuation, and glycoprotein E (gE) as the antigenic marker, as a vaccine candidate strain (Brum et al. 2010a). The present article reports an investigation on the attenuation and immunogenicity of this recombinant in calves. In a first experiment, 80 to 90-day-old seronegative calves (n=6) inoculated intranasally with the recombinant (titer of 107.5TCID50) shed virus in low to moderate titers in nasal secretions for up to 6 days, yet did not develop any respiratory, systemic or neurological signs of infection. At day 30 post-infection (pi) all calves had BoHV-5 specific neutralizing (VN) antibodies in titers of 4 to 8 and were negative for anti-gE antibodies in a commercial ELISA test. Administration of dexamethasone (0.1mg/kg/day during 5 days) to four of these calves at day 42 pi did not result in virus shedding or increase in VN titers, indicating lack of viral reactivation. Secondly, a group of 8-month-old calves (n=9) vaccinated intramuscularly (IM) with the recombinant virus (107.5TCID50/animal) did not shed virus in nasal secretions, remained healthy and developed VN titers from 2 to 8 at day 42 post-vaccination (pv), remaining negative for gE antibodies. Lastly, 21 calves (around 10 months old) maintained under field conditions were vaccinated IM with the recombinant virus (titer of 107.3TCID50). All vaccinated animals developed VN titers from 2 to 16 at day 30 pv. A boost vaccination performed at day 240 pv resulted in a rapid and strong anamnestic antibody response, with VN titers reaching from 16 to 256 at day 14 post-booster. Again, serum samples remained negative for gE antibodies. Selected serum samples from vaccinated animals showed a broad VN activity against nine BoHV-5 and eight BoHV-1 field isolates. These results show that the recombinant virus is attenuated, immunogenic for calves and induces an antibody response differentiable from that induced by natural infection. Thus, the recombinant BoHV-5gE/TKD is an adequate candidate strain for a modified live vaccine.

Além de conferirem proteção, vacinas contra os herpesvírus animais devem permitir a diferenciação sorológica entre animais vacinados e infectados naturalmente. Foi recentemente relatada a construção e caracterização in vitro de um recombinante do BoHV-5 defectivo na enzima timidina quinase (tk) para atenuação e, na glicoproteína E (gE) como marcador antigênico, como potencial cepa vacinal (Brum et al. 2010a). O presente artigo relata uma investigação sobre da atenuação e imunogenicidade do recombinante BoHV-5gE/TKD em bezerros. Em um primeiro experimento, bezerros soronegativos, com 80 a 90 dias de idade (n=6), inoculados pela via intranasal (IN) com o vírus recombinante (título de 107,5TCID50) excretaram baixos títulos de vírus nas secreções nasais por até seis dias, mas não desenvolveram sinais sistêmicos, respiratórios ou neurológicos. No dia 30 pós-infecção (pi), todos os animais possuíam anticorpos neutralizantes contra o BoHV-5, em títulos entre 4 e 8, mas permaneceram negativos para anticorpos contra a gE. Administração de dexametasona (Dx) a quatro desses bezerros no dia 42 pi (0.1mg/kg/dia durante 5 dias) não resultou em excreção viral ou em aumento dos títulos de anticorpos, indicando ausência de reativação viral. Em um segundo experimento, vacinação intramuscular (IM) de bezerros com 8 meses de idade (n=9) com o recombinante (107,5TCID50/animal) não resultou em excreção viral ou em manifestações clínicas. Os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 8 no dia 42 pós-vacinação (PV) e permaneceram negativos para anticorpos anti-gE. Finalmente, 21 bezerros (aproximadamente 10 meses de idade) mantidos a campo foram vacinados com o recombinante (107,3TCID50) pela via IM. Todos os animais vacinados desenvolveram anticorpos neutralizantes em títulos de 2 a 16 no dia 30pv. Revacinação desses animais no dia 240 pv provocou uma resposta anamnéstica rápida e intensa, resultando em títulos neutralizantes entre 16 e 256 no dia 14 pós-revacinação. O soro de todos os animais permaneceu negativos para anticorpos contra a gE. Amostras de soro dos animais vacinados apresentaram atividade neutralizante cruzada frente a nove isolados de BoHV-5 e oito de BoHV-1. Esses resultados demonstram que o recombinante é atenuado e imunogênico para bezerros e induz uma resposta sorológica diferenciável da resposta à infecção natural. Portanto, o recombinante se constitui em um potencial candidato a cepa vacinal.

E-mail: isdutra@fmva.unesp.br The serological response of beef calves was evaluated with different vaccination regimens against blackleg, using an official strain (MT) and a field-collected strain of Clostridium chauvoei as antigens. Sixty calves were randomly allocated to four different groups and were submitted to distinct vaccination protocols with a commercial polyvalent vaccine. Group G1 was first vaccinated at four months of age and a booster shot was given after weaning, at eight months. Group G2 was given the first dose at eight months and a booster shot 30 days later. Group G3 was vaccinated only once at eight months and the control group was not vaccinated. These alternative vaccination regimens were proposed in an effort to adequately protect cattle under open-field farming conditions. Serological evaluations were made by Elisa at 4, 8, 9 and 10 months of age. Both groups receiving booster shots had a significantly increased serological response 30 days later. However, the serum IgG levels against C. chauvoei were significantly higher in the calves that were first vaccinated at four months. At 10 months, the two booster shot groups (G1 and G2) had similar serological responses, while the calves that were treated with a single dose of vaccine at weaning (G3) had a response that was similar to that of the control group. The serological response of the calves was significantly inferior at several of the evaluation times when the field strain of the bacteria was used as a challenge antigen instead of the official MT strain. The serological response of calves that are vaccinated twice was found to be satisfactory, independent of the first injection being made at four or eight months of age. It was also concluded that it would be useful to include local bacterial strains in commercial vaccine production.

E-mail: isdutra@fmva.unesp.br Foi avaliada a resposta sorológica de bezerros de corte submetidos a diferentes esquemas de vacinação contra o carbúnculo sintomático, empregando-se como antígenos duas cepas distintas de Clostridium chauvoei: uma oficial (MT) e a outra uma cepa de campo. Os animais (n=60) foram randomizados em quatro grupos (G1, G2, G3 e Controle) e submetidos a três protocolos distintos de vacinação com um produto comercial polivalente. O G1 foi primovacinado aos 4 meses de idade e recebeu o reforço após desmama (8 meses de idade). O G2 recebeu a primeira dose aos 8 meses de idade e reforço 30 dias após. O G3 foi vacinado somente aos 8 meses de idade e o Controle não foi vacinado. As avaliações sorológicas pelo ensaio imunoenzimático (Elisa) foram realizadas aos 4, 8, 9 e 10 meses de idade dos bezerros. Nos dois esquemas em que os animais receberam o booster (G1 e G2), houve um aumento significativo (p<0,05) da resposta sorológica quando foram avaliados 30 dias após. No entanto, os valores séricos de IgG contra C. chauvoei foram significativamente maiores nos animais primovacinados aos 4 meses de idade. Aos 10 meses, os dois grupos que receberam o reforço vacinal (G1 e G2) não diferiram entre si na resposta sorológica e os bezerros que receberam uma única dose de vacina na desmama (G3) não diferiu do Controle. A resposta sorológica dos bezerros foi significativamente inferior (p<0,05) em diversos momentos da avaliação, quando a cepa de campo foi empregada como antígeno e quando comparada à da cepa MT. Pode-se deduzir que a resposta sorológica dos bezerros vacinados aos 4 e 8 meses de idade foi satisfatória e que existiram diferenças significativas nos valores séricos de anticorpos contra C. chauvoei quando na avaliação foi empregada cepa de campo.


#43 - Canine distemper virus detection in asymptomatic and non vaccinated dogs, 30(2):139-144

E-mail: helendelpuerto@hotmail.com A quantitative real time polymerase chain reaction (PCR) revealed canine distemper virus presence in peripheral blood samples from asymptomatic and non vaccinated dogs. Samples from eleven domestic dogs with no signs of canine distemper and not vaccinated at the month of collection were used. Canine distemper virus vaccine samples in VERO cells were used as positive controls. RNA was isolated with Trizol®, and treated with a TURBO DNA-free kit. Primers were designed for canine distemper virus nucleocapsid protein coding region fragment amplification (84 bp). Canine b-actin (93 bp) was utilized as the endogenous control for normalization. Quantitative results of real time PCR generated by ABI Prism 7000 SDS Software showed that 54.5% of dogs with asymptomatic canine distemper were positive for canine distemper virus. Dissociation curves confirmed the specificity of the real time PCR fragments. This technique could detect even a few copies of viral RNA and identificate subclinically infected dogs providing accurate diagnosis of this disease at an early stage.

E-mail: helendelpuerto@hotmail.com A reação em cadeia da polimerase (PCR) em tempo real revelou a presença do vírus da cinomose canina em amostra de sangue de cães assintomáticos e não vacinados. Amostra de onze cães domésticos sem nenhum sinal clínico de cinomose e que não foram vacinados no mês da coleta de sangue foram utilizados para análise. Amostra vacinal do vírus da cinomose canina em células VERO foi utilizada como controle positivo. O RNA total foi isolado utilizando-se Trizol®, e tratadas com o Kit TURBO DNA-free. Os iniciadores foram desenhados para amplificar a região do nucleocapsídeo viral com 319pb e 84pb para a PCR convencional e PCR em tempo real, respectivamente. O fragmento alvo da b-actina canina com 93pb foi utilizado como controle endógeno e normalizador. Resultados quantitativos da PCR em tempo real gerados pelo programa ABI Prism 7000 SDS demonstraram que 54,5% dos cães assintomáticos foram positivos para o vírus da cinomose canina. As curvas de dissociação confirmaram a especificidade dos fragmentos da PCR em tempo real. A detecção precoce do RNA viral é importante para a identificação de cães subclinicamente infectados e limitar a difusão da doença.

E-mail: bcs85@hotmail.com The efficacy of a polyvalent bacterin vaccine against Aeromonas hydrophila, Pseudomonas aeroginosa and Enterococcus durans administered by different routes in Nile tilapia was assessed by analyzing hematological and immunological parameters 7 and 21 days after vaccination. Treatments consisted of: non-vaccinated tilapia; tilapia vaccinated by intraperitoneal injection with 2x108 formalin-inactivated bacteria·mL-1; tilapia vaccinated orally with 2x107 formalin-inactivated bacteria·g-1, feed for 5 days; tilapia vaccinated by immersion bath in 2x107 formalin-inactivated bacteria·mL-1, for 20 minutes. Vaccinated fish groups presented higher hematocrit, number of erythrocytes and leukocytes than the non-vaccinated group. Serum agglutination titer of intraperitoneally vaccinated fish was higher on both evaluation periods for the three bacteria strains. Only on day 21 post-vaccination fish from the oral and immersion vaccination groups presented higher serum agglutination titer than the non-vaccinated fish for A. hidrophyla and E. durans. Serum antimicrobial activity in vaccinated fish was higher for P. aeroginosa and E. coli than in non-vaccinated fish on both evaluation periods. The different vaccine administration routes stimulated hematological and immunological responses in Nile tilapia 21 days post-vaccination, but intraperitoneal vaccination presented higher total number of leukocytes, lymphocytes and serum agglutination titer.

Os tratamentos consistiram de: tilápias não vacinadas; tilápias vacinadas via injeção intraperitoneal (i.p.) contendo 2x108 bactérias inativadas·mL-1; tilápias alimentadas com ração contendo vacina na proporção de 2x107 bactérias inativadas·g-1, durante 5 dias; tilápias vacinadas por banho de imersão em 2x107 bactérias inativadas·mL-1, durante 20 minutos. Os peixes vacinados apresentaram maior porcentagem de hematócrito, número de eritrócitos e leucócitos durante o período do experimento, em relação aos não vacinados. O título de aglutinação do soro dos peixes vacinados intraperitonealmente foi superior nos dois períodos de avaliação para todas as bactérias. Apenas no 21º dia os peixes vacinados oralmente e por imersão apresentaram título de aglutinação superior aos peixes não vacinados para A. hidrophyla e E. durans. O soro dos peixes vacinados apresentou maior atividade antimicrobiana para P. aeroginosa e E. coli, do que os peixes não vacinados, nos dois períodos. As diferentes vias de administração da vacina estimularam a resposta hematológica e imunológica da tilápia do Nilo após 21 dias de vacinadas, sendo que o número total de leucócitos, linfócitos, título aglutinante do soro dos vacinados i.p. foi maior do que os demais.

The vaccine titers were monitored using the microscopic agglutination test (MAT) for Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, Pomona and Wolffi serovars. All vaccines used were capable to product agglutinins for the Hardjo and Wolffi serovars observed at 3 days after vaccination, remaining until the 150th day; those serovars induced the highest titres of agglutinins. Vaccine D, in spite of not containing the Wolffi serovar, induced the production of agglutinins to this serovar. Agglutinins to the Canicola serovar were only observed in the animals vaccinated with the D bacterine. Vaccine D induced the highest average titers of antibodies to all tested serovars.

E-mail: flores@ccr.ufsm.br This paper reports the antibody response and fetal protection in pregnant cows conferred by an experimental vaccine containing two attenuated strains of bovine viral diarrhea virus (BVDV-1 and BVDV-2). Cows (n=19) were vaccinated twice, with a 34-days-interval, with the experimental vaccine and together with non-vaccinated controls (n=18), were mated and challenged between days 60 and 90 of gestation by intranasal inoculation of four heterologous BVDV-1 and BVDV-2 isolates. The antibody response was evaluated by serum-neutralization tests performed at different intervals after vaccination (days 34, 78 and 138 post-vaccination [pv]). Fetal protection was monitored by ultrassonographic and clinical examination of the dams and fetuses during the rest of gestation; and through virological and serological examination of pre-colostral blood obtained from aborted and/or recently born fetuses/calves. At the day of challenge (day 138 pv), all vaccinated cows had neutralizing antibodies in high titers against BVDV-1 (1,280- ³10,240), and with one exception (titer 20), presented moderate to high titers to BVDV-2 (80-1,280). At the end of the monitoring, only three out of 18 control cows (16.6%) delivered healthy, virus-free calves. Fifteen non-vaccinated cows (83.3%) presented signs of fetal infection and/or had reproductive losses. Seven of these cows (38.8%) delivered virus-positive calves; five were healthy and survived (27.7%); two were premature or weak and lasted three and 15 days, respectively. The other eight cows (44.4%) aborted between day 30 post-challenge and the parturition; or delivered premature or stillbirth calves. In contrast, 17 out of 19 (89.4%) vaccinated cows delivery virus-free, healthy calves. One vaccinated cow aborted around day 130 post-challenge, yet this fetus could not be examined for the presence of virus. Another cow delivered a virus-positive calf (5.2%). In summary, the experimental vaccine induced adequate antibody titers in most animals and the immunological response induced by vaccination was able to prevent fetal infection and reproductive losses upon challenge with a pool of heterologous BVDV isolates. Hence, this experimental vaccine may be an attractive alternative for the prevention of reproductive losses associated with BVDV infection.

Cows (n=19) were vaccinated twice, with a 34-days-interval, with the experimental vaccine and together with non-vaccinated controls (n=18), were mated and challenged between days 60 and 90 of gestation by intranasal inoculation of four heterologous BVDV-1 and BVDV-2 isolates. The antibody response was evaluated by serum-neutralization tests performed at different intervals after vaccination (days 34, 78 and 138 post-vaccination [pv]). Fetal protection was monitored by ultrassonographic and clinical examination of the dams and fetuses during the rest of gestation; and through virological and serological examination of pre-colostral blood obtained from aborted and/or recently born fetuses/calves. At the day of challenge (day 138 pv), all vaccinated cows had neutralizing antibodies in high titers against BVDV-1 (1,280- ³10,240), and with one exception (titer 20), presented moderate to high titers to BVDV-2 (80-1,280). At the end of the monitoring, only three out of 18 control cows (16.6%) delivered healthy, virus-free calves. Fifteen non-vaccinated cows (83.3%) presented signs of fetal infection and/or had reproductive losses. In vitro characterization of biologically cloned viruses obtained at passages 0, 1, 10, 20 and 30 demonstrated that the attenuation process did not significantly affect the phenotypic and antigenic properties of the viruses. No major differences in plaque size and morphology and in the growth kinetics in tissue culture were observed among the viruses obtained at different passages. Likewise, the antigenic profile of these viruses did not change upon successive passages in tissue culture, as ascertained by the pattern of binding by 48 monoclonal antibodies (mAbs). Intramuscular inoculation of both viruses (IBSP-2: 107.3 TCID50; SV-253: 106.8 TCID50) at passage 30 (p30) in twelve 15 months old heifers did not produce clinical signs, demonstrating the attenuation of the viruses. Following inoculation, infectious virus was detected in leucocytes of most inoculated animals (10/12) between days 3 and 6 post-inoculation (pi) and in nasal secretions of three animals (days 4, 7 and 8pi). However, the vaccine viruses were not transmitted to three seronegative calves maintained as sentinels. All vaccinated calves seroconverted at day 14 post-vaccination. A moderate to high serum neutralizing response against five Brazilian BVDV-1 (titers from 80 to ³1,280) and four Brazilian BVDV-2 isolates (titers from 20 to 640) was observed at day 33 post-vaccination (pv). In general, the highest titers were observed against the Brazilian BVDV-1 isolates. At day 240 post-vaccination, the animals received a booster administration (IBSP-2: 107.3 TCID50 and SV-253: 106.8 TCID50). E-mail: proehe@ufrgs.br The authors previously reported the construction of a glycoprotein E-deleted (gE·) mutante of bovine herpesvirus type 1.2a (BHV-1.2a). This mutant, 265gE·, was designed as a vacinal strain for differential vaccines, allowing the distinction between vaccinated and naturally infected cattle. In order to determine the safety and efficacy of this candidate vaccine virus, a group of calves was inoculated with 265gE·. The virus was detected in secretions of inoculated calves to lower titres and for a shorter period than the parental virus inoculated in control calves. Twenty one days after inoculation, the calves were challenged with the wild type parental virus. Only mild signs of infection were detected on vaccinated calves, whereas nonvaccinated controls displayed intense rhinotracheitis and shed virus for longer and to higher titres than vaccinated calves. Six months after vaccination, both vaccinated and control groups were subjected to reactivation of potentially latent virus. The mutant 265gE· could not be reactivated from vaccinated calves. The clinical signs observed, following the reactivation of the parental virus, were again much milder on vaccinated than on non-vaccinated calves. Moreover, parental vírus shedding was considerably reduced on vaccinated calves at reactivation. In view of its attenuation, immunogenicity and protective effect upon challenge and reactivation with a virulent BHV-1, the mutant 265gE· was shown to be suitable for use as a BHV-1 differential vaccine vírus.

Para determinar a atenuação e eficácia do 265gE· como imunógeno, bezerros foram inoculados por via intranasal com 106,9 DICC50 do mesmo. O vírus foi detectado em secreções dos animais inoculados em títulos mais baixos e por um período mais curto do que a amostra virulenta parental, inoculada em animais controle. Vinte e um dias após, os animais inoculados com o vírus mutante foram desafiados com a amostra parental, apresentando somente sinais leves de infecção. Os animais controle apresentaram intensa rinotraqueíte e excretaram vírus em títulos mais elevados e por mais tempo do que os vacinados. Seis meses após a vacinação, foi examinada a capacidade de reativação da infecção nos bezerros, através da administração de corticosteróides. O mutante 265gE- não foi reativado dos animais vàcinados. Os sinais clínicos consequentes à reativação do vírus parental foram muito atenuados nos animais vacinados, em comparação com os não vacinados. Além disso, a excreção de vírus de campo foi consideravelmente reduzida nestes últimos. E-mail: flores@ccr.ufsm.br Two isolates of bovine viral diarrhea virus (BVDV) submitted to multiple passages in tissue culture associated with ultraviolet irradiation were evaluated as vaccine virus candidates. The attenuation of the modified viruses was assessed in calves and in pregnant ewes. Intramuscular inoculation of the viruses in four seronegative calves produced only a mil d and transient rise in body temperature, followed by the production of high titers of neutralizing antibodies. The viruses were not detected in nasal secretions or in the blood following inoculation. However, intramuscular inoculation of these viruses in four pregnant ewes resulted in transplacental transmission and infection of ali fetuses. To assess fetal protection conferred by immunization, pregnant ewes immunized twice with the modified viruses were subsequently challenged by intranasal inoculation of BVDV-1 (SV-126.8, n=6) or BVDV-2 (SV-260, n=5). At the day of challenge (134 days after the second immunization), ali ewes had high titers of neutralizing antibodies (256 to >4096) to the vaccine viruses and variable titers (8 to >4096) to Brazilian BVDV-1 and BVDV-2 field isolates. Fifteen days after challenge, the ewes were euthanized and fetal tissues were examined for infectivity. Ali fetuses from non-vaccinated, challenged ewes (n=4) were infected. In contrast, none of the fetuses from the immunized dams (n = 11) were positive for virus, indicating that the immunological response induced by immunization with the vaccine candidate viruses was capable of preventing fetal infection. These results indicate that it is possible to achieve fetal protection to BVDV by induction of a strong immunological response using modified live vaccines.

As amostras foram testadas quanto à sua atenuação para bezerros e fetos ovinos, reatividade antigênica contra isolados de campo, e capacidade de induzir proteção fetal em ovelhas prenhes. Inoculação intramuscular (IM) dos vírus modificados em quatro bezerros produziu apenas uma elevação discreta e passageira da temperatura corporal, seguida de produção de altos títulos de anticorpos neutralizantes. O vírus não foi detectado em secreções nasais ou sangue nos dias seguintes à inoculação. Porém, a inoculação IM desses vírus em quatro ovelhas prenhes foi seguida de transmissão transplacentária e infecção em todos os fetos. Para os testes de proteção fetal, ovelhas prenhes previamente imunizadas com duas doses vacinais, foram inoculadas por via intranasal com amostras de BVDV-1 (SV-126.8, n=6) ou BVDV-2 (SV-260, n=5). No dia do desafio (134 dias após a segunda dose), todos os animais apresentavam altos títulos de anticorpos neutralizantes (256 a >4096) contra os vírus vacinais; além de títulos variados (8 a >4096) contra várias isolados brasileiros de BVDV-1 e BVDV-2. Quinze dias após o desafio, as ovelhas foram sacrificadas e os tecidos fetais foram examinados para a presença de vírus. Todos os fetos das ovelhas controle não-vacinadas apresentaram-se (n=4) positivos para os vírus utilizados no desafio. Em contraste, nenhum feto das ovelhas imunizadas (n = 11) foi positivo para vírus, indicando que a resposta imunológica induzida pela vacinação com os vírus modificados foi capaz de prevenir a infecção fetal. Estes resultados indicam que é possível obter-se forte resposta imunológica e proteção fetal contra o BVDV com o uso de vacinas vivas modificadas.
