#81 - Histopathological changes in liver and gill epithelium of Nile tilapia, Oreochromis niloticus, exposed to waterborne copper, p.103-109

E-mail: fontain@utad.pt Nile tilapia, Oreochromis niloticus, of both sexes were reared in freshwater and exposed to 0.5, 1.0 and 2.5mg L-1 of waterborne copper for a period of 21 days. Liver and gill samples were collected after 21 days of exposure to copper and lesions were analyzed by light microscopy. The main histopathological changes observed in gills exposed to the highest concentration were edema, lifting of lamellar epithelia and an intense vasodilatation of the lamellar vascular axis. Although less frequent, lamellar fusion caused by the filamentar epithelium proliferation and some lamellar aneurisms were also found. The liver of control group exhibited a quite normal architecture, while the fish exposed to copper showed vacuolation and necrosis. These hepatic alterations were more evident in fish exposed to 1.0 and 2.5mg L-1 copper concentrations. The number of hepatocytes nucleus per mm2 of hepatic tissue decreased with the increase of copper concentration. In contrast, the hepatic somatic index was high in fish exposed at 2.5mg L-1 of copper. In short, this work advance new knowledge as influence of copper in the gill and liver histology of O. niloticus and demonstrated that their effects could be observed at different concentrations.

This experimental simulation of sensibilization by Mycobacterium bovis and Mycobacterium avium in cattle aimed to verify the immune response by both the cervical comparative test and the evolution of the specific production of gamma-interferon, and also to identify interference of unspecified reactions by M. avium on the test results. The results support that the experimental animals started a response of delayed hypersensitivity to the inactivated bacilli, and that both diagnostic tests for bovine tuberculosis were efficient for the identification of animals sensitized with M. bovis and for discrimination of reactions generated by inoculation of cattle with M. avium.

[Equine monocytic Ehrlichiosis in Rio Grande do Sul: clinical, pathological and epidemiological aspects.] Ehrlichiose monocítica eqüina no Rio Grande do Sul: aspectos clínicos, anátomo-patológicos e epidemiológicos. Pesquisa Veterinária Brasileira 26(2):97-101. Laboratório de Doenças Infecciosas, Faculdade de Veterinária, Universidade Federal de Pelotas, Campus Universitário, Pelotas, RS 96010-900, Brazil. E-mail: bitoxu@ig.com.br A spontaneous outbreak of equine monocytic Ehrlichiosis (EME) is described. The disease occurred from November 2001 to February 2002 on a farm in the municipality of Arroio Grande, located at the western edge of the Mirim Lake, state of Rio Grande do Sul. Out of 50 horses 13 were affected and 6 died. The morbidity was 26% and the mortality 46.1%. The affected horses were 1-5 years old. The most important clinical sign was a profuse acute diarrhea. At necropsy of a horse that died with clinical signs of the disease, the main alterations were characterized by hemorrhagic areas of the mucosa of the large intestine with liquid contents, and congested and hemorrhagic areas alternating with normal areas as well as nodules with purulent secretion in the mucosa of the small intestine. The histological changes were characterized by moderate mononuclear infiltration of the mucosa, characterizing a lympho-histiocytic enteritis with predominance of macrophages. In the jejunum and ileum, related to the invagination of crypts and villi, a granulomatous enteritis was observed. To confirm the diagnosis of infection by Neorickettsia (Ehrlichia) risticii, blood samples of seven affected horses were taken and a polimerase chain reaction (PCR) was performed. Three tested blood samples were positive (42.8%) with observation of a band of 529pb characteristic of N. risticii.

[Histological and ultrastructural evolution of liver lesions in experimental Myoporum laetum poisoning of sheep and cattle.] Evolução das lesões histológicas e ultra-estruturais no fígado de ovinos e bovinos experimentalmente intoxicados por Myoporum laetum. E-mail: bonel-raposo@brturbo.com Green leaves of Myoporum laetum were collected during spring and summer, and administered to five sheep and six steers at dosages of 20 and 30 g/kg. Liver biopsies were taken before (Controls) and 1, 3 and 7 days after dosage. ln sheep, the clinicai signs were depression, rumen hypomotility, dried feces, tenesmus, teeth grinding, dyspnea and typical lesions of photosensitization. ln cattle, the clinicai picture was much less pronounced. The main histological findings in sheep were vacuolization of hepatocytes, portal ftbrosis, bile duct proliferation and necrosis of periportal hepatocytes; the ultrastructural examination revealed hyperplasia of the smooth endoplasmic reticulum, hepatocellular hydropic degeneration, presence of crystals and severa! other degenerative changes. ln cattle both, the histological and the ultrastructural findings, were less evident.

E-mail: bonel-raposo@brturbo.com Amostras de Myoporum laetum foram colhidas durante a primavera e verão e administradas a cinco ovinos e seis bovinos em doses únicas de 20 e 30 g/kg. Biópsias hepáticas foram colhidas antes (controles) e 1, 3 e 7 dias após a dosagem da planta. Estas biópsias foram analisadas histológica e ultraestruturalmente. Os sinais clínicos, em ovinos, caracterizaramse, especialmente; pordepressão, diminuição dos movimentos ruminais, fezes ressequidas, tenesmo, ranger de dentes, dispnéia e lesões típicas de fotossensibilização. Em bovinos, o quadro clínico foi discreto. Os principais achados histológicos, em ovinos, incluíram vacuolização de hepatócitos, fibrose portal, proliferação de duetos biliares e necrose de hepatócitos periportais. Os estudos ultra-estruturais, em ovinos, revelaram hiperplasia do retículo endoplasmático liso, tumefação de hepatócitos, degranulação e vesiculação do retículo endoplasmático rugoso, presença de cristais aciculares, retenção biliar, tumefação de mitocôndrias e várias outras alterações degenerativas. Em bovinos, tanto os achados histológicos, quanto os ultra-estruturais foram menos evidentes.

ABSTRACT.- Santos D.M.S., Berchieri Jr A., Fernandes S.A., Tavechio A.T. & Amaral LA. 2000. [Salmonella in broiler frozen carcasses.] Salmonella em carcaças de frango congeladas. Pesquisa Veterinária Brasileira 20(1):39-42. Depto Patologia Veterinária, Fac. Ciências Agrárias e Veterinárias, Campus de Jaboticabal, Via de acesso Paulo Donato Castellane s/n, Jaboticabal, SP 14870-000, Brazil. Hundred and fifty frozen broiler carcasses of four commercial brands, purchased at retail stores for Salmonella research, were examined: 43 of the carcasses referred to each of the brands A, B, D and 21 of brand C. Thirty-two percent of the samples were found positive; 11 serotypes were identified as S. Agona, S. Anatum, S. Enteritidis, S. Hadar, S. Havana, S. Mbandaka, S. Montevideo, S. Ouakam, S. Poona, S. Schwarzengrund and S.14, 5, 12:-. Antibiogram testing of the isolated strains showed 100% resistance to ampicilin, 75.0% to cefhalotin, 52.1 % to cephoxitin, 22.9% to tobramicin, 6.2% to polimixin B and to tetracyclines, 4.2% to gentamicin, and 2.1% to netilmicin, to aztreonam and to amicacin. All strains showed total sensibility to chloramphenicol and to sulfazotrim.

RESUMO.- Santos D.M.S., Berchieri Jr A., Fernandes S.A., Tavechio A.T. & Amaral LA. 2000. [Salmonella in broiler frozen carcasses.] Salmonella em carcaças de frango congeladas. Pesquisa Veterinária Brasileira 20(1):39-42. Depto Patologia Veterinária, Fac. Ciências Agrárias e Veterinárias, Campus de Jaboticabal, Via de acesso Paulo Donato Castellane s/n, Jaboticabal, SP 14870-000, Brazil. Um total de 150 carcaças de frango congeladas, de quatro marcas comerciais, foram analisadas para pesquisa de Salmonella. Foram examinadas 43 carcaças de cada uma das marcas A, B, D e 21 da marca C. Observou-se um percentual de 32,0% de contaminação. Foram identificados 11 sorotipos: S. Agona, S. Anatum, S. Enteritidis, S. Hadar, S. Havana, S. Mbandaka, S. Montevideo, S. Ouakam, S. Poona, S. Schwarzengrund e S. I 4, 5, 12: -. O antibiograma das cepas mostrou 100% de resistência à ampicilina, 75,0% à cefalotina, 52, 1 % à cefoxitina, 22,9% à tobramicina, 6,2% à polimixina B e à tetraciclina, 4,2% à gentamicina e 2,1% à netilmicina, ao aztreonam e à amicacina. Todas as cepas apresentaram sensibilidade total ao cloranfenicol e ao sulfazotrim.


#86 - Efficacy of selenite-novobiocin and tetrathionate-novobiocin. broths in survey of Salmonella from meat meal

Abstract in English:

Selenite-novobiocin (SN) and tetrathionate-novobiocin (TN) broths were compared for the isolation of Salmonella from meat meal, It was also investigated the correlation between the occurrence of the indicator organisms and of Salmonella. SN and TN were equally effective. It was verified that none of these indicator groups (total coliforms and fecal coliforms) are reliable as an index for Salmonella contamination in meat meals. There was no correlation between Salmonella counts and the number of isolated Salmonella colonies.

Os caldos selenito-novobiocina (SN) e tetrationat-onovobiocina (TN) foram comparados durante o isolamento de Salmonella de amostras de farinha de carne utilizadas para fabricação de ração animal. Também, foram realizadas contagens de Salmonella e de coliformes (total e fecal) para verificar se é possível estabelecer alguma correlação entre a presença de coliformes e a presença e o isolamento de Salmonella. De acordo com os resultados obtidos, o caldo SN e o caldo TN demonstraram rendimento semelhante. As contagens de coliformes (total e fecal) segundo a análise estatística, não apresentaram correlação alguma (positiva ou negativa) com a contagem de Salmonella e com o número de cepas isoladas através dos caldos SN e TN. Também não foi constatada correlação entre a contagem de Salmonella e o número de cepas isoladas.


#87 - Preenrichment and direct enrichment in survey of Salmonella

Abstract in English:

A comparison on several different culture procedures for Salmonella isolation from meat meals was made, using lactose broth, buffered peptone water, quarter strength Ringer's solution for preenrichment and selenite-novobiocin for direct enrichment in cultures incubated at 37ºC and -43°C, with or without a layer of vaseline. All preenrichment procedures using cultures incubated at 43°C were superior to preenrichment ones using cultures incubated at 37ºC and direct enrichment procedures using both 37ºC and 43ºC incubation temperatures. Nothing seemed to be gained by the use of a vaseline layer over for the purpose of stopping aerobic incubation. In what concerns lactose broth, buffered peptone water and quarter strength Ringer's solution media, no significant differences for efficiency were found.

O presente estudo teve o propósito de avaliar, durante investigação da presença de Salmonella em farinha de carne, o aproveitamento da solução de Ringer 1/4 (SR 1/4), como pré-enriquecimento, em comparação com o caldo lactosado e a água peptonada tamponada (APT) e também com o caldo selenito-novobiocina (SN) como enriguecimento direto, incubados a 37ºC ou a 43ºC, com ou sem uma camada de vaselina. Os resultados alcançados demonstraram que a 37ºc é indiferente utilizar o pré-enriquecimento ou o enriquecimento direto. Mas a 43ºC o pré-enriquecimento apresenta rendimento superior ao enriquecimento direto incubado a 37ºC e a 43ºC e ao pré-enriquecimento incubado a 37ºC. A adição de vaselina é dispensável, por que não melhora o índice de isolamento. Quanto ao pré-enriquecimento, constatou-se que a SR 1/4 pode ser aproveitada em substituição ao caldo lactosado e à APT.
