#1 - Ultrastructural and morphometric description of the ear skin and cartilage of two South American wild histricognate rodents (Dasyprocta leporina and Galea spixii)

Abstract in English:

Skin and cartilage have been the main source for the recovery of somatic cells to be used in conservation strategies in wild mammals. In this sense, an important step for the cryopreservation of these samples is to recognize the properties of the skin and cartilage. Thus, knowing that the skin may differ among species and aiming to contribute to the establishment of cryobanks, the study examined the differences in the ear skin and cartilage of wild rodents from South America, agouti (Dasyprocta leporina) and spix’s yellow-toothed cavy (Galea spixii). Ultrastructural and quantitative methods were used to measure skin and cartilage thickness, density of collagen and elastic fibers, cell type number and distribution, and proliferative activity. Although ultrastructural analysis revealed a similar pattern between species, morphometric analysis of the skin and cartilage showed differences between agoutis and cavies regarding thickness of epidermis layers (corneum: 5.3±2.5µm vs. 3.9±0.6µm; intermediate: 16.4±6.2µm vs. 23.4±8.1µm; basal: 9.9±2.1µm vs. 4.8±0.5µm), dermis (183.1±44.0µm vs. 258.2±22.9µm), total skin (211.8±46.0µm vs. 290.3±23.7µm) and perichondrium (27.6±6.1µm vs. 10.5±1.8µm). A greater number of epidermal cells (61.7±15.2 vs. 24.8±7.6) and chondrocytes (32.7±9.0 vs. 27.5±4.7) were observed in agouti, while the cavy presented a greater number of melanocytes (12.6±4.7 vs. 29.9±6.2), keratinocytes (14.7±4.2 vs. 29.8±7.6), and fibroblasts (103.6±24.7 vs. 112.2±11.3). Moreover, a higher percentage of collagen fibers and proliferative activity was observed in the skin of cavies, when compared to the skin of agoutis. Therefore, there are differences between agouti and cavy for ear skin and cartilage, requiring the establishment of species-specific cryopreservation protocols.

A pele e cartilagem têm sido uma importante fonte de recuperação de células somáticas a serem utilizadas em estratégias de conservação em mamíferos silvestres. Nesse contexto, uma importante etapa para criopreservação é conhecer, inicialmente, as propriedades que compõem a pele e cartilagem. Sabendo, então, que a pele pode diferir-se entre espécies e com o objetivo de contribuir para o estabelecimento de criobancos, o estudo evidenciou as diferenças da pele e da cartilagem do pavilhão auricular apical de cutias (Dasyprocta leporina) e preás (Galea spixii) que são roedores silvestres presentes na América do Sul. Para tanto, métodos ultraestruturais e quantitativos foram utilizados para mensurar a espessura da pele e da cartilagem, densidade de fibras colágenas e elásticas, número e distribuição dos tipos celulares e atividade proliferativa. Embora as propriedades ultraestruturais em cutias e preás tenham se mostrado semelhantes, avaliações acerca da morfometria da pele e da cartilagem demonstrou diferenças, especialmente nas camadas epidérmicas (córnea: 5,3±2,5µm vs. 3,9±0,6µm; espinhosa: 16,4±6,2µm vs. 23,4±8,1µm; basal: 9,9±2,1µm vs. 4,8±0,5µm), derme (183,1±44,0µm vs. 258,2±22,9µm), pele total (211,8±46,0µm vs. 290,3±23,7µm) e pericôndrio (27,6±6,1µm vs. 10,5±1,8µm). Além disso, um número maior de células epidérmicas (61,7±15,2 vs. 24,8±7,6) e condrócitos (32,7±9,0 vs. 27,5±4,7) foram observados em cutias, enquanto em preás um maior número de melanócitos (12,6±4,7 vs. 29,9±6,2), queratinócitos (14,7±4,2 vs. 29,8±7,6) e fibroblastos (103,6±24,7 vs. 112,2±11,3) foram evidenciados. Ainda, em preás, uma maior porcentagem de fibras colágenas e da atividade proliferativa foram observadas quando comparadas a pele de cutias. Portanto, existem diferenças entre cutias e preás para pele e cartilagem do pavilhão auricular, exigindo desta forma um estabelecimento de protocolos de criopreservação específica para cada uma destas espécies.


#2 - Histomorphometric and immunohistochemical evaluation of the frontal cerebral cortex in diabetic rats after treatment with melatonin

Abstract in English:

The central nervous system is vulnerable to complications caused by diabetes. These complications lead to increased oxidative stress in the brain, resulting in damage to the cerebral cortex, among other regions. Insulin and hypoglycemic agents are still the most widely used treatments. However, current research with an experimental model of diabetes suggests the use of antioxidants, such as melatonin. Thus, we tested the hypothesis that exogenous melatonin may decrease or prevent the effects of diabetes in the frontal cortex of the rat brain. Fifty albino rats were allocated into five groups: GC = rats without diabetes induction, GD = diabetic rats induced by streptozotocin, GDM = streptozotocin-induced and melatonin-treated diabetic rats, GDI = diabetic rats induced by streptozotocin and treated with insulin, GDMI = diabetic rats induced by streptozotocin and treated with melatonin and insulin simultaneously. Diabetes was induced by intraperitoneal administration of streptozotocin (60mg/kg). Insulin (5U/day) was administered subcutaneously and melatonin (10mg/kg) by drinking water; both treatments last days after. We analyzed animals’ weight, the cytokines IL-6 and TNF-α, apoptosis, glycogen, and did morphometry and histopathology of the frontal cortex were analyzed. The results showed that the cerebral cortex of the diabetic animals presented axonal degeneration, reduced number of neurons in the cortex, reduced glycogen, increased IL-6 and TNF-α expression, high apoptotic index, and reduced animal weight and the brain. Treatment with melatonin associated or not with insulin prevented such effects. Thus, we conclude that melatonin associated with insulin may be an alternative for avoiding the impact of diabetes in the brain’s frontal cortex.

O sistema nervoso central é vulnerável a complicações originadas pelo diabetes estresse oxidativo no cérebro e resultando em lesões no córtex cerebral, dentre outras regiões. A insulina e hipoglicemiantes ainda são os tratamentos mais utilizados, entretanto, pesquisas atuais com modelo experimental do diabetes sugerem a utilização de antioxidantes como, por exemplo, a melatonina. Assim, testamos a hipótese de que a melatonina exógena pode diminuir ou prevenir os efeitos do diabetes no córtex frontal do cérebro de ratos. Foram utilizados 50 ratos albinos, divididos em 5 grupos: GC = ratos sem indução ao diabetes, GD = ratos induzidos ao diabetes pela estreptozotocina, GDM = ratos induzidos ao diabetes pela estreptozotocina e tratados com melatonina, GDI = ratos induzidos ao diabetes pela estreptozotocina e tratados com insulina, GDMI = ratos induzidos ao diabetes pela estreptozotocina e tratados com melatonina e insulina simultaneamente. O diabetes foi induzido pela administração intraperitoneal de estreptozotocina (60mg/kg). A insulina (5U/dia) foi administrada por via subcutânea e a melatonina (10mg/kg) pela água de beber. Ambos tratamentos foram realizados durante 30 dias após a indução. Foram analisados o peso dos animais, do cerebro, as citocinas IL-6 e TNF-α, apoptose, glicogênio, além da morfometria e histopatologia do córtex frontal. Os resultados mostraram que o córtex cerebral dos animais diabéticos apresentou degeneração axonal, redução do número de neurônios no córtex, redução do glicogênio, aumento da expressão do IL-6 e TNF-α, elevação do índice apoptótico, além da redução do peso dos animais e do cérebro. O tratamento com melatonina associada ou não a insulina preveniu tais efeitos. Assim, concluímos que a melatonina associada ou não a insulina pode ser uma alternativa na prevenção dos efeitos do diabetes no córtex frontal do cérebro.


#3 - Comparative retinal histomorphometry and visual acuity of three bat species of the genus Artibeus (Phyllostomidae: Stenodermatinae)

Abstract in English:

This study performed a histomorphometric analysis of the retina and estimated the visual acuity of three fruit-eating bats of the genus Artibeus, including Artibeus lituratus, Artibeus planirostris and Artibeus obscurus. In total, 13 animals were used for this study, in which the retinas were hemidisected, fixed, cut, and stained. The visual acuity was determined by the density of ganglion cells in the retina and the retinal layers were also measured from histological sections. The retinas of these bats are avascular, do not present tapetum lucidum in any quadrant, and have the 10 retinal layers common in mammals. Moreover, it was observed that the thickness of the retina in the central region was significantly higher in all measured parameters (p<0.001), except for the outer plexiform layer (OPL) which was significantly higher in the peripheral region (p<0.001). The retinas of the three species showed a horizontal visual streak with a higher concentration of retinal ganglion cells (RGCs) at the inferotemporal region. In addition, the species A. lituratus exhibited extras areas of high cell density in the retina. Thus, A. lituratus showed the highest visual acuity (1.92 cycles/degree), while A. planirostris showed a visual acuity of (1.77 cycles/degree) and A. obscurus exhibited the lowest visual acuity (1.50 cycles/degree). All these characteristics are related to the echolocation system and the eating habits of each species. Therefore, it can be concluded that bats of the genus Artibeus have a high visual acuity value compared to other echolocating bats and all these differences might be directly linked to the phylogeny of the genus.

Este estudo realizou uma análise histomorfométrica da retina e determinou os valores de acuidade visual de três espécies de morcegos do gênero Artibeus, incluindo Artibeus lituratus, Artibeus planirostris e Artibeus obscurus. Em total, 13 animais foram usados para esse estudo, no qual as retinas foram hemidissecadas, fixadas, cortadas e coradas. A acuidade visual foi determinada pela densidade de células ganglionares na retina e também foi realizado a mensuração das camadas retinianas a partir de cortes histológicos. As retinas dessas três espécies são avasculares, não apresentaram tapetum lucidum em nenhum quadrante e possuem as 10 camadas retinianas comum em mamíferos. Verificou-se também que a espessura da retina na região central foi significativamente maior em todos os parâmetros mensurados (p<0,001), com exceção da camada plexiforme externa (OPL) que foi significativamente maior na região periférica (p<0,001). As retinas das três espécies exibiram uma especialização retiniana em forma de faixa visual horizontal apresentando a maior concentração de células ganglionares na região inferotemporal. Além disso, a espécie A. lituratus exibiu áreas extras de alta densidade celular na retina. Assim, a espécie A. lituratus exibiu a maior acuidade visual (1,92 ciclos/grau), A. planirostris mostrou um valor intermediário entre essas duas espécies (1,77 ciclos/grau) e A. obscurus exibiu a menor acuidade visual (1,50 ciclos/grau). Todas estas características estão relacionadas aos padrões de ecolocalização e hábitos alimentares de cada espécie. Portanto, pode-se concluir que os morcegos do gênero Artibeus têm um alto valor de acuidade visual em comparação com outros morcegos ecolocalizadores e todas essas diferenças devem estar diretamente ligadas à filogenia do gênero.


#4 - Reproductive biology, morphology, and morphometry of ovaries and oviducts of Trachemys scripta elegans in Brazilian Cerrado

Abstract in English:

Although Trachemys scripta elegans is an exotic species popular as a pet in Brazil, studies on reproductive biology and capacity are non-existent in the Brazilian Cerrado. This study analyzed ovarian and oviduct characteristics and the egg production capacity of T. scripta elegans grown in this biome. The findings will associate with the size of the specimens and the sexual maturity, aiming at comparisons with native and exotic populations, as well as interspecific and contributing to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. Thus, 39 females had evaluated the body biometry and the morphology and morphometry of the ovaries and oviducts. G2 (N=20): with Class I (>5-10mm) follicles, with Class I and Class II (>10-fold) follicles, 25mm) and G3 (N=9) with Class I, Class II and Class III (>25mm) follicles. Analysis of variance, Scott-Knott’s test, and Pearson’s correlation analysis showed that there was no significant difference between the groups in body biometry; in the mean gonadosomatic index and gonadal morphometry, only the width of the oviducts in the right antimer and the mass and width in the left antimer were higher in G3, the only one that presented eggs. There was positive and harmonic development between body mass, carapace, and plastron, and gonadal growth occurred concomitantly with body growth, indicating a higher reproductive potential and a positive relationship between the size of the litter and the female litter. The gonadosomatic index proved to be an excellent reproductive indicator, and the ovarian evaluation was a better indicator of sexual maturity than the maximum carapace length. Ovaries were irregular structures, without delimitation between the cortical and medullary regions and filled with vitelogenic follicles of different diameters, atresic follicles, and corpora lutea, which reflected the ovarian complexity of the species and the presence of follicular hierarchy. In the scarce stroma, two germinative beds were observed per ovary and the presence of gaps very close to the follicles and associated with the blood vessels. Analysis of gonadal tissue revealed three types of oocytes according to cytoplasmic characteristics: homogeneous, vesicular or vesicular in the cortex with apparent granules. Oviducts were functional and separated, joining only in the final portion to form the cloaca and subdivided into infundibulum, tuba, isthmus, uterus, and vagina. The structure of the uterine tube was composed of serosa, muscular and mucous, which was full of glands. The presence of eggs in the oviducts indicated that the specimens can reproduce in the Brazilian Cerrado. This study provides necessary and relevant information on the reproductive biology and capacity of T. scripta elegans in the Brazilian Cerrado and can contribute to the understanding of its impact on the invaded ecosystems and the establishment of eradication programs. The extraction of females with capacity can reduce the annual reproductive yield of the species and decrease its effect on local biodiversity.

Embora Trachemys scripta elegans seja uma espécie exótica popular como animal de estimação no Brasil, estudos sobre biologia e capacidade reprodutivas são inexistentes no Cerrado brasileiro. Este estudo analisou características ovarianas e do oviduto e a capacidade de produção de ovos em T. scripta elegans criadas neste bioma, correlacionando estes achados ao tamanho dos espécimes e a maturidade sexual, visando comparações com populações nativas e exóticas, bem como interespecíficas e contribuir para a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação. Assim, 39 fêmeas tiveram avaliadas a biometria corporal e a morfologia e morfometria dos ovários e ovidutos. De acordo com o tamanho dos folículos ovarianos as fêmeas foram separadas em G1 (N= 10): com folículos Classe I (>5-10 mm), G2 (N= 20): com folículos Classe I e Classe II (>10-25 mm) e G3 (N= 9) com folículos Classe I, Classe II e Classe III (>25 mm). À análise de variância, teste de Scott-Knott e à análise de correlação de Pearson verificou-se que não houve diferença significativa entre os grupos na biometria corporal; no índice gonadossomático médio e na morfometria gonadal, apenas a largura dos ovidutos no antímero direito e a massa e a largura no antímero esquerdo foram maiores no G3, o único que apresentou ovos. Houve desenvolvimento positivo e harmônico entre massa corporal, carapaça e plastrão e o crescimento gonadal ocorreu concomitante ao crescimento corporal, indicando maior potencial reprodutivo e relação positiva entre o tamanho da ninhada de ovos e o da fêmea. O índice gonadossomático mostrou-se um bom indicador reprodutivo e a avaliação ovariana um melhor indicador da maturidade sexual que o comprimento máximo da carapaça. Ovários foram estruturas irregulares, sem delimitação entre a região cortical e medular e repletos de folículos vitelogênicos de diferentes diâmetros, folículos atrésicos e corpos lúteos, que refletiram a complexidade ovariana da espécie e a presença de hierarquia folicular. No estroma escasso foram observados dois leitos germinativos por ovário e a presença de lacunas muito próximas aos folículos e associadas aos vasos sanguíneos. A análise do tecido gonadal revelou três tipos de oócitos de acordo com as características do citoplasma: homogêneo, vesicular ou vesicular no córtex com grânulos aparentes. Ovidutos eram funcionais e separados, unindo-se apenas na porção final para formar a cloaca e subdividiam-se em infundíbulo, tuba uterina, istmo, útero e vagina. A estrutura da tuba uterina era constituída de serosa, muscular e mucosa, a qual era repleta de glândulas. A presença de ovos nos ovidutos indicou que os espécimes podem se reproduzir no cerrado brasileiro. Este estudo fornece informações básicas e relevantes da biologia e capacidade reprodutivas de T. scripta elegans no Cerrado brasileiro e pode contribuir com a compreensão de seu impacto nos ecossistemas invadidos e com o estabelecimento de programas de erradicação, uma vez que a extração de fêmeas com capacidade reprodutiva pode contribuir com a diminuição do rendimento reprodutivo anual da espécie e diminuir seu efeito sobre a biodiversidade local.


#5 - Seasonal influence on testicular morphophysiological parameters of bat Carollia perspicillata in fragments of the Atlantic Forest, northeastern Brazil

Abstract in English:

Bats belong to the order Chiroptera, family Phyllostomidae, and present a wide diversity of reproductive strategies. However, information on the reproductive biology of male bats is scarce, mainly in the Northeast Region of Brazil. Thus, this study evaluated the seasonal testicular histomorphometry of the bat Carollia perspicillata in fragments of the Atlantic Forest in Pernambuco state. To this end, adult males were collected, euthanized for removal of the testicles, and later submitted to a routine histological technique. Histomorphometric analysis included assessment of the areas of tubular and intertubular compartment occupation, as well as quantification of spermatocytes, rounded spermatids, elongated spermatids, and Sertoli and Leydig cells. Results indicated that this bat species presents reproductive seasonality, because significantly higher averages of the testicular parameters were observed in the rainy season, which is a period of greater availability of food resources. Such inferences indicate that there is a synchrony between peak spermatogenesis and hormonal inversion in the months of high precipitation; furthermore, a higher carrying capacity of the Sertoli cells is noted. C. perspicillata males possibly present greater sperm and androgenic activity in the rainy season, associated with increased tubular area and number of spermatogenic cells, as well as with the intertubular area and number of Leydig cells, respectively.

Os morcegos pertencem a ordem Chiroptera, família Phyllostomidae, e apresentam ampla diversidade de estratégias reprodutivas. Entretanto, as informações relacionadas à biologia reprodutiva dos machos são escassas, principalmente no Nordeste do Brasil. Dessa forma, o trabalho avaliou a histomorfometria sazonal testicular de Carollia perspicillata em fragmentos de Mata Atlântica de Pernambuco. Para tanto, os machos adultos foram coletados, eutanasiados para a remoção dos testículos, e posteriormente submetidos à técnica histológica de rotina. As análises histomorfométricas avaliaram as áreas de ocupação do compartimento tubular e intertubular, assim como a quantificação dos espermatócitos, espermátides arredondadas, espermátides alongadas, células de Sertoli e de Leydig. Os resultados indicaram que a espécie apresenta sazonalidade reprodutiva, visto que maiores médias significativas dos parâmetros testiculares foram encontradas na estação chuvosa, que é um período de maior disponibilidade de recursos alimentares. Tais inferências indicam que existe uma sincronia entre o pico de espermatogênese e investimento hormonal nos meses de alta precipitação, atrelado a isso, nota-se ainda, uma maior capacidade de suporte das células de Sertoli. Os machos de C. perspicillata possivelmente apresentam uma maior atividade espermática e androgênica na estação chuvosa, associadas ao aumento da área tubular e do número de células espermatogênicas, assim como da área intertubular e do número das células de Leydig, respectivamente.


#6 - Morphometry of the mandibular foramen applied to local anesthesia in hoary fox (Lycalopex vetulus)

Abstract in English:

Great part of the veterinary care in wild carnivores is intended to treat the dental disorders, and oral cavity disorders may generally affect the animal health as a whole. It is factual that knowing the location of the mandibular foramen is vital for local anesthetic block of the inferior alveolar nerve, however, there is still no data on the morphometry of the hoary fox mandibular foramen. The aim was describing morphometry of the mandibular foramen of this species and associating its position with anatomic reference points in the mandible, thus providing support for more effective local anesthetic block of the inferior alveolar nerve in such species. Four adult jaws of Lycalopex vetulus were used. Radiography and biometrics of the hemimandibulas were performed. The rostral third of the jaw body in a lateral view presented three mental foramens, being a rostral, a medium and a caudal. Each hemimandibula presented I3/C1/PM4/M3. The angle of the mandible was marked by the masseteric fossa, the angular incisure, the angular process and the mandibular foramen, and this last one located perpendicularly to the dorsal edge of the angular incisures in medial view. In this view, the crevice to the milohyoid nerve, projected in the caudodorso rostral direction, was also evidenced. The ramus of the mandible was characterized by the presence of the condylar and coronoid processes, and by the dorsal and ventral mandibular incisions. Statistical analyzes did not present significant differences between the antimeres of the studied animals, and the penetration of the needle perpendicularly to the dorsal end of the angular incision on average 8.79mm, overlapped to the medial face of the angle of the mandible could be indicated. Alternatively, the access may also be achieved by inserting the needle in an average of 17.69mm perpendicular to the dorsal end of the angular process, in contact with the medial aspect of the angle of the mandible, and in caudo-rostral projection, also allowing a better anesthetic blockade of the inferior alveolar nerve in L. vetulus. It can also be concluded that the masseteric fossa, the dorsal and ventral mandibular angles, the crevice to the milohyoid nerve, the ramus of the mandible and the mandibular foramen presented differences in their topographic descriptions when compared to the other canids.

A maior parte dos atendimentos veterinários em carnívoros silvestres destina-se ao tratamento das afecções dentárias, sendo que doenças de cavidade oral podem afetar a saúde do animal como um todo. É factível que o conhecimento da localização do forame mandibular é essencial para o bloqueio anestésico local do nervo alveolar inferior, entretanto, ainda são inexistentes dados sobre a morfometria do forame mandibular de raposa-do-campo. Objetivou-se descrever a morfometria do forame mandibular desta espécie, e correlacionar sua posição com pontos de referência anatômica na mandíbula, oferecendo subsídio para um bloqueio anestésico local mais efetivo do nervo alveolar inferior nesta espécie. Foram utilizadas quatro mandíbulas de cadáveres adultos de Lycalopex vetulus. Realizou-se a radiografia e as biometrias das hemimandíbulas. O terço rostral do corpo da mandíbula em uma vista lateral apresentou três forames mentuais, sendo um rostral, um médio e um caudal. Cada hemimandíbula apresentou I3/C1/PM4/M3. O ângulo da mandíbula foi marcado pela fossa massetérica, pela incisura angular, pelo processo angular e pelo forame mandibular, e este último localizado perpendicularmente ao extremo dorsal da incisura angular em vista medial. Nesta vista, também se evidenciou o sulco para o nervo milohióideo, projetado em sentido caudodorsorostral. O ramo da mandíbula foi caracterizado pela presença dos processos condilar e coronóide, e pelas incisuras mandibulares dorsal e ventral. As análises estatísticas não apresentaram diferenças significantes entre os antímeros dos animais estudados, podendo-se indicar a penetração da agulha perpendicularmente ao extremo dorsal da incisura angular em média 8,79mm, justaposto à face medial do ângulo da mandíbula. Alternativamente, o acesso também poderá ser realizado introduzindo a agulha em média 17,69mm de forma perpendicular ao extremo dorsal do processo angular, em contato com a face medial do ângulo da mandíbula, e em projeção caudorostral, permitindo também um melhor bloqueio anestésico do nervo alveolar inferior na L. vetulus. Também se pode concluir que a fossa massetérica, as incisuras angular, mandibulares dorsal e ventral, o sulco para o nervo milohióideo, o ramo da mandíbula e o forame mandibular apresentaram diferenças em suas descrições topográficas quando comparados aos demais canídeos.

Morphophysiological adaptations are necessary to allow the maintenance of migratory cycles and, therefore, studies with this focus can help clarify biological aspects related to migration. We analyzed the morphology variation in pectoral muscles and intestinal mucosa of Calidris pusilla, during different phases of the wintering period on the coast of Brazil. Fragments of pectoral muscles and duodenal were collected, fixed and processed for histology according to standard procedure, from specimens captured in a locality on the Brazilian coast. Modifications were found in the measured parameters among the three phases of wintering, arrival in Brazil (October, mid-period), January and departure to the Northern Hemisphere - May. The registered structural dynamism characterizes the growth of flight musculature and intestinal changes related to nutrition. Such changes occur temporarily due to the activities of preparation and migration between the northern and southern hemispheres.

O dinamismo estrutural registrado caracteriza o crescimento da musculatura de vôo e as alterações intestinais relacionadas à nutrição. As mudanças nos parâmetros medidos entre as três fases do inverno, chegada ao Brasil (outubro, meio período), janeiro e saída para o Hemisfério Norte. Tais mudanças ocorrem temporariamente devido às atividades de preparação e migração entre os hemisférios norte e sul.

E-mail: dalilian.an@gmail.com The objective of this study was to evaluate the occurrence of main gastrointestinal parasites and to quantify and identify the species of Eimeria present in young goats and adults in the city of Quixadá, Ceará, during the dry and rainy season. Six hundred fifty-four animals were used, with 334 animals during the dry season, which were 149 mothers and 185 offspring and 320 animals during the rainy season, with 106 mothers and 214 offspring, independent of breed standard and creation system. Samples were collected in the months of November and December 2012 and May 2013 in the city of Quixadá, Ceará and sent to the Parasitology Laboratory of Embrapa Goats and Sheep at Sobral, Ceará, feces were collected for counting the number of eggs per gram of feces (EPG), counting oocysts per gram of feces (OOPG), coproculture and identification of Eimeria species based on the morphometry. Matrices showed higher means egg type Strongyloidea (372.03) and Strongyloides (8.45) in the dry season and the rainy season had higher mean only to Strongyloidea (502.85). The inverse happened to OOPG, where the offspring showed higher means in the dry season (9387.41) and rainy (9630.37). The genus most frequent of nematode in coprocultures was Trichostrongylus. (63.67%) in the dry season and Haemonchus sp. the rainy (66.67%). One hundred percent of the herds, oocysts of Eimeria were found, with 81% of infected animals. The occurrence of eimeriosis in young and adult categories was 64.3% and 83.2% in the dry season and 87.4% and 92.4% during the rainy season. The species identified were: Eimeria alijevi, E. apsheronica, E. arloingi, E. caprina, E. caprovina, E. christenseni, E. hirci, E. and E. jolchijevi ninakohlyakimovae. In the offspring E. alijevi was the most frequent in the dry season and the rainy E. ninakohlyakimovae. Already in matrices the species E. Alijevi showed the highest frequency in both periods. In herds studied it evidenced the presence of gastrointestinal nematodes eggs and oocysts of Eimeria spp. independent of animal category.

Foram utilizados 654 animais, sendo 334 animais no período seco, dos quais eram 149 matrizes e 185 crias e 320 animais no período chuvoso, sendo 106 matrizes e 214 crias, independente de padrão racial e sistema de criação. As coletas foram realizadas nos meses de novembro e dezembro de 2012 e maio de 2013 no município de Quixadá-Ceará e encaminhadas ao Laboratório de Parasitologia da Embrapa Caprinos e Ovinos na cidade de Sobral-Ceará. Foram coletadas fezes para contagem do número de ovos por gramas de fezes (OPG), contagem de Oocistos por gramas de fezes (OOPG), coprocultura e identificação das espécies de Eimeria com base na morfometria. As matrizes apresentaram maiores médias de ovos do tipo Strongyloidea (372,03) e Strongyloides (8,45) no período seco e no período chuvoso apresentaram maiores médias apenas para Strongyloidea (502,85). O inverso aconteceu com o OOPG, onde as crias apresentaram maiores médias no período seco (9387,41) e no chuvoso (9630,37). O gênero de nematódeo mais frequente na coprocultura foi o Trichostrongylus sp. (63,67%) no período seco e o Haemonchus sp. no chuvoso (66,67%). Em 100% dos rebanhos, foram encontrados oocistos de Eimeria, com 81% dos animais infectados. A ocorrência da eimeriose nas categorias jovens e adultas foi de 64,3% e 83,2% no período seco e de 87,4% e 92,4% no período chuvoso. As espécies identificadas foram: Eimeria alijevi, E. apsheronica, E. arloingi, E. caprina, E. caprovina, E. christenseni, E. hirci, E. jolchijevi e E. ninakohlyakimovae. Nas crias a E. alijevi foi a mais freqüente no período seco e a E. ninakohlyakimovae no chuvoso. Já nas matrizes a espécie E. alijevi apresentou a maior frequência em ambos os períodos. Nos rebanhos estudados foi evidenciado a presença de ovos de nematódeos gastrointestinais e oocistos de Eimeria spp. independente da categoria animal.


#9 - Analysis of the reproductive stage and ovarian histomorphometry of Dermanura cinerea (Chiroptera: Phyllostomidae) in an Atlantic Forest fragment of Pernambuco, northeastern Brazil

Abstract in English:

This study aimed to analyze the reproductive stage, histology and morphometry of the ovary of Dermanura cinerea in an Atlantic Forest fragments in the Biological Reserve of Saltinho, Pernambuco, Brazil. Adult females were captured monthly by mist net, during two consecutive nights from June/2014 to November/2015. The meteorological data were provided by the National Institute of Meteorology and grouped together with the reproductive data, in six periods: period I (June to August/2014), period II (September to November/2014), period III (December/2014 to February/2015), period IV (March to May/2015), period V (June to August/2015) and period VI (September to November/2015). The females were classified according to their reproductive stages in: Inactive, pregnant, lactating and postlactating. Among the 25 females caught, 10 inactive females were selected for the histomorphometric analysis of the ovaries and then they were euthanized. After that, the specimens had their ovaries removed, fixed, processed and included in Paraplast. The organs were cut and stained by Hematoxylin – Eosin and the histological slides produced were submitted to the appropriate optical microscope analysis. Each type of ovarian follicle was estimated according to the histological characteristics evidenced for other mammals. In the morphometric analysis, it was considered: The area of the ovary (measured in square millimeter), Quantification of different types of ovarian follicles (unilaminar and multilaminar primary follicles, antral and matures) and corpus luteum, as well as the area of each by those different follicles (measured in square micrometer) The results indicated that 16 females were inactive, 5 were lactating, 4 were pregnant and no post-lactating. The histological analysis revealed that the ovaries of D. cinerea exhibit characteristics, structures and cellular types similar to other mammals. The morphometric data of the study area were statistically evaluated using the Student’s t-test. Therefore, ovarian histomorphometric analysis are very important to infer with greater certainty about the reproduction of a species. Thus, it was observed that females of D. cinerea exhibit two reproductive peaks, that is, they can give birth either at the end of dry periods or from the beginning to the middle of the rainy season. The main feature observed in the ovary of this species, found in the Atlantic Forest area of the state of Pernambuco, is the preparation of the ovarian cortex to ovulate in months of high rainfall throughout the year. Therefore, this species tries to adjust its reproductive period with the variations of the environmental conditions, so that the reproduction might occur during the period more favorable for the maintenance of their offspring.

Esse estudo objetivou analisar o estágio reprodutivo, histologia e morfometria do ovário de Dermanura cinerea em um fragmento de Mata Atlântica da Reserva Biológica de Saltinho (Pernambuco, Brasil). Foram utilizadas fêmeas adultas capturadas por redes de neblina, de modo que as coletas ocorreram mensalmente, durante duas noites consecutivas de Junho/2014 a Novembro/2015. Os dados meteorológicos foram fornecidos pelo Instituto Nacional de Meteorologia e agrupados, juntamente com os dados reprodutivos, em seis períodos, sendo estes: período I (Junho a agosto/2014), período II (Setembro a novembro/2014), período III (Dezembro/2014 a fevereiro/2015) período IV (Março a maio/2015), período V (Junho a agosto/2015) e período VI (Setembro a novembro/2015). As fêmeas foram classificadas, de acordo com o estágio reprodutivo em: inativas, grávidas, lactantes e pós-lactantes. Das 25 fêmeas capturadas, foram selecionadas, para as análises histomorfométricas dos ovários, 10 fêmeas inativas, as quais foram eutanasiadas. Depois disso, os espécimes tiveram os seus ovários removidos, fixados, processados e inclusos em paraplast. Os cortes foram corados por Hematoxilina – Eosina e as lâminas histológicas produzidas foram submetidas às devidas análises em microscópio óptico. Cada tipo de folículo ovariano foi estimado de acordo com as características histológicas evidenciadas para outros mamíferos. Nas análises morfométricas, considerou-se: a área do ovário (mensurada em milímetro quadrado), quantificação dos diferentes tipos de folículos ovarianos (primários unilaminar e multilaminar, antrais e maduros) e corpo lúteo, bem como foi medida a área de cada um desses diferentes tipos de folículos (mensurada em micrômetro quadrado). Os resultados indicaram a presença de 16 fêmeas inativas, 5 lactantes, 4 grávidas e nenhuma pós-lactante. As análises histológicas destacaram que os ovários de D. cinerea possui características, estruturas e tipos celulares padrões semelhantes ao dos demais mamíferos. Os dados morfométricos da área do ovário foram avaliados estatisticamente por meio do teste t de Student. Assim, as análises histomorfométricas ovariana são de suma importância para inferir com maior segurança sobre a reprodução de uma dada espécie. Portanto, ficou constatado que as fêmeas de D. cinerea apresentam dois picos reprodutivos, isto é, podem parir no final de períodos secos ou do início para metade do período chuvoso. A principal característica observada no ovário dessa espécie, encontrada em área de Mata Atlântica do estado de Pernambuco, foi a preparação do córtex ovariano para ovular em meses de elevados índices pluviométricos ao longo do ano. Logo, essa espécie procura ajustar seu período reprodutivo com as variações das condições ambientais, de forma que a reprodução ocorra em períodos mais favoráveis para manutenção da prole.


#10 - Estimation, morphometry and ultrastructure of ovarian preantral follicle population in agouti (Dasyprocta leporina)

Abstract in English:

The aim of this study was to characterize the preantral ovarian follicular population in agoutis (D. leporina) by estimating the number of follicles at each developmental category, and also describe the morphometry and the specific features of the follicle and the oocyte by using light and transmission electron microscopy. The length of each ovary was measured using a caliper rule, longitudinally sectioned into two halves and both were immediately fixed to perform the estimation of follicular population and ultrastructural analysis. The mean (±S.E.M.) population of follicular per pair of ovary was estimated at 4419.8±532.26 and 5397.52±574.91 for right and left ovaries, respectively, but no differences were observed between them. The diameters for follicles, oocyte and nuclei were: 18.62±3.40μm, 12.28±2.37μm and 6.10±0.93μm for primordial, 23.75±5.70μm, 14.22±3.00μm and 6.70±1.24μm for primary and 88.55±17.61μm, 52.85±17.56μm and 22.33±17.61μm for secondary follicles, respectively. The most of the follicles found belonged to the primordial category (86.63%), followed by primary (13.01%) and secondary (0.35%) one. Additionally, polyovular follicles were observed in all the animals and they represented 7.51% of the total follicles counted. The ultrastructural analysis showed that the oocyte presented a central and regular nuclei, displaying a homogenous mass. Among the organelles, the mitochondria were the most abundant and the oocyte Golgi apparatus was rarely observed. In conclusion, this work shows for the first time the characterization of the population of preantral follicles in the ovary of Dasyprocta leporina. Those information will be useful for further development and adaptation of biotechniques such as germplasm cryopreservation and in vitro gametes manipulation.

O objetivo deste trabalho foi caracterizar a população folicular ovariana pré-antral em cutias (D. leporina) estimando o número de folículos em cada categoria de desenvolvimento, e também descrever a morfometria e as características específicas do folículo e oócito usando microscopia de luz e eletrônica de transmissão. O comprimento de cada ovário foi medido utilizando um paquímetro, seccionados longitudinalmente em duas metades e ambos foram imediatamente fixados para realizar a estimativa da população folicular e análise ultraestrutural. A média (±S.E.M.) da população folicular por par de ovário foi estimada em 4419,8±532,26 e 5397,52±574,91 nos ovários direito e esquerdo, respectivamente, mas não foram observadas diferenças entre eles. Os diâmetros dos folículos, oócito e núcleos, respectivamente, foram: 18,62±3,40μm, 12,28±2.37μm e 6,10±0,93μm para primordial, 23,75±5,70μm, 14,22±3,00μm e 6,70±1,24μm para primário e 88,55±17,61μm, 52,85±17,56μm e 22,33±17,61μm de folículos secundários. A maioria dos folículos encontrados pertencia à categoria primordial (86,63%), seguido pelo primário (13,01%) e um secundário (0,35%). Adicionalmente, os folículos poliovulares foram observados em todos os animais e representavam 7,51% do total de folículos contados. A análise ultra-estrutural mostrou que o oócito apresentou núcleos centrais e regulares, exibindo uma massa homogênea. Dentre as organelas, as mitocôndrias foram as mais abundantes e o aparelho de Golgi do oócito foi raramente observado. Em conclusão, este trabalho mostra pela primeira vez a caracterização da população de folículos pré-antrais do ovário da Dasyprocta leporina. Essas informações serão úteis para o desenvolvimento e adaptação de biotécnicas, como a criopreservação de germoplasma e manipulação de gametas in vitro.
