#1 - Nervous form of listeriosis in buffaloes

Abstract in English:

Listeriosis is a disease that affects several animal species, including humans, and has three different forms of presentation: encephalic, reproductive, or septicemic. The nervous form is caused mainly by the bacterium Listeria monocytogenes. In Brazil, this disease has already been described in sheep, goats, and cattle. There are no reports of the disease in buffaloes in Brazil and worldwide. The objective of this study was to describe an outbreak of listeric meningoencephalitis in buffaloes in the state of Pará, Brazil. The outbreak occurred in a property located in the municipality of Bujaru, in the eastern Amazon, from May to July 2016. In a herd of 47 buffaloes, three animals (Cases 1, 2 and 3), aged <40 days, presented a neurological condition with locomotion difficulty characterized by paralysis of the four limbs, hypoesthesia, lateral recumbency, and death. Morbidity was 6.38% and lethality was 100%. At necropsy, no significant macroscopic lesions were found. Samples of the central nervous system were collected, fixed in 10% buffered formalin, and routinely processed for histopathological analysis. The main microscopic changes observed were unilateral microabscesses in the brainstem composed predominantly of mononuclear cells, with fewer polymorphonuclear cells, and perivascular cuffs composed mostly of mononuclear cells and few neutrophils. Samples of Cases 1 and 2 revealed Gram-positive bacteria in the areas of necrosis by the Gram’s stain technique. Samples of Case 1 were positive in immunohistochemistry for L. monocytogenes. Diagnosis of the nervous form of listeriosis was based on epidemiological data, clinical profile, and immunostaining for Listeria monocytogenes. Results showed that listeriosis should be considered in the differential diagnosis in buffaloes with nervous signs.

A listeriose é uma doença que afeta várias espécies animais, incluindo o homem, e possui três formas diferentes de apresentação: nervosa, abortiva ou septicêmica. A forma nervosa é causada principalmente pela bactéria Listeria monocytogenes. No Brasil a doença já foi descrita em bovinos, ovinos e caprinos, mas não foram encontrados relatos desta doença em búfalos no Brasil e no mundo. O objetivo deste trabalho foi descrever um surto de listeriose nervosa em búfalos no estado do Pará, Brasil. O surto ocorreu de maio a julho de 2016, em uma propriedade localizada no município de Bujaru, na Amazônia Oriental. Três bubalinos de um total de 47 animais (Casos 1, 2 e 3), menores de 40 dias, apresentaram um quadro clínico neurológico caracterizado por dificuldade de locomoção, paralisia dos quatro membros, diminuição da sensibilidade cutânea, decúbito lateral e morte. A morbidade foi de 6,38% e a letalidade de 100%. Na necropsia não foram encontradas lesões macroscópicas significativas. Amostras do sistema nervoso central foram coletadas e fixadas em formalina tamponada a 10% e processadas rotineiramente para análise histopatológica. As principais alterações microscópicas observadas foram microabscessos unilaterais no tronco encefálico, compostos predominantemente por células mononucleares, com menor número de polimorfonucleares, e manguitos perivasculares compostos predominantemente por células mononucleares e poucos neutrófilos. Amostras dos Casos 1 e 2 revelaram bactérias Gram positivas nas áreas de necrose na técnica de Gram. Amostras do Caso 1 resultaram positivas na imuno‑histoquímica para L. monocytogenes. O diagnóstico da forma nervosa da listeriose foi baseado nos dados epidemiológicos, no quadro clínico patológico e na imunomarcação para Listeria monocytogenes. Os resultados demostram que a listeriose deve ser considerada no diagnóstico diferencial em bubalinos com sinais nervosos.

A two-stage sampling was conducted, 10 farms were selected by probability proportional to size. A single visit was made to each farm. Samples from bovine faeces, feedstuffs, bulk tank milk, drinking water and soil from the entry and exit pens of the milking parlour were collected for bacteriological studies. PCR assays were used to confirm species and determine the serotype profile of L. monocytogenes isolates. AscI-pulsed-field gel electrophoresis was done to genetically compare them. Listeria spp. were isolated from eight of ten dairy farms, whereas L. monocytogenes in three of them. Serotype distribution in L. monocytogenes was as follows: 1/2a, three isolates; 4b, one isolate. L. monocytogenes or L. innocua excreted from clinically healthy milking cows was detected via faeces. In feedstuffs, only one L. monocytogenes 1/2a isolate from a pasture was obtained. The strain was identical by PFGE to an isolate 1/2a obtained from a pool of milking cow feces that grazed on this farm. No isolation of Listeria spp. was retrieved from the bulk tank milk or drinking water from any of the farms. Listeria innocua was detected in 13 feedstuffs and seven samples of soil from the entry and exit pens of the milking parlour. This is a first local study that confirms the presence of Listeria spp. including L. monocytogenes in healthy cattle and environment of pasture-based dairy farms. These results suggest the potential role that healthy cattle and their sub-products would play as a source of these agents for humans and/or others animals. More detailed studies that include genetic comparison of human and animal isolates are required in order to clearly establish the epidemiological relationship.

Foram coletadas amostras para cultura bacteriológica de matéria fecal bovina, além de alimentos, leite do tanque de resfriamento, água e solo na entrada e saída da sala de ordenha. Com os isolados de L. monocytogenes foi realizado PCR para a confirmação da espécie e determinação do perfil do serotipo. AscI-elctroforese em gel de campo pulsado foi realizado para compará-los geneticamente. Listeria spp. foram isoladas de oito de dez estabelecimentos, enquanto L. monocytogenes foram detectadas em três deles. A distribuição dos serotipos nos isolados de L. monocytogenes recuperados foi: 1/2a três isolados, 4b um isolado. Foram detectadas vacas leiteras clinicamente sadias &#8203;&#8203;que excretaram L. monocytogenes ou L. innocua nas fezes. Dos alimentos do gado houve só um isolamento de L. monocytogenes 1/2a em uma pastagem. Esta estirpe foi idêntica no PFGE a um isolado 1/2a obtido de uma “piscina” de fezes de vacas leiteiras do mesmo estabelecimento. Não houve isolamento de Listeria no leite do tanque de resfriamento ou na água de nenhum dos estabelecimentos. Listeria innocua foi detectada em 13 alimentos para o gado e sete amostras de solo na entrada e saída da sala de ordenha. Este parece ser o primeiro estudo local que confirma a presença de Listeria spp. incluindo L. monocytogenes em vacas leiteiras sadias e no meio ambiente de propriedades leiteiras com base alimentícia na pastagem. Esses resultados sugerem o potencial de vacas sadias e seus subprodutos como possível fonte desses agentes para humanos e/ou outros animais. São necessários estudos mais detalhados que incluem a comparação genética de isolados humanos e de animais para estabelecer claramente seu relacionamento epidemiológico.


#3 - Suppurative infectious diseases of the central nervous system in domestic ruminants, 37(8):820-828

E-mail: saulo.pavarini@ufrgs.br This study describes suppurative infectious diseases of the central nervous system (CNS) in domestic ruminants of southern Brazil. Reports from 3.274 cattle, 596 sheep and 391 goats were reviewed, of which 219 cattle, 21 sheep and 7 goats were diagnosed with central nervous system inflammatory diseases. Suppurative infectious diseases of the CNS corresponded to 54 cases (28 cattle, 19 sheep and 7 goats). The conditions observed consisted of listerial meningoencephalitis (8 sheep, 5 goats and 4 cattle), suppurative leptomeningitis and meningoencephalitis (14 cattle, 2 goats and 1 sheep), cerebral (6 cattle and 2 sheep), and spinal cord (7 sheep) abscesses, and basilar empyema (4 cattle and 1 sheep). Bacterial culture identified Listeria monocytogenes (9/54 cases), Escherichia coli (7/54 cases), Trueperella pyogenes (6/54 cases) and Proteus mirabilis (1/54 cases). All cases diagnosed as listeriosis through histopathology yielded positive immunostaining on immunohistochemistry, while 12/17 of the cases of suppurative leptomeningitis and meningoencephalitis presented positive immunostaining for Escherichia coli. Meningoencephalitis by L. monocytogenes was the main neurological disease in sheep and goats, followed by spinal cord abscesses in sheep. In cattle, leptomeningitis and suppurative meningoencephalitis was the most frequent neurological disease for the species, and E. coli was the main cause of these lesions. Basilar empyema, mainly diagnosed in cattle, is related to traumatic injuries, mainly in the nasal cavity, and the main etiologic agent was T. pyogenes.

Foram avaliados laudos de 3.274 bovinos, 596 ovinos e 391 caprinos, dos quais 219 bovinos, 21 ovinos e sete caprinos foram diagnosticados como doenças inflamatórias no sistema nervoso central. As doenças neurológicas infecciosas supurativas corresponderam a 54 casos (28 bovinos, 19 ovinos e sete caprinos). As enfermidades observadas foram meningoencefalite por Listeria monocytogenes (oito ovinos, cinco caprinos e quatro bovinos), leptomeningite e meningoencefalite supurativa (14 bovinos, dois caprinos e um ovino), abscessos cerebrais (seis bovinos e dois ovinos) e medulares (sete ovinos); e empiema basilar (quatro bovinos e um ovino). Através do isolamento bacteriano foram identificados: L. monocytogenes (9/54 casos), Echerichia coli (7/54 casos), Trueperella pyogenes (6/54 casos) e Proteus mirabilis (1/54 casos). Todos os casos diagnosticados como listeriose por histologia foram positivos na imuno-histoquímica para L. monocytogenes, e 12/17 casos de leptomeningite e meningoencefalite supurativa foram positivos na imuno-histoquímica para E. coli. A meningoencefalite por L. monocytogenes representou a principal enfermidade neurológica em ovinos e caprinos, seguido dos abscessos medulares em ovinos. A leptomeningite e meningoencefalite supurativa foi a doença neurológica supurativa mais frequente em bovinos e o principal agente causador da lesão foi E. coli. O empiema basilar, frequentemente, diagnosticado em bovinos, foi relacionado com lesões traumáticas, principalmente, de cavidade nasal e o principal agente causador foi T. pyogenes.


#4 - Molecular characterization of Listeria monocytogenes from beef samples and cattle slaughterhouses located in the Federal District, Brazil, 36(10):957-964

[Molecular characterization of Listeria monocytogenes from beef samples and cattle slaughterhouses located in the Federal District, Brazil.] Caracterização molecular de Listeria monocytogenes oriundas de cortes cárneos bovinos e de abatedouros frigoríficos de bovinos localizados no Distrito Federal, Brasil. Pesquisa Veterinária Brasileira 36(10):957-964. Faculdade de Agronomia e Medicina Veterinária, Universidade de Brasília, Brasília, DF 70910-900, Brazil. E-mail: joanamarchesini@gmail.com The aim of the study was the analysis of Listeria monocytogenes strains in beef samples as well as slaughterhouse environment, located in the Federal District, promote serotyping by polymerase chain reaction (PCR), perform antibiotic susceptibility and submit the strains to Pulsed-field gel electrophoresis (PFGE). A total of 125 beef samples were analyzed, 45 samples of carcasses swabs and 43 swab samples. It detected 13 strains of Listeria monocytogenes, 11 in beef samples. and 2 in slaughterhouse environment. No carcass swabs strains were isolated. Among the 13 strains of L. monocytogenes six strains of serotype 4b were found, five serotype 1/2c and two strains of serotype 1/2a. Among the 11 strains of L. monocytogenes found in beef, one (9.1%) strain showed resistance to erythromycin, one (9.1%) strain to gentamicin, one to ciprofloxacin (9.1%) and all strains (100%) were resistant to nalidixic acid. The two strains coming from the slaughterhouse drains, all (100%) were resistant to nalidixic acid and Sulfonamides. The analysis by pulsed field gel electrophoresis (PFGE) showed 13 different pulsotypes; they were grouped into three different clonal groups, coincidentally correlated with the three different serotypes found, what suggests a widespread dissemination of these profiles in the Federal District, Brazil.

Foram analisados um total de 125 cortes cárneos bovinos, 45 amostras de swabs de carcaças e 43 amostras de swabs em que foram detectados 13 cepas de Listeria monocytogenes, sendo 11 em cortes cárneos bovinos e 2 swabs de ambiente em um abatedouro frigorifico. Não foram isoladas cepas de swabs de carcaça. Dentre as 13 cepas de Listeria monocytogenes foram encontradas seis cepas do sorotipo 4b, cinco do sorotipo 1/2c e duas cepas do sorotipo 1/2a. Dentre as 11 cepas de L. monocytogenes encontradas em cortes cárneos bovino, uma (9,1%) cepa apresentou resistência a eritromicina, outra (9,1%) cepa a gentamicina e outra a ciprofloxacina (9,1%) e todas as cepas (100%) apresentaram resistência ao Ác. Nalidíxico. Das duas (2) cepas oriundas de ralos de abatedouro frigorífico, todas (100%) apresentaram resistência ao Ác. Nalidíxico e a sulfonamidas. A análise por eletroforese de campo pulsante (PFGE) demonstrou 13 diferentes pulsotipos, em que foram agrupados em 3 diferentes grupos clonais, que coincidentemente se correlacionavam com os 3 diferentes sorotipos encontrados sugerindo uma ampla disseminação desses perfis no Distrito Federal.

E-mail: aclcamara@yahoo.com.br In addition to listeriosis which is relatively common in ruminants, there are three other uncommon suppurative intracranial processes (SIP) identifiable in adult ungulates as brain abscess, basilar empyema and suppurative meningitis. The present paper reports the epidemiological, clinical, laboratorial, pathological and microbiological findings of 15 domestic ruminants with SIP. A total of 15 animals were selected (eight sheep, four cattle and three goats); with the definitive diagnoses of basilar empyema (n=3), brain abscess (n=1), listeriosis (n=5) and suppurative meningitis (n=6). Hematology revealed leukocytosis with inversion of the lymphocyte/ neutrophil ratio in 4 cases. In the majority of animals, cerebrospinal fluid (CSF) presented light yellow coloration and cloudy aspect due to neutrophilic pleocytosis (15 – 997 leukocytes/µL). Microbiological culture of CSF or central nervous system (CNS) fragments resulted on isolation of Trueperella (Arcanobacterium) pyogenes, Listeria monocytogenes, Escherichia coli and Stenotrophomonas sp. In a goat with thalamic abscess, microbiological assay was not performed, but Gram positive bacilli type bacteria were observed in histology. The diagnosis of these outbreaks was based on the association of epidemiological, clinical, pathological and bacteriological findings; reiterating that the infectious component remains an important cause of CNS disease in domestic ruminants and also shows the need for dissemination of information about the most effective preventive measures for the ranchers.

O presente trabalho tem como objetivo relatar os achados epidemiológicos, clínicos, laboratoriais, patológicos e microbiológicos de 15 ruminantes domésticos com PSI. O total de 15 animais foi selecionado (oito ovinos, quatro bovinos e três caprinos), com o diagnóstico definitivo de empiema basilar (n=3), abscesso cerebral (n=1), listeriose (n=5) e meningite supurativa (n=6). A hematologia revelou leucocitose com inversão da relação linfócito/ neutrófilo em quatro animais. Na maioria dos animais, a análise do líquido cefalorraquidiano (LCR) revelou coloração amarelado clara e aspecto turvo devido à pleocitose neutrofílica (15 – 997 leucócitos/µL). A cultura microbiológica de LCR ou de fragmentos do sistema nervoso central (SNC), permitiu o isolamento de Trueperella (Arcanobacterium) pyogenes, Listeria monocytogenes, Escherichia coli e Stenotrophomonas sp. No caprino com abscesso de tálamo, a cultura microbiológica não foi realizada, mas bactérias Gram positivas tipo bacilos foram observadas na histologia. O diagnóstico desses surtos foi baseado na associação dos achados epidemiológicos, clínicos, patológicos e bacteriológicos; reiterando que o componente infeccioso continua sendo uma causa importante de doença do SNC em ruminantes domésticos e também mostra a necessidade de disseminação da informação sobre as medidas preventivas mais eficazes para os criadores.

Clinical signs included head tilt, torticollis, nystagmus, staggering, falls and eventually recumbency and paddling, with stiff limbs. Treatment of Goat C with antibiotics resulted in a temporary remission of the clinical signs. The clinical courses where 5, 10 and 30 days respectively for Goats A, B and C. Goat A died spontaneously and the other two where euthanatized in extremis. No gross changes were observed. Histologic lesions were predominantly unilateral, with inflammatory and degenerative changes, extending from the medulla oblongata to the thalamus. In all 3 cases there were perivascular cuffings of one or more types of mononuclear cells (lymphocytes, plasma cells, activated macrophages) and occasionally neutrophils associated with areas of malacia where Gitter cells filled spaces of parenchymal loss. Additionally, Goat B had microabscesses in the medulla, pons, and mesencephalon and multifocal neutrophilic and lymphocytic infiltrates within the fascicles of the trigeminal nerve and in the cerebellar leptomeninges. In the brainstem of Goat C, there was multifocal granulomatous inflammation which included epithelioid macrophages and occasional multinucleated giant cells. Listeria sp antigen was detected by imunohistochemistry in routinely processed sections of mesencephalon from Goats A and C and of pons from Goat B.

In all 3 cases there were perivascular cuffings of one or more types of mononuclear cells (lymphocytes, plasma cells, activated macrophages) and occasionally neutrophils associated with areas of malacia where Gitter cells filled spaces of parenchymal loss. Additionally, Goat B had microabscesses in the medulla, pons, and mesencephalon and multifocal neutrophilic and lymphocytic infiltrates within the fascicles of the trigeminal nerve and in the cerebellar leptomeninges. In the brainstem of Goat C, there was multifocal granulomatous inflammation which included epithelioid macrophages and occasional multinucleated giant cells. Listeria sp antigen was detected by imunohistochemistry in routinely processed sections of mesencephalon from Goats A and C and of pons from Goat B.
