#1 - Molecular epidemiology of Clostridioides difficile obtained from fecal samples of wild animals in Brazil

Abstract in English:

Clostridioides difficile is a strictly anaerobic, spore-forming Gram-positive bacterium associated with diarrhea, known as C. difficile infection (CDI). In domestic animals, C. difficile is considered an important pathogen mostly in pigs and horses, but there are also reports in other domestic species. In wild animals, the epidemiology of C. difficile is largely unknown, and the role of the bacterium as a cause of diarrhea is unclear. The aim of this study was to determine the prevalence of C. difficile in the feces of wild animals referred to the Center of Medicine and Research in Wild Animals (CEMPAS). Fecal samples obtained from 100 animals of 34 different species were subjected to qPCR for the detection of the C. difficile 16S rRNA gene and two major toxin genes (tcdA and tcdB) and to anaerobic bacterial isolation. A total of 63 animals (63%) were positive for C. difficile by qPCR, and 16 isolates were recovered. The opossum (Didelphis spp.) had the highest number of positive animals in both tests (from 21 samples, 19 were qPCR positive, and four isolates were recovered). Three toxigenic strains (RT 002, 004, and 014), all previously described as infecting humans and animals, were isolated in the following species: bearded dragon (Pogona vitticeps), pampas fox (Lycalopex vetulus), and marmoset (Callithrix sp.). The presence of C. difficile in the feces of wild animals highlights the importance of wildlife as potential carriers of infection for production animals or humans.

O Clostridioides difficile é uma bactéria Gram-positiva estritamente anaeróbica e formadora de esporos, associada à diarreia e conhecida como infecção por C. difficile (CID). Em animais domésticos, o C. difficile é considerado um patógeno importante principalmente em porcos e cavalos, mas também há relatos em outras espécies domésticas. Em animais selvagens, a epidemiologia do C. difficile é amplamente desconhecida, e o papel da bactéria como causa de diarreia não está claro. O objetivo deste estudo foi determinar a prevalência do C. difficile nas fezes de animais selvagens encaminhados ao Centro de Medicina e Pesquisa em Animais Selvagens (CEMPAS). Amostras de fezes obtidas de 100 animais de 34 espécies diferentes foram submetidas à qPCR para a detecção do gene 16S rRNA do C. difficile e dois principais genes de toxina (tcdA e tcdB), além de isolamento bacteriano anaeróbico. Um total de 63 animais (63%) foram positivos para C. difficile por qPCR, e 16 isolados foram recuperados. O gambá (Didelphis spp.) apresentou o maior número de animais positivos em ambos os testes (de 21 amostras, 19 foram positivas na qPCR, e quatro isolados foram recuperados). Três cepas toxigênicas (RT 002, 004 e 014), todas previamente descritas como infectando humanos e animais, foram isoladas nas seguintes espécies: dragão barbado (Pogona vitticeps), raposa-pampas (Lycalopex vetulus) e sagui (Callithrix sp.). A presença de C. difficile nas fezes de animais selvagens destaca a importância da vida selvagem como potencial portadora de infecção para animais de produção ou seres humanos.


#2 - Occurrence of Salmonella spp. in fecal samples from foals with and without diarrhea in the state of São Paulo: microbiological diagnosis, antimicrobial susceptibility profile, and molecular detection

Abstract in English:

The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.

O presente estudo investigou a ocorrência de Salmonella spp. em fezes de 200 potros com até um ano de idade (100 com sinais clínicos de diarreia e 100 sem sinais clínicos de diarreia), utilizando as técnicas de cultivo bacteriológico e PCR em tempo real (qPCR) pelos métodos de corante fluorescente (SYBR® Green) e sonda específica (Taqman®) para a detecção do gene invA com e sem etapa de pré-enriquecimento seletivo em caldo de tetrationato. O cultivo bacteriológico revelou 15% (n=30) de animais positivos (21 animais com diarreia e nove animais sem diarreia). Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multirresistência foi constatada em 43,33% (n=13) dos isolados, sendo um isolado obtido de animal sem diarreia e 12 isolados de animais com diarreia. Todas as técnicas de qPCR empregadas no estudo apresentaram maior número de amostras classificadas como positivas para Salmonella spp. comparadas ao cultivo bacteriológico de fezes. Adicionalmente, em todas as técnicas de qPCR houve maior número de animais detectados como positivos no grupo de animais com diarreia em relação aos animais sem diarreia. Os resultados confirmaram a utilidade do método qPCR sem a etapa de pré-enriquecimento em tetrationato, como um teste rápido para detecção de Salmonella spp. em animais portadores ou em animais com sinais clínicos de diarreia. O cultivo bacteriológico deve ser associado para a realização do teste de sensibilidade aos antimicrobianos e sorotipificação. O isolamento de Salmonella spp. em nove animais sem diarreia, confirma a importância dos animais portadores assintomáticos na epidemiologia da doença. A multirresistência observada evidencia a importância do uso racional de antimicrobianos em equinos e a importância da adoção de protocolos de biossegurança que sejam eficazes para controlar a disseminação de infecções entre animais e a transmissão zoonótica nas fazendas.


#3 - Stx1 and Stx2 subtyping and antimicrobial resistance in Shiga toxin-producing Escherichia coli (STEC) isolates from cattle and sheep feces in the Southeastern region of the State of Goiás, Brazil

Abstract in English:

imicrobial resistance in 106 Shiga toxin-producing Escherichia coli (STEC) strains isolated from cattle and sheep feces. PCR was used to determine the subtypes, and the disk-diffusion method was used to evaluate the antimicrobial resistance. Ten antibiotics from five different classes were tested. Among the isolates of bovine origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2d) were identified. In isolates of sheep origin, two subtypes of Stx1 (Stx1a and Stx1c), and four subtypes of Stx2 (Stx2a, Stx2b, Stx2c, and Stx2 g) were identified. The results obtained suggest the presence of high diversity in Stx1 and Stx2 genes. Further, 96.6% (57/59) of bovine fecal strains and 89.4% (42/47) of sheep fecal strains showed resistance to at least one tested antibiotic. In both animal species, most strains were multidrug-resistant (MDR) (67.8% in cattle and 59.6% in sheep), with no significant difference between host animals. Adult animals were eight times more likely to have STEC with greater pathogenic potential. STEC with the highest pathogenic potential were three times more likely to be multidrug-resistant than STEC with the lowest pathogenic potential. The data reported in this study suggests the occurrence of strains with high potential pathogenicity in the region studied. Therefore, the ruminants of this region are carriers of strains that can cause infections in humans.

O presente estudo teve como objetivo subtipar os genes Stx1 e Stx2 e caracterizar a resistência antimicrobiana em 106 isolados de Escherichia coli produtoras de toxinas Shiga (STEC) isoladas de fezes de bovinos e ovinos. A PCR foi utilizada para determinar os subtipos e o método de difusão em disco foi utilizado para avaliar a resistência antimicrobiana. Dez antibióticos de cinco classes diferentes foram testados. Entre os isolados de origem bovina, foram identificados dois subtipos de Stx1 (Stx1a e Stx1c) e quatro subtipos de Stx2 (Stx2a, Stx2b, Stx2c e Stx2d). Nos isolados de origem ovina, foram identificados dois subtipos de Stx1 (Stx1a e Stx1c) e quatro subtipos de Stx2 (Stx2a, Stx2b, Stx2c e Stx2g). Os resultados obtidos sugerem a presença de alta variabilidade nos genes Stx1 e Stx2. Além disso, 96,6% (57/59) dos isolados fecais de bovinos e 89,4% (42/47) dos isolados de ovinos mostraram resistência a pelo menos um antibiótico testado. Em ambas as espécies animais, a maioria das cepas foi multirresistente (MDR) (67,8% em bovinos e 59,6% em ovinos), sem diferença significativa entre as espécies animais do reservatório. Os animais adultos tiveram oito vezes mais chances de apresentar STEC com maior potencial patogênico. STEC com o maior potencial patogênico teve três vezes mais chances de ser multirresistente do que o STEC com o menor potencial patogênico. Os dados relatados neste estudo sugerem a ocorrência de cepas com alto potencial de patogenicidade na região estudada. Portanto, os ruminantes dessa região são hospedeiros de isolados que podem causar infecções em humanos.


#4 - Investigation of Norovirus genogroups (GI, GII and GIV) in stool of pet dogs with diarrhea

Abstract in English:

In this study, we searched the existence of human norovirus (NoV) GI, GII and GIV in the stool of 128 pet dogs with diarrhea, of different sex, age and breed, in Burdur, Turkey, using Real-Time PCR method. Human NoV GII was found in only 5 of the 128 dog stool samples (3.91%). It was discovered that human NoV existed most in crossbreed, female and aged 24 months or over dogs. These dogs found with human NoV GII were either bought from pet shops, stray dogs or taken as puppy of another pet dog. The sheltering conditions of these dogs were moderate and they were fed with home food residue and dry food. It was also found that most of them were vaccinated and had certain walking sites. The owners of the animals detected with infection generally did not have the habit of washing their hands or changing their clothes before or after caring their pets. We strongly advice that dog owners’ personal hygiene, the necessity of changing their clothes during their contact with animals, the environment provided for the dog, the sensitivity in caring, use of strong and effective disinfectant, keeping the dogs away from toilets and sewerage systems, as well as not feeding them with food residues are crucial issues in dogs’ care. Owners of the dogs with NoV GII were middle aged or elderly people, male, and there were no children in their houses. As these dogs are treated like the owner’s child, it is assumed that they could be transmitted with NoV GII as a result of close interaction with their owner.

Neste estudo pesquisamos a existência de norovírus humano (NoV) GI, GII e GIV nas fezes de 128 cães com diarréia, de diferentes sexos, idades e raças, em Burdur, Turquia, utilizando o método de PCR em tempo real. NoV GII humano foi encontrado em apenas 5 das 128 amostras de fezes de cães (3,91%). Foi descoberta NoV humana, principalmente em cruzamentos, fêmeas e cães com idade igual ou superior a 24 meses. Os  cães encontrados com NoV GII humano foram comprados de lojas de animais, eram vira-latas ou foram tomados como filhotes de outro cão de estimação. As condições de abrigo desses cães eram moderadas. Os cães foram alimentados com restos de comida caseira e comida seca. Verificou-se também que a maioria dos animais foi vacinada e tinham locais adequados para caminhada. Os donos dos animais detectados com infecção geralmente não tinham o hábito de lavar as mãos ou trocar de roupa antes ou depois de cuidar de seus animais de estimação. Aconselhamos que a higiene pessoal dos donos, a necessidade de trocar de roupa durante o contato com animais, o ambiente fornecido para o cão, a sensibilidade no cuidado, o uso de desinfetantes eficazes, manter os cães longe de banheiros e esgotos, assim como evitar alimentá-los com resíduos alimentares, são questões cruciais no cuidado dos cães. Os proprietários dos cães com NoV GII são de meia-idade ou idosos, a maioria do sexo masculino, e não havia crianças em suas casas. Como esses cães são tratados como um filho, presume-se que eles foram infectados com o NoV GII como resultado de uma interação próxima com o proprietário.

E-mail: cmatto@mgap.gub.uy The aim of the study was to detect Listeria spp., particularly Listeria monocytogenes, in cattle and environment of pasture based dairy farms in Paysandú, Uruguay. A two-stage sampling was conducted, 10 farms were selected by probability proportional to size. A single visit was made to each farm. Samples from bovine faeces, feedstuffs, bulk tank milk, drinking water and soil from the entry and exit pens of the milking parlour were collected for bacteriological studies. PCR assays were used to confirm species and determine the serotype profile of L. monocytogenes isolates. AscI-pulsed-field gel electrophoresis was done to genetically compare them. Listeria spp. were isolated from eight of ten dairy farms, whereas L. monocytogenes in three of them. Serotype distribution in L. monocytogenes was as follows: 1/2a, three isolates; 4b, one isolate. L. monocytogenes or L. innocua excreted from clinically healthy milking cows was detected via faeces. In feedstuffs, only one L. monocytogenes 1/2a isolate from a pasture was obtained. The strain was identical by PFGE to an isolate 1/2a obtained from a pool of milking cow feces that grazed on this farm. No isolation of Listeria spp. was retrieved from the bulk tank milk or drinking water from any of the farms. Listeria innocua was detected in 13 feedstuffs and seven samples of soil from the entry and exit pens of the milking parlour. This is a first local study that confirms the presence of Listeria spp. including L. monocytogenes in healthy cattle and environment of pasture-based dairy farms. These results suggest the potential role that healthy cattle and their sub-products would play as a source of these agents for humans and/or others animals. More detailed studies that include genetic comparison of human and animal isolates are required in order to clearly establish the epidemiological relationship.

Com os isolados de L. monocytogenes foi realizado PCR para a confirmação da espécie e determinação do perfil do serotipo. AscI-elctroforese em gel de campo pulsado foi realizado para compará-los geneticamente. Listeria spp. foram isoladas de oito de dez estabelecimentos, enquanto L. monocytogenes foram detectadas em três deles. A distribuição dos serotipos nos isolados de L. monocytogenes recuperados foi: 1/2a três isolados, 4b um isolado. Foram detectadas vacas leiteras clinicamente sadias ​​que excretaram L. monocytogenes ou L. innocua nas fezes. Dos alimentos do gado houve só um isolamento de L. monocytogenes 1/2a em uma pastagem. Esta estirpe foi idêntica no PFGE a um isolado 1/2a obtido de uma “piscina” de fezes de vacas leiteiras do mesmo estabelecimento. Não houve isolamento de Listeria no leite do tanque de resfriamento ou na água de nenhum dos estabelecimentos. Listeria innocua foi detectada em 13 alimentos para o gado e sete amostras de solo na entrada e saída da sala de ordenha. Este parece ser o primeiro estudo local que confirma a presença de Listeria spp. incluindo L. monocytogenes em vacas leiteiras sadias e no meio ambiente de propriedades leiteiras com base alimentícia na pastagem. Esses resultados sugerem o potencial de vacas sadias e seus subprodutos como possível fonte desses agentes para humanos e/ou outros animais. São necessários estudos mais detalhados que incluem a comparação genética de isolados humanos e de animais para estabelecer claramente seu relacionamento epidemiológico.


#6 - Detection of coronavirus in diarrhea samples from water buffaloes (Bubalus bubalis) raised in the State of São Paulo, Brazil, 37(8):802-804

E-mail: lgregory@usp.br Neonatal diarrhea can be a consequence of bacterial, endoparasite and viral infections. Although virus involved in diarrhea is frequently studied in cattle herds, there is lack of studies in Brazilian buffalo herds. The aim of this study was evaluate the presence of rotavirus and coronavirus in diarrhea samples of buffaloes (Bubalus buballis) raised on eight farms in Pariquera-açu, Registro, Pirassununga, Dourado, São João da Boa Vista e Congonhas, State of São Paulo, Brazil. We collected 40 diarrhea samples from water buffalo calves. While coronavirus was detected using nested polymerase chain reaction, rotavirus was detected using Polyacrylamide Gel Electrophoresis (PAGE) with silver stain. Rotavirus was not detected, while two samples (2/40, 5.0%) were positive for coronavirus. Although we did not detect rotavirus, a low percentage of coronavirus was observed; possible interference of these viruses in the development of diarrhea should not be discarded. Considering the lack of literature about diarrhea in water buffalo calves, particularly the one related with coronavirus, our results encourage new studies to enhance buffalo health in our country.

O objetivo deste trabalho foi avaliar a presença de rotavírus e coronavírus em amostras de fezes diarreicas de búfalos (Bubalus buballis) criados em oito propriedades localizadas em Pariquera-açu, Registro, Pirassununga, Dourado, São João da Boa Vista e Congonhas, Estado de São Paulo. Foram coletadas 40 amostras de fezes diarreicas de bezerros búfalos (Bubalus bubalis). A detecção de coronavírus foi realizada pela nested-PCR, enquanto que a detecção de rotavírus foi realizada pela Eletroforese em Gel de Poliacrilamida (PAGE) com coloração com nitrato de prata. Enquanto rotavírus não foi identificado, duas amostras (2/40, 5,0%) foram positivas para coronavírus. Embora no presente trabalho tenha havido baixa detecção de coronavírus e a ausência de rotavírus nos rebanhos estudados, a possível interferência desses vírus no desenvolvimento dos quadros diarreicos não deve ser descartada. Considerando o escasso material encontrado na literatura a respeito da diarreia em bezerros búfalos, principalmente aquele relativo ao coronavírus, nossos resultados são um incentivo para que novos estudos sejam realizados para impulsionar o desenvolvimento da bubalinocultura em nosso país.

E-mail: josepaes@fmvz.unesp.br Salmonellosis is a foodborne disease caused by bacteria of the genus Salmonella, being pigs and pork-products potentially important for its occurrence. In recent decades, some serovars of Salmonella have shown increase of resistance to conventional antimicrobials used in human and animal therapy, with serious risks for public health. The aim of this study was to evaluate feces (n=50), mediastinal (n=50), mesenteric (n=50) and mandibular (n=50) lymph nodes obtained from slaughter houses for Salmonella spp. Positive samples were serotyped and subjected to an in vitro antimicrobial susceptibility test, including the extended-spectrum beta-lactamase (ESBL) production. Salmonella species were identified in 10% (20/200) of total samples. From these, 20% (10/50) were identified in the submandibular lymph nodes, 18% (9/50) in the mesenteric lymph nodes, 2% (1/50) in feces and 0% (0/50) in the mediastinal lymph nodes. The serotypes found were Salonella Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg and S. Derby with 5% (5% each). All strains showed resistance to at least one antimicrobial; 90% were resistant to four or more antimicrobials, and 15% were multidrug-resistant. Resistance to ciprofloxacin, tetracycline and nalidixic acid was particularly prevalent amongst the tested serovars. Here, we highlighted the impact of pigs in the epidemiological chain of salmonellosis in domestic animals and humans, as well as the high antimicrobial resistance rates of Salmonella strains, reinforcing the necessity for responsible use of antimicrobials for animals as an emergent One Health issue, and to keep these drugs for human therapy approaches.

E-mail: josepaes@fmvz.unesp.br Nas últimas décadas, o aumento de cepas circulante de Salmonella concomitantemente a resistência microbiana tem despertado a preocupação dos órgãos de Saúde Pública. Deste modo, o objetivo do presente trabalho foi pesquisar a presença de Salmonella a partir de fezes (n=50), linfonodos mediastinos (n=50), mesentéricos (n=50) e submandibular (n=50) oriundos de um abatedouro suíno. As cepas isoladas foram sorotipadas e testadas quanto a resistência antimicrobiana. A presença de Salmonella isolada foram em 10% (20/200) do total de amostras, sendo 20% dos linfonodos submandibulares, 18% dos linfonodos mesentéricos e 2% das fezes. Os sorotipos encontrados foram S. Typhimurium (55%), S. enterica subsp. enterica 4,5,12: i: - (35%), S. Brandenburg (5%) e S. Derby (5%). Todas a cepas apresentaram resistência a pelo menos um antimicrobiano testado, sendo 90% resistente pelo menos quatro antimicrobianos. Destes, 15% foram classificadas como multidrogas resistentes. Os antimicrobianos mais resistentes entre os sorovares isolados foram a ciprofloxacina, tetraciclina e o ácido nalidixico. A presença de cepas de Salmonella resistente a antimicrobianos na espécie suína tem gerado um grande impacto epidemiológico entre homem e animal, reforçando cada vez mais a necessidade do uso adequado de drogas principalmente relacionado com o tema “One Health”.

However, this method is less sensitive and requires months of incubation before colony growth occurs. In this study, culture method was used on sheep faeces to diagnose paratuber-culosis in animals with compatible signs of the disease. A comparison of two culture media used to isolation was also investigated. Culture was positive in 2.0% of faecal samples. Isolation was obtained using Löwenstein Jensen® with mycobactin® J, and the Middlebrook® 7H11 with OADC®. The Löwenstein Jensen® with mycobactin® J was that provided highest amount of isolations. The percentages of isolation in each culture media were 2.0% (6/300) to Löwenstein Jensen® with micobactina J, and 1.0% (3/300) to Middlebrook® 7H11/OADC. The three positive samples in Middlebrook® 7H11/OADC were also positive in Löwenstein Jensen® with micobactina J. In the Middlebrook® 7H11/OADC alone there was no sample growth. The results of this study suggest that culture media of Löwenstein-Jensen® with micobactina® J is more effective for the isolation of sheep strains in Portugal.

#9 - Occurrence of Salmonella sp in finishing pigs in Rio Grande do Sul, Brazil, 22(3):104-108

[Occurrence of Salmonella sp in finishing pigs in Rio Grande do Sul, Brazil.] Ocorrência de Salmonella sp em suínos de terminação no Rio Grande do Sul. Pesquisa Veterinária Brasileira 22(3):104-108. Depto Medicina Veterinária Preventiva, Faculdade de Veterinária, UFRGS, Av. Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. Pen feces samples were taken in ten finishing farms in Rio Grande do Sul (Brazil) and submitted to Salmonella isolation protocol. Two farms negative and one positive in the preliminary screening were chosen, and feces from 25 randomly selected pigs in each of them were individually collected. The sarne animals were later sampled (rectal swab, intestinal content and mesenteric lymph nodes) at the slaughterhouse. After the introduction of new animals into farm Nl and Pl, other 25 pigs on each farm were examined as described above. In the preliminary screening, Salmonella was isolated from pen feces samples of 3 of the 10 investigated farms. Two of these herds showed a high level contamination. Eight Salmonella serotypes (Agona, Bredeney, Lexington, London, Mbandaka, Panama, Schwartzengrund, Salmonella sp) were found, with serotypes Agona and Bredeney being the most frequent. When individual animals were sampled, Salmonella was isolated in all selected farms. Salmonella was isolated in 6.4% of feces collected on the farm, 5.3% of intestinal contents and 5.6% of lymph nodes. Antibiogram testing of the isolated strains showed 97.8% resistance to sulphonamides, 82.6% to streptomycin, 36.9% to tetracyclin and 15.2% to sulfazotrim.

Foram colhidas amostras de fezes de lote de suínos em 10 granjas terminadoras do Estado do Rio Grande do Sul para pesquisa de Salmonella sp. Duas granjas consideradas negativas (Nl e N2) e uma positiva (Pl) nesta primeira etapa foram escolhidas para colheita de amostras de fezes individuais de 25 animais escolhidos aleatoriamente. No abatedouro foram coletados swab retal, conteúdo intestinal e linfonodos mesentéricos dos mesmos animais amostrados na granja. Após a introdução de novos animais nas granjas Nl e Pl, outros 25 animais foram amostrados em cada granja, da mesma forma descrita acima. Três granjas tiveram amostras de fezes de lote positivas, sendo que em duas foi constatado um alto nível de contaminação. Foram encontrados 8 sorotipos de Salmonella (Agona, Bredeney, Lexington, London, Mbandaka, Panama, Schwartzengrund, Salmonella sp), sendo os sorotipos Agona e Bredeney os mais encontrados. Na colheita individual realizada, todas as granjas amostradas foram positivas. Em 6,4% das amostras de fezes colhidas na granja, 5,3% das amostras de conteúdo intestinal e 5,6% dos linfonodos mesentéricos foi possível isolar Salmonella. O antibiograma das linhagens de Salmonella isoladas demonstrou 97,8% de resistência à sulfonamida, 82,6% à estreptomicina, 36,9% à tetraciclina e 15,2% à sulfazotrim.


#10 - Emergence of Haematobia irritans in cattle dung pats in Seropédica county, Rio de janeiro, 21(2):77-80

[Emergence of Haematobia irritans in cattle dung pats in Seropédica county, Rio de janeiro] Emergência de Haematobia irritans em fezes bovinas no município de Seropédica, Rio de Janeiro. Pesquisa Veterinária Brasileira 21(2):77-80. Depto Parasitologia Animal, Univ. Fed. Rural do Rio de janeiro, Seropédica, RJ 23851-970, Brazil. This studywas carried out in order to investigate the emergence of adult flies of Haematobia irritans in cattle dung maintained in the field and in the laboratory, as well as other flies associated with dung pats. Two groups of cattle dung were used, one in the field and another in the laboratory, each group consisting of five fresh dung pats ofmedium size (about 30 cm in diameter), opaque appearance, presence of a thin externai crust· and firm consistency. The dung pats were covered with emergence cages in pyramidal format, presenting a hole in the superior part where a removable flask was coupled, and the substitution of the dung was done every 2 weeks. A total of 355 specimens of H. irritans was collected, 151 of which were obtained in the field and 204 specimens in the laboratory. These differences probably were dueto smaller temperature variations in the laboratory than in the field, besides the action of arthropodal predators, competitors and parasites of horn fly pupae that probably interfered in H. irritans emergence in cattle dung under field conditions. A higher number of females were observed in both conditions. Besides H. irritans, there were collected other flies associated with bovine manure belonging to the following families: Aulacigastridae, Muscidae, Psychodidae, Sarcophagidae, Sepsidae, Tachinidae and Ulidiidae. Sepsidae was the most abundant, with 5,224 specimens of the total of 8,928 flies collected, followed by Sarcophagidae with 2,235, Muscidae with 1,357, Aulacigastridae with 54, Psychodidae with 46, Ulidiidae with 6 and Tachinidae with 5 specimens.

Foram utilizados dois grupos de fezes bovinas, um à campo e outro em laboratório, sendo cada grupo formado por cinco bolos fecais frescos de tamanho mediano (cerca de 30 cm de diâmetro), aparência opaca, cor esverdeada, presença de fina crosta externa e consistência firme. As fezes foram cobertas com gaiolas de emergência de formato piramidal, apresentando na parte superior um orifício onde foi acoplado um frasco removível; a substituição das fezes foi realizada quinzenalmente. Foram coletados um total de 355 espécimens de H. irritans, sendo 151 provenientes das gaiolas mantidas no campo e 204 em laboratório. Estas diferenças deveram-se provavelmente à menores oscilações de temperatura verificadas no laboratório do que àquelas ocorridas no campo, além da ação de artrópodes predadores, competidores e parasitóides que, provavelmente, interferiram na emergência de H. irritans nas fezes bovinas mantidas no campo. Foi observado maior número de fêmeas em comparação com machos em ambas as condições investigadas. Além de H. irritans, obteve-se outros dípteros associados às fezes bovinas, pertencentes as seguintes famílias: Aulacigastridae, Muscidae, Psychodidae, Sarcophagidae, Sepsidae, Tachinidae e Ulidiidae.
