E-mail: rinaldo.mota@hotmail.com The objective of the investigation was to evaluate the in vitro antimicrobial sensibility of 291 isolates of Staphylococcus spp., taken from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife (A), Agreste (B) and Zona da Mata (C) in the state of Pernambuco, Brazil. From the 291 isolates, 170 (58.4%) were identified as negative coagulase Staphylococcus (SCN), 84 (28.9%) as Staphylococcus aureus, and 37 (12.7%) as positive coagulase Staphylococcus (SCP). To study sensitivity to antimicrobials, the diffusion in disks method was used with 16 antimicrobial drugs commonly employed in the treatment of mastitis. The most efficient antibiotic in vitro was the combination of neomicine + bacitracine + tetracycline with percentages of 98.4%, 99.3%, and 89.7% for the A, B, and C regions, respectively. The least efficient was ampicillin, which was resistant to 56.5% of the isolates taken from region A, 72.8% from region B, and 71.8% from region C. These results indicate the need for periodic testing of sensitivity in vitro, as these variations can compromise the treatment of animals as well as control programs for bovine mastitis caused by Staphylococcus spp.

All samples were processed at the Laboratory of Bacteriology, Department of Preventive Veterinary Medicine, UFSM. From 51 bacteria isolated from jundiá fishes (Rhamdia quelen) 51 (100%) were sensitive to gentamycin, 49 (96,08%) to sulphazotrin, 47(92,16%) to chloramphenicol, 43 (84,31%) to tetracylin, 43 (84,31%) to naldixic acid, 31 (60,78%) to nitrofurantoin, 22 (43,14%) to erytromycin, 22 (43,14%) to ampicillin, 15 (29,41%) spiramycin, 13 (25,50%) to cholystin, and 5 (3%) to penicillin G. With exception of an isolate of Staphylococcus spp., the bacteria analyzed in the present study were resistant to one or more antimicrobial agents tested. Knowledge of the sensitivity profile of bacteria involved in infectious processes in fish will allow rational antimicrobial treatment that will contribute to the control of fish diseases in Rhamdia quelen without causing great risks to public health and the environment.

The commercial drugs used were moxidectin 0.2%, albendazole, levamisol chlorhydrate, ivermectin and an aqueous extract of the purgative potato (Operculina hamiltonii). The reduction test was applied through egg counts per gram of feces and larval culture to evaluate resistance. The fecal samples were collected in the day when medication was accomplished (day base), at 7, 14 and 21 days after treatment. The following results for the reduction of Trichostrongyloidea eggs were obtained: Treatment of the females with Moxidectina reduced the egg counts to 92.8, 88.7 and 89.8%, of the males to 92.6, 96.2 and 98.1%; with Levamisol of the females to 96, 97.1 and 91%, of the males to 85.7, 94,2 and 100%; with Albendazol of the females 65, 60.3 and 75.4%, of the males 88.8, 88.8 and 55.5%; with Ivermectin of the females to 92.2, 68.6 and 70.6%, of the males to 41.7, 73.6 and 59.7%; and with the purgative potato of the females to 31.8, 34.1 and 49.4%, of the males to 61.5, 80.7 and 50%. In larval culture Haemonchus, followed by Bunostomum, Trichostrongylus, and Oesophagostomum, were identified even after treatments.

#24 - Comparison of the sensibility of cattle and buffaloes to poisoning by Palicourea marcgravii (Rubiaceae), 23(4):167-172

[Comparison of the sensibility of cattle and buffaloes to poisoning by Palicourea marcgravii (Rubiaceae).] Comparação da sensibilidade de bovinos e búfalos à intoxicação por Palicourea marcgravii (Rubiaceae). Pesquisa Veterinária Brasileira 23(4): 167-172. Escola de Medicina Veterinária, Campus Castanhal, Universidade Federal do Pará, Rua Maximino Porpino 1000, Castanhal, PA 68743- 080, Brazil. E-mail: diomedes@ufpa.br In order to verify if buffaloes are more resistant than cattle to poisoning by Palicourea marcgravii, the plant was given per os simultaneously to buffaloes and cattle. Seven buffaloes and three head of cattle were used. Doses of 0.5, 1.0 and 2.0 g/kg did not cause signs of poisoning in buffaloes; but doses of 3.0, 4.0 and 6.0 g/kg caused death. In cattle 0.25 g/kg did not cause symptoms; but 0.5 and 2.0 g/kg were lethal. The influence of exercise on the onset of symptoms, the time which elapsed from the beginning of the administration of the plant to the appearance of symptoms, and the symptoms themselves, were similar in the two animal species. The clinical course was longer in buffaloes; whilst in cattle the period between the onset of severe symptoms and death lasted 9 to 17 minutes; in buffaloes it varied from 10 minutes to 1 hour 28 minutes. It can be concluded, that buffaloes are about six times more resistant than cattle to the toxic action of P. maracgravii. The lower incidence of deaths in buffaloes by poisonous plants in the Amazon region is, at least in part, due the lower sensibility of buffaloes to poisoning by this plant. Another factor responsible for the smaller number of buffaloes which die due to plant poisoning in the Amazon region, is possibly that buffaloes prefer the Jowlands, e.g. temporarily flooded areas, which are the habitat of Arrabidaea bilabiata, the second most important poisonous plant of the Amazon region. This plant is less poisonous than P. marcgravii, and its habitat are the higher not flooded areas. So it would be safer, in order to diminish losses, to breed buffaloes instead of cattle in areas where P. marcgravii occurs. The reason for the greater resistance of buffaloes should be further investigated as a possible prophylactic measure to avoid poisoning by P. marcgravii in cattle. On the other hand, Australian investigators modified genetically the ruminai bacteria Butyrivibrio fibrisolvens by introducing a gene, isolated from a Moraxella sp, which codifies a dehalogenase, capable of hydrolyzing fluoroacetate, the poisonous principle of P. marcgravii. Transferring the genetically modified B. fibrisolvens into the rumen of animal, which consume plants that contain fluoroacetate, would be a viable method to control that poisoning through a ruminal detoxifying process for the toxin. Through preliminary contacts, the director of the consorcium in Australia, responsible for the modification of the bacteria, declared that he is interested to sell the technology to our country, but it would be necessary to know if it is permitted to introduce into Brazil the genetically modified bacteria. If allowed, it would be necessary to perform investigations on the viability and the methodology for using this bacteria to control P. marcgravii poisoning in our country.

E-mail: diomedes@ufpa.br O presente estudo foi realizado para verificar se búfalos são mais resistentes do que bovinos à ação tóxica de Palicourea marcgravii, mediante a administração da planta por via oral, simultaneamente, a bovinos e a búfalos. Foram usados sete búfalos e três bovinos. Verificou-se que em búfalos doses de 0,5 g/kg, 1,0 g/kg e 2,0 g/kg não causaram sintomas de intoxicação. As doses de 3,0 g/kg, 4,0 g/kg e 6,0 g/kg causaram a morte dos búfalos. Em bovinos, a dose de 0,25g/kg não causou sinais clínicos de intoxicação, enquanto que doses de 0,5 g/kg e 2,0 g/kg causaram a morte. A influência do exercício sobre o aparecimento dos sintomas, o prazo decorrido desde o começo da administração da planta até o início de sintomas, e os próprios sintomas, foram semelhantes nas duas espécies animais. O curso clínico foi mais longo nos búfalos. Enquanto nos bovinos o período entre o aparecimento dos sintomas graves e a morte foi de 9 a 17 minutos, nos búfalos variou de 10 minutos a 1 hora e 28 minutos. Pode se concluir que os bubalinos são aproximadamente seis vezes mais resistentes do que os bovinos à ação tóxica de P. marcgravii. O menor índice de mortes pela ação de plantas tóxicas na Amazônia em búfalos é, pelo menos em parte, devido à maior resistência do búfalo à intoxicação por essa planta. Outro fator responsável pelo menor número de mortes em búfalos pela intoxicação por plantas, na Amazônia, poderia ser que os búfalos preferem a várzea, que é o habitat de Arrabidaea bilabiata, a segunda planta tóxica mais importante da Amazônia, menos tóxica do que P. marcgravii, e com habitat na terra firme. Em áreas onde ocorre P. marcgravii seria mais prudente, para diminuir os prejuízos, criar búfalos em lugar de bovinos. A causa dessa maior resistência do búfalo merece ser investigada para a eventual elaboração de métodos profiláticos da intoxicação por P. marcgravii em bovinos. Por outro lado, pesquisadores australianos modificaram geneticamente a bactéria ruminal Butyrivibrio fibrisolvens, mediante a introdução de um gene, isolado de Moraxella sp, que codifica uma dehalogenase, capaz de hidro-lizar fluoroacetato. A transferência de B.fibrisolvens geneticamente modificado para o rúmen de animais ingerindo plantas que contêm fluoroacetato, como é o caso de P. marcgravii, seria um método viável para o controle da intoxicação mediante a detoxificação ruminal do princípio ativo. Em contatos preliminares o diretor responsável do consórcio na Austrália responsável pela modificação da bactéria, declarou o interesse em vender a tecnologia ao nosso país, porém seria necessário saber se é possível importar essa bactéria geneticamente modificada no Brasil. Caso positivo, seria indispensável realizar pesquisas sobre a viabilidade e a metodologia para o uso dessa bactéria em nosso meio.

E-mail: gustalima@hotmail.com Many studies about Haematobia irritans have been made due to the damages and worries it causes to cattle breeding in many countries. The common name of the fly is related to its behavior. 1111 Europe and the United States of America it is named horn-fly because it stays right on the base of the horns in order to protect itself from low temperatures; in Latin America, however, it is known as "mosca-da-paleta" because of its preferred localization on the shoulder of cattle. Biological aspects of the bovines can lead to different levels of fly infestation in the herd. Mean numbers of flies on different anatomic regions and individual infestations were evaluated in 60 Nelore bulls with the sarne zootechnic features during 1998 in Araçatuba city, State of São Paulo, Brazil. Both sides of the animals were filmed, and the number of flies was recorded in cassette tapes. Fly counts and their delimitation on 15 anatomic regions were performed from film viewing. During rainy days a significant increase (p<0.05) in number of flies in the abdominal region was observed. Shoulders, forelegs, back and ribs showed the highest number of flies (p<0.05). The evaluation of individual infestation revealed less than 50 flies on 50% of the cattle, 50 to 100 flies on 38% and over 100 flies on 12% of them.

Seu nome está relacionado com o local de permanência nos bovinos. É conhecida como "horn fly" (mosca-dos-chifres) na Europa e nos Estados Unidos da América e mosca-da-paleta na América Latina. Os fatores biológicos podem produzir em bovinos de um único rebanho, diferentes níveis de infestação da mosca. Durante o ano de 1998 em Araçatuba, estado de São Paulo, foram avaliados o número médio de mosca por região anatômica; bem como os diferentes níveis de infestação em 60 bovinos da raça Nelore. Os bovinos foram filmados de ambos os lados do corpo para registrar o número de mosca em fitas cassetes. As fitas foram assistidas para a contagem e demarcação da mosca em 15 regiões anatômicas. O maior número de mosca (p<0,05) foi observado nas regiões escapular, interescapular e costal; nos períodos chuvosos observou-se um aumento significativo (p<0,05) na região ventral. As avaliações individuais, demonstraram infestação com menos de 50 moscas em 50% dos bovinos, 50 a 100 moscas em 38% e acima de 100 moscas em 12% dos bovinos.


#26 - Toxicological sensitivity and specificity of the micro-complement fixation test for the detection of botulinum toxins C and D in culture medium and liver of mice, 18(2):47-52

The toxicological sensitivity and specificity of the micro-complement fixation test (MCF) for the detection of botulinum toxins C and D were studied in supernatants of the bacterial cultures and in livers of mice inoculated with lethal and sublethal doses. Botulinum toxins C and D were produced in Hemoline culture medium, titered through the determination of LD50 by the mouse test and adjusted to dilutions of 10, 1, 0.1, 0.01 and 0.001 LD50 Two experimental models were used to determine the toxicological sensitivity of MCF in the supernatant of the culture medium with the dilutions described, and also in liver extracts of mice weighing 20 g and inoculated with the same dilutions. Detection of the botulinum toxins was attempted in liver extracts of mice which had received lethal doses of the respective toxins, and in others which had been inoculated with sublethal doses and were sacrificed in intervals of 5 days. The results show that the toxicological sensitivity of MCF, regarding the two types of toxins at the level of 0.001 LD50, was 100% when the supernatants of the culture medium were tested; this means that the sensitivitywas 100 times higher than with the mouse test. The toxicological sensitivity of MCF in the liver extracts of mice inoculated with 1 and 10 LD50 of botulinum toxins C and D was inferior, giving values of 100, 80, 89 and 72% respectively. By this test it was also possible to detect botulinum toxins type C and D in liver extracts of mice inoculated with sublethal doses, up to 15 days after the injection. The specificity of MCF was 88% and 92%, when liver extracts of healthy contral mice were tested and when challenged with antitoxins C and D; and 100% when challenged with the supernatant of the culture medium. These results indicate that MCF could be of importance for research and could substitute in vivo tests.

[Toxicological sensitivity and specificity of the micro-complement fixation test for the detection of botulinum toxins C and D in culture medium and liver of mice.] Sensibilidade toxicológica e especificidade do teste de microfixação de complemento na detecção de toxinas botulínicas C e D em meio de cultura e fígado de camundongo. Pesquisa Veterinária Brasileira 18(2):47-52. Depto Apoio, Produção e Saúde Animal, Unesp-Campus de Araçatuba, Cx. Postal 533, Araçatuba, SP 16015-050, Brazil. No presente estudo pretendeu-se verificar a sensibilidade toxicológica e especificidade do Teste de Microfixação de Complemento (MCF) na detecção de toxinas botulínicas C e D no sobrenadante de cultivas bacterianos e em fígados de camundongos inoculados com doses letais e subletais. As toxinas foram produzidas em meio de cultura Hemoline, tituladas através da determinação da DL50 pelo Bioensaio em Camundongo e diluídas nas concentrações de 10, 1, 0, 1, 0,01 e 0,001 DL50. Desta forma, foram utilizadas em dois modelos experimentais, onde foi determinada a sensibilidade toxicológica do MCF no sobrenadante do meio de cultura com as diluições descritas acima e ainda em extratos hepáticos de camundongos com peso corporal de 20g, inoculados com as mesmas diluições. A tentativa de evidenciação das toxinas botulínicas nos extratos hepáticos de camundongos foi realizada através da sua extração após a morte pela administração das doses letais e ainda pelo sacrifício dos animais inoculados -com doses subletais, em intervalos de 5 dias. Os resultados evidenciaram uma sensibilidade toxicológica para o MCF de 100% para os dois tipos de toxinas ao nível de 0,01 DL50, quando testados os sobrenadantes de meio de cultura, portanto 100 vezes superior ao Bioensaio em Camundongo. A sensibilidade toxicológica do MCF, quando examinados extratos hepáticos de camundongos inoculados com 1 e 10 DL50 de toxinas botulínicas C e D, foi inferior, com valores de 100, 80, 89 e 72%, respectivamente. Pelo teste foi possível detectar toxinas botulínicas tipos C e D nos extratos hepáticos de camundongos inoculados com doses subletais até 15 dias após a sua inoculação. A especificidade do MCF foi de 88 e 92%, quando testados extratos hepáticos de camundongos sadios, e confrontados com as antitoxinas C e D; e 100% no sobrenadante do meio de cultura. Os resultados apontam para uma possível utilização do teste como importante instrumento de pesquisa e ainda na eventual substituição dos testes in vivo pelas suas implicações éticas e limitações práticas.


#27 - Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens, 18(1):39-44

[Evaluation of the sensitivity of bulk tank milk cultures for the isolation of contagious bovine mastitis pathogens.] Avaliação da sensibilidade da cultura de leite do tanque para isolamento de agentes contagiosos da mastite bovina. Pesquisa Veterinária Brasileira 18(1):39-44. Embrapa-Centro Nacional de Pesquisa de Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. Samples of bulk tank milk from 33 herds were collected at the dairy processing plant and cultured, as a means of detecting specific (contagious) bovine mastitis pathogens. Somatic cell counts (SCC) were made on a Fossomatic 90. Two and three weekly consecutive samples were obtained from 13 and 12 herds, respectively. Only one sample was examined from eight herds. Three daily consecutive samples of bulk milk and individual quarter samples from all lactating cows from four herds (A, B, C and D) were also examined. Milk from individual quarters were cultured on blood agar while tank milk samples were cultured on TKT, Mannitol Salt, MacConkey agars and Sabouraud containing chloramphenicol. Staphylococcus aureus was recovered from 26 of the 33 herds sampled in the dairy processing plant. Nine of these samples also contained Streptococcus agalactiae. Nine herds had SCC above 500,000 ml1 The remaining 23 herds had SCC levels below 400,000 ml1 S. aureus and S. agalactiae were isolated from five of the nine herds with high SCC, S. agalactiae from one and S. aureus from three. Six herds had SSC below 200,000 ml1 S. aureus and S. agalactiae were isolated from one, S. aureus from three, while the other two were negative for both pathogens. The results of herds A, B, C and D sampled at the farms showed that S. aureus was isolated from 1.8%, 19.2%, 17.0% and 8.4% of the animals and 0.9%, 5.9%, 5.4% and 2.2% of the mammary quarters, respectively. S. agalactiae was isolated from herds A, C and D. Within these herds the percentages of isolation were, respectively, 1.8%, 10.6% and 8.4% for the cows and 0.46%, 3.8% and 3.7% for the mammary quarters. S. aureus was recovered from all three bulk tank cultures from herds A, B and D. Only the third sample from herd C was positive for S. aureus. S. agalactiae was recovered from all samples collected from herd D, two samples from herd C and one sample from herd A. Coliforms were isolated from all tank samples from herds A, B, C and D and from all but one sample collected in the processing plant. Yeasts were recovered from 16 herds sampled at the processing plant and from all tank samples from herds A, B, C, and D. Neither coliforms or yeasts were isolated from the individual animals of herds A, B, C and D. These findings indicate that the milk was contaminated during or after milking, probably due to deficient hygiene and cleaning procedures. The analysis of the bulk tank milk cultures showed that the test was sensitive enough to detect contagious mastitis pathogens. The sensitivity of the test increased when more than two consecutive samples were examined.

Pesquisa Veterinária Brasileira 18(1):39-44. Embrapa-Centro Nacional de Pesquisa de Gado de Leite, Juiz de Fora, MG 36038-330, Brazil. Amostras de leite total (leite do tanque) de 33 rebanhos foram coletadas na plataforma de recepção da indústria laticinista e cultivadas para detectar patógenos específicos (contagiosos) da mastite. Foi feita a contagem de células somáticas (CCS) das amostras utilizando o equipamento Fossomatic 90. Em 13 e 12 rebanhos avaliaram-se duas e três amostras semanais consecutivas, respectivamente, e em oito avaliou-se apenas uma. Foram também examinadas três amostras diárias consecutivas do leite do tanque e amostras dos quartos mamários individuais, coletadas na própria fazenda, de todas as vacas em lactação de quatro rebanhos (A, B, C e D). As amostras de leite dos quartos mamários individuais foram cultivadas em ágar sangue e as amostras do tanque, em placas de TKT, Sal Manitol, MacConkey e Sabouraud contendo cloranfenicol. Dos 33 rebanhos cujas amostras foram obtidas na plataforma de recepção da indústria, isolou-se Staphylococcus aureus de 26, nove desses em associação com Streptococcus agalactiae e em três rebanhos isolou-se somente S. agalactiae. Nove rebanhos tiveram CCS acima de 500.000 ml1 e 21, abaixo de 400.000 ml1 Em cinco dos nove rebanhos com CCS acima de 500.000 m1-1 foram isolados S. aureus e S. agalactiae, em três, apenas S. aureus e em um, apenas S. agalactiae. Seis rebanhos apresentaram CCS abaixo de 200.000 ml·1; de um deles foram isolados S. aureus e S. agalactiae, de três, S. aureus e os outros dois foram negativos para estes dois patógenos. Os resultados encontrados nos quatro rebanhos cujas amostras foram coletadas na própria fazenda mostraram que S. aureus foi isolado nas seguintes porcentagens dos animais: 1,8%, 19,2%, 17,0% e 8,4% e dos quartos mamários: 0,9%, 5,9%, 5,4% e 2,2%, respectivamente, para os rebanhos A, B, C e D. S. agalactiae foi isolado dos rebanhos A, C e D. Nestes três rebanhos, as porcentagens de isolamento foram, respectivamente, 1,8%, 10,6% e 8,4% para as vacas e 0,46%, 3,8% e 3,7% para os quartos mamários. S. aureus foi isolado de todas três amostras do tanque dos rebanhos A, B e D. Somente a terceira amostra do rebanho C foi positiva para S. aureus. S agalactiae foi recuperado de todas as amostras do rebanho D, duas do rebanho C e de uma do rebanho A. Todas as amostras do tanque dos rebanhos A, B, C e D apresentaram contaminação com coliformes e somente uma das amostras coletadas na plataforma de recepção da indústria foi negativa para coliformes. Leveduras foram isoladas de 16 amostras coletadas na indústria e de todas amostras do tanque dos rebanhos A, B, C e D. Não foram isolados coliformes ou leveduras dos quartos mamários dos animais destes rebanhos, sugerindo que ocorreu contaminação do leite durante ou após a ordenha, provavelmente devido a deficiências nos processos de limpeza e higienização. A análise dos resultados das culturas do leite do tanque mostrou que o exame foi específico para detectar os patógenos contagiosos da mastite. A sensibilidade do teste aumentou quando se examinaram mais de duas amostras consecutivas.


#28 - Sensitivity and specificity of the Califomia Mastitis Test as a diagnostic tool for subclinical mastitis in quarter somatic cell count estimation

Abstract in English:

The California Mastitis Test (CMT) is a simple and widely used diagnostic tool for subclinical mastitis. lt is used even in areas where laboratory facilities are available for diagnosis and monitoring program purposes. CMT usually score 1-5, where 1 indicates a completely negative reaction and 2-5 increasing degrees of inflammatory reaction of the udder. The reactions 2-5, 3-5 or 4-5 may be considered as indicative of subclinical mastitis, because they are related to an increase of the somatic cell count (SCC) in the milk. This variation, however, may produce either false-positive or false-negative results. The aim of this study was to assess the sensitivity and specificity of the CMT in comparison with SCC. A total of 3,012 quarter milk samples from 760 lactating cows were examined. CMT was evaluated at the moment of sampling at cow side and SCC in the laboratory, with a fluoro-opto-electronic method (Fossomatic 90). The average CCS (x 1,000 cells/ml) for CMT scores were: 1 (79.9), 2 (333.5), 3 (670.3), 4 (1,354.0) and 5 (4,455.6). Three options for CMT interpretation were evaluated in relation to a range of CCS, starting from 100,000 cells/ml: (a) 1 versus 2, 3, 4, and 5; (b) 1 and 2 versus 3, 4 and 5; and (e) 1, 2 and 3 versus 4 and 5. The sensitivities of CMT scoring to detect quarters with SCC >200,000/ml were 79%, 61% e 34%for options a, b ande, respectively. The sensitivities of CMT scores for SCC >500,000/ml for options a, b ande, were, respectively, 93%, 82% e 54%. The sensitivity of CMT scores to identify subclinical mastitis was satisfactory (above 80%) when options b ande were used. The interpretation of CMT score 3 as negative for subclinical mastitis was considered as adequate (sensitivity around 80%) only when CCS ranged between 1,200,000 and 1,400,000 cells/ml. The specificities of CMT scoring for CCS of 200,000 and 500,000 were, respectively, 90% and 80% (option a), 97% and 90% (option b) and 99% and 97% (option e).

O "California Mastitis Test" (CMT) estima o conteúdo de células somáticas no leite e é interpretado subjetivamente, estabelecendo-se escores que, na maioria dos casos, variam de 1 a 5. O escore 1 indica uma reação completamente negativa e os de 2-5 indicam graus crescentes de resposta inflamatória do úbere, sendo normalmente considerados como indicativos de mastite subclínica. Dependendo da interpretação dos escores, o CMT pode produzir resultados falso-positivos ou falso-negativos. Esse trabalho teve o objetivo de avaliar a sensibilidade e a especificidade do CMT em relação à contagem de células somáticas (CCS). Foram utilizadas 3.012 amostras de leite provenientes de 760 vacas Holandesas ou mestiças Holandês-Zebu, pertencentes a 15 rebanhos. Todas as amostras foram submetidas ao CMT e processadas para CCS em equipamento Fossomatic 90. Os valores médios de CCS (x 1.000 células/mi) obtidos para os escores de CMT foram 1 (79,9), 2 (333,5), 3 (670,3), 4 (1.354,0) e 5 (4.455,6). Três opções de interpretação (doente/não-doente) para o CMT foram testadas, em relação aos valores de CCS, iniciando com 100.000 células/ml: (a) 1 versus 2, 3, 4, e 5; (b) 1 e 2 versus 3, 4 e 5; (c) 1, 2, 3 versus 4 e 5. As sensibilidades do CMT em identificar corretamente quartos mamários acima de 200.000 células/mi foram 79%, 61% e 34%, para as opções a, b e e, respectivamente. Para identificar corretamente contagens acima de 500.000 células/mi, as sensibilidades do CMT, para as opções a, b e e, foram, respectivamente: 93%, 82% e 54%. A sensibilidade do CMT em identificar quartos mamários com mastite subclínica foi adequada (acima de 80%) quando a interpretação do teste foi mais rigorosa (opções a e b). A interpretação da reação 3 como negativa (opção e) só alcançou sensibilidade de 80% para contagens entre 1.200.000 e 1.400.000 células/mi. As especificidades do CMT, para CCS de 200.000 e 500.000 foram, respectivamente, 90% e 80% (opção a), 97% e 90% (opção b) e 99% e 97% (opção e).


#29 - Sensitivity of the rabbit to the fresh and dried leaves of Lantana camara var. aculeata (Verbenaceae)

Abstract in English:

The leaves of Lantana camara var. aculeata were given to rabbits in order to test the sensitivity of this animal species to the plant, collected at Quatis, Rio de Janeiro. The experiments were also performed to test the sensitivity of the rabbit in comparison with cattle and sheep. The main purpose of this study was to find the best methodology for surveys on the toxicity of the diferente lantanas in Brazil. Fifteen adult rabbits were used, two of them as controls. Two rabbits ate by themselves 11, within five days a total of 37.5 and 44.44g/kg of the plant, preserved in its fresh stage at 4° to 6ºC. The other 11 rabbits received the dried and powdered leaves of L. camara var. aculeata by stomach tube at doses of 6, 8, 10, 12 and 15g/kg, given within one day. The symptoms in the rabbits were discrete to slight and consisted in apathy, lack of apetite and anorexia, few dried faeces in form of sibalas, sometimes covered by mucus, as well as icterus and loss of weight. None of the animals showed photosensitivity and none of them died. The only post-mortem finding in six rabbits which were killed at different phases of the experimental poisoning, was icterus. Histopathological exames revealed only lesions of slight severity, which were cloudy swelling of the liver cells at the periphery of the lobules, discrete vacuolization in the intermediate zone which resulted in lysis of a few liver cells, and discrete apoptosis. Because of the low sensitivity of the rabbit to the toxicity of Lantana, when compared with that of cattle and sheep, it is concluded that this animal species is not suitable for the proposed survey.

A finalidade deste estudo foi testar a sensibilidade do coelho à Lantana camara var. aculeata procedente de Quatis, Rio de Janeiro, e compará-la com a dos bovinos e ovinos, no intuito de encontrar a melhor metodologia para o trabalho de levantamento da toxidez das diversas lantanas do Brasil. Utilizaram-se 15 coelhos adultos, dos quais dois serviram como controle. Dois coelhos ingeriram voluntariamente um total de 37,5g/kg e 44,44g/kg de peso vivo das folhas frescas e conservadas à temperatura de 4° a 6ºC, durante 5 dias. Os demais coelhos receberam doses de 6, 8, 10, 12 e 15g/kg de peso vivo das folhas dessecadas de L. camara var. aculeata, administradas em um dia, por via intra-gástrica. Os sintomas foram discretos a leves e resumiram-se em apatia, inapetência e anorexia, fezes escassas, ressecadas, em pequenas sílabas e, às vezes, envoltas por muco, icterícia e perda de peso; não observou-se fotossensibilização nos animais. Nenhum animal morreu. A única alteração macroscópica observada em 6 coelhos sacrificados em diversas fases de intoxicação, foi icterícia. As lesões histológicas foram muito suaves e restringiram-se apenas a tumefação dos hepatócitos da periferia dos lóbulos, discreta vacualização na zona intermediária, com raras células hepáticas terminando em lise e discreta apoptose. Diante da baixa sensibilidade do coelho a Lantana, quando comparada a do bovino e ovino, conclui-se que esta espécie animal não se presta para o levantamento proposto.


#30 - Sensitivity of microorganisms isolated from bovine mastitis to antibiotics and chemotherapeutic drugs

Abstract in English:

Antibiotic and chemotherapeutic drug sensitivity of 951 cultures of Staphylococcus sp., 590 cultures of Streptococcus sp. and 550 cultures of Corynebacterium sp., isolates of bovine mastites were studied. Staphylococcus sp. showed the highest levels of sensitivity (75-90%) to Cephalothin, Nitrofurantoin Vancomycin and Novobiocion: Complete resistance was seen against Phosphomycin, Polymyxin B, Colestin Sulphate and Rifamycin. In relation to Streptococcus sp., the most effective antibiotics were Chloramphenicol, Novobiocin and Vancomycin (72-77%). High levels of resistance (90-100%) were seen with Rifamycin, Streptomycin, Colestin Sulfate, Sulfomethoxazole, Phosphomycin and Polymyxin B. As for the cultures of Corynebacterium sp., the greatest sensitivity was seen with Cephalosporin (100%), followed by Erythromycin, Chloramphenicol, Gentamycin, Kanamycin, Tetracycline, Carbenicillin and Tobramycin (80-97%). None of the samples of Corynebacterium sp. studied showed complete resistance to any of the drugs tested.

Estudou-se in vitro a sensibilidade a antimicrobianos de 951 amostras de bactérias do gênero Staphylococcus, 590 amostras do gênero Streptococcus e 550 do gênero Corynebacterium, isoladas de mastites bovinas. Obtiveram-se os seguintes resultados em relação às bactérias do gênero Staphylococçus: os maiores índices de sensibilidade (75 a 90%) foram os observados frente à cefalosporina, nitrofurantoína, vancomicina e novobiocina. Observou-se 100% de resistência frente à fosfomicina, polimixina B, sulfato de colistina e rifamicina. Em relação a Streptococcus sp., os antimicrobianos mais efetivos foram cloranfenicol, novobiocina e vancomicina (sensibilidade de 72 a 77%). Altos índices de resistência (90 a 100%) foram observados frente à rifamicina, estreptomicina, sulfato de colistina, sulfametoxazol, fosfomicina e polimixina B. Quanto às amostras de Corynebacterium sp., a maior sensibilidade foi observada frente à cefalosporina (100%), seguida da eritromicina, cloranfenicol, gentamicina, canamicina, tetraciclina, carbenicilina e tobramicina (97 a 80%). Não foi observada resistência máxima (100%) aos antimicrobianos nas amostras de Corynebacterium sp. estudadas.
