E-mail: rinaldo.mota@hotmail.com RESUMO.- A paratuberculose (doença de Johne) é uma das doenças de maior importância econômica para ruminantes em vários países e pode representar uma ameaça ao desenvolvimento da pecuária brasileira. É uma doença infecto-contagiosa que provoca enterocolite granulomatosa crônica, incurável e de difícil controle, cujo agente é o Mycobacterium avium subsp. paratuberculosis (MAP). Descreve-se a ocorrência de paratuberculose em um rebanho de búfalos no Estado de Pernambuco, Brasil. Não foi encontrado registro, na literatura, da ocorrência de paratuberculose em búfalos no país. De 100 búfalos, cinco mostravam sinais clínicos característicos da doença. À necropsia de dois animais as lesões estavam restritas ao intestino delgado com evidente espessamento da mucosa, aumento de linfonodos mesentéricos e vasos linfáticos proeminentes e dilatados. À microscopia, observaram-se na mucosa do intestino, infiltrado inflamatório granulomatoso com numerosos macrófagos epitelióides e células gigantes de Langhans, além de bacilos álcool-ácido resistentes (BAAR) visualizados através da coloração de Ziehl-Neelsen (ZN). Nos linfonodos mesentéricos, havia espessamento da cápsula e marcada inflamação granulomatosa. O exame direto pela técnica de ZN para pesquisa do bacilo em esfregaços de fezes, raspado de mucosa intestinal e imprint de linfonodos mesentéricos resultou positivo. A PCR IS900 específico de linfonodo mesentérico e mucosa intestinal revelou amplificação de um fragmento de aproximadamente 110pb, confirmada pela comparação com outras sequências de M. avium subsp. paratuberculosis disponíveis no GenBank.

E-mail: rinaldo.mota@hotmail.com Paratuberculosis (PTB) is a disease of great economical importance for ruminant in several countries and represents a threat to the development of Brazilian livestock. The contagious disease caused by chronic PTB leads to incurable granulomatous enterocolitis of difficult control. PTB is caused by the Mycobacterium avium subsp. paratuberculosis (MAP). No record on the occurrence of paratuberculosis in buffaloes in Brazil could be found. Five of 100 buffaloes in a herd in Pernambuco-Brazil showed clinical signs characteristic of PTB. At necropsy, of two animals the lesions were restricted to the small intestine with thickening and corrugation of the mucosa, increase of mesenteric lymph nodes and prominent lymph vessels. Histopathology revealed granulomatous inflammation infiltrated with numerous epithelioid macrophages, Langhans type giant cells, and clusters of Ziehl-Neelsen (ZN) positive organisms within the intestinal mucosa. In the mesenteric lymph nodes there was thickening of the capsule and marked granulomatous inflammation. Smears of feces and scrapping smears were prepared from intestinal mucosa and cut surface of mesenteric lymph nodes and, stained by the Ziehl-Neelsen method for research of acid fast bacilli, with positive results. Lymph nodes and intestinal mucosa revealed at IS900 specific polymerase chain reaction amplification of a fragment of about 110pb, confirmed by the comparison with other sequences of M. avium subsp. paratuberculosis available in GenBank.

E-mail: franklin.riet@pq.cnpq.br This paper reports paratuberculosis in sheep and goats for the first time in Brazil. On Farm 1 in a flock of 33 goats and 13 sheep, one adult female goat presented progressive weight loss during about one year, and soft feces during one month before death. All the animals from the flock were submitted to the comparative tuberculin test. One sheep was positive to the avian tuberculin (2.2%) and in two the results were inconclusive. On Farm 2, in a flock of 200 sheep and 80 goats, one adult ewe presented progressive weight loss during about one year, and soft feces during 20 days before death. All sheep older than four months of age and 23 goats were submitted to tuberculin test with avian tuberculin; 47 (25.4%) sheep were positive, 115 (61.5%) had inconclusive results, and 25 (13.4%) were negative. No positive tuberculin test were observed in the goats, but in 11 (47.8%) the test was inconclusive and in 12 (52.2%) was negative. At necropsy both affected animals had enlarged mesenteric lymph nodes. In the affected sheep, the gut wall was thickened and corrugated, mainly in the ileum and ileocecal valve. Histologically, granulomas with mainly foamy macrophages were observed in the lamina propria and submucosa of the gut, in mesenteric lymph nodes, and in the goat also in the liver. One tuberculin positive sheep and one tuberculin negative goat from Farm 1 were euthanized and necropsied. A mild thickened of the intestinal wall was observed grossly in both animals. Inflammatory infiltrate with mainly lymphocytes was observed on the submucosa and lamina propria of the gut. In the four cases acid-alcohol positive rods, positive for Mycobacterium spp. on immunohistochemical examination were observed in macro-phages and lymphocytes. New studies are necessary to know the prevalence and economical impact of paratuberculosis in goats and sheep in Brazil, to determine control measures, and to evaluate the risk of human transmission.

Na Fazenda 1, de um rebanho de 33 caprinos e 13 ovinos, uma cabra adulta apresentou emagrecimento progressivo por aproximadamente um ano e fezes pastosas um mês antes da morte. Todos os animais do rebanho foram tuberculinizados com a prova comparativa. Um ovino (2,2%) teve resultado positivo à tuberculina aviar e em dois o teste foi inconclusivo. Na Fazenda 2, com 200 ovinos e 80 caprinos, foi afetada uma ovelha adulta que apresentou emagrecimento progressivo por aproximadamente um ano e fezes pastosas por aproximadamente 20 dias. Todos os ovinos com mais de 4 meses de idade e 23 caprinos foram tuberculinizados com tuberculina aviar; em 47 (25,4%) o resultado foi positivo, em 115 (61,5%) inconclusivo e em 25 (13,4%) negativo. Entre as cabras não houve nenhuma positiva à tuberculina aviar, mas em 11 (47,8%) o teste foi inconclusivo e em 12 (52,2%) foi negativo. Na necropsia dos dois animais com sinais clínicos os linfonodos mesentéricos estavam aumentados de tamanho e edemaciados. O ovino afetado da Fazenda 2, apresentou espessamento e enrugamento da mucosa do intestino, principalmente no íleo e válvula íleo-cecal. Microscopicamente o caprino e o ovino com sinais clínicos apresentaram lesões semelhantes, caracterizadas por granulomas com predominância de macrófagos espumosos, na lâmina própria e submucosa do intestino e linfonodos mesentéricos. O ovino positivo à tuberculina e um caprino negativo na Fazenda 1 foram eutanasiados e apresentaram discreto espessamento da mucosa do íleo. Na histologia foi observado infiltrado preferentemente linfocítico. Em todos os casos dentro de macrófagos e linfócitos havia bacilos ácool-ácidos resistentes, positivos na imuno-histoquímica para Mycobacterium spp. Sugere-se a necessidade de estudar a prevalência de paratuberculose em rebanhos de caprinos e ovinos no Brasil, determinar medidas de controle e avaliar a possibilidade de transmissão para humanos.

E-mail: alage@vet.ufmg.br This paper describes the clinical, pathological, and microbiologic aspects of paratuberculosis (Johne’s disease) in a dairy Gyr herd in the State of Paraíba, northeastern Brazil. An eight years old cow with chronic unresponsive diarrhea was clinically examined and euthanized for pathological evaluation. Fecal samples from all 160 animals over 12 months of age from the herd were collected for isolation of Mycobacterium avium subsp. paratuberculosis. Clinically, the index case cow was severely dehydrated, cachectic, with profuse mucous diarrhea. The main post-mortem findings were emaciation and thickened intestinal wall. Microscopically, the intestinal lamina propria and submucosa were infiltrated by macrophages, epithelioid cells, and Langhans giant cells with numerous alcohol-acid resistant bacilli in the cytoplasm. Two fecal samples displayed growth in slants of Herrold’s egg-yolk agar supplemented with mycobactin J, 150 days after incubation. No growth was noticed in slants without mycobactin J. Microscopic examination of the isolated microorganisms stained by Ziehl-Neelsen revealed considerable amounts of alcohol-acid resistant bacilli, morphologically compatible with Mycobacterium spp. Based on the clinical signs, gross and histological lesions, growth time, bacterial morphology in Ziehl-Neelsen staining, and dependence of mycobactin J, the first diagnosis of paratuberculosis in Zebu cattle was made.

Uma vaca de oito anos que apresentava diarréia persistente, refratária a tratamento foi necropsiada para estudo anátamo-histopatológico. Também foram coletadas amostras de fezes de todos os 160 animais do plantel, com idade superior a 12 meses, para tentativa de isolamento de Mycobacterium avium subsp. paratuberculosis. Ao exame clínico, o animal caso índice apresentou caquexia, diarréia profusa e desidratação grave. À necropsia, o animal apresentou-se emaciado e, ao exame detalhado do trato digestivo, foi observado espessamento da parede e superfície mucosa do íleo e intestino grosso. À microscopia, verificou-se intensa infiltração de macrófagos espumosos associado a raras células epiteliódes e gigantes do tipo Langerhans na lâmina própria e submucosa. À coloração de Ziehl-Neelsen foram observadas miríade de bacilos álcool-ácido resistentes no citoplasma destas células. Houve crescimento de colônias bacterianas em duas das 160 amostras de fezes após 150 dias de incubação em tubos com meio Herrold’s egg-yolk suplementados com micobactina J e ausência de crescimento nos tubos com mesmo meio, mas sem suplementação. Os microrganismos isolados foram corados pelo Ziehl-Neelsen observando-se presença de grande quantidade de bacilos álcool-ácido resistente, com morfologia compatível ao gênero Mycobacterium. Baseado na história clínica, achados anátomo-histopatológicos e histoquímicos (Ziehl-Neelsen), e microbiológicos, firmou-se o primeiro diagnóstico de paratuberculose em Zebu na Paraíba.

Interpretation of the CCT was checked by calculation of the difference obtained between skin measures correspondent to respective tuberculins, before and 72 hours after inoculation: negative, when bovine PPD is smaller than avian PPD or larger in up to 1.8mm; suspicious or inconclusive, reaction to bovine PPD larger than avian PPD between 1.9 and 2.4mm; positive, reaction to bovine PPD larger than or equal to avian PPD in 2.5mm. From goats submitted to CCT, 0.47% (9/1866) reacted positively (95% CI = 0.23%-0.94%). The prevalence of positive herds to caprine tuberculosis was 10.71% (95% CI = 5.02-19.37%). When submitted to necropsy and histopathologic examination, a positive goat at tuberculin test presented tuberculosis-like lesions. The presence of Acid-Alcohol Resistant Bacilli (BAAR) was also detected by direct bacterioscopy; in relation to herd size, herds with larger number of animals (>25) with high incidence of the disease, and presence of the infection was evidenced in properties that raised cattle in association with goats and did not carry out tuberculin tests in cattle.

A comparison of two culture media used to isolation was also investigated. Culture was positive in 2.0% of faecal samples. Isolation was obtained using Löwenstein Jensen® with mycobactin® J, and the Middlebrook® 7H11 with OADC®. The Löwenstein Jensen® with mycobactin® J was that provided highest amount of isolations. The percentages of isolation in each culture media were 2.0% (6/300) to Löwenstein Jensen® with micobactina J, and 1.0% (3/300) to Middlebrook® 7H11/OADC. The three positive samples in Middlebrook® 7H11/OADC were also positive in Löwenstein Jensen® with micobactina J. In the Middlebrook® 7H11/OADC alone there was no sample growth. The results of this study suggest that culture media of Löwenstein-Jensen® with micobactina® J is more effective for the isolation of sheep strains in Portugal.

[Clinical and pathological aspects of bovine paratuberculosis in Rio Grande do Sul, southern Brazil.] Aspectos clínicos e patológicos da paratuberculose em bovinos no Rio Grande do Sul. Pesquisa Veterinária Brasileira 19(3/4):109-115. Setor de Patologia Veterinária, Depto Patologia Clínica Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Av. The clinical and pathological aspects of bovine paratuberculosis in a dairy herd in the county of Capela de Santana, Rio Grande do Sul, southern Brazil, are described. Clinical signs in eight cows out of 345 cattle included chronic diarrhea refractory to treatment, progressive emaciation and decreased milk production. Necropsy findings included severe and diffuse thickening of the mucosa of the small intestine with exaggerated reticular pattern, reflected in the serosal surface, similar to cerebral gyri. Intestinal loops were filled with whitish watery contents. The lymphatic vessels of the intestinal serosa and of the mesentery were thickened and tortuous. The mesenteric lymph nodes were enlarged and, on cut surface, there were oozing of milky fluid. Plaques of mineralization were evident in the intima of larger arteries, in the endocardium of the heart valves and in the ruminal serosa. There was also edema of the abomasal folds and atrophy of the caudate lobe of the liver. The main histopathological findings were granulomatous enteritis, lymphadenitis and lymphangitis. The inflammatory infiltrate was composed of macrophages, epithelioid cells and Langhans’ giant cells containing large numbers of acid-fast bacilli. Vascular lesions included degeneration, calcification and collagen proliferation of the intima and media of larger arteries. Other microscopk lesions included calcification of the ruminal serosa, diffuse hepatocellular atrophy and multifocal granulomatous hepatitis. Mycobacterium paratuberculosis was cultured in Herrold’s médium enriched with mycobactin from all the eight samples of intestinal scrapings submitted to bacteriological examination.

Descrevem-se os achados clínicos e patológicos da para tuberculose em uma criação intensiva de bovinos· de leite no município de Capela de Santana, RS. Sinais clínicos foram observados em oito de um total de 345 bovinos e consistiam em diarréia crônica refratária ao tratamento, emagrecimento progressivo e queda da produção de leite. As principais lesões macroscópicas, observadas nos oito animais eutanasiados e necropsiados, incluíam intestino delgado com acentuado espessamento da parede e superfície mucosa de aspecto reticulado, semelhante às circunvoluções cerebrais, lesão essa perceptível através da serosa. A luz intestinal estava preenchida por conteúdo fluido e de aspecto leitoso. Os vasos linfáticos do mesentério mostravam-se mais evidentes, sendo que alguns tinham aspecto varicoso. Os linfonodos mesentéricos estavam aumentados de volume e, ao corte, fluía grande quantidade de líquido leitoso: Focos de mineralização foram observados na íntima das artérias, nas válvulas cardíacas e na serosa do rúmen. Havia também edema das dobras do abomaso e do mesentério e atrofia do lobo caudado do fígado. As principais lesões microscópicas incluíam enterite, linfadenite e linfangite granulomatosas que se caracterizavam por infiltrado inflamatório composto por macrófagos, células epitelióides e células gigantes de Langhans que continham grande quantidade de bacilos álcool-ácido resistentes. As lesões vasculares consistiam em degeneração e mineralização das túnicas íntima e média das artérias de grande calibre associadas à proliferação de colágeno. Havia calcificação da serosa do rúmen, atrofia hepatocelular difusa e hepatite granulomatosa multifocal. Mycobacterium paratuberculosis foi cultivado em meio de Herrold enriquecido com micobactina a partir de raspados do intestino em todas as oito amostras enviadas para exame bacteriológico.


#47 - Comparison of the allergic diagnosis of bovine tuberculosis using the comparative and Stormont tests

Abstract in English:

The comparison of the comparative tuberculin test and the Stormont method, performed at the sarne groups of animals, gave highly convergent results in bovines free from tuberculosis and also in infected groups with strong reactors. In weak reactors the results were highly discordant. A group of 21 bovines with negative results at the comparative tuberculin test, but reacting to avian tuberculin, was negative with the Stormont test. Twenty six bovines which reacted strongly positive (4.0mm or more) at the comparative test, with or without non-specific reactions, showed also positive reactions in 88.4%, suspicious reactions in 7.7% and negative reactions in 3.9% at the Stormont test. A group of 19 bovines, initially tested by the Stormont method and considered as infected, when retested by the comparative test after 90 days, showed 84.2% positive reactors and 15.7% negative ones. Groups of bovines with suspicious (2.0 to 2.9mm) or weakly positive reactions (3.0 to 3.9 mm) at the comparative test, showed 20.0% positive, 20.0% suspicious and 60.0% negative reactors in the suspicious group and 53.3% positive, 13.3% suspicious and 33.3% negative reactions in the weakly positive reactors by the Stormont test. Another group of bovines gave suspicious reactions when initially tested by the Stormont method, but showed 50.0% positive, 9.3% suspicious and 40.6 negative reactions when retested by the comparative test. The discordant results of the present and other investigations do not allow to recommend the Stormont test for use in routine procedures for the diagnosis of bovine tuberculosis before a critical evaluation has been made, with slaughter and bacteriological exarnination of reacting animals.

A comparação entre a tuberculinização intradérmica comparada (tuberculina bovina e aviária) e a tuberculinização dupla (com 7 dias de intervalo) conhecida por teste de Stormont, revelou resultados altamente concordantes em grupos de bovinos sem tuberculose e em grupos de animais infectados com fortes reações alérgicas, no entanto, apresentou resultados bem discordantes nos reagentes suspeitos e fracamente positivos em ambos os testes cujas causas não puderam ser esclarecidas por exames necroscópicos e bacteriológicos por razões econômicas. Em um grupo de 21 bovinos com resultados negativos na tuberculização comparada, mas portadores de reações à tuberculina aviária, o teste de Storment revelou também resultados negativos. Em 26 bovinos reagentes positivos no teste comparativo com aumento de espessura da dobra da pele (AEDP) acima de 4,0 mm, com ou sem reações inespecíficas, o teste de Stormont acusou também 88,4% de reações positivas, 7,7% reações suspeitas e 3,9% reações negativas. Em outro experimento, um grupo de 19 bovinos reagentes positivos no teste de Stormont, quando retestado, 90 dias depois pelo teste comparativo, este revelou 84,2% de reagentes positivos e 15,7% de reações negativas. Em grupos de bovinos com reações suspeitas (AEDPs entre 2,0 e 2,9 mm) ou fracamente positivos (AEDPs entre 3,0 e 3,9mm) no teste comparativo, o teste de Stormont revelou 20,0% de reações positivas, 20,0% de reações suspeitas e 60,0% negativas, e 53,3% positivas, 13,3% suspeitas e 33,3% negativas respectivamente. Num grupo de bovinos que no teste de Stormont acusou reações suspeitas e que, 90 dias após foi retestado pela tuberculização comparativa, esta acusou 50,0% de reagentes positivos, 9,3% suspeitos e 40,6% negativos. Os resultados discordantes da presente pesquisa e da de outras não recomendam o uso rotineiro do teste de Stormont no diagnóstico da tuberculose bovina antes de uma avaliação científica criteriosa, com abate e exames bacteriológicos de animais reagentes.


#48 - Intermittent treatment of bovine tuberculosis with isoniazid

Abstract in English:

An intennittent treatment regímen for bovine tuberculosis with isoniazid was used. Fifty five positive and 13 suspect reactors were treated orally with cristalized isoniazid (25 mg/kg bw) mixed in a concentrated ration given 3 times a week, during 6 months. The cure of the tuberculosis was evaluated after the treatment by allergic desensitization with comparative tuberculinizations performed in 3 months intervals, during a year. The monitoring showed progressive reduction of the allergic sensitivity until definitive disappearance, At 90 days pos-treatment there were 31 negative, 19 suspect and 18 positive reactors; at 180 days, 47 were negative, 10 suspect and 11 positive; at 270 days, 51 were negative, 12 suspect and 5 weakly positive, and at 360 days, 63 were negative, 5 suspect and no one anymore positive. As it was demonstrated experimentally, the pos-treatment suspect reactors may be considered infection-free and so the cure rate was 100 percent. This experiment showed that the intennittent 3 week dosages were as good as the continuous regímen with the advantage of about 40 percent cost reduction.

Foi adotado um regime de tratamento intermitente da tuberculose com isoniazida em 55 bovinos reagentes positivos e 13 suspeitos, A isoniazida em forma de sal cristalizado foi fornecida aos animais misturada em ração concentrada, em doses de 25 mg/kg p.v., nas 2. as, 4. as e 6. as feiras, durante 6 meses. A cura da tuberculose foi avaliada pela dessensibilização alérgica, após o término do tratamento através de tuberculinizações comparativas realizadas de 3 em 3 meses, durante um ano. Este monitoramento revelou progressiva redução da sensibilidade alérgica até o desaparecimento definitivo. Aos 90 dias pós-tratamento já havia 31 (45,5%) bovinos negativos, 19 (27,9%) suspeitos e 18 (26,4%) positivos; aos 180 dias, 47 (69,1%) negativos, 10 (14,7%) suspeitos e 11 (15,1%) positivos; aos 270 dias, 51 (75,0%) negativos, 12 (17,6%) suspeitos e 5 (7,3%) positivos e aos 360 dias, 63 (92,6%) negativos, 5 (7,3%) suspeitos e nenhum mais positivo. Como os reagentes suspeitos, no final do tratamento podem ser considerados livres da infecção, o percentual de curas foi de 100%, mostrando que o tratamento intermitente com 3 dosagens de isoniazida semanais foi tão eficiente quanto o regime contínuo. O regime intermitente reduziu de 120 para 80 o número de doses administradas e reduziu o custo em 40%.


#49 - Histologic study and isolation of mycobacteria from tuberculosis-like lesions in cattle in southern Brazil

Abstract in English:

One hundred and fifty-nine lesions macroscopically similar to tuberculosis, observed in cattle slaughtered from 13 counties located in southem Rio Grande do Sul, were studied histologically. Tuberculosis was diagnosed in 147 (92.45%), lesions, club-forming granulomas in 7 (4.4%), eosinophilic abscess in 2 (1.26%), and abscess in 3 (1.89%). Fifty-nine specimens with histological lesions of tuberculosis were cultured for mycobacteria, obtaining 40 isolates, all of them classified as Mycobacterium bovis. Based in these results and the data related with condemnation due to tuberculosis in Rio Grande do Sul, the prevalence of the disease in beef cattle in the State during 1980-1987 was estimated in 0.6%. However, marked differences were observed between different areas. The higher prevalences (2.23% to 3.39%) occurred in the regions focated in the coastland of the "Lagoa dos Patos", middle prevalences (0.97% to 1.95%) in the regions located West and Northwest near Porto Alegre, and the lower prevalences in the other regions of the State (0.12% to 0.71 %). The annual prevalence of the disease has been decreasing sistematically from 1980 (0.95%) to 1987 (0.42).

Cento e cinqüenta e nove lesões observadas em frigoríficos como macroscopicamente similares à tuberculose, de bovinos provenientes de 14 municípios da região sul do Rio Grande do Sul, foram estudadas histologicamente. Tuberculose foi diagnosticada em 147 (92,45%) lesões, actinogranuloma em 7 (4,4%), abscesso eosinofílico em 2 (1,26%) e abscesso em 3 (1,89%). Em 59 desses materiais, com lesões histológicas de tuberculose, foram realizadas culturas para micobactérias, obtendo-se 40 isolamentos, todos eles tipificados como Mycobacterium bovis. Com base nos resultados do estudo histológico e bacteriológico foram analisados os dados de condenação por tuberculose de todos os municípios do Rio Grande do Sul, no período 1980-1987, estimando-se a prevalência da doença, em gado de corte no Estado, em aproximadamente 0,6%. No entanto detectaram-se marcadas variações, entre regiões, na freqüência de lesões macroscopicamente similares à tuberculose, observando-se maiores prevalências (2,23% a 3,39%) nas microrregiões homogêneas localizadas no litoral da Lagoa dos Patos, prevalências médias nas 6 microrregiões próximas a Porto Alegre, no sentido oeste e noroeste (0,97% a 1,95%), e as menores prevalências nas restantes microrregiões do Estado (0,12% a 0,71%).


#50 - Massal treatment of bovine tuberculosis with isoniazid

Abstract in English:

There were 409 bovines in a Swiss Brown-cattle herd in Mossoró, Rio Grande do Norte, Brazil, reacting positively to the tuberculin test. The tuberculosis was confirmed by necropsy, histopathological and bacteriological procedures. The infected animals were treated with purê crystalised isoniazid, administered orally at a dose of about 25 mg/kg/day, during 60 days consecutively and other 60 doses 3 times a week. The large number of reactors did not allow individual medication. Therefore treatment at a large scale was used in groups of animals varying in weight up to 100 kg. Isoniazid was mixed mechanically, daily, and applied at 12 g per 1 kg of high palatable concentrated ration, furnished in long double troughs. The quantity of feed was calculated based on the media of live weight and the numbers of animals in each group. The clinically affected animals showed recuperation starting with the second month of treatment. Thirty days after the end of the treatment the allergic monitoration began by comparative tuberculinizations at intervals of 2 to 3 months. The cure was evaluated by the gradual tuberculin desensitization. In most animals the tuberculin reactions became negative after 2 to 4 months post-treatment. At the end of this experiment 96.4% of the treated animals were cured. In slaughtered animals no lesions were found.

Num rebanho de bovinos da raça suíça parda, em Mossoró, Rio Grande do Norte, foram encontrados 409 animais reagentes positivos na tuberculinização comparativa com tuberculinas PPDs bovina e aviária. A tuberculose foi confirmada por exames necroscópicos, histopatológicos e bacteriológicos em animais vitimados pela doença. Os bovinos infectados foram submetidos ao tratamento com isoniazida pura, cristalizada, administrada por via oral, na dose de aproximadamente 25 mg/kg pv, durante 60 dias consecutivos e mais 60 doses dadas, em dias alternados, 3 vezes por semana. Face ao grande número, não foi possível medicar cada animal individualmente, optando-se pelo tratamento massal, em lotes de até 80 animais cujo peso variava até 100 kg. A isoniazida era misturada mecanicamente na proporção de 12 g para 1 kg de ração concentrada com alta palatibilidade, fornecida aos animais em cochos coletivos duplos em que cada animal livremente procurava o alimento. A quantidade de ração era calculada a base do peso médio e do número de animais do lote, de modo que cada animal, ao comer o alimento, estava ingerindo sua dose terapêutica de isoniazida. Durante o período da medicação, os animais clinicamente portadores de sintomas de tuberculose, mostravam sua recuperação a partir do segundo mês em diante. Trinta dias após o término da medicação foi iniciado o monitoramento alérgico pela tuberculinização comparativa com intervalos de 2 a 3 meses. A cura da tuberculose foi avaliada pela gradual dessensibilização à tuberculina. Na maioria dos animais tratados a reação caiu para níveis negativos entre 2 e 4 meses após o término da medicação. No final do trabalho experimental verificou-se que 96,4% dos animais tratados estavam curados. A cura foi confirmada pela inspeção sanitária em animais abatidos em matadouro.
