E-mail: relisampa@terra.com.br Due to the increasing use of cortical bone allografts in orthopedic surgeries, de knowledge of its biomechanics characteristics during preservation time is needed. The present study consisted in the analyses of the resistance power to the micro-traction of samples of rabbit cortical bones preserved in several means for up to 180 days and fresh. The results showed that the resistance and the preservation time presented an inversely proportional relation, meaning that, the longer the time of preservation, the shorter the physical resistance evaluated in the biomechanical rehearsal of resistance to the micro-traction. The glycerin has presented lower values in relation to the resistance test, showing, after 30 days, only 24.58% of the present power in the fresh bone, and by 180 days, 1.76%. The samples submitted to autoclavation also showed low values by the end of the experiment, while they remained with just 12.31% of the power present in the fresh bone. The bones preserved in homologous plasma, Dakin liquid and the ones cryopreserved showed the best levels of resistance at the end of the experiment, remaining, respectively with 82.47, 70.34 and 66.72% of the maximum power, while compared with the resistance of the fresh bones. The conclusion is that the choice of methodology and time of preservation interfered directly in the biomechanics of the cortical bones, promoting decrease of the resistance capacity to the traction along the period of preservation.

E-mail: deferro@usp.br The development of biologic modalities designed to enhance bone regeneration and wound healing of specific periodontal sites continues to be a challenge and has been accomplished through the cell binding activity of Type-I collagen. These have been provided by a synthetic cell biding peptide (P-15), associated to a anorganic bone matrix (ABM) to form ABM/P-15. The aim of this study was to evaluate the attachment loss and periodontal pocket response in teeth after 3 and 6 months with ABM/P-15 graft application. Twenty one dogs from the Veterinary Hospital, University of São Paulo, were anesthetized in order to accomplish periodontal treatment and 132 teeth faces with attachment loss were treated. From these, 36.4% (48 faces) received cell binding peptide and 63.6% (84 faces) compounded the control group that received conventional treatment (muco-gingival flap and root planning). The procedure was documented by intra-oral radiography and all periodontal probings were photographed. After 3 and 6 months, the animals were re-anesthetized in order to accomplish new photography, radiography and periodontal probing exams. The 48 attachment loss faces that received graft material exhibited 40% of regeneration rate after 6 months. The control faces did not change their attachment level. The palatal face presented the better regeneration rates (40%) and the canines and molars teeth showed the better responses (57.14% and 65%, respectively). There was no post-surgical infection related to absence of oral home care. It can be concluded that ABM/P-15 helps a more rapidly periodontal structure re-attachment and regeneration, including alveolar bone. Its application was easy and practical, and the post-surgical complications incidence was low. Nevertheless, more work is necessary to evaluate the amount and the quality of formed bone and periodontal ligament.

Mimosoideae).] Intoxicação aguda e abortos em cobaias pelas favas de Enterolobium contortisiliquum (Leg. Mimosoideae). Pesquisa Veterinária Brasileira 28(12):593-596. Pods of E. contortisiliquum were administered orally to 4 groups of 3 guinea pigs each. Another group of 3 guinea-pigs was used as control. Group 1 and 2 were fed with one dose of 5 and 10g of pods for kg body weight, respectively. The guinea pigs of Group 3 and 4 received 10 and 15g/kg, respectively, divided into daily doses of 5g/kg. One guinea pig from Group 2 and one from Group 4 died 12 and 18 hours after the end of the administration. Gross lesions were hemorrhages of the stomach and of the large and small gut, enlarged liver, and dilated gall bladder. Histologically, the liver had severe vacuolation and necrosis of periportal hepatocytes. In another experiment a ration containing 4% of pods of E. contortisiliquum was fed to 2 groups of 4 guinea-pigs, 35 days after mating. Four of the 8 guinea pigs aborted 6-15 days after the beginning of ingestion. The other 4 guinea pigs were not pregnant. All guinea pigs were euthanized after abortion or at the end of the experiment. Histologically all animals had mild to severe periportal hemorrhagic necrosis. All fetuses had variable degree of autolysis. In 4 fetuses studied no significant histologic lesions were observed. The acute lesions observed in guinea-pigs are similar than those observed in the spontaneous poisoning by Enterolobium spp. in cattle. Similar lesions are observed in guinea-pigs poisoned experimentally with saponins from E. gummiferum. The results of the experiments in pregnant guinea pigs suggest that E. contortisiliquum can be used to study the abortive effect of its pods or its toxic compounds.

Twelve 5-month-old male lambs maintained on alfalfa hay (3% BW) and non-fluorinated water ad libitum were used. Animals were allocated into Control, receiving 5g NaCl/animal/day + 0.2mg I/kg dry matter) and Treated group, receiving the same treatment plus sodium fluoride (4.7mg F/kg body weight). Mineral treatment was given by gavage, daily for 150 days. Blood, urine and fecal samples were collected during and the end of the experiment. At the end of treatment period animals were euthanized and kidney, pineal and bone samples were collected. Urine F was higher in treated animals throughout the experiment. Bone F levels were also increased in treated animals; pineal F content however, was not different between groups. Kidney histology revealed no differences. It is concluded that chronic F administration induces accumulation of the element in the skeleton. However such fact appears not to be detrimental to animals. Rates of F accumulation in bone and urine excretion obtained in other species can not be used in growing lambs.

Therefore this study aimed to compare the velocity of bone callus formation in the treatment of fracture repair with plates and screws alone or with plates and screws plus BMP. Thirty three dogs with radius and ulna fractures were distributed into two groups, where animals of the control group received the conservative treatment performed with screws and plates alone, whilst the other group received the conservative treatment and BMP. The time of bone callus formation was evaluated comparatively through radiographic exams 30, 60, 90, 120, 180 and 210 days after the surgical procedure. Animals treated with BMP showed a healing time of 32±15 days, which was significantly different (p <0.001) from the control group which required 127±34 days. With the results obtained, it can be concluded that the distal radio-ulna fractures of dogs weighing less than 6 kg suffered a significant reduction of the bone callus formation time, which was around 90 days.

S. glaucophyllum) (Sm) during 9 days. The Sm-poisoned rabbits had an increase of bone resorption in the endosteal surface of the cortical zone and also in the surface covered by osteoblasts of the primary and secondary spongiosa of the trabecular bone compartment. Moreover, the epiphyseal growth plates in long bones appeared narrower than in the control rabbits, with reduction of the proliferative and hyperthrophic chondrocyte zones. The electron microscopic study revealed a significant decrease of proteoglycans in the hyperthrophic chondrocyte zone evidenced by a significant reduction of rutenium red positive granules in the poisoned rabbit. Altogether, these data suggest that cell differentiation may play a pivotal role in the pathogenesis of Sm-induced bone lesions.

E-mail: bonel-raposo@brturbo.com Green leaves of Myoporum laetum were collected during spring and summer, and administered to five sheep and six steers at dosages of 20 and 30 g/kg. Liver biopsies were taken before (Controls) and 1, 3 and 7 days after dosage. ln sheep, the clinicai signs were depression, rumen hypomotility, dried feces, tenesmus, teeth grinding, dyspnea and typical lesions of photosensitization. ln cattle, the clinicai picture was much less pronounced. The main histological findings in sheep were vacuolization of hepatocytes, portal ftbrosis, bile duct proliferation and necrosis of periportal hepatocytes; the ultrastructural examination revealed hyperplasia of the smooth endoplasmic reticulum, hepatocellular hydropic degeneration, presence of crystals and severa! other degenerative changes. ln cattle both, the histological and the ultrastructural findings, were less evident.

E-mail: bonel-raposo@brturbo.com Amostras de Myoporum laetum foram colhidas durante a primavera e verão e administradas a cinco ovinos e seis bovinos em doses únicas de 20 e 30 g/kg. Biópsias hepáticas foram colhidas antes (controles) e 1, 3 e 7 dias após a dosagem da planta. Estas biópsias foram analisadas histológica e ultraestruturalmente. Os sinais clínicos, em ovinos, caracterizaramse, especialmente; pordepressão, diminuição dos movimentos ruminais, fezes ressequidas, tenesmo, ranger de dentes, dispnéia e lesões típicas de fotossensibilização. Em bovinos, o quadro clínico foi discreto. Os principais achados histológicos, em ovinos, incluíram vacuolização de hepatócitos, fibrose portal, proliferação de duetos biliares e necrose de hepatócitos periportais. Os estudos ultra-estruturais, em ovinos, revelaram hiperplasia do retículo endoplasmático liso, tumefação de hepatócitos, degranulação e vesiculação do retículo endoplasmático rugoso, presença de cristais aciculares, retenção biliar, tumefação de mitocôndrias e várias outras alterações degenerativas. Em bovinos, tanto os achados histológicos, quanto os ultra-estruturais foram menos evidentes.

[Clinical and hematological evaluation of Nelore calves experimentally infected with isolates of Babesia bigemina from the Southeastern, Northeastern and Northern regions of Brazil.] Avaliação clínica e hematológica em bezerros Nelore infectados experimentalmente com isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil. Pesquisa Veterinária Brasileira 23(2):52-60. Clínica de Bovinos, Campus Garanhuns, Univ. Fed. Rural de Pernambuco, Cx. Postal 152, Garanhuns, PE 55292-901, Brazil. A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animais were previously inoculated with 2.0x109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animais each, that were subinoculated with 1.0x1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen,reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 151 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low leveis of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

O presente trabalho teve por objetivo estudar comparativamente as alterações clínicas e hematológicas desencadeadas por isolados de Babesia bigemina das regiões Sudeste, Nordeste e Norte do Brasil em bezerros Nelore infectados experimentalmente. Foram utilizados 18 bezerros com idade entre sete e nove meses, isentos de anticorpos contra Babesia sp. e criados livres de carrapatos. Três animais foram previamente inoculados com 2,0x109 eritrócitos parasitados (EP) para cada isolado. Os outros 15 bezerros foram subdivididos em três grupos de cinco animais, que foram subinoculados com 1,0x1010 EP dos respectivos isolados. Foram avaliadas as alterações clínicas e hematológica por meio da determinação da parasitemia, do hemograma, do fibrinogênio plasmático, da contagem de reticulócitos, da análise descritiva da medula óssea e da fragilidade osmótica eritrocitária, no decorrer de 30 dias, perfazendo um total de sete momentos de observação. O acompanhamento da resposta imunológica pelo teste de imunofluorescência indireta foi realizado diariamente até o 10º dia pós-inoculação (DPI) e posteriormente no 15º, 20º, 25º e 30º DPI. Clinicamente, observou-se uma manifestação muito branda da doença. Os achados laboratoriais revelaram baixos níveis de parasitemia; decréscimo nos valores do eritrograma; ausência de reticulócitos; diminuição inicial na contagem total dos leucócitos, neutrófilos e linfócitos com posterior elevação do número destas células; hipercelularidade da série eritrocítica e decréscimo da relação mielóide:eritróide mais acentuada entre o 8º e 12º DPI e um aumento da fragilidade osmótica eritrocitária nos grupos inoculados com os isolados sudeste e nordeste. Nenhum dos três isolados de B. bigemina desencadeou a forma clínica característica da enfermidade, apesar de induzirem uma resposta imune humoral.

The main limitation in the use of rock phosphate in swine diets is the fluorine contente that may be toxic when used for long periods. The objective of this study was to evaluate the effects of monocalcium phosphate (MP) and Tapira phosphate (TP) to sows and their offspring when fed during four reproductive cycles, as compared to the traditional use of dicalcium phosphate (DP). The study involved 13, 16 and 13 sows for DP, MP and TP phosphates, respectively. The litters were also fed with the sarne respective phosphate treatments until they reached 95 kg of body weight. The variables analyzed were: macroscopic tooth lesions score in sows, histological evaluation of bone tissue, the thyroid and parathyroid glands, and the evaluation of bone mineral contents and bone morphometrical characteristics. The results showed that TP is toxic for sows when used continuously during 4 reproductive cycles and causes lesions of chronic fluorosis in the teeth and bone tissue. TP also affected the litters with some minor lesions of fluorosis in bone tissue at the 4th reproductive cycle. MP induced some minor inespecific lesions only in bone tissue of the sows. No deleterious effects were observed in sows and litters fed DP.

A limitação principal do uso de fosfatos naturais na alimentação de suínos é a presença de flúor que pode ser tóxico quando ingerido por longos períodos. O objetivo deste trabalho foi avaliar os eventuais efeitos deletérios dos fosfatos Monocálcico (MP) e Tapira (TP), sobre porcas e suas proles alimentadas com esses fosfatos durante quatro ciclos reprodutivos consecutivos, comparativamente com o tradicional fosfato bicálcico (DP). Foram usadas 13 porcas alimentadas com FB (testemunha) 16 com MP e 13 com TP. As leitegadas oriundas de cada ciclo também foram alimentados com os respectivos fosfatos até a idade de abate. As seguintes variáveis foram avaliadas: lesões macroscópicas nos dentes das porcas, alterações histológicas no tecido ósseo e nas glândulas tireóide e paratireóide, conteúdo de minerais no tecido ósseo e características morfométricas do osso. Os resultados obtidos mostraram que o uso de TP por 4 ciclos reprodutivos, na alimentação de suínos, provocou lesões de fluorose crônica nos dentes e tecido ósseo das porcas. As leitegadas alimentadas com este fosfato também apresentaram discretas lesões de fluorose, mas somente evidentes no 4° ciclo reprodutivo. Somente algumas alterações inespecíficas ocorreram nos ossos das porcas que receberam o MP.O DP não provocou alterações patológicas no tecido ósseo, tanto nas porcas como nas suas leitegadas.


#60 - Morphologicmorphometrical and microradiographical studies on ribs of cattle supplemented with rock phosphate

Abstract in English:

The present study was conducted to evaluate possible bone changes in cattle due to the ingestion of different levels of fluorine from rock phosphate used as a phophorus supplement. The bone material studied were ribs collected from cattle of three experiments where Tapira rock phosphate was used at different levels and during different periods as a partial or sole source of supplementary phosphorus. In the first experiment feedlot cattle ingested during a 6 month period 63 and 128g/day of Tapira rock phosphate containing 1.3% fluorine. In the second experiment cattle maintained in Brachiaria decumbens pastures during 33 months ingested several levels of Tapira rock phosphate in the mineral mixtures. In the third experimente heifers averaging 14 months of age ingested a mineral supplement with Tapira rock phosphate until the 5th lactation period. Careful examination of the rib samples by histological, morphometric and microradiographic techniques revealed no abnormalities which could be related to fluorine ingestion, neither any differences among tretaments regarding the bone structure. These findings support the conclusion that, as far as bone alterations are concemed, Tapira rock phosphate could be used as a source of phosphorus for cattle under the conditions described in the experiments, a practice which would markedly reduce the cost of phosphorus supplementation.

O objetivo do presente trabalho foi o de avaliar, através de técnicas especializadas, as possíveis alterações ósseas produzidas em bovinos em decorrência da ingestão prolongada de diferentes níveis de flúor contido no fosfato de rocha de Tapira utilizado como fonte suplementar de fósforo. No primeiro experimento bovinos confinados ingeriram, durante 6 meses, quantidades variáveis (63 e 128g/dia) de fosfato de Tapira contendo 1.3% de flúor. No segundo experimento, bovinos em pastos de Brachiaria decumbens ingeriram, durante 33 meses, misturas minerais contendo diferentes níveis de fosfato de rocha de Tapira. No terceiro experimento, novilhas com idade inicial média de 14 meses ingeriram mistura mineral com fosfato de Tapira até a quinta lactação inclusivamente. Através de exames histológicos, morfométricos e microrradiográficos das amostras de costelas, não se observaram alterações da normalidade óssea, bem como não foram registradas diferenças entre amostras provenientes de diferentes tratamentos. Tais achados permitem inferir que, do ponto de vista de alterações ósseas, o fosfato de rocha de Tapira pode ser utilizado como fonte suplementar de fósforo para bovinos, nas dosagens, períodos e manejos alimentares estudados, sem risco de produzir alterações patológicas nos esqueleto dos animais.
