RESUMO.- Rissi D.R. & Barros C.S.L. 2010. [Spontaneous poisoning by ionophores in sheep in Rio Grande do Sul, Brazil.] Intoxicação espontânea por antibióticos ionóforos no Rio Grande do Sul. E-mail: claudioslbarros@uol.com.br Um surto de intoxicação espontânea por antibióticos ionóforos em ovinos da região Central do Rio Grande do Sul é descrito. Os 16 ovinos afetados estavam em campo nativo e ingeriram acidentalmente um aditivo alimentar para frangos contendo 250g/kg de narasina. Os sinais clínicos consistiam de fraqueza, incoordenação, dispnéia, secreção nasal, decúbito e morte em poucas horas. Um ovino apresentou urina escura. Macroscopicamente havia ascite, hidrotórax, edema pulmonar e palidez hepática. Discreto grau de degeneração muscular na musculatura esquelética dos membros pélvicos e torácicos foi observado histologicamente. O diagnóstico de intoxicação por narasina foi realizado com base no histórico (ingestão de aditivo alimentar contendo narasina) e nos achados clinico-patológicos.

E-mail: claudioslbarros@uol.com.br An outbreak of spontaneous ionophore toxicity in sheep grazing in native pasture in Rio Grande do Sul state, Brazil is described. Sixteen sheep which had accidental access to a chicken feed additive containing 250g/kg of narasin were affected. Clinical signs consisted of weakness, incoordination, dyspnea, nasal discharge, recumbency, and death in a few hours. One sheep showed dark red urine. Grossly there were ascites, hydrothorax, pulmonary edema, and hepatic paleness. Discrete skeletal muscle degeneration was observed histologically in the muscles of the pelvic and thoracic limbs. The diagnostic of narasin toxicosis was based on history (ingestion of feed additive containing narasin), clinical, and pathological findings.

Ionophores are used as food additives as coccidiostacts in several animal species and growth promoter and bloat prevention in ruminants. The most often used ionophores are monensin, lasalocid, narasin and salinomycin. There is a great variation in the susceptibility to the toxic effect of ionophores in different animal species. Poisoning can occur when the dosage is too high or when not correct doses for a certain animal species are given. Cases of poisoning have been described in sheep, swine, horses, dogs and poultry. For horses ionophores are extremely toxic. The use of ionophores is only safe when used accordingly to the instructions of the manufacturer and especially for each animal species. In this paper the most important data regarding clinical-pathological and pathogenic aspects, and also the conditions in which the poisoning may occur are critically reviewed.

#3 - Experimental narasin poisoning in sheep

Abstract in English:

Experimental poisoning by narasin was induced in 13 sheep through oral administration of the drug. Six sheep died, three were killed in extremis and four recovered. The onset of clinical sigos was 2 hours to 7 days after the administration of the drug, and the clinical course lasted from 7 hours to 14 days. Initially there were laborious breathing, tachypnea, fever and ruminal atony. These sigos were followed by incoordinated gait, stiffness, reluctance to move and changes in frequency and intensity of cardiac sounds. Grunting, grinding of teeth, myoglobinuria, and sternal and lateral recumbency were also observed. One sheep had sudden death. Necropsy findings were observed in seven of the sheep that died and consisted of pale areas in the skeletal muscles and myocardium. More frequently affected muscles were masseter, semitendinosus, sternocephalicus, brachiocephalicus, extrinsec muscles of the tongue, pectoral, supraspinatus and teres major. Hydropericardium, edema of the lungs and the wall of the gall bladder were also observed. Ocasionally the cut surface of the lymph nodes was reddened and moist. In one sheep there was edema surrounding the esophagus and within the cervical muscles; in another one there was a bolus of partially chewed food in the oral cavity. Histologically, the pale areas in the skeletal muscles and myocardium corresponded to multifocal or focal extensive degeneration and necrosis of myofibers; these lesions were sometimes associated with processes of regeneration in skeletal muscles and reparation in the myocardium. The histological muscle lesions were more pronounced in the tongue and muscles of the limbs.

Intoxicação pelo antibiótico ionóforo narasina foi induzida experimentalmente em 13 ovinos; desses, seis morreram espontaneamente, três foram sacrificados in extremis e quatro se recuperaram. O início dos sinais clínicos ocorria entre 2 horas e 7 dias após a administração da droga e persistiam por 7 horas a 14 dias. Inicialmente havia respiração ofegante e entrecortada, taquipnéia, febre, redução temporária do apetite ou anorexia e atonia ruminal seguidos de alterações no andar, como arrastar das pinças, apoio sobre os boletos dos membros posteriores, incoordenação, rigidez, relutância em movimentar-se e alteração na frequência e intensidade dos sons cardíacos. Ocorriam também gemidos, ranger de dentes, urina acastanhada e decúbito esternal ou lateral. Um animal apresentou morte súbita. Alterações macroscópicas foram observadas em sete dos ovinos necropsiados e consistiam de áreas pálidas nos músculos esqueléticos e no miocárdio. Os músculos mais frequentemente atingidos foram masseter, semitendíneo, esternocefálico, braquicefálico, extrínsecos da língua, peitoral, supra-espinhal e redondo maior. Havia hidropericárdio, edema e congestão pulmonares, fígado pálido, edema da parede da vesícula biliar, linfonodos ocasionalmente avermelhados e suculentos ao corte, edema periesofágico e entre os feixes musculares da região cervical e acúmulo de alimento semimastigado acumulado na cavidade oral. Histologicamente as áreas pálidas dos músculos esqueléticos e do miocárdio correspondiam a lesões degenerativo-necróticas multifocais a focalmente extensas associadas ou não a processos regenerativos, nos músculos esqueléticos, e reparativos, no miocárdio. As lesões histológicas foram mais acentuadas nos músculos da língua e dos membros.


#4 - Experimental narasin poisoning in cattle

Abstract in English:

Experimental poisoning by narasin was induced in 11 calves through oral administration of the drug. Five calves died, one was killed in extremis and 5 recovered after presenting mild clinical signs of the toxicosis. The onset of clinical signs was 12 hours to 4 days after the administration of the drug and clinical courses lasted from 24 hours to 9 days. Clinical signs were anorexia, decreased intensity and frequency of ruminal movements. Consistency of the feces varied from dry to semiliquid or liquid. Affected calves were reluctant to move, had incoordinated and stiff gait, and dragged the point of the hooves while walking. There was sluggish masticatory movements with drooling of saliva and dysphagia. The intensity of cardiac sounds were increased and, in one case, the jugular was distended and pulsating. Some animais were depressed, presented dyspnea, polydipsia and groanings. One calf presented myoglobinuria. Main necropsy findings were in the myocardium and consisted of focal or diffuse pai e irregular areas. Signs of heart failure such as pulmonary edema, hydrotorax and clotted blood in the left ventricle of the heart were also observed. Degenerative lesions characterized by pai e areas were seen in the skeletal muscles of three calves, but were marked in only one. Most affected muscles were masseter, extrinsec muscles of the tongue, tensor fasciae latae, semitendinosus, semimembranosus, psoas, quadriceps femoris, infraspinatus and gastrocnemius. Main histopathological changes consisted of degenerative myopathy and cardiomyopathy characterized by vacuolization of myofibers, hyalin and flocular necrosis either associated or not with reparative events. Myofiber regeneration and fibrosis were observed respectively in the skeletal muscles and in the myocardium.

Intoxicação por narasina foi induzida pela administração oral da droga a 11 bovinos. Desses, cinco tiveram morte espontânea, um foi sacrificado in extremis e outros cinco apresentaram sinais clínicos da toxicose, mas se recuperaram. Os sinais clínicos iniciaram entre 12 horas e 4 dias após ingestão da narasina e a duração do curso clínico variou de 24 horas até 9 dias. Houve anorexia, redução na intensidade e número de movimentos ruminais, fezes pastosas, ressequidas ou diarréicas. Os animais apresentavam relutância em movimentar-se, tinham andar incoordenado e rígido, e caminhavam arrastando as pinças dos membros posteriores. Em alguns a mastigação era lenta, com salivação abundante e havia disfagia. A intensidade dos sons cardíacos estava aumentada e, em um caso, ocorreu discreto pulso positivo da jugular. Alguns animais apresentaram depressão, gemidos, dispnéia, respiração abdominal e polidipsia. Em um caso houve mioglobinúria. As principais alterações observadas à necropsia localizavam-se no miocárdio e consistiam de áreas pálidas irregulares focais ou palidez difusa. Sinais de insuficiência cardíaca como edema pulmonar, hidrotórax, edema subcutâneo de declive e sangue coagulado no ventrículo esquerdo foram também observados. Alterações degenerativas representadas por áreas pálidas foram vistas nos músculos esqueléticos de 3 bovinos, mas de forma marcada somente em um. Os músculos mais envolvidos foram masseter, extrínsecos da língua, tensor da fáscia lata, semitendíneo, semimembranáceo, psoas, quadriceps femoris, músculos da face medial da escápula, grande dorsal e gastrocnêmio. A histopatologia consistiu de miopatia e cardiomiopatia degenerativas. Regeneração de miofibras e fibrose foram observadas, respectivamente, nos músculos esqueléticos e no miocárdio.


#5 - An outbreak of narasin poisoning in swine

Abstract in English:

An outbreak of narasin poisoning in swine is described. The diagnosis was based on the history, clinical-pathological findings, the reproduction of the disease by the administration of the feed originally given to the animals and by chemical analysis which showed doses sufficiently high to cause poisoning. Inspite that the clinical-pathological picture of the natural and experimental poisoning was of the sarne nature, there were some differences. In the natural outbreak lethality was high, but in the experiments none of the animals died. In the natural cases besides the lesions in the muscles, also lesions of the heart muscle fibres were seen, not detected in the esperimental animals. lt is suggested that these differences could be due to the stress to which the animals were submitted in the piggery by the great number of animals per box, whereas the experimental animals were kept individually.

Descreve-se um surto de intoxicação por narasina em suínos, cujo diagnóstico baseou-se no histórico, quadro clínico-patológico, na reprodução da enfermidade pela experimentação com a ração comercial originalmente oferecida aos suínos, bem como pelo exame químico posterior que detectou narasina em quantidades suficientemente elevadas para causar intoxicação. Apesar de os quadros clínico-patológicos das intoxicações natural e experimental serem da mesma natureza, houve algumas diferenças. Nos casos naturais o índice de letalidade foi elevado, enquanto que na intoxicação experimental nenhum animal morreu. Os exames histopatológicos revelaram, em parte dos casos naturais, além das lesões dos músculos estriados, também lesões nas fibras musculares cardíacas, últimas não verificadas nos casos experimentais. Sugere-se que essas diferenças poderiam ser explicadas pelo estresse ao qual os animais eram submetidos na granja pelo grande número de animais nas baias, ao contrário dos animais experimentais que eram mantidos em compartimentos individuais.
