#1 - Biochemical, endocrine, and histopathological profile of liver and kidneys of sheep with pregnancy toxemia

Abstract in English:

The objective of the present study was to characterize the biochemical, hormonal, and mineral profile, and histopathology of the liver and kidneys, related to the severity of natural cases of pregnancy toxemia (PT) in sheep. A total of 45 sheep with PT were analyzed at the “Clínica de Bovinos”, Campus Garanhuns-UFRPE. The animals were submitted to clinical examination, followed by the collection of blood and urine. A necropsy was performed on thirteen animals that died and a histopathological examination was performed on samples of liver and kidneys. Increased creatinine, urea, glucose, fructosamine, non-esterified fatty acids, β-hydroxybutyrate, cortisol, chlorine, amylase, aspartate aminotransferase, gamma-glutamyltransferase, and folic acid were found, whereas insulin, potassium, and total and ionizable calcium presented low values for the species. Total protein, albumin, globulin, sodium, magnesium, and vitamin B12 remained within the normal range. In the macroscopic analysis of the liver, it was possible to observe an increase in organ and border size, yellowish coloration and parenchyma that varied from firm to friable. Vacuolation of the hepatocytes and renal tubular cells was observed. Metabolic disorders in sheep associated with hepatic and renal lesions are more apparent in overweight animals. With the greater impairment in hepatic and renal function, the clinical prognosis of animals with a high body score should be considered reserved.

Objetivou-se caracterizar o perfil bioquímico, hormonal, mineral e a histopatologia hepática e renal, relacionando com a severidade de casos naturais de toxemia da prenhez (TP) em ovelhas. Analisou-se 45 ovelhas na Clínica de Bovinos, Campus Garanhuns-UFRPE, com TP. Foram submetidas ao exame clínico, seguido de coleta de sangue e urina. Treze animais que vieram a óbito foram necropsiados, nas amostras de fígado e rins realizou-se exame histopatológico. Revelou-se elevação de creatinina, ureia, glicose, frutosamina, ácidos graxos não esterificados, β-hidroxibutirato, cortisol, cloro, amilase, aspartato aminotransferase, gama glutamiltransferase e ácido fólico, enquanto insulina, potássio, cálcio total e ionizável apresentaram valores abaixo para os da espécie. A proteína total, albumina, globulina, sódio, magnésio e vitamina B12 mantiveram-se dentro da normalidade. Na análise macroscópica do fígado, pôde-se observar aumento de tamanho do órgão e dos bordos, coloração amarelada e parênquima que variava de firme a friável. Observou-se vacuolização dos hepatócitos e das células tubulares renais. Os transtornos metabólicos nas ovelhas associados às lesões hepáticas e renais são mais aparentes nos animais com sobrepeso. Com o maior comprometimento da função hepática e renal, o prognóstico clínico dos animais com escore corporal elevado há de ser considerado reservado.


#2 - Evaluation of serum concentrations of cortisol and thyroxine in bitches with early-stage mammary carcinoma, 38(10):1949-1954

E-mail: sofiabcrivellenti@ufu.br In view of the fact that cancer is considered a chronic disease that can interfere with hormonal homeostasis by means of cytokines, we hypothesized that, even at early stages, mammary carcinoma is able to alter the balance of the hypothalamic-pituitary-thyroid and hypothalamic-pituitary-adrenal axes. To test this hypothesis, the serum concentrations of basal cortisol, thyroxine (T4), free thyroxine (fT4), and thyroid-stimulating hormone (TSH) were evaluated in 20 unspayed bitches that had a histopathological diagnosis of grade 1 mammary carcinoma at clinical stage I according to the World Health Organization (WHO) classification (T1N0M0). The control animals comprised 10 unspayed bitches in perfect health conditions that were matched with those with mammary carcinoma by age. No significant differences regarding the concentrations of basal cortisol, TSH, t4, and fT4 were found between the bitches carrying early stage mammary carcinoma when compared to the control group. This suggests that, even if malignant, early-stage mammary carcinomas do not exhibit the ability to alter the concentrations of hormones produced by the hypothalamic-pituitary-adrenal or hypothalamic-pituitary-thyroid axes.

Para tal, foram avaliadas as concentrações séricas de cortisol basal, tiroxina (T4), tiroxina livre (fT4) e tireotrofina (TSH) de 20 fêmeas caninas, inteiras, com diagnóstico histopatológico de carcinoma mamário grau 1 e estadiamento clínico I segundo a classificação da Organização Mundial da Saúde - OMS (T1N0M0). Os animais controle constituíram-se por 10 fêmeas caninas inteiras, em perfeitas condições de higidez, as quais foram pareadas, por idade, com aquelas portadoras de carcinoma mamário. Não foram encontradas diferenças significativas nas concentrações de cortisol basal, TSH, T4 e fT4 das cadelas portadoras de carcinoma mamário em estádio inicial quando comparadas às controles sugerindo que, mesmo considerados malignos, ainda não apresentam a capacidade de alterar as concentrações dos hormônios produzidos pelos eixos hipotalâmico-hipofisário-adrenal e tireóideo.


#3 - Adrenal lesions in 300 dogs

Abstract in English:

The adrenal glands of 300 dogs, underwent to necropsy in the Laboratório de Patologia Veterinária at the Universidade Federal de Santa Maria (LPV/UFSM), were studied from March 2012 to June 2013. The collected tissues were fixed in 10% buffered formalin and sent to the routine histological processing in order to preparation and analysis of histological slides. Subsequently, it was reviewed the necropsy reports and the epidemiological data of the dogs which had the adrenal glands collected. Out of the 300 dogs examined, 82% had one or more adrenal lesions that were classified according to the pathogenesis and prevalence. The hyperplastic lesions were the most prevalent, comprising 91.5% of the cases, followed by degenerative (26%), circulatory (20.3%), neoplastic (15%), inflammatory (4.1%) and other lesions (14.2%). Grossly, hyperplasia was characterized by a variable amount of whitish nodules, ranging from 0.2-1.5 cm of diameter. In 72.9% of the hyperplasia cases, the nodules were composed by cells of the glomerulosa zone. Most of the animals that showed hyperplastic lesions consisted of females (60.4%), elderly (46.9%) and purebred dogs (55.6%). Fatty degeneration was the most prevalent degenerative lesions (54.7%), characterized by cells of the fasciculata and reticularis zones with large cytoplasmic vacuoles. Among the neoplastic changes, primary tumors were the most prevalent (75.7%), with predominance of adenomas (57.1%), followed by pheochromocytomas (25%) and cortical carcinomas (17.9%). Metastatic tumors corresponded to 24.3% of all cases of neoplastic changes. The adrenal glands were the site of metastasis of mammary gland carcinoma, osteosarcoma, melanoma, lymphoma, cholangiocarcinoma, histiocytic sarcoma and lung adenocarcinoma. Under “other lesions”, adrenal of stress was the most prevalent change (37.1%). It was characterized histologically by a cortex with irregular appearance, with areas of mild cell vacuolation in the fasciculata zone, interspersed with hypereosinophilic cell groups. This study allowed the conclusion that the lesions in adrenal glands of dogs are extremely common, highlighting the importance of a careful evaluation of this organ in the routine of pathological diagnostic.

Foram estudadas as adrenais de 300 cães submetidos à necropsia no Laboratório de Patologia Veterinária da Universidade Federal de Santa Maria, no período de março de 2012 a junho de 2013. Os tecidos coletados foram fixados em formol 10% e encaminhados ao processamento his­tológico de rotina para confecção e análise de lâminas histológicas. Posteriormente, foram revisados os protocolos de necropsia e dados epidemiológicos referentes aos cães que tiveram as adrenais coletadas foram anotados. Dos 300 cães analisados, 82% tinham uma ou mais alterações nas adrenais, que foram classificadas de acordo com a patogênese e a prevalência. Lesões hiperplásicas foram as mais prevalentes, com 91,5% das ocorrências, seguido de lesões degenerativas (26,%), circulatórias (20,3%), neoplásicas (15%), inflamatórias (4,1%) e outras lesões (14,2%). Macroscopicamente, as hiperplasias caracterizavam-se por variável quantidade de nódulos brancacentos, de até 1,5cm de diâmetro. Em 72,9% dos casos de hiperplasia, os nódulos eram constituídos de células da camada glomerulosa. A maioria dos animais que apresentou lesões hiperplásicas era constituída de fêmeas (60,4%), idosos (46,9%) e cães com raça definida (55,6%). A degeneração gordurosa foi a lesão degenerativa mais prevalente (54,7%) e era caracterizada por células da zona fasciculada e da zona reticular com grandes vacúolos citoplasmáticos. Entre as alterações neoplásicas, tumores primários foram mais prevalentes (75,7%), com predomínio de adenomas (57,1%), seguido de feocromocitomas (25%) e carcinomas corticais (17,9%). Os tumores metastáticos perfizeram 24,3% dos casos de alterações neoplásicas. As adrenais foram sítio de metástase de carcinoma mamário, osteossarcoma, melanoma, linfoma, colangiocarcinoma, sarcoma histiocítico e adenocarcinoma pulmonar. No item “outras lesões”, adrenal de estresse foi a alteração mais prevalente (37,1%). Essa alteração era caracterizada histologicamente por córtex de aspecto variegado, com áreas de fina vacuolização citoplasmática das células da camada fasciculada, intercaladas por grupos de células hipereosinofílicas. Este estudo possibilitou concluir que lesões nas adrenais de cães são extremamente frequentes, de modo que a avaliação cuidadosa desse órgão se faz necessária na rotina de diagnóstico em patologia.

E-mail: jcferreira@fmvz.unesp.br The aim of this study was to evaluate the response of blue-fronted parrots to restraint procedures and separation by non-invasive methods such as measurement of glucocorticoid metabolites in droppings. For this, we utilized 24 birds, 17 males and 7 females, initially kept in large adjacent aviaries, separated by sex. After capture and 3 minutes of manual contention, by random, 13 birds returned to the aviary and 11 animals were housed in individual cages in the same facility of the aviaries allowing the maintenance of auditory and visual contact between them. In order to evaluate if the physically isolated birds isolated or in groups would react in different ways, all droppings samples were collected at 3-hours intervals during 24 hours to evaluation of excreted glucocorticoid metabolites by enzimeimmunoassay. There were no significant effects of sex (P=0.5850), treatment (P=0.6805) and time (P=0.2293) and the glucocorticoid metabolites concentrations were within the diurnal range expected for this specie. Therefore, the endocrine response of both groups was similar and stress of capture and physical separation was not significant for the birds.

Para tanto, foram utilizadas 24 aves, 17 machos e sete fêmeas, inicialmente mantidas em viveiros amplos e adjacentes, separadas por sexo. Após captura e contenção de três minutos, 13 aves voltaram para os viveiros coletivos e 11 foram alojadas em gaiolas individuais no mesmo recinto dos viveiros, de modo a permitir que as aves isoladas mantivessem contato visual e auditivo com as demais. Para avaliar se os animais responderiam de maneira diferente quando fisicamente isolados ou em grupo, amostras de excretas foram coletadas sequencialmente em intervalos de três horas durante 24 horas para avaliação dos metabólitos de glicocorticoides por enzimaimunoensaio. Não houve efeito significativo de sexo (P=0,5850), tratamento (P=0,6805) e tempo (P=0,2293), e as concentrações de metabólitos de glicocorticoides mantiveram-se dentro da variação diurna esperada para esta espécie. Portanto, ambos os grupos responderam endocrinologicamente de forma semelhante e o estresse de captura e separação física não foi significativo para as aves.

E-mail: manuelafraga@hotmail.com The communication of the female reproductive status in Callithrichidae relies mainly on the socio-sexual behavior, as generally the ovulation is concealed in this primate family by a primary signaling system. In this study the socio-sexual behavior patterns was analyzed in association with the concentration of fecal metabolites of sex steroid hormones progesterone (MFP), estradiol (MFE) and testosterone (MFT) in captive couples of Black-Tufted-Marmoset (Callithrix penicillata), during the different phases of the ovarian cycle. The studied group was composed of four adult couples kept in the São Paulo City Wild Animals Rehabilitation Center. The behavioral patterns were record by focal samplings, with 30 seconds intervals for each observation, five days a week, totalizing 14.400 registers per animal. The measurement of fecal metabolites of progesterone (MFP), estradiol (MFE) and testosterone (MFT) proceeded by enzyme immune assay (EIA). The results allowed to determine the duration of the ovarian cycle and to characterize three different phases (follicular, periovulatory and luteal). It was possible to determine 31 complete cycles that lasted 24.3±4.1 days (Mean ± SD). The follicular and luteal phases lasted 13.04±4.8 and the luteal phase 11.2±4.2 days. The behavioral patterns (scent marking, sniff genitals, grooming and sexual presentation) were more prevalent in the periovulatory phase as the behavioral variable “proximity” as well. There were no variations in the concentration of MFT in the males during the period studied. The associated analyses of the fecal metabolite of sex steroids and the socio-sexual behaviors led to a better understanding of the factors involved in the reproduction of C. penicillata.

Neste trabalho, os padrões de comportamentos sócio-sexuais foram analisados em conjunto com as concentrações de metabólitos fecais dos esteróides sexuais progesterona (MFP), estradiol (MFE) e testosterona (MFT) em casais cativos de Sagüi-de-tufos-pretos (Callithrix penicillata), nas diferentes fases do ciclo ovariano. O grupo estudado era composto por quarto casais adultos, mantidos no Centro de Reabilitação de Animais Selvagens da prefeitura de São Paulo. Os padrões comportamentais foram registrados pelo método de amostragem focal por intervalo de tempo a cada 30 segundos, cinco vezes por semana, totalizando 14.400 registros por animal. A mensuração das concentrações de metabólitos fecais dos esteroides sexuais foram realizados pelo método de enzima imunoensaio (EIE). Os resultados obtidos dessas concentrações possibilitaram a determinação endócrina das fases do ciclo ovariano (folicular e luteal) e de suas respectivas durações, assim como a determinação da fase periovulatória. Foram caracterizados 31 ciclos ovarianos completos, com duração de 24,3±4,1 dias (média ±DP), sendo que a fase folicular compreendeu 13,04±4,8dias e a fase lútea 11,2±4,2 dias. Os comportamentos sócio-sexuais (marcação por cheiro, cheirar genitália, catação e apresentação sexual) e a variável “proximidade” mostraram-se significativamente mais prevalentes na fase periovulatória do que nas demais fases do ciclo. Não houve alteração das concentrações de MFT dos machos ao longo de todo o período estudado. A análise conjunta das concentrações de metabólitos fecais de esteróides sexuais e dos comportamentos sócio-sexuais possibilitou um melhor entendimento das relações endócrino-comportamentais e reprodutivas de C. penicillata.

Under physiological conditions the number of SOM-LI endocrine cells has been shown to constitute 3,30±0,22. All applied pathological processes resulted in changes in the SOM–like immunoreactivity, which varied in particular processes studied. The number of SOM-LI endocrine cells increased to 6,28±0,31 and 4,43±0,35 during chemically driven inflammation and proliferative enteropathy, respectively, and decreased to 1,17%±0,16 after axotomy. The obtained results suggest that SOM-LI endocrine cells may participate in various pathological states within porcine descending colon and their functions probably depend on the type of pathological factor.
