#1 - Effect of pre-anesthetic fasting on gastric emptying and plasma glucose in healthy dogs of different age groups

Abstract in English:

Gastric emptying and plasma glucose were evaluated in young and adult dogs, fed with dry and wet food, submitted to different periods of pre-anesthetic fasting (6, 8, and 12 hours). Forty healthy dogs were selected, which were segmented into four groups according to the age group and type of diet. It was evaluated the gastric emptying by ultrasound and serum glycemia. Only 17.5% presented complete gastric emptying, and no significant differences were found between the 6 and 8-hour fasting evaluations, or between the age groups and the diets, considering significance level p<0.05. Mean plasma glucose values from the groups indicated normal glycemia at all times of evaluation. A significant difference was found between the means of glycemia in young and adult dogs, with the 8-hour fasting with wet diet (p=0.03) and with 12 hours with dry diet (p=0.04). Healthy young and adult dogs, in physiological equilibrium, maintain average values of plasma glucose despite prolonged periods of pre-anesthetic fasting, which may be necessary, since 8-hour fasting for solid food is not enough to provide complete gastric emptying.

Avaliou-se o esvaziamento gástrico e a glicemia plasmática em cães jovens e adultos, alimentados com ração seca e úmida, submetidos a diferentes períodos de jejum pré-anestésico (6, 8 e 12 horas). Foram selecionados 40 cães hígidos, os quais foram segmentados em 4 grupos de acordo com a faixa etária e o tipo de dieta administrada. Foi avaliado o esvaziamento gástrico por ultrassonografia e a glicemia sérica. Apenas 17,5% apresentaram completo esvaziamento gástrico, não sendo encontradas diferenças significativas entre as avaliações com 6 e 8 horas de jejum, ou entre as faixas etárias e dietas, considerando nível de significância p<0,05. Os valores médios da glicose plasmática dos grupos indicaram normoglicemia em todos os momentos de avaliação. Foi encontrada diferença significativa entre as médias da glicemia dos cães jovens e adultos, no período de 8 horas de jejum com dieta úmida (p=0,03) e com 12 horas nos animais com dieta seca (p=0,04). Conclui-se que cães hígidos jovens e adultos, em equilíbrio fisiológico, mantêm valores normais de glicemia plasmática apesar de períodos prolongados de jejum pré-anestésico, os quais podem ser necessários, tendo em vista que 8 horas de jejum alimentar de sólidos não é suficiente para proporcionar completo esvaziamento gástrico.


#2 - Glucose and insulin curve in pregnant mares and its relationship with clinical and biometric features of newborn foals

Abstract in English:

The aim of the present study was to describe the dynamics of glucose and insulin curves in pregnant mares, and to evaluate the curves according to body condition score, identifying the presence of insulin resistance and correlating these values with the weight, height and clinical changes of the neonates. For this, pregnant mares were evaluated and then grouped according to body condition score during the gestation length until lactation. GrM corresponds to mares with moderate body score (BCS 5-6); GrOv were mares with overweight body score (BCS 7) and GrOb were obese mares (BCS 8-9). A two-step oral sugar test (OST) was used to determine the data. Cortisol analysis was performed with 300-320 days of gestation, at foaling and after parturition. For evaluation of the neonate, a general clinical examination and, weight and height measurements were performed. The results showed hyperglycemia in response to OST with normal insulin values at foaling with a subsequent fall in both values at lactation disregarding group division. Baseline glucose was decreased in GrM compared to GrOv and GrOb with 70-100 days of gestation and with 130-160 days of gestation. With 270-300 days of gestation and post-partum GrOb had increased baseline glucose than GrM. After OST, glucose at foaling day in GrOb presented increased values than GrM. Baseline insulin values did not differ between groups. Post OST insulin levels were higher in GrOb than GrM and GrOv at parturition. No difference in cortisol between moments was identified. GrOb and GrOv maintained increased concentrations after foaling while GrM had a decrease. No correlation was found between maternal glucose and insulin values with foal weight and height, however, a lower ratio between neonatal weight and mare’s weight in GrOb and GrOv was identified in relation to the GrM. At foaling, mares presented glucose dysregulation, with obese and overweight mares presenting a greater response to OST.

O objetivo do presente estudo foi descrever a dinâmica das curvas de glicose e insulina em éguas gestantes e avaliar as curvas de acordo com o escore de condição corporal, identificando a presença de resistência insulínica e correlacionando esses valores com o peso, altura e alterações clínicas dos neonatos. Para isso, as éguas prenhes foram avaliadas em conjunto e agrupadas de acordo com o escore de condição corporal durante a gestação até o pós-parto. GrM pertenciam éguas com escore corporal moderado (EC 5-6); GrOv, grupo de éguas com escore corporal acima do peso (EC 7) e GrOb, grupo de éguas obesas (EC 8-9). O teste de glicose oral em duas etapas (OST) foi usado para determinar os dados. A análise do cortisol também foi realizada nos 300-320 dias de gestação, no dia do parto e após o parto. Para avaliação do neonato, foram realizados exame clínico geral e medidas de peso e altura. Os resultados mostraram hiperglicemia em resposta ao OST com valores normais de insulina no momento parto, com uma queda subsequente em ambas as variáveis na lactação, desconsiderando a divisão do grupo. A glicemia basal diminuiu no GrM em comparação com GrOv e GrOb com 70-100 dias de gestação e com 130 160 dias de gestação. Com 270-300 dias de gestação e no pós-parto, o GrOb apresentou aumento na glicemia basal em relação ao GrM. Após OST, a glicose no dia do parto no GrOb apresentou valores aumentados em relação ao GrM. Os valores basais de insulina não diferiram entre os grupos. Após OST níveis de insulina foram maiores no GrOb do que GrM e GrOv no momento do parto. Não houve diferença nos valores de cortisol entre os momentos. O GrOb e GrOv mantiveram cortisol aumentado após o parto enquanto o GrM diminuiu. Não foi encontrada correlação entre os valores de glicemia e insulina materna com o peso e a altura do potro, entretanto, foi identificada uma relação menor entre o peso neonatal e o peso da égua no GrOb e GrOv em relação ao GrM. No parto, as éguas apresentaram desregulação da glicose, sendo que as éguas obesas e com sobrepeso apresentaram uma resposta maior ao OST.

Email: mege@fmvz.unesp.br Clinical evaluations of newborns, as well as defining the therapeutic decision, represent significant challenges to the veterinarian. This study aimed to evaluate neonatal vitality using the modified Apgar score and blood glucose, lactate, and cortisol concentrations in newborn equines. Twenty foals of the Paint Horse breed born by normal delivery were evaluated during the first 48 hours of life. The modified Apgar score was determined at birth and 10 minutes later, with values ranging from 0 to 10. Blood samples for the laboratory tests were collected at birth and at 4, 8, 12, 16, 20, 24, 36, and 48 hours. The mean Apgar score was significantly lower at birth than at 10 minutes (p < 0.001). Blood glucose concentrations increased significantly between birth (98.90 ± 35.97 mg/dL) and 8 hours (127.90 ± 27.15 mg/dL), stabilizing after the first 12 hours (148.95 ± 29.59 mg/dL). Serum lactate concentrations showed a significant decrease across the time points tested, with the highest value occurring at birth (5.48 ± 2.17 mmol/L) and stabilizing after 24 hours (2.76 ± 1.31 mmol/L). Cortisol concentrations significantly decreased (p < 0.01) between 4 (10.46 ± 7.46 µg/dL) and 48 hours (5.21 ± 3.35 µg/dL), with the highest value occurring 4 hours after birth. The common occurrence of laboratory abnormalities in healthy newborn foals reinforces the need to conduct regular clinical examinations and to develop laboratory reference values &#8203;&#8203;for different breeds and ages.

O objetivo deste estudo foi avaliar a vitalidade neonatal pelo escore de Apgar modificado, glicemia, lactatemia e cortisol em neonatos equinos. Para tal, foram utilizados 20 potros da raça Paint Horse, nascidos em eutocia, durante as primeiras 48 horas de vida. O escore de Apgar modificado foi realizado ao nascimento e 10 minutos após, atribuindo-se notas de 0 a 10. As colheitas das amostras de sangue, para a realização das análises laboratoriais, ocorreram nos momentos ao nascimento, 4, 8, 12, 20 16, 20, 24, 36 e 48 horas de vida. A média do escore de Apgar ao nascimento foi significativamente menor que aos 10 minutos (p<0,001). A glicemia aumentou significativamente entre o nascimento (98,90 ± 35,97 mg/dL) e às 8 horas (127,90 ± 23 27,15 mg/dL), com estabilização após as primeiras 12 horas (148,95 ± 29,59 mg/dL). A concentração sérica de lactato apresentou decréscimo significativo entre os momentos estudados, sendo o maior valor encontrado ao nascimento (5,48 ± 2,17 mmol/L), com estabilização a partir das 24 horas (2,76 ± 1.31 mmol/L). As concentrações de cortisol, reduziram significativamente (p<0,01), entre as 4 (10,46 ± 7,46 µg/dL) e as 48 horas (5,21 ± 3,35 µg/dL), sendo o maior valor encontrado as 4 horas após o nascimento. A ocorrência comum de anormalidades laboratoriais em potros recém-nascidos, aparentemente saudáveis, reforça a necessidade da realização de exames clínicos regulares e o desenvolvimento de valores laboratoriais de referência, em diferentes raças e idades.

E-mail: clarisse.coelho@uvv.br The aim of this study was evaluate the influence of physical exercise of submaximal intensity (marcha gait) on biochemical variables used for the evaluation of energetic metabolism in Mangalarga Marchador horses trained in Espirito Santo, Brazil. Serum and plasma samples were obtained from 15 horses in four different moments: before (T0), 5 minutes (T1), 30 minutes (T2) and 2 hours (T3) after the exercise. Results showed that the imposed exercise did not influence plasmatic glucose, with mean values of 117.1±5.8g/dL, 122.6±59.6mg/dL, 124.8±48.6mg/dL and 112.9±49.1mg/dL, and serum insuline, with median values of 6.50mUI/mL, 2.00mUI/mL, 5.85mUI/mL and 11.60mUI/mL, respectively on the moments T0, T1, T2 and T3. Differently, it was possible to observe a positive influence of the exercise over serum triglycerides, with mean values of 25.4±14.9mg/dL, 42.3±17.8mg/dL, 31.4±17.7mg/dL and 25.1±15.1mg/dL, and serum cortisol, with mean values of 7.46±4.37mg/dL, 12.45±3.08mg/dL, 11.40±2.52mg/dL and 6.89±1.78mg/dL, respectively on the moments T0, T1, T2 and T3. It was possible to conclude that the marcha gait leaded to an elevation of serum triglycerides and cortisol levels. Also, as the values found were within reference levels, it was possible to conclude that the horses were used to this physical activity imposed on the occasion.

A análise dos resultados demonstrou a não influência do exercício físico imposto sobre a glicose plasmática, com valores médios de 117,1±35,8mg/dL, 122,6±59,6mg/dL, 124,8± 48,6mg/dL e 112,9±49,1mg/dL, e sobre a insulina sérica, com valores de mediana de 6,50mUI/mL, 2,00mUI/mL, 5,85mUI/mL e 11,60mUI/mL, respectivamente, nos tempos T0, T1, T2 e T3. De forma oposta, foi possível observar uma influência significativa sobre triglicérides séricos, com valores médios de 25,4±14,9mg/dL, 42,3±17,8mg/dL, 31,4±17,7mg/dL e 25,1±15,1mg/dL, e sobre o cortisol sérico, com valores médios de 7,46±4,37mg/dL, 12,45±3,08mg/dL, 11,40±2,52mg/dL e 6,89±1,78mg/dL, respectivamente nos tempos T0, T1, T2 e T3. A interpretação destes resultados permitiu concluir que a marcha elevou as concentrações séricas de triglicérides e cortisol. Também foi possível destacar que, por tais valores encontrarem-se dentro de intervalos fisiológicos, os equinos usados estavam aptos ao nível de exercício físico imposto na ocasião.


#5 - Assessment of NADPH-diaphorase stained myenteric neurons of the jejunum of diabetic rats supplemented with ascorbic acid, p.95-102

Among the theories proposed for its etiology the oxidative stress stands out. The performance of nitric oxide as a link between the metabolic and vascular neuropathogenic factors that triggers the diabetic neuropathy has already been put forward. This study aimed to assess the quantification and measurements of the cell body profile area (CBPA) of NADPH-diaphorase reactive (NADPH-dp) myenteric neurons of the jejunum of diabetic rats (induced by streptozotocin) supplemented with Ascorbic Acid (AA). These changes in the myenteric neurons seem to be related to the gastrointestinal disturbances observed in diabetes mellitus (DM). Twenty male Wistar rats (Rattus norvegicus) were distributed in 4 groups (n=5): controls (C), control supplemented (CS), diabetic (D), and diabetic suplemented (DS). DM was induced by estreptozotocin (50mg/kg body wt). One week after the induction and confirmation of the DM (glycemia exam), animals of the groups CS and DS received 50mg of AA three times a week by gavage. After 90 days of experiment, the animals were anesthetized with lethal thiopental dose (40mg/kg) and the collected jejunum processed for the histochemistry NADPH-diaphorase technique. Whole-mount preparations were obtained for quantitative and morphometric analysis of the myenteric neurons. A quantity of jejunum neurons in the Group D (96±7.5) was not different (P>0.05) from Group DS (116±8.08), C (92±9.7), and CS (81±5.4), but in Group DS the quantity was higher (P<0.05) than in Group C and CS. The CBPA of neurons from Group D (189.50±2.68µm2) and DS (195.92±3.75µm2) were lower (P<0.05) than from Group C (225.13±4.37µm2) and CS (210.23±3.15µm2). The streptozotocin-induced DM did not change the jejunum-ileum area, the jejunum myenteric plexus space organization and the density of NADPH-dp neurons. The 50g AA-supplementation, three times a week, during 90 days, did not decrease hyperglycemia; however, it had a neuroprotective effect on the myenteric neurons, minimizing the increase on the CBPA of NADPH-dp neurons and increasing the amount of NADPD-dp neurons.
